The association between plants and arbuscular mycorrhizal fungi (AMF) can be used to bioremediate areas contaminated by metals. The objectives of this work were to evaluate the lead (Pb2+) phytoaccumulation capacity, morpho-physiology and nutrition responses of Vernonia polyanthes exposed to a solution amended with concentrations of lead nitrate and arbuscular mycorrhizal fungi. The treatments consisted of increasing doses of Pb2+ as lead nitrate [Pb(NO3)2], two strains of AMF and an absolute control without lead and AMF. Lead negatively affected some morphophysiological variables, reduced 27.3, 25.63, 30.60, and 56.60% shoot length, root collar diameter, number of leaves and leaf area, respectively, besides reducing decreasing chlorophyll a. Lead accumulated in the shoot and roots, the latter at the highest concentrations. However, the translocation factor was above 1, indicating low efficiency. The bioaccumulation factor referring to the roots were above 1. The fungi colonization rate was low, 3.31% for Gigaspora margarita and 2.33% for Acaulospora morrowiae. However, the absorption of lead increased, reflecting in lower values of chlorophyll a, dry mass of root and diameter. Results indicated that the arboreal species V. polyanthes tolerate high concentrations of lead and can accumulate significant amounts in the roots. AMF increase the accumulation of lead in the shoot and can be used in projects aimed at the phytoextraction of metals.

Abstract: This article describes the chemical composition of Vernonia chalybaea essential oil, and investigates its antimicrobial, antioxidant and hemolytic activities. The evaluation of the antifungal activity was performed by the broth microdilution method using strains of yeasts and dermatophytic fungi. The checkerboard technique to find antimicrobial modulatory effects was performed using ketoconazole as standard drug. The antioxidant activity was evaluated by DPPH scavenging assay and β-carotene/linoleic-acid system. The toxicity was characterized by the brine shrimp lethality test and hemolysis bioassays. The essential oil was obtained by hydrodistillation and analyzed by GC-MS method, showing to be rich in the sesquiterpenes β-caryophyllene (39.06%) and bicyclogermacrene (19.69%), and also demonstrated a relevant antifungal activity against strains of Trichophyton rubrum. In the modulatory activity assay, the essential oil of V. chalybaea and β-caryophyllene demonstrated a synergistic interaction with ketoconazole, with increasing of its antifungal action. The antioxidant activity was evidenced mainly by β-carotene/linoleic acid system, with IC50 value of 35.87 ± 0.32 µg/mL. The results suggest that V. chalybaea essential oil and β-caryophyllene are valuable natural medicinal agents with antioxidant and antimicrobial activities.

Objetivou-se avaliar a composição química e produtividade dos principais componentes do óleo essencial de Baccharis dracunculifolia DC. em função de doses de composto orgânico (0, 10, 20, 30, 40 e 50 t ha-1). Foi realizada uma colheita, aos 150 dias após o transplante das mudas. O óleo essencial, da massa seca útil da parte aérea, foi extraído por hidrodestilação e analisado em cromatógrafo a gás acoplado a espectrômetro de massas (Shimadzu, QP-5000). A identificação dos constituintes químicos foi realizada através da análise comparativa dos espectros de massas das substâncias com o banco de dados do sistema CG-EM (Nist 62.lib), literatura e índice de retenção. Os resultados foram submetidos à análise de variância pelo teste F, às médias obtidas foram submetidas à análise de regressão e o teste Tukey para o efeito das doses de composto orgânico. Os três componentes sesquiterpênicos, E-nerolidol, espatulenol e óxido de cariofileno, perfazem 58,44% da média relativa da composição química do óleo essencial de B. dracunculifolia, composto pela presença de 28 substâncias. Na produtividade dos componentes γ-muroleno, valenceno, δ-cadineno e E-nerolidol as dosagens estudadas influenciaram as plantas, que na dosagem 30 t ha-1 obtiveram os melhores resultados. Se o objetivo no cultivo de B. dracunculifolia for o componente espatulenol as dosagens 30 e 40 t ha-1 obtiveram os melhores resultados. Para a produtividade do componente óxido de cariofileno as dosagens estudadas influenciaram as plantas, que na dosagem 40 t ha-1 obtiveram os melhores resultados.


The objective was to evaluate the chemical composition and productivity of the main components of the essential oil of Baccharis dracunculifolia DC. due to organic compound (0, 10, 20, 30, 40 and 50 t ha-1). We performed a harvest, 150 days after transplanting. The essential oil of the dry mass useful in the aerial part was extracted by hydrodistillation and analyzed in gas chromatograph coupled to a mass spectrometer (Shimadzu, QP-5000). The identification of chemical constituents was performed by comparative analysis of mass spectra of substances with the database of the GC-MS (Nist 62.lib), literature and retention rates. The results were subjected to analysis of variance by F test, the averages obtained were subjected to regression analysis and Tukey test for the effect of doses of organic compound. The three components sesquiterpene, E-nerolidol, espatulenol and caryophyllene oxide, account for 58.44% of the average for the chemical composition of essential oil of B. dracunculifolia, composed by the presence of 28 substances. Productivity of components γ-murolene, valencene, δ-cadinene and E-nerolidol was influenced by doses, which in dose 30 t ha-1 obtained the best results. If the goal in the cultivation of B. dracunculifolia is the component espatulenol dosages 30 and 40 t ha-1 obtained the best results. To the productivity of caryophyllene oxide doses studied influenced the plants, which in dose 40 t ha-1 obtained the best results.

Vernonia scorpioides has been widely used in Brazil to treat skin problems and chronic wounds, such as ulcers of the lower limbs and diabetic lesions. In the present study, we investigated the effect of a dichloromethane (DCM) fraction of V. scorpioides leaf extract on Ehrlich ascitic and solid tumor-bearing mice. The animals were treated once a day with the DCM fraction at a concentration of 5 mg/kg, administered ip during and after the development of the tumor. The lifespan, weight, number and type of leukocytes, number of tumor cells, volume of solid and ascitic tumors were measured. The development of the tumor with pre-treated tumor cells in vitro with the DCM fraction (5 mg/kg) was analyzed and the animals were sacrificed after 7 days. The DCM fraction (5 mg/kg) totally inhibited tumor development when in direct contact with tumor cells, and also ascitic tumor development with in vitro treatment or when administered ip, in loco (after 7 days). Animals treated with the DCM fraction increased their lifespan ca. 2 weeks and maintained their body weight for 30 days. When applied immediately after the inoculation of the tumor cells in vivo, it totally abolished tumor development, with tumor development only decreasing when treatment was started 3 days after the tumor challenge. These data suggest an antineoplastic activity of the fraction. Oral or ip administration of DCM fraction (5 mg/kg) for 7 days did not reduce the solid tumor volume. The cytotoxic activity described here differs from the conventional immune suppressing profile of standard chemotherapy because it increases neutrophil influx to the peritoneal cavity. These results show that, besides exhibiting a tumoricidal activity, the DCM fraction also exhibits inflammatory activity.

RESUMO

Some informations about the sesquiterpene lactones isolated from Brazilian species of Vernonia are described, as well the results of tests developed with such compounds with respect to their anti-feedant, molluscicide, antimicrobial and analgesic properties.
