Objectives: Non-alcoholic fatty liver disease (NAFLD) has been linked to an increased risk of kidney stones in prior observational studies, However, the results are inconsistent, and the causality remains to be established. We aimed to investigate the potential causal relationship between NAFLD and kidney stones using two-sample Mendelian randomization (MR). Methods: Genetic instruments were used as proxies for NAFLD. Summary-level data for the associations of exposure-associated SNPs with kidney stones were obtained from the UK Biobank study (6536 cases and 388,508 controls) and the FinnGen consortium (9713 cases and 366,693 non-cases). MR methods were conducted, including inverse variance weighted method (IVW), MR-Egger, weighted median, and MR-PRESSO. MR-Egger Regression Intercept and Cochran's Q test were used to assess the directional pleiotropy and heterogeneity. Results: cALT-associated NAFLD did not exhibit an association with kidney stones in the Inverse variance weighted (IVW) methods, in both the FinnGen consortium (OR: 1.02, 95%CI: 0.94-1.11, p = 0.632) and the UKBB study (OR: 1.000, 95%CI: 0.998-1.002, p = 0.852). The results were consistent in European ancestry (FinnGen OR: 1.05, 95%CI: 0.98-1.14, p = 0.144, UKBB OR: 1.000, 95%CI: 0.998-1.002, p = 0.859). IVW MR analysis also did not reveal a significant causal relationship between NAFLD and the risk of kidney stone for the other three NAFLD-related traits, including imaging-based, biopsy-confirmed NAFLD, and more stringent biopsy-confirmed NAFLD. The results remained consistent and robust in the sensitivity analysis. Conclusions: The MR study did not provide sufficient evidence to support the causal associations of NAFLD with kidney stones.

Graphene (GE) is an emerging type of two-dimensional functional nanoparticle with a tunable passageway for oil molecules. Herein, polyvinylidene fluoride (PVDF)/GE composite membranes with controllable pore structure were fabricated with a simple non-solvent-induced phase separation method. The change of crystallinity and crystal structure (α, ß, γ, etc.) generated is due to the addition of GE, which benefits the design of a suitable pore structure for oil channels. Meanwhile, the hydrophobicity and thermal stability of the composite membrane were obviously enhanced. With 3 wt % GE, the contact angle was 124.6°, which was increased greatly compared to that of the GE-0 sample. Moreover, the rate of the phase transition process was affected by the concentration of casting solution, temperature, and composition of the coagulation bath. For example, the composite membrane showed better oil-water separation properties when the coagulation bath was dioctyl phthalate. In particular, the oil flux and separation efficiencies were up to 2484.08 L/m2·h and 99.24%, respectively. Consequently, PVDF/GE composite membranes with excellent lipophilicity may have good prospects for oily wastewater treatment.

BACKGROUND: Mesenchymal stromal cells (MSCs) therapy for acute respiratory distress syndrome (ARDS) is an emerging treatment, but most of the current trials of MSCs stay in the animal experimental stage, and the safety and efficacy of MSCs in clinical application are not clear. We aimed to analyze the safety, efficacy and biomarkers of mesenchymal stromal cells in the treatment of ARDS. METHODS: For this systematic review and meta-analysis, we searched PubMed, Embase, Cochrane Central Register of Controlled Trials, Web of science, CNKI, VIP and Wan Fang data, studies published between database inception and Mar 17, 2022. All randomized controlled trials (RCT) of stem cell interventions for ARDS were included, without language or date restrictions. We did separate meta-analyses for mortality, subjects with adverse events (AEs) and subjects with serious adverse events (SAEs). Since the trials data are dichotomous outcomes, the odds ratio (OR) is adopted for meta-analysis. The quality of the evidence was assessed with the Cochrane risk of bias tool. FINDINGS: In total, 5 trials involving 171 patients with ARDS were included in this meta-analysis. A total of 99 individuals were randomly assigned to receive MSCs treatment, and 72 were randomly assigned to receive placebo treatment. Treatment with MSCs appeared to increase the occurrence of adverse events, but this result was not statistically significant (OR, 1.58; 95%CI, 0.64-3.91; P = 0.32). The occurrence of serious adverse events was lower in the MSCs group than in the placebo group (OR, 0.57; 95%CI, 0.14-2.32; P = 0.43); there seems to be no significant difference between the two groups in terms of 28 days mortality (OR, 0.93; 95%CI, 0.45-1.89); oxygenation index and biomarkers showed a tendency to improve in treatment, but there was a lack of more statistically significant clinical evidence to support them. INTERPRETATION: Based on the current clinical trials, MSCs intervention has some safety for ARDS patients, but its effectiveness and predictive value of airspace biomarkers need to be determined by more large-scale, standard randomized controlled trials.

In this work, polyvinylidene fluoride (PVDF) loose nanofiltration (NF) hollow fiber membranes with multilayer structure were prepared successfully based on a solvent-free process. Graphene oxide (GO) was used to cover the interface pores of the pristine PVDF membranes via vacuum filtration, and polypyrrole (PPy) was polymerized on the surface to further decorate the membrane structure. Interestingly, the modified membranes exhibited a multilayer structure due to synergistic effect of GO and PPy. The structure and property of PVDF loose NF membranes were investigated in detail. After modifying by GO and PPy, the hydrophilicity improved obviously. Moreover, the molecular weight cut off (MWCO) was about 3580 Da, and the smallest pore size of skin layer decreased to 2.5-4 nm. Furthermore, the PVDF loose NF hollow fiber membranes presented a high dye rejection (˃98.5%) for negative dyes, whereas a low salt rejection for NaCl (about 4%), showing a great potential for separating dye/salt accurately. Specifically, there were not any solvent used in all the preparation processes. The work offered a novel strategy for green preparation of loose NF membranes.

This study investigated the effect of L-theanine on carcass traits, meat quality, muscle antioxidant capacity, and amino acid (AA) profiles of broilers. Three hundred 1-day-old Ross 308 male broilers were randomly allotted to five groups with six replicates. Birds were fed the basal diet or basal diet with 300, 600, 900, or 1,500 mg/kg L-theanine for 42 consecutive days. The results showed that L-theanine quadratically increased dressing percentage, eviscerated percentage, and leg muscle yield (p < .05). Meanwhile, drip loss, cooking loss, shear force, L*24h, and muscle lactate content decreased quadratically in response to dietary L-theanine supplementation (p < .05), while pH24h and muscle glycogen content were quadratically improved by L-theanine (p < .05). Notably, the contents of muscle malondialdehyde and protein carbonyl, and the activities of muscle total antioxidant capacity, catalase, and glutathione peroxidase decreased quadratically in response to dietary L-theanine supplementation (p < .05), suggesting that the oxidative stress level of muscle was decreased quadratically. Moreover, L-theanine quadratically increased the concentrations of most of muscle essential AA, nonessential AA, and flavor AA (p < .05). In conclusion, L-theanine can be used as a valuable feed additive to modulate carcass traits, meat quality, muscle antioxidant status, and AA profiles of boilers, and its optimum addition level is 600 mg/kg based on the present study.

Biosurfactant plays an important role in bioremediation of crude oil contamination and microbial enhanced oil recovery (MEOR). In the present study, a salt-tolerant, biosurfactant-producing bacterium, designated A-8, was isolated from wastewater contaminated with petroleum collected from the Changqing reservoir in China. A phylogenetic analysis based on the 16S rRNA sequence suggests that strain A-8 belongs to the genus Achromobacter. The optimal growth conditions for strain A-8 in mineral salt (MS) medium were 30°C, pH 7, and 10 g/L NaCl, while the optimal conditions for biosurfactant production in a fermentation medium were 40-45°C, pH 7, and more than 70 g/L NaCl. Better biosurfactant production was obtained from strain A-8 when edible oil and liquid paraffin were used as carbon sources and when (NH4)2SO4 was used as an inorganic nitrogen source compared with other tested carbon and nitrogen sources. The biodegradation of petroleum in MS medium in different optimized conditions reached 56.23-73.87% for 20 days. The biodegradation of petroleum, together with the production of organic acid and biosurfactant, decreased the viscosity of petroleum by about 45%. The decrease in petroleum viscosity and the biodegradation of petroleum suggest the potential use of strain A-8 for MEOR and bioremediation of petroleum-contaminated environments.

G-quadruplexes (Gqs) are guanine-rich DNA structures formed by single-stranded DNA. They are of paramount significance to gene expression regulation, but also drug targets for cancer and human viruses. Current ensemble and single-molecule methods require fluorescent labels, which can affect Gq folding kinetics. Here we introduce, a single-molecule Gq nanopore assay (smGNA) to detect Gqs and kinetics of Gq formation. We use ∼5 nm solid-state nanopores to detect various Gq structural variants attached to designed DNA carriers. Gqs can be identified by localizing their positions along designed DNA carriers, establishing smGNA as a tool for Gq mapping. In addition, smGNA allows for discrimination of (un)folded Gq structures, provides insights into single-molecule kinetics of Gq folding, and probes quadruplex-to-duplex structural transitions. smGNA can elucidate the formation of Gqs at the single-molecule level without labeling and has potential implications on the study of these structures both in single-stranded DNA and in genomic samples.

Graphene (GE) has attracted significant attention on account of its unique structure and superior performance, arousing a new research field for materials science. Herein, a novel GE-coated poly(ethylene terephthalate) nonwoven (PGNW) hollow tube (PGNW-T) was fabricated for continuous and highly effective oil collection from the water surface. The PGNW was prepared via a dip-spray coating method, which possessed superhydrophobicity-superoleophilicity and could absorb a variety of oils or organic solvents with the absorption capacity (Q) value of 18-34 times its own weight. Then, PGNW-T was obtained through winding the PGNW on the surface of a porous polypropylene hollow tube. As-prepared PGNW-T was competent for dynamic oil collection with high flux (18 799.94 L/m2 h), outstanding separation efficiency (97.14%), and excellent recyclability (>96% after 10 cycles) from the oil/water mixture. In particular, a miniature device based on as-prepared PGNW-T was developed for continuous thin oil film collection, which could dynamically "catch up" floated oils or organic solvents from the water surface. Finally, our strategy is extremely facile to scale up, showing its huge potential application in practical oil-spill remediation.

BACKGROUND: Resveratrol has been shown to prevent high ambient temperature (HT)-induced spleen dysplasia, but the mechanisms of action are not clear. This study aims to examine the hypothesis that HT-induced spleen dysplasia may be associated with HT-induced oxidative stress and apoptosis, and resveratrol may activate the nuclear factor erythroid 2-related factor 2 (Nrf2) signaling pathway, thus reducing oxidative stress and apoptosis. RESULTS: Results showed that HT caused spleen dysplasia in broilers, reflecting the lower relative weight of the spleen (P < 0.05). Compared with birds in a normal ambient temperature group, birds in the HT group exhibited higher (P < 0.05) malondialdehyde (MDA), protein carbonyl (PC), 8-hydroxydeoxyguanosine (8-OHdG) and Bcl-2 associated X protein (Bax) content, higher Bax, caspase-3 and caspase-9 mRNA levels, and caspase-3 and caspase-9 activity, and a higher Bax/B-cell lympoma/leukemia-2 (Bcl-2) ratio, but they exhibited lower (P < 0.05) glutathione (GSH) and Bcl-2 content, and lower Nrf2, glutathione peroxidase (Gpx), MnSOD, heme oxygenase 1, glutathione reductase (GR) and Bcl-2 mRNA levels, and lower total antioxidant capacity (T-AOC), T-SOD and catalase and maganese superoixide dismutase (CAT) activity, indicating HT-induced oxidative stress and apoptosis. Compared with birds in the HT group, birds in the HT + Res group exhibited higher (P < 0.05) GSH and Bcl-2 content, higher Nrf2, CAT, MnSOD, GR and Bcl-2 mRNA levels, and higher T-AOC, T-SOD and CAT activity, but lower (P < 0.05) MDA content, and Bax and caspase-3 mRNA levels, lower caspase-3 and caspase-9 activities, and Bax/Bcl-2 ratio, indicating that resveratrol activated the Nrf2 signaling pathway and decreased apoptosis in the spleen. CONCLUSION: Resveratrol was effective in ameliorating HT-induced spleen dysplasia in broilers through the activation of the Nrf2 signaling pathway, thereby decreasing apoptosis, suggesting that resveratrol may offer a potential nutritional strategy to protect against some HT-induced detriments. © 2018 Society of Chemical Industry.

Polysulfone (PSF)/fluorinated ethylene propylene (FEP) mixed matrix membranes (MMMs) with super hydrophobic surface were successfully fabricated via non-solvent induced phase separation (NIPS) method. The effects of FEP content on the morphology, roughness, wettability, pore size, and mechanical property of PSF/FEP MMMs were characterized by scanning electron microscope, confocal microscopy, contact angle goniometer, mercury porosimetry, and tensile testing instrument, respectively. When the FEP content was 9 wt%, the average roughness of M-4 reached 0.712 µm. Meanwhile, the water contact angle (CA) and the water sliding angle (SA) was 153.3° and 6.1°, respectively. M-4 showed super hydrophobicity with a micro- and nanoscale structure surface. Then, M-4 was used for separating of water-in-oil emulsion, showing high separation efficiency for water-in-kerosene and water-in-diesel emulsions of 99.79% and 99.47%, respectively. The flux and separation efficiency changed slightly after 10 cycles. Therefore, this study indicated that the obtained PSF/FEP MMM with super hydrophobic surface could be used for efficient water-in-oil emulsion separation.

Nanopore sensors show great potential for rapid, single-molecule determination of DNA sequence information. Here, we develop an ionic current-based method for determining the positions of short sequence motifs in double-stranded DNA molecules with solid-state nanopores. Using the DNA-methyltransferase M.TaqI and a biotinylated S-adenosyl-l-methionine cofactor analogue we create covalently attached biotin labels at 5'-TCGA-3' sequence motifs. Monovalent streptavidin is then added to bind to the biotinylated sites giving rise to additional current blockade signals when the DNA passes through a conical quartz nanopore. We determine the relationship between translocation time and position along the DNA contour and find a minimum resolvable distance between two labeled sites of ∼200 bp. We then characterize a variety of DNA molecules by determining the positions of bound streptavidin and show that two short genomes can be simultaneously detected in a mixture. Our method provides a simple, generic single-molecule detection platform enabling DNA characterization in an electrical format suited for portable devices for potential diagnostic applications.
