RESUMEN

Nano zero-valent iron (nZVI) is widely used in soil remediation due to its high reactivity. However, the easy agglomeration, poor antioxidant ability and passivation layer of Fe-Cr coprecipitates of nZVI have limited its application scale in Cr-contaminated soil remediation, especially in high concentration of Cr-contaminated soil. Herein, we found that the carboxymethyl cellulose on nZVI particles could increase the zeta potential value of soil and change the phase of nZVI. Along with the presence of biochar, 97.0% and 96.6% Cr immobilization efficiency through CMC-nZVI/BC were respectively achieved in high and low concentrations of Cr-contaminated soils after 90-days remediation. In addition, the immobilization efficiency of Cr(VI) only decreased by 5.1% through CMC-nZVI/BC treatment after 10 weeks aging in air, attributing to the strong antioxidation ability. As for the surrounding Cr-contaminated groundwater, the Cr(VI) removal capacity of CMC-nZVI/BC was evaluated under different reaction conditions through column experiments and COMSOL Multiphysics. CMC-nZVI/BC could efficiently remove 85% of Cr(VI) in about 400 hr when the initial Cr(VI) concentration was 40 mg/L and the flow rate was 0.5 mL/min. This study demonstrates that uniformly dispersed CMC-nZVI/BC has an excellent remediation effect on different concentrations of Cr-contaminated soils.

RESUMEN

An efficient and selective method for cysteine conjugation utilizing para-quinone methides (p-QMs) was developed. p-QM labeling exhibits high specificity toward the cysteine residue, as evidenced by its reactivity with various amino acid derivatives, peptides, and proteins. Notably, the p-QM-cysteine reactions display robust kinetics with rate constants up to 1.67 × 104 M-1·s-1. Furthermore, p-QM conjugation enables us to attach a fluorescent probe to a HER2 nanobody, resulting in selective labeling of HER2-positive SK-BR-3 cells.

The properties of different organic fertilizers and their potential for stabilizing toxic metals(loids) in soil have not been fully investigated. This study characterized and evaluated three organic fertilizers from different raw materials. The mushroom residue organic fertilizer (MO) had higher C, H, and O contents and more functional groups (-OH, C-H, and C = O). Its application significantly increased pH (1.00 ~ 1.32 units), organic matter (OM) content (26.58 ~ 69.11%), and cation exchange capacity (CEC) (31.52 ~ 39.91%) of soil. MO treatments can simultaneously reduce the bioavailable TCLP-Cd and TCLP-As in soil, solving the difficulties of remediating the combined Cd and As pollution. MO treatments inhibited the migration of Cd and As from soil to plant, promoting plant growth. Redundancy analysis (RDA) revealed that metal(loid) variations in plants were related to soil properties (40.09%) and TCLP-Cd/As (44.74%). Furthermore, the toxic metals(loids) risk assessment for all organic fertilizers was at safe levels. This study provided a valuable reference for choosing organic fertilizers and presented a novel option for the "producing while remediating" of farmlands with low pollution.

Mitigating the adverse effects of stressful environments on crops and promoting plant recovery in contaminated sites are critical to agricultural development and environmental remediation. Iron-based nanomaterials (Fe NMs) can be used as environmentally friendly nano-fertilizer and as a means of ecological remediation. A meta-analysis was conducted on 58 independent studies from around the world to evaluate the effects of Fe NMs on plant development and antioxidant defense systems in stressful environments. The application of Fe NMs significantly enhanced plant biomass (mean = 25%, CI = 20%-30%), while promoting antioxidant enzyme activity (mean = 14%, CI = 10%-18%) and increasing antioxidant metabolite content (mean = 10%, CI = 6%-14%), reducing plant oxidative stress (mean = -15%, CI = -20%∼-10%), and alleviating the toxic effects of stressful environments. The observed response was dependent on a number of factors, which were ranked in terms of a Random Forest Importance Analysis. Plant species was the most significant factor, followed by Fe NM particle size, duration of application, dose level, and Fe NM type. The meta-analysis has demonstrated the potential of Fe NMs in achieving sustainable agriculture and the future development of phytoremediation.

Hexavalent chromium (Cr(VI)) is carcinogenic and widely presented in soil. In this study, modified zero-valent iron (ZVI) with oxalic acid on biochar (OA-ZVI/BC) was prepared using wet ball milling method for the remediation of Cr(VI)-contaminated soil. Microscopic characterizations showed that ZVI were distributed on the biochar uniformly and confirmed the enhanced interface interaction between biochar and ZVI by wet ball milling. Electrochemical analysis indicated the strong electron transfer ability and enhanced corrosion behavior of OA-ZVI/BC. Moreover, inhibitory efficiencies of Cr(VI) removal with the addition of 1,10-phenanthroline suggested abundant Fe2+ generation in OA-ZVI/BC, which might facilitate the reduction of Cr(VI) to Cr(III). Theory calculation further demonstrated the ZVI modified by oxalic acid was more susceptible to solid-solid interfacial reactions with Cr(VI), and more electrons were transferred to Cr(VI). When applied to Cr(VI)-contaminated soil, OA-ZVI/BC could passivate 96.7 % total Cr(VI) and maintained for 90 days. The toxicity characteristic leaching procedure (TCLP) and simple based extraction test (SBET) were used to evaluate the leaching toxicity and bioaccessibility of Cr(VI), respectively. The TCLP-Cr(VI) decreased to 0.11 mg·L-1 after OA-ZVI/BC treatment, much lower than that of soils with ZVI/BC and OA-ZVI remediation (1.5 mg·L-1 and 4.1 mg·L-1). The bioaccessibility of Cr(VI) reduced by 93.5 % after 3-month remediation. Sequential extraction showed that Cr fractions in the soil after OA-ZVI/BC remediation was converted from acetic acid-extractable (HOAc-extractable) to more stable forms (e.g., residual and oxidizable forms). Benefiting from the synergies of oxalic acid, biochar and wet ball milling, OA-ZVI/BC exhibited an excellent performance on the remediation of Cr(VI)-contaminated soil, whose mechanisms involved adsorption, reduction (Fe0/Fe2+, Fe2+/Fe3+) and co-precipitation. This study herein develops a promising ZVI technology in the remediation of heavy metal-contaminated soil.

A comprehensive assessment of the heavy metal system in the rehabilitated saline-alkali land holds significant importance, as the in-situ remediation process utilizing amendments substantially alters the initial physicochemical properties of the soil, which could lead to the migration or reactivation of previously stabilized heavy metals. In this context, the present study aims to evaluate the heavy metal content and health risk within the improved saline-alkali soil-plant system. Moreover, a comprehensive evaluation based on the TOPSIS-RSR method is carried out to accurately gauge the soil health status. The findings indicate that the modification process has an impact on the concentrations of heavy metals in the soil and crops, causing either an increase or decrease. However, the level of heavy metal pollution in the improved saline-alkali soil and rape remains within safe limits. The results of the migration of heavy metals after amendment application indicated that the migration of heavy metals in the soil was influenced by the properties of the heavy metals, the composition of the amendment, and leaching. Furthermore, the total non-carcinogenic hazard quotients in the soil and rape were within the safe threshold for all populations. The findings provided novel insights into the status and risk assessment of the pollution of improved saline-alkali soil.

In order to determine the ideal conditions for the application of nanoscale zero-valent iron (nZVI) in agricultural production, this review studies the effects of nZVI application on plant physiological parameters, presents its mechanism and prospective outcomes. In this research, it was observed that the application of nZVI had both favorable and unfavorable effects on plant growth, photosynthesis, oxidative stress, and nutrient absorption levels. Specifically, the application of nZVI significantly increased the biomass and length of plants, and greatly reduced the germination rate of seeds. In terms of photosynthesis, there was no significant effect for the application of nZVI on the synthesis of photosynthetic pigments (chlorophyll and carotenoids). In terms of oxidative stress, plants respond by increasing the activity of antioxidant enzyme under mild nZVI stress and trigger oxidative burst under severe stress. In addition, the application of nZVI significantly increased the absorption of nutrients (B, K, P, S, Mg, Zn, and Fe). In summary, the application of nZVI can affect the plant physiological parameters, and the degree of influence varies depending on the concentration, preparation method, application method, particle size, and action time of nZVI. These findings are important for evaluating nZVI-related risks and enhancing nZVI safety in agricultural production.

How to manage potato peel waste sustainably has been an issue faced by the potato industry. This work explored the feasibility of potato peel waste for biohydrogen production via dark fermentation, and investigated the effects of various inoculum enrichment methods (acid, aeration, heat-shock and base) on the process efficiency. It was observed that the hydrogen production showed a great variation when using various inoculum enrichment methods, and the aeration enriched inoculum obtained the maximum hydrogen yield of 71.0 mL/g-VSadded and VS removal of 28.9 %. Different enriched cultures also exhibited huge variations in the bacterial community structure and metabolic pathway. The highest abundance of Clostridium sensu stricto fundamentally contributed to the highest process efficiency for the fermenter inoculated with aeration treated culture. This work puts forward a promising strategy for recycling potato peel waste, and fills a gap in the optimal inoculum preparation method for biohydrogen fermentation of potato peel waste.

The quality of agricultural soils is important for agricultural production and food safety. The contamination of agricultural soils by heavy metal(loid)s (HMs) has aroused global attention. Fifty-two topsoil samples with 8 HMs were gathered to assess the health risks of farmland soil in Huairou District, Beijing. As a significantly enriched pollutant, the results revealed that Hg had greater ecological risks relative to other HMs. We found that the positive matrix factorization (PMF) model appears to be more physically plausible in identifying complex pollution sources compared to the absolute principal components score-multiple linear regression (APCS-MLR) model, which had a higher fit coefficient (r2 = 0.69-0.99). Five HMs from pollution sources, including agricultural activities, traffic source, natural source, fuel burning, and industrial production, were identified by integrating the PMF model with Pearson's correlation analysis, revealing corresponding contribution rates of 29.40%, 22.54%, 20.16%, 15.20%, and 12.70%, respectively. The probabilistic health risk evaluation results showed an absence of non-carcinogenic risks in all populations, but the carcinogenic risk could not be ignored, especially in children. In addition, the source-oriented health risks showed that agricultural activities made the largest contribution to the health risks of all populations. This research provides scientific evidence for preventing HMs contamination and control of farmland.

Aberrant activation of the Wnt signaling pathway has been identified in numerous types of cancer. One common feature of oncogenic Wnt regulation involves an increase in the cellular levels of ß-catenin due to interference with its constitutive ubiquitin-dependent degradation. Targeting ß-catenin has therefore emerged as an appealing approach for the treatment of Wnt-dependent cancers. Here, we report a strategy that employs multifunctional stapled peptides to recruit an E3 ubiquitin ligase to ß-catenin, thereby rescuing ß-catenin degradation by hijacking the endogenous ubiquitin-proteasome pathway. Specifically, we designed, synthesized, and evaluated a panel of multifunctional stapled peptides that have a ß-catenin binding moiety (StAx-35) covalently linked to a second stapled peptide moiety (SAH-p53-8), which is capable to interact with the E3 ubiquitin ligase MDM2. We found that in vitro these multifunctional peptides can recruit the MDM2 protein to ß-catenin and induce poly-ubiquitination on ß-catenin. In cellulo, treatment of the human colorectal cancer cell line SW480 with the multifunctional stapled peptides showed dose-dependent degradation of endogenous ß-catenin levels. In addition, a luciferase reporter assay showed that the multifunctional stapled peptides can suppress ß-catenin-mediated gene expression via the Wnt signaling pathway. Therefore, these multifunctional stapled peptides provide a unique research tool for examining the Wnt signaling pathway by targeted knockdown of ß-catenin at the protein level, and may serve as leads for potential drug candidates in the treatment of Wnt-dependent cancers.

Chemotherapy remains the primary treatment of advanced solid cancer, including lung cancer. However, as first-line treatment, cisplatin-based therapy is restricted by the frequent development of drug resistance. Increasing data showed that the programmed cell death protein ligand 1 (PD-L1) plays a vital role in regulating cisplatin resistance. However, the underlying mechanisms are not fully understood. We found that miR-526b-3p expression declined while PD-L1 was elevated in cisplatin-resistant lung cancer compared to that in cisplatin-sensitive lung cancer by analyzing clinical samples. Significantly, miR-526b-3p was associated with response to cisplatin negatively. We further demonstrated that miR-526b-3p reversed cisplatin resistance, suppressed metastasis, and activated CD8+ T cells in a STAT3/PD-L1-dependent manner. Thus, our findings extended the knowledge of PD-L1-mediated cisplatin resistance of lung cancer. In addition, the introduction of miR-526b-3p provided a new clue to improve the anti-tumor effects of the combination of immunotherapy and chemotherapy.

PURPOSE: In order to solve the problem of accurate and effective segmentation of the patient's lung computed tomography (CT) images, so as to improve the efficiency of treating lung cancer. METHOD: We propose a U-Net network (DC-U-Net) fused with dilated convolution, and compare the results of segmented lung CT with DC-U-Net, Otsu and region growth. We use Intersection over Union (IOU), Dice coefficient, Precision and Recall to evaluate the performance of the three algorithms. RESULTS: Compared with the common segmentation algorithm Otsu and region growing, the segmented image of DC-U-Net is closer to the Ground truth. The IOU of DC-U-Net is 0.9627, and the Dice coefficient is 0.9743, which is close to 1 and much higher than the other two algorithms. CONCLUSION: We propose that the model can directly segment the original image automatically, and the segmentation effect is good. This model speeds up the segmentation, simplifies the steps of medical image segmentation, and provides better segmentation for subsequent lung blood vessels, trachea and other tissues.

PURPOSE: Esophageal cancer is a common malignant tumor in life, which seriously affects human health. In order to reduce the work intensity of doctors and improve detection accuracy, we proposed esophageal cancer detection using deep learning. The characteristics of deep learning: association and structure, activity and experience, essence and variation, migration and application, value and evaluation. METHOD: The improved Faster RCNN esophageal cancer detection in this paper introduces the online hard example mining (OHEM) mechanism into the system, and the experiment used 1520 gastrointestinal CT images from 421 patients. Then, we compare the overall performance of Inception-v2, Faster RCNN, and improved Faster RCNN through F-1 measure, mean average precision (mAP), and detection time. RESULTS: The experiment shows that the overall performance of the improved Faster RCNN is higher than the other two networks. The F-1 measure of our method reaches 95.71%, the mAP reaches 92.15%, and the detection time per CT is only 5.3s. CONCLUSION: Through comparative analysis on the esophageal cancer image data set, the experimental results show that the introduction of online hard example mining mechanism in the Faster RCNN algorithm can improve the detection accuracy to a certain extent.

Long-term retention and accumulation of heavy metals in rivers pose a great threat to the stability of ecosystems and human health. In this study, Beiyun River was taken as the example to quantitatively identify pollution sources and assess the pollution source-oriented health risk. A total of 8 heavy metals (Mn, Ni, Pb, Zn, As, Cr, Cd, and Cu) in Beiyun River were measured. Ordinary kriging (OK) and inverse distance weight (IDW) methods were used to predict the distribution of heavy metals. The results showed that the OK method is more accurate, and heavy metal pollution in the midstream and downstream is much more serious than that in the upstream. Principal component analysis-multiple linear regressions (PCA-MLR) and positive matrix factorization (PMF) methods were used to quantitatively identify pollution sources. The coefficient of determination (R2) of PMF is closer to 1, and the analyzed pollution source is more refined. Furthermore, the result of source identification was imported into the health risk assessment to calculate the hazard index (HI) and carcinogenic risk (CR) of various pollution sources. The results showed that the HI and CR of As and Ni to local residents were serious in the Beiyun River. Industrial activities (23.0%) are considered to be the largest contribution of heavy metals in Beiyun River, followed by traffic source (17%), agricultural source (16%), and atmospheric deposition (16%). The source-oriented risk assessment indicated that the largest contribution of HI and CR is agricultural source in the Beiyun River, followed by industrial activities. This study provides a "target" for the precise control of pollution sources, which is of great significance for improving the fine management of the water environment in the basin.

Triple-negative breast cancer (TNBC) is one of the most highly invasive and metastatic breast cancers without safe and effective therapeutic drugs. The natural product oridonin is reported to be a potential anti-TNBC agent. However, its moderate activity and complex structure hampered its clinical application. In this study, the novel oridonin analogues were first identified by removal of multiple hydroxyl groups and structural simplification of oridonin. The representative analogue 20 exhibited potent anticancer effects. Further structural modification on 20 generated the most potent derivative 56, which possessed 120-fold more potent antiproliferative activity than oridonin in the TNBC cell line HCC1806. Importantly, compound 56 exhibited more potent anticancer activity than paclitaxel in TNBC xenograft nude mice. Moreover, 56 could attenuate the expression of MMP-2, MMP-9, p-FAK, and integrin ß1 to inhibit TNBC cell metastasis. All results suggest that compound 56 may warrant further investigation as a promising candidate agent for the treatment of TNBC.

The chitosan-stabilized ferrous sulfide nanoparticles were loaded on biochar to prepare a composite material FeS-CS-BC for effective removal of hexavalent chromium in water. BC and FeS-CS-BC were characterized by Brunauer-Emmett-Teller (BET), scanning electron microscopy (SEM), X-ray diffraction (XRD), and Fourier transform infrared spectroscopy (FTIR) analyses. Batch experiments were employed to evaluate the Cr(VI) removal performance. The experimental results showed that the removal rate of Cr(VI) by FeS-CS-BC(FeS:CS:BC = 2:2:1) reached 98.34%, which was significantly higher than that of BC (44.58%) and FeS (79.91%). In the pH range of 2-10, the removal of Cr(VI) by FeS-CS-BC was almost independent of pH. The limitation of coexisting anions (Cl-、SO42-、NO3-) on Cr(VI) removal was not too obvious. The removal of Cr(VI) by FeS-CS-BC was fitted with the pseudo-second-order dynamics, which was a hybrid chemical-adsorption reaction. The X-ray photoelectron spectroscopy (XPS) analysis result showed that Cr(VI) was reduced, and the reduced Cr(VI) was fixed on the surface of the material in the form of Cr(VI)-Fe(III). In this study, corn stalk was modified by manganese (Mn) before (MBC1) and after (MBC2) pyrolysis at different temperatures (400~600 °C) under anaerobic conditions for Cd sorption in both water and soil. Batch experiments in aqueous solution were conducted to evaluate the optimum sorption capability by biochar with and without manganese-modified. Both types of manganese modification can improve the sorption capacity of Cd(II) on biochar, which is superior to the corresponding pristine biochar without modification, especially, pyrolyzed at 500 °C with 5:1 modification ratio. Under the optimal preparation conditions, the sorption percentage on MBC2 was 11.01% higher than that of MBC1. The maximum sorption capacity of MBC2 was 191.94 mg g-1 calculated by isotherm model. The performance of MBC2 was also verified in soil stabilization experiments in Cd-contaminated soil. We can conclude from the results of BCR extraction that all the application rates of MBC2 (1%, 2%, and 3%) can reduce the mild acid-soluble fraction Cd. The reducible, oxidizable, and residual fraction Cd showed an upward trend, thus controlling the migration, transformation, and enrichment of Cd in soil. The characteristic analysis showed biochar has more irregular fold and more particle-aggregated surface after modification. The main components of these aggregated particles are manganese oxides (MnOx) with high sorption capacity, such as the MnOx crystal structure loaded on MBC2 is a mixed structure of δ-MnO2 and MnO. However, these particles may block the biochar pores, or some of the pores may collapse at high temperatures during the modification process. The specific surface area was reduced, even if the sorption effect of MBC was strongly enhanced. Meanwhile, under the action of the secondary pyrolysis of MBC2 modification process, the MBC2 has a higher degree of aromatization with more potential active sorption sites for Cd. The study concluded that the MBC2 could be a promising amendment for Cd in both water and soil real field applications.

In this study, polyethylene glycol (PEG)-stabilized nano zero-valent iron (nZVI) supported by biochar (BC) (PEG-nZVI@BC) was prepared to remedy Cr(VI) with high efficiency. The morphology, functional groups, and crystalline structure of PEG-nZVI@BC composites were characterized, revealing that when PEG was added, a large number of -OH functional groups were introduced, and nZVI was effectively dispersed on the BC surface with a smaller particle size. The results of Cr(VI) remediation experiments showed Cr(VI) removal rate by PEG-nZVI@BC (97.38%) was much greater than that by BC-loaded nZVI (nZVI@BC) (51.73%). The pseudo second-order and Sips isotherm models provide the best simulation for Cr(VI) removal experimental data, respectively. The main remediation mechanism of Cr(VI) was reduction and co-precipitation of Cr-containing metal deposits onto PEG-nZVI@BC. Ecotoxicity assessment revealed PEG-nZVI@BC (1.00 g/L) has little influence on rice germination and growth, but resisted the toxicity of Cr(VI) to rice. The modified Community Bureau of Reference (BCR) sequential extraction showed pyrolysis could increase the percentage of oxidizable and residual Cr and diminish the environmental risk of Cr release from post-removal composites.

Nowadays, the remediation of polycyclic aromatic hydrocarbons (PAHs)-contaminated soil has received wide attention. In this work, Ochrobactrum sp. (PW) was isolated through selective enrichment from PAHs-contaminated soil in coking plant of Beijing, and the effects of PW on phytoremediation of that soil by alfalfa (Medicago sativa L.) and ryegrass (Lolium multiflorum Lam.) were investigated through pot experiments. Plant biomass, peroxidase (POD) activity, malondialdehyde (MDA) contents, soil enzyme activity (polyphenol oxidase and dehydrogenase activity), and residual concentration of PAHs in soils were determined to illustrate the ability of PW for enhancing the degradation of PAHs by plants. The results showed that the fresh weight of ryegrass and alfalfa inoculated with PW was significantly (p < 0.05) increased while the activity of POD and MDA contents were notably (p < 0.05) reduced than that without inoculation. Additionally, PW enhanced the activity of polyphenol oxidase and dehydrogenase in soil significantly (p < 0.05), and further enhanced the degradability of the system to PAHs. Different treatment methods could be ranked by the following order according to the degradability: SP (alfalfa + PW) > RP (ryegrass + PW) > PW (PW) > S (alfalfa) > R (ryegrass). The combined action of PW and alfalfa/ryegrass could accelerate the degradability of PAHs from soil contaminated by coking plants. PW could be used as potential bacteria to promote phytoremediation of the soil contaminated by PAHs.

Nanometer zero-valent iron (nZVI) has been widely used in the treatment of heavy metals such as hexavalent chromium (Cr(VI)). A novel composite of bead string-structured nZVI on modified activated carbon (nZVI-MAC) is prepared here, using polyethylene glycol as the stable dispersant rather than traditional ethanol during the loading process. The microstructure characterization shows that nZVI particles are loaded on MAC with a bead string structure in large quantity and stably due to the addition of hydroxyl functional groups on the surface by polyethylene glycol. Experiments on the treatment of Cr(VI) in wastewater show that the reaction process requires only 20 min to achieve equilibrium. The removal rate of Cr(VI) with a low concentration (80-100 mg/L) is over 99% and the maximum saturation removal capacity is up to 66 mg/g. The system converts Cr(VI) to trivalent chromium (Cr(III)) through an oxidation-reduction effect and forms an insoluble material with iron ions by coprecipitation, which is then adsorbed on the surface of the nZVI-MAC. The process conforms to the quasi-second order adsorption kinetics equation (mainly chemical adsorption process).
