RESUMO

Pollen tubes are tip-growing cells that create safe routes to convey sperm cells to the embryo sac for double fertilization. Recent studies have purified and biochemically characterized detergent-insoluble membranes from tobacco pollen tubes. These microdomains, called lipid rafts, are rich in sterols and sphingolipids and are involved in cell polarization in organisms evolutionarily distant, such as fungi and mammals. The presence of actin in tobacco pollen tube detergent-insoluble membranes and the preferential distribution of these domains on the apical plasma membrane encouraged us to formulate the intriguing hypothesis that sterols and sphingolipids could be a "trait d'union" between actin dynamics and polarized secretion at the tip. To unravel the role of sterols and sphingolipids in tobacco pollen tube growth, we used squalestatin and myriocin, inhibitors of sterol and sphingolipid biosynthesis, respectively, to determine whether lipid modifications affect actin fringe morphology and dynamics, leading to changes in clear zone organization and cell wall deposition, thus suggesting a role played by these lipids in successful fertilization.

Recent studies have documented the presence of microplastics (MPs) in remote areas, including soils or sediments collected in mountain and glacier environments, but information on their presence in snow is scant. The present study aimed at exploring the presence of MPs in residual snow collected in four locations of the Aosta Valley (Western Italian Alps), with different accessibility and human presence. Overall, the µ-FTIR analyses confirmed the presence of 18 MPs in snow, 7 (39%) items were fibres, while 11 (61%) were fragments. Polyethylene (PE; 7 MPs) was the main polymer, followed by polyethylene terephthalate (PET; 3 MPs), high density PE (HDPE; 3 MPs), polyester (2 MPs), while only 1 MP made by low density PE, polypropylene and polyurethane were found. The mean (± SE) concentration of MPs in snow ranged between 0.39 ± 0.39 MPs/L and 4.91 ± 2.48 MPs/L, with a mean of 2.32 ± 0.96 MPs/L for the sampling locations. The concentration of MPs did not statistically differ among locations. Our results suggest that MPs presence in high-mountain ecosystems might depend on deposition through atmospheric precipitations or local sources due to human activities. For these reasons, policies aiming at reducing plastic use and dispersal in mountain areas may be effective in preventing local MP contamination.

Contamination by plastic debris has been documented in most regions of the world, but their occurrence in high mountain areas has not been investigated to date. Here we present the first report of the occurrence and amount of microplastic in any terrestrial glacier environment. In the supraglacial debris of the Forni Glacier (Italian Alps), we observed the occurrence of (mean ±â€¯standard error) 74.4 ±â€¯28.3 items kg-1 of sediment (dry weight). This amount is within the range of variability of microplastic contamination observed in marine and coastal sediments in Europe. Most plastic items were made by polyesters, followed by polyamide, polyethylene and polypropylene. We estimated that the whole ablation area of Forni Glacier should host 131-162 million plastic items. Microplastic can be released directly into high elevation areas by human activities in the mountain or be transported by wind to high altitude. The occurrence of microplastic on Forni Glacier may be due to the gathering of debris coming from the large accumulation area into the relatively smaller ablation area of the glacier, as a consequence of its flow and melting.

OBJECTIVE: Autonomic nervous system is involved at the onset of Parkinson disease (PD), and alpha-synuclein (α-Syn) and its phosphorylated form (p-αSyn) have been detected in dermal autonomic nerve fibers of PD. We assessed disease specific conformation variant of α-Syn immunoreactivity in cutaneous nerves and characterized skin denervation patterns in PD and atypical parkinsonism (AP). METHODS: We enrolled 49 subjects, 19 with PD, 17 age-matched healthy controls, and 13 with AP. The manifestations of disease were rated on clinical scales. Skin biopsies from ankle, thigh, and neck were analyzed by immunofluorescence for p-αSyn, 5G4 as a conformation specific antibody to pathogenic α-Syn and PGP9.5 as axonal marker. Intraepidermal nerve fiber density was measured in all anatomical sites as marker of neurodegeneration. Thirteen of the 19 PD underwent a 1 year follow-up visit plus skin biopsies. RESULTS: PD subjects displayed more severe cervical skin denervation (P < 0.03), which correlated to disease duration and worsened between initial and follow-up examination (P < 0.001). p-αSyn and 5G4 were equally sensitive and specific for the diagnosis of PD (area under the ROC was 0.839 for p-αSyn and 0.886 for 5G4). PD and AP with possible alpha-synucleinopathies share the features of marked cervical denervation and the presence of 5G4. In contrast AP with possible tauopathies were normal. INTERPRETATION: Conformational specific forms of α-Syn are detectable in skin biopsy by immunofluorescence in PD, with a promising diagnostic efficiency similar to p-αSyn. Cervical cutaneous denervation correlates with disease duration and increases over time standing out as a potential biomarker of PD progression.

Migratory behaviour is controlled by endogenous circannual rhythms that are synchronized by external cues, such as photoperiod. Investigations on the genetic basis of circannual rhythmicity in vertebrates have highlighted that variation at candidate 'circadian clock' genes may play a major role in regulating photoperiodic responses and timing of life cycle events, such as reproduction and migration. In this comparative study of 23 trans-Saharan migratory bird species, we investigated the relationships between species-level genetic variation at two candidate genes, Clock and Adcyap1, and species' traits related to migration and geographic distribution, including timing of spring migration across the Mediterranean Sea, migration distance and breeding latitude. Consistently with previous evidence showing latitudinal clines in 'circadian clock' genotype frequencies, Clock allele size increased with breeding latitude across species. However, early- and late-migrating species had similar Clock allele size. Species migrating over longer distances, showing delayed spring migration and smaller phenotypic variance in spring migration timing, had significantly reduced Clock (but not Adcyap1) gene diversity. Phylogenetic confirmatory path analysis suggested that migration date and distance were the most important variables directly affecting Clock gene diversity. Hence, our study supports the hypothesis that Clock allele size increases poleward as a consequence of adaptation to the photoperiodic regime of the breeding areas. Moreover, we show that long-distance migration is associated with lower Clock diversity, coherently with strong stabilizing selection acting on timing of life cycle events in long-distance migratory species, likely resulting from the time constraints imposed by late spring migration.
