RESUMO

Food adulteration is a global concern, drawing attention from safety authorities due to its potential health risks. Detecting and categorizing oil adulteration is crucial for consumer safety and food industry integrity. This research explores hyperspectral imaging (HSI) analysis to identify substandard oil adulteration at different stages. Using the non-destructive HSI Specim Fx 10 system, a method for precise and easy imaging-based fraud detection and classification was proposed. The 670 oil samples, including pure (Almond, Mustard, Coconut, Olive) and adulterated (Sunflower, Castor, Liquid Paraffin), were analyzed. The Savitzky-Golay filter preprocessed the images to remove noise and smooth spectral signatures. The oils were identified using various machine learning approaches, including Support Vector Machines, Logistic Regression, Linear Discriminant Analysis, Random Forests, Decision Trees, K-Nearest Neighbors, and Naïve Bayes with Linear Discriminant Analysis excelling in identification. Performance parameters, including precision, recall, F1-score, and overall accuracy, were calculated. The proposed method achieved a validation accuracy of 100%, outperforming numerous state-of-the-art approaches. This study introduces a robust pipeline for effective oil adulteration detection, offering a significant advancement in food safety and quality control.

Induction followed by concurrent chemoradiation (CCRT) is the standard of care for locally advanced nasopharyngeal carcinoma (LANPC). This study evaluated and compared the efficacy of two regimens of neoadjuvant chemotherapy along with CCRT in LANPC. Patients with LANPC were randomly divided in Group I (receiving neoadjuvant gemcitabine and cisplatin) and Group II (receiving neoadjuvant docetaxil, cisplatin and fluorouracil). Both groups also received concurrent single agent (i.e., cisplatin) chemotherapy and radiotherapy (70Gy). Treatment response was assessed at 8 weeks after the completion of CCRT using RECIST criteria. A total of 68 LANPC patients were enrolled. Group I comprised of 32 patients, with male to female ratio of 2.2, a mean (range, median) age of 38.6±11.3 (19-58, 36) years. Group II comprised of 36 patients, with male to female ratio of 3.5, mean (range, median) age of 40.9 ±11.6 (17-63, 40) years. The complete response was higher whereas the partial response was lower in Group I as compared to Group II (23/32 versus 16/36 and 06/32 versus 18/36, respectively). LANPC patients receiving gemcitabine plus cisplatin based neoadjuvant chemotherapy showed higher response, as compared with docetaxil, cisplatin and fluorouracil based neoadjuvant chemotherapy.

Cationic dyes present in industrial effluents significantly reduce the effectiveness of remediation operations. Considering the terrible impact of these pollutants on environment and biodiversity, investigating strategies to remove potentially harmful compounds from water is becoming an increasingly intriguing issue. In this work, we employed a simple hydrothermal technique to synthesize Fe-doped CdO (2, 4, and 6 wt %) nanostructures and assessed their efficacy in degrading methylene blue (MB) dye and inhibiting the growth of Staphylococcus aureus and Escherichia coli, respectively. Structural, morphological, and optical characterization of produced nanomaterials was also performed using X-ray diffraction, TEM, and UV absorption spectra. The photocatalytic decomposition of MB was significantly enhanced (58.8%) by using Fe (6 wt %)-doped CdO catalysts for 80 min under irradiation. In addition, 2.05-5.05 mm inhibitory zones were seen against Gram-positive bacteria (S. aureus), whereas the range for Gram-negative bacteria (E. coli) was 1.65-2.75 mm. These nanostructures were shown to be very effective inhibitors of beta-lactamase, d-alanine-d-alanine ligase B, and fatty acid synthase inhibitor by in silico molecular docking investigations.

BACKGROUND: Oxidative stress and inflammation play crucial roles in macro/microvascular complications. Phenolic compounds and their derivatives show promise as therapeutic agents for diseases like cancer, metabolic disorders, and cardiovascular diseases. With their antioxidant and anti-inflammatory properties, these compounds hold potential for mitigating vascular complications and improving overall health. METHODOLOGY: This study aimed to assess the therapeutic potential of five 2-methoxy phenol derivatives (T2, T5, T6, T7, and T8) as antioxidants, anti-inflammatory agents, and vasorelaxants using in vitro, in silico, and in vivo approaches. RESULTS: Among all, T2 exhibited substantial antioxidant potential against 2, 2-diphenyl-1-picrylhydrazyl (DPPH) radicals with IC50 (27.97 µg/mL), nitric oxide (NO) radicals (IC50 = 34.36 µg/mL), hydroxyl (OH) radicals (IC50 = 34.83 µg/mL) and Iron chelation (IC50 = 24.32 µg/mL). Molecular docking analysis confirms that all derivatives, particularly T2, exhibit favorable binding energies with the target proteins, ACE (-7.7 Kcal/mol), ECE-1 (-7.9 Kcal/mol), and COX-1 (-7.8 Kcal/mol). All of the compounds demonstrated satisfactory physicochemical and pharmacokinetic characteristics, and showed minimal to no toxicity during in silico, in vitro, and in vivo assessments. In isolated aortic rings from Sprague Dawley rats, pre-contracted with high K+ (80 mM), T2 induced vasorelaxation in dose dependent manner and shifted calcium response curves to the right as compared to verapamil. T2 also showed substantial platelet aggregation inhibition in a dose dependent manner with IC50 21.29 µM. All derivatives except T7 exhibited significant conservation of endogenous antioxidants i.e. catalase (CAT), peroxidase (POD), superoxide dismutase (SOD) and reduced glutathione (GSH) and significantly suppressed serum levels of inflammatory markers i.e. nitric oxide (NO), peroxides (TBARS), interleukin-6 (IL-6) and cyclooxygenase-2 (COX-2). CONCLUSION: The study concludes that T2 has significant antioxidant potential and vasorelaxant effects with adequate pharmacokinetics, making it a promising lead compound for further molecular investigation in cardiovascular disorders.

Gastrointestinal (GI) intestinal stromal tumors account for 60% of mesenchymal GI tract tumors commonly located in the stomach and small intestine, predominantly solid tumors that rarely undergo cystic degeneration. A 65-year-old patient with increasing upper abdominal swelling and a computed tomography scan abdomen showed a large unilocular 17 × 16 × 15 cm lesion. A colossal cystic swelling in the lesser omentum, anterior to the stomach, was found upon exploration. Histopathological examination showed a spindle cell tumor turned out to be CD117 positive and S100 negative on immunostains. The tumor was moderate risk gastric gastrointestinal intestinal stromal tumor (GIST) based on the site; Stomach, Size >10 cm; Mitosis <5/5 mm2 according to risk assessment of GIST, 2006. GISTs are predominantly solid tumors and rarely undergo cystic transformation. The primary differential diagnoses of spindle cell neoplasm are GISTs, Leiomyoma, Leiomyosarcoma and Schwannoma. These spindle cell neoplasms are differentiated by applying a panel of Immunohistochemical stains, CD117, SMA and S100.

Oryza sativa is grown worldwide and exhibit sensitivity to different stresses. Exponential increase in microplastics in agroecosystems is damaging and demand pragmatic strategies to protect growth and yield losses. We evaluated exogenous application of different doses of glutathione (GSH) for mediation of physiological traits of rice plants experiencing two different MPs i.e. PET and HDPE in root zone. All the rice seedlings exhibited MP-induced significant (P ≤ 0.001) growth inhibition compared to the control plants. GSH application (T3) significantly increased the shoot fresh weight (8.80%), root fresh weight (19.22%), shoot dry weight (13.705%), root dry weight (25.52%), plant height (5.75%) and 100-grain weight (9.36%), compared to control plants. GSH treated plants (T4) showed a recovery mechanism by managing the marginal rate of reduction of all photosynthetic and gas exchange attributes by showing 34.44, 20.98, 34.83, 42.16, 39.70, and 51.38% reduction for Chl-a, Chl-b, total cholophyll, photosynthetic rate (A), transpiration rate (E), and stomatal conductance (Gs), respectively, compared to control plants. Under 5 mg L-1 HDPE and PET, rice seedlings without GSH treatment responded in terms of increase in total soluble sugar, total free amino acid, glutathione, glutathione disulfide contents, while total soluble protein and ascorbic acid contents decreased significantly, compared with control plants. Corrleation matrix revealed positive relationship between GSH and improvement in studied attributes. Moreover, exogenous GSH improved rice growth and productivity to counter the negative role of MPs. This unique study examined the effects of GSH on rice plants growing in MP-contaminated media and revealed how exogenous GSH helps plants survive MP-stress.

Soil heavy metal contamination is increasing rapidly due to increased anthropogenic activities. Lead (Pb) is a well-known human carcinogen causing toxic effects on humans and the environment. Its accumulation in food crops is a serious hazard to food security. Developing environment-friendly and cost-efficient techniques is necessary for Pb immobilization in the soil. A pot experiment was executed to determine the role of biochar (BC), zero-valent iron nanoparticles (n-ZVI), and zero-valent iron nanoparticles biochar composite (n-ZVI-BC) in controlling the Pb mobility and bioaccumulation in wheat (Triticum aestivum L.). The results showed that BC and n-ZVI significantly enhanced the wheat growth by increasing their photosynthetic and enzymatic activities. Among all the applied treatments, the maximum significant (p ≤ 0.05) improvement in wheat biomass was with the n-ZVI-BC application (T3). Compared to the control, the biomass of wheat roots, shoots & grains increased by 92.5, 58.8, and 49.1%, respectively. Moreover, the soil addition of T3 amendment minimized the Pb distribution in wheat roots, shoots, and grains by 33.8, 26.8, and 16.2%, respectively. The outcomes of this experiment showed that in comparison to control treatment plants, soil amendment with n-ZVI-BC (T3) increased the catalase (CAT), superoxide dismutase (SOD) activity by 49.8 and 31.1%, respectively, ultimately declining electrolyte leakage (EL), malondialdehyde (MDA) and hydrogen peroxide (H2O2) content in wheat by 38.7, 33.3, and 38%respectively. In addition, applied amendments declined the Pb mobility in the soil by increasing the residual Pb fractions. Soil amendment with n-ZVI-BC also increased the soil catalase (CAT), urease (UR), and acid phosphatase (ACP) activities by 68, 59, and 74%, respectively. Our research results provided valuable insight for the remediation of Pb toxicity in wheat. Hence, we can infer from our findings that n-ZVI-BC can be considered a propitious, environment friendly and affordable technique for mitigating Pb toxicity in wheat crop and reclamation of Pb polluted soils.

Parthenium hysterophorus L. is a vigorous plant species with cosmopolitan distribution. It can uptake considerable quantities of heavy metals from the soil and accrue these metals in its different tissue. The use of chelating agent i.e., Ethylenediaminetetraacetic acid (EDTA) can boost up metal uptake capacity. Pot experiment was performed to evaluate phytoextraction potential of P. hysterophorus for lead (Pb) and cadmium (Cd) with and without the aid of EDTA chelator. Shoot length, weight of root and shoot (both fresh and dry), leaves number, and chlorophyll contents of P. hysterophorus got reduced with an increase in metal uptake. The results revealed the highest concentration of Cd in shoot without and with EDTA was 283.6 and 300.1 mg kg-1, correspondingly. Increase in Pb concentration was also boosted up by the EDTA from its maximum concentration in shoot 4.30-9.56 mg kg-1. Generally, Pb and Cd concentrations were greater in shoots of P. hysterophorus than the roots regardless of EDTA in the treatments. EDTA also impacted positively the accumulation of essential ions K+, Na+, and Ca+2 in P. hysterophorus. The capacity of P. hysterophorus to accumulate Pb and Cd found to be increased with EDTA in the soil. Bringing metals level in the soil in accordance to the WHO standards can improve the ecosystem as well as public health.

Melanoma is one of the most common skin infections, has triggered significant morbidity and mortality across the globe. Previous studies have reported that mutations in CDKN2A signalling network is associated with cutaneous malignant melanoma. In the present study, initially, the BioGrid database was utilized, and then hierarchical clustering was performed to identify the CDKN2A signature pathways. In addition, a GO Enrichment analysis was investigated using DAVID (n=187 genes) toolkit. Subsequently, the cBioPortal cancer genomic platform was exploited using alteration ranked frequency to determine the role of the CDKN2A signaling network in 363 samples of cutaneous malignant melanoma patients and we find that CDKN2A and its close interactors PTEN and HUWE1 show highest mutations. Further, we systematically employed molecular docking approach via MOE to target PTEN, CDKN2A and HUWE1 with chloroquine which is naturally occurring in medicinal plant Nigella sativa (NS) and observed virtuous interactions between all receptors and ligand molecules with a binding energy of -11.379, -10.324 and -9.06 Kcal/mol, respectively. The outcomes obtained stipulate a vigorous research resource for using chloroquine as a multitargeted anticancer drug. This novel evidence should help the development of effective therapeutic compounds for the treatment of cancer. Our results reveal that chloroquine is a relevant and novel potential therapeutic drug for the treatment of melanoma.

This study was aimed to develop novel in situ forming gels based on N-vinylcaprolactam, sodium alginate, and N,N-methylenebisacrylamide. The in situ Poly (NVRCL-g-NaAlg) gels were developed using the cold and free radical polymerization method. The structure formation, thermal stability, and porous nature of gels was confirmed by FTIR, NMR, DSC, TGA, and SEM. The tunable gelation temperature was evaluated by tube titling and rheological analysis. Optical transmittance showed that all formulations demonstrated phase transition around 33 °C. The swelling and release profile showed that gels offered maximum swelling and controlled 5-FU release at 25 °C and pH (7.4), owing to a relaxed state. Porosity and mesh size showed an effect on swelling and drug release. The in vitro degradation profile demonstrated a controlled degradation rate. An MTT assay confirmed that formulations are safe tested against Vero cells. In vitro cytotoxicity showed that 5-FU loaded gels have controlled cytotoxic potential against HeLa and MCF-7 cells (IC50 = 39.91 µg/mL and 46.82 µg/mL) compared to free 5-FU (IC50 = 50.52 µg/mL and 53.58 µg/mL). Histopathological study demonstrated no harmful effects of gels on major organs. The in vivo bioavailability in rabbits showed a controlled release in gel form (Cmax, 1433.59 ± 45.09 ng/mL) compared to a free drug (Cmax, 2263.31 ± 13.36 ng/mL) after the subcutaneous injection.

OBJECTIVE: To examine the association of perception of osteoarthritis with adverse pain anxiety, symptoms of depression, positive and negative affects in patients with knee osteoarthritis, and to identify the prevalence of these parameters in male and female patients. METHOD: The cross-sectional study was conducted at the Department of Orthopaedic Surgery, Pakistan Institute of Medical Sciences, Islamabad, Pakistan, from September 2017 to December 2018, and comprised osteoarthritis patients, aged 30-60 years. Psychiatric evaluation consisted of a semi-structured interview based on the Present Status Examination, and assessment was done using Beck Depression Inventory, Positive and Negative Affect Scale, Osteoarthritis Scale, and the Pain Anxiety Scale. Radiographic examination was done to evaluate knee osteoarthritis. Data was analysed using SPSS 20. RESULTS: Of the 250 subjects 144(57.6%) were males and 106(42.4%) were females. The overall mean age was 55.09+/-10.60 years. The patients had a strong inclination to enhance psychiatric illnesses (p<0.05), and gender differences were significant in this regard (p<0.05). CONCLUSIONS: Osteoarthritis patients revealed high prevalence of pain anxiety, depression and mood swings. Male patients were predisposed more towards psychiatric features compared to females.

Six ecotypes of Typha domingensis Pers. Jahlar (E1), Sheikhupura (E2), Sahianwala (E3), Gatwala (E4), Treemu (E5) and Knotti (E6) from different ecological regions were collected to evaluate the leaf anatomical and biochemical attributes under different levels of salinity and nickel stress viz; L0 (control), L1 (100 mM + 50 mg kg-1), L2 (200 mM + 100 mg kg-1) and L3 (300 mM + 150 mg kg-1). Presence of salt and Ni in rooting medium consistently affected growth, anatomical and physio-biochemical attributes in all Typha ecotypes. Discrete anatomical modifications among ecotypes such as reduced leaf thickness, increased parenchyma area, metaxylem cell area, aerenchyma formation and improved metaxylem vessels were recorded with increasing dose of salt and Ni. The minimum anatomical damages were recorded in E1 and E6 ecotypes. In all ecotypes, progressive perturbations in ionic homeostasis (Na+, K+, Cl-, N) due to salt and metal toxicity were evident along with reduction in photosynthetic pigments. Maximum enhancement in Catalase (CAT), Superoxide dismutase (SOD), Peroxidase (POD) and modulated Malondialdehyde (MDA) activity was recorded in E1 and E6 as compared to other ecotypes. Accumulation of large amounts of metabolites such as total soluble sugars, total free amino acids content in Jahlar, Knotti, Treemu and Sahianawala ecotypes under different levels of salt and Ni prevented cellular damages in T. domingensis Pers. The correlation analysis exhibited a close relationship among different levels of salinity and Ni with various plant attributes. PCA-Biplot verified our correlational analysis among various attributes of Typha ecotypes. An obvious separation of Typha characters in response to different salinity and Ni levels was exhibited by PC1. We recommend that genetic potential of T. domingensis Pers. To grow under salt and Ni stresses must be investigated and used for phytoremediation and reclamation of contaminated soil.

Contamination of soils with heavy metals (HMs) caused serious problems because plants tend to absorb HMs from the soil. In view of HM hazards to plants as well as agro-ecosystems, we executed this study to assess metal toxicity to mung bean (Vigna radiata) plants cultivated in soil with six treatment levels of cadmium (Cd) and nickel (Ni) and to find metal tolerant variety, i.e., M-93 (V1) and M-1(V2) with multifarious plant biochemical and physiological attributes. Increasing doses of Cd and Ni inhibited plant growth and photosynthesis and both varieties showed highly significant differences in the morpho-physiological attributes. V2 showed sensitivity to Cd and Ni treatments alone or in combination. Tolerance indices for attributes presented a declined growth of Vigna plants under HM stress accompanied by highly significant suppression in gas exchange characteristics. Of single element applications, the adverse effects on mung bean were more pronounced in Cd treatments. V1 showed much reduction in photosynthesis attributes except sub-stomatal CO2 concentration in all treatments compared to V2. The yield attributes, i.e., seed yield/plant and 100-seed weight, were progressively reduced in T5 for both varieties. In combination, we have observed increased mobility of Cd and Ni in both varieties. The results showed that water use efficiency (WUE) generally increased in all the treatments for both varieties compared to control. V2 exhibited less soluble sugars and free amino acids compared to V1 in all the treatments. Similarly, we recorded an enhanced total free amino acid contents in both varieties among all the metal treatments against control plants. We conclude that combinatorial treatment proved much lethal for Vigna plants, but V1 performed better than V2 in counteracting the adverse effects of Cd and Ni.

The aim of the study was to evaluate the radiation levels, radiological doses and excess lifetime cancer risk possessed by the urban soils that were collected from the vicinity of the exclusive mining and excavation centers of Dera Ghazi Khan. The high purity germanium detector was utilized for assessment of naturally occurring radionuclides (NORMs) in soil and results showed that the average activity concentrations of 226Ra, 232Th, and 40K (37 Bq/kg, 43.07 Bq/kg, 737 Bq/kg respectively) surpassed the world's average documented values (35 Bq/kg, 30 Bq/kg, and 400 Bq/kg respectively). Moreover, the average values of radiological hazards assessment like radium equivalent, internal and external hazard indices, absorbed dose rate, annual gonadal dose equivalent and excess lifetime cancer risk were 155.70 (Bq/kg), 0.4, 0.5, 73.96 (nGy/h) 90.73 (µSv/y), 476.24 (µSv/y) and 0.31(10-3) respectively. The data acquired was analyzed using descriptive statistics, cluster analysis and principal component analysis. ArcGIS (10.5) software was utilized for developing maps of radionuclide's concentration for the study area. Results of the study may serve as an important baseline radiometric data for future epidemiological studies and monitoring initiatives in the study area.
