RESUMO

Background: Agmination and/or satellitosis in pigmented blue lesions is a phenomenon rarely mentioned in the literature and not well known. This phenomenon can be expressed by several benign and malignant pigmented blue lesions, such as blue nevi, Spitz nevi, melanocytoma and melanoma. On this spectrum, dermoscopy, reflectance confocal microscopy (RCM) and dynamic Optical coherence tomography (D-OCT) represent non-invasive imaging technologies, which may help clinicians in the diagnosis of melanoma and non-melanoma skin cancers in daily clinical practice. Methods: Currently, in the literature there is a lack of new data about agminated blue lesions and blues lesions with satellitosis, as well as the lack of a recent and updated review of the literature about this topic. Therefore, considering that clinicians must be confident with the diagnosis of these rare skin lesions, we decided to carry out this work. Results: In this paper, four new cases of agminated pigmented cutaneous lesions were described. Moreover, a review of the current literature on this topic was performed. Conclusions: A clinical-pathological correlation is often needed to reach a correct diagnosis; currently, dermoscopy and non-invasive diagnostic techniques, such as reflectance confocal microscopy and optical coherence tomography, due to the depth of these skin lesions in the dermis, can only make a partial and limited contribution.

RESUMO

Primary myelofibrosis (PMF) is a chronic myeloproliferative neoplasm (MPN) characterized by progressive bone marrow sclerosis, extra-medullary hematopoiesis, and possible transformation to acute leukemia. In the last decade, the molecular pathogenesis of the disease has been largely uncovered. Particularly, genetic and genomic studies have provided evidence of deregulated oncogenes in PMF as well as in other MPNs. However, the mechanisms through which transformation to either the myeloid or lymphoid blastic phase remain obscure. Particularly, it is still debated whether the disease has origins in a multi-potent hematopoietic stem cells or instead in a commissioned myeloid progenitor. In this study, we aimed to shed light upon this issue by using next generation sequencing (NGS) to study both myeloid and lymphoid cells as well as matched non-neoplastic DNA of PMF patients. Whole exome sequencing revealed that most somatic mutations were the same between myeloid and lymphoid cells, such findings being confirmed by Sanger sequencing. Particularly, we found 126/146 SNVs to be the e same (including JAK2V617F), indicating that most genetic events likely to contribute to disease pathogenesis occurred in a non-commissioned precursor. In contrast, only 9/27 InDels were similar, suggesting that this type of lesion contributed instead to disease progression, occurring at more differentiated stages, or maybe just represented "passenger" lesions, not contributing at all to disease pathogenesis. In conclusion, we showed for the first time that genetic lesions characteristic of PMF occur at an early stage of hematopoietic stem cell differentiation, this being in line with the possible transformation of the disease in either myeloid or lymphoid acute leukemia.

RESUMO

Polycyclic Aromatic Hydrocarbons (PAHs) are an important class of pollutants which mostly come out from incomplete combustion of organic materials including fossil fuels. For this reason, they are often found at high concentrations in cities, contaminating air with their gas and particle phase. While European Union policies try to reduce their concentrations, huge efforts are still devoted to mitigate the pollution by PAHs. One such measure of mitigation is the use of plants in capturing PAHs and other chemicals due to the ability of leaves in filtering both gas and particle fractions. In this work thirteen conifer species among those frequently living in temperate and Mediterranean cities were selected and concentrations of 16 PAHs were measured in needles. The data for spruce were used to back-calculate air concentrations of PAHs which were later used to obtain specific equations of plant-air partition coefficient (KPA). The results showed that the difference in total PAH uptake between the most (Pinus pinaster) and the least (Picea abies) efficient tree was close to an order of magnitude. Looking to the capability of the different species in enriching the particulate matter (PM) associated fraction of PAHs on their needles, Pinus pinaster was instead the least important (3.4% of total PAHs), while Cupressus lusitanica was the most important (34% of total PAHs). The new KPA equations can be used to fine tune the PAH uptake of a specific amount of plant biomass on air concentration reduction.

RESUMO

Estimating KDOC (dissolved organic carbon/water partition coefficient) and DDOC (dissolved organic carbon/water distribution coefficient) of neutral and ionizable organic chemicals is a crucial task for assessing mobility, modelling transport, environmental fate of a variety of chemicals and for evaluating their bioavailability in terrestrial and aquatic environments. A critical literature search of reliability-selected KDOC and DDOC values was performed to setup novel predictive relationships for KDOC and DDOC of neutral and ionizable organic chemicals. This goal was pursued by using: 1) LSER (linear solvation energy relationship) models to predict KDOC for neutral chemicals using Abraham solute parameters calculated for different DOC sources (all DOC sources together, soil porewater, surface water, wastewater and Aldrich humic acid (HA)); 2) linear regressions for predicting DDOC of organic acids from the octanol/water partition coefficient (Log KOW or Log P) and the dissociation constant (pKa), accounting separately for the contribution of the neutral and ionic fraction. The proposed models predicted Log KDOC and DDOC values within a root mean square deviation (RMSD) generally smaller than 0.3 log units.

RESUMO

The partitioning tendency of chemicals, in this study pesticides in particular, into different environmental compartments depends mainly on the concurrent relevance of the physico-chemical properties of the chemical itself. To rank the pesticides according to their distribution tendencies in the different environmental compartments we propose a multivariate approach: the combination, by principal component analysis, of those physico-chemical properties like organic carbon partition coefficient (Koc), n-octanol/water partition coefficient (Kow), water solubility (Sw), vapour pressure and Henry's law constant (H) that are more relevant to the determination of environmental partitioning. The resultant macrovariables, the PC1 and PC2 scores here named leaching index (LIN) and volatality index (VIN), are proposed as preliminary environmental partitioning indexes in different media. These two indexes are modeled by theoretical molecular descriptors with satisfactory predictive power. Such an approach allows a rapid pre-determination and screening of the environmental distribution of pesticides starting only from the molecular structure of the pesticide, without any a priori knowledge of the physico-chemical properties.
