RESUMO

OBJECTIVE: Eyebrow supraorbital craniotomy is a versatile keyhole technique for treating intracranial pathologies. The eyelid supraorbital approach, an alternative approach to an eyebrow supraorbital craniotomy, has not been widely adopted among most neurosurgeons. The purpose of this systematic review and meta-analysis was to perform a pooled analysis of the complications of eyebrow or eyelid approaches for the treatment of aneurysms, meningiomas, and orbital tumors. METHODS: A systematic review of the literature in the PubMed, Embase, and Cochrane Review databases was conducted for identifying relevant literature using keywords such as "supraorbital," "eyelid," "eyebrow," "tumor," and "aneurysm." Eyebrow supraorbital craniotomies with or without orbitotomies and eyelid supraorbital craniotomies with orbitotomies for the treatment of orbital tumors, intracranial meningiomas, and aneurysms were selected. The primary outcomes were overall complications, cosmetic complications, and residual aneurysms and tumors. Secondary outcomes included five complication domains: orbital, wound-related, scalp or facial, neurological, and other complications. RESULTS: One hundred three articles were included in the synthesis. The pooled numbers of patients in the eyebrow and eyelid groups were 4689 and 358, respectively. No differences were found in overall complications or cosmetic complications between the eyebrow and eyelid groups. The proportion of residuals in the eyelid group (11.21%, effect size [ES] 0.26, 95% CI 0.12-0.41) was significantly higher (p < 0.05) than that in the eyebrow group (6.17%, ES 0.10, 95% CI 0.08-0.13). A subgroup analysis demonstrated significantly higher incidences of orbital, wound-related, and scalp or facial complications in the eyelid group (p < 0.05), but higher other complications in the eyebrow group. Performing an orbitotomy substantially increased the complication risk. CONCLUSIONS: This is the first meta-analysis that quantitatively compared complications of eyebrow versus eyelid approaches to supraorbital craniotomy. This study found similar overall complication rates but higher rates of selected complication domains in the eyelid group. The literature is limited by a high degree of variability in the reported outcomes.

RESUMO

PURPOSE: There is a growing body of literature documenting glioma heterogeneity in terms of radiographic, histologic, molecular, and genetic characteristics. Incomplete spatial specification of intraoperative tumor samples may contribute to variability in the results of pathological and biological investigations. We have developed a system, termed geo-tagging, for routine intraoperative linkage of each tumor sample to its location via neuronavigation. METHODS: This is a single-institution, IRB approved, prospective database of undergoing clinically indicated surgery. We evaluated relevant factors affecting data collection by this registry, including tumor and surgical factors (e.g. tumor volume, location, grade and surgeon). RESULTS: Over a 2-year period, 487 patients underwent craniotomy for an intra-axial tumor. Of those, 214 underwent surgery for a newly diagnosed or recurrent glioma. There was significant variation in the average number of samples collected per registered case, with a range of samples from 2.53 to 4.75 per tumor type. Histology and grade impacted on sampling with a range of 2.0 samples per tumor in Grade four, IDH-WT gliomas to 4.5 samples in grade four, IDH-mutant gliomas. The range of cases with sampling per surgeon was 6 to 99 with a mean of 47.6 cases and there was a statistically significant differences between surgeons. Tumor grade did not have a statistically significant impact on number of samples per case. No significant correlation was found between the number of samples collected and enhancing tumor volume, EOR, or volume of tumor resected. CONCLUSION: We are using the results of this analysis to develop a prospective sample collection protocol.

RESUMO

Atherosclerosis, chronic non-communicable diseases, and metabolic syndrome are highly interconnected and collectively contribute to global health concerns that reduce life expectancy and quality of life. These conditions arise from multiple risk factors, including inflammation, insulin resistance, impaired blood lipid profile, endothelial dysfunction, and increased cardiovascular risk. Adopting a plant-based diet has gained popularity as a viable alternative to promote health and mitigate the incidence of, and risk factors associated with, these three health conditions. Understanding the potential benefits of a plant-based diet for human health is crucial, particularly in the face of the rising prevalence of chronic diseases like diabetes, hypertension, dyslipidemia, atherosclerosis, and cancer. Thus, this review focused on the plausible advantages of consuming a type of food pattern for the prevention and/or treatment of chronic diseases, emphasizing the dietary aspects that contribute to these conditions and the evidence supporting the benefits of a plant-based diet for human health. To facilitate a more in-depth analysis, we present separate evidence for each of these three concepts, acknowledging their intrinsic connection while providing a specific focus on each one. This review underscores the potential of a plant-based diet to target the underlying causes of these chronic diseases and enhance health outcomes for individuals and populations.

RESUMO

PURPOSE: Despite strong induction chemotherapy response rates, only 30%-40% of patients with adult B-cell acute lymphoblastic leukemia (ALL) attain long-term remission. This study analyzes ALL in Latin America (LA) and recommends diagnosis, treatment, and management protocols. METHODS: The Americas Health Foundation organized a panel of hematologists from Argentina, Brazil, Chile, Colombia, and Mexico to examine ALL diagnosis and therapy and produce recommendations. RESULTS: Lack of regional data, unequal access to diagnosis and therapy, inadequate treatment response, and uneven health care distribution complicate adult ALL management. The panel recommended diagnosis, first-line and refractory treatment, and post-transplantation maintenance. Targeted treatments, including rituximab, blinatumomab, and inotuzumab ozogamicin, are becoming available in LA and must be equitably accessed. CONCLUSION: This review adapts global information on treating ALL to LA. Governments, the medical community, society, academia, industry, and patient advocates must work together to improve policies.

SUMMARY: The regular practice of a certain type of training is one of the strategies used to optimize body composition. Consequently, nutritional intervention is not always considered as part of interdisciplinary work. This study aimed to identify the effects of a nutritional intervention program associated with concurrent training on body composition, assessed through bioelectrical impedance (BIA) in physically active men. The intervention lasted 6 months, with the participation of 12 subjects (23.4 ± 4.9 years). During the first 5 months, food and training planning was applied, adjusted to the individual characteristics of each of the participants. At the beginning of the 6th month, food planning was eliminated and only training was maintained, to analyze the possible changes in body composition of the participants. Prior to (before the intervention program), during (at the end of month 5), and after the intervention (end of month 6), adipose tissue, muscle, and fat-free mass were evaluated through electrical bioimpedance (BIA). The results showed that in the first five months, this training program generated a decrease in the percentage of adipose tissue (PRE= 16.20 ± 4.75; POST= 9.52 ± 3.70; p= 0.000; ES= 1.51) and adipose tissue index (PRE= 5.34 ± 1.75; POST= 3.16 ± 1.34; p=0.001; ES=1.35), increase in the percentage of muscle tissue (PRE= 46.05 ± 3.02; POST= 49.90 ± 2.90; p=0.004; ES= -1.25) and increased muscle/fat index (PRE= 2.53 ± 0.75; POST = 4.85 ± 2.64, p=0.005, SE= -1.15). The above effects were significantly reduced when the nutritional intervention was withdrawn from the program (p>0.05). It is concluded that nutritional intervention is a key factor to generate positive effects in the optimization of body composition regardless of the level of training. These results show the importance of individualized food planning, carried out by a professional nutritionist with regard to changes in the body composition of physically active subjects. The results of this study could be useful for nutritionists when forming disciplinary teams to improve eating habits and body composition.

SUMMARY: Soccer is the most popular sport in the world. Thus, the variables that benefit performance and success have sparked great interest and occupy an essential role in the preparation of athletes. Therefore, the aim of the present study was to analyze the body composition and proportionality according to categories and playing position. 111 male soccer players of the Chilean national soccer team under-15 (14.6±0.3 years), under-17 (16.3±0.5 years), under-20 (19.0±0.7 years), and elite (24.3±4.7 years) categories were evaluated through anthropometry. Values of body composition and proportionality were recorded and organized by position and category; significance was established at p<.05. For weight, goalkeepers had the highest for their position; under-20 players, the highest for their category (83.7±3.8 kg). Regarding height, elite and under-17 central defenders had the highest values (187.0±4.2 cm and 179.9±3.5 cm, respectively). Goalkeepers in all categories have the highest BMI values; the highest were observed in elite (25.1±1.3 kg/m2). Under-17 forwards/extremes had the highest values in terms of muscle mass percentage (60.5±8.0 %), whereas under-15 fullbacks have the lowest values in terms of fat mass (9.6±0.7 %). The players of the Chilean national soccer team have similar values to other researches with players of the same age and category; however, the younger categories have significant differences in the variables that provide advantages during a game with the elite category. Therefore, emphasis should be placed on equating physical conditions before promoting a player to the older categories.

RESUMO

BACKGROUND: the present study aimed to investigate the effects of a single nutritional preventive session previous to a critical period linked to fat gain in university students with overweightness and obesity, emulating a nutritional session of a public health system. METHODS: In this single-blind randomized controlled trial, 23 students met all the criteria to be included (20.91 ± 2.52-year-old; 52.2% women) who were divided into two groups: intervention group (IG) and control group (CG). Fat mass (FM) by dual-energy X-ray absorptiometry (DXA), physical activity by accelerometry, feeding evaluation through three questionnaires, and a set of healthy lifestyle recommendations were evaluated before and after the national holidays (NH). RESULTS: Our findings showed that FM increased significantly in the CG, but not in the IG (CG = 428.1 g; IG = 321.9 g; Δ = 106.2 g; p = 0.654 [95% CI = -379.57, 591.92]). However, no differences were found during the NH between them (Hedges' g effect size = 0.19; p = 0.654). In addition, no statistical differences were observed between groups in feeding evaluations, the set of recommendations performed, and physical activity. CONCLUSION: a single preventive session before a critical period, using a similar counselling approach as used in the public health system, might not be enough to promote changes in eating and physical activity patterns and preventing fat gain in overweight/obese university students. Long-term interventions are a must.

Soccer is the most practiced sport in the world. This popularity is since practically anyone can play it amateurishly, but as it becomes professionalized it requires the valuation of ananthropometric profile that favors a certain role within the playing field. The objective of the study was to compare the anthropometric characteristics of Chilean youth soccer players sub 14, sub 15 and sub 16 according to the game position and category. The number of individuals evaluated in this study was 180 male youth players in the sub 14 categories (n = 60), sub 15 (n = 58) and sub 16 (n = 62), belonging to the lower divisions of the soccer clubs: Audax Italiano (n = 62), Cobresal (n = 57) and Magallanes (n =61) from the Metropolitan Region. The sample was chosen for convenience and consists of the total of players belonging to these three categories and these three soccer teams. These players had an average training of 4 days a week with a duration of each practice session of 120 minutes, plus a match on weekends with a duration of 90 minutes. There were significant differences in all of them between different game positions. We can emphasize that the central defenses presented the greatest statistical differences in all the lengths corresponding to the lower limbs: iliospinal height (97.6 ± 3.6), trochanteric height (92.1 ± 3.3), trochanteric-tibial lateral length (45.2 ± 1.7), lateral tibial height (46.3 ± 1.8) and medial-malleolar medial tibial length (38.7 ± 2.0), whereas the archers had them in the lengths of the train superior: acromio-radial (33.0 ± 1.6), radial-styloid (27.8 ± 1.7) and styloid-dactyly medium (20.2 ± 1.0). It was also found that there are significant differences in weight among players of different categories, with those of sub 16 having the highest weight (65.7 ± 7.4) and those of sub 14 the lowest (58.7 ± 8.1). At the same time, it is shown how there are statistical differences in the size between the sub 14 division who obtained the lowest measurement on average and the other 2 series confronted, but not so between sub 15 and sub 16, the latter being those who had the highest stature on average. It is also shown as the percentage of fat mass, ranging between 24.2 % of the sub 15 division in its lower limit, and 25.2% of the sub 14 in its upper limit being all considered as "acceptable" and without statistical significance between any categories. Statistical differences were found between the sub 14 category and the sub 16 category in all body lengths except in the radial-styloid. It is also shown that there are significant differences between the sub 14 and sub 15 in the lengths: acromio-radial and trochanteric-lateral tibial and in the heights: ilio-spinal, trochanteric and lateral tibial. This is directly related to the lower average stature they present in the sub-division 14. In turn, no statistical significance was found at any length between sub-15 and sub-16 players. Likewise, statistical differences were found in the weight, height, and weight of: skin, fat, bone mass, muscle, residual mass and in all body lengths less than radial-styloid among players of different categories. The results show the importance of dividing the young players in a greater number of positional roles when defining the ideal anthropometric characteristics according to their location in the field of play. The differences found in the body composition between categories suggest paying attention to the development of maturity when promoting a player to a larger division.

Foundation: classical dance requires specific capacities at the level of strength, speed and flexibility. Training implies a long-term training overload and stress; therefor, an imbalance load-recovery may cause a syndrome of overtraining. There are no specific procedures to diagnose it, and then it is necessary to have markers, which provide information previously. Objective: to determine the influence of training phases at the premiere of the season on the autonomic control of cardiac rate, energy consumption and sleep quality in dancers of classical dance. Method: a cross-descriptive study was realized in the Santiago de Chile city, during previous training of classical dance dancers, for the season 2015, which was performed during July. The universe constituted by 25 professional dancers, the sample selected was nine healthy dancers without lesions and/or pharmacological treatment. Variations of cardiac rate food consumption in 24 hours were registered and a survey for quality of sleep. A descriptive statistic was used for the measurements of central trend. For comparisons of phases, the test of t student was uses. For correlation analysis, Spearman test was used. Results: there were no significant differences among the parameters evaluated. It was found a correlation between pNN50 and the score for quality of sleep by Pittsburgh in the phase 1 (r= -0, 77; p= 0, 02). Conclusion: from the statistical point of view, the results suggest that the training phases before the premiere do not influence the variations of cardiac rate, energy intake and quality of sleep. It was clinically observed that the variability tends to decrease while the energy consumption increases and the quality of sleep tends to improve.

Foundation: In team sports as soccer, knowing the player´s profiles may allow a better choice and training. Female soccer, even though it started in the 70´s, only in the last decades it has had a considerable development. Objective: to describe the anthropometric and somatotype of soccer female players. Methods: descriptive study which consisted on evaluating 72 female soccer players pertaining to the Chilean Female Soccer Federation. For obtaining and analyzing their morphologic profile, their body mass, standing height, sitting height, the total sum of six cutaneous folders and eight cutaneous folders were measured according to the protocol of marking and measuring of the International Society for developing Cineanthropometry. There were also calculated the three components of Health-Carter somatotype for each game position. Results: There were not found significant differences of body mass, height, total of six and eight cutaneous folders, there were in the variable sitting height (p<0,05). There were not significant differences at absolute and relative levels of adipose and body mass but there was in relative bone mass (p<0, 05). The 66, 7 % of the players showed a mesomorphism as the main component. Conclusion: Mesomorphic somatotype predominated in all categories and there are significant differences among categories. That is why it is necessary to consider specific strategies for each of them aiming at optimizing the athlete's sport output

RESUMO

In this study, we observed that mice lacking the IL-1 receptor (IL-1R) (IL1r-/-) or deficient in IL1-ß developed multiple epidermal cysts after chronic UVB exposure. Cysts that developed in IL1r-/- mice were characterized by the presence of the hair follicle marker Sox 9, keratins 10 and 14, and normal melanocyte distribution and retinoid X receptor-α expression. The increased incidence of cysts in IL1r-/- mice was associated with less skin inflammation as characterized by decreased recruitment of macrophages, and their skin also maintained epidermal barrier function compared with wild-type mice. Transcriptional analysis of the skin of IL1r-/- mice after UVB exposure showed decreased gene expression of proinflammatory cytokines such as tumor necrosis factor-α and IL-6. In vitro, primary keratinocytes derived from IL1r-/- mice were more resistant to UVB-triggered cell death compared with wild-type cells, and tumor necrosis factor-α release was completely blocked in the absence of IL-1R. These observations illustrate an unexpected yet prominent phenotype associated with the lack of IL-1R signaling in mice and support further investigation into the role of IL-1 ligands in epidermal repair and innate immune response after damaging UVB exposure.

ntroduction: Two important factors have been described as contributing to the onset of fatigue during physical exercise: Decreased muscle glycogen stores and the loss of water with electrolytes through sweating, favoring dehydration. Because of this, fluid and electrolyte replacement after physical exercise becomes the priority to restore altered body homeostasis during physical exercise. Due to its nutritional composition, milk appears to be a good natural alternative as a rehydration beverage after exercise. Objective: To compare the effectiveness chocolate skim milk as pots-exercise rehydration beverage with an artificial sports drink in the military, based on the hypothesis that chocolate skim milk has equal or better effects on post-exercise hydration than an artificial sports drink. Method: 30 soldiers (23.0 ± 3.2 years) were divided into 2 groups: 1) rehydration with sports drink with 6 percent carbohydrates and ratio of Na / K 2: 1 (ISO group). Rehydration with chocolate skim milk (LECHE group). Fluid retention was assessed through body weight and the specific gravity of urine (GEO) before and after physical exercise and after 2 hours of rehydration. Results: significant differences were found (p < 0.05) in fluid retention in the LECHE group compared to the ISO group. There were no significant differences between the GEO of both groups. Conclusion: The results show that rehydration with skim milk is more effective in fluid retention and weight recovery prior to exercise than sports drink(AU)

RESUMO

In acute myeloid leukemia (AML), mutations of the Fms-like tyrosine kinase 3 receptor (FLT3) and its overexpression are related with hyperleukocytosis, higher risk of relapse, and decrease of both disease-free survival and overall survival. It has been suggested that this phenomenon confers proliferative and survival advantages to the malignant blast cells. As a consequence, it is an attractive therapeutic target. As the best treatment strategy for mutated FLT3 AML remains to be defined, the addition of FLT3 inhibitor drugs to chemotherapy or to the bone marrow transplant approach has become a growing strategy. With encouraging results, this combination seems to be an attractive option. Relevant data regarding the current treatment trends on mutated FLT3 AML is reviewed here.

RESUMO

Recent successes in the development of new therapies for metastatic melanoma, such as mitogen-activated protein kinase pathway inhibitors, anticytotoxic T lymphocyte-associated antigen-4, and programmed cell death protein 1/programmed cell death ligand 1 (PD-L1) pathway-blocking antibodies, as well as combination strategies, all yielded promising results, changing the continually evolving landscape of therapeutic options for patients with melanoma. One promising new treatment modality is based on the use of immunomodulatory monoclonal antibodies that enhance the function of components of the antitumor immune response such as T cells or block immunologic checkpoints that restrain effective antitumor immunity. Program death-1 receptor and its ligand, PD-L1, is a major mechanism by which a tumor suppresses T cell-mediated antitumor immune responses. Studies in mice have shown that GK-1, an 18 amino acid peptide from Taenia crassiceps cisticerci, has the potential to be used as a primary or adjuvant component for the treatment of cancers by stimulating proinflammatory cytokines. The authors hypothesized that treatment with GK-1 in combination with anti-PD-L1 will increase survival in mice bearing melanoma tumors. C57BL/6 mice were injected with B16-F10-luc2 cells and separated into four groups: control, GK-1, anti-PD-L1, and GK-1/anti-PD-L1. The tumor sizes were measured and monitored using calipers and bioluminescence. The GK-1 peptide in combination with anti-PD-L1 showed significantly longer survival (34 days) compared with the other groups (23-27 days). This means an increase; survival increased 47.82% in the mice treated with GK-1+anti-PD-L1, 21.7% in mice treated with GK-1 alone, and 6.08% in those mice treated with anti-PD-L1 only. Blood samples were collected at days 0, 14, and at euthanization or end of the experiment and monitored for cytokines using mouse-specific V-PLEX Proinflammatory Panel. A decrease in TNF-α, IL-4, IL-5, IL-6, and IL-10 serum levels was observed in the GK-1/anti-PD-L1 combination group that may explain the beneficial effects of the combination treatment in prolonging the life of mice bearing melanoma. The data indicate that GK-1/anti-PD-L1 combined therapy affectively increases survival and warrants further clinical investigations.

RESUMO

BACKGROUND: The first reported case of intestinal perforation secondary to metastatic lung carcinoma was reported in 1957. Intestinal metastases are present in up to 1.8% of the cases, with small bowel obstruction as the most common clinical presentation. CLINICAL CASE: An 89 year-old male, who was diagnosed with a high-grade pulmonary mucoepidermoid tumour 2 months previously. The patient was admitted to the hospital for 3 days due to diffuse colic abdominal pain of moderate to severe intensity, accompanied by nausea and gastric vomiting, as well as 2 episodes of bloody bowel movements. On physical examination, the patient was noted to have tachycardia and tachypnoea, as well as clinical signs of acute abdomen. He had white cells of 24,900 per mm3, and 87% neutrophils. Exploratory laparotomy was performed, which showed a bowel perforation associated with a tumour mass 15cm beyond the angle of Treitz. Bowel resection and primary anastomosis were performed. The histopathological analysis reported the diagnosis of a high-grade mucoepidermoid tumour with small bowel and mesentery with disease-free surgical margins. Unfortunately the patient had a fatal outcome secondary to hospital-acquired pneumonia. CONCLUSION: The cases of metastases to small bowel are extremely rare, and to our knowledge this is first case reported in Mexico. The patient described went to the emergency room with gastrointestinal bleed and intestinal perforation that required urgent surgical intervention with small bowel resection and primary anastomosis. Unfortunately the patient died secondary to hospital acquired pneumonia.

RESUMO

BACKGROUND: The incidence of acute leukemia (AL) has increased. Its prognosis is variable and depends on several baseline characteristics with a highly heterogeneous presentation. In Mexico, large-scale descriptive studies have not yet been published; the objective of this study was to analyze the initial basic characteristics of patients diagnosed with AL in our population. PATIENTS AND METHODS: In this multicenter, retrospective study, 1018 patients ≥ 16 years of age and diagnosed with AL between 2009 and 2014, were included. We described age, gender, complete blood count, and AL subtype according to flow cytometry analysis. RESULTS: Acute lymphoblastic leukemia (ALL) was as common as acute myeloid leukemia (AML) (51% vs. 49%). The median age was 31 years. Only 9.6% of patients with ALL were positive for the Philadelphia chromosome. No gender differences were observed. The median age at presentation of AML was 43 years. Acute promyelocytic leukemia was the most frequent AML subtype (38.3%), with a median age of 37 years. CONCLUSION: ALL is equally as frequent as AML in patients ≥16 years of age. Philadelphia-positive prevalence is less frequent than that reported in literature. AML cases occur in a younger age in comparison with other countries. There is a higher rate of acute promyelocytic leukemia among our patients compared with other non-Latin American populations. This study is the largest ever performed in Mexico regarding descriptive AL data.

RESUMO

BACKGROUND: Acute myeloid leukemia is typically a disease of the older population and presents mostly in the fifth decade of life. Myeloid sarcoma is a rare initial presentation of acute myeloid leukemia. Previously it has only been documented in children and younger patients. CASE PRESENTATION: We present an unusual case of retro-orbital myeloid sarcoma as an initial presentation of acute myeloid leukemia in a 43-year-old Caucasian man, with rearrangement of chromosome 11q23 involving the MLL gene. CONCLUSIONS: We present an unusual case of retro-orbital myeloid sarcoma as an initial presentation of acute myeloid leukemia in a 43-year-old man, with rearrangement of chromosome 11q23 involving the MLL gene.

RESUMO

OBJECTIVE: In a previous study, we demonstrated the therapeutic efficacy of a subcutaneous injection of GK1 peptide in a melanoma mouse model, effectively increasing the mean survival time by 42.58%, delaying tumor growth, and increasing intratumoral necrosis compared with the control. As a first approach to investigate the anti-melanoma effect of GK1, this study was carried out to determine the hematological effects along with both serum and lung cytokine profiles in a melanoma lung metastatic model. MATERIALS AND METHODS: Thirteen C57BL6 female mice were transfected in the lateral tail vein with 2×10(5) B16-F0 melanoma cells. After 7 days, mice were separated in two different groups and treatments were initiated (day 0): The GK1-treated group (seven mice) were injected every 5 days intravenously with GK1 (10 µg) in the lateral tail vein, and the control group (six mice) were injected every 5 days with intravenous saline solution. Blood samples were collected every 5 days from day 0; tumor samples were obtained for cytokine measurements on the day of sacrifice. RESULTS: In the peripheral blood, mice treated with GK1 presented a statistically significant decrease in IFN-γ (p<0.05), and lymphocytes tended to be lower compared with the control mice (p=0.06). Lung metastatic analysis demonstrated a significant increase in IFN-γ and IL-12p70 (p<0.05); a significant decrease in IL-17, IL-4, IL-22, IL-23, and IL-12p40 (p<0.05); and a marginal decrease in IL-1ß (p=0.07) compared with the control. DISCUSSION: Our results suggest that an intratumoral increase of cytokines with antitumor activity along with an intratumoral decrease of cytokines with protumor activity could explain, in part, the anti-melanoma effects of GK1 in a lung metastatic melanoma mouse model. Further studies must be performed to elucidate the precise mechanisms of action for GK1 peptide against melanoma, and their eventual application in humans.

RESUMO

T leukemogenesis is a multistep process, where the genetic errors during T cell maturation cause the healthy progenitor to convert into the leukemic precursor that lost its ability to differentiate but possesses high potential for proliferation, self-renewal, and migration. A new misdirecting "leukemogenic" signaling network appears, composed by three types of participants which are encoded by (1) genes implicated in determined stages of T cell development but deregulated by translocations or mutations, (2) genes which normally do not participate in T cell development but are upregulated, and (3) nondifferentially expressed genes which become highly interconnected with genes expressed differentially. It appears that each of three groups may contain genes coding ion channels. In T cells, ion channels are implicated in regulation of cell cycle progression, differentiation, activation, migration, and cell death. In the present review we are going to reveal a relationship between different genetic defects, which drive the T cell neoplasias, with calcium signaling and ion channels. We suggest that changes in regulation of various ion channels in different types of the T leukemias may provide the intracellular ion microenvironment favorable to maintain self-renewal capacity, arrest differentiation, induce proliferation, and enhance motility.
