RESUMEN

22q11.2 deletion syndrome is a condition with complex multisystem involvement, and many clinicians will encounter patients living with the condition. 22q11.2 deletion syndrome is known to significantly increase the risk of psychosis, and there is some emerging evidence that 22q11.2 deletion syndrome may be associated with an increased risk of malignancy. We report on a case of an adolescent female who had a delayed diagnosis of 22q11.2 deletion syndrome after she developed severe psychosis at an early age. She was subsequently diagnosed in late adolescence with papillary thyroid carcinoma. This case contributes to the limited body of evidence regarding the treatment of psychosis secondary to 22q11.2 deletion syndrome and the potential increased risk of malignancy associated with the genetic condition.

Synergies to achieve high phosphorus (P) use efficiency (PUE) and mitigate greenhouse gas (GHG) emissions are critical for developing strategies aimed toward agricultural green development. However, the potential effects of such synergies in the entire P supply chain through optimizing P management in crop production are poorly understood. In this study, a partial life cycle of a GHG emissions model was developed to quantify the P-related GHG emissions in the entire P supply chain in China. Our results showed that 16.3 kg CO2-equivalent (CO2-eq) was produced from the entire P supply chain per unit of P used for grain agriculture (maize, rice, and wheat). P-related GHG emissions in China increased more than five-fold from 1980 (7.2 Tg CO2-eq) to 2018 (44.9 Tg CO2-eq). GHG emissions were found to be strongly associated with the intensity of grain production in China, and they varied considerably across production regions owing to the differences in the P fertilizer production efficiency. Mineral P fertilizer use in crop production was the primary source of P-related GHG emissions. The results suggest that sustainable P management by matching mineral P fertilizer rates and fertilizer types with crop needs can mitigate GHG emissions by 10.8-27.7 Tg (24.0-65.1%). Moreover, this can improve PUE and reduce mineral P input by 0.7-1.4 Tg (24.0-46.0%). These findings highlight that potential synergies between high PUE and low P-related GHG emissions can be achieved via sustainable P management, thereby enhancing green agricultural development in China and other regions worldwide.

An increasing number of studies have demonstrated that arbuscular mycorrhizal fungi can cooperate with other soil microorganisms, e.g., bacteria, which develop near or on the surface of the extraradical hyphae where they perform multiple functions. However, the mechanisms involved in this privileged relationship are still poorly known. In the present study, we investigated how the arbuscular mycorrhizal fungus Rhizophagus irregularis MUCL 43194 influences the three pace-making enzymes (i.e., citrate synthase, isocitrate dehydrogenase, and α-oxoglutarate dehydrogenase) of the tricarboxylic acid (TCA) cycle in the phosphate-solubilizing bacterium Rahnella aquatilis HX2. The study was conducted under strict in vitro culture conditions and analysis made at the transcriptional level. Results showed that R. irregularis induced the expression of the gene-encoding citrate synthase (gltA), the pace-making enzyme involved in the first step of the TCA cycle, in R. aquatilis at all time points of observation (i.e., 1, 6, 12, 24, 48, and 72 h). The expression of the gene-encoding isocitrate dehydrogenase (icd) significantly decreased at 6, 12, 24, 48, and 72 h and the expression of the gene-encoding α-oxoglutarate dehydrogenase E1 component (kgdhc) significantly increased at 1, 6, and 48 h. The above results suggested that R. irregularis may influence the level of adenosine triphosphate production in R. aquatilis and thus the metabolism of the bacterium by stimulating the expression of gltA involved in the TCA cycle. Our results suggest a fine-tuned dialog between R. irregularis MUCL 43194 and R. aquatilis HX2 and emphasize the complexity of the interactions that might take place at the hyphal surface of arbuscular mycorrhizal fungi hosting communities of microbes.

Diarrhoeal disease is responsible for 8.6% of global child mortality. Recent epidemiological studies found the protozoan parasite Cryptosporidium to be a leading cause of paediatric diarrhoea, with particularly grave impact on infants and immunocompromised individuals. There is neither a vaccine nor an effective treatment. Here we establish a drug discovery process built on scalable phenotypic assays and mouse models that take advantage of transgenic parasites. Screening a library of compounds with anti-parasitic activity, we identify pyrazolopyridines as inhibitors of Cryptosporidium parvum and Cryptosporidium hominis. Oral treatment with the pyrazolopyridine KDU731 results in a potent reduction in intestinal infection of immunocompromised mice. Treatment also leads to rapid resolution of diarrhoea and dehydration in neonatal calves, a clinical model of cryptosporidiosis that closely resembles human infection. Our results suggest that the Cryptosporidium lipid kinase PI(4)K (phosphatidylinositol-4-OH kinase) is a target for pyrazolopyridines and that KDU731 warrants further preclinical evaluation as a drug candidate for the treatment of cryptosporidiosis.

Plants associated with arbuscular mycorrhizal fungi (AMF) acquire phosphorus via roots and extraradical hyphae. How soil P level affects P accumulation within hyphae and how P in hyphae influences the accumulation of metal minerals remains little explored. A bi-compartmented in vitro cultivation system separating a root compartment (RC), containing a Ri T-DNA transformed carrot root associated to the AMF Rhizophagus irregularis DAOM 197198, from a hyphal compartment (HC), containing only the extraradical hyphae, was used. The HC contained a liquid growth medium (i.e., the modified Strullu-Romand medium containing P in the form of KH2PO4) without (0 µM) or adjusted to 35, 100, and 700 µM of KH2PO4. The accumulation of P and metal minerals (Ca, Mg, K, Na, Fe, Cu, Mn) within extraradical hyphae and AMF-colonized roots, and the expression of the phosphate transporter gene GintPT were assessed. The expression of GintPT in the extraradical hyphae did not differ in absence of KH2PO4 or in presence of 35 and 100 µM KH2PO4 in the HC but was markedly reduced in presence of 700 µM KH2PO4. Hyphal P concentration was significantly lowest in absence of KH2PO4, intermediate at 35 and 100 µM KH2PO4 and significantly highest in presence of 700 µM KH2PO4 in the HC. The concentrations of K, Mg, and Na were positively associated with the concentration of P in the extraradical hyphae developing in the HC. Similarly, P concentration in extraradical hyphae in the HC was related to P concentration in the growth medium and influenced the concentration of K, Mg, and Na. The accumulation of the metal mineral K, Mg, and Na in the extraradical hyphae developing in the HC was possibly related to their function in neutralizing the negative charges of PolyP accumulated in the hyphae.

Chronic stress can suppress natural killer (NK) cell activity; this may also be related to the effect of stress on the neuroendocrine-immune network. Sea buckthorn (SBT) (Hippophae rhamnoides L.) is a thorny nitrogen fixing deciduous shrub, native to both Europe and Asia. It has been used as a medicinal plant in Tibetan and Mongolian traditional medicines. SBT has multifarious medical properties, including anti-fatigue as well as immunoregulatory effects. This study reports the effects of SBT oil with regard to the cytotoxicity and quantity of NK cells in the blood of a chronic-stress rat model, in addition to its mechanisms on the neuroendocrine-immune network. These results show that SBT oil, given by gavage to rats with chronic stress, could increase the following: body weight, NK cell quantities, and cytotoxicity, as well as the expression of perforin and granzyme B. The results also show that SBT oil in rats with chronic stress could suppress cortisol, ACTH, IL-1ß and TNF-α levels, in addition to increasing 5-HT and IFN-γ serum levels. This leads to suggest that SBT oil, in rats with chronic stress, can increase NK cell cytotoxicity by upregulating the expression of perforin and granzyme B, thus causing associated effects of SBT oil on the neuroendocrine-immune network.

The effect of salinity on brown seeds/black seeds ratio, seed weight, endogenous hormone concentrations, and germination of brown and black seeds in the euhalophyte Suaeda salsa was investigated. The brown seeds/black seeds ratio, seed weight of brown and black seeds and the content of protein increased at a concentration of 500 mM NaCl compared to low salt conditions (1 mM NaCl). The germination percentage and germination index of brown seeds from plants cultured in 500 mM NaCl were higher than those cultured in 1 mM NaCl, but it was not true for black seeds. The concentrations of IAA (indole-3-acetic acid), ZR (free zeatin riboside) and ABA (abscisic acid) in brown seeds were much greater than those in black seeds, but there were no differences in the level of GAs (gibberellic acid including GA1 and GA3) regardless of the degree of salinity. Salinity during plant culture increased the concentration of GAs, but salinity had no effect on the concentrations of the other three endogenous hormones in brown seeds. Salinity had no effect on the concentration of IAA but increased the concentrations of the other three endogenous hormones in black seeds. Accumulation of endogenous hormones at different concentrations of NaCl during plant growth may be related to seed development and to salt tolerance of brown and black S. salsa seeds. These characteristics may help the species to ensure seedling establishment and population succession in variable saline environments.

AIM: To evaluate the concentrations of eight cytokines in order to identify potential biomarkers for assisting in the detection of colorectal cancer. MATERIALS & METHODS: The concentrations of IFN-γ, IL-10, IL-6, IL-8, TNF-α, MMP-2, MMP-7 and MMP-9 were detected in the sera of 69 healthy controls, 93 colorectal adenoma patients and 149 colorectal cancer (CRC) patients. RESULTS: Multivariate logistic regression analyses, which included CEA, CA199, IL-8, TNF-α and MMP-7, were used to evaluate the diagnostic value for differentiating between colorectal adenoma and CRC. The area under the curve was 0.945 (95% CI: 0.909-0.981). The sensitivity and specificity were 85.86 and 96.78%, respectively. Compared with the conventional biomarkers CEA and CA199, multivariate logistic regression showed significant improvement. CONCLUSION: Our data demonstrated that testing using a panel of three serum cytokines, CEA and CA199 may have strong potential to assist in the detection of CRC.

PABCREATIC neuroendocrine tumours are uncommon neoplasms of the pancreas. They may cause a clinical syndrome due to hormone overproduction. Glucagonoma is a rare kind of pancreatic tumors. Here we report a case of glucagonoma. Hypercalcemia occurred when the patient underwent octreotide acetate long-acting release.

Breast cancer surgical options now include nipple-sparing mastectomy (NSM), but there has been much controversy regarding the oncologic safety of the preserved nipple. This study evaluates frequency and patterns of occult nipple involvement in a large contemporary cohort of patients, aiming to improve patient selection for NSM. A total of 2323 consecutive mastectomy specimens with grossly unremarkable nipples were evaluated by sagittal sections through the entire nipple and subareolar tissue. Sixteen different clinical and tumor parameters were examined to predict cancerous nipple involvement. Nipple involvement was noted in 331 of 2323 (14.2%) mastectomy specimens. Occult nipple involvement rate was 10.7% (248 cases). Occult nipple involvement usually occurs as ductal carcinoma in situ. In univariate analysis, patient age, tumor size, tumor-to-nipple distance, tumor central location, tumor type, lymph node status, lymphatic vascular invasion, histologic grade, estrogen receptor, progesterone receptor, human epidermal growth factor receptor 2 (HER-2) amplification, and multicentric or multifocal tumor were associated with positive nipple involvement. By multivariate logistic regression analysis with the entire selection process, tumor size, tumor-to-nipple distance, central location, lymph node status, lymphatic vascular invasion, HER-2 amplification, and multicentric or multifocal tumor were shown to be associated with nipple involvement by carcinoma. Nearly 90% women undergoing mastectomy did not have occult nipple involvement. NSM may be a viable option in appropriate patient selection and setting. NAC preservation would be appropriate in HER-2 negative, axillary lymph node, and lymphatic vascular negative patients with small, solitary tumors located on the periphery of the breast.

Neural stem cells are multipotent progenitor cells that show self-renewal activity. In this study, we assessed the use of neural stem cells for ameliorating ischemia-reperfusion injury of the gerbil cochlea. Neural stem cells were injected into one inner ear through the round window 1 day after ischemic insult. Immunostaining for nestin showed that the distribution of neural stem cells was concentrated within the organ of Corti. Seven days after ischemia, the injury-induced auditory brainstem response threshold shift and progressive inner hair cell damage were markedly less on the neural stem cell-transplanted side. These results suggest that the transplantation of neural stem cells is therapeutically useful for preventing damage to hair cells that occurs after transient ischemia of the cochlea.
