BACKGROUND: Selection for sound conformation has been widely used as a primary approach to reduce lameness and leg weakness in pigs. Identification of genomic regions that affect conformation traits would help to improve selection accuracy for these lowly to moderately heritable traits. Our objective was to identify genetic factors that underlie leg and back conformation traits in three Danish pig breeds by performing a genome-wide association study followed by meta-analyses. METHODS: Data on four conformation traits (front leg, back, hind leg and overall conformation) for three Danish pig breeds (23,898 Landrace, 24,130 Yorkshire and 16,524 Duroc pigs) were used for association analyses. Estimated effects of single nucleotide polymorphisms (SNPs) from single-trait association analyses were combined in two meta-analyses: (1) a within-breed meta-analysis for multiple traits to examine if there are pleiotropic genetic variants within a breed; and (2) an across-breed meta-analysis for a single trait to examine if the same quantitative trait loci (QTL) segregate across breeds. SNP annotation was implemented through Sus scrofa Build 10.2 on Ensembl to search for candidate genes. RESULTS: Among the 14, 12 and 13 QTL that were detected in the single-trait association analyses for the three breeds, the most significant SNPs explained 2, 2.3 and 11.4% of genetic variance for back quality in Landrace, overall conformation in Yorkshire and back quality in Duroc, respectively. Several candidate genes for these QTL were also identified, i.e. LRPPRC, WRAP73, VRTN and PPARD likely control conformation traits through the regulation of bone and muscle development, and IGF2BP2, GH1, CCND2 and MSH2 can have an influence through growth-related processes. Meta-analyses not only confirmed many significant SNPs from single-trait analyses with higher significance levels, but also detected several additional associated SNPs and suggested QTL with possible pleiotropic effects. CONCLUSIONS: Our results imply that conformation traits are complex and may be partly controlled by genes that are involved in bone and skeleton development, muscle and fat metabolism, and growth processes. A reliable list of QTL and candidate genes was provided that can be used in fine-mapping and marker assisted selection to improve conformation traits in pigs.

When exposed to small amounts of bacterial endotoxin, matrices of cholesterol crystals, but not cholesterol itself, primed human monocytes/macrophages to a highly augmented (>10-fold) production of inflammatory tumor necrosis factor-α. Priming also sensitized the cells, as 10- to 100-fold lower levels of endotoxin were needed for TNF-α production equivalent to that of unprimed cells. The pro-inflammatory effect was selective as endotoxin-induced production of other pro-inflammatory cytokines was unaffected while production of anti-inflammatory interleukin-10 was diminished. These findings suggest that cholesterol matrix formation may play a pathogenic role in atherosclerotic inflammation, and they indicate a mechanism by which bacteria and/or bacterial products may play a role in processes leading to arteriosclerosis.

Immulina®, a commercial extract of Arthrospira (Spirulina) platensis is a potent activator of THP-1 monocytes and CD4+ T cells IN VITRO and enhances several immunological functions in mice. We further characterized Immulina® by determining that Braun-type lipoproteins are responsible for a major portion of the IN VITRO monocyte activation exhibited by this material. In order to understand the effect of Immulina® on NK cell activity, a pilot study was conducted on ten healthy North American individuals who supplemented their diet with Immulina® (400 mg/day) for seven days. We observed a 40% average increase in the killing of K562 tumor cells by NK cells (p < 0.01) after Immulina® supplementation. In a separate placebo-controlled, crossover study involving 11 healthy Danish subjects, we observed increased mRNA expression of the NK cell marker NKG2D by 37% (p = 0.02) and by 55% (p = 0.0003) after administration of Immulina® (200 mg and 400 mg per day, respectively) for seven days. The mRNA expression of the NK- and T-cell marker perforin increased by 75% (p = 0.008) after administration of 400 mg Immulina® per day. Both markers displayed significant dose-dependent effects (p = 0.0003 and p = 0.02, respectively). The ratio between CD56 (bright) and CD56 (dim) NK cells was not affected by Immulina® administration. In summary, two independent studies showed enhancement of NK cell activity following administration of Immulina® for seven days.

INTRODUCTION: According to nurses' assessment, physician-nurse collaboration is problematic. The aim of the study was to investigate whether nurses believe physicians' appearances is significant for their ability to collaborate. MATERIAL AND METHODS: This is a single-blinded, quasi-experimental intervention study. All physicians included will undergo a total plastic surgical body makeover. Half of the included physicians are to be operated with a view to enhance their physical features, half to weaken their physical features. Multifactorial ANOVA analysis will be used to secure the division. The Jefferson Scale of Attitudes Toward Physician-Nurse Collaboration will be used for baseline and follow-up study of the nurses' assessment. RESULTS: Due to ethical considerations, researchers had difficulties finding surgeons prepared to perform procedures aiming at weakening the physicians' physical features. Furthermore, it proved difficult to find physicians willing to undergo surgery with a view to weaken their physical features. Consequently, the study was never realized. Additionally, the study experienced difficulties raising the funding needed to cover surgery costs for all included physicians. CONCLUSION: We conclude that physicians often have difficulties collaborating with nurses, regardless of their appearance. It seems that physicians are not interested in improving the collaboration with nurses, and further studies are therefore needed to improve physician-nurse collaboration. We recommend in-depth interviews to achieve harmonic interaction leading to a prolific and close future collaboration.

BACKGROUND: Carcinoma in situ (CIS) is believed to be a precursor of invasive bladder cancer. Identification of CIS is a valuable prognostic factor since radical treatment strategies can be offered these patients before the disease becomes invasive. METHODS: We developed a pathway based classifier approach to predict presence or absence of CIS in patients suffering from non muscle invasive bladder cancer. From Ingenuity Pathway Analysis we considered four canonical signalling pathways (p38 MAPK, FGF, Calcium, and cAMP pathways) with most coherent expression of transcription factors (TFs) across samples in a set of twenty-eight non muscle invasive bladder carcinomas. These pathways contained twelve TFs in total. We used the expression of the TFs to predict presence or absence of CIS in a Leave-One-Out Cross Validation classification. RESULTS: We showed that TF expression levels in three pathways (FGF, p38 MAPK, and calcium signalling) or the expression of the twelve TFs together could be used to predict presence or absence of concomitant CIS. A cluster analysis based on expression of the twelve TFs separated the samples in two main clusters: one branch contained 11 of the 15 patients without concomitant CIS and with the majority of the genes being down regulated; the other branch contained 10 of 13 patients with concomitant CIS, and here genes were mostly up regulated. The expression in the CIS group was comparable to the expression of twenty-three patients suffering from muscle-invasive bladder carcinoma. Finally, we validated our results in an independent test set and found that prediction of CIS status was possible using TF expression of the p38 MAPK pathway. CONCLUSION: We conclude that it is possible to use pathway analysis for molecular classification of bladder tumors.

UVB-TL01 is proven to be efficient in whole body treatment for several chronic inflammatory dermatoses. Chronic hand dermatoses of different and often mixed genesis represent a great problem in clinical practice. We have performed a prospective, open study with patients suffering from psoriasis, eczema or pustulosis of the hands treated with hand UVB-TL01. Thirty patients (23 women and 7 men) were included, mean age 43 years (range 25-68 years). The psoriatics were the best responders, as 9 of 11 were improved or much improved after 20-38 treatments, compared to 11 of 16 patients with eczema (11-31 treatments). One of three patients with palmar pustulosis experienced improvement after 23-27 treatments. UVB-TL01 seems to be helpful in the majority of patients with chronic inflammatory diseases of the hands. More extensive studies with regard to proper dosing and length of treatment period ought to be carried out.

When sewage sludge is applied to arable land, linear alkylbenzene sulfonate (LAS) is released into the environment. In soils, LAS has been shown to impede microbial processes, such as bacterial iron reduction. The aim of the present study was to quantify LAS adsorption and desorption to agricultural soils and iron oxides and relate this to the inhibition of microbial iron reduction. Two agricultural soils were used, namely, Askov (coarse sandy loam soil) and Lundgaard (coarse sandy soil). In both soils, LAS inhibited microbial iron reduction even at low LAS concentrations with 10% effect concentrations of 6 to 7 and 26 to 32 mg LAS/kg dry-weight soil for Lundgaard and Askov soil, respectively. The sorption isotherms showed that sorption of LAS to iron oxides was 10 to 100 times stronger than sorption to the agricultural soils. Also, it appeared that at low LAS concentrations (< 10 mg/kg dry-wt soil), Lundgaard soil adsorbed approximately 10 times more LAS than Askov soil. Thus, the inhibitory effect of LAS on microbial iron reduction was highest in the Lundgaard soil, which exhibited both the strongest sorption and the lowest desorption of the two soils. A possible hypothesis to explain this correlation was that LAS toxicity toward bacterial iron reduction was, at least partly, caused by LAS adsorbed to iron oxides, which could interfere with transfer of electrons between the bacteria and their respiratory electron acceptor.
