RESUMO

Seaweed polysaccharides can be used for protective skin photoaging which is caused by long-term exposure to ultraviolet B (UVB). In this study, a multifunctional composite hydrogel (FACP5) is prepared using sulfated galactofucan polysaccharides, alginate oligosaccharides as active ingredients, and polyacrylonitrile modified κ-Carrageenan as substrate. The properties of FACP5 show that it has good water retention, spreadability, and adhesion. The antiphotoaging activity is evaluated in vitro and in vivo. In vitro experiments demonstrate that the components of FACP5 exhibit good biocompatibility, antioxidant, and anti-tyrosinase activities, and could reduce the cell death rate induced by UVB. In vivo experiments demonstrate that, compared with the mice skin in model group, the skin water content treated with FACP5 increases by 29.80%; the thicknesses of epidermis and dermis decrease by 53.56% and 43.98%, respectively; the activities of catalase and superoxide dismutase increase by 1.59 and 0.72 times, respectively; the contents of interleukin-6 and tumor necrosis factor-α decrease by 19.21% and 17.85%, respectively; hydroxyproline content increases by 32.42%; the expression level of matrix metalloproteinase-3 downregulates by 42.80%. These results indicate that FACP5 has skin barrier repairing, antioxidant, anti-inflammatory, and inhibiting collagen degradation activies, FACP5 can be used as a skin protection remedy for photoaging.

BACKGROUND: Glutathione S-transferase (GST) is a crucial enzyme for metabolism, detoxification, and stress resistance in organisms. Many GSTs have been identified in seaweeds, but the isolation and functional analysis of GSTs in Saccharina japonica have not been completed. RESULT: In this study, a total of 32 SjGST genes, localized on 10 scaffolds and 6 contigs, were identified and categorized into three groups. Most of these SjGSTs were presumed to be distributed in the cytoplasm. Tandem duplication had a significant influence on the expansion of the SjGST gene family. Functional analysis of cis-acting elements in the promoter regions demonstrated that SjGSTs enhance the stress resistance of the kelp. Quantitative real-time PCR tests confirmed that SjGSTs positively influence S. japonica sporophytes under stress from low salinity, drought, and high temperature. Recombinant yeast tests further affirmed the role of SjGSTs in stress resistance; SjGSTs improved the growth rate of recombinant yeast under 1.5 M NaCl or 8 mM H2O2. Analysis of biochemical parameters indicated that the optimum temperatures for SjGST20 and SjGST22 were 20 °C, and the optimum pH values were 7.0 and 8.0 for SjGST20 and SjGST22, respectively. The Km values for the substrate 1-chloro-2,4-dinitrobenzene (CDNB) were 2.706 mM and 0.674 mM and were 6.146 mM and 3.559 mM for the substrate glutathione (GSH) for SjGST20 and SjGST22, respectively. CONCLUSION: SjGSTs are important stress resistant genes in S. japonica. This research results will enhance our understanding the function of GSTs in brown seaweeds, and explained its functional roles in stress resistance in marine environments.

Inflammation is a complicated host-protective response to stimuli and toxic conditions, and is considered as a double-edged sword. A sulfated Saccharinajaponica polysaccharide (LJPS) with a sulfate content of 9.07% showed significant inhibitory effects against lipopolysaccharide (LPS)-induced inflammation in RAW 264.7 macrophage cells and zebrafish. Its chemical and structural properties were investigated via HPLC, GC, FTIR, and NMR spectroscopy. In vitro experiments demonstrated that LJPS significantly inhibited the generation of nitric oxide (NO) and prostaglandin E2 (PGE2) via the downregulation of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) expression and suppressed pro-inflammatory cytokines tumor necrosis factor (TNF)-α and interleukin (IL)-1ß production via the nuclear factor-kappa B (NF-κB) and mitogen-activated protein kinase (MAPK) signal pathways in LPS-induced RAW 264.7 cells. Moreover, LJPS showed strong protective effects against LPS-induced inflammatory responses in zebrafish, increasing the survival rate, reducing the heart rate and yolk sac edema size, and inhibiting cell death and the production of intracellular reactive oxygen species (ROS) and NO. Its convenience for large-scale production and significant anti-inflammatory activity indicated the potential application of LJPS in functional foods, cosmetics, and pharmaceutical industries.

A fucose-rich fucoidan was purified from brown seaweed Saccharina japonica, of which the UVB protective effect was investigated in vitro in keratinocytes of HaCaT cells and in vivo in zebrafish. The intracellular reactive oxygen species levels and the viability of UVB-irradiated HaCaT cells were determined. The results indicate that the purified fucoidan significantly reduced the intracellular reactive oxygen species levels and improved the viability of UVB-irradiated HaCaT cells. Furthermore, the purified fucoidan remarkably decreased the apoptosis by regulating the expressions of Bax/Bcl-xL and cleaved caspase-3 in UVB-irradiated HaCaT cells in a dose-dependent manner. In addition, the in vivo UV protective effect of the purified fucoidan was investigated using a zebrafish model. It significantly reduced the intracellular reactive oxygen species level, the cell death, the NO production, and the lipid peroxidation in UVB-irradiated zebrafish in a dose-dependent manner. These results suggest that purified fucoidan has a great potential to be developed as a natural anti-UVB agent applied in the cosmetic industry.

Tic20 is an important translocon protein that plays a role in protein transport in the chloroplast. The sequence of Tic20 was determined in the lower brown alga Saccharina japonica. Structural analysis of SjTic20 revealed a noncanonical structure consisting of an N-terminal non-cyanobacterium-originated EF-hand domain (a helix-loop-helix structural domain) and a C-terminal cyanobacterium-originated Tic20 domain. Subcellular localization and transmembrane analysis indicated that SjTic20 featured an "M"-type Nin-Cin-terminal orientation, with four transmembrane domains in the innermost membrane of the chloroplast in the microalga Phaeodactylum tricornutum, and the EF-hand domain was entirely extruded into the chloroplast stroma. Our study provides information on the structure, localization, and topological features of SjTic20, and further functional analysis of SjTic20 in S. japonica is needed.

Parkinson's disease (PD) is a neurodegenerative movement disorder that is caused by a selective loss of dopaminergic neurons. Current PD treatments provide symptomatic relief but do not prevent or decelerate disease progression. Previous studies have suggested that acetylated and phosphorylated porphyran, derived from Porphyra, produces a neuroprotective effect against 6-OHDA-induced damage. Due to its antioxidant and neuroprotective potential, this study evaluates whether oligo-porphyran (OP) could be beneficial in an experimental model of PD in mice. The drug 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) was intraperitoneally injected (20 mg/kg body weight) for seven days to simulate PD, followed by OP administration. We found that the behavioral deficits in spontaneous motor activity, latency to descend in a pole test, and suspension in a traction test were ameliorated, and excessive dopamine (DA) metabolism was suppressed after OP treatment. Additionally, we found that OP protected dopaminergic neurons by preventing MPTP-induced decreases in dopaminergic transporter and tyrosine hydroxylase protein levels. We speculated whether OP regulates a signaling pathway that affects the behavioral changes seen in PD mice. In this study, the PI3K/Akt/Bcl-2 pathway was detected. Our results demonstrate that OP increased the phosphorylation of PI3K/Akt/GSK-3ß and inhibited the activation of caspase-3 and poly (ADP-ribose) polymerase, with changes in the Bax/Bcl-2 ratio. These results showed that OP might promote DA neuron survival in vivo by regulating the PI3K/Akt/Bcl-2 pathway, thereby ameliorating the neurobehavioral deficits in a PD mouse model and suggesting OP as a neuroprotective treatment for PD.

Phlorotannins are a group of complex polymers of phloroglucinol (1,3,5-trihydroxybenzene), which are unique compounds from marine brown algae. In our present study, a procedure for extraction and enrichment of phlorotannins from S. fusiforme with highly antioxidant potentials was established. After comparison of different extraction methods, the optimal extraction conditions were established as follows. The freeze-dried seaweed powder was extracted with 30% ethanol-water solvent with a solid/liquid ratio of 1:5 at temperature of 25 °C for 30 min. After extraction, the phlorotannins were fractioned by different solvents, among which the ethyl acetate fraction exhibited both the highest total phlorotannin content (88.48 ± 0.30 mg PGE/100 mg extract) and the highest antioxidant activities. The extracts obtained from these locations were further purified and characterized using a modified UHPLC-QQQ-MS method. Compounds with 42 different molecular weights were detected and tentatively identified, among which the fuhalol-type phlorotannins were the dominant compounds, followed by phlorethols and fucophlorethols with diverse degree of polymerization. Eckol-type phlorotannins including some newly discovered carmalol derivatives were detected in Sargassum species for the first time. Our study not only described the complex phlorotannins composition in S. fusiforme, but also highlighted the challenges involved in structural elucidation of these compounds.

A bacterial strain capable of hydrolyzing sulfate ester bonds of p-nitrophenyl sulfate (pNPS) and agar was isolated from the coast area of Qingdao, China. It was identified as Marinomonas based on its 16S rRNA gene sequence and named as Marinomonas sp. FW-1. An arylsulfatase with a recovery of 13 % and a fold of 12 was purified to a homogeneity using ion exchange and gel filtration chromatographies. The enzyme was composed of a single polypeptide chain with the molecular mass of 33 kDa estimated using SDS-PAGE. The optimal pH and temperature of arylsulfatase were pH 9.0 and 45, respectively. Arylsulfatase was stable over pH 8-11 and at temperature below 55 °C. The K m and V max of this enzyme for the hydrolysis of pNPS were determined to be 13.73 and 270.27 µM/min, respectively. The desulfation ratio against agar from red seaweed Gelidium amansii and Gracilaria lemaneiformis were 86.11 and 89.61 %, respectively. There was no difference between the DNA electrophoresis spectrum on the gel of the arylsulfatase-treated G. amansii agar and that of the commercial agarose. Therefore, this novel alkaline arylsulfatase might have a great potential for application in enzymatic conversion of agar to agarose.

Photosynthetic stramenopile have chloroplasts of secondary endosymbiotic origin and are significant as aquatic primary productivity and biomass production. In marine environments, many photosynthetic stramenopiles utilize blue light to regulate growth, development, and organelle movement. Aureochrome (AUREO) is a new type blue light photoreceptor specific in photosynthetic stramenopiles. Previously, several AUREO orthologs were reported in genomes of stramenopile members, but the full-length cDNA sequences were completed only in Vaucheria frigida (Xanthophyceae), Fucus distichus (Phaeophyceae), and Ochromonas danica (Chrysophyceae). In this study, the full-length cDNA of AUREO from Saccharina japonica (designated as SjAUREO) was isolated based on homologous cloning and the rapid amplification of cDNA ends (RACE). It characterized by the full length of 1,013 bp with an open reading frame of 612 bp, which encoded a polypeptide of 203 amino acids with predicted molecular weight of 23.08 kDa and theoretical isoelectric point of 7.63. The deduced amino acid sequence of SjAUREO contained one N-terminal basic region/leucine zipper (bZIP) transcription regulation domain and a single light-, oxygen-, or voltage-sensitive (LOV) domain near the C-terminus. Homologous analysis showed that SjAUREO shared 40-92 % similarities with those of other photosynthetic stramenopiles. Phylogenetic analysis revealed close phylogenetic affinity between SjAUREO and AUREO4 of brown alga Ectocarpus siliculosus. Real-time PCR detection revealed that the SjAUREO transcription was markedly increased under BL exposure and dramatically upregulated in the 1-month juvenile sporophyte than those in the 2 and 3-month materials, which indirectly reflected the SjAUREO associated with the BL-mediated photomorphogenesis during the growth and early development of juvenile sporophytes. In vitro expression showed one distinct band existed at ∼27 kDa, and western blot detection proved that it was positive to the anti-His antibody with high specificity. Our results enriched the knowledge of AUREO properties in S. japonica and provided clues to explore the mechanisms underlying diverse physiological responses mediated by BL photoreceptors AUREO in the photosynthetic stramenopiles.

In this study, a full-length cytosolic heat shock protein 70 complementary DNA (cDNA) of Laminaria japonica (designated as LJHsp70) was obtained by reverse transcriptase-polymerase chain reaction (RT-PCR) coupled with rapid amplification of cDNA ends. The full length of LJHsp70 cDNA was 2,918 bp, with a 5' untranslated region of 248 bp, a 3' untranslated region of 696 bp, and an open reading frame of 1,974 bp encoding a polypeptide of 657 amino acids with an estimated molecular mass of 72.03 kDa and an estimated isoelectric point of 4.97. There was highly repeated sequence of CAA in 5' untranslated region of LJHsp70. The result of phylogenetic tree of Hsp70s, the BLAST program, analysis and cytosolic Hsp70-specific motif of LJHsp70 verified that the cloned LJHsp70 belonged to cytosolic Hsp70 family. Three typical Hsp70 signature motifs were detected in LJHsp70 by InterPro analysis. Under different stress conditions, messenger RNA (mRNA) expression levels of LJHsp70 were quantified by quantitative RT-PCR. To L. japonica sporophytes kept in different temperatures for 1 h, the expression level of LJHsp70 at 30 degrees C was highest and twofold higher than that at 10 degrees C. To L. japonica sporophytes kept at 25 degrees C for different times, the mRNA expression level of LJHsp70 reached a maximum level after 7 h and then dropped progressively. The expression level of LJHsp70 at 0 or 5 per thousand salt concentration for 2 h was twofold higher than that at 30 per thousand salt concentration for 2 h. The results showed that LJHsp70 may be a kind of potential biomarker used to monitor environment conditions.
