The European eel (Anguilla anguilla) is a critically endangered species. The impact of environmental contamination on this species has been highlighted as contributing to the decline in recruitment. The Mar Menor hypersaline coastal lagoon (SE Spain) is one of the most productive fisheries of European eel in Europe, making it a critical habitat for species conservation. The present study aimed to provide an initial overview of the impact of organic chemical contaminants on the European eel and the potential sublethal effects of chemical pollution on pre-migrating eels in this hypersaline habitat. We investigated muscle bioaccumulation of main persistent and hazardous organic contaminants (including some current-use pesticides) and genotoxicity, neurotoxicity, and xenobiotic detoxification system responses. The findings show that lagoon eels were exposed to high levels of legacy organochlorine contaminants, recently banned pesticides (chlorpyrifos), and some emerging chemicals. Some individuals surpassed the maximum levels of CBs authorized by the European Commission for human consumption. In this species, residuals of chlorpyrifos, pendimethalin, and chlorthal dimethyl have been reported for the first time. This field study provides relevant data to stock management and human health consumption and provides the first biomarker responses in European eel under permanent hypersaline conditions. Furthermore, the high frequency of micronuclei in peripheral erythrocytes of lagoon eels indicates sublethal genotoxic effects on the organism. Overall, the European eels growing and maturing in the Mar Menor lagoon are exposed to toxic and carcinogenic chemicals. The lack of seafood safety regulations for human consumption for some legacy chemicals that were measured in high concentrations in our study requires special action. Further biomonitoring and research are recommended to protect the animal, public, and environmental health.

A 9-year-old boy diagnosed with acute myeloblastic leukemia and undergoing chemotherapy, was admitted with febrile neutropenia. During his admission, several violaceous plaques appeared on the upper extremities and anterior left hemithorax, which worsened and acquired a necrotic center. We performed a biopsy and histology showed a cutaneous infarction at the dermoepidermal and subcutaneous level. We observed abundant wide hyphae with right-angled branching and a culture isolated Rhizopus oryzae. A plastic surgery consultant performed a surgical debridement of the lesions and treatment was started with intravenous amphotericin B. The patient did well on treatment and after almost a month of hospitalization, he was discharged with oral posaconazole. Mucormycosis is an opportunistic fungal infection associated with immunosuppression, particularly involving prematurity and hematological diseases in the pediatric age group. Multiple lesions, as in our case, are infrequent. The clinical presentation is variable. Direct smear or histological observation is the quickest diagnostic technique whereas culture is the most definitive. The combination of surgical debridement and amphotericin B is the treatment with the highest survival rates.

BACKGROUND AND AIMS: Patients with colonic inflammatory bowel disease (IBD) have a high risk of colorectal cancer (CRC). Current guidelines recommend endoscopic surveillance, yet epidemiological studies show poor compliance. The aims of our study were to analyse adherence to endoscopic surveillance, its impact on advanced colorectal lesions, and risk factors of non-adherence. METHODS: A retrospective multicentre study of IBD patients with criteria for CRC surveillance, diagnosed between 2005 and 2008 and followed up to 2020, was performed. Following European guidelines, patients were stratified into risk groups and adherence was considered when surveillance was performed according to the recommendations (±1 year). Cox-proportional regression analyses were used to compare the risk of lesions. p-values below 0.05 were considered significant. RESULTS: A total of 1031 patients (732 ulcerative colitis, 259 Crohn's disease and 40 indeterminate colitis; mean age of 36 ± 15 years) were recruited from 25 Spanish centres. Endoscopic screening was performed in 86% of cases. Adherence to guidelines was 27% (95% confidence interval, CI = 24-29). Advanced lesions and CRC were detected in 38 (4%) and 7 (0.7%) patients respectively. Adherence was associated with increased detection of advanced lesions (HR = 3.59; 95% CI = 1.3-10.1; p = 0.016). Risk of delay or non-performance of endoscopic follow-up was higher as risk groups increased (OR = 3.524; 95% CI = 2.462-5.044; p < 0.001 and OR = 4.291; 95%CI = 2.409-7.644; p < 0.001 for intermediate- and high- vs low-risk groups). CONCLUSIONS: Adherence to endoscopic surveillance allows earlier detection of advanced lesions but is low. Groups at higher risk of CRC are associated with lower adherence.

Tissue engineering (TE) is a therapeutic option within regenerative medicine that allows to mimic the original cell environment and functional organization of the cell types necessary for the recovery or regeneration of damaged tissue using cell sources, scaffolds, and bioreactors. Among the cell sources, the utilization of mesenchymal cells (MSCs) has gained great interest because these multipotent cells are capable of differentiating into diverse tissues, in addition to their self-renewal capacity to maintain their cell population, thus representing a therapeutic alternative for those diseases that can only be controlled with palliative treatments. This review aimed to summarize the state of the art of the main sources of MSCs as well as particular characteristics of each subtype and applications of MSCs in TE in seven different areas (neural, osseous, epithelial, cartilage, osteochondral, muscle, and cardiac) with a systemic revision of advances made in the last 10 years. It was observed that bone marrow-derived MSCs are the principal type of MSCs used in TE, and the most commonly employed techniques for MSCs characterization are immunodetection techniques. Moreover, the utilization of natural biomaterials is higher (41.96%) than that of synthetic biomaterials (18.75%) for the construction of the scaffolds in which cells are seeded. Further, this review shows alternatives of MSCs derived from other tissues and diverse strategies that can improve this area of regenerative medicine.

BACKGROUND: Infection in acute pancreatitis will worsen the disease prognosis. The aim of our study was to analyze the role of procalcitonin as a prognostic biomarker for infections and clinical severity. METHOD: A prospective single-cohort observational study of patients diagnosed of acute pancreatitis (n = 152) was designed. PCT determination was tested on admission (first 72 h). Infections (biliary, extrapancreatic and infected pancreatic necrosis), need for antibiotics, urgent ERCP and severity scores for acute pancreatitis was assessed. ROC curves were designed and the area under the curve was calculated. Logistic regression for multivariate analysis was performed to evaluate the association between procalcitonin optimal cut-off level and major complications. RESULTS: PCT >0.68 mg/dL had higher incidence of global infection, acute cholangitis, bacteraemia, infected pancreatic necrosis, use of antibiotics in general, and need for urgent ERCP. In the multivariate regressions analysis, PCT >0.68 mg/dL at admission demonstrated to be a strong risk factor for complications in acute pancreatitis. DISCUSSION: PCT levels can be used as a reliable laboratory test to predict infections and the clinical severity of acute pancreatitis. High levels of PCT predict antibiotics prescription as well as the need for urgent ERCP in patients with concomitant clinically severe cholangitis.

BACKGROUND: Malnutrition is frequent in patients with cirrhosis and has been associated with poor prognosis. The Model for End-stage Liver Disease (MELD) score was created to predict survival after Transjugular Intrahepatic Porto-systemic Shunt (TIPS) but lacks a nutritional parameter. AIMS: To evaluate the prognostic value of serum cholesterol in patients with cirrhosis undergoing TIPS and to develop a prognostic score to predict survival. METHODS: An explorative cross-sectional study was conducted of cirrhotic patients undergoing TIPS from 2008 until 2019. Exclusion criteria were liver transplantation or hepatocellular carcinoma before TIPS. Risk analysis was used to compare survival according to clinical and analytical data. The diagnostic performance of serum cholesterol added to MELD was evaluated and confirmed in an external validation cohort. RESULTS: The final cohort of 100 patients had a mean MELD score of 14±5 and cholesterol of 122±51 mg/dL. MELD (p < 0,05) and both cholesterol (p < 0,05) and low-density lipoprotein levels (LDL-C) (p < 0,05) were independent predictors of post-TIPS transplant-free survival with an optimal cut-off of 106 mg/dL for serum cholesterol. The combined MELD-cholesterol risk score improved diagnostic accuracy of each parameter separately, and this was confirmed in the external cohort. CONCLUSION: Serum cholesterol and LDL-C are independent predictors of transplant-free survival in cirrhotic patients undergoing TIPS. The MELD-cholesterol score slightly improved prognostic accuracy. LAY SUMMARY: As an objective and easily measured indicator of both nutritional status and hepatic function, serum cholesterol could be useful to predict transplant-free survival in patients with cirrhosis undergoing TIPS. It can enable health care providers to identify high-risk patients and to optimize nutritional status before TIPS.

Garlic (Allium sativum) has been used in alternative medicine to treat several diseases, such as cardiovascular and neurodegenerative diseases, cancer, and hepatic diseases. Several publications have highlighted other features of garlic, including its antibacterial, antioxidative, antihypertensive, and antithrombotic properties. The properties of garlic result from the combination of natural compounds that act synergistically and cause different effects. Some garlic-derived compounds have been studied for the treatment of several types of cancer; however, reports on the effects of garlic on neuroblastoma are scarce. Neuroblastoma is a prevalent childhood tumor for which the search for therapeutic alternatives to improve treatment without affecting the patients' quality of life continues. Garlic-derived compounds hold potential for the treatment of this type of cancer. A review of articles published to date on some garlic compounds and their effect on neuroblastoma was undertaken to comprehend the possible therapeutic role of these compounds. This review aimed to analyze the impact of some garlic compounds on cells derived from neuroblastoma.

The occurrence of bioactive compounds and contaminant-associated effects was assessed by means of in vivo and in vitro assays using different extractable fractions of surface sediments from a contaminated coastal lagoon (Mar Menor, SE Spain). Sediment elutriates and clean seawater, previously exposed to whole sediment, were used for assessing the in vivo toxicity on embryo development of the sea urchin Paracentrotus lividus. Agonist and antagonist activities relating to estrogen and androgen receptors and agonist activities on aryl hydrocarbon receptor (expressed as ethoxyresorufin-O-deethylase (EROD) activities) were investigated in sediment extracts by using HER-Luc, AR-EcoScreenTM and fibroblast-like RTG-2 cell lines. Embryotoxicity effects were greater for sediment elutriates than those incubated in sediment-water interphase, implying that diffusion of bioactive chemicals can occur from sediments to sea water column, favoured by sediment disturbance events. In vitro results show the occurrence in extracts of compounds with estrogen antagonism, androgen antagonism and dioxin-like activities. Multidimensional scaling analysis classified the sampling sites into four sub-clusters according to their chemical-physical and biological similarities, relating in vitro bioactivity with the total organic carbon and known organic chemical load, with particular reference to total sum of PAHs, PCB 180, p,p-DDE and terbuthylazine. Overall, results pointed to the presence of unknown or unanalyzed biologically-active compounds in the sediments, mostly associated with the extracted polar fraction of the Mar Menor lagoon sediments. Our findings provide relevant information to be considered for the environmental management of contaminated coastal lagoons.

Gliomas are the most common and most lethal primary malignant adult brain tumors, and glioblastomas are the most frequent. Several risk factors are involved in their pathogenesis; these include environmental factors as well as host factors. The etiology of most gliomas remains unknown. Epstein-Barr Virus (EBV), a member of the Herpesviridae family, was the first tumoral virus to be described, and several viruses in connection with cancer were discovered thereafter. During the complex interaction between host and EBV, several events take place. In the context of survival, EBV can drive its host cells with subsequent disruption of the cellular machinery, leading to tumorigenesis as the final outcome. Thus, the EBV infection has been associated with different tumors. In this review, we discuss EBV and cancer. We have analyzed previously published papers and have conducted a critical analysis on the role of the viral infection in glioblastoma. Several works have described the presence of the virus, but none have shown a conclusive association. Thus, there is need to continue analyzing the interaction between host and virus to determine whether the viral presence is incidental or has some association with glioblastoma.

Burkholderia cenocepacia is an opportunistic pathogen that infects individuals with cystic fibrosis, chronic granulomatous disease, and other immunocompromised states. B. cenocepacia survives in macrophages in membrane-bound vacuoles; however, the mechanism by which B. cenocepacia gains entry into macrophages remains unknown. After macrophage internalization, survival of B. cenocepacia within a bacteria-containing membrane vacuole (BcCV) is associated with its ability to arrest the maturation of the BcCV. In this study, we show that B. cenocepacia induces localized membrane ruffling, macropinocytosis, and macropinosomes-like compartments upon contact with the macrophage. The Type 3 Secretion System (T3SS) of B. cenocepacia contributes to macrophage entry and macropinosome-like compartment formation. These data demonstrate the ability of Burkholderia to enter macrophages through the induction of macropinocytosis.

Introducción. Los defectos septales ventriculares graves, asociados con un ventrículo hipoplásico y otro hiperplásico o con la ausencia total del tabique interventricular, suelen ser incompatibles con la vida embrionaria y fetal. Pese a su gravedad, no se conocen sus causas. Por marcaje in vivo en embriones de pollo se confirmó la importancia de las trabéculas en la morfogénesis del tabique interventricular. Por manipulación genética en embriones de ratón y aves se determinó que la ausencia de la Neuregulina 1 (NRG1) o sus receptores, además de provocar escasa diferenciación de los miocitos ventriculares y deficiente formación de las trabéculas, determina la muerte prematura del embrión. Con base en estos antecedentes, el objetivo fue determinar el papel real de NRG1 en la trabeculogénesis temprana y su importancia en la regulación de la proliferación y apoptosis. Métodos. Se estableció un modelo de órgano cultivo de corazón de embrión de pollo previo al inicio de la trabeculogénesis. Se realizaron ensayos de inhibición total de la actividad de NRG1 endógena y posterior adición de la proteína exógena a diferentes concentraciones, determinando la actividad cíclica de los miocitos ventriculares con el antígeno nuclear de proliferación celular y la apoptosis con lisotraker. Resultados. El suero fetal bovino promueve la proliferación, pero impacta negativamente la trabeculogénesis. La adición de NRG1 a concentraciones bajas y períodos cortos de incubación no induce trabeculogénesis. En contraste, a concentraciones medias y períodos de cultivo no mayores a 24 horas, tiene un efecto positivo sobre este proceso. También promueve la proliferación y evita la apoptosis del miocardio ventricular. El incremento en la concentración de NRG1 posiblemente provoca un desbalance molecular que favorece la proliferación desordenada pero no la trabeculogénesis. Conclusiones. El entendimiento del papel de NRG1 en la trabeculogénesis aporta datos para conocer las redes moleculares involucradas también en el desarrollo del tabique interventricular, información indispensable para entender el origen de los defectos septales ventriculares graves.


Background. Serious ventricular septal defects associated with hypoplastic and hyperplasic ventricles or total absence of the interventricular septum (IVS) are usually incompatible with embryonic and fetal life. Despite the importance of these cardiac diseases, their causes are not yet known. Using in vivo labeling in the chick embryo, the importance of ventricular trabeculation was confirmed in IVS morphogenesis. Using knockout mice and retrovirus in birds, it was determined that lack of function of neuregulin 1 (NRG1) or their ErbB receptors not only causes deficient differentiation of ventricular myocytes and poor formation of trabeculae, but also determines premature death of the embryos. Based on this background, the aim of this work was to determine the actual role of NRG1 in early trabeculogenesis and its importance in proliferation and apoptosis regulation. Methods. An embryonic chicken heart organ culture system at the age prior to the beginning of the trabeculogenesis process was established. Endogenous activity of NRG1 was inhibited in the organ cultures that were then stimulated with NRG1 at different concentrations. Myocyte proliferation was determined using the proliferating cell nuclear antigen and apoptosis with LysoTracker (LTR). Results. Fetal bovine serum promotes proliferation but negatively impacts trabeculogenesis. Low concentration of NRG1 and short periods of incubation do not induce trabeculogenesis. In contrast, average NRG1 concentrations and cultivation periods not exceeding 24 h have a positive effect on the onset of this process. This also promotes myocardial proliferation but avoids apoptosis. Higher concentrations of NRG1 possibly cause a molecular imbalance that favors untidy proliferation but not trabeculogenesis. Conclusions. Understanding of the role of NRG1 on ventricular trabeculogenesis provides valuable information for the molecular pathways also involved in IVS development. This information is essential for understanding the origin of serious ventricular septal defects.

A biological screening was performed to establish the total exposure to estrogenic compounds of red mullet (Mullus barbatus) collected at several sites along the Spanish Mediterranean coast by testing male fish bile extracts using the in vitro ER-LUC reporter gene assay. In addition, major metabolites were identified and measurements of OH-PAHs (1-naphthol, 9-phenantrol, 9-fluorenol, 1-pyrenol, 1OH-BaP and 3OH-BaP) and alkylphenols (4-n-nonylphenol (4-n-NP) and 4-tertoctylphenol (4-tert-OP)) in the same fish bile extracts were taken by gas chromatography-mass spectrometry in electron ionization mode (GC-EI-MS). Relative in vitro estrogenic potencies of the chemically quantified compounds were also tested. The highest biliary concentrations of 1-pyrenol, 9-fluorenol and 4-n-NP were found in fish from Barcelona and from the Mar Menor coastal lagoon. However, these concentrations can be considered relatively low compared to values reported in red mullet from other polluted waters in the Mediterranean Sea. The contribution of 1-pyrenol, 4-n-NP and 4-tert-OP to the total estrogenic potency measured in male fish bile was found to be negligible, indicating the presence of other estrogenic compounds in red mullet bile. Estrogenic potency in bile from male fish was markedly elevated in Mar Menor lagoon (234.8±5.7 pg E2EQ/µl), and further research will be necessary to explain whether the presence of natural and synthetic-hormones in the lagoon contributed to this finding. Values of approximately 15-16E2EQ pg/mg bile can be regarded as preliminary baseline levels of bile estrogenicity in male red mullet from the western Mediterranean Sea.

Zinc oxide nanoparticles (ZnO-NPs) are inevitably released into the environment and are potentially dangerous for aquatic life. However, the potential mechanisms of cytotoxicity of zinc nanoparticles remain unclear. Studying the toxicity of ZnO-NPs with In vitro systems will help to determine their interactions with cellular biomolecules. The aim of this study was to evaluate the cytotoxic potentials of ZnO-NPs in established fish cell lines (RTG-2, RTH-149 and RTL-W1) and compare them with those of bulk ZnO and Zn(2+) ions. Membrane function (CFDA-AM assay), mitochondrial function (MTT assay), cell growth (KBP assay), cellular stress (ß-galactosidase assay), reductase enzyme activity (AB assay), reactive oxygen species (ROS), total glutathione cellular content (tGSH assay) and glutathione S-transferase (GST) activities were assessed for all cell lines. ZnO-NPs cytotoxicity was greater than those of bulk ZnO and Zn(2+). ZnO-NPs induced oxidative stress is dependent on their dose. Low cost tests, such as CFDA-AM, ROS, GST activity and tGSH cell content test that use fish cell lines, may be used to detect oxidative stress and redox status changes. Particle dissolution of the ZnO-NPs did not appear to play an important role in the observed toxicity in this study.

We have previously reported that Salmonella infects B cells and survives within endosomal-lysosomal compartments. However, the mechanisms used by Salmonella to enter B cells remain unknown. In this study, we have shown that Salmonella induces its own entry by the induction of localized ruffling, macropinocytosis, and spacious phagosome formation. These events were associated with the rearrangement of actin and microtubule networks. The Salmonella pathogenesis island 1 (SPI-1) was necessary to invade B cells. In contrast to macrophages, B cells were highly resistant to cell death induced by Salmonella. These data demonstrate the ability of Salmonella to infect these non-professional phagocytic cells, where the bacterium can find an ideal intracellular niche to support persistence and the possible dissemination of infection.

RESUMO

The ample development of diagnostic echocardiography in pediatric cardiology has demanded precise knowledge of the abnormal anatomy of hearts that present congenital cardiac diseases. As a result, the information on morphologic and molecular aspects of cardiac embryogenesis has become fundamental to understand the anomalous anatomy of the malformed hearts. Based on these facts, in this paper we reviewed normal cardiogenesis, integrating the new information obtained experimentally in the chick embryo and from classic descriptive knowledge in humans. The age at which each cardiac segment appears is specified. At the same time, the changes in shape, relationships and position of these cardiac segments are detailed. Some implications of this process in the production of congenital cardiac defects and the importance of some specific genes are also discussed. This information is useful in the diagnosis of congenital cardiac diseases, as well as in discussing their embryogenesis. It is also beneficial in studying the possible mechanisms and genes implicated in normal morphogenesis of cardiac chambers, septa and valves. All this knowledge is important to plan strategies to avoid the production of this type of congenital pathologies.

Se define el septum atrioventricular y sus características anatomicas. Este septum se divide en dos regiones: muscular y membranosa. Se describen los tipos de comunciación atrioventricular aisladas. Se discute la embriogénesis normal y patológica del septum atrioventricular. Se define el septum interventricular y se describen sus características anatómicas. Este septum se divide en dos regiones: fibrosa y muscular. La muscular se divide en tres regiones: porción de la cámara de salida o infundibular (outlet). Las comunicaciones inteventriculares se clasifican de acuerdo con las regiones septales en que están ubicadas. Se establecen las características anatómicas de identidad de cada una de ellas.
