RESUMO

Spontaneous odontogenic tumors are neoplasms characterized by a mixed odontogenic ectomesenchymal and odontogenic epithelial origin; they are rare in both humans and animals. A 3-year-old male Alpine Chamois (Rupicapra rupicapra) was found dead in north-west Italy, and was referred for the necropsy to the Department of Veterinary Sciences of the University of Turin (Italy). At the external examination a 10 × 8 cm, exophytic, red-pink, smooth, firm and ulcerated mass was observed on the inferior lip. Histologically the tumor was characterized by spindle shaped cells arranged in bundles in an abundant hyaline matrix. Multifocal and rare chords of odontogenic epithelium mixed with rare melanocytes that penetrate the neoplasia were visible. Immunohistochemistry showed a clear cytokeratin positivity of epithelial clusters. Macroscopical, histological and immunohistochemical findings were consistent with a diagnosis of locally infiltrative ameloblastic fibroma. To our best knowledge, this is the first report of this tumor in a wild ungulate and in Alpine Chamois.

A wild Beech Marten (Martes foina), was referred for necropsy to the Department of Animal Pathology of the University of Turin (Italy). At gross examination, whitish and firm masses, 10-mm in diameter, were found on the heart and in the kidney. Spleen showed lighter color and greater consistency, and the cut surface of the liver appeared scattered with whitish-yellow coalescing foci homogeneously distributed. Amyloid deposits were present in the perivascular and intercellular spaces of the visceral organs, such as the heart, liver, and kidneys. Amyloid stained positively with Congo red with and without 5% potassium permanganate pretreatment and showed green birefringence observable under polarized light. A diagnosis of systemic AL amyloidosis was made. This is the first description of systemic AL amyloidosis in a wild Stone Marten.

RESUMO

We describe a diffuse, multicentric, large B-cell lymphoma in a wild boar (Sus scrofa) involving the abdomen, head, and nose and invading the frontal leptomeninges. The tumor was predominantly composed of dense, basophilic, round-to-polygonal cells. Immunohistochemistry for CD79 and Ki-67 was positive in all masses.

An 18-year-old female lion (Panthera leo) was referred to the Department of Animal Pathology of the University of Turin (Italy). At necropsy, multiple nodular, 4-20-mm, confluent white firm nodules were scattered throughout the pleural surfaces of the thoracic wall and of the lungs. Histological lesions were represented by proliferations of papillary structures lined by cuboidal basophilic mesothelial cells with large, oval nuclei and abundant granular eosinophilic cytoplasm. Immunohistochemistry revealed immunoreactivity for pancytokeratin and vimentin. None of the cells expressed calretinin antigen. Asbestos fibers and asbestos bodies were not detected respectively by light microscopy and by Scanning Electron Microscope-Energy Dispersive Spectrometer investigations. On the contrary, chrysotile asbestos were identified in samples from shelter material. Histological and immunohistochemical findings were consistent with the diagnosis of an epithelial malignant mesothelioma. To our best knowledge, this is the first report of a pleural mesothelioma in a lion.

Small ruminant lentivirus genotype E lacks the dUTPase subunit and vpr-like gene. Two strains (Roccaverano and Seui) with identical genetic organization have been described, with the env HV1-HV2 domains being the most divergent. Although dUTPase and vpr-like deletions have been involved in the RT fidelity in non dividing cells, both strains were able to replicate efficiently in blood derived macrophages (BDM), while virus production of E1 subtype was reduced or abrogated in replicating fibroblastic-like cells. The transcriptional activity of genotype E was similar in these two cellular populations. When viral pseudotypes were generated with the env of both viruses, Roccaverano pseudotype displayed a paranuclear localization on BDM, suggesting a different mechanism of entry. Polymorphic GAS and TAS sites in the U3 region, further suggest that a population different from classically activated macrophages can be infected by these viruses, opening new insights into lentiviruses with low or null pathogenic potential.

The effects of 17beta-oestradiol (E2) on gene expression in cultures of bovine primary prostate stromal cells (BPSCs) and prostate gland tissue were studied. In the first part of the study, BPSCs were grown in the presence of E2 from the first passage to the end of the experiment; a second group was treated in the same way but the treatment was suspended for 48 hours before the end of the experiment; a third group of BPSCs served as a control. In the second part of the study, five male veal calves, aged 130 days, were treated four times intramuscularly with 10 mg of E2 at intervals of two weeks and then euthanased two weeks after the last treatment. Quantitative PCR and immunohistochemistry were used to evaluate the expression of fibroblast growth factor (FGF) receptors (FGFRs), FGFs, progesterone receptor, androgen receptor and oestrogen receptor in BPSCs and prostate tissue. E2 induced a significant over-expression of progesterone receptor in both BPSCs and prostate tissue. There was also a marked up-regulation of FGFR types 1, 2 and 3 genes observed in the BPSCs.

Ninety trapped nutria (Myocastor coypus) from a protected area of Piedmont (Italy), including the Po river, were examined for the prevalence for lesions in major viscera, selected serum antibodies and enteric bacteria. Coccidial lesions in the liver included cholangitis, calcification and necrosis. Renal lesions were nonsuppurative interstitial nephritis and a single case of renal adenocarcinoma. The lungs had a 41.1% prevalence of nonsuppurative interstitial pneumonia. Ten of 87 sera (11.5%) had antibodies against Leptospira bratislava, 3 of 87 (3.4%) against Leptospira ichterohaemorrhagiae, 15 of 41 (36.6%) against Toxoplasma gondii, and antibodies against encephalomyocarditis virus were detected in 5 of 78 sera (6.4%). All fecal samples were negative for Salmonella, Shigella, and Pseudomonas, and growth of enterobacteriaceae was in the normal range.

BACKGROUND: An upregulation of the cell-cycle associated proteins p53 and p21/Waf1/Cip1 induced by mycobacteria was previously reported. We aimed to evaluate the expression of such proteins in peripheral blood human monocyte cultures infected with strains of different mycobacterial pathogens. METHODS: The study relied on the immunocytochemical determination of p53, p21/Waf1/Cipl, bcl-2 and on the Tunel detection of apoptosis in monocytes populations cultured on four-welled chamber slides (10(6) cells/well) infected with Mycobacterium tuberculosis, M. bovis and M. avium for four consecutive days (mycobacterium/monocyte ratio 10:1). The results were expressed as mean values and SD of the percentages of stainings recorded in five fields per slide. RESULTS: The statistical analysis with Fischer test demostrated that at most sampling times the p53 and p21/Waf1/Cip1 expression and the apoptosis index were significantly higher in M. tuberculosis infected cultures than in controls (p<0.05). The M. bovis related picture diverged from the previous one for a lower p53 expression (p<0.05) at all sampling times. The M. avium infected culture values did not diverge significantly from the controls. CONCLUSIONS: The p53 and p21/Wafl/Cipl upregulation is compatible with both host defense strategies and pathogen strategies (safeguard of intracellular sanctuaries). The discrepancies among different cultures suggest a direct relationship between p53 activation and mycobacterial ability to enter host cells.

Organotin compounds (OTs) are used worldwide in industry and in agricultural practice. In Italy, the OT mostly used is the fungicide triphenyltin acetate (TPTA), although in past years it has been submitted to revision for occupational and environmental grounds. In the present study, the possible effect of the fungicide on cell proliferation has been investigated, using [(3)H]thymidine incorporation radiometric assay. Mouse thymocytes in primary culture were stimulated for 24 h with T- and B-cell mitogens (concanavalin A, CON A and phytohemagglutinin, PHA or pokeweed mitogen, PWM, respectively). Cultures were then exposed 24 h to 0, 1 and 8 microM TPTA. At the end of incubation, cells were pulsed with methyl-[(3)H]thymidine and harvested for total radioactivity counts after a further 24 h of incubation. An overall dose-dependent significant (P<0.05) reduction of proliferative response was observed with all mitogens tested. Interestingly, CON A proved to be more sensitive (P<0.05) to the TPTA toxic effect compared with PWM and PHA. TPTA is cytotoxic to mouse thymocytes in primary culture, particularly towards the mature lymphocytes (CD4(+) and CD8(+)). The present results support the hypothesis of a TPTA-induced decrease of lymphoproliferative response to T- and B-cell mitogens, previously observed with other OTs.

A detailed macroscopic and histological description is given of tuberculous-like lesions in mandibular lymph nodes obtained from wild boar (Sus scrofa) collected in Italy during the 1995/1996 hunting season, as well as a correlation with results obtained from a Mycobacterium tuberculosis complex target amplified test system. According to macroscopic appearance, lesions were largely represented by caseous-necrotic-calcified granulomata (n = 272/285; 95.4%), while histological investigations showed mainly necrotic-calcified (n = 82/218; 37.6%) and fibronecrotic-calcified (n = 81/218; 37.2%) lesions. When tested with a target rRNA amplification/hybridization technique to detect M. tuberculosis complex, 112 (43.6%) samples out of 275 tested gave positive results.

With the aim of describing the effect of severe feed restriction on the liver histology, morphometrical analysis of liver sections of 10 alpine chamois (Rupicapra rupicapra) was performed. Five animals were found dead during the winter season 1995-96 and five were collected during the hunting season 1996. Hepatocyte nuclear size was measured in squared micrometers using Image-Pro Plus software. A significant decrease in the mean size of the nuclei of hepatocytes in emaciated chamois, as compared to harvested animals was observed. The reduction in cell nuclear size may be linked to the mobilization of body protein to prevent ketosis during severe food restriction, as hypothesized for other wild ungulates. The change in hepatocyte size may be the consequence of a strategy to minimize energy expenditure and may be proposed as an index of metabolic stress during winter undernutrition.

Cerebrospinal fluid (CSF) obtained from 20 clinically healthy dogs and from 15 dogs affected by neurological disorders were examined for total and differential cell counts, immunocytochemistry for canine distemper virus antigen, total protein concentration and electrophoretic separation, and glucose and enzyme determination. Dogs affected by canine distemper showed an increase in macrophages, presence of specific inclusion bodies, and an increase in total protein concentration and gamma-globulin fraction. In bacterial meningoencephalomyelitis pleocytosis, increase of total proteins with special regard to high molecular weight proteins, elevated levels of lactate dehydrogenase (LDH), alterations of LDH isoenzyme profile, and decrease in glucose levels were detected; inflammatory disorders were more often characterized by an increase in LDH level, while in non-inflammatory disorders (hydrocephalus and spinal cord neoplasia) no variation in LDH levels was detected. Analysis of CSF in dogs appears relatively easy to perform and may help in establishing the condition of the blood-brain barrier as well as in the diagnosis of neurological disorders.

A serological study was undertaken by means of indirect hemagglutination (IHA) in 57 households of eleven patients with confirmed pulmonary hydatidosis (by serology and surgical procedure). Serum samples from 40 blood donors were used as control group. The IHA positivity rates were 90.8%, 40.3% (23/57) and 2.5% (1/40) in patients with hydatidosis, households of these patients and blood donors (control group). Hydatidosis was confirmed in 4 out of 23 cases of IHA positive households. The high incidence by IHA in households living with patients with hydatid disease can be a good screening parameter to identify a high-risk asymptomatic population carrying the disease at an early phase.

Pathologic findings, lectin histochemistry, and nuclear estrogen receptors were studied in the reproductive organs of gilts treated with clenbuterol. A ration containing 1 ppm of clenbuterol was fed for 40 days to four Landrace x Large white, 9-month-old gilts, weighing 134 to 172 kg at slaughter (gilt Nos. 5-8). Four gilts (Nos. 1-4) served as controls. Treated animals had macroscopic lesions characterized by microcystic ovaries and uterine atrophy. Histopathologic lesions included atretic degeneration of many ovarian follicles, complete absence of functional corpora lutea, a reduction in the number of endometrial glands, and a decrease in cytoplasmic volume of endometrial and glandular epithelial cells. In ovaries, uterus, and vagina lectin histochemistry, performed with thirteen different biotinylated lectins, revealed a different staining distribution between control and treated gilts. The binding pattern of Ricinus communis agglutinin-I (RCA-I) and -II (RCA-II) in the ovaries of control gilts, displayed labeling of cytoplasm in theca interna cells of Graafian follicles. There was no labeling of the same cells in treated gilts. Labeling patterns with Griffonia simplicifolia agglutinin-I (GS-I), Phaseolus vulgaris agglutinin (PHA), RCA-I and RCA-II documented a difference in the vascularity of the theca interna between Graafian follicles of control and treated gilts. The GS-I and Ulex europaeus agglutinin-I (UEA-I) binding patterns in uterus and vagina of treated gilts when compared to control gilts suggested that there was a block of the cycling activity in the proliferative stage. Immunohistochemical staining for estrogen receptors in the endometrium was positive in all but one treated gilts, and negative to weakly positive in control gilts. Serum progesterone concentrations were decreased in treated animals when compared to control; estradiol concentrations were similar in both group of gilts. Cystic ovaries, uterine atrophy, and reduction in progesterone concentrations suggested that clenbuterol changed ovarian hormonal activity in treated animals.

Genital tracts from 15 cows with catarrhal and purulent inflammation of the uterus and uterine tubes, cystic hyperplasia of the endometrium, or hydrosalpinx were evaluated by the use of scanning electron microscopy to determine epithelial changes associated with these conditions. Uterine epithelium was revealed to be easily damaged, even in the course of mild inflammation, whereas epithelium of the uterine tube was more resistant.
