RESUMO

Perfluoroalkyl and polyfluoroalkyl substances (PFASs) are synthetic chemicals, encompassing compounds like perfluorooctane sulfonate (PFOS), which have widespread applications across various industries, including food packaging and firefighting. In recent years, China has increasingly employed 6:2 Cl-PFESA as an alternative to PFOS. Although the association between PFAS exposure and hepatocellular carcinoma (HCC) has been demonstrated, the underlying mechanisms that promote HCC proliferation are uncleared. Therefore, we aimed to investigate the effects and differences of PFOS and 6:2 Cl-PFESA on HCC proliferation through in vivo and in vitro tumor models. Our results reveal that both PFOS and 6:2 Cl-PFESA significantly contribute to HCC proliferation in vitro and in vivo. Exposure led to reduced population doubling times, enlarged cell colony sizes, enhanced DNA synthesis efficiency, and a higher proportion of cells undergoing mitosis. Furthermore, both PFOS and 6:2 Cl-PFES) have been shown to activate the PI3K/AKT/mTOR signaling pathway and inhibit necroptosis. This action consequently enhances the proliferation of HCC cells. Our phenotypic assay findings suggest that the tumorigenic potential of 6:2 Cl-PFESA surpasses that of PFOS; in a subcutaneous tumor model using nude mice, the mean tumor weight for the 6:2 Cl-PFESA-treated cohort was 2.33 times that observed in the PFOS cohort (p < 0.01). Despite 6:2 Cl-PFESA being considered a safer substitute for PFOS, the pronounced effects of this chemical on HCC cell growth warrant a thorough assessment of hepatotoxicity risks linked to its usage.

Perfluorooctanoic acid (PFOA), a synthetic alkyl chain fluorinated compound, has emerged as a persistent organic pollutant of grave concern, casting a shadow over both ecological integrity and humans. Its insidious presence raises alarms due to its capacity to bioaccumulate within the human liver, potentially paving the treacherous path toward liver cancer. Yet, the intricate mechanisms underpinning PFOA's role in promoting the growth of hepatocellular carcinoma (HCC) remain shrouded in ambiguity. Here, we determined the proliferation and transcription changes of HCC after PFOA exposure through integrated experiments including cell culture, nude mice tests, and colony-forming assays. Based on our findings, PFOA effectively promotes the proliferation of HCC cells within the experimental range of concentrations, both in vivo and in vitro. The proliferation efficiency of HCC cells was observed to increase by approximately 10% due to overexposure to PFOA. Additionally, the cancer weight of tumor-bearing nude mice increased by 87.0% (p < 0.05). We systematically evaluated the effects of PFOA on HCC cells and found that PFOA's exposure can selectively activate the PI3K/AKT/mTOR/4E-BP1 signaling pathway, thereby playing a pro-cancer effect on HCC cells Confirmation echoed through western blot assays and inhibitor combination analyses. These insights summon a response to PFOA's dual nature as both an environmental threat and a promoter of liver cancer. Our work illuminates the obscured domain of PFOA-induced hepatoxicity, shedding light on its ties to hepatocellular carcinoma progression.

Monoaromatic hydrocarbons (MACHs) are a ubiquitous category of volatile compounds found in various environmental media. Despite their prevalence, systematic studies of MACHs on a large regional scale are still lacking. Herein, a comprehensive investigation of the occurrence, seasonal variations, distribution characteristics, and health risks of MACHs was carried out by analyzing soil samples (372 surface soils and 96 soil columns) from 33 typical industrial parks in the Yangtze River Delta (YRD) region. MACHs were detected in all surface soil samples. BTEXS (benzene, toluene, ethylbenzene, xylene, and styrene) were the five predominant congeners with the highest detection frequencies (90.9 %-100 %), collectively accounting for >78.2 % of the total MACHs content. Higher residual levels of MACHs were observed in winter compared to summer (P < 0.01), with total concentrations of 24 MACHs ranging from 30.9 ng/g to 1536 ng/g (median: 135 ng/g) in winter and 16.3 ng/g to 931 ng/g (median: 87.9 ng/g) in summer. Soils collected from the northeast of Jiangsu Province and the southwest of Anhui Province exhibited relatively higher levels of MACHs. On the basis of principal component analysis, we proposed that industrial emissions and vehicle exhaust may be the main sources of MACHs contamination in the soils of YRD industrial parks. Vertically, the concentrations of total MACHs decreased with the soil depth. Soil organic matter (OM) content and the concentration of MACHs in the surface soil layer (0-15 cm) were significant factors influencing the vertical migration and distribution of MACHs (P < 0.05). It was verified that residual MACHs in the soils posed lower lifetime non-carcinogenic and carcinogenic risks to the inhabitants of the study area. The field study provides valuable evidence for the formulation of MACHs pollution control policies in the YRD region.

Pollution of microplastics (MPs) has become a potential threat to Antarctic marine ecosystems. However, the occurrence of MPs in Antarctic krill (Euphausia superba), a keystone species in Antarctic ecosystems, remains unclear. In this study, the abundance and characteristics of MPs were examined in Antarctic krill samples (n = 437) collected from two Antarctic regions. MPs were extracted using an alkali digestion method and analyzed using Fourier-transform infrared spectroscopy. The mean abundance of MPs in Antarctic krill samples from the South Shetland Islands (n = 355) and the South Orkney Islands (n = 82) were 0.29 ± 0.14 and 0.20 ± 0.083 items/individual, respectively. >90 % of MPs found in Antarctic krill were < 150 µm in size. Fibers represented 77 % and 87 % of the MPs in Antarctic krill samples from the South Shetland Islands and the South Orkney Islands, respectively. Black, blue, and red were the predominant colors of MPs in Antarctic krill, accounting for 32 %, 22 %, and 21 % of the total MPs, respectively. Seven polymer compositions were identified for the MPs in Antarctic krill, with the predominance of polyethylene (37 % of total MPs), followed by polypropylene (22 %) and polyester (21 %). To our knowledge, this is the first study to investigate the occurrence of MPs in Antarctic krill samples. The results of this study are important for evaluating the risks of MP exposure in Antarctic krill.

Neonicotinoids (NEOs) are widely applied in agricultural lands and are widespread in different environments, accelerating threats to ecosystems and human health. A number of in vitro/in vivo studies have reported adverse effects of NEOs on mammalian health, but the link between NEO exposure and toxic effects on human liver remains unclear. We randomly recruited 201 participants and quantified eight commercialized NEOs in bile. High frequency and concentration of detection indicate low degradation of human liver on NEOs. The main NEOs are nitenpyram and dinotefuran, which contribute to about 86% of the total residual levels of eight NEOs, due to the highest solubility in bile and are not degraded easily in liver. In contrast, imidacloprid and thiacloprid are major compounds in human blood, according to previous studies, suggesting that individual NEOs behave differently in blood and bile distribution. There was no statistical difference in NEO residues between cancer and non-cancer participants and among the different participant demographics (e.g., age, gender, and body mass index). The serum hematological parameters -bile acid, total bilirubin, cholesterol and alkaline phosphatase -were positively correlated with individual NEO concentrations, suggesting that NEO exposure affects liver metabolism and even enterohepatic circulation. The study first examined the NEO residues in human bile and provided new insights into their bioavailability and hepatoxicity risk.

Inhalation exposure to polycyclic aromatic hydrocarbons (PAHs) from indoor solid fuel combustion poses a high health risk, and PAHs bound to particles with smaller sizes (e.g., PM1.0, aerodynamic diameter ≤ 1.0 µm) should be of particular concern since they can penetrate deep into pulmonary alveoli. However, PAHs bound to PM1.0 was less studied compared with PAHs in total suspended particles or PM2.5. In this study, multiple provincial field measurements were conducted to investigate 28 PAHs bound to PM1.0 in rural Chinese homes. Daily averaged PM1.0-PAH28 concentrations ranged from 27 ng/m3 to 3795 ng/m3 (median: 233 ng/m3) and from 10 ng/m3 to 2978 ng/m3 (median: 87 ng/m3) in indoor and outdoor air, respectively. Higher concentrations were found in northern China in winter due to increased solid fuels consumption for space heating. The ambient pollution was lower during the non-heating season in Eastern China, where clean energy was preferred. Highly toxic congeners were more abundant in indoor air compared with outdoor air. The results of source apportionment revealed that solid fuel combustion was the primary contributor to rural household PM1.0-PAHs, but other sources such as vehicles cannot be overlooked. The transition to cleaner energy can reduce the indoor PM1.0-PAH28 and BaPeq-28 concentrations by 87% and 98%, respectively, and more efficient reduction was observed for highly toxic congeners. The estimated Incremental Lifetime Cancer Risk (ILCR) based on PM1.0-PAH28 ranged from 4.6 × 10-5 to 3.4 × 10-2, far exceeding the acceptable level of 10-6. Over 60% of the ILCR could be attributed to inhalation exposure during childhood and adolescence.

Per-/polyfluoroalkyl substances (PFASs) are widespread in global human blood, and have some toxic effects on liver. However, effects of PFAS exposure on human liver cancer (LC) risk are still not known. In this study, 203 LC patients and 203 controls were recruited, and their serum samples were collected between 2019 and 2021. We determined the residues of 12 PFASs in serum from all participants and quantified their association with LC incidence and tumor markers. PFOS (9.8 ng/mL) had the highest mean concentration in human serum, followed by PFOA (8.3 ng/mL) and 6:2 Cl-PFESA (3.9 ng/mL). We found that concentrations of PFOS and 6:2 Cl-PFESA in human serum were significantly correlated with the levels of alpha fetoprotein (AFP) (ßPFOS = 0.13, 95% confidence interval (CIPFOS): 0.088, 0.17; ß6:2 Cl-PFESA = 0.070, CI6:2 Cl-PFESA: 0.036, 0.10). A positive association of PFOS and 6:2 Cl-PFESA with odds ratios (OR) of LC (ORPFOS = 0.609, CIPFOS: 1.179, 4.029, P = 0.001; OR6:2 Cl-PFESA = 1.844, CI6:2 Cl-PFESA: 1.176, 2.512, P = 0.02) were found, after adjusting for different covariates. Moreover, serum PFOA concentrations were associated with carcinoembryonic antigen (CEA), but their correlation with the LC incidence was not statistically significant. This new finding supports the evidence for the positive associations among PFAS exposure, change of specific tumor marker, and LC risks.

Black carbon (BC) emissions, derived primarily from incomplete fuel combustion, significantly affect the global and regional climate. Mass absorption efficiency (MAE) is one important parameter in evaluating the climate impacts of BC. Here, values and variabilities in the MAE of BC (MAEBC) from real-world residential emissions were investigated from a field campaign covering 163 burning events for different fuel-stove combinations. MAEBC (average: 12 ± 5 m2/g) was normally distributed and varied greatly by 2 orders of magnitude. Statistically significant differences in MAEBC were found for various fuels, while no significant differences were observed among different stoves. The fuel difference explained 72 ± 7% of the MAEBC variation. MAEBC did not correlate with the modified combustion efficiency but positively correlated with the ratio of organic carbon (OC) to elemental carbon (EC) and negatively correlated with char-EC. The OC/EC ratio was not always lower in coal emissions in comparison to biomass burning emissions. Coal- and biomass-burning emissions had different profiles of carbon fractions. Char-EC, OC, OC/EC, and char-EC/soot-EC can explain 68.7% of the MAEBC variation, providing the potential for predicting MAEBC from the carbon fractions, since they are more commonly measured and available.

BACKGROUND: Household air pollution (HAP) from residential combustion considerably affects human health in rural China. Large-scale population migration and rural lifestyle changes during the Spring Festival are supposed to change the household air pollution and health risks; however, limited field study has determined its impacts on HAP and short-term health outcomes. METHODS: A field study was conducted in rural areas of Southern China before and during the Spring Festival to explore the associations between HAP and blood pressure considering different factors such as cooking fuel, heating fuel, and smoking. Stationary real-time PM2.5 monitors were used to measure PM2.5 concentrations of the kitchen, living room, and yard of 156 randomly selected households. Personal exposure to PM2.5 was calculated based on the results of stationary samplers and corresponding time local residents spent in different microenvironments, and one adult resident was recruited of each family for the blood pressure measurement. RESULTS: Both personal exposure to PM2.5 and blood pressures of local residents increased during Spring Festival compared to the days before the holiday. Based on generalized linear model coupled with dominance analysis approach, it was found that personal PM2.5 exposure was positively associated with the factors of population size and the types of cooking and heating fuels with the relative contributions of approximately 82%, and systolic blood pressure (SBP, 100-120 mmHg as normal range for adults) was positively and significantly associated with personal PM2.5 exposures with the relative contribution of 11%. CONCLUSION: The findings in this study demonstrated that Spring Festival can give rise to increase of HAP and hypertension risks, also related to tremendous solid fuel use, suggesting further policy making on promoting cleaner energy in rural areas and more attention on large population migration during national holidays.

Polyhalogenated carbazoles (PHCZs) are an emerging class of persistent, bioaccumulative compounds that are structurally and chemically related to dioxins. They have been detected widely in sediment, river, and soil samples, but their environmental risks are largely unknown. Therefore, seven common PHCZs were tested for their endocrine disrupting potential in silico, in vitro, and in vivo. A dual-luciferase reporter gene assay was used to detect receptor-mediated (agonist or antagonistic) activity (concentration range: 10-9-10-5 M) against the estrogen receptor α (ERα), glucocorticoid receptor α (GRα), and mineralocorticoid receptor (MR). The alterations in the steroidogenesis pathway were investigated in H295R cells. Antagonistic effects against GRα were observed with five PHCZs, along with an increase in the cortisol levels of H295R cells. The most common effect observed was that of the agonistic activity of ERα, with the molecular docking analysis further indicating that hydrogen bonding and hydrophobic interactions may stabilize the interaction between PHCZs and the estrogen receptor binding pocket. In addition, a seven-day exposure of young female rats to three PHCZs (27-BCZ, 3-BCZ, and 36-BCZ) resulted in changes in serum E2 levels, uterine epithelium cell heights, and relative uterus weights. In conclusion, endocrine-disrupting effects, especially the estrogenic effects, were observed for the tested PHCZs. Such adverse effects of PHCZs on humans and wildlife warrant further thorough investigation.

Indoor air contributes significantly to overall exposure, particularly for rural Chinese who often use solid fuels for cooking and/or heating. Unfortunately, overlooked rural indoor air leads to a critical knowledge gap. Simultaneous measurements in the kitchen, living room, and immediately outside of houses using six-channel particle counters were carried out in 18 biomass-burning rural and 3 non-biomass-burning urban households (as a comparison) in winter to characterize dynamic change patterns indoor air pollution and indoor-outdoor relationship. The rural households mainly used wood or crop residues for cooking and heating, while the urban households used pipelined natural gas for cooking and air conditioners for heating. In rural households with significant solid-fuel burning internal sources, the highest concentration was found in the kitchen (101 ±â€¯56 µg/m3), with comparable levels in the living room (99 ±â€¯46 µg/m3) and low levels in outdoor air (91 ±â€¯39 µg/m3). A generally opposite direction of indoor-outdoor exchange was found between the rural and urban households. PM in kitchen air is smaller than that in living rooms and outdoors because solid fuel burning (mainly in rural households) and cooking oil heating (in rural and urban households). Indoor and outdoor PM concentration changed synchronously, with a slight delay in indoor air in urban households but a slight delay in outdoor air in rural households. Cooking, heating, and smoking elevated indoor PM significantly, but different from the cooking activity that produced peaks lasting for about 30 min, emissions from heating created a series of peaks due to frequent disturbance and fuel-feeding and had more significant impacts on the daily average concentration. Distinct indoor-outdoor relationships and dynamic change patterns between the two household categories w/o strong internal biomass burning sources imply that totally different model schemes are needed to quantitatively address indoor air pollution and inhalation exposure.

In recent years, ambient air has been severely contaminated by particulate matters (PMs) and some gas pollutants (nitrogen dioxide (NO2) and sulfur dioxide (SO2)) in China, and many studies have demonstrated that exposure to these pollutants can induce great adverse impacts on human health. The concentrations of the pollutants were much higher in winter than those in summer, and the average concentrations in this studied area were lower than those in northern China. In the comparison between high-resolution emission inventory and spatial distribution of PM2.5, significant positive linear correlation was found. Though the pollutants had similar trends, NO2 and SO2 delayed with 1 h to PM2.5. Besides, PM2.5 had a lag time of 1 h to temperature and relative humidity. Significant linear correlation was found among pollutants and meteorological conditions, suggesting the impact of meteorological conditions on ambient air pollution other than emission. For the 24-h trend, lowest concentrations of PM2.5, NO2, and SO2 were found around 15:00-18:00. In 2015, the population attributable fractions (PAFs) for ischemic heart disease (IHD), cerebrovascular disease (stroke), chronic obstructive pulmonary disease (COPD), lung cancer (LC), and acute lower respiratory infection (ALRI) due to the exposure to PM2.5 in Zhejiang province were 25.82, 38.94, 17.73, 22.32, and 31.14%, respectively. The population-weighted mortality due to PM2.5 exposure in Zhejiang province was lower than the average level of the whole country-China.

Personal exposure to size-segregated particles among rural residents in Shanxi, China in summer, 2011 were investigated using portable carried samplers (N = 84). Household air pollution was simultaneously studied using stationary samplers in nine homes. Information on household fuel types, cooking activity, smoking behavior, kitchen ventilation conditions etc., were also collected and discussed. The study found that even in the summer period, the daily average concentrations of PM2.5 and PM1.0 in the kitchen were as high as 376 ± 573 and 288 ± 397 µg/m3 (N = 6), that were nearly 3 times of 114 ± 81 and 97 ± 77 µg/m3 in the bedroom (N = 8), and significantly higher than those of 64 ± 28 and 47 ± 21 µg/m3 in the outdoor air (N = 6). The personal daily exposure to PM2.5 and PM1.0 were 98 ± 52 and 77 ± 47 µg/m3, respectively, that were lower than the concentrations in the kitchen but higher than the outdoor levels. The mass fractions of PM2.5 in TSP were 90%, 72%, 65% and 68% on average in the kitchen, bedroom, outdoor air and personal inhalation exposure, respectively, and moreover, a majority of particles in PM2.5 had diameters less than 1.0 µm. Calculated time-weighted average exposure based on indoor and outdoor air concentrations and time spent indoor and outdoor were positively correlated but, was ∼33% lower than the directly measured exposure. The daily exposure among those burning traditional solid fuels could be lower by ∼41% if the kitchen was equipped with an outdoor chimney, but was still 8-14% higher than those household using cleaning energies, like electricity and gas. With a ventilator in the kitchen, the exposure among the population using clean energies could be further reduced by 10-24%.

The total concentrations and component profiles of polycyclic aromatic hydrocarbons (PAHs) in ambient air, surface soil and wheat grain collected from wheat fields near a large steel-smelting manufacturer in Northern China were determined. Based on the specific isomeric ratios of paired species in ambient air, principle component analysis and multivariate linear regression, the main emission source of local PAHs was identified as a mixture of industrial and domestic coal combustion, biomass burning and traffic exhaust. The total organic carbon (TOC) fraction was considerably correlated with the total and individual PAH concentrations in surface soil. The total concentrations of PAHs in wheat grain were relatively low, with dominant low molecular weight constituents, and the compositional profile was more similar to that in ambient air than in topsoil. Combined with more significant results from partial correlation and linear regression models, the contribution from air PAHs to grain PAHs may be greater than that from soil PAHs.

Little information exists on emission factors (EFs, quantities of pollutants emitted per unit of fuel consumed) for brick kilns in China, although brick kilns are important emission sources of many air pollutants, and 45% of the world's bricks are produced in China. In this study, EFs of carbon dioxide (CO2), carbon monoxide (CO), sulfur dioxide (SO2), nitrogen oxides (NOx), particulate matters (PMs), black carbon (BC), organic carbon (OC), and polycyclic aromatic hydrocarbons (PAHs) for brick kilns were derived based on field measurements of a total of 18 brick kilns of major types in China. This was the first study to quantify EFs of both stack and fugitive sources based on a modified carbon balance method that was developed for this study. The EFs of most pollutants, especially the incomplete combustion products in fugitive emissions, were much higher than those for stack emissions, indicating a substantial underestimation of total emissions when leakage is not taken into consideration. This novel method can be applied to quantify emissions from other similar sources with both stack and fugitive emissions.

Endocrine-disrupting chemicals (EDCs), which can threaten ecological safety and be harmful to human beings, have been cause for wide concern. There is a high demand for efficient methodologies for evaluating potential EDCs in the environment. Herein an evaluation platform was developed using novel and statistically robust ternary models via different machine learning models (i.e., linear discriminant analysis, classification and regression tree, and support vector machines). The platform is aimed at effectively classifying chemicals with agonistic, antagonistic, or no estrogen receptor (ER) activities. A total of 440 chemicals from the literature were selected to derive and optimize the three-class model. One hundred and nine new chemicals appeared on the 2014 EPA list for EDC screening, which were used to assess the predictive performances by comparing the E-screen results with the predicted results of the classification models. The best model was obtained using support vector machines (SVM) which recognized agonists and antagonists with accuracies of 76.6% and 75.0%, respectively, on the test set (with an overall predictive accuracy of 75.2%), and achieved a 10-fold cross-validation (CV) of 73.4%. The external predicted accuracy validated by the E-screen assay was 87.5%, which demonstrated the application value for a virtual alert for EDCs with ER agonistic or antagonistic activities. It was demonstrated that the ternary computational model could be used as a faster and less expensive method to identify EDCs that act through nuclear receptors, and to classify these chemicals into different mechanism groups.

In this study, paired surface soil and mature wheat grain samples were collected in the cornfields near the large Handan Steel Manufacturer; and the total concentrations and compositional profiles of the parent PAHs were measured, then the spatial distribution characteristics and correlation with total organic carbon fractions in soil were determined. Accordingly, a preliminary source identification was performed, and the association between PAHs in surface soil and wheat grain was briefly discussed. The median concentration of total PAHs in surface soils from the cornfields of Handan was 398.9 ng x g(-1) (ranged from 123.4 ng x g(-1) to 1626.4 ng x g(-1), where around 18% and 10% of all the studied soil samples were over the corresponding quality criteria for total PAHs and B [a] P in soils, respectively. The MMW and HMW species were the main components in the compositional profiles of surface soils. Based on the specific isomeric ratios of PAHs species, coal/biomass combustion and transportation fuel (tail gas) were the dominant mixed sources for the local PAHs emission. The fractions of surface soil TOC had significant positive correlations with the total PAHs and also with the individual components with different rings. In addition, the median concentration of total PAHs in wheat grains collected in the cornfields near the Handan Steel Manufacture was 27.0 ng x g(-1) (ranged from 19.0-34.0 ng x g(-1)). The levels in wheat grains were not high, and lower than the related hygienic standards of food proposed by EU and China. The LMW and MMW PAHs with 2 to 4 rings occupied a larger proportion, more than 84% of the total PAHs, which was largely different from the component profiles in surface soils. This situation suggested that the local sources of PAHs in wheat grains may originate not only from surface soil via root absorption and internal transportation, but also from ambient air through dry and wet deposition on the leaf surface (stoma).

There is wide concern about polycyclic aromatic hydrocarbons (PAHs) because of their carcinogenic and mutagenic potential. The coking industry is an important source of PAHs. In this study, 36 arable soil samples, a sensitive medium from the perspective of food safety and health, were collected from one of the largest coke production bases in China. The concentration of total 21 PAHs ranged from 294 to 1665 ng g(-1), with a mean of 822±355 ng g(-1). Approximately 60% of the soil samples were heavily polluted with the level higher than 600 ng g(-1). Particularly high abundances of high molecular weight PAHs were found, and the calculated BaPeq was as high as 54.3 ng g(-1). Soil PAH levels were positively correlated with soil organic matter content. The soil PAHs were from complex mixture sources, and high-temperature pyrogenic sources were most likely responsible for the heavy PAH contamination. Effective control strategies and probable remediation approaches should be proposed to improve soil quality.

The health impacts of polycyclic aromatic hydrocarbons (PAHs), the most concerning organic pollutants, depend not only on the locations and strengths of emission sources, but also on individual susceptibility. Moreover, trans-boundary transport makes them a global concern. In this study, a comprehensive analysis of the global health impacts of polycyclic aromatic hydrocarbons (PAHs) in ambient air is presented. Model resolution is critical in exposure modelling. Globally, incremental lifetime lung cancer risk (ILCR) induced by ambient PAH exposure is 3.1 × 10(-5). If the individual susceptibility was not taken into consideration, the overall risk would be underestimated by 55% and the proportion of highly vulnerable population would be underestimated by more than 90%. Emphasizing on individual susceptibility, our study provides an instrumental revision of current risk assessment methodology. In terms of lung cancer risk, the most important sources are combustion of biomass fuels (40%) and fossil fuels (14%) in the residential/commercial sector, coke (13%) and aluminium (12%) production, and motor vehicles (9%). PAHs can travel long distance globally especially within the Eurasian continent. Still, the risk is dominantly contributed by local.

In vitro tests using human adenocarcinomic alveolar epithelial cell line A549 and small mouse monocyte-macrophage cell line J774A.1 were conducted to test toxicity of six PM (particulate matter) samples from Beijing. The properties of the samples differ significantly. The production of inflammatory cytokine (TNF-α for J774A.1) and chemokine (IL-8 for A549) and the level of intracellular reactive oxygen species (ROS) were used as endpoints. There was a positive correlation between water soluble organic carbon and DTT-based redox activity. Both cell types produced increased levels of inflammatory mediators and had higher level of intracelllar ROS, indicating the presence of PM-induced inflammatory response and oxidative stress, which were dose-dependent and significantly different among the samples. The releases of IL-8 from A549 and TNF-α from J774A.1 were significantly correlated to PM size, Zeta potential, endotoxin, major metals, and polycyclic aromatic hydrocarbons. No correlation between ROS and these properties was identified.
