RESUMO

Cadmium (Cd) accumulates in the vegetative tissues of rice and wheat crops, posing a serious threat in the food chain. A long-term field experiment was conducted to investigate the effects of rice husk biochar (RHB), farm manure (FM), press mud (PrM), and poultry manure (PM) on the growth, yield, and economics of wheat and rice crops grown with sewage water. The results showed that RHB increased wheat plant height (27%, 66%, 70%), spike-length (33%, 99%, 56%), straw yield (21%, 51%, 49%), and grain yield (42%, 63%, 65%) in year-1, year-2, and year-3, than respective controls. For rice crop, RHB showed the maximum increase in plant height (64%, 92%, 96%), spike length (55%, 95%, 90%), straw yield (34%, 53%, 55%), and grain yield (46%, 66%, 69%) each year (2019-2021), compared to their respective controls. The Cd immobilization was increased by the application of RHB while other treatments followed FM > PrM > PM > control in each year of wheat and rice crops. For year-1, benefit-cost ratio remained maximum with the application of FM while for the 2nd and 3rd years in sequence, RHB proved more economical than other treatments and consistently produced wheat and rice with lower Cd concentration than FM, PrM, and PM in grains. This long-term experiment suggested that the application of organic amendments consistently increased biomass of rice and wheat and decreased the Cd concentration in tissues. The RHB remained more effective compared with FM, PrM, and PM in terms of yield, low Cd accumulation and economics of rice and wheat crops.

RESUMO

Melatonin (MT) has been demonstrated to provide defense against both biotic and abiotic stressors. Boron toxicity (BT) can significantly limit the growth and production of plants. However, few studies have been conducted on whether MT is effective in attenuating B toxicity in different plants. In order to evaluate the efficacy of exogenous MT treatment in reducing the negative impact of BT on rice seedlings, this study examined the influence of MT on growth, antioxidant capacity, cell wall composition, and proline metabolism in rice seedlings under hydroponics. Four treatments were established: MT (50 µM), MT + BT (50 µM MT + 800 µM B), BT (800 µM), and CK (control) in a completely randomized design. The results indicate that BT had a significant detrimental effect on the shoot length, root length, and root and shoot fresh weights of rice seedlings by 11.96%, 27.77%, 25.69%, and 18.67%, respectively as compared to the control treatment. However, exogenous MT application increased these parameters and reduced B accumulation in aboveground parts (14.05%) of the plant. Exogenous MT also increased the endogenous melatonin content and antioxidant enzyme activities (64.45%, 71.61%, 237.64%, and 55.42% increase in superoxide dismutase, ascorbate peroxidase, and peroxidase activities, respectively), while decreasing reactive oxygen species levels and oxidized forms of glutathione and ascorbic acid. Additionally, MT enhanced the biosynthesis of proline by decreasing proline dehydrogenase (ProDH) and increasing the GSH (glutathione) and ASA (ascorbic acid) contents. Exogenous MT also increased cell wall components that can increase B adsorption to the cell wall. Overall, these findings suggest that MT application can be a potential solution for strengthening the stress tolerance of rice seedlings, particularly under conditions of B toxicity. In regions where soil contains high levels of boron, the use of MT could enhance rice crop yields and quality.

RESUMO

An 82-year-old male presented with progressive dysphagia and simultaneous hoarseness of voice for the past 6 months. He had mitral valve repair and a permanent pacemaker for a heart block 5 years ago. A computed tomographic scan of the neck demonstrated a cervical esophageal diverticulum. Oral Gastrogrifin contrast study confirmed esophageal diverticulum in Killian-Jamieson space. Open surgical diverticulectomy was performed safely. Patient's dysphagia resolved immediately, and he regained his normal voice after 6 weeks. Introduction: Killian-Jamieson's esophageal diverticulum is a rare form of pulsion diverticulum which originate through a muscular gap in the anterolateral wall of the esophagus, inferior to the cricopharyngeus muscle and superior to the circular and longitudinal muscle of the esophagus. Killian-Jamieson defined this area where the recurrent laryngeal nerve enters the pharynx, called Killian-Jamieson triangle. Ekberg and Nylander, in 1983, described an esophageal diverticulum in Killian-Jamieson space. The most common clinical manifestations in such patients are dysphagia, cough, epigastric pain, recurrent respiratory tract infections, and rarely hoarseness of voice. In symptomatic patients, surgical or endoscopic resection of the diverticulum is mandatory. We report this case in line with SCARE (Surgical CAse REport) criteria. Case Report: An 82-year-old male presented to our outpatient clinic with a history of progressive dysphagia for solid food and hoarseness of voice for the last 6 months. He denied gastroesophageal reflux, cough, and shortness of breath. On examination of the neck, there was swelling on the left side but no tenderness or lymphadenopathy. Basic blood investigations, including complete blood count, liver, and renal panels, were normal. An echocardiogram showed mild impairment of left ventricle function and normally functioning mitral valve. Chest X-ray showed a pacemaker in position. Computed tomography scan of the neck showed esophageal diverticulum. Gastrogrifin contrast study showed esophageal Killian-Jamieson diverticulum (KJD). Discussion: The acquired esophageal diverticulum is categorized into three types based on its anatomical location. Zenker's and Killian-Jamieson (pulsion diverticulum) in the proximal part, traction diverticulum in the middle part, due to pulling from fibrous adhesions following the lymph node infection and epiphanic pulsion type in the distal esophagus. Although the KJD and Zenker's diverticulum (ZD) arise close to each other in the pharyngoesophageal area, they are anatomically distinct. Although ZD and KJD have the same demographic features, they are more commonly found in older men (60-80 years) and women, respectively. The incidence of ZD is 0.01-0.11%, and KJD is 0.025% of the population. Rubesin et al. reported radiographic findings in 16 KJD cases. They found the majority of them were on the left side (72%), followed by 20% on the right side and 8% bilateral. Conclusion: In conclusion, we report a rare case of dysphagia and simultaneous hoarseness of voice in an octogenarian due to KJD, who was treated with open diverticulectomy, and dysphagia resolved; he regained his voice back after 6 weeks. In our opinion, endoscopic surgery in such a patient with KJD can put recurrent laryngeal at risk of injury since an endoscopic approach operator cannot visualize and dissect away the recurrent laryngeal nerve, particularly when KJD already compresses it.

RESUMO

Pleomorphic adenoma (PA) of the parotid gland is considered one of the commonest benign salivary gland neoplasms in both adults and pediatrics. However, metastatic pleomorphic adenoma (MPA) is extremely rare. In the past three decades, multiple cases were reported in the literature of MPA, where the metastatic phase has been preceded by a local recurrence for the majority of the cases. Metastases to the lungs, liver, bone, head and neck were reported. This paper will discuss a rare case presentation of MPA that metastasised solely to the face and scalp subcutaneous tissue with no other sites of metastases in a male adult.

RESUMO

A 42-year-old male pediatrics physician was admitted with a history of acute chest pain and sudden severe dysphagia to solids and liquids. He denied any history of abdominal pain, vomiting, dyspnea, nausea weight loss. He could not even swallow saliva. The patient denied any history of drug abuse. A computed tomographic scan of the chest (CT)showed a posterior mediastinal mass inseparable from the esophagus and descending aorta. Magnetic resonant imaging (MRI)scan revealed a cystic mass full of blood inseparable from the esophagus and adherent to the aorta. The mass was resected entirely through the left thoracotomy; post-operative recovery was uneventful; dysphagia resolved as the post-operative contrast swallow study showed a free flow of contrast to the stomach patient resumed his regular diet.

RESUMO

Osteosarcoma is a kind of bone cancer which generally starts to develop in the lengthy bones in the legs and arms. Because of an increase in occurrence of cancer and patient-specific treatment options, the detection and classification of cancer becomes a difficult process. The manual recognition of osteosarcoma necessitates expert knowledge and is time consuming. An earlier identification of osteosarcoma can reduce the death rate. With the development of new technologies, automated detection models can be exploited for medical image classification, thereby decreasing the expert's reliance and resulting in timely identification. In recent times, an amount of Computer-Aided Detection (CAD) systems are available in the literature for the segmentation and detection of osteosarcoma using medicinal images. In this view, this research work develops a wind driven optimization with deep transfer learning enabled osteosarcoma detection and classification (WDODTL-ODC) method. The presented WDODTL-ODC model intends to determine the presence of osteosarcoma in the biomedical images. To accomplish this, the osteosarcoma model involves Gaussian filtering (GF) based on pre-processing and contrast enhancement techniques. In addition, deep transfer learning using a SqueezNet model is utilized as a featured extractor. At last, the Wind Driven Optimization (WDO) algorithm with a deep-stacked sparse auto-encoder (DSSAE) is employed for the classification process. The simulation outcome demonstrated that the WDODTL-ODC technique outperformed the existing models in the detection of osteosarcoma on biomedical images.

RESUMO

A 46 years old male smoker was admitted to our hospital with a three-month history of chest discomfort and burning sensations due to regurgitation of food. The gastroenterologist tried multiple attempts to pass the endoscope through the lower end of the esophagus but failed. Post endoscopy Chest -X-ray showed right hemithorax fluid collection. A 28Fr chest drain was inserted, and fluid analysis revealed chyle. A contrast computed tomographic scan of the chest (CT) revealed esophageal perforation. The patient was managed conservatively by the primary physician on TPN, Antibiotics, and keeping him nil by mouth. After two weeks of failed conservative management, they referred the patient to the thoracic surgeon. We planned two-stage surgery because the patient was critically sick, septic, and hemodynamically unstable on inotropic support.

RESUMO

L-cystine (L-cys) functionalized plasmonic silver nanomaterial (Ag NPs) was fabricated toward the selective and sensitive detection of paracetamol and cadmium. The prepared L-cys-Ag nanoparticles (NPs) were characterized by transmission electron microscopy (TEM), scanning electron microscopy (SEM), X-ray diffraction spectroscopy (XRD) and fourier transform infrared spectroscopy (FTIR) analyses. SEM imaging show that Ag NPs was decorated on the surface of L-cysteine 3D cubic nanosheet. L-cys-Ag NPs showed selective and sensitive detection towards paracetamol and cadmium. The interference study confirms that the presence of other metal ions didn't inhibit the detection of cadmium by L-cys-Ag NPs. The limit of detection of paracetamol and cadmium by L-cys-Ag NPs was calculated to be 1.2 and 2.82 nM respectively. In addition, the real sample detection of paracetamol on blood serum and urine, and cadmium on STP were performed and the recovery percentage was above 97%. Further, the real sample analysis was performed in tap and drinking water and the recovery percentage was more than 98%. The analytic logic gate on the multicolour detection of cadmium and paracetamol was performed for the semi-quantitative monitoring of paracetamol and cadmium by L-cys-Ag NPs. The developed L-cys-Ag NPs were found to be an effective tool for the monitoring of cadmium in environmental water bodies and paracetamol in blood and urine.

RESUMO

Salinity is one of the largest stresses blocking horizontal and vertical expansion in agricultural lands. Establishing salt-tolerant genotypes is a promising method to benefit from poor water quality and salinized lands. An integrated method was developed for accomplishing reliable and effective evaluation of traits stability of salt-tolerant wheat. The study aims were to estimate the genetic relationships between explanatory traits and shoot dry matter (SDM), and determine the traits stability under three salinity levels. Morphophysiological and biochemical traits were evaluated as selection criteria for SDM improvement in wheat for salinity tolerance. Three cultivars and three high-yielding doubled haploid lines (DHLs) were used. Three salt (NaCl) levels (control (washed sand), 7 and 14 dS m-1) were applied for 45 days (at the first signs of death in the sensitive genotypes). All morphophysiological traits gradually decreased as salinity levels increased, excluding the number of roots. Decreases were more visible in sensitive genotypes than in tolerant genotypes. All biochemical traits increased as salinity levels increased. Variance inflation factors (VIFs) and condition number exhibited multicollinearity for membrane stability index and polyphenol oxidase activity. After their removal, all VIFs were <10, thereby increasing path coefficient accuracy. Total chlorophyll content (CHL) and catalase (CAT) provided significant direct effects regarding genetic and phenotypic correlations for the three salinity levels and their interactions in path analysis on SDM, indicating their stability. CHL and CAT had high heritability (>0.60%) and genetic gain (>20%) and highly significant genetic correlation, co-heritability, and selection efficiencies for SDM. CHL and CAT could be used as selection criteria for salinity tolerance in wheat-breeding programs. The tolerated line (DHL21) with the check cultivar (Sakha 93) can be also recommended as novel genetic resource for improving salinity tolerance of wheat.

RESUMO

We report a case of 16 -year male who presented with nonproductive cough, chest pain, and hemoptysis. His chest -x-ray and computerized tomographic scan (CT) of the thorax with contrast enhancement revealed a large mediastinal mass mostly occupying the left hemithorax. Percutaneous CT scan-guided biopsy of the mediastinal mass was reported as an endodermal sinus tumor (EDST). Serum alpha-fetoprotein levels were markedly raised (120,000 ng/ml). After completion of chemotherapy repeat, CT scan of the thorax revealed a large residual mass. Radical resection of the tumor was carried out. Twelve-year post-surgical resection follow-up with serial serum alpha-fetoprotein (AFP) tumor marker levels and CT scan of the thorax showed no recurrence.

RESUMO

Hydrogen sulfide (H2S) and nitric oxide (NO) have been known to affect vast number of processes in plants under abiotic stresses. Also, calcium (Ca) works as a second messenger in plants, which underpins the abiotic stress-induced damage. However, the sequence of action of these signaling molecules against cadmium (Cd)-induced cellular oxidative damage remains unidentified. Therefore, we studied the synergistic actions and/or relationship of signaling molecules and Ca-dependent activation of tolerance mechanisms in Vigna radiata seedlings under Cd stress. The present study shows that exogenous Ca supplemented to Cd-stressed V. radiata seedlings reduced Cd accumulation and improved the activity of nitrate reductase, and L/D-cysteine desulfhydrase (LCD/DCD) that resulted in improved synthesis of NO and H2S content. Application of Ca also elevated the level of cysteine (Cys) by upregulating the activity of Cys-synthesizing enzymes serine acetyltransferase and O-acetylserine(thiol)lyase in Cd-stressed seedlings. Maintenance of Cys pool under Cd stress contributed to improved H2S content which together with Ca and NO improved antioxidant enzymes and components of ascorbate-glutathione (AsA-GSH) cycle. All these collectively regulated the activity of NADPH oxidase and glycolate oxidase, resulting in the inhibition of Cd-induced generation of reactive oxygen species. The elevated level of Cys also assisted the Cd-stressed seedlings in maintaining GSH pool which retained normal functioning of AsA-GSH cycle and led to enhanced content of phytochelatins coupled with reduced Cd content. The positive effect of these events manifested in an enhanced rate of photosynthesis, carbohydrate accumulation, and growth attributes of the plants. On the contrary, addition of NO scavenger cPTIO [2-(4-carboxyphenyl)-4,4,5,5-tetramethylimidazoline-1-oxyl-3-oxide], H2S scavenger HT (Hypotaurine) and Ca-chelator EGTA (Ethylene glycol-bis(b-aminoethylether)-N,N,N',N'-tetraacetic acid) again developed a condition similar to stress and positive effect of the signaling molecules was abolished. The findings of the study postulate that Ca in association with NO and H2S mitigates Cd-induced impairment and enhances the tolerance of the V. radiata plants against Cd stress. The results of the study also substantiate that Ca acts both upstream as well as downstream of NO signals whereas, H2S acts downstream of Ca and NO during Cd-stress responses of the plants.

RESUMO

Amygdalin a naturally occurring compound, predominantly in the bitter kernels of apricot, almond, apple and other members of Rosaceae family. Though, amygdalin is used as an alternative therapy to treat various types of cancer but its role in cancer pathways has rarely been explored yet. Therefore, present study was intended with the aim to investigate the alleged anti-cancerous effects of amygdalin specifically on PI3K-AKT-mTOR and Ras pathways of cancer in human body. Computational modelling and simulation techniques were used to assess the effect of amygdalin on PI3K-AKT-mTOR and Ras pathways using different level of dosage. It was observed that amygdalin had direct and substantial contribution to regulate PI3K-mTOR activities on threshold levels while the other caner pathways were effected indirectly. Consequently, amygdalin is a down-regulator of a cancer within a specified amount and contribute considerably to reduce various types of cancer in human. Furthermore, in-vitro and in-vivo analyses of amygdalin could be of helpful to authenticate its pharmacological effects.

RESUMO

Phytochemical composition, in vitro antioxidant and antiproliferative activity against HepG2 cells were studied in the kernels of apricot cultivars grown in the northern areas of Pakistan. Relatively, the kernel of Habbi cultivar/AP-12 depicted significant potential to scavenge DPPH and ABTS+ free radicals as well as oxygen radical absorbance capacity along with highest contents of total flavonoids, phenolics, chlorogenic and syringic acids on dry weight basis. The average concentration of quercetin ranged 0.072-1.343 mg/100 g, and of EGCG from 0.713 to 6.521 mg/100 g with maximum concentration in Hulappa/AP-3 and Kho Chali-Khatta 3/AP-17, respectively. Amygdalin content was highest (1145 mg/100 g) in the kernel of Balaani/AP-14. Highest inhibition of HepG2 cells was found in the kernel of Waflu Chuli/AP-9 (EC50 = 15.70 ±â€¯3.77 mg/mL). The PCA showed significant contributions of polyphenols and flavonoids towards biochemical assays, while CA revealed similarities and associations among various cultivars. Our study revealed that Habbi, Waflu Chuli, Thukdeena and Balaani kernels are rich sources of bioactive compounds and possess significant antioxidant and anticancer activity and can contribute considerably in the prevention and treatment of chronic health disorders.

RESUMO

Cedrus deodara is economically and ethnobotanically an important forest tree and is shown to be at decline in Northern areas of Pakistan in recent years mainly due to high concentration of Nitrogen in forests. Ectomycorrhizal (ECM) association forming fungi enables the forest trees to develop optimally by absorbing water from the rhizosphere through their absorptive hyphae and by making available the nutrients by mobilization of N and P from the organic substrates. This study was conducted to identify the ECM strains from C. deodara rhizosphere and to analyse the impact of high N load on the C. deodara seedlings to establish N critical load value for coniferous forests of Pakistan. Six new fungal strains were identified from the rhizosphere of C. deodara and were registered at GenBank (NCBI) as Emmia latemarginata strain ACE1, Aspergillus terreus strain ACE2, Purpureocillium lilacinum strain ACE3, Talaromyces pinophilus strain ACE4, A. fumigatus strain ACE5 and T. pinophilus strain ACE6 with accession numbers MH145426, MH145427, MH145428, MH145429, MH145430 and MH547115. Four out of six isolated strains were inoculated with seedlings of C. deodara singly and in consortium (CN) in combination with nitrogen load of 0 (C), 25 (T1), 50 (T2), 100 kg N ha-1 yr-1 (T3). Agronomic, physiological and gene expression studies for ExpansinA4 (EXPA4) and Cystatins (Cys) were made to analyse the impact of fungal strains in relation to high N stress. This study suggests a positive impact of T1 (25 kg N ha-1 yr-1) Nitrogen load and a negative impact of T3 (100 kg N ha-1 yr-1) on growth parameters and expression patterns of EXPA4 and Cys genes. Peroxidase (POX) activity decreased in the order ACE5 > ACE2 > C > ACE3 > ACE1 > CN. However, the results of Superoxide dismutase (SOD) showed decreasing trend in the order ACE5 > C > CN > ACE1 > ACE2 > ACE3. Strain ACE3 was shown to have a positive impact on the seedlings in terms of growth, physiology and expression of genes. Present study suggests that newly identified fungal strains showing positive impact on the growth and physiology of C. deodara could be used for the propagation of this economically important plant in Pakistan after pathogenicity test.

RESUMO

Despite numerous reports on the role of nitric oxide (NO) in regulating plants growth and mitigating different environmental stresses, its participation in sulfur (S) -metabolism remains largely unknown. Therefore, we studied the role of NO in S acquisition and S-assimilation in tomato seedlings under low S-stress conditions by supplying NO to the leaves of S-sufficient and S-deficient seedlings. S-starved plants exhibited a substantial decreased in plant growth attributes, photosynthetic pigment chlorophyll (Chl) and other photosynthetic parameters, and activity of enzymes involved in Chl biosynthesis (δ-aminolevulinic acid dehydratase), and photosynthetic processes (carbonic anhydrase and RuBisco). Also, S-deficiency enhanced reactive oxygen species (ROS) (superoxide and hydrogen peroxide) and lipid peroxidation (malondialdehyde) levels in tomato seedlings. Contrarily, foliar supplementation of NO to S-deficient seedlings resulted in considerably reduced ROS formation in leaves and roots, which alleviated low S-stress-induced lipid peroxidation. However, exogenous NO enhanced proline accumulation by increasing proline metabolizing enzyme (Δ1-pyrroline-5-carboxylate synthetase) activity and also increased NO, hydrogen sulfide (a gasotransmitter small signaling molecule) and S uptake, and content of S-containing compounds (cysteine and reduced glutathione). Under S-limited conditions, NO improved S utilization efficiency of plants by upregulating the activity of S-assimilating enzymes (ATP sulfurylase, adenosine 5-phosphosulfate reductase, sulfide reductase and O-acetylserine (thiol) lyase). Under S-deprived conditions, improved S-assimilation of seedlings receiving NO resulted in improved redox homeostasis and ascorbate content through increased NO and S uptake. Application of 2-(4-carboxyphenyl)-4,4,5,5-tetramethylimidazoline-1-oxy l-3-oxide (an NO scavenger) invalidated the effect of NO and again caused low S-stress-induced oxidative damage, confirming the beneficial role of NO in seedlings under S-deprived conditions. Thus, exogenous NO enhanced the tolerance of tomato seedlings to limit S-triggered oxidative stress and improved photosynthetic performance and S assimilation.

RESUMO

AIM: To stratify the malignancy risks in thyroid nodules in a tertiary care referral center using the Bethesda system. METHODS: From January, 2012 to December, 2014, a retrospective analysis was performed among 1188 patients (15-90 years) who had 1433 thyroid nodules and fine-needle aspiration at Prince Sultan Military Medical City, Saudi Arabia. All thyroid cyto-pathological slides and ultra sound reports were reviewed and classified according to the Bethesda System for Reporting Thyroid Cytopathology. Age, gender, cytological features and histological types of the thyroid cancer were collected from patients' medical chart and cytopathology reports. RESULTS: There were 124 total cases of malignancy on resection, giving an overall surgical yield malignancy of 33.6%. Majority of the thyroid cancer nodules (n = 57, 46%) in Bethesda VI category followed by Bethesda IV (n = 25, 20.2%). Almost 40% of the cancer nodules in 31-45 age group in both sex. Papillary thyroid carcinoma (PTC) was the most common form of thyroid cancer among the study population (111, 89.6%) followed by 8.9% of follicular thyroid carcinoma (FTC), 0.8% of medullary carcinoma and 0.8% of anaplastic carcinoma. Among the Bethesda IV category 68% thyroid nodules were PTC and 32% FTC. CONCLUSION: The malignancy values reported in our research were constant and comparable with the results of other published data with respect to the risk of malignancy. Patients with follicular neoplasm/suspicious for follicular neoplasm and suspicious of malignancy categories, total thyroidectomy is indicted because of the substantial risk of malignancy.

RESUMO

INTRODUCTION: Paragangliomas are rare tumors arising from neural crest tissue located outside the adrenal gland. Primary seminal vesicle paraganglioma is extremely rare entity. PRESENTATION OF CASE: A 26-year-old male patient presented with symptoms and signs of acute appendicitis where a CT of abdomen and pelvis showed an inflamed appendix and incidental finding of left seminal vesicle mass. The patient underwent uneventful laparoscopic appendectomy followed by transrectal ultrasound (TRUS) guided seminal vesicle biopsies. Histopathology revealed a neuroendocrine neoplasm consistent with paraganglioma. Surgical excision of the left seminal vesicle was carried out. DISCUSSION: Paraganglioma of genitourinary tract is rare. The urinary bladder is the most common site, followed by the urethra, pelvis and ureter. Seminal vesicle paragangliomas were reported in association with other genitourinary organ involvement such as bladder and prostate. Isolated seminal vesicle paraganglioma is extremely rare and surgical excision remains the standard treatment for localized paraganglioma. CONCLUSION: Primary tumors of seminal vesicle are rare and represent a diagnostic challenge. Differential diagnosis includes a list of benign and malignant tumors. Primary seminal vesicle paraganglioma is a rare but important diagnosis to be included in the differential diagnosis.
