Drosophila melanogaster is a widely used animal model in human diseases and to date it has not been applied to the study of the impact of tobacco use on human sexual function. Hence, this report examines the effects of different concentrations of cigarette smoke extract (CSE) exposure on the size and sexual behavior of D. melanogaster. Wild-type flies were held in vials containing CSE-infused culture media at concentrations of 10%, 25%, and 50% for three days, and their offspring were reared under the same conditions before measuring their body size and mating behavior. CSE exposure during development reduced the tibia length and body mass of emerging adult flies and prolonged the time required for successful courtship copulation success, while courtship behaviors (wing extension, tapping, abdomen bending, attempted copulation) remained largely unchanged. Our findings indicate that CSE exposure negatively affects the development of flies and their subsequent reproductive success. Future experiments should investigate the CSE effect on male female fertility.

Anastrepha ludens is a polyphagous frugivorous tephritid that infests citrus and mango. Here, we report the establishment of a laboratory colony of A. ludens reared on a larval medium that is a waste for the citrus industry, specifically, orange (Citrus × sinensis) fruit bagasse. After 24 generations of rearing on a nutritionally poor orange bagasse diet, pupae weighed 41.1% less than pupae from a colony reared on a nutritionally rich artificial diet. Larvae from the orange bagasse diet had 6.94% less protein content than larvae from the artificial diet, although their pupation rate was similar. Males from the orange bagasse diet produced a scent bouquet with 21 chemical compounds and were sexually competitive, but they had significantly shorter copulations when compared to males from the artificial diet and from the wild host, Casimiroa edulis, which had relatively simple scent bouquets. The chemical complexity in the odors of males from the orange bagasse diet might initially have attracted females to novel scent combinations, but, once in the copula, they may have been able to sense negative characteristics in males, leading them to terminate copulations soon after they began. We conclude that A. ludens can adjust morphological, life history, nutritional, and chemical traits when adapted to a larval environment consisting of fruit bagasse.

A trade-off hypothesis pertains to the biased allocation of limited resources between two of the most important fitness traits, reproduction and survival to infection. This quid pro quo manifests itself within animals prioritizing their energetic needs according to genetic circuits balancing metabolism, germline activity and immune response. Key evidence supporting this hypothesis includes dipteran fecundity being compromised by systemic immunity, and female systemic immunity being compromised by mating. Here, we reveal a local trade-off taking place in the female Drosophila midgut upon immune challenge. Genetic manipulation of intestinal motility, permeability, regeneration and three key midgut immune pathways provides evidence of an antagonism between specific aspects of intestinal defense and fecundity. That is, juvenile hormone (JH)-controlled egg laying, lipid droplet utilization and insulin receptor expression are specifically compromised by the immune deficiency (Imd) and the dual oxidase (Duox) signaling in the midgut epithelium. Moreover, antimicrobial peptide (AMP) expression under the control of the Imd pathway is inhibited upon mating and JH signaling in the midgut. Local JH signaling is further implicated in midgut dysplasia, inducing stem cell-like clusters and gut permeability. Thus, midgut JH signaling compromises host defense to infection by reducing Imd-controlled AMP expression and by inducing dysplasia, while midgut signaling through the Imd and Duox pathways compromises JH-guided metabolism and fecundity.

Concentrative nucleoside transporters (Cnts) are unidirectional carriers that mediate the energy-costly influx of nucleosides driven by the transmembrane sodium gradient. Cnts are transmembrane proteins that share a common structural organization and are found in all phyla. Although there have been studies on Cnts from a biochemical perspective, no deep research has examined their role at the organismal level. Here, we investigated the role of the Drosophila melanogaster cnt1 gene, which is specifically expressed in the testes. We used the CRISPR/Cas9 system to generate a mutation in the cnt1 gene. The cnt1 mutants exhibited defects in the duration of copulation and spermatid maturation, which significantly impaired male fertility. The most striking effect of the cnt1 mutation in spermatid maturation was an abnormal structure of the sperm tail, in which the formation of major and minor mitochondrial derivatives was disrupted. Our results demonstrate the importance of cnt1 in male fertility and suggest that the observed defects in mating behavior and spermatogenesis are due to alterations in nucleoside transport and associated metabolic pathways.

Prolactin, a hormone involved in vertebrate parental care, is hypothesized to inhibit reproductive hypothalamic-pituitary-gonadal (HPG) axis activity during parenting, thus maintaining investment in the current brood as opposed to new reproductive efforts. While prolactin underlies many parental behaviors in birds, its effects on other reproductive behaviors, such as courtship, remain unstudied. How prolactin affects neuropeptide and hormone receptor expression across the avian HPG axis also remains unknown. To address these questions, we administered ovine prolactin (oPRL) or a vehicle control to both sexes in experienced pairs of the biparental rock dove (Columba livia), after nest removal at the end of incubation. We found that oPRL promoted parental responses to novel chicks and stimulated crop growth compared to controls, consistent with other studies. However, we found that neither courtship behaviors, copulation rates nor pair maintenance differed with oPRL treatment. Across the HPG, we found oPRL had little effect on gene expression in hypothalamic nuclei, but increased expression of FSHB and hypothalamic hormone receptor genes in the pituitary. In the gonads, oPRL increased testes size and gonadotropin receptor expression, but did not affect ovarian state or small white follicle gene expression. However, the oviducts of oPRL-treated females were smaller and had lower estrogen receptor expression compared with controls. Our results highlight that some species, especially those that show multiple brooding, may continue to express mating behavior despite elevated prolactin. Thus, mechanisms may exist for prolactin to promote investment in parental care without concurrent inhibition of reproductive function or HPG axis activity.

Insects are exposed to cadmium stress since cadmium pollution has increasingly become a serious global environmental issue. However, until now few studies have paid attention to the effect of heavy metals on insect reproductive behaviors. In our study, the courtship behaviors, mating behaviors and fecundity of beet armyworm Spodoptera exigua (Lepidoptera: Noctuidae) exposed to different concentrations of cadmium in artificial diets at larval stage were studied. The results showed that cadmium stress changed the courtship rhythm by significantly advancing or delaying the courtship starting time. Low dose of cadmium (0.2 mg/kg) increased the courtship frequency in the first two scotophases, but in the fourth phase, the two cadmium treatments reduced the frequency. The total courtship duration was significantly shortened in the first six scotophases except high dose of cadmium treatment (51.2 mg/kg) in the sixth dark phase. Paired adults did not mate after the seventh scotophase under low cadmium exposure, while high cadmium stress made the paired adults just copulate in the first four scotophases. The daily mating rate and total mating rate decreased with the increase in cadmium concentration. The number of eggs of low cadmium treatment was higher than that of control, but the difference was not significant; the number of eggs in high cadmium treatment was lower than that of control and low cadmium treatment. Our results indicate that cadmium exposure can disrupt the courtship rhythm for females and has negative influences on copulation behavior and high cadmium stress can reduce fecundity. Hence, the insect population increase will be affected by heavy metal pollution. Our study will provide scientific reference for environmental risk assessment of heavy metal pollution.

Evidence suggests an important role of Pavlovian learning in sexual partner selection. Female rats that experience paced copulation with a male scented with a neutral odor selectively solicit and receive ejaculations from the scented male relative to an unscented male. Exposure to the conditioned odor alone induces Fos protein in regions of the brain associated with sexual excitation. Here we tested whether female rats can be conditioned to show a sexual preference for an unscented male rat of the same strain. Female Long-Evans rats were given 10 copulatory trials with either a one-hole pacing divider or a four-hole pacing divider in a unilevel chamber with the same conspecific male (n = 16). Females were then given an open-field partner preference test with the paired male versus a novel male. After two reconditioning trials females were exposed to the partner or a novel male to induce Fos expression. Females that paced with the one-hole divider received the first ejaculation and more ejaculations overall from the paired compared to novel male. Fos immunoreactivity within oxytocin neurons in the PVN, mPOA, and VMH was increased in females with a preference that were exposed to the paired male. These data indicate that female rats can form selective sexual preferences for an individual conspecific and that their formation depends on the type of pacing during conditioning. These findings further suggest the involvement of oxytocin in the display of conditioned preferences. Thus, early copulatory experience appears to determine the mating strategy used by female rats.

BACKGROUND Nyssorhynchus deaneorum is a potential malaria vector because it has been shown to be competent to transmit Plasmodium vivax and Plasmodium falciparum, and because it exhibits antropophilic and endophilic behaviors in some regions of the Amazon. This profile makes Ny. deaneorum a useful mosquito for experiments that model Plasmodium-vector interactions in the Amazon. OBJECTIVE Herein we describe how a free-mating colony of Ny. deaneorum has been established using an automated light stimulation system. METHODS Mosquitoes were captured in São Francisco do Guaporé, Rondônia. The F1 generation was reared until adult emergence at which point copulation was induced using an automatic copulation induction system (ACIS). FINDINGS After four generations, natural mating and oviposition began to occur without light stimulation. The number of pupae and adult mosquitoes increased from the F5 to F10 generations. The new Ny. deaneorum colony exhibited susceptibility to P. vivax. MAIN CONCLUSIONS Automated light stimulation is an effective method for establishing an Ny. deaneorum colony under laboratory conditions as it produces enough adults to create a stenogamic colony. The establishment of a stable, P. vivax-susceptible colony of Ny. deaneorum makes it possible to model parasite-vector interactions and to test novel drug therapies that target parasite development in mosquitoes.

The medial amygdala (MeA), bed nucleus of the stria terminalis (BNST), and medial preoptic area (mPOA) are important for the regulation of male sexual behavior. Sexual experience facilitates sexual behaviors and influences activity in these regions. The goal of this study was to determine whether sexual experience or copulation induces plasticity in the MeA, BNST, or mPOA of male rats, as indicated by changes in levels of Arc, which is indicative of activity-dependent synaptic plasticity in the brain. To this end, sexually naïve or experienced males were placed in mating arenas either alone, with an inaccessible estrus female, or with an accessible estrus female. Arc protein levels were then quantified in these three regions using immunohistochemistry. As expected, sexual experience facilitated copulation, as evidenced by a reduction in latencies to mount, intromit, and ejaculate. Copulation also increased the number of Arc-positive cells in the MeA, anterior BNST, posterior BNST, and the posterior mPOA, but not in the central-rostral region of the mPOA. Surprisingly, prior sexual experience did not impact levels of Arc, suggesting that copulation-induced Arc occurs in both sexually naïve and experienced males.

The present study evaluates the role of social factors in the transition from infanticidal to paternal male behavior and its association with T concentration, presence of estrogen receptor alpha (ERα) and androgen receptor (AR) in the olfactory bulb (OB), medial preoptic area (mPOA) and medial amygdala (MeA) of Mongolian gerbils (Meriones unguiculatus). This study included thirty-six sexually inexperienced males displaying aggressive behavior toward foreign pups. The selected animals were mated and organized into four groups. The paternal behavior tests were performed on the day of copulation (DCOPUL), during cohabitation with a pregnant female (CPREG), on the day of birth (DBIRTH), and on day 6 postpartum (DPP6). Eight sexually inexperienced males (CTL (male-male cohabitation) were used as control. After paternal behavior tests, blood samples were obtained to quantify T by radioimmunoassay; the brains were removed and analyzed for immunoreactivity (ir) of ERα and AR. All males of the DCOPUL, DBIRTH, and DPP6 groups exhibited paternal behavior, whereas the males of CPREG and CTL groups were aggressive with the pups. Paternal behavior was associated with high T concentrations, and the presence of ERα-ir and AR-ir in the OB, MeA, and mPOA. These results suggest that the transition from aggressive to paternal response to pups is facilitated by copulation, and that in this transition is involved an increase in T concentration. Moreover, the presence of ERα-ir and AR-ir in the OB, mPOA, and MeA could indicate that estrogenic and androgenic pathways participate in the regulation of paternal behavior of the Mongolian gerbils.

AIMS: We hypothesized that copulation-induced temporary anti-nociception in female rats is mediated by the activation of central and/or peripheral oxytocin receptors. To test this hypothesis, we assessed the effects of intraperitoneal (ip), intrathecal (it), and intra-cerebroventricular (icv) administration of an oxytocin receptor antagonist (atosiban), on copulation-induced temporary anti-nociception in estrous rats. MAIN METHODS: The treatment groups were ovariectomized rats pre-treated subcutaneously (sc) with 10 µg of estradiol benzoate (EB) followed 24 h later by an sc injection of 5 µg EB, and 4 h later, by an sc injection of 2 mg progesterone (P4). Rats were then administered saline vehicle (ip, it, or icv: control groups) or atosiban (500 µg/kg ip; 500 ng it; or 500 ng icv: experimental groups). Thirty minutes after drug or saline administration, their sexual behavior was tested by pairing with a sexually-experienced male rat. Brief pulse trains of 50 Hz, 300 ms duration, supra-threshold tail electrical shocks (STS) were delivered before and during copulatory activity i.e., while the female was receiving mounts, intromissions, or ejaculations, and we recorded whether vocalization occurred in response to each STS. KEY FINDINGS: Replicating our previous findings, the vocalization response to STS in control rats was significantly attenuated during intromissions and ejaculations, compared to their baseline (pre-mating) response, indicative of anti-nociception. By contrast, rats pre-treated with atosiban (each route of administration) failed to show an attenuation of the vocalization response to shock. SIGNIFICANCE: These findings provide evidence that the temporary anti-nociceptive effect of copulation in female rats is mediated by copulation-induced release of endogenous oxytocin in brain, spinal cord and periphery.

PURPOSE: We determined the effect of chronic bilateral neurectomy of the dorsal nerve of the clitoris on urinary parameters and sexual behavior of conscious female rats. MATERIALS AND METHODS: A total of 18 anesthetized virgin female Wistar rats were used in this study, including 11 that underwent bilateral neurectomy of the dorsal nerve of the clitoris and 7 that underwent sham surgery. Urinary parameters were determined in awake animals preoperatively, and 3 and 10 days postoperatively. Sexual behavior was tested 14 days postoperatively to determine whether the females expelled urine during sexual encounters. After male ejaculation the females were anesthetized with urethane to record external urethral sphincter electromyogram activity in response to clitoris, perigenital skin and vaginal stimulation. Neurectomy was corroborated anatomically. RESULTS: Sham surgery did not significantly modify urinary parameter values. However, bilateral neurectomy of the dorsal nerve of the clitoris significantly increased voiding frequency and voiding duration (p <0.05). It did not significantly affect the flow rate, voided volume or voiding interval. Of females that underwent bilateral neurectomy of the dorsal nerve of the clitoris 67% expelled urine just after male ejaculation. CONCLUSIONS: These results suggest that the pudendal nerve is an important neural pathway in the convergence and crosstalk of female urogenital neural circuits, and genital deafferentation may be a causal factor of coital urinary incontinence. Rats with bilateral transection of the dorsal nerve of the clitoris may serve as an animal model of coital incontinence.

Mating events and aggregations of vulnerable nurse sharks Ginglymostoma cirratum were recorded in the insular protected area of Fernando de Noronha (FEN), Brazil, between April and August 2015. Female sharks were observed clustering in groups of up to 14 individuals in shallow water adjacent to the shore. Several evasive mating behaviours in the presence of males were noticed, including shoreward movement, positioning ventral side up at the sea surface with emerged pectoral and pelvic fins, body rolling and caudal fin thrashing. Fresh bite marks indicative of male courtship and coupling attempts were visible in female's pectoral and caudal fins. Altogether, the observed behaviours match previous reports of non-cooperative female nurse sharks during mating opportunities. An extended mating season coupled with a persistent use of inshore habitats result in nurse sharks being particularly vulnerable to human pressure during a most sensitive stage of their life cycle. The effective conservation of nurse shark populations from the western South Atlantic may thus depend on the protection of critical habitats where this species aggregates to reproduce. Explicitly addressing environmental requirements by vulnerable species in local management strategies is indispensable to ensure that human pressure, including ecotourism development, does not collide with stipulated conservation aims.(AU)


Agregações e eventos reprodutivos do vulnerável tubarão-lixa, Ginglymosytoma cirratum , foram registrados na área insular protegida do Arquipélago de Fernando de Noronha (FEN), Brasil, entre Abril e Agosto de 2015. Grupos constituídos por até 14 fêmeas foram observados em águas rasas adjacentes à linha costeira. Perante a presença de machos, diversos comportamentos evasivos por parte das fêmeas foram identificados, incluíndo movimentação para águas ainda mais rasas, posicionamento na superfície do mar com a face ventral orientada para cima e emersão das nadadeiras peitorais e pélvicas, enrolamento do corpo e batimento da nadadeira caudal na superfície do mar. Foram identificadas marcas recentes de mordida nas nadadeiras peitorais e caudais das fêmeas, sugerindo interações de acasalamento com machos. No geral, os comportamentos observados coincidem com os comportamentos exibidos por fêmeas não aquiescentes à cópula reportados no hemisfério norte. Em FEN, a duração relativamente longa do período de acasalamento, juntamente com a utilização persistente dos habitats mais costeiros, promovem a vulnerabilidade do tubarão lixa às pressões antrópicas durante uma das fases mais sensíveis do seu ciclo de vida. A conservação efetiva das populações de tubarão-lixa do Oceano Atlântico Sul ocidental poderá, assim, depender da proteção dos habitats críticos de agregação onde esta espécie se reproduz. Por conseguinte, a abordagem explícita dos requisitos ambientais associados a espécies vulneráveis no âmbito dos planos de manejo locais deve ser considerada como indispensável para assegurar que as pressões antrópicas, incluíndo o desenvolvimento ecoturístico, não comprometerão os objetivos conservacionistas estipulados.(AU)

A species' mating system depends on its spatial distribution and temporal availability of mating opportunities, as well as on the resources that create these opportunities. In addition, for many species, courtship is driven by specific behaviors that precede and follow copulation. Although Sphex ingens is a taxonomically well known species of digger wasp, its ecology and behavior remain poorly known. Hence, we analyzed patterns and trends of sexual behavior, in order to understand whether courtship can persist in a polygamous mating system. We monitored by video wasp populations in Ilha Grande, southeastern Brazil. Based on the observed behaviors, we calculated stochastic probabilities with a Markov chain to infer on behavioral trends. We recorded four behavioral phases based on 19,196 behavioral acts observed in 224 copulation attempts. There were no significant differences in common behavioral acts between males and females. The copulation patterns, conflicts, and trends observed in S. ingens clearly show the influence of sexual selection in its promiscuous mating system.

Nitric oxide (NO) acts in the medial preoptic area (mPOA) of the hypothalamus to facilitate the expression of male sexual behavior and has also been widely implicated in mechanisms of experience, learning, and memory. Using immunohistochemistry for Fos, as a marker for neural activity, and nitric oxide synthase (NOS), the enzyme that catalyzes the production of nitric oxide (NO), we examined whether sexual activity and sexual experience influence Fos co-expression in NOS-containing neurons in the mPOA of male rats. Consistent with previous findings, results indicate that mating increased activity in the mPOA, and that sexual experience facilitated the expression of sexual behaviors, together with increased mating-induced Fos and NOS in the mPOA. Results also indicate that mating increased co-expression of Fos in NOS-containing neurons, and that this increase was highest in animals undergoing their first sexual encounter, indicating that initial sexual experience increases NO production in the mPOA of male rats.

RESUMEN

Extra group copulations are rarely observed among the Alouatta genus, which lives in a fragmented area in Brazil. Here, we describe the second extra group copulation in A. caraya in literature, which was the first from Brazil. This happened in the southeast urban island forest, which measures 25 ha. In this forest there are two troops: one of them with thirteen howlers and the other with four. These howlers may be extinct in a few years since in the region there is only 3.89 % of native forests and high anthropic action due to agribusiness. We discuss the possible issues that led to the extra group copulation.


Cópulas extra grupo raramente são observados em animais do gênero Alouatta, que vivem em áreas fragmentadas do Brasil. Aqui, descrevemos a segunda cópula extra grupo em A. caraya na literatura, que foi a primeira do Brasil. Isso aconteceu na floresta ilha urbana sudeste, que mede 25 ha. Nesta floresta existem dois bandos: um deles com treze bugios e o outro com quatro. Estes bugios podem ser extintos em poucos anos, uma vez que na região há apenas 3,89% de florestas nativas e alta ação antrópica, devido ao agronegócio. Nós discutimos os possíveis aspectos que levaram à cópula extra grupo.

RESUMEN

Forced copulation is characterized by jerky and rapid movements performed by a female in an apparent attempt to escape from a mounting male. This short communication reports a forced copulation attempt by the gymnophthalmid lizard Micrablepharus maximiliani in a forest enclave inside the Caatinga biome, Tenente Laurentino Cruz municipality, Rio Grande do Norte, Brazil. The rejection behavior of the female consisted of running, pushing and hiding from the mounting male. Although the causes of female mate rejection remain unclear, it may be possible that on this occasion the female M. maximiliani may not have been reproductively active and/or avoided the male deliberately.


A cópula forçada é caracterizada por movimentos bruscos e rápidos realizados por uma fêmea em uma aparente tentativa de escapar de um macho no ato da monta. Esta comunicação relata uma tentativa de cópula forçada pelo lagarto gimnoftalmídeo Micrablepharus maximiliani em um enclave de floresta dentro do bioma Caatinga, município de Tenente Laurentino Cruz, Rio Grande do Norte, Brasil. O comportamento de rejeição da fêmea consistiu de correr, empurrar e se esconder do macho durante a monta. Embora as causas da rejeição do parceiro pela fêmea não sejam claras, pode ser possível que nesta ocasião a fêmea de M. maximiliani não estava reprodutivamente ativa e/ou evitou o macho deliberadamente.

RESUMEN

Copulatory linking of male and female genitalic structures in 11 Neotropical species of Chrysomelidae and one species of Megalopodidae was studied by freezing and then dissecting pairs of beetles in copula. In Megalopus armatus (Megalopodidae) the male has a long endophallus with complex membranous protuberances and a terminal flagellum that probably reaches the spermatheca. In the subfamily Eumolpinae the females have telescoping ovipositors through which the male endophalli pass, reaching to or near the mouth of the spermathecal duct. A long thin flagellum is probably inserted into the spermathecal duct. The male endophalli are braced inside the female using various structures, including two pairs of lateral appendages and apical appendages (both lateral pairs sclerotized in Colaspis sanjoseana and only the basal pair in Brachypnoea irazuensis), a pair of membranous swellings (in Metaxyonycha amasia), and apical microspicules on the endophallus (in Xanthonia). In the subfamily Galerucinae, males of Metrioidea and Diabrotica (tribe Galerucini) have relatively short endophalli ornamented with sclerotized hooks, spines and needles. In Metrioidea elongata the long needle-like endophallic spines of the male were erected inside the female and penetrated the wall of her bursa. In the tribe Alticini, the male endophallus is very short and does not enter the female in two species, Alagoasa gemmata and Walterianella sp. Instead, the apical parts of the female bursae were everted, and were clamped by sclerites of the male median lobe. The male genitalia of Plectrotetra hirsuta reached deep into the female, to the median oviduct, far beyond the mouth of the spermathecal duct. Both rigid and membranous structures meshed in complex ways. When these observations are combined with published descriptions of mating in other Chrysomelidae, the great diversity of copulatory morphology in these families comes into sharper focus.


Se describen los coajustes mecánicos entre las estructuras genitales de machos y hembras de 11 especies neotropicales de Chrysomelidae y una especie de Megalopodidae. Las parejas fueron congeladas durante la cópula y después disectadas. En Megalopus armatus (Megalopodidae) el macho tiene un endofalo largo, con protuberancias membranosas complejas, y un flagelo terminal que probablemente penetra el largo del ducto de la espermateca hasta el lumen de la espermateca. En la subfamilia Eumolpinae, las hembras tienen ovipositores telescópicos, a través de los cuales los endofali de los machos pasan para llegar hasta o cerca a la boca del ducto de la espermateca. Es probable que el largo y delgado flagelo se inserta en el ducto de la espermateca. El endofalo del macho se apoya dentro de la hembra por varias estructuras, las cuales incluyen dos pares de apéndices laterales y apéndices apicales (ambos pares laterales son esclerotizados en Colaspis sanjoseana, únicamente el par basal en Brachypnoea irazuensis), un par de hinchamientos membranosos (en Metaxyonycha amasia), y espículas apicales sobre el endofalo (en Xanthonia). RESUMEN

Copulation in Opisthogonimus fonsecai Ruiz & Leão, 1942 (Trematoda, Digenea, Plagiorchiidae), parasite of Bothrops moojeni Hoge, 1966 (Serpentes, Viperiade), was reported. Data on the histology of the cirrus and metraterm of the copulants were presented. Morphology of the cirrus was also examined by scanning electron microscopy.(AU)
