RESUMEN

In order to load fish oil for potential encapsulation of fat-soluble functional active substances, fish oil-loaded multicore submillimeter-sized capsules were prepared with a combination method of three strategies (monoaxial electrospraying, chitosan-tripolyphosphate ionotropic gelation, and Tween blending). The chitosan-tripolyphosphate/Tween (20, 40, 60, and 80) capsules had smaller and evener fish oil cores than the chitosan-tripolyphosphate capsules, which resulted from that Tween addition induced smaller and evener fish oil droplets in the emulsions. Tween addition decreased the water contents from 56.6 % to 35.0 %-43.4 %, increased the loading capacities from 10.4 % to 12.7 %-17.2 %, and increased encapsulation efficiencies from 97.4 % to 97.8 %-99.1 %. In addition, Tween addition also decreased the highest peroxide values from 417 meq/kg oil to 173-262 meq/kg oil. These properties' changes might result from the structural differences between the chitosan-tripolyphosphate and chitosan-tripolyphosphate/Tween capsules. All the results suggested that the obtained chitosan-tripolyphosphate/Tween capsules are promising carriers for fish oil encapsulation. This work also provided useful knowledge to understand the preparation, structural, and physicochemical properties of the chitosan-tripolyphosphate capsules.

Using gas chromatography-tandem mass spectrometry and electrospun nanofibrous membrane, we developed and validated a simple, rapid, and sensitive methodology for quantifying eugenol residues in fish tissue and water samples. Fish tissue extract and water samples (315 samples) collected from three southeastern China provinces (Shanghai, Zhejiang, and Fujian), originating from eight provinces of Zhejiang, Jiangsu, Shandong, Guangdong, Fujian, Anhui, Shanghai, and Jiangxi, from April 2021 to April 2023 were filtered with an electrospun nanofiber membrane, extracted with trichloromethane/n-hexane, and directly concentrated to dry after simple purification. An internal standard of p-terphenyl in n-hexane and 5-µL injection volumes of the solutions was used to analyze eugenol via internal calibration with a minimum concentration of 0.5 µg/L in water samples and 0.1 µg/kg in aquatic product samples. The highest amount of eugenol was detected in Fujian province, possibly due to the higher temperature during transportation, while the lowest amount was found in Shanghai, which mainly uses temporary fish-culture devices. This is a fast, inexpensive, and effective method for testing large quantities of fish water and meat samples.

Until now, the molecular mechanisms underlining sperm motility defect causing male infertility are still poorly understood. Safe and effective compounds or drugs that can improve sperm motility are also very limited. Lysophosphatidic acid (LPA) is a naturally occurring phospholipid and a bioactive intermediate with multiple biological activities. It has been detected in various body fluids such as serum, plasma, saliva, tears, blister fluids, hen egg white, and ascites from patients with ovarian cancer. LPA is also abundant in seminal plasma and follicular fluid. It enhances follicle stimulation, improves oocyte fertilization, and promotes early embryonic development and embryo implantation. However, the physiological role of LPA in the male reproductive system remains unknown. Here, our study showed that LPA significantly improved the motility parameters of human sperm hyperactivation in a dose-dependent manner. The LPA-induced elevation of sperm motility is dependent on bovine serum albumin (BSA) but independent of the classical BSA-induced sAC/cAMP/PKA signaling pathway. The enhancement of sperm motility by LPA could not be blocked by CCCP, a respiratory inhibitor suppressing mitochondrial ATP production. Moreover, LPA improved the activity of triosephosphate isomerase in glycolysis. Meanwhile, LPA treatment significantly increased ATP and phosphoenolpyruvate levels and decreased ADP content during sperm glycolysis. Notably, none of known or identified LPA receptors was detected in human sperm. Further investigations showed that LPA promoted sperm motility through L-type calcium channels. In summary, this study revealed the involvement of LPA in the regulation for human sperm motility by enhancing glycolysis and activating L-type calcium channels. The current findings may shed new light on the understanding of causes of asthenozoospermia, and indicate that LPA could be used as a novel therapeutic agent to improve sperm function and fertilizing capacity.

Although the potential corrosive microbial communities of the refined oil pipelines can cause pipeline failure which directly threatens on soil and water environment, few studies have been published in this field. Therefore, the long-distance on-site internal corrosion detection and high-throughput sequencing techniques were employed in this study to investigate the distribution shifts of the corrosive microbial communities on the inner wall of a refined oil pipeline and its impact on the internal corrosion. The microorganisms colonizing on the inner wall of the pipeline showed significant distribution differences between the axial direction of the relative elevation and radial direction of the cross-section. On the inner wall, the high diversity and the abundance of the corrosive microbial communities induced serious microbiologically influenced corrosion (MIC), while the chemical corrosion and the synergy of the corrosive microbial communities accelerated the internal corrosion of the refined oil pipeline. A corrosion zone model has been proposed, which divides the pipeline cross-section into the sediment, the water-oil interface, the gas-oil interface, and the oil fully immersed zones. Therefore, the relationships between the environment, corrosion degree, and distribution characteristics of the corrosive microbial communities in the pipeline were analyzed. This research exhibited the importance of the distribution characteristics of the corrosive microorganisms on the inner wall of the refined oil pipelines. Its internal corrosion behavior was accurately explored, while providing a basis for controlling the corrosive microbial communities.

Seminal plasma contains a high concentration of extracellular vesicles (EVs). The heterogeneity of small EVs or the presence of nonvesicular extracellular matter (NV) pose major obstacles in understanding the composition and function of seminal EVs. In this study, we employed high-resolution density gradient fractionation to accurately characterize the composition and function of seminal EVs and NV. We found that the seminal EVs could be divided into 3 different subtypes-namely, high-density EV (EV-H), medium-density EV (EV-M), and low-density EV (EV-L)-after purification using iodixanol, while NV was successfully isolated. EVs and NV display different features in size, shape, and expression of some classic exosome markers. Both EV-H and NV could markedly promote sperm motility and capacitation compared with EV-M and EV-L, whereas only the NV fraction induced sperm acrosome reaction. Proteomic analysis results showed that EV-H, EV-M, EV-L, and NV had different protein components and were involved in different physiological functions. Further study showed that EV-M might reduce the production of sperm intrinsic reactive oxygen species through glutathione S-transferase mu 2. This study provides novel insights into important aspects of seminal EVs constituents and sounder footing to explore their functional properties in male fertility.

BACKGROUND: Although the latest Gleason grading system in 2014 has distinguished between Gleason 3 + 4 and 4 + 3, Gleason 8 and Gleason 9-10 are remained systemically classified. METHODS: A total of 261,125 patients diagnosed with prostate cancer (PCa) were selected between 2005 and 2015 from the Surveillance, Epidemiology, and End Results (SEER) database. We used propensity score matching to balance clinical variables and then compared overall survival (OS) and cancer-specific survival (CSS) between Gleason score subgroups. We further establish a new Gleason survival grading system based on the hazard ratio (HR) values of each Gleason subgroup. Cox proportional hazards models and Kaplan-Meier curves were used to compare patient survival. RESULTS: Among PCa patients with Gleason score 8 disease, patients with Gleason 5 + 3 had significantly worse OS and CSS than those with Gleason 3 + 5 (OS: HR = 1.26, p = 0.042; CSS: HR = 1.42, p = 0.005) and 4 + 4 (HR = 1.50 for OS and HR = 1.69 for CSS, p < 0.001 for all). PCa patients with Gleason 5 + 3 and Gleason 4 + 5 may have the similar OS and CSS (reference Gleason score <=6, 5 + 3: OS HR = 2.44, CSS HR = 7.63; 4 + 5: OS HR = 2.40, CSS HR = 8.92; p < 0.001 for all). The new Gleason survival grading system reclassified the grades 4 and 5 of the 2014 updated Gleason grading system into three hierarchical grades, which makes the classification of grades more detailed and accurate. CONCLUSION: PCa patients with Gleason 8-10 may have three different survival subgroups, Gleason 3 + 5 and 4 + 4, Gleason 5 + 3 and 4 + 5, and Gleason 5 + 4 and 5 + 5. Our results maximize risk stratification for PCa patients, provide guidance for clinicians to assess their survival and clinical management, and make a recommendation for the next Gleason grading system update.

To investigate the role of previous cancer on overall survival in patients with bladder cancer (BCa) and to establish an effective prognostic tool for individualized overall survival prediction.A total of 78,660 patients diagnosed with BCa between 2000 and 2013 were selected from the Surveillance, Epidemiology, and End Results (SEER) database, among which 8915 patients had a history of other cancers. We compared the overall survival between patients with and without previous cancer after propensity score matching and we further established a nomogram for overall survival prediction.Univariate and multivariate Cox analyses were used to determine independent prognostic factors. The calibration curve and concordance index (C-index) were used to assess the accuracy of the nomogram. Cox proportional hazards models and Kaplan-Meier analysis were used to compare survival outcomes.BCa patients with previous cancer had worse overall survival compared with those without previous cancer (HR = 1.37; 95%CI = 1.32-1.42, P < .001). Cancers in lung prior to BCa had the most adverse impact on overall survival (HR = 2.35; 95%CI = 2.10-2.63; P < .001), and the minimal impact was located in prostate (HR = 1.16; 95%CI = 1.10-1.22; P < .001) for male and in gynecological (HR = 1.15; 95%CI = 1.02-1.30; P = .027) for female. The shorter interval time between 2 cancers and the higher stage of the previous cancer development, the higher risk of death. Age, race, sex, marital status, surgery, radiation, grade, stage, type of previous cancer as the independent prognostic factors were selected into the nomogram. The favorable calibration curve and C-index value (0.784, 95%CI = 0.782-0.786) indicated the nomogram could accurately predict the 1-, 3-, and 5-year overall survival rate of BCa patients.Previous cancer has a negative impact on the overall survival of BCa patients and requires more effective clinical management. The nomogram provides accurate survival prediction for BCa patients and might be helpful for clinical treatment selection and follow-up strategy adjustment.

The longhorned beetles, Rhaphuma horsfieldi and Xylotrechus quadripes, are two polyphagous insects with larvae feeding on different host plants. In this study, we identified and characterized three gene superfamilies of cytochrome P450s (CYPs), carboxylesterases (COEs) and glutathione-S-transferases (GSTs) involved in the detoxification of endobiotics (e.g., hormones and steroids) and xenobiotics (e.g., insecticides, sex pheromones and plant allelochemicals) through a combination approach of bioinformatics, phylogenetics, expression profiles and genomics. Transcriptome analyses led to the identification of 281 transcripts encoding 135 P450s, 108 COEs and 38 GSTs from the two beetles, coupled with comparative studies of detoxification genes among coleopteran species, suggesting a correlation between host range and the sizes of P450 or COE gene repertoires. The P450s of two beetles were phylogenetically classified into four clades, representing the majority of genes in the CYP3 clan. The COEs from R. horsfieldi and X. quadripes were separately grouped into 11 and 10 clades, and the GST superfamily was assigned into six clades. Expression profiles revealed that the detoxification genes were broadly expressed in various tissues as an implication of functional diversities. Ultimately and more importantly, five alternative splicing events in the Epsilon GSTs, including RhorGSTe7.1/GSTe7.2 and XquaGSTe3.1/GST3.2, were acquired in Coleoptera, in which these genes and their orthologs shared highly conserved gene structure. Our current study has complemented the resources for the detoxification genes in the family Cerambycidae, and allows for functional experiments to identify candidate molecular targets involved in pest resistance to insecticides like organophosphates, organochlorines and pyrethroids.

BACKGROUND: Currently, the prognosis of kidney cancer depends mainly on the pathological grade or tumor stage. Clinicians have few effective tools that can personalize and adequately evaluate the prognosis of kidney cancer patients. METHODS: A total of 70 481 kidney cancer patients were selected from the Surveillance, Epidemiology, and End Results database, among which patients diagnosed in 2005-2011 (n = 42 890) were used to establish nomograms for overall survival (OS) and cancer-specific survival (CSS), and those diagnosed in 2012-2015 (n = 24 591) were used for external validation. Univariate and multivariate Cox analyses were used to determine independent prognostic factors. Concordance index (C-index), receiver operating characteristic curve, and calibration curve were used to evaluate the predictive capacity of the nomograms. We further reduced subgroup classification and used propensity score matching to balance clinical informations, and analyzed the effect of other variables on survival. We established a new kidney cancer prognostic score system based on the effect of all available variables on survival. Cox proportional hazard model and Kaplan-Meier curves were used for survival comparison. RESULTS: Age, gender, marital status, surgery, grade, T stage, and M stage were included as independent risk factors in the nomograms. The favorable area under the curve (AUC) value (for OS, AUC = 0.812-0.858; and for CSS, AUC = 0.890-0.921), internal (for OS, C-index = 0.776; and for CSS, C-index = 0.856), and external (for OS, C-index = 0.814-0.841; and for CSS, C-index = 0.894-0.904) validation indicated that the proposed nomograms could accurately predict 1-, 3-, and 5-year OS and CSS of kidney cancer patients. The Aggtrmmns prognostic scoring system based on age, gender, race, marital status, grade, TNM stage, and surgery of kidney cancer patients could stage patients more explicitly than the AJCC staging system. CONCLUSION: The nomogram and Aggtrmmns scoring system can predict OS and CSS in kidney cancer patients effectively, which may help clinicians personalize prognostic assessments and clinical decisions.

Takifugu obscurus is famous for its delicious taste. In this study, three peptide fractions from Takifugu obscurus muscle hydrolysates were used to evaluate and compare physicochemical and sensory characteristics after Maillard reaction. Maillard reaction products (MRPs) were analyzed by detecting the changes of browning intensity, free amino acids, volatile compounds and taste profile. Results showed that intermediate flavor products in three fractions of MRPs from different molecular weight peptides were much stronger than in control groups. The free amino acids of MRPs from 0.3 to 3.0 kDa and whole fraction showed a similar trend. The contents of volatile compounds (furans, phenols, pyrazines and so on) also increased in the three MRP fractions. The taste characteristics of three fractions showed stronger kokumi effect and umami taste compared to their control groups. To sum up, this investigation lays the groundwork for the development of a flavoring agent from Takifugu obscurus.

Fermentation of soybean is a process in which soy proteins are broken down into small peptides to exert various physiological functions beyond their nutritional value and to improve food source bioactive components responsible for health benefits. Enzymatic hydrolysis could speed up the degradation of proteins during fermentation of soybean, thus resulting in higher peptide production. In the present study, fermented soy meal (fermented with Bacillus subtilis from Douchi) was hydrolyzed by thermolysin, and the water extraction was then separated into four fractions using ultrafiltration membranes. Their vasorelaxation activities were screened, and the most potent fraction was further isolated and purified to obtain four peptides. Briefly, three peptides exerted a dose-dependent vasorelaxation (0.01-4.10 µM) in the phenylephrine preconstricted thoracic aorta ring of Sprague-Dawley rat (relaxation actions were all endothelium-independent), while one peptide induced vasoconstriction. Furthermore, an independent causal relationship between vasorelaxation and angiotensin converting enzyme (ACE) inhibition activities was found.

In this study, we challenged to identify vasoactive peptides in soybean 11S glycinin hydrolysate by thermolysin to regulate intracellular Ca(2+) concentration ([Ca(2+)]i) that can induce constrictive vascular tension. As a function of the inhibition of elevated [Ca(2+)]i by 10 µM angiotensin (Ang) II in vascular smooth muscle cells (VSMCs), eleven peptides were successfully identified from the hydrolysate, among which His-Gly-Lys exhibited the most potent inhibition against [Ca(2+)]i elevation in Ang II-stimulated VSMCs (inhibition at 300 µM: 46.5±8.0% vs. control). The biological capacity of His-Gly-Lys analogues as an [Ca(2+)]i inhibitor was also proven when His-Lys and His-Gly-Arg elicited a significant reduction in [Ca(2+)]i. In contrast, less reduction of [Ca(2+)]i by His-Gly-Ile and His-(3-methyl)-Gly-Lys indicated the importance of the imino proton in His, along with basic amino acids positioned at C-terminal for the effect.

A modified QuEChERS-GC-MS method for analysis of procymidone residue in leeks and soil was developed and validated. Procymidone residue dynamics and residues in supervised field trials at GAP conditions in leeks and soil in greenhouses were studied. Leek samples were treated under microwave radiation for 1min before homogenization, followed by extracting with acetonitrile and clean-up with reverse solid phase dispersion by primary and secondary amine (PSA). Procymidone residue was determined by GC-MS in selected ion monitoring (SIM) mode. At fortification levels of 0.02, 0.2 and 2mg/kg in leeks and soil, it was shown that recoveries ranged from 74.9% to 100.8% with relative standard deviations of 1.3-8.5%. At four geographical experimental plots, procymidone residue in leeks and soil showed a relatively fast dissipation rate, with half-lives of 4.52-8.76 days for leeks and 3.76-5.65 days for soil. At pre-harvest intervals of 21-30 days, procymidone residue ranged from 0.033 to 0.17mg/kg in leeks, and 0.020-1.75mg/kg in soil. Residues persistence varied in leeks and soil in four field trials, suggesting that it might be affected by some physical and chemical factors, growth dilution factor, soil characteristics and microorganisms.
