BACKGROUND: Ameloblastoma and ameloblastic carcinoma are epithelial odontogenic tumors that can be morphologically similar. In the present study, we evaluated the DNA content and Ki-67 index in the two tumors. METHODS: The paraffin blocks of the tumors were selected to obtain sections for the immunohistochemical reactions and preparation of the cell suspension for acquisition in a flow cytometer. The Random Forest package of the R software was used to verify the contribution of each variable to classify lesions into ameloblastoma or ameloblastic carcinoma. RESULTS: Thirty-two ameloblastoma and five ameloblastic carcinoma were included in the study. In our sample, we did not find statistically significant differences in Ki-67 labeling rates. A higher fraction of cells in 2c (G1) was correlated with the diagnosis of ameloblastoma, whereas higher rates of 5c-exceeding rate (5cER) were correlated with ameloblastic carcinoma. The Random Forest model highlighted histopathological findings and parameters of DNA ploidy study as important features for distinguishing ameloblastoma from ameloblastic carcinoma. CONCLUSION: Our findings suggest that the parameters of the DNA ploidy study can be ancillary tools in the classification of ameloblastoma and ameloblastic carcinoma.

BACKGROUND: Establishing the risk of malignant transformation (MT) in oral leukoplakia is usually based on grading oral epithelial dysplasia (OED) on biopsy tissue, for which two systems are proposed: a 3-tier and a binary system. Only very few actuarial studies have tested the accuracy of such methods in predicting MT, especially for the binary system. This study aimed to assess the accuracy of the two grading systems in predicting MT in a cohort of oral leukoplakia (OL) from Brazil, with follow-up data. METHODS: The sample comprised 878 individuals diagnosed with OL from 2005 to 2018. Follow-up data were obtained both locally and from the regional cancer registry. All lesions were graded using both the 3-tier and the binary systems. Kaplan-Meier curves (Log-rank Mantel-Cox) were used to assess risk and kappa to assess interobserver agreement. RESULTS: Thirty-five individuals underwent MT (4%). Both systems demonstrated prognostic value, though the 3-tier system proved superior, with OR 9.23 (3.42-23.69), PPV 0.152, NPV 0.98, compared to binary OR 3.49 (1.79-6.79), PPV 0.079, NPV 0.976. Interobserver agreement was also superior in the 3-tier system (0.47, p < 0.05) compared to the binary system (0.139, p = 0.39). Combining the two systems enhanced prognostic values (OR 14.28, PPV 0.217, NPV 0.981). CONCLUSION: The 3-tier system presented superior prognostic value to the binary system. Combining both systems to double-grade intermediate lesions might enhance risk assessment.

Despite many progresses in the development of new systemic therapies for oral squamous cell carcinoma (OSCC), the five-year survival rate of OSCC is low. The traditional chemotherapies approach (cisplatin - CDDP) shows some limitations like drug toxicity, limited efficacy, and drug resistance. Promising studies suggested OSCC cancer stem cells (CSC) presented resistance to CDDP. We have previously studied many targets, and we extensively showed the efficacy of the NFκB signaling and the role of histones acetylation, on different malignant tumors, including adenoid cystic carcinoma and mucoepidermoid carcinoma, but until then the effects of the NFkB inhibitor and histone deacetylase (HDAC) inhibitor on the biology of OSCC were not evaluated. Here we assessed the pharmacological inhibitor of NFκB emetine and HDAC inhibitor SAHA on the behavior of CSC derived from OSCC. Our data suggested that CDDP administration resulted in reduced viability of bulk OSCC cells and increased CSC. A single and isolated shot of emetine and SAHA were able to disrupt CSC by inhibiting the NFκB pathway and increasing the histone acetylation levels, respectively. Further, the combined administration of emetine and SAHA presented the same CSC disruption as seen in emetine alone.

There is currently no clear understanding on the pathways involved in the process of cell inhibition by photobiomodulation (PBM). The present study evaluated the influence of PBM on the expression of autophagy markers in vitro in an in situ model of oral carcinoma. Oral squamous cell carcinoma (Cal27) and stromal fibroblasts (FG) cultures were used. The independent variables were 'cell type' (FG and CAL27) 'culture condition' (monocultures or co-cultures) and PBM (placebo and 36 J/cm2). The cultures were irradiated from a red LED source for mRNA expression and protein expression analyses. The autophagy markers evaluated were Beclin-1, LC3B and p62 as well as adjuvant markers (BAX Bcl-2, VEGF, CD105, CD34, PRDX1, PRDX4 and GRP78). The Cal27 cells upregulated the autophagy markers upon exposure to PBM both at the mRNA and protein expression levels, providing evidence to explain malignant cell inhibition by PBM.

Objective: This article reports the case of a patient with oral manifestation of coronavirus disease 2019 (COVID-19) treated with photobiomodulation (PBM) and photodynamic therapy (PDT). Background: Some dermatological and oral mucosal lesions have recently been linked to severe acute respiratory syndrome coronavirus 2 infection. Methods: A 65-year-old female patient with a confirmed real-time reverse transcriptase-polymerase chain reaction diagnosis of COVID-19 presented with dry edematous lips, edema with mucosal desquamation, ulceration and blood crust on the inner aspect of the lips, gingival petechiae and erythematous and pseudomembranous lesions on the dorsum of the tongue. The treatment protocol was three sessions of antimicrobial PDT (aPDT) (660 nm diode laser + methylene blue) to the lips and tongue, every 24 h to control contamination, followed by PBM (low-power laser, 100 mW, 2 J/point) to the lips, tongue, and oral mucosa for additional four sessions every 24 h. Results: Therapy association promoted pain control and healing of oral mucosal lesions in 7 days of treatment. Conclusions: PBM and aPDT could be an interesting approach to manage COVID-19 patients.

ABSTRACT Melatonin (MLT) is a potential signaling molecule in the homeostasis of bone metabolism and may be an important mediator of bone formation and stimulation. The aim of this in vitro study was to evaluate the effect of MLT on the viability, mRNA/protein expression and mineralization of pre-osteoblastic cells. The concentrations 5, 2.5, 1, 0.1 and 0.01 mM MLT were tested on pre-osteoblastic cells (MC3T3) compared to control (no MLT), evaluating proliferation and cell viability (C50), gene expression (RT-PCR) and secretion (ELISA) of COL-I and OPN at 24h, 48h and 72h, and the formation of mineral nodules (alizarin red and fast red) after 10 days of treatment. MLT at 5 and 2.5 mM proved to be cytotoxic (C50), so only 0.01, 0.1 and 1 mM were used for the subsequent analyses. OPN mRNA expression increased with MLT at 0.1 mM - 1 mM, which was followed by increased secretion of OPN both at 24h and 72h compared to the remaining groups (p <0.05). COL-I mRNA and COL-1 secretion followed the same pattern as OPN at 0.1 mM MLT at 72h of treatment (p <0.05). Regarding mineralization, all MLT doses (except 1mM) caused an increase (p <0.05) in the formation of mineral nodules compared to the control. Melatonin at 0.01mM - 1mM had a stimulatory effect on osteoblasts by upregulating COL-I and OPN expression/ secretion and mineralization, thereby fostering osteogenesis.


RESUMO A melatonina (MLT) é uma molécula potencial de sinalização na homeostase do metabolismo ósseo e pode ser um importante mediador da formação e estimulação óssea. O objetivo deste estudo in vitro foi avaliar o efeito da MLT na viabilidade, na expressão do mRNA da proteína e mineralização de células préosteoblásticas. As concentrações de MLT 5, 2,5, 1, 0,1 e 0,01 mM foram testadas em células pré-osteoblásticas da linhagem MC3T3 em comparação ao controle (sem MLT), avaliando a proliferação e a viabilidade celular (C50), expressão gênica (rtPCR) e secreção (Elisa) de Colágeno tipo 1 (COL-I) e osteopontina (OPN) às 24, 48 e 72 horas, além da formação de nódulos minerais por meio do teste vermelho de Alizarina fast red após 10 dias de tratamento. MLT a 5 e 2,5 mM provou ser tóxico (C50). Portanto, as concentrações de 0,01, 0,1 e 1 mM foram utilizadas para as análises subsequentes. A expressão do mRNA da OPN aumentou com MLT a 0,1 mM-1mM, seguida pela secreção aumentada de OPN às 24 e 72 horas em comparação aos demais grupos (p<0,05). O mRNA de COL-I e a secreção de COL-I seguiram o mesmo padrão do OPN a 0,1 mM de MLT em 72 horas de tratamento (p<0,05). Em relação à mineralização, todas as doses de MLT (exceto 1mM) causaram aumento (p<0,05) na formação de nódulos minerais em comparação ao controle. A MLT na concentração entre 0,01mM a 1 mM teve um efeito estimulador sobre os osteoblastos, ao regular positivamente a expressão e secreção de COL-I e OPN, além da mineralização, favorecendo a osteogênese.

Abstract This report describes the diagnostic process of a rare disorder in a Brazilian female child. The patient presented initially as a 7-year-old with multiple whitish submucosal nodules of a fibrous consistency in the lower lip, but with an inconclusive pathology report. When she turned 9 years of age, she presented with exacerbation of the original clinical findings, which then involved the upper lip, buccal mucosa, tongue and lingual frenulum. In addition, dermatological lesions were noted on the child's limbs and face, as well as a hoarse voice. Histopathological examination of the buccal mucosa revealed dense connective tissue with hyaline foci, which were positive with periodic acid-Schiff (PAS) staining and resistant to diastase digestion. Clinical and histopathological findings led to the diagnosis of a rare genetic disease with fewer than 300 reported cases - lipoid proteinosis. Magnetic resonance imaging revealed calcium deposits in her amygdaloid region of the brain, and nasopharyngolaryngoscopy revealed lesions in her vocal cords. The patient currently is stable and under multidisciplinary follow-up, but no treatment has been recommended to date.


Resumo Este relato descreve o processo diagnóstico de uma doença rara em uma criança brasileira do sexo feminino. A paciente, inicialmente com 7 anos de idade, apresentava múltiplos nódulos submucosos esbranquiçados, de consistência fibrosa, no lábio inferior, mas com um laudo patológico inconclusivo. Quando completou 9 anos de idade, ela apresentou exacerbação dos achados clínicos originais, que envolveram o lábio superior, mucosa bucal, língua e frênulo lingual. Além disso, lesões dermatológicas foram observadas nos membros e no rosto da criança, assim como rouquidão. O exame histopatológico da mucosa bucal revelou tecido conjuntivo denso com focos hialinos, que foram positivos com coloração periódica com ácido-Schiff (PAS) e resistente à digestão da diástase. Os achados clínicos e histopatológicos levaram ao diagnóstico de uma doença genética rara com menos de 300 casos relatados - proteinose lipoide. A ressonância magnética revelou depósitos de cálcio em amígdala cerebral e a nasofaringolaringoscopia revelou lesões em cordas vocais. Atualmente, a paciente está estável e em acompanhamento multidisciplinar, mas nenhum tratamento foi recomendado até o momento.

Current management of oral potentially malignant disorders is careful monitoring. Unfortunately, the 'watch and wait' approach only generates anxiety and a feeling of powerlessness, especially to those caring for patients. Photobiomodulation (PBM) has emerged as a potential strategy to inhibit possible transforming cells. The aim of this study was to investigate the effect of LED-based PBM on the progression of malignant invasion into a fibroblast-based stroma. An in vitro model of carcinoma in situ (CIS) containing stromal fibroblasts and carcinoma cells in co-culture was used to study the effect of PBM on the expansion of CIS colonies. A second model of co-culture (cells separated by membrane), was used to study cell counts, viability and apoptosis following PBM at high doses (36 J/cm2). The data was analyzed using Kruskal-Wallis, Dunn's test and non-linear regression, wherever appropriate. PBM was able to inhibit the expansion of CIS colonies as well as the total number of colonies after 72 h of treatment (p < 0.05). Cell viability, apoptosis and death assays revealed an overall advantage of stromal fibroblasts over carcinoma cells after high-dose PBM. In conclusion, LED-based PBM at high doses inhibited the progression and number of oral squamous cell carcinoma colonies without affecting the surrounding stromal fibroblasts in vitro.

BACKGROUND: Adenoid cystic carcinoma (AdCC) and polymorphous adenocarcinoma (PAC) are included among the most common salivary gland cancers. They share clinical and histological characteristics, making their diagnosis challenging in specific cases. MicroRNAs (miRNA) are short, non-coding RNA sequences of 19-25 nucleotides in length that are involved in post-transcriptional protein expression. They have been shown to play important roles in neoplastic and non-neoplastic processes and have been suggested as diagnostic and prognostic markers. METHODS: This study, using quantitative RT-PCR, investigated miR-150, miR-455-3p and miR-375 expression, in order to identify a possible molecular distinction between AdCC and PAC. RESULTS: miRNA-150 and miRNA-375 expression was significantly decreased in AdCC and PAC compared with salivary gland tissue controls, whilst miRNA-455-3p showed significantly increased expression in AdCC when compared to PAC, (P < 0.05). miR-150, miR-357 and miR-455-3p expression in AdCC, PAC and control was not associated with age, gender nor with anatomic site (major and minor salivary glands) (P > 0.05). CONCLUSION: MiR-455-3p could be used as a complimentary tool in the diagnosis of challenging AdCC cases.

The present study examined the efficacy of the topical 15d­PGJ2­poloxamer 407 hydrogel in an atopic dermatitis (AD) animal model. The 15d­PGJ2 hydrogel was prepared and characterized. The examined rats possessed AD­Like cutaneous lesions, which were induced using 2,4­dinitrochlorobenzene, the rats were then treated with a hydrogel vehicle, 15d­PGJ2 hydrogel or tacrolimus for 14 days. The rats were sacrificed and blood samples were collected to quantify the IgE levels. Subsequently, skin biopsies were stained with toluidine blue to identify mast cells and immunohistochemistry was performed for ROR­Î³t and TNF­α. Histological analyses demonstrated that 15d­PGJ2 hydrogel significantly decreased mast cell infiltration (P<0.05) when compared with the AD­group. Tacrolimus at 0.1% exhibited decreased mast cell infiltration; however, this difference was not statistically significant from the AD­group. Topical 15d­PGJ2 hydrogel and Tacrolimus 0.1% significantly reduced the serum levels of IgE (P<0.05) compared with the AD­group. Immunohistochemistry revealed a significant decrease in ROR­Î³t and TNF­α positive cell expression (P<0.05) in the 15d­PGJ2 hydrogel group compared with the AD­group. In summary, topical administration of 15d­PGJ2 hydrogel had a beneficial effect on AD symptoms, suggesting that this formulation may be a useful strategy for the treatment of AD.

ABSTRACT Objective: The objective of this review was to evaluate the outcomes of the treatment of peri-implant defects, using Guided Bone Regeneration. Methods: A literature search was performed based on the PICO methodology in the PubMed/Medline, SciELO, Lilacs electronic databases, CAPES periodicals and the Cochrane Library. We included studies using bovine mineral matrix, associated to a collagen membrane for the treatment of peri-implantitis by Guided Bone Regeneration. Results: Of 1,163 studies, 10 were included in this review after applying the evaluation criteria. A total of 269 implants were treated in 260 patients. The follow-up period ranged from 6 to 48 months. The studies evaluated outcome in terms of reduction in probing depth, gain of clinical attachment and healing of the bony defect. Due to the heterogeneity of the studies, it was not possible to perform meta-analysis. Conclusion: Treatment of peri-implant lesions with Guided Bone Regeneration is a viable modality of treatment, providing reduction in bleeding on probing, as well as gain of clinical attachment. Complete filling of the defect is, however, an unpredictable result.


RESUMO Objetivo: O objetivo desta revisão sistemática foi avaliar os desfechos do tratamento dos defeitos peri-implantares, por meio da técnica da Regeneração Óssea Guiada. Métodos: Uma pesquisa bibliográfica, baseada na metodologia PICO, foi realizada nas bases de dados eletrônica PubMed/Medline, SciELO, Lilacs periódicos Capes e Cochrane Library. Foram incluídos estudos que utilizaram matriz mineral bovina, associado a uma membrana de colágeno para o tratamento da peri-implantite por Regeneração Óssea Guiada. Resultados: De 1.163 estudos, 10 foram incluídos nesta revisão, após aplicação dos critérios de avaliação. Um total de 269 implantes foram tratados em 260 pacientes. O período de acompanhamento variou de 6 a 48 meses. Os estudos avaliados reportaram redução média da profundidade de sondagem, ganho de inserção clínica e preenchimento ósseo do defeito. Devido à heterogeneidade dos estudos não foi possível realizar metanálise. Conclusão: O tratamento das lesões peri-implantares, com a técnica da Regeneração Óssea Guiada é uma modalidade viável de tratamento, proporcionando redução do sangramento à sondagem, bem como o ganho de inserção clínica. Porém, o completo preenchimento do defeito, é um resultado imprevisível.

ABSTRACT The aim of this study was to assess marginal bone loss in teeth restored with endodontic posts on all four aspects of the teeth using cone beam tomography. Bone loss was assessed via scores (0-5) in relation to root length and bone height. The results showed that the scores for the contralateral and restored sides, respectively, ranged from 0 to 3 and 0 to 5 buccally, 0 to 4 and 0 to 4 palatally, 0 to 3 and 0 and 4 mesially and 0 to 3 and 0 to 4 distally. A significant difference in bone loss was observed between the post-restored teeth and their contralaterals for the buccal, lingual/palatal and mesial aspects (p<0.05), with teeth bearing endodontic posts scoring highest. In conclusion, marginal bone loss was higher in teeth restored with intra-canal posts when compared to their sound contralaterals.


RESUMO O objetivo deste estudo foi avaliar a perda óssea marginal nas quatro faces de dentes restaurados com pinos endodônticos, usando tomografia por feixe cônico. A perda óssea foi avaliada através de escores (0-5) em relação ao comprimento da raiz e altura do osso. Os resultados mostraram que os escores dos lados contralateral e restaurado, respectivamente, variaram de 0 a 3 e 0 a 5 bucalmente, 0 a 4 e 0 a 4 palatalmente, 0 a 3 e 0 e 4 mesialmente e 0 a 3 e 0 a 4 distalmente. Uma diferença significativa na perda óssea foi observada entre os dentes pós-restaurados e seus contralaterais para os aspectos vestibular, lingual / palatal e mesial (p <0,05), com os dentes com pinos endodônticos pontuando mais alto. Em conclusão, a perda óssea marginal foi maior em dentes restaurados com pinos intra-canal quando comparados aos seus contralaterais sonoros.

DNA aneuploidy is an imbalance of chromosomal DNA content that has been highlighted as a predictor of biological behavior and risk of malignant transformation. To date, DNA aneuploidy in oral potentially malignant diseases (OPMD) has been shown to correlate strongly with severe dysplasia and high-risk lesions that appeared non-dysplastic can be identified by ploidy analysis. Nevertheless, the prognostic value of DNA aneuploidy in predicting malignant transformation of OPMD remains to be validated. The aim of this meta-analysis was to assess the role of DNA aneuploidy in predicting malignant transformation in OPMD. The questions addressed were (i) Is DNA aneuploidy a useful marker to predict malignant transformation in OPMD? (ii) Is DNA diploidy a useful negative marker of malignant transformation in OPMD? These questions were addressed using the PECO method. Five studies assessing aneuploidy as a risk marker of malignant change were pooled into the meta-analysis. Aneuploidy was found to be associated with a 3.12-fold increased risk to progress into cancer (RR=3.12, 95% CI 1.86-5.24). Based on the five studies meta-analyzed, "no malignant progression" was more likely to occur in DNA diploid OPMD by 82% when compared to aneuploidy (RR=0.18, 95% CI 0.08-0.41). In conclusion, aneuploidy is a useful marker of malignant transformation in OPMD, although a diploid result should be interpreted with caution.

Despite the overall benefits of immunization, vaccine hesitancy has been a growing trend and has been associated with the resurgence of vaccine-preventable diseases. The aim of this study was to assess vaccine confidence and hesitancy in Brazil, as part of a wider project to map vaccine confidence globally. One thousand subjects were interviewed, either online or face-to-face, based on a general questionnaire regarding perceptions on vaccines and vaccination. Further exploratory questions were used with the subset of respondents who were parents of children aged under 5. Such questions extracted information regarding vaccination behavior, opinions on vaccination and government health services, and vaccine hesitancy. Reasons for hesitancy were classified as relating to confidence, convenience and/or complacency, and the population was also analyzed socio-demographically. The results showed that overall confidence in immunization was higher than confidence in family planning services, community health workers and emergency services. Seventy-six people reported hesitancy to vaccinate. The commonest reasons for hesitancy were issues with confidence (41.4%), efficacy/safety of the vaccine (25.5%) and concerns about adverse events (23.6%). The sociodemographic analysis revealed that vaccine hesitancy was associated with marital status, level of education and income. Despite overall vaccine confidence being high, a clear trend toward lower levels of confidence was associated with higher levels of hesitancy, which warrants on-going monitoring, due to the dynamic and changing nature of vaccine hesitancy.


Apesar dos benefícios globais da imunização, a hesitação em vacinar é uma tendência crescente que tem sido associada ao ressurgimento das doenças imunopreveníveis. O estudo teve como objetivo avaliar a confiança nas vacinas e a hesitação em vacinar no Brasil, como parte de um projeto mais amplo para mapear a confiança em vacinas em nível global. Foram entrevistadas mil pessoas, direta ou virtualmente, usando um questionário geral sobre percepção em relação às vacinas e à vacinação. Foram utilizadas perguntas exploratórias adicionais no subconjunto de entrevistados que eram pais de crianças abaixo de cinco anos de idade. Essas perguntas produziram informações sobre o comportamento em relação à vacinação, opiniões sobre vacinação e serviços públicos de saúde e hesitação em vacinar. Os motivos da hesitação foram classificados em relação à confiança, conveniência e/ou acomodação, e a população também foi analisada conforme as características sociodemográficas. Os resultados mostraram que a confiança geral na imunização foi maior do que nos serviços de planejamento familiar, agentes comunitários de saúde e serviços de emergência. Setenta e seis pessoas relataram hesitação em vacinar. Os motivos mais frequentes da hesitação diziam respeito a confiança (41,4%), eficácia/segurança da vacina (25,5%) e preocupações com eventos adversos (23,6%). A análise sociodemográfica mostrou que a hesitação em vacinar estava associada ao estado civil, escolaridade e renda. Apesar da alta confiança geral na vacinação, uma clara tendência para níveis de confiança mais baixos esteve associada a níveis mais altos de hesitação, o que justifica o monitoramento permanente dessa tendência, em função da natureza dinâmica da hesitação em vacinar.


A pesar de lo beneficios generales de la inmunización, la renuencia a la vacunación ha sido una tendencia en crecimiento que ha sido asociada con el resurgimiento de las enfermedades prevenibles por vacunación. El objetivo de este estudio fue evaluar la confianza y renuencia a las vacunas en Brasil, como parte de un proyecto más amplio para mapear la confianza en las vacunas globalmente. Mil sujetos fueron entrevistados, bien en línea o cara-a-cara, mediante un cuestionario general respecto a sus percepciones sobre las vacunas y la vacunación. Se utilizaron otras preguntas exploratorias con el subconjunto de encuestados, que eran padres de niños con una edad inferior a los cinco años. Tales preguntas recabaron información respecto al comportamiento sobre la vacunación, opiniones sobre vacunación y servicios de salud gubernamentales, así como su renuencia a las vacunas. Las razones para esta última se clasificaron como aquellas relacionadas con la confianza, conveniencia y/o complacencia, y la población fue también analizada sociodemográficamente. Los resultados mostraron que la confianza general en inmunización fue más alta que la confianza en servicios de planificación familiar, trabajadores de salud comunitarios y servicios de emergencia. Setenta y seis personas informaron de renuencia a la vacunación. Las razones más comunes para la renuencia fueron temas relacionados con la confianza (41,4%), eficacia/seguridad de la vacuna (25,5%) y preocupaciones sobre efectos adversos (23,6%). El análisis sociodemográfico reveló que la renuencia a la vacunación estaba asociada con el estado civil, nivel de educación e ingresos. A pesar de que la confianza general en las vacunas es alta, existe una clara tendencia hacia niveles más bajos de confianza, que estaba asociada con altos niveles de renuencia, lo que garantiza una supervisión permanente, debido a la dinámica y naturaleza cambiante del rechazo a las vacunas.

The aim of the present study was to analyze the in vitro effect of various doses of epidermal growth factor (EGF; 5 and 10 ng/ml) on matrix metalloproteinase-2 (MMP-2) secretion and E-cadherin/ß-catenin expression by co-cultured cells that mimic an in situ carcinoma ex-pleomorphic adenoma, where benign myoepithelial cells from a pleomorphic adenoma surround malignant epithelial cells. EGF was supplemented in various doses and the effects were evaluated following four days of cell culture. ELISA was performed to determine MMP-2 secretion levels. Gene expression for E-cadherin and ß-catenin was analyzed using quantitative polymerase chain reaction. The results revealed that E-cadherin expression decreased when the cells were supplemented with 5 ng/ml EGF. ELISA results indicated that MMP-2 secretion increased when EGF was supplemented at concentrations of 5 and 10 ng/ml. The present findings demonstrated that EGF may be involved in the epithelial-mesenchymal transition process via altering the E-cadherin/ß-catenin complex and increasing MMP-2 secretion, which may then favor the dissolution of the basement membrane to the benefit of malignant cell clusters, contributing to the development of an invasive phenotype in this in vitro model of tumorigenesis.

Multiple myeloma (MM) is characterised by intense protein folding and, consequently endoplasmic reticulum (ER) stress. The prostaglandin 15d-PGJ2 is able to raise oxidative stress levels within the cell and potentially trigger cell death. The aim of this study was to evaluate the antineoplastic effect of 15d-PGJ2 on MM in vitro and in vivo via ER and oxidative stress pathways. MM.1R and MM.1S cell lines were treated with 15d-PGJ2 at 1-10µM and evaluated with regard to proliferation, mRNA expression of PRDX1, PRDX4, GRP78, GRP94, CHOP, BCL-2 and BAX. Stress data was validated via oxidized glutathione assays. MM.1R cells were inoculated into NOD/SCID mice, which were subsequently treated daily with 15d-PGJ2 at 4mg/kg or vehicle (control), with tumour volume being monitored for 14days. 15d-PGJ2 reduced cell proliferation, induced cell death and apoptosis at 5µM and 10µM and Stress-related genes were upregulated at the same doses. Oxidized glutathione levels were also increased. 15d-PGJ2 at 4mg/kg in vivo halted tumour growth. In conclusion, 15d-PGJ2 induced myeloma cell death via ER stress in vitro. 15d-PGJ2 in vivo also inhibited tumour growth.

RESUMEN

Gingival cyst of the adult is a rare slow growing and asymptomatic lesion that arises from the rests of the dental lamina. The present report describes the case of a miniscule adult gingival cyst in the lower anterior gingiva in a 51-year-old male with agenesis of lower premolars and lateral incisors. This paper contrasts the literature concerning the differentiation between the gingival cyst of the adult and the lateral periodontal cysts as well as the possible misguided concept that the former may be such rare an occurrence.

There is mounting evidence on the importance of some biological processes in tumor growth, such as vascular supply, apoptosis, autophagy, and senescence. We have investigated these processes in polymorphous low-grade adenocarcinoma (PLGA), in an attempt to identify those that are relevant for this particular lesion. We analyzed 31 cases of PLGA using immunohistochemistry to antibodies against CD34 and CD105 to detect blood vessels; against D2-40 to detect lymphatic vessels; against Bax, Bcl-2, and survivin to explore cell apoptosis; and against Beclin and LCB3 to investigate autophagy and against p21 and p16 to assess senescence. Our results showed that PLGA growth does not depend on newly formed vessels but only on preexisting vasculature. Furthermore, PLGA is promoted by autophagy, sustained by both anti-apoptotic and anti-senescence signals, and stimulated by Bcl-2 and survivin.
