RESUMO

Adrenal responsiveness was tested in nonpregnant, lactating Holstein dairy cows fed diets supplemented with OmniGen-AF (OG; Phibro Animal Health Corp., Teaneck, NJ), an immune modulator, and in nonsupplemented control (CON) cows following bolus infusions of a combination of corticotropin-releasing hormone (CRH; 0.3 µg/kg of BW) and arginine vasopressin (VP; 1.0 µg/kg of BW) or ACTH (0.1 IU/kg of BW) in 2 environments: thermoneutral [TN; temperature-humidity index (THI) <60] for 24 h/d and heat stress (HS; THI >68 for 17 h/d). Cows (506) were initially fed OG (n = 254) or CON (n = 252) diets for 44 d before selection of a subgroup of cows (n = 12; 6 OG, 6 CON) for the study. The 2 subgroups were balanced for parity, milk yield, and days in milk. All cows were transported to and housed in 2 environmentally controlled rooms at the University of Arizona Agricultural Research Complex (Tucson). Cows were given 3 d to acclimate to the rooms and then underwent 12 d of TN conditions and then 8 d of HS conditions for a total of 24 d on experiment. Cows were infused with CRH-VP on d 9 of TN and on d 1 of HS and with ACTH on d 10 of TN and on d 2 of HS. Hormone infusions took place at 1000 h (0 h) on each infusion day. Blood samples, taken in 30-min intervals, were first collected at 0800 h (-2 h) and were drawn until 1800 h (8 h). Before infusion, serum progesterone was elevated in OG cows compared with CON cows. Infusion of releasing factors (CRH-VP or ACTH) caused increases in serum cortisol and progesterone, but cortisol release was greater in CON cows than in OG cows during HS, whereas progesterone did not differ between the 2 treatments. Serum ACTH increased following infusion of releasing factors, but this increase was greater following CRH-VP infusion than ACTH infusion. Serum bovine corticosteroid-binding globulin also increased following infusion of releasing factors in both treatment groups, but this increase was greater during HS in cows fed OG. The free cortisol index (FCI) increased following CRH-VP and ACTH and was higher in HS than in TN for both OG and CON cows. However, the FCI response was blunted in OG cows compared with CON cows during HS. Heat stress enhanced the adrenal response to releasing factors. Additionally, the adrenal cortisol and FCI response to releasing factors was reduced during acute heat stress in cows fed OG. Collectively, these data suggest that OG supplementation reduced the adrenal responsiveness to factors regulating cortisol secretion during acute HS.

The objectives of this study were to determine whether plane of nutrition (PON) of milk replacer previously provided to calves, and dosage level of Mannheimia haemolytica (MH), influenced inflammatory responses to a combined viral-bacterial respiratory challenge. Holstein calves (1 d of age; n = 30) were assigned to treatments in a 2 × 3 factorial with pre-weaning PON and MH dose as main effects (n = 5 per treatment). Calves were fed either a low (LPN; n = 15) or a high PON (HPN; n = 15) from birth through weaning. Calves fed LPN were fed 436 g of dry matter (DM) per day of milk replacer until weaning, and HPN calves were fed 797 g of DM per day of milk replacer from d 1 to 10 and 1,080 g of DM per day from d 11 until weaning. Calf starter and water were offered ad libitum. Calves were step-down weaned beginning at d 54 and moved into an enclosed barn at d 70. Indwelling rectal temperature (RT) recording devices and jugular catheters were inserted at d 80. Calves were challenged with 1.5 × 108 plaque-forming units (pfu) per mL of bovine herpesvirus-1 (BHV-1) in each nostril at d 81 and with either 106, 107, or 108 cfu of MH at d 84. Blood samples were collected at varying intervals relative to BHV-1 and MH challenges. Four LPN calves either died or were euthanized soon after the 144-h observation period, whereas all HPN calves survived the entire observation period. As dosage of MH administered increased, acute and systemic inflammatory responses increased. Higher doses of MH resulted in increased leukocyte, neutrophil, and haptoglobin concentrations in infected calves. Data from the current study suggest that the highest dose, 108 cfu, triggered weaned calves' acute disease response, whereas the lower doses, 106 and 107 cfu, caused more moderate inflammation and disease. The effects of PON on inflammation responses to the disease challenge indicated that calves previously fed the LPN diet had more severe pathophysiological responses. Calves fed LPN showed higher peripheral neutrophil and leukocyte counts and serum haptoglobin concentrations following the BHV-1 challenge. Additionally, following the MH challenge, LPN calves had higher peripheral neutrophil counts, neutrophil-to-lymphocyte ratios, and serum tumor necrosis factor-α concentrations. These data demonstrate that higher doses of MH increase the acute inflammatory response and prolong inflammation, and that calves previously fed LPN responded more severely to the combined viral-bacterial respiratory challenge.

This study examined the relationship of prenatal transportation stress (PNS) with exogenous GnRH-induced LH and testosterone secretion in sexually mature Brahman bulls. Brahman cows (n = 96; 48 were stressed by transportation at 5 stages of gestation and 48 were controls) produced a calf crop of 85 calves. All bulls (n = 46) from this calf crop were electroejaculated every 2 wk beginning at a scrotal circumference of 24 cm until sexual maturity (SM; i.e., 500 million sperm/ejaculate). The initial 11 control and 12 PNS bulls to reach SM were selected for the experiment. Within 7-21 d after reaching SM, bulls were fitted with jugular cannulas, from which blood samples were collected at 15-min intervals for 6 h prior to exogenous GnRH administration (10 ng/kg BW; i.v.) and for 6 h after GnRH. Serum concentrations of LH, testosterone, and cortisol were determined by RIA. Age and body weight did not differ ( > 0.1) between PNS and control bulls at the time of the experiment. All bulls responded similarly to exogenous GnRH, indicating no influence of PNS on LH or testosterone response to GnRH. More ( < 0.01) PNS (9 of 11) than control (3 of 12) bulls exhibited an endogenous pre-GnRH LH pulse, and more ( = 0.02) PNS (9 of 11) than control bulls (4 of 12) exhibited a pre-GnRH testosterone response to LH. The average concentration of testosterone during the 60 min (time -60, -45, -30, -15, and 0 min relative to GnRH) immediately preceding GnRH, tended to be greater ( = 0.07) in PNS (1.46 ± 0.30 ng/mL) than control (0.68 ± 0.28 ng/mL) bulls. During that time span serum cortisol was lower ( < 0.01) in PNS (4.00 ± 0.91 ng/mL) than control (7.8 ± 0.87 ng/mL) bulls. A treatment by time interaction ( = 0.03) affected testosterone concentrations from time -240 to 360 min relative to GnRH. Results from this study indicate that PNS did not affect pituitary responsiveness to GnRH or testicular responsiveness to GnRH-induced LH secretion.

Two experiments investigated how plane of nutrition influences performance, leukocyte responses, and resistance to an oral Salmonella enterica serotype Typhimurium challenge. In experiment 1, 46 (2±1 d of age) calves were randomly assigned to 2 diets: a low (LPN; n=23) and high plane of nutrition (HPN; n=23). The LPN calves were fed 409 g/d of dry matter (DM) of a 20% crude protein and 20% fat milk replacer, whereas HPN calves were fed 610 and 735 g/d of DM of a 28% crude protein and 25% fat milk replacer during wk 1 and 2 to 6, respectively. In experiment 2, 20 bull calves (LPN; n=11 and HPN; n=9) were orally challenged on d 80 with 1.5×10(7) cfu of Salmonella Typhimurium (ATCC #14028). The HPN calves had a greater incidence (87.5 vs. 45.5%) and duration of days with high fecal scores (5.5 vs. 3.5 d). The LPN calves had greater neutrophil surface expression of L-selectin on d 7, 21, and 42. Following the Salmonella Typhimurium challenge, calf starter DM intake was greater among the HPN calves. The percentage of neutrophils producing an oxidative burst was also greater among HPN calves on d 1 to 5 after the challenge. Similarly, the intensity of the oxidative burst tended to be greater among the HPN calves on d 2 and 3 postchallenge. The secretion of tumor necrosis factor-α from whole-blood cultures stimulated with lipopolysaccharide tended to be greater on d 1 and was greater on d 5 and 6 among HPN calves. The median ranks of haptoglobin concentrations were greater and plasma zinc concentrations tended to be decreased among LPN calves. These data indicate that feeding a HPN to Jersey calves improved average daily gain and feed efficiency, but increased the incidence of high fecal scores during the first few weeks of life; however, the HPN Jersey calves may be more resistant to Salmonella Typhimurium after weaning.

The difference in the acute phase response of a heat-tolerant and a heat-sensitive Bos taurus breed to a lipopolysaccharide (LPS) challenge when housed at different air temperatures (Ta) was studied. Angus (ANG; heat-sensitive; n = 11; 306 ± 26 kg BW) and Romosinuano (RO; heat-tolerant; n = 10; 313 ± 32 kg BW) heifers were transported from the USDA Agricultural Research Service SubTropical Agricultural Research Station in Florida to the Brody Environmental Chambers at the University of Missouri, Columbia. Heifers were housed in stanchions in 4 temperature-controlled environmental chambers. Initially, Ta in the 4 chambers was cycling at thermoneutrality (TN; 18.5°C-23.5°C) for a 1-wk adjustment period, followed by an increase in 2 of the 4 chambers to cycling heat stress (HS; 24°C-38°C) for 2 wk. On day 19, heifers were fitted with jugular catheters and rectal temperature (RT) recording devices. On day 20, heifers were challenged with LPS (0.5 µg/kg BW; 0 h), sickness behavior scores (SBSs) were recorded, and blood samples were collected at 0.5-h intervals from -2 to 8 h and again at 24 h relative to LPS challenge at 0 h. Serum was isolated and stored at -80°C until analyzed for cortisol and cytokine concentrations. A breed by Ta interaction (P < 0.001) was observed for RT such that the post-LPS average RT in RO heifers housed at TN was lower than the RT of all other treatment groups (P < 0.001), whereas ANG heifers housed at HS had greater post-LPS average RT than all other treatment groups (P < 0.001). In response to LPS, HS increased SBS after LPS in RO heifers compared to RO heifers housed at TN (P < 0.001), whereas HS decreased SBS after LPS in ANG heifers compared to ANG heifers housed at TN (P = 0.014). The cortisol response to LPS was greater in TN than in HS heifers (P < 0.01) and was also greater in RO than in ANG heifers (P = 0.03). A breed by Ta interaction (P < 0.01) was observed for tumor necrosis factor-α (TNF-α) concentration such that HS increased post-LPS serum concentrations of TNF-α in ANG heifers compared to ANG heifers housed at TN (P = 0.041), whereas HS decreased post-LPS concentrations of TNF-α in RO heifers compared to RO heifers housed at TN (P = 0.008). A tendency (P < 0.06) was observed for a breed by Ta interaction for IL-6 concentrations such that RO heifers had greater post-LPS concentrations of IL-6 than ANG heifers when housed at HS (P = 0.020). A breed by Ta interaction was observed for interferon-γ (IFN-γ; P < 0.01) concentrations such that HS decreased post-LPS concentrations of IFN-γ in ANG heifers compared to ANG heifers housed at TN (P < 0.001), and HS increased post-LPS concentrations of IFN-γ in RO heifers compared to RO heifers housed at TN (P = 0.017). These data indicate differences in the acute phase response between the heat-tolerant RO and heat-sensitive ANG heifers under different Ta which may aid in elucidating differences in productivity, disease resistance, and longevity among cattle breeds.

Thirty-three colostrum-deprived Holstein bull calves (initial BW of 131 ± 4 kg) were used to determine the effect of timing of anthelmintic administration relative to vaccination on antibody titer response to vaccine component antigens. When calves were at least 3 mo of age, they were sorted randomly into individual pens and assigned to 1 of 3 treatment groups, treatments consisted of 1) dewormed 2 wk before vaccination (DPV), 2) dewormed at the time of vaccination (DV), or 3) control, vaccinated but not dewormed (CONT). All calves were inoculated with infective larvae of brown stomach worms (Ostertagia ostertagi) and intestinal worms (Cooperia spp.) on d 1, 7, 10, 14, and 18 for a total dose of 235,710 infective larvae per calf. Calves (DPV and DV) were dewormed on d 21 or 35 with a 10% fenbendazole suspension at 5 mg/kg of BW. On d 35, all calves were vaccinated with a modified-live virus respiratory vaccine containing IBRV (infectious bovine rhinotracheitis virus), BVDV-1 (bovine viral diarrhea virus genotype 1), BVDV-2 (BVDV genotype 2), PI-3 (parainfluenza-3), and BRSV (bovine respiratory syncytial virus). During the 103-d experiment, weekly fecal egg counts, blood, and rectal temperatures were collected and health status was recorded daily. Blood samples were obtained weekly to determine serum neutralizing (SN) antibody titers to IBRV, BVDV-1, BVDV-2, and PI-3 and cytokine levels for IL-4, IL-6, TNF-α (tumor necrosis factor-α), and IFN-γ (interferon-gamma). There was a tendency (P < 0.09) for CONT calves to have greater IL-4 concentrations. By design, control calves had greater (P < 0.01) fecal egg counts during the experiment. All calves developed antibody titers to IBRV, BVDV-1, BVDV-2, and PI-3 by d 15 postvaccination. On d 88, all calves were challenged with IBRV and blood samples were obtained on d 88, 89, 90, 93, 95, 98, 99, and 103. All calves had increased rectal temperatures during the final 7 d of the IBRV challenge. However, the CONT group had greater (P < 0.01) rectal temperatures on each sampling day except d 90 compared with the DPV and DV treatments. Therefore, deworming before or at vaccination reduced parasite burden and decreased rectal temperature increase after an IBRV challenge. Deworming strategy had no effect on antibody response to vaccination or IBRV challenge.

The objective was to compare the acute-phase response of steers receiving different doses of bovine corticotropin-releasing hormone (CRH). Fourteen weaned Angus steers (BW = 191 ± 2.1 kg, age = 167 ± 4.7 d) fitted with an indwelling jugular catheter and a rectal temperature (RT) monitoring device were assigned to receive 1 of 3 treatments (intravenous infusion): 1) 0.1 µg of CRH/kg of BW (CRH1; n = 5), 2) 0.5 µg of CRH/kg of BW (CRH5; n = 5), and 3) 10 mL of saline (0.9%; n = 4). Blood samples were collected via catheters, relative to treatment infusion (0 h), hourly from -2 to 0 h and 4 to 8 h and every 30 min from 0 to 4 h. Rectal temperatures were recorded every 30 min from -2 to 8 h. Blood samples were also collected via jugular venipuncture and rectal temperatures assessed using a digital thermometer every 6 h from 12 to 72 h and every 24 h from 96 to 168 h. All plasma samples collected during the study were analyzed for concentrations of haptoglobin. All plasma samples collected from -2 to 8 h were analyzed for cortisol concentrations. Serum samples collected hourly from -2 to 8 h were analyzed for concentrations of NEFA, IL-6, tumor necrosis factor (TNF)-α, and interferon-γ. Cortisol peaked at 0.5 h for CRH1 steers but returned to baseline concentrations at 1 h relative to infusion (time effect; P < 0.01). In CRH5 steers, cortisol peaked at 0.5 h and returned to baseline concentrations 3.5 h relative to infusion (time effect; P < 0.01). Cortisol concentrations did not change after treatment infusion for saline steers (time effect; P = 0.42). In CRH1 steers, NEFA concentrations peaked 5 h after treatment infusion (time effect; P = 0.01). Conversely, serum NEFA concentrations did not change for CRH5 and saline steers after treatment infusion (time effect; P > 0.37). Mean serum TNF-α concentrations in CRH1 steers after treatment infusion were greater compared with saline (P = 0.02), tended to be greater (P = 0.08) compared with CRH5, and were similar (P = 0.40) between CRH5 and saline steers. Mean RT in CRH1 steers after treatment infusion were greater (P < 0.04) compared with saline and CRH5 and similar (P = 0.50) between CRH5 and saline steers. Haptoglobin increased and peaked 72 h after treatment infusion for CRH1 steers (time effect; P = 0.01) but did not change for CRH5 and saline steers (time effect; P > 0.45). In conclusion, the bovine acute-phase response stimulated by CRH infusion is dependent on the CRH dose and the subsequent response in circulating cortisol.

Our primary objective of this experiment was to evaluate potential genetic differences between two diverse Bos taurus breeds [Angus (AG) and Romosinuano (RO)] in response to an endotoxin challenge. Eighteen steers (n = 9 steers/breed; 299.4 ± 5.2 kg BW) were acclimated to environmentally controlled chambers maintained at thermoneutrality (19.7 °C) and then fitted with indwelling jugular catheters and rectal temperature (RT) recording devices 1 d before the endotoxin challenge. The next day, blood samples were collected at 30-min intervals from -2 to 8 h, and RT was measured continuously at 1-min intervals throughout the study. At time 0, all steers received an intravenous bolus injection of lipopolysaccharide (LPS; 2.5 µg/kg BW). Serum samples were stored at -80 °C until analyzed for cortisol, proinflammatory cytokines [tumor necrosis factor-alpha (TNF-α), interleukin-1 beta (IL-1ß), IL-6, and interferon gamma (IFN-γ)], and acute phase proteins (serum amyloid A, acid soluble protein, ceruloplasmin, and α-acid glycoprotein). Rectal temperatures increased in both breeds within 1 h after LPS, with RO producing a greater increase in RT than AG steers (P < 0.001). Serum cortisol and TNF-α increased (P < 0.01) in both breeds within 1 h after the LPS challenge. For cortisol, an overall breed effect (P < 0.02) was detected, such that AG steers had a higher cortisol response than RO steers. A breed × time interaction (P < 0.01) was observed for TNF-α, such that the response was delayed and extended in the RO steers compared with the AG steers. At 2 and 2.5 h after LPS, TNF-α concentrations were greater (P < 0.03) in RO steers than in AG steers. For IL-1ß, a breed × time interaction (P < 0.04) was also observed. At 3 h after LPS, IL-1ß concentrations were greater (P < 0.01) in RO steers than in AG steers. Serum IL-6 and IFN-γ increased (P < 0.01) in a similar manner in both groups after the LPS challenge. These data show differences in the innate immune response between two diverse Bos taurus breeds which may provide insight about differences observed in productivity, heat tolerance, disease resistance, and longevity among cattle breeds.

The objectives of this study were to compare innate immune responses of calves weaned early (EW; n=23; weaned at 23.7 ± 2.3 d of age) with those of conventionally weaned calves (CW; n=22; weaned at 44.7 ± 2.3 d of age). All calves were fed 3.8L of colostrum within 12h of birth and were subsequently fed milk replacer twice daily. The weaning process began by withdrawal of the afternoon milk-replacer feeding. Milk was fully withdrawn, and the calf was considered completely weaned when it consumed 900 g of calf starter as-fed for 2 consecutive days. Blood samples were collected from all calves at 24, 27, 31, 45, 48, 52, and 66 ± 2.3 d of age. Early weaned calves took a variable amount of time to completely wean from milk replacer; therefore, data were also analyzed by comparing calves grouped by latency to completely weaned (fast=1 to 5 d; intermediate=6 to 8 d; slow=15 to 17 d). Slow-EW calves weighed less than either the fast- or intermediate-EW calves before initiating weaning. At 27 d of age, circulating neutrophils were greater among EW calves than CW calves. Moreover, fast-EW calves had lower neutrophil:mononuclear cell ratios at 45 d of age than other EW calves. Slow-EW calves had lower TNF-α concentrations from whole blood stimulated with endotoxin at 27 and 31 d of age compared with fast- and intermediate-EW calves. All EW calves had decreased neutrophil L-selectin at d 27 and increased neutrophil L-selectin at 31 d of age. At 31 d of age, neutrophil ß(2)-integrin was the greatest among the fast-EW calves. All EW calves had decreased neutrophil oxidative burst at 27 and 31 d of age. Three days after CW calves were weaned they had higher neutrophils, hematocrit percentages, and circulating cortisol than EW calves. In addition, 3 d after CW calves were weaned they had decreased neutrophil oxidative burst responses to Escherichia coli. Weaning, irrespective of age, suppressed many innate immune responses. In addition, early weaning transiently suppressed L-selectin expression on neutrophils; however, the immunological significance in the context of the resistance to disease is unknown because EW calves likely had greater protection from passively derived immunoglobulins when they were weaned. Finally, calves with lower BW around 24 d of age may not be suitable for early weaning programs as evident in the suppressed secretion of TNF-α from whole blood cultures during the week following the initiation of weaning.

The objectives of this study were to determine the effects of switching Holstein calves to once-daily feeding during the fourth week of life (24 ± 2.3 d of age; once-fed n=22; twice-fed n=22) on innate immune responses, and to evaluate whether carry-over effects occurred when the calves were weaned during the seventh week of life. Peripheral blood samples were taken immediately before the change in feeding strategy (24 d of age) and at 27, 31, 45, 48, 52, and 66 d of age and were analyzed for circulating cortisol, haptoglobin, total leukocyte counts, neutrophil:mononuclear cells, and hematocrit percentage. Heparinized whole blood was also stimulated with lipopolysaccharide (LPS) for 24h and the concentration of tumor necrosis factor-α (TNF-α) in the supernatant was analyzed. Neutrophil L-selectin and ß(2)-integrin expression were analyzed by flow cytometry. Simultaneous neutrophil phagocytic and oxidative burst responses to a heat-killed Escherichia coli were quantified by dual-color flow-cytometry. Treatment (once-daily or twice daily feeding) had no effect on pre- or postweaning performance. Once-fed calves tended to have more circulating neutrophils at 27 d of age, greater expression of L-selectin on neutrophils at 31 and 45 d of age, and greater intensity of phagocytosis at 45 d of age. Once-fed calves secreted less TNF-α in LPS-stimulated whole blood cultures at 45 d of age compared with twice-fed calves and this tended to persist through the immediate postweaning period. None of the other immune parameters differed after weaning between the preweaning feeding strategies. Consolidating calf milk replacer into one feeding during the fourth week of life was likely a mild and acute stressor, as evidenced by transient neutrophilia in the absence of suppressed functional capacities of neutrophils. Future research should address the mechanism and immunological significance of the persistent decreased TNF-α response in once-fed calves.

The impact of gestational dam restraint stress on progeny immune and neuroendocrine temporal hormone responses to lipopolysaccharide (LPS) challenge was assessed. Maternal stress (5-min snout snare restraint stress during days 84 to 112 of gestation) increased (P < 0.05) the magnitude of tumor necrosis factor (TNF)-α, interleukin-6, epinephrine (E), norepinephrine, and serum amyloid A (SAA) production following LPS infusion in the offspring. Moreover, these effects appear to be dependent on gender for TNF-α, E, and cortisol production. However, maternal stress did not affect (P > 0.05) the normalization of proinflammatory cytokines or neuroendocrine hormones produced following LPS. Collectively, these results indicate that maternal stress impacts aspects of the proinflammatory cytokine and stress hormone response in their progeny following LPS dosing of the offspring. This response is potentially responsible in part for the resultant changes to SAA production. Because several of the changes observed here are dependent on pig gender, these results are also the first evidence that inherent epigenetic factors coupled with maternal stress impact the cumulative response to stress and LPS in young pigs.

The effects of active dry yeast, Saccharomyces cerevisiae boulardii (Scb), on the immune/cortisol response and subsequent mortality to Escherichia coli lipopolysaccharide (LPS) administration were evaluated in newly weaned piglets (26.1 ± 3.4 d of age). Barrows were assigned to 1 of 2 treatment groups: with (Scb; n = 15) and without (control; n = 15) the in-feed inclusion of Scb (200 g/t) for 16 d. On d 16, all piglets were dosed via indwelling jugular catheters with LPS (25 µg/kg of BW) at 0 h. Serial blood samples were collected at 30-min intervals from -1 to 6 h and then at 24 h. Differential blood cell populations were enumerated hourly from 0 to 6 h and at 24 h. Serum cortisol, IL-1ß, IL-6, tumor necrosis factor-α (TNF-α), and interferon-γ (IFN-γ) concentrations were determined via porcine-specific ELISA at all time points. In Scb-treated piglets, cumulative ADG increased (P < 0.05) by 39.9% and LPS-induced piglet mortality was reduced 20% compared with control piglets. White blood cells, lymphocytes, and neutrophils were increased (P < 0.05) in Scb-treated animals before LPS dosing compared with control piglets before being equally suppressed (P < 0.05) from baseline in both treatments after LPS dosing with a return to baseline by 24 h. Suppression of circulating cortisol concentrations (P < 0.05) was observed in Scb-treated piglets from -1 h to 1 h relative to LPS dosing compared with control animals before both peaked equally and subsequently returned to baseline. Peak production (P < 0.05) of IL-1ß and IL-6 was less in Scb-treated piglets after LPS administration compared with controls before both equally returned to baseline. Peak TNF-α production in Scb-treated animals was accelerated 0.5 h and was greater (P < 0.05) than peak production in control piglets, after which both equally returned to baseline. The peak production of IFN-γ was greater and had increased (P < 0.05) amplitude persistence for 3 h in Scb-treated animals compared with control piglets before both equally returned to baseline. These results highlight the previously unidentified effects of Scb administration on immune and cortisol responses and the subsequent impact on growth and endotoxin-induced mortality in weaned piglets.

Previous research indicates that early weaning before shipment can reduce transportation-induced increases in acute-phase proteins (APP) and can increase feedlot performance in beef calves. These data suggest that the combination of weaning and transport stress may compromise the immune system of calves, thus hindering subsequent performance and health. Therefore, our objective was to determine if the innate immune response of early weaned calves (EW; 80 d of age) differed from normal-weaned calves (NW; 250 d of age) in response to an endotoxin challenge. Eighteen Brahman x Angus calves (8 and 10 EW and NW, respectively; 233 +/- 5 kg of BW) were used. Calves were maintained on pasture with supplement and then moved into individual pens for 1 wk of acclimation before the start of the study. Calves were fitted with an indwelling jugular catheter 1 d before LPS challenge (0 h; 1.0 microg/kg of BW, intravenously). Blood samples were collected at 30-min intervals from -2 to 8 h. Serum samples were stored at -80 degrees C until analyzed for cortisol, tumor necrosis factor-alpha (TNF), IL-1 beta, IL-6, interferon-gamma (IFN), ceruloplasmin, and haptoglobin. The temporal pattern and sex effect of immune and stress hormone responses to a lipopolysaccharide (LPS) challenge were assessed using a pig model. Secretion of the pro-inflammatory cytokines tumor necrosis factor (TNF)-alpha, interleukin (IL)-1beta, and IL-6 increased in a time-dependent manner following LPS infusion. There was also a time-dependent increase in secretion of the stress-related hormones cortisol, epinephrine (E), and norepinephrine (NE) following LPS, with peak concentrations attained within 30 min. The magnitude of the TNF-alpha and IL-1beta responses were both positively associated (P < 0.05) with the magnitude of cortisol response following LPS, whereas serum IL-1beta and IL-6 were positively correlated with the magnitude of E and NE responses following LPS. Acute-phase protein production was also time-dependently increased following LPS. The concentration of immune cells in circulation was decreased (P < 0.05) at 5.5h post-LPS and negatively correlated with pro-inflammatory cytokine production. By 24h post-LPS, immune cell counts increased (P < 0.05) and were positively associated with both pro-inflammatory cytokine and stress hormone production. The amplitude of pro-inflammatory cytokine response following LPS was affected (P < 0.05) by sex classification; however, the magnitude of elevated cytokine concentrations was not. The magnitude of the NE response, but not of the E and cortisol responses, to LPS was influenced by sex (P < 0.05). Similar to the pro-inflammatory cytokines, the magnitude of exposure to the stress hormones following LPS was not influenced by sex. The production of serum amyloid A (SAA) was influenced by sex, with barrows producing more SAA than gilts at 24h post-LPS (P < 0.05). Collectively, these results demonstrate sex-specific, concomitant temporal changes in innate immune- and stress-related hormones.

Full term crossbred sows were selected to study the interaction of the immune system, hypothalamus-pituitary-adrenal axis, and growth in pigs born by Cesarean section (c-section; n=4 sows) or vaginal birth (n=4 sows). Gestation length and birth weight did not differ between vaginal birth and c-section pigs (P=0.34 and 0.62, respectively). Blood and tissue samples were collected from 44 pigs at birth. Forty-five pigs were weaned at 13 d. On d 14, pigs received an i.p. injection of lipopolysaccaride (LPS; 150 microg/kg) or saline at min 0, and blood samples were collected at -20, -10, 0, 5, 10, 20, 40, 60, 90, and 120 min. Vaginal birth pigs had 21% greater average daily gain than c-section pigs on d 14 (P<0.01). Basal serum concentrations of adrenocorticotrophin (ACTH) and cortisol were greater in c-section than vaginal birth pigs at birth (P<0.01) but were not different at 14 d (P=0.99 and 0.80, respectively). LPS increased serum concentrations of ACTH, cortisol, interferon-gamma (IFN-gamma), and tumor necrosis factor-alpha (TNF-alpha; P<0.01) but the response was not different between c-section and vaginal birth (P>0.22). Basal serum concentrations of TNF-alpha tended to be greater in c-section vs vaginal birth pigs at 14 d (P=0.0967); however, basal serum concentrations of IFN-gamma tended to be lower in c-section pigs vs vaginal birth pigs at 14 d (P=0.0787). Expression of interleukin (IL)-6, IL-6 receptor, IL-1beta, and TNF-alpha mRNA did not differ between vaginal birth and c-section pigs but changed in an age and tissue dependent manner. Thus, reduced growth rate of c-section pigs is associated with altered immune system function.

Twenty-four Angus x Hereford crossbred steers (247 kg BW; SE = 2.4 kg) were used in a completely random design to evaluate the effect of energy source and level with or without antibiotic administration on measures of immune function. Steers were fed 1 of 3 dietary treatments: a 70% concentrate diet ad libitum (70AL), a 30% concentrate diet ad libitum (30AL), and a 70% concentrate diet offered in an amount calculated to provide NE(g) intake equal to the 30AL treatment (70RES). Half the steers in each dietary treatment received a s.c. injection of tilmicosin phosphate (ANTI; 1 mL/30 kg of BW); the other half received an equal volume of saline s.c. (SAL). Steers were offered the treatment diets for 28 d before and were administered the ANTI or SAL injections 2 d before indwelling catheters were placed in the jugular vein and 2.0 microg/kg of BW of Escherichia coli lipopolysaccharide (LPS) was administered i.v. Blood serum was collected at 30-min intervals from -2 to 6 h and at 8, 12, 24, 48, and 72 h relative to the LPS challenge. Increased energy intake (70AL) increased (P < or = 0.04) DMI, ADG, and rectal temperature (RT) after the challenge compared with the 70RES treatment. The 30AL treatment increased the maximum concentrations and area under the response curve of the proinflammatory cytokines (PIC) interferon-gamma, tumor necrosis factor-alpha, and IL-6 (P < or = 0.05) compared with the average of the 70AL and 70RES treatments. Decreased energy intake (70RES vs. 70AL) increased IL-6 (P < or = 0.003) but did not significantly increase interferon-gamma and tumor necrosis factor-alpha (P > or = 0.14) after LPS administration. Tilmicosin administration decreased the time to attain maximal RT (P = 0.01) by 1 h without altering the peak RT (P = 0.85), and tilmicosin interacted with energy intake to increase prechallenge PIC in 70RES vs. 70AL (P < or = 0.05). Results indicate that increased PIC response, presumably resulting from a combination of decreased energy intake and from direct effects of roughage, may be a mode of action for the slight decrease in morbidity that often occurs when newly received, stressed calves are fed roughage-based receiving diets. Tilmicosin phosphate might have immunomodulatory capacity beyond its direct effects on pathogenic bacteria, and these effects could interact with dietary energy intake in cattle.

To test possible dietary immune modulators, 32 crossbred male pigs were given 1 of 4 dietary treatments (8 pigs/treatment): control, Saccharomyces cerevisiae with beta-glucan (Energy Plus, Natural Chem Industries LTD, Houston, TX; 0.312 g/kg of BW, 2.5% of diet), vitamin C (Stay C 35, DSM Nutritional Products Inc., Prisippany, NJ; 75 ppm), or beta-glucan plus vitamin C together (combination; 0.312 g/kg of BW and 75 ppm, respectively). Supplements were given in whole milk within 36 h of birth and then daily for 2 wk until weaning, when the supplement was given in feed for an additional 2 wk. Growth was recorded during the 4 wk of supplement delivery. An i.v. lipopolysaccharide challenge (LPS; 150 microg/kg) was given 14 d postweaning at 0900. Behavior was observed, and blood samples were collected every 30 min for 4 h via a jugular catheter from -1 (0800) to 3 (1200) h relative to challenge (-60, -30, 0, 30, 60, 90, 120, 150, and 180 min), and tissues were collected after exsanguination. Beta-glucan (glucan and combination) increased (P < 0.05) BW and ADG compared with vitamin C and control. Cortisol concentrations showed an interaction (P < 0.05) of the beta-glucan and vitamin C. Intestinal expression of tumor-necrosis factor (TNF)-alpha mRNA was greatest for vitamin C and beta-glucan compared with control and combination, and liver TNF-alpha mRNA expression showed a main effect (P < 0.01) of beta-glucan. Lung expression of TNF-alpha mRNA exhibited a vitamin C effect (P < 0.01). In contrast, spleen had greater (P < 0.01) relative abundance of TNF-alpha mRNA in beta-glucan pigs. Intestinal expression of IL-1Ra mRNA was greater (P < 0.05) for vitamin C and beta-glucan treatments compared with the control and combination pigs. Liver expression of IL-1 receptor antagonist mRNA exhibited a vitamin C effect (P < 0.01). Lying and sleeping behaviors differed (P < 0.05) among treatments early in the observations (0700 to 0720), then sporadically until 50 min after the LPS injection. The vitamin C group slept less (P < 0.05) on those occasions. The time spent lying was least (P < 0.05) for the glucan and combination pigs immediately after the injection. These results show a complex interaction between vitamin C and this yeast product after LPS challenge, with differential expression in tissues by 2 h after LPS injections. The combination enhanced postweaning growth and reduced TNF-alpha expression of the intestinal and liver tissues, suggesting an important immunomodulatory role of the combination treatment.

The objectives were to determine relative ADG, ADFI, behavior, and endocrine responses in weaned pigs receiving exogenous ghrelin. Twenty-four barrows weaned at 18 d of age (d 0 of the experiment) were catheterized via the jugular vein, weighed, and assigned to either a ghrelin (n = 12) or saline (control; n = 12) infusion group. Initial pig BW did not differ between treatments (7.87+/-0.39 vs. 7.92+/-0.35 kg for ghrelin and control treatments, respectively). Pig BW and feed intakes were measured once daily throughout the experiment. Starting on d 1, the ghrelin pigs were intravenously infused three times daily for 5 d with 2 microg/kg BW of human ghrelin, and the control pigs were similarly infused with saline. Activity observations and blood samples were taken at -15, 0, 15, 30, 60, 90, 120, 240, and 480 min relative to the first infusion and then three times daily (0800, 1600, and 2400) for 8 d. Weight gain during the 5-d infusion period was greater by the ghrelin than by control pigs (0.57+/-0.10 vs. 0.21+/-0.13 kg, respectively; P < 0.04); however, there was no increase in feed intake. During two behavioral observation periods, more pigs in the ghrelin treatment were observed eating compared with control pigs (P < 0.05). The initial infusion of exogenous ghrelin increased serum ghrelin, GH, insulin, and cortisol concentrations (P < 0.05). Endogenous serum ghrelin increased from d 1 to 8 of the experiment in control animals (P < 0.05). Serum IGF-I initially fell in both treatment groups from d 1 to 2 (P < 0.05) but then increased from d 5 to 8 (P < 0.05). Peripheral concentrations of glucose in the ghrelin pigs were greater on d 2, 3, 7, and 8 than on d 1 (P < or = 0.05). In both treatment groups, peripheral concentrations of leptin increased from d 7 to 8, and cortisol decreased from d 1 to 5 of the experiment. These observations provide evidence that ghrelin may positively influence weight gain and concomitantly increase GH, insulin, and cortisol secretion in weaned pigs.

Seventy-two crossbred pigs (7.58 +/- 0.30 kg BW) weaned at 28 +/- 3 d of age were used to investigate the effects of fish oil supplementation on pig performance and on immunological, adrenal, and somatotropic responses following an Escherichia coli lipopolysaccharide (LPS) challenge in a 2 x 2 factorial design. On d 14 and 21, pigs were injected intraperitoneally with either 200 microg/kg BW of LPS or an equivalent amount of sterile saline. Blood samples were collected 3 h after injection for analysis of interleukin-1beta (IL-1beta), prostaglandin E2 (PGE2), cortisol, growth hormone (GH), and insulin-like growth factor (IGF)-I. On d 2 after LPS challenge, peripheral blood lymphocyte proliferation (PBLP) was determined. Lipopolysaccharide challenge decreased ADG (487 vs. 586 g; P < 0.05) and ADFI (as-fed, 776 vs. 920 g; P < 0.05) from d 14 to 21 and ADG (587 vs. 652 g; P < 0.10) from d 21 to 28. Fish oil improved ADG (554 vs. 520 g; P < 0.10) and ADFI (891 vs. 805 g; P < 0.10) from d 14 to 21. On d 14, LPS challenge x diet interactions were observed for IL-1beta (P < 0.10), PGE2 (P < 0.001), and cortisol (P < 0.05) such that these measurements responded to the LPS challenge to a lesser extent (IL-1beta: 93 vs. 114 pg/mL, P < 0.05; PGE2: 536 vs. 1,285 pg/mL, P < 0.001; cortisol: 143 vs. 206 ng/mL, P < 0.05) in pigs receiving the FO diet than in pigs fed the CO diet. In contrast, among LPS-treated pigs, pigs fed the FO diet had higher IGF-I (155 vs. 101 ng/mL; P < 0.10) than those fed the CO diet. On d 21 among LPS-treated pigs, pigs fed FO had lower IL-1beta (70 vs. 84 pg/mL; P < 0.10) and cortisol (153 vs. 205 ng/mL; P < 0.05) than those fed CO. Pigs fed FO had lower PGE2 (331 vs. 444 pg/mL; P < 0.05) and higher IGF-I (202 vs. 171 ng/mL; P < 0.10) compared with those fed CO. Lipopolysaccharide challenge decreased GH (0.27 vs. 0.33 ng/mL; P < 0.05) on d 14, whereas it had no effect on GH on d 21. During both LPS challenge periods, the challenge increased PBLP when these cells were incubated with 8 (1.46 vs. 1.32; P < 0.10) or 16 microg/mL (1.46 vs. 1.30; P < 0.05) of concanavalin A. Fish oil had no effect on PBLP. These results suggest that FO alters the release of proinflammatory cytokines, which might lead to improved pig performance during an immunological challenge.

The study objective was to assess endocrine and tIssue responses to feed deprivation in weaned pigs. In experiment 1, eight crossbred castrated male pigs were either fed on a continual basis (CON; n=4) or were feed deprived for 24 h and then re-fed until 30 h (FD; n=4). Relative serum concentrations of ghrelin tended to be lower in FD pigs at 12 h (P=0.08) when compared with CON pigs, but was higher at 24 h and 30 h compared with 12 h (P<0.05). Serum IGF-I was lower in FD pigs from 12 to 30 h as compared with CON pigs (P<0.05) and increased following re-feeding (P<0.06). Experiment 2 consisted of 32 pigs that were either fed for 72 or 96 h (CON72 and CON96; n=16), feed deprived for 72 h (FD72; n=8), or FD72 and then re-fed from 72-96 h (FD72/RF24; n=8). Ghrelin in the FD72 and FD72/RF24 groups was lower at 12 h (P<0.03) compared with CON pigs, but then increased from 12 h to 36 h (P<0.01). Serum IGF-I and leptin decreased following feed deprivation (P<0.001) and remained low until re-feeding. Cortisol was elevated from 12 h to 72 h during feed deprivation (P<0.01) but was not different from CON96 pigs following refeeding (P>0.88). Expression of ghrelin mRNA tended to be lower in the FD72 pigs' stomachs, pituitary glands, and hypothalami (P=0.06, 0.07, and 0.08 respectively) compared with CON pigs. These results provide evidence that feed deprivation is accompanied by multiple changes in the endocrine and neuroendocrine axis which influences feed intake, somatotropic response, and hypothalamic-pituitary-adrenal axis hormone concentrations.
