RESUMEN

The possible interference of resistant pest’s populations to insecticides in natural enemies in the action thas not been clarified yet. Thus, this study aimed to evaluate Trichogramma pretiosum Riley (Hymenoptera: Trichogrammatidae) performance on Spodoptera frugiperda (J. E. Smith) (Lepidoptera: Noctuidae) eggs with resistance frequency to the Metaflumizone over six generations of product exposure. Egg cards (2.0 x 7.0 cm) containing eggs from two populations of S. frugiperda, (resistant to Metaflumizone and the other susceptible), were exposed to T. pretiosum females for 24 hours in free-choice and no-choice testing in three generations (G1, G4, and G6). A completely randomized experimental design was used with 25 replications, each consisting of an egg card (experimental unit) containing 20 eggs. The parameters evaluated were: parasitism (%), emergence (%), sex ratio, number of emerged parasitoids per egg and males/females longevity. ANOVA and Tukey test (P≤ 0.05) were applied on the results. Results showed a reduction in parasitism [41.0% (G1) and 28.4% (G4)], egg emergence (17.5%) and parasitoids/egg [16.2 (G4) and 17.2 (G6)] in eggs originating from the population with resistance frequency. Females emerging from G6 populations eggs without exposure to Metaflumizone had greater longevity (3.5 days more) than the resistant population. The sex ratio and male longevity were not affected. The results indicate a reduction in T. pretiosum activity if S. frugiperda populations have some frequency of resistance to Metaflumizone.


A possível interferência de populações de pragas resistentes na ação de inimigos naturais ainda não foi esclarecida. Assim, este trabalho teve como objetivo avaliar o desempenho de Trichogramma pretiosum Riley (Hymenoptera: Trichogrammatidae) em ovos de Spodoptera frugiperda (J. E. Smith) (Lepidoptera: Noctuidae) com frequência de resistência à Metaflumizona ao longo de seis gerações de exposição ao produto. Cartelas (2,0 x 7,0 cm) com ovos de duas populações de S. frugiperda, (resistente à Metaflumizona e outra suscetível), foram expostas às fêmeas de T. pretiosum por 24 horas em condições de livre escolha e sem chance de escolha por três gerações (G1, G4 e G6). O delineamento experimental foi inteiramente casualizado com 25 repetições, sendo cada repetição composta por uma cartela (unidade experimental) contendo 20 ovos. Os parâmetros avaliados foram: parasitismo (%), emergência (%), razão sexual, número de parasitoides emergidos por ovo e longevidade de machos e fêmeas. ANOVA e teste de Tukey (P≤ 0,05) foram aplicados aos dados coletados. Os resultados mostraram redução do parasitismo [41,0% (G1) e 28,4% (G4)], emergência de ovos (17,5%) e parasitoides/ovo [16,2 (G4) e 17,2 (G6)] em ovos oriundos da população com frequência de resistência. As fêmeas emergidas de ovos da população G6 sem exposição à Metaflumizona, tiveram maior longevidade (3,5 dias a mais) do que a população exposta ao inseticida. A razão sexual e a longevidade de machos não foram afetadas. Os resultados indicam uma redução na atividade de T. pretiosum se as populações de S. frugiperda apresentarem alguma frequência de resistência à Metaflumizona.

Microplitis pallidipes Szépligeti (Hymenoptera: Braconidae) is an important parasitic wasp of second and third-instar noctuid larvae such as the insect pests Spodoptera exigua, Spodoptera litura, and Spodoptera frugiperda. As in other insects, M. pallidipes has a chemosensory recognition system that is critical to foraging, mating, oviposition, and other behaviors. Odorant-binding proteins (OBPs) are important to the system, but those of M. pallidipes have not been determined. This study used PacBio long-read sequencing to identify 170,980 M. pallidipes unigenes and predicted 129,381 proteins. Following retrieval of possible OBP sequences, we removed those that were redundant or non-full-length and eventually cloned five OBP sequences: MpOBP2, MpOBP3, MpOBP8, MpOBP10, and MpPBP 429, 429, 459, 420, and 429 bp in size, respectively. Each M. pallidipes OBP had six conserved cysteine residues. Phylogenetic analysis revealed that the five OBPs were located at different branches of the phylogenetic tree. Additionally, tissue expression profiles indicated that MpOBP2 and MpPBP were mainly expressed in the antennae of male wasps, while MpOBP3, MpOBP8, and MpOBP10 were mainly expressed in the antennae of female wasps. MpOBP3 was also highly expressed in the legs of female wasps. Temporal profiles revealed that the expression of each M. pallidipes OBP peaked at different days after emergence to adulthood. In conclusion, we identified five novel odorant-binding proteins of M. pallidipes and demonstrated biologically relevant differences in expression patterns.


Title: Identification et profil d'expression des protéines de liaison aux odeurs chez la guêpe parasite Microplitis pallidipes à l'aide du séquençage à lecture longue PacBio. Abstract: Microplitis pallidipes Szépligeti (Hymenoptera : Braconidae) est une importante guêpe parasite des larves de noctuelles de deuxième et troisième stades telles que les insectes ravageurs Spodoptera exigua, Spodoptera litura et Spodoptera frugiperda. Comme d'autres insectes, M. pallidipes possède un système de reconnaissance chimiosensoriel, essentiel à la recherche de nourriture, à l'accouplement, à la ponte et à d'autres comportements. Les protéines de liaison aux odeurs (PLO) sont importantes pour le système, mais celles de M. pallidipes n'ont pas été déterminées. Cette étude a utilisé le séquençage à lecture longue PacBio pour identifier 170 980 unigènes de M. pallidipes et prédit 129 381 protéines. Après la récupération des séquences de PLO possibles, nous avons supprimé celles qui étaient redondantes ou pas de pleine longueur et avons finalement cloné cinq séquences de PLO, MpOBP2, MpOBP3, MpOBP8, MpOBP10 et MpPBP, respectivement de taille 429, 429, 459, 420 et 429 pb. Chaque PLO de M. pallidipes avait six résidus de cystéine conservés. L'analyse phylogénétique a révélé que les cinq PLO étaient situés à différentes branches de l'arbre phylogénétique. De plus, les profils d'expression tissulaire ont indiqué que MpOBP2 et MpPBP étaient principalement exprimés dans les antennes des guêpes mâles, tandis que MpOBP3, MpOBP8 et MpOBP10 étaient principalement exprimés dans les antennes des guêpes femelles. MpOBP3 était également fortement exprimé dans les pattes des guêpes femelles. Les profils temporels ont révélé que l'expression de chaque PLO de M. pallidipes culminait à différents jours après l'émergence à l'âge adulte. En conclusion, nous avons identifié cinq nouvelles protéines de liaison aux odeurs de M. pallidipes et démontré des différences biologiquement pertinentes dans les profils d'expression.

The trade-off between plant growth and resistance to herbivory is thought to be at least partly mediated by the interactions between jasmonates and gibberellins (GAs). Insect herbivory activates jasmonate biosynthesis and signaling, and plant growth is concomitantly inhibited. Whether or not the herbivory-induced jasmonates suppress the accumulation of GAs and thus reduce plant growth, and which jasmonates are functional in this process, remain unclear. In this study, we show that herbivory-induced stunted growth of Nicotiana attenuata was completely dependent on allene oxide cyclase (AOC) and coronatine insensitive1 (COI1), which encode a JA biosynthetic enzyme and the receptor, respectively, but only partially dependent on jasmonic acid-isoleucine conjugate (JA-Ile), the bioactive jasmonate. Quantification of GAs and exogenous treatments indicated that herbivory-induced growth inhibition was caused by GA4 deficiency, and that the reduction in GA4 accumulation was strongly associated with both decreased concentrations of GA biosynthetic gene transcripts and transcriptional activation of GA catabolic genes. We further show that JA-Ile only positively regulated the levels of GA catabolic genes, while the accumulation of GA biosynthetic gene transcripts was controlled by certain AOC-derived jasmonate(s) rather than by JA-Ile. This work sheds light on the mechanisms by which plants adapt to herbivory by using intricate phytohormone signaling and transcriptional regulatory networks.

Sufficient energy supply to the host immune system is important for resisting pathogens. Therefore, during pathogen infection, the host metabolism is reassigned from storage, growth, and development to the immune system. Previous studies in Drosophila melanogaster have demonstrated that systemic metabolic switching upon an immune challenge is activated by extracellular adenosine signaling, modulating carbohydrate mobilization and redistributing energy to the hemocytes. In the present study, we discovered that symbiotic virus (SmBV) of the parasitoid wasp Snellenius manilae is able to down-regulate the extracellular adenosine of its host, Spodoptera litura, to inhibit metabolism switching. The decreased carbohydrate mobilization, glycogenolysis, and ATP synthesis upon infection results in the host being unable to supply energy to its immune system, thus benefitting the development of wasp larvae. When we added adenosine to the infected S. litura larvae, we observed enhanced host immune responses that decreased the pupation rate of S. manilae. Previous studies showed that after pathogen infection, the host activates its adenosine pathway to trigger immune responses. However, our results suggest a different model: we found that in S. manilae, SmBV modulates the host adenosine pathway such that wasp eggs and larvae can evade the host immune response.

Abstract The fall armyworm, Spodoptera frugiperda (J.E. Smith) (Lepidoptera: Noctuidae), is an important maize pest. Due to the environmental impact and emergence of resistance caused by chemical pesticides and transgenic events, the use of baculoviruses becomes a safe and useful alternative for its control in integrated pest management strategies. Here we report the identification of a novel isolate of a granulovirus of S. frugiperda native to the central region of Argentina, named SfGV ARG. We observed that larvae infected with SfGV ARG showed a yellowish coloration, swollen body and, in some cases, severe lesions in the last abdominal segments. We confirmed the identity of the isolate by sequencing fragments of the lef-8, lef-9 and granulin genes and by calculating evolutionary distances using the Kimura-2-Parameter model. SfGV ARG DNA restriction pattern allowed to estimate a genome of at least 135 kb.


Resumen La oruga militar tardía, Spodoptera frugiperda (J.E. Smith) (Lepidoptera: Noctuidae), es una plaga importante del maíz. Debido al impacto ambiental y a la aparición de resistencia causados por los pesticidas químicos y los eventos transgénicos, el uso de baculovirus resulta una alternativa útil y saludable para su control en estrategias de manejo integrado de plagas. En este trabajo reportamos la identificación de un nuevo aislamiento del granulovirus de la S. frugiperda nativo de la región central de Argentina, SfGV ARG. Se observó que larvas infectadas con SfGV ARG mostraron coloración amarillenta, hinchazón y, en algunos casos, lesiones graves en los últimos segmentos abdominales. Se confirmó la identidad del aislamiento por secuenciación de fragmentos de los genes lef-8, lef-9y granulina, y por cálculo de distancias evolutivas usando el parámetro de Kimura-2. El patrón de restricción generado con el ADN genómico de SfGV ARG permitió estimar un tamaño de genoma de al menos 135 kb.

Fall armyworm, Spodoptera frugiperda (Smith) (Lepidoptera: Noctuidae) is one of the main pests in maize crop with developing resistance to chemical products and Bt technology. Therefore, alternative control methods such as essential oils are important steps in the implementation management strategies for this pest. This study aimed to evaluate the efficiency of essential oils (EOs) of Corymbia citriodora, Myrciaria dubia (Myrtaceae), Lippia microphylla (Verbenaceae) and Piper umbelattum (Piperaceae) in controlling S. frugiperda. The OEs were extracted and mortality tests were conducted with topic and volatile applications, in 30 second-instar caterpillars originated from insect rearing and artificial diet. As a control, we conducted tests with distilled water and acetone. EOs that provided mortality rates above 80% were submitted to chemical analysis for constituent identification. The efficient EOs were only those of C. citriodora and L. microphylla. For EO of C. citriodora, the LD80 was 7.06 ± 0.73 mg.g-1 in topical application and 5.85 ± 0.75 µL via volatile application. On the other hand, for EO of L. microphylla, DL80 was 9.95 ± 1.25 mg.g-1 in topical application and 18.56 ± 3.55 µL via volatile application. Chemical analysis showed that the main constituents were citronella for the EO of C. citriodora and (E)-caryophyllene and (E)-nerolidol to the EO of L. microphylla. EOs of C. citriodora and L. microphylla are promising for controlling S. frugiperda, with emphasis on the volatile effect of C. citriodora oil.(AU)


A lagarta-do-cartucho, Spodoptera frugiperda (Lepdoptera: Noctuidae), é uma das principais pragas na cultura do milho e nos últimos anos vem desenvolvendo resistência a produtos químicos e à tecnologia Bt. Métodos alternativos de controle, como o emprego de óleos essenciais, são um passo importante na implementação de estratégias de manejo para essa praga. O objetivo deste trabalho foi avaliar a eficiência dos óleos essenciais de Corymbia citriodora, Myrciaria dubia (Myrtaceae), Lippia microphylla (Verbenaceae) e Piper umbelattum (Piperaceae) no controle de S. frugiperda. Os óleos essenciais foram extraídos e testes de mortalidade com aplicação tópica e de voláteis foram conduzidos com 30 lagartas de segundo instar provenientes de criação massal e em dieta artificial. Como testemunha, foram conduzidos testes com água destilada e acetona. Os óleos essenciais que proporcionaram taxas de mortalidade acima de 80% foram submetidos à análise química para identificação de seus constituintes. Entre os óleos essenciais, mostraram-se eficientes apenas os de C. citriodora e L. microphylla. Para o óleo essencial de C. citriodora, a DL80 foi de 7,06 ± 0,73 mg.g-1 em aplicação tópica e 5,85 ± 0,75 µL via aplicação do volátil. Já para o óleo essencial de L. microphylla, a DL80 foi de 9,95 ± 1,25 mg.g-1 em aplicação tópica e 18,56 ± 3,55 µL via aplicação do volátil. A análise química demonstrou que os principais constituintes foram citronelal para o óleo essencial de C. citriodora, e (E)-cariofileno e (E)-nerolidol para o de L. microphylla. Os óleos essenciais de C. citriodora e L. microphylla são promissores para o controle de S. frugiperda, com destaque para o efeito volátil do óleo de C. citriodora.(AU)

Early instar larvae of the tobacco cutworm Spodoptera litura (Lepidoptera: Noctuidae) and the beet armyworm Spodoptera exigua (Lepidoptera: Noctuidae) are recognized hosts of the parasitic wasp Microplitis prodeniae Rao and Kurian (Hymenoptera: Braconidae), although M. prodeniae has previously been regarded as monophagous. In this study, we found the immature period and longevity of M. prodeniae developing in S. exigua was similar to that in S. litura. It was shown that the development time of M. prodeniae in S. exigua was 15.1 ± 0.3 d, not significantly different from 15.0 ± 0.2 d in S. litura. The parasitism rate of M. prodeniae attacking S. exigua was significantly lower than on S. litura (65.48 ± 2.29 and 43.83 ± 2.20%, respectively), whilst the female ratio of the wasp's offspring was not significantly different when developing on the two species. M. prodeniae females prefer to oviposit on the second- and third-instar host larvae of S. exigua, rather than other instars. The effects of development of M. prodeniae on two important lepidopterous pests are discussed.

Two physiological changes of Spodoptera litura parasitized by Microplitis bicoloratus are hemocyte-apoptosis and retarded immature development. ß-Chain of Fo F1 -ATPase was found from a S. litura transcriptome. It belongs to a conserved P-loop NTPase superfamily, descending from a common ancestor of Lepidopteran clade. However, the characterization of ß-chain of ATPase in apoptotic cells and its involvement in development remain unknown. Here, the ectopic expression and endogenous Fo F1 -ATPase ß-chain occurred on S. litura cell membrane: in vivo, at the late stage of apoptotic hemocyte, endogenous Fo F1 -ATPase ß-chain was stably expressed during M. bicoloratus larva development from 4 to 7 days post-parasitization; in vitro, at an early stage of pre-apoptotic Spli221 cells by infecting with M. bicoloratus bracovirus particles, the proteins were speedily recover expression. Furthermore, endogenous Fo F1 -ATPase ß-chain was localized on the apoptotic cell membrane. RNA interference (RNAi) of Fo F1 -ATPase ß-chain led to significantly decreased head capsule width. This suggested that Fo F1 -ATPase ß-chain positively regulated the development of S. litura. The RNAi effect on the head capsule width was enhanced with parasitism. Our research found that Fo F1 -ATPase ß-chain was expressed and localized on the cell membrane in the apoptotic cells, and involved in the development of S. litura.

The endoparasitoid Microplitis prodeniae Rao and Chandry is an important potential augmentative biological control agent for lepidopteran pests of vegetables and tobacco. However, cold storage of pupae is required to ensure that sufficient parasitoids are available when they are needed in the field. In this study, pupae were maintained at 0, 4 or 10°C for 5-50 days after which the adults were evaluated for emergence, pre-emergence period, sex ratio, female longevity, oviposition period, and fecundity. Cold storage did not affect the pre-emergence period or proportion of females; however, there was a significant reduction in emergence, female longevity, oviposition period, and fecundity with increased exposure to cold. The pre-emergence period was approximately 5 days, and approximately 50% of the emergent parasitoids were females. A cold storage regime of 10 days at 10°C had no effect on the parasitoids and adult emergence was greater than 50% even after 20 days at 10°C. There was no carryover of the cold treatment from parental to F1 and F2 generations. Thus, M. prodeniae can be stockpiled for field release by exposing the pupae to a cold regime and subsequently holding them for adult emergence at 28°C.

Abstract The fall armyworm Spodoptera frugiperda (Lepidoptera; Noctuidae) is a voracious pest of numerous crops of economic importance throughout the New World. In Brazil, its larvae are attacked by several species of parasitoid wasps, making them potential candidate as biological control agents against this pest. A survey of the parasitoid fauna on S. frugiperda in maize crops throughout Brazil reveals two species of Campoletis, which are morphologicaly very similar species. In this paper we combine these data with pictures from the type material of C. sonorensis and C. flavicincta, as well as their descriptions to provide a redescription to Campoletis sonorensis (Cameron, 1886) using for this both morphological characters and DNA Barcoding (Hebert et al., 2003) information, in an attempt to help with the correct identification of the taxa to improve biological control studies.


Resumo Spodoptera frugiperda é uma praga voraz de diversas culturas de importância econômica no Novo Mundo. No Brasil, suas larvas podem ser atacadas por diversas espécies de vespas parasitóides que são candidatos a agentes de controle biológico contra essa praga. Pesquisando os parasitóides da fauna de Spodoptera frugiperda em cultivos de milho no Brasil foram encontradas duas espécies do gênero Campoletis, muito similares morfologicamente. Este trabalho apresenta uma redescrição para Campoletis sonorensis usando caracteres morfológicos e DNA Barcoding (Hebert et al., 2003) com o objetivo de evitar erros de identificação desse grupo, aprimorando estudos de controle biológico.

Insect cellular immune responses include encapsulation, nodule formation, and phagocytosis. Hemichannels and gap junctions are involved in these cellular actions. Innexins (Inxs: analogous to the vertebrate connexins) form hemichannels and gap junctions, but the molecular mechanisms underlying their biology is still unclear. In this article, we reported a steady-state level of Inxs (SpliInxs) in hemocytes of Spodoptera litura, which formed nonfunctional hemichannels on the cell surface to maintain normal metabolism. We also reported that two innnexins (SpliInx2 and SpliInx3) were expressed significantly higher in hemocytes compared to other tissues, suggesting that they play important roles in hemocytes. Amino acid analysis found that two cysteine residues in two extracellular loops provided the capability for SpliInx2 and SpliInx3 hemichannels to dock into gap junctions. Western blotting demonstrated that both extracellular and intracellular loops of SpliInx3 and the extracellular loops of SpliInx2 might undergo posttranslational modification during the formation of a steady-state hemichannel. During hemichannel formation, SpliInx2 presented as one isoform, while SpliInx3 presented as three isoforms. These results provide fundamental knowledge for further study of how steady-state levels of SpliInxs are dynamically adjusted to perform cellular immune responses under immune challenge.

In the context of the modern agriculture, pest control is important in order to increase productivity in maize (Zea mays L.). However, this control should be done rationally, prioritising environmentally safer methods such as biological control. This paper aims to report the occurrence of Dolichozele koebelei Viereck, 1911 (Hymenoptera: Braconidae) in Spodoptera frugiperda (J. E. Smith, 1797) (Lepidoptera: Noctuidae) larvae collected in maize subjected to different cropping systems. The experiment was conducted at the Centro Nacional de Pesquisa de Milho e Sorgo (CNPMS) in Sete Lagoas, Minas Gerais State, Brazil, using organic and conventional production. Ten plants were sampled from each of the 24 plots and for each production system, three times a week during the entire cycle of maize (variety BR 106). In the laboratory, larvae were distributed in individual rearing containers with artificial diet until the end of the biological cycle. An increased number of S. frugiperda larvae was observed in organic single crop maize; hence a higher percentage of S. frugiperda larvae parasitised by Hymenoptera and Diptera also occurred in the maize under this production system. Dolichozele koebelei had not yet been described in association with larvae of S. frugiperda. The percentage of parasitism of S. frugiperda larvae was high in both experiments, indicating the importance of natural control agents in reducing the population density of S. frugiperda, and especially the importance of an appropriate crop management.


No contexto agrícola moderno, o controle de pragas é importante para se obter aumento de produtividade na cultura do milho (Zea mays L.). No entanto, esse controle deve ser feito racionalmente, e deve priorizar métodos ambientalmente mais seguros, como o controle biológico. Este trabalho teve como objetivo relatar a ocorrência de Dolichozele koebelei Viereck, 1911 (Hymenoptera: Braconidae) em larvas de Spodoptera frugiperda (J. E. Smith, 1797) (Lepidoptera: Noctuidae) coletadas em milho sob diferentes formas de cultivo. O experimento foi conduzido no Centro Nacional de Pesquisa de Milho e Sorgo (CNPMS) em Sete Lagoas, Minas Gerais, Brasil, em áreas de produção orgânica e convencional. Durante todo o ciclo do milho (cultivar BR 106), três vezes por semana, foram amostradas em cada uma das 24 parcelas, 10 plantas, que foram avaliadas no laboratório. As larvas encontradas foram distribuídas individualmente em recipientes de criação com dieta artificial até o final do ciclo biológico. Maior número de larvas de S. frugiperda foi obtido no milho orgânico solteiro, com maior percentual de larvas de S. frugiperda parasitadas por Hymenoptera ou Diptera. Dolichozele koebelei ainda não havia sido descrita em associação com larvas de S. frugiperda. De uma forma geral nos dois experimentos o percentual de larvas de S. frugiperda parasitadas, foi alto, indicando a importância dos agentes de controle natural na redução da densidade populacional de S. frugiperda e, principalmente, a importância do manejo adequado da cultura do milho.

Although apomixis is the most common form of parthenogenesis in diplodiploid arthropods, it is uncommon in the haplodiploid insect order Hymenoptera. We found a new type of spontaneous apomixis in the Hymenoptera, completely lacking meiosis and the expulsion of polar bodies in egg maturation division, on the thelytokous strain of a parasitoid wasp Meteorus pulchricornis (Wesmael) (Braconidae, Euphorinae) on pest lepidopteran larvae Spodoptera litura (Fabricius) (Noctuidae). The absence of the meiotic process was consistent with a non-segregation pattern in the offspring of heterozygous females, and no positive evidence was obtained for the induction of thelytoky by any bacterial symbionts. We discuss the conditions that enable the occurrence of such rare cases of apomictic thelytoky in the Hymenoptera, suggesting the significance of fixed heterosis caused by hybridization or polyploidization, symbiosis with bacterial agents, and occasional sex. Our finding will encourage further genetic studies on parasitoid wasps to use asexual lines more wisely for biological control.

The tropical armyworm, Spodoptera litura (F.) (Lepidoptera: Noctuidae), is an important pest of tobacco, Nicotiana tabacum L. (Solanales: Solanaceae), in South China that is becoming increasingly resistant to pesticides. Six potential trap crops were evaluated to control S. litura on tobacco. Castor bean, Ricinus communis L. (Malpighiales: Euphorbiaceae), and taro, Colocasia esculenta (L.) Schott (Alismatales: Araceae), hosted significantly more S. litura than peanut, Arachis hypogaea L. (Fabales: Fabaceae), sweet potato, Ipomoea batata Lam. (Solanales: Convolvulaceae) or tobacoo in a greenhouse trial, and tobacco field plots with taro rows hosted significantly fewer S. litura than those with rows of other trap crops or without trap crops, provided the taro was in a fast-growing stage. When these crops were grown along with eggplant, Solanum melongena L. (Solanales: Solanaceae), and soybean, Glycines max L. (Fabales: Fabaceae), in separate plots in a randomized matrix, tobacco plots hosted more S. litura than the other crop plots early in the season, but late in the season, taro plots hosted significantly more S. litura than tobacco, soybean, sweet potato, peanut or eggplant plots. In addition, higher rates of S. litura parasitism by Microplitis prodeniae Rao and Chandry (Hymenoptera: Bracondidae) and Campoletis chlorideae Uchida (Ichnumonidae) were observed in taro plots compared to other crop plots. Although taro was an effective trap crop for managing S. litura on tobacco, it did not attract S. litura in the seedling stage, indicating that taro should either be planted 20-30 days before tobacco, or alternative control methods should be employed during the seedling stage.

Both Microplitis prodeniae Rao and Chandry (Hymenoptera: Bracondidae) and Campoletis chlorideae Uchida (Hymenoptera: Ichnumonidae) are major parasitoids of Spodoptera litura (Fabricious) (Lepidoptera: Noctuidae) in tobacco, Nicotiana tabacum L. (Solanales: Solanaceae) at Nanxiong, Guangdong Province, South China. The niches and interspecific competition relationships of the two species were studied. The results show that the competition between the two species for spatial and food resources was very intense, and C. chlorideae was always dominant when the two species compete for spatial and food resources in different periods. Thus C. chlorideae may drive M. prodeniae away when they occupy the same spatial or food resource. The adaptability of C. chlorideae to the environment in the tobacco fields may be greater than that of M. prodeniae, so C. chlorideae can maintain a higher population compared to that of M. prodeniae.

Spodoptera frugiperda (J E Smith) is an important pest of several crops, but especially on maize in Brazil. The implementation of biological control measures hinges on the identification of its predators and other natural enemies. As a means of identifying predators, antibodies against S. frugiperda eggs were generated by inoculating rabbits with macerated S. frugiperda eggs, and the production of antibodies against S. frugiperda egg proteins was verifi ed by double immunodiffusion (DID). These antibodies were then utilized in another serological technique, counterimmunoeletrophoresis (CIE), to identify insects that could have ingested S. frugiperda eggs. Macerates of entire insects collected in maize plantations and of individual parts of their digestive tract, including the crop, were the source of antigens in the CIE, while predators fed S. frugiperda eggs in the laboratory served as the control. Antibodies produced by the inoculated rabbits were effective in detecting S. frugiperda egg proteins, especially if crop macerates were used as antigens. Among the species of insects collected from maize plantations, Lagria villosa Fabricius (Coleoptera: Lagriidae) and a species of Lygaeidae (Hemiptera) were identified as possible S. frugiperda predators.

We assessed in the laboratory the attraction of the parasitoid Cotesia marginiventris (Cresson) toward odors emitted by conventional maize (Zea mays L. ssp. mays) and Bt (Bacillus thuringiensis) maize seedlings following actual or simulated injury by Spodoptera frugiperda (Smith), the parasitoid's host, and emitted by the host's frass, produced following consumption of conventional or Bt maize seedlings. Females of C. marginiventris exhibited similarly strong responses to conventional and Bt maize seedlings injured by the host or with simulated injury, and these were stronger than responses to clean air. In contrast, the responses of C. marginiventris females were consistently weaker toward host frass derived from Bt maize tissue compared to frass derived from conventional maize tissue. We hypothesized that the weakened response was due to a detrimental effect of Bt endotoxins, present in the Bt maize tissue, on the bacterial community present in the host's gut and frass, including bacteria that produce odors attractive to C. marginiventris. As an initial test of our hypothesis, we compared between the responses of C. marginiventris females to host frass produced following consumption of Bt maize and frass produced from conventional maize which had been treated with an antibiotic (tetracycline) to eliminate host gut bacteria. Our results showed that C. marginiventris females responded similarly weakly to host frass derived from conventional maize tissue treated with antibiotic and to frass derived from Bt maize tissue, treated or untreated with antibiotic, while they responded strongly to frass derived from conventional maize untreated with antibiotic, so provided initial, partial support for our hypothesis. We discussed the weakened response of C. marginiventris females to host frass derived from Bt maize in the context of plausible impacts of transgenic crop cultivars on parasitoid foraging and populations, and the implications for biological control of non-target, polyphagous pests, such as S. frugiperda.

Este trabalho teve por objetivo verificar a interação entre o parasitoide Telenomus remus e o predador Doru luteipes. Para tanto, posturas deS. frugiperda parasitadas porT. remus foram ofertadas a D. luteipes e ovos de D. luteipes foram ofertados a T. remus. Essa oferta levou em consideração diferentes fases do desenvolvimento embrionário do parasitoide e do predador. Constatou-se que quando as posturas de S. frugiperda foram expostas primariamente a T. remus. D. luteipes as consumiu, mas somente até o terceiro dia após o parasitismo.T. remus parasitou ovos deD. luteipes com até 48h de desenvolvimento embrionário, mas apenas quando as fêmeas do predador estavam ausentes. Diante disso, a interação intraguilda parece não ser um fator importante de regulação populacional destas espécies.


This work aimed at evaluating the interaction between Telenomus remus andD. luteipes. S. frugiperda eggs previously parasitized by T. remus were offered to D. luteipes and D. luteipes eggs were offered to T. remus. To study the influence of different embryonic development stages of the parasitoid, we offered S. frugiperda previously parasitized eggs at different embryonic development stages to the predator. And we offered predator eggs at different embryonic development stages to the parasitoid. The behavior of insects was observed. When S. frugiperda eggs were exposed to T. remus previously, D. luteipes eat the eggs until three days after parasitism. T. remus parasitized D. luteipes eggs until 48h they were laid, but only when predator females were absent.

Eighteen Xenorhabdus isolates associated with Spanish entomopathogenic nematodes of the genus Steinernema were characterized using a polyphasic approach including phenotypic and molecular methods. Two isolates were classified as Xenorhabdus nematophila and were associated with Steinernema carpocapsae. Sixteen isolates were classified as Xenorhabdus bovienii, of which fifteen were associated with Steinernema feltiae and one with Steinernema kraussei. Two X. bovienii Phase II were also isolated, one instable phase isolated from S. feltiae strain Rioja and one stable phase from S. feltiae strain BZ. Four representative bacterial isolates were chosen to study their pathogenicity against Spodoptera littoralis with and without the presence of their nematode host. The four bacterial isolates were pathogenic for S. littoralis leading to septicemia 24h post-injection and killing around 90% of the insect larvae 36 h post-injection, except for that isolated from S. kraussei. After 48 h of injection, this latter isolate showed a lower final population in the larval hemolymph (10(7) instead of 10(8)CFU per larvae) and a lower larval mortality (70% instead of 95-100%). The virulence of the nematode-bacteria complexes against S. littoralis showed similar traits with a significant insect larvae mortality (80-90%) 5 days post-infection except for S. kraussei, although this strain reached similar of larval mortality at 7 days after infection.

Leishmania actin was cloned, overexpressed in baculovirus-insect cell system, and purified to homogeneity. The purified protein polymerized optimally in the presence of Mg2+ and ATP, but differed from conventional actins in its following properties: (i) it did not polymerize in the presence of Mg2+ alone, (ii) it polymerized in a restricted range of pH 7.0-8.5, (iii) its critical concentration for polymerization was found to be 3-4-fold lower than of muscle actin, (iv) it predominantly formed bundles rather than single filaments at pH 8.0, (v) it displayed considerably higher ATPase activity during polymerization, (vi) it did not inhibit DNase-I activity, and (vii) it did not bind the F-actin-binding toxin phalloidin or the actin polymerization disrupting agent Latrunculin B. Computational and molecular modeling studies revealed that the observed unconventional behavior of Leishmania actin is related to the diverged amino acid stretches in its sequence, which may lead to changes in the overall charge distribution on its solvent-exposed surface, ATP binding cleft, Mg2+ binding sites, and the hydrophobic loop that is involved in monomer-monomer interactions. Phylogenetically, it is related to ciliate actins, but to the best of our knowledge, no other actin with such unconventional properties has been reported to date. It is therefore suggested that actin in Leishmania may serve as a novel target for design of new antileishmanial drugs.
