RESUMO

Four experiments were designed to evaluate the effect of different circulating progesterone (P4) concentrations during a synchronization of ovulation protocol for the timed artificial insemination (TAI) of Bos indicus (Nelore) beef cattle. In the first trial, 13 ovariectomized Nelore heifers were randomly allocated into one of three groups using new P4 devices (New; 1.0 g P4), previously used P4 devices for 8 days (Used1x), and previously used P4 devices for 16 days (Used2x), in a crossover experimental design. The circulating P4 concentrations during the P4 device treatment were lower for Used1x (2.3 ± 0.1 ng/mL) and Used2x (2.0 ± 0.1 ng/mL) than those for New (3.8 ± 0.2 ng/mL; P = 0.001). In the second trial, the ovarian follicular dynamics of 60 anestrous cows were evaluated after the cows received the treatments described previously (New [n = 20], Used1x [n = 20], and Used2x [n = 20]). During the insertion of the P4 device, the cows were administered 2.0-mg estradiol benzoate. Eight days later, the P4 device was removed, and the cows were administered 0.53-mg sodium cloprostenol, 300 IU eCG, and 1-mg estradiol cypionate. There were no differences among the groups during the interval from P4 device removal to ovulation (73.7 ± 2.9 vs. 69.8 ± 2.4 vs. 68.4 ± 2.3 hours) or regarding the ovulation rate (70.0% vs. 80.0% vs. 85.0%). However, the maximum diameter of the largest follicle was greater (P = 0.06) in the Used2x (15.3 ± 0.4 mm) than that of New (13.5 ± 0.8 mm) and Used1x (14.9 ± 0.5 mm). In experiment 3, 443 anestrous cows were randomly assigned into one of the three treatments (New [n = 144] vs. Used1x [n = 167] vs. Used2x [n = 132]) and received a TAI 48 hours after the P4 device removal. The diameter of the largest follicle during the device removal (10.7 ± 0.3 vs. 11.2 ± 0.2 vs. 11.3 ± 0.3 mm) and the 30-day pregnancy rates (51.4% vs. 53.9% vs. 43.2%) did not differ among the experimental groups. In experiment 4, a field trial with 593 B indicus (Nelore) cows was conducted to evaluate the pregnancy per AI using different levels of P4 in a TAI protocol (New [n = 189] vs. Used1x [n = 203] vs. Used2x [n = 201]). The pregnancy per AI was similar between the treatment groups (63.5% vs. 57.6% vs. 62.7%). In conclusion, the low circulating P4 concentrations that were released from a used P4 device efficiently controlled the ovarian follicular growth and exhibited no detrimental effects on the pregnancy rates of the B indicus (Nelore) beef cattle.

The effects of estradiol benzoate (EB) and estradiol cypionate (EC) on induction of ovulation after a synchronized LH surge and on fertility of Bos indicus females submitted to timed AI (TAI) were evaluated. In Experiment 1, ovariectomized Nelore heifers were used to evaluate the effect of EB (n = 5) and EC (n = 5) on the circulating LH profile. The LH surge timing (19.6 and 50.5 h; P = 0.001), magnitude (20.5 and 9.4 ng/mL; P = 0.005), duration (8.6 and 16.5 h; P = 0.001), and area under the LH curve (158.6 and 339.4 ng/mL; P = 0.01) differed between the EB and EC treatments, respectively. In Experiment 2 (follicular responses; n = 60) and 3 (pregnancy per AI; P/AI; n = 953) suckled Bos indicus beef cows submitted to an estradiol/progesterone-based synchronization protocol were assigned to receive one of two treatments to induce synchronized ovulation: 1 mg of EB im 24 h after progesterone (P4) device removal or 1 mg of EC im at P4 device removal. There was no difference (P > 0.05) between EB and EC treatments on follicular responses (maximum diameter of the ovulatory follicle, 13.1 vs. 13.9 mm; interval from progesterone device removal to ovulation, 70.2 vs. 68.5 h; and ovulation rate, 77.8 vs. 82.8%, respectively). In addition, P/AI was similar (P < 0.22) between the cows treated with EB (57.5%; 277/482) and EC (61.8%; 291/471). In conclusion, despite pharmacologic differences, both esters of estradiol administered either at P4 device removal (EC) or 24 h later (EB) were effective in inducing an LH surge which resulted in synchronized ovulations and similar P/AI in suckled Bos indicus beef cows submitted to TAI.

The present study aimed to evaluate the efficacy of different inducers of new follicular wave emergence (FWE) and ovulation in fixed-time artificial insemination (FTAI) synchronization protocols using norgestomet ear implants (NORG) in Bos indicus cattle. In Experiment 1, the synchronization of FWE was evaluated when two different estradiol esters in different doses [2mg estradiol benzoate (EB), 2.5mg EV or 5mg estradiol valerate (EV)] were administered with NORG implant insertion in B. indicus cattle (estrous cyclic heifers and cows with suckling calves; n=10 per treatment). After estradiol treatment, ovarian ultrasonic exams were performed once daily to detect the interval between treatment and FWE. There were significant treatment-by-animal category interaction (P=0.05) on the interval from the estradiol treatment to FWE. An earlier (P<0.0001) and less variable (P=0.02) interval from estradiol treatment to FWE was observed in heifers treated with EB (2.5±0.2; mean±SE) than in those treated with 2.5mg EV (4.2±0.3) or 5mg EV (6.1±0.6). Cows treated with 5mg EV (4.0±0.5) had longer (P=0.05) interval than cows receiving EB (2.5±0.2), however, there was an intermediate interval in those cows treated with 2.5mg EV (3.1±0.4). In Experiment 2, the number of uses of the NORG implant (new; n=305 or previously used once; n=314) and three different ovulation induction hormones [0.5mg estradiol cypionate (EC) at implant removal (n=205), 1mg EB given 24h after implant removal (n=219), or 100µg gonadorelin (GnRH) given at FTAI (n=195)] were evaluated in Nelore heifers (2×3 factorial design). Similar pregnancy per AI (P/AI; 30 days after FTAI; P>0.05) were achieved using each of the three ovulation induction hormones (EB=40.6%; EC=48.3%, or GnRH=48.7%) and with a new (47.2%) or once-used NORG implant (44.3%). In Experiment 3, the effect of different ovulation induction hormones for FTAI [1mg EC at NORG implant removal (n=228), 10µg buserelin acetate at FTAI (GnRH; n=212) or both treatments (EC+GnRH; n=215)] on P/AI was evaluated in suckled beef cows treated with a once-used NORG implant and EB to synchronize the FWE. Similar P/AI (P=0.71) were obtained using GnRH (50.9%), EC (51.8%) or both treatments (54.9%) as ovulation induction hormones. Therefore, both doses of EV (2.5 or 5.0mg) with NORG implant delayed and increased the variation of the day of new FWE compared with EB in B. indicus cattle. These effects were more pronounced in B. indicus heifers than cows. Synchronization protocols for FTAI with either a new or once-used NORG implant with EB at insertion to induce a new FWE and either the use of EB, EC or GnRH as ovulation induction hormones may be successful in B. indicus heifers. Also, when a once-used NORG implant was used, either the administration of EC, GnRH or both as ovulation inducers resulted in similar P/AI in suckled B. indicus cows, showing no additive effect of the combination of both ovulation induction hormones.

Medidas de controle de doenças por meio do uso de extratos e óleos essenciais vêm sendo investigadas como alternativa aos fungicidas convencionais. Este trabalho objetivou estudar o efeito antimicrobiano de extratos brutos aquosos e óleo essencial de Achillea millefolium no crescimento micelial (CM), na esporulação e na germinação de conídios de Corynespora cassiicola bem como a atividade de peroxidase em plantas de pepino. Para isto, o extrato bruto aquoso (EBA) (1; 5; 10; 20 e 25%) foi incorporado ao meio de cultura BDA (batata-dextrose-ágar) e o óleo essencial (OE) (20, 40, 60, 100 e 200 mL) foi distribuído sobre a superfície do meio de cultura. Disco de micélio foi repicado para os diferentes meios e o crescimento micelial avaliado, diariamente, por 7 dias. Para os ensaios in vivo, plantas de pepino foram pulverizadas com EBAs nas concentrações 1, 10 e 25%, Saccharomyces cereviseae 20% e água, aos quatro e dois dias antes e concomitantemente à inoculação com C. cassiicola. Foi avaliada a severidade da doença e a atividade da enzima peroxidase. Para EBA, não houve inibição do crescimento micelial, da germinação e da esporulação em todas as concentrações testadas. Entretanto, nos tratamentos com OE, houve inibição de até 63% do crescimento micelial. Na esporulação e germinação, a inibição foi de 100% e 98%, respectivamente, na alíquota de 200 ?L para OE. No controle da doença, o tratamento mais efetivo foi EBA a 25% quando realizado concomitante à inoculação. No intervalo de tempo avaliado, não se constatou atividade da enzima peroxidase.


The control of diseases by using crude extracts (CE) and essential oils (EO) are being investigated as an alternative to conventional fungicides. The objective of this work was to evaluate the antimicrobial effect of aqueous extracts and essential oil of Achillea millefolium on mycelial growth, sporolution and spores germination of Corynespora cassiicola, as well as peroxidase activity in cucumber plants. For this, crude extract (1; 5; 10; 20 and 25%) was incorporated into potato dextrose agar medium (PDA), and essential oil (20, 40, 60, 100 and 200 mL) was distributed over the surface of the PDA. The mycelial growth was measured daily for 7 days. In the in vivo test, cucumber plants were sprayed with crude extracts at concentrations 1; 10 and 25%, and Saccharomyces cereviseae 20% (control), 4 and 2 days before and the same time as inoculation with C. cassiicola. The severity of disease and peroxidase activity were evaluated. For crude extract, neither mycelial growth nor spore germination and sporulation was inhibited at any of the tested concentrations. In the treatments with essential oil there was inhibition up to 63% of the mycelial growth. In the sporulation and spores germination the inhibition was 100% and 98%, respectively, for 200uL of EO. In the control of the disease the mosteffective treatment was the crude extract 25% when carried out at the same time as the inoculation. In the evaluated period, no peroxidase activity was found.

The objective of this study was to evaluate the effects of treatment with an intravaginal progesterone-releasing device (CIDR) and estradiol benzoate (EB) on follicular dynamics in Bos indicus (n=23), Bos taurus (n=25), and cross-bred (n=23) heifers. To assess the influence of reduced serum progesterone concentrations during 8 days of treatment with a progesterone-releasing device on follicular dynamics, half of the heifers received PGF at CIDR insertion (Day 0; 3 x 2 factorial design). Mean (+/-S.E.M.) serum progesterone concentrations during CIDR treatment varied (P<0.05) among genetic groups: B. indicus (5.4+/-0.1 ng/mL), B. taurus (3.3+/-0.0 ng/mL), and cross-bred (4.3+/-0.1 ng/mL). Maximum diameter of the dominant follicle (DF) was smaller (P<0.01) in B. indicus heifers (9.5+/-0.5 mm) than in cross-bred (12.3+/-0.4 mm) or B. taurus heifers (11.6+/-0.5 mm). B. indicus experienced lower (P<0.01) ovulation rate (39.1%) than did B. taurus (72.7%) and cross-bred (84.0%). Heifers treated with PGF on Day 0 had lower (P<0.05) serum progesterone concentrations during progesterone treatment. The PGF treatment on Day 0 increased (P<0.01) the diameter of the DF (11.9+/-0.4 mm vs. 10.5+/-0.4 mm). Moreover, greater (P=0.02) ovulation rates (78.8 vs. 54.0%) occurred in heifers treated with PGF on Day 0. In summary, B. indicus heifers had greater serum progesterone concentrations, smaller DF diameter, and a lower ovulation rate compared to B. taurus heifers. Prostaglandin treatment on the day of CIDR insertion reduced serum progesterone during treatment, and resulted in increased maximum DF diameter and ovulation rate.

RESUMO

C. chauvoei presence was detected by means of polymerase chain reaction (PCR) from supernatant of culture in cooked meat medium of liver, muscle and metatarsian bone marrow samples of seven calves with blackleg symptoms. The isolation under anaerobic conditions of one muscle sample revealed Clostridium perfringens in pure culture.


Foi detectada presença de Clostridium chauvoei pela reação de PCR a partir de cultivo em cooked meat medium de amostras de fígado, músculo e medula óssea metatarsiana de sete bezerros acometidos de carbúnculo sintomático. O isolamento de uma amostra de músculo sob condições anaeróbias revelou Clostridium perfringens em cultura pura.

Dez vacas multíparas, secas, foram distribuídas aleatoriamente em dois grupos de cinco animais cada. Nos dias 8 a 12 do diestro, o primeiro grupo recebeu 100 ml de anti-soro contra líquido folicular livre de esteróides (anti-LFb) produzido em ovelhas ovariectomizadas. O segundo grupo (controle) recebeu 100 ml de soro de ovelhas näo-imunizadas. Seis horas após a aplicaçäo, os dois grupos foram superovulados com FSH (18 NIH-FSH-S1 unidades) e LH (0,29 NIH-LH-S1 unidades) administrados em quantidades decrescentes durante quatro dias. Na manhä do terceiro dia, foi administrada uma dose luteolítica de cloprostenol. Duas inseminaçöes foram realizadas 48 e 60 horas após. Os embriöes foram recuperados pelo método cervical 7 dias após a primeira inseminaçäo. Amostras de sangue foram coletadas durante todo o período experimental para determinar, por radioimunoensaio, as concentraçöes plasmáticas de FSH, LH e progesterona. Todas as vacas do grupo imunizado e 3 do grupo controle apresentaram mais de 2 CL. Näo existiu diferença significativa (p>0,05) na taxa de ovulaçäo entre os grupos imunizado e controle (14,4 e 9,9, respectivamente). O número de embriöes recuperado näo foi significativamente diferente (p>0,05) entre os grupos, embora o grupo imunizado tenha apresentado maior número de embriöes transferíveis (3,4 ñ 1,0 versus 0,8 ñ 0,4, p<0,05). As concentraçöes de gonadotrofinas plasmáticas näo foram correlacionadas com a taxa de ovulaçäo ou com o número de embriöes recuperados. As concentraçöes de progesterona plasmática foram positivamente correlacionadas (r = 0,88, p<0,01) com a taxa de ovulaçäo. Os resultados sugerem que o anti-LFb, aplicado antes da superovulaçäo, näo reduz a variabilidade da resposta ovariana

RESUMO

Neste trabalho fez-se um estudo comparativo das propriedades fisicoquimicas, quimicas e organolepticas de queijos Mozzarelas elaborados com leite de bufala, utilizado isoladamente ou em combinacao com o leite de vaca na proporcao de l:l, com aqueles convencionalmente fabricados com leite de vaca. Apos a obtencao dos queijos procedeu-se a avaliacao organoleptica e as determinacoes de pH, acidez titulavel, umidade, gordura, residuo mineral fixo, proteina bruta e nitrogenio soluvel
