RESUMEN

Children can get affected by polycyclic aromatic hydrocarbons (PAHs) while they interact with play area soil/rubber surfacing and exposed to PAHs by dermal contact, inhalation and hand-to-mouth activity. A comparative study has been conducted on PAHs profiling and probable cancer risk of children from PAHs present in uncovered playground surface soil and poured rubber surfaced playground dust. Surface soil and dust samples have been collected from 14 different children parks around the Korea University campus, Seoul, Republic of Korea. Concentrations of 16 PAHs in the soils/dust were found to be in a range of 2.82-57.93 µg g-1. Profiling of the PAHs from the playground soils/dust reveals 3-ring PAHs are dominating with 79.9% of total PAHs content, on an average. The diagnostic ratio analysis confirms that vehicular exhaust and fossil fuel burning are likely the main sources of high molecular weight carcinogenic PAHs, whereas low molecular weight PAHs have pyrogenic origin. The probabilistic health risk assessment using Monte Carlo simulations for the estimation of the 95% cancer risk exposed to the PAHs from the surfaced playgrounds shows a little higher value than the USEPA safety standard (1.3 × 10-5). Sensitivity analysis revealed exposure duration and relative skin adherence factor for soil as the most influential parameters of the assessment. Noticeably, cancer risk is approximately 10 times higher in poured rubber surfaced playgrounds than in uncovered soil playgrounds.

RESUMEN

Hydroxyethyl starch (HES) solutions are synthetic non-protein colloid solutions used to treat hypovolemia. However, their use is not free from the risk of allergic reactions. A 42-year-old male was scheduled to undergo aortic-iliac-femoral bypass surgery for the treatment of arteriosclerosis obliterans. He had no history of allergy. Two hours after the start of surgery, and within minutes after HES administration, facial erythema, hypotension and bronchospasm developed. HES infusion was discontinued under the estimation of anaphylaxis. The patient received phenylephrine, ephedrine, diphenhydramine and hydrocortisone with hydration. After restoration of vital signs, surgery was performed without complications.

Scutellaria barbata D. Don (Lamiaceae; SB) inhibited the growth of leiomyomal cells (LM). A time-dependent antiproliferative effect was noted when 10(-5) m buserelin, gonadotrophin-releasing hormone (GnRH) agonist or 20-40 microg/mL SB was added. The inhibition of cell growth decreased with the addition of the PKC activator (12-O-tetradecanoylphorbor-13-acetate; TPA) much as it did with the addition of SB, and the decreases in the viable cells caused by the addition of SB were reversed completely by pretreatment with a protein kinase C (PKC) inhibitor (calphostin C). The findings suggest that SB inhibits cell proliferation in cultured human uterine leiomyoma cells accompanied by PKC activation. Next, the study investigated the effect of SB on fetal development for toxicity. Pregnant Sprague-Dawley rats, from gestation day 6-15, were administered 20 g/L or 50 g/L SB in the drinking water and then killed on day 20. No maternal toxicity was observed, however, embryonic loss in the treatment groups was double that of the controls (p < 0.05). No gross morphologic malformations were seen in the treated fetuses. Fetuses exposed to SB were found to be significantly heavier than the controls, an effect that was greater in female fetuses and was not correlated with increased placental size. The results suggest that the SB had no toxicity and that in utero exposure to SB resulted in increased early embryo loss with increased growth in surviving fetuses. On the other hand, Western blot analyses revealed that Bcl-2 protein of a 26 kDa was abundant in leiomyomal cells, but not in normal myometrial cells. The addition of progesterone (100 ng/mL) resulted in a striking increase in Bcl-2 protein expression in the cultured leiomyoma cells. However, the addition of SB (20 microg/mL) resulted in a significant reduction in Bcl-2 protein expression in the cells. The results indicated that human uterine leiomyomal cells express Bcl-2 protein and progesterone enhances its expression, however, SB reduces the expression of Bcl-2 protein in human uterine leiomyoma cells.
