In this study, the recombinant gut protein rRa92A produced in Pichia pastoris yeast cells was used to immunize cattle in two experiments, one in Brazil and the other in Uganda. In both experiments, the animals were intramuscularly (IM) injected with 200 µg of recombinant protein in Brazil on days 0, 30 and 51 and in Uganda on days 0, 30. Blood samples for sera separation were collected from different days in both experiments. These samples were analyzed by ELISAs. In Brazil, ticks collected from the animals during the experimental period were analyzed for biological parameters. At Uganda, blood was collected to assess blood parameters, clinical signs were recorded and adult tick (Rhipicephalus appendiculatus) counts were performed. All animals of the vaccinated groups were shown to produce antibodies, and it was not possible to detect an effect on Rhipicephalus microplus. All the clinical parameters were considered within the normal ranges for both the experimental and control groups in Uganda. Antibody absorbance was elevated after each immunization and remained high until the end of the experiments, remaining low in the control animals. The results of stall test carried out in Brazil using R. microplus tick showed efficacy of 21.95%. The rRa92A immunization trial experiments in Uganda showing a decrease of 55.2% in the number of engorged adult ticks, which was statistically significant (p<0.05). Assessment of the immunogenicity of Ra92A produced in the P. pastoris expression system in bovines is reported for the first time, and the protein acted as a concealed antigen.

Abstract Amblyomma scalpturatum is a tick species that is unique to South America. It is commonly associated with the Amazon biome and has been reported in some Brazilian states. This tick species exhibits host specificity: it parasitizes tapirs and suidae. Its role in transmitting pathogens to humans is still unknown. Amblyomma scalpturatum is known to be a human-biting tick; however, there is only one report showing that humans make suitable hosts for this species. The knowledge of tick fauna is lacking in the Acre State. This study collected free-living ticks with the aim of finding new records in Acre State. Collections were carried out in Amazon forest fragments in Rio Branco municipality. An A. scalpturatum specimen was identified and submitted sequencing of the ITS-2 gene. This study presents the first molecular confirmation of A. scalpturatum collected in situ in Acre State, North Region, Brazil. This study also presents the first record of a successfully completed feeding by an A. scalpturatum nymph on a human host in the North region of Brazil. Further investigation is needed to ascertain the complete life cycle of this tick species, its seasonality in the environment, and its relationship to pathogens and competence to transmit them.


Resumo Amblyomma scalpturatum é uma espécie de carrapato que ocorre apenas na América do Sul. Está comumente associada ao bioma da Amazônia e tem sido relatada em alguns estados brasileiros. Esta espécie de carrapato apresenta especificidade a hospedeiros: parasita antas e suídeos. Sua competência em transmitir agentes patogênicos a seres humanos é ainda desconhecida. Amblyomma scalpturatum é também conhecido por picar seres humanos; entretanto, há apenas um relato que evidencie que seres humanos sejam capazes de serem hospedeiros adequados. Ainda existem lacunas no conhecimento sobre a ixodofauna no estado do Acre. Neste estudo foram coletados carrapatos de vida livre com o objetivo de encontrar novos registros para o estado do Acre. Coletas de carrapatos foram realizadas em fragmentos de floresta Amazônica no município de Rio Branco. Um exemplar de A. scalpturatum foi identificado e submetido a sequenciamento do gene ITS-2. Este estudo apresenta a primeira confirmação molecular de A. scalpturatum coletado in situ no estado do Acre, região Norte, Brasil. Este estudo também apresenta o primeiro relato de parasitismo completo de uma ninfa de A. scalpturatum em um hospedeiro humano na região Norte do Brasil. Mais investigações são necessárias para elucidar o ciclo de vida completo dessa espécie de carrapato, a sazonalidade de seus estádios no meio ambiente, sua relação a agentes patogênicos e competência em transmiti-los.

The success of cattle tick fixation largely depends on the secretion of substances that alter the immune response of the host. The majority of these substances are expressed by the parasite salivary gland and secreted in tick saliva. It is known that hosts can mount immune responses against ticks and bovine European breeds, and bovine industrial crossbreeds are more susceptible to infestations than are Bos indicus cattle. To identify candidates for the development of novel control strategies for the cattle tick Rhipicephalus (Boophilus) microplus, a salivary gland transcriptome analysis of engorged females fed on susceptible or resistant hosts was performed. Using RNA-Seq, transcriptomes were de novo assembled and produced a total of 235,451 contigs with 93.3% transcriptome completeness. Differential expression analysis identified 137 sequences as differentially expressed genes (DEGs) between ticks raised on tick-susceptible or tick-resistant cattle. DEGs predicted to be secreted proteins include innexins, which are transmembrane proteins that form gap junction channels; the transporters Na+/dicarboxylate, Na+/tricarboxylate, and phosphate transporter and a putative monocarboxylate transporter; a phosphoinositol 4-phosphate adaptor protein; a cysteine-rich protein containing a trypsin inhibitor-like (TIL) domain; a putative defense protein 3 containing a reeler domain; and an F-actin-uncapping protein LRRC16A with a CARMIL_C domain; these genes were upregulated in ticks fed on tick-susceptible cattle. DEGs predicted to be non-secreted proteins included a small heat shock protein and the negative elongation factor B-like, both acting in a coordinated manner to increase HSP transcript levels in the salivary glands of the ticks fed on tick-susceptible cattle; the 26S protease regulatory subunit 6B and another chaperone with similarity to calnexin, also upregulated in ticks fed on tick-susceptible cattle; an EF-hand calcium binding protein and a serine carboxypeptidase (SCP), both involved in the blood coagulation cascade and upregulated in ticks fed on tick-susceptible cattle; and two ribosomal proteins, the 60S acidic ribosomal protein P2 and the 60S ribosomal protein L19. These results help to characterize cattle tick salivary gland gene expression in tick-susceptible and tick-resistant hosts and suggest new putative targets for the control of tick infestations, as those genes involved in the mechanism of stress response during blood feeding.

Abstract The cattle tick Rhipicephalus microplus causes significant economic losses in agribusiness. Control of this tick is achieved mainly through the application of chemical acaricides, often resulting in contamination of animal food products and of the environment. Another major concern associated with acaricide use is the increasing reports of resistance of this tick vector against the active ingredients of many commercial products. An alternative control method is vaccination. However, the commercially available vaccine based on a protein homologous to Bm86 exhibits variations in efficacy relative to the different geographical locations. This study aimed to identify antigenic determinants of the sequences of proteins homologous to Bm86. Phylogenetic analyses were performed to determine the extent of divergence between different populations of R. microplus to identify the sequence that could be used as a universal vaccine against the multiple geographically distinct populations of R. microplus and related tick species. Considering the extensive sequence and functional polymorphism observed among strains of R. microplus from different geographical regions, we can conclude that it may be possible to achieve effective vaccination against these cattle ticks using a single universal Bm86-based antigen.


Resumo O carrapato Rhipicephalus microplus é responsável por perdas significativas no agronegócio. O controle deste carrapato é feito principalmente por meio da aplicação de acaricidas químicos, geralmente resultando na contaminação de produtos de origem animal e do meio ambiente. Outra preocupação importante associada ao uso de acaricidas é o crescente aumento de relatos sobre a resistência deste carrapato a princípios ativos de vários produtos comerciais. Uma alternativa de controle é por meio de vacinação. Porém, a vacina comercializada contendo proteína homóloga à Bm86, apresenta variações de eficácia em relação às diferentes localizações geográficas. Este estudo buscou identificar determinantes antigênicos das sequencias de proteínas homólogas a Bm86. As análises filogenéticas foram feitas para determinar a extensão da divergência entre diferentes populações de R. microplus com o objetivo de identificar a sequência que poderia ser usada como vacina universal contra as múltiplas populações geograficamente distintas de R. microplus e espécies de carrapatos relacionados. Considerando-se a extensa sequência e o polimorfismo observados entre linhagens de R. microplus de diferentes regiões geográficas, podemos concluir que pode ser possível obter uma vacinação efetiva contra esses carrapatos bovinos utilizando um único antígeno universal baseado em Bm86.

The fatal cases of spotted fever reported in some Brazilian municipalities are generally associated with the presence of ticks and capybaras (Hydrochoerus hydrochaeris). The urban forest fragments where this rodent is present are of great concern. We investigated the diversity of free ticks as well as the occurrence of ticks infected by Rickettsia in one of these fragments, in Campo Grande city, Mato Grosso do Sul state, Brazil. From winter 2012 until the autumn of 2015, using CO2 traps, we collect 1219 ticks, of which 1045 belonged to the species Amblyomma sculptum and 174 belonged to A. dubitatum. Both species were more abundant in spring (September to December) and summer (December to March). Rickettsial DNA was not detected in any of the specimens collected; however, one of the species found, A. sculptum is considered the main vector of Brazilian Spotted Fever, thus deserving constant vigilance by the public health organs.


Casos fatais de febre maculosa que foram relatados em alguns municípios brasileiros geralmente estão associados à presença de carrapatos e capivaras (Hydrochoerus hydrochaeris). Os fragmentos florestais urbanos onde este roedor está presente são de grande preocupação. Investigou-se aqui a diversidade de carrapatos livres e a ocorrência de carrapatos infectados por riquétsias em um desses fragmentos em Campo Grande, Mato Grosso do Sul. Desde o inverno de 2012 até o outono de 2015, usando armadilhas de CO2, foram coletados 1219 carrapatos, dos quais 1045 pertenciam à espécie Amblyomma sculptum e 174 pertenciam a A. dubitatum. Ambas as espécies foram mais abundantes na primavera (Setembro a Dezembro) e no verão (Dezembro a Março) . O DNA riquetsial não foi detectado em nenhum dos espécimes coletados; entretanto, uma das espécies encontradas, o A. sculptum é considerado o principal vetor da Febre Maculosa Brasileira, merecendo assim vigilância constante pelos Orgãos de Saúde Pública.

Abstract The objective of this work was to evaluate the diversity of ticks associated with free-living animals and to investigate new host records for ticks. Ticks were collected from animals rescued during the flood of the Jamari River in the municipality of Ariquemes, state of Rondônia, North Region of Brazil. A total of 39 animals were captured, out of which 10 were amphibians, 19 were reptiles and 10 were mammals. A total of 127 ticks of the Amblyomma genus were collected from these animals, distributed among seven species: Amblyomma dissimile, Amblyomma geayi, Amblyomma humerale , Amblyomma longirostre, Amblyomma nodosum , Amblyomma rotundatum and Amblyomma varium. In addition, one specimen of Rhipicephalus (Boophilus) microplus was collected. Among these specimens, 85 were adults and 42 were nymphs, with A. rotundatum being the most prevalent species. An Amblyomma spp. larvae was also collected from a lizard (Uranoscodon superciliosus), and one Amblyomma calcaratum and one Amblyomma dubitatum were recovered from the environment, thus totaling 130 ticks. Among the Ixodidae collected from different hosts, we provide the first report for the species A. rotundatum parasitizing Rhinella major, U. superciliosus, Leptophis ahaetulla, Chironius multiventris, and Mastigodryas boddaerti, as well as of A. humerale parasitizing U. superciliosus, A. geayi parasitizing Choloepus didactylus, and Rhipicephalus (B.) microplus parasitizing Alouatta puruensis.


Resumo O objetivo deste trabalho foi avaliar a diversidade de carrapatos associados à animais de vida livre e investigar novos registros de hospedeiros. Coletas foram feitas em animais resgatados durante a cheia do Rio Jamari, localizado no município de Ariquemes, estado de Rondônia, Região Norte do Brasil. Um total de 39 animais foi capturado, dos quais dez eram anfíbios, 19 eram répteis e dez eram mamíferos. 127 carrapatos do gênero Amblyomma foram coletados destes animais, distribuídos em sete espécies: Amblyomma dissimile , Amblyomma geayi, Amblyomma humerale, Amblyomma longirostre, Amblyomma nodosum, Amblyomma rotundatum e Amblyomma varium. Adicionalmente, um exemplar de Rhipicephalus (Boophilus) microplus foi coletado. Dentre estes espécimes, 85 eram adultos e 42 eram ninfas, com A. rotundatum sendo a espécie mais prevalente. Uma larva de Amblyomma spp. também foi coletada de um lagarto (Uranoscodon superciliosus), um Amblyomma calcaratum e um Amblyomma dubitatum foram recuperados do ambiente, assim totalizando 130 carrapatos. Dentre os ixodídeos coletados de diferentes hospedeiros, este trabalho fornece o primeiro registro da espécie A. rotundatum parasitando Rhinella major, U. superciliosus, Leptophis ahaetulla, Chironius multiventris e Mastigodryas boddaerti, assim como da espécie A. humerale parasitando U. superciliosus , a espécie A. geayi parasitando Choloepus didactylus e R. microplus parasitando Alouatta puruensis .

Abstract One generation of Amblyomma coelebs life cycle under experimental conditions was evaluated. Ten tick pairs were allowed to feed on rabbits under laboratory conditions (LC), resulting six engorged females with a mean weight of 1,403.9 mg. Two females were maintained in a forest reserve under natural conditions (NC), and four were maintained in incubators (LC). The engorgement period lasted 10.33 days. Pre-oviposition periods were 10.75 (NC) and 22 days (LC). The mean egg-mass weight was 514.76 mg, and the blood meal conversion index was 36.67% (LC). Incubation period under NC and LC were 91 and 56.33 days and hatching rates were 50% and 28.33%, respectively. Larval engorgement period ranged from 4 to 10 days, with average weight of 1.1 mg. Engorged larvae were incubated under NC and LC, with a premolt period of 27 to 36 days and molting rate of 7.1% and 28.7%, respectively. Nymphal engorgement period ranged from 5 to 7 days, with a mean weight of 18.8 mg and a recovery rate of 54.54%. In LC, the ecdysis mean period was 24.5 days, and molting rate was 44.44%, resulting in 24 adult A. coelebs. Our results show a life cycle of 187.45 (NC) and 149 (LC) days.


Resumo Uma geração do ciclo de vida de Amblyomma coelebs em condições laboratoriais foi avaliada. Dez casais de carrapatos foram alimentados em coelhos sob condições laboratoriais (CL), resultando em seis fêmeas ingurgitadas, com um peso médio de 1.403,9 miligramas (mg). Duas fêmeas foram mantidas em uma reserva florestal sob condições naturais (CN), e quatro foram mantidas em incubadoras (CL). O período de ingurgitamento durou 10,33 dias. Períodos de pré-postura foram de 10,75 (CN) e 22 dias (CL). O peso médio das massas de ovos foi de 514,76 mg e o índice de conversão alimentar foi de 36,67% (CL). O período de incubação em CN e CL foi de 91 e 56,33 dias e os percentuais de eclosão foram de 50% e 28,33%, respectivamente. O período de ingurgitamento larval variou de quatro a 10 dias, com peso médio de 1,1 mg. Larvas ingurgitadas foram incubadas em CN e CL, com período de pré-muda de 27 a 36 dias e percentual de ecdise de 7,1% e 28,7%, respectivamente. O período de ingurgitamento das ninfas oscilou de cinco a sete dias, com peso médio de 18,8 mg e uma taxa de recuperação de 54,54%. Em CL, o período médio de ecdise foi de 24,5 dias, e o percentual de muda foi 44,44%, resultando em 24 adultos de A. coelebs. Estes resultados demonstram um ciclo de vida de 187,45 (CN) e 149 (CL) dias.

The bovine tick Rhipicephalus (Boophilus) microplus is found in several tropical and subtropical regions of the world. This parasite transmits pathogens that cause disease, such as babesiosis (Babesia bovis and B. bigemina) and anaplasmosis (Anaplasma marginale). Tick infestations cause enormous livestock losses, and controlling tick infestations and the transmission of tick-borne diseases remains a challenge for the livestock industry. Because the currently available commercial vaccines offer only partial protection against R. (B.) microplus, there is a need for more efficient vaccines. Several recombinant antigens have been evaluated using different immunization strategies, and they show great promise. This work describes the construction and immunological characterization of a multi-antigen chimera composed of two R. (B.) microplus antigens (RmLTI and BmCG) and one Escherichia coli antigen (B subunit, LTB). The immunogenic regions of each antigen were selected and combined to encode a single polypeptide. The gene was cloned and expressed in E. coli. For all of the experiments, two groups (treated and control) of four Angus heifers (3-6 months old) were used. The inoculation was performed via intramuscular injection with 200 µg of purified recombinant chimeric protein and adjuvated. The chimeric protein was recognized by specific antibodies against each subunit and by sera from cattle inoculated with the chimera. Immunization of RmLTI-BmCG-LTB cattle reduced the number of adult female ticks by 6.29% and vaccination of cattle with the chimeric antigen provided 55.6% efficacy against R. (B.) microplus infestation. The results of this study indicate that the novel chimeric protein is a potential candidate for the future development of a more effective vaccine against R. (B.) microplus.

Abstract The present study aimed to contribute towards identification and registration of tick species that parasitize dogs in rural and urban areas of three mesoregions of Paraná, southern Brazil, and to estimate the rate of occurrence of each species. Fifty-six dogs with ticks living in three mesoregions: Metropolitana de Curitiba (MC), Centro Oriental (COP) and Centro Sul Paranaense (CSP), were used in the study. From these 56 dogs, 253 ticks were collected and were identified and morphologically characterized according to the species. Among all the ticks, 69.6% were identified as belonging to the species Rhipicephalus sanguineus sensu lato (s. l.); 28.1% as Amblyomma aureolatum and 2.4% as Amblyomma ovale. Among the dogs in MC that were evaluated, 57.7% were parasitized by R. sanguineus s. l., 38.5% by A. aureolatum and 3.8% by A. ovale; while in COP, 72.4% of the dogs were parasitized by A. aureolatum and 27.6% by R. sanguineus s. l.. In CSP, one tick was obtained, which was identified as A. aureolatum.


Resumo O presente estudo objetivou contribuir com a identificação e o registro das espécies de carrapatos que parasitam cães de áreas rurais e urbanas de três mesorregiões do Paraná, Sul do Brasil, e estimar a taxa de ocorrência de cada espécie. Cinquenta e seis cães com carrapatos, provenientes das mesorregiões: Metropolitana de Curitiba (MC), Centro Oriental (COP) e Centro Sul Paranaense (CSP) foram utilizados no estudo. Dos 56 cães, foram coletados 253 carrapatos que foram identificados e caracterizados morfologicamente de acordo com a espécie. Do total de carrapatos, 69,6% foram identificadas como pertencentes à espécie Rhipicephalus sanguineus s. l.; 28,1% como Amblyomma aureolatum e 2,4% como Amblyomma ovale. Dentre os animais avaliados, provenientes da MC, 57,7% estavam parasitados por R. sanguineus s. l., 38,5% por A. aureolatum e 3,8% por A. ovale; enquanto na COP 72,4% dos cães foram parasitados por A. aureolatum e 27,6% por R. sanguineus s. l.. Na CSP foi obtido um carrapato, identificado como A. aureolatum.

Ticks from Rhipicephalus sanguineus complex are widely distributed in the world and one species from this complex is the most common tick on dogs in Brazil, notably in urban areas. This tick is a vector of several diseases. Among others it transmits the agent of canine Ehrlichiosis, a major dog infectious disease and the agent of Rocky Mountain spotted fever. This tick can spread rapidly and develop intolerable infestations within no time. Currently tick control is done with acaricides and demand for such drugs has grown fast. However, R. sanguineus has already developed resistance to the main active compounds and the development of new acaricides is necessary. Many essential oils of plants have acaricidal effect and may be an important source of molecules for the synthesis of new acaricide products. In this study, we evaluated the effectiveness of a new herbal phytotherapic, consisting of the essential oil of Tagetes minuta L., against R. sanguineus in vitro and on dogs undergoing experimental infestations. The product displayed 100% efficacy against larvae, nymphs and adults of the tick on all tested conditions.

Abstract The Rhipicephalus (Boophilus) microplus complex currently consists of five taxa, namely R. australis, R. annulatus, R. (B.) microplus clade A sensu, R. microplus clade B sensu, and R. (B.) microplus clade C sensu. Mitochondrial DNA-based methods help taxonomists when they are facing the morpho-taxonomic problem of distinguishing members of the R. (B.) microplus complex. The purpose of this study was to perform molecular characterization of ticks in all five regions of Brazil and infer their phylogenetic relationships. Molecular analysis characterized 10 haplotypes of the COX-1 gene. Molecular network analysis revealed that haplotype H-2 was the most dispersed of the studied populations (n = 11). Haplotype H-3 (n = 2) had the greatest genetic differentiation when compared to other Brazilian populations. A Bayesian phylogenetic tree of the COX-1 gene obtained strong support. In addition, it was observed that the population of R. (B.) microplus haplotype H-3 exhibited diverging branches among the other Brazilian populations in the study. The study concludes that the different regions of Brazil have R. (B.) microplus tick populations with distinct haplotypes.


Resumo Carrapatos do complexo R. (B.) microplus se distribuem em cinco taxa: R. australis, R. annulatus, R. (B.) microplus clado A sensu R. microplus clado B sensue e R. (B.) microplus clado C sensu. Métodos baseados no DNA mitocondrial podem auxiliar taxonomistas quando há dificuldades em estabelecer diferenças morfológicas para distinguir membros do complexo R. (B.) microplus. O objetivo deste estudo foi a caracterização molecular e a inferência de relações filogenéticas em carrapatos de todas as cinco regiões geográficas do Brasil. Para o gene COX-1, a análise molecular caracterizou 10 haplótipos. Na análise molecular em rede foi observado que o haplótipo H-2 é o mais disperso entre as populações (n=11). O haplótipo H-3 (n=2) foi o que obteve maior diferenciação genética ao ser comparado com outras populações brasileiras. A árvore filogenética Bayesiana de gene COX-1 gerou suporte robusto e foi observado que a população de R. (B.) microplus haplótipo H-3 apresentou ramificação com divergência entre as outras populações brasileiras apresentadas neste estudo. Conclui-se que as populações brasileiras possuem diversidade haplotípica com divergência entre as diversas populações de R. (B.) microplus no Brasil.

Abstract The Rhipicephalus (Boophilus) microplus tick is responsible for considerable economic losses in Brazil, causing leather damage, weight loss and reduced milk production in cattle and results in the transmission of pathogens. Currently, the main method for controlling this tick is using acaricides, but their indiscriminate use is one of the major causes of resistance dissemination. In this study, the adult immersion test (AIT) was used to evaluate resistance in ticks from 28 properties located in five different states (Mato Grosso do Sul, Mato Grosso, Ceará, São Paulo, e Minas Gerais) and the Distrito Federal (DF) of Brazil. The resistance was found in 47.64% of the repetitions demonstrating an efficacy of less than 90% in various locations throughout the country. The larvae packet test was used to evaluate samples from ten properties in four states (Mato Grosso do Sul, Mato Grosso, Minas Gerais and Espírito Santo). Spray products belonging to the main classes of acaricides, including combination formulations, were used in both types of test. New cases of resistance were found on properties within the states of Ceará, Espírito Santo and Mato Grosso, where such resistance was not previously reported.


Resumo O carrapato Rhipicephalus (Boophilus) microplus é responsável por consideráveis perdas econômicas no Brasil, causando, nos bovinos, danos ao couro, perda de peso, redução na produção de leite, o que resulta na transmissão também de patógenos. Atualmente, o principal método de controle para esse carrapato é o uso de acaricidas, porém seu uso indiscriminado é uma das principais causas da disseminação da resistência. Neste estudo, o teste de imersão de adultos (TIA) foi utilizado para avaliar a resistência em carrapatos provenientes de 28 propriedades, localizadas em cinco diferentes estados brasileiros (Mato Grosso do Sul, Mato Grosso, Ceará, São Paulo, e Minas Gerais) e no Distrito Federal (DF). A resistência foi encontrada em 47,64% das repetições, demonstrando uma eficácia de menos de 90% em vários lugares do país. O teste de pacote de larva foi utilizado para avaliar amostras de 10 propriedades em quatro estados (Mato Grosso do Sul, Mato Grosso, Minas Gerais e Espírito Santo). Produtos de pulverização, pertencentes às principais classes de acaricidas, incluindo-se associações, foram utilizados em ambos os testes. Novos casos de resistência foram encontrados em propriedades nos estados do Ceará, Espírito Santo e Mato Grosso, nas quais a resistência ainda não havia sido previamente reportada.

Tick infestation may cause several problems including affecting domestic animal health and reducing the production of meat and milk, among others. Resistance to several classes of acaricides have been reported, forcing researchers to search for alternative measures, such as vaccines against ticks, to ensure tick control while having no or at least low negative impacts on the environment and public health. However, the current commercially available vaccines in different strains of Rhipicephalus microplus are reported to be of low efficacy. Fortunately, reverse vaccinology approaches have shown positive results in the new generation of vaccines. On this basis, a synthetic peptide from the ATAQ protein, which is present in the gut and Malpighi tubes of R. microplus, was synthesized. The ATAQ proteins were isolated, characterized and sequenced from several species of the genus Rhipicephalus. The alignment showed 93.3% identity among DNA sequences of ATAQs from these species. Because of this, immunization trials with this peptide were conducted on mice, rabbits and cattle to evaluate the humoral immune response and the efficacy against Rhipicephalus sanguineus in addition to R. microplus. Based on recent results, we conclude that reverse vaccinology is a promising approach because it is more accurate and faster than conventional methods in the detection of potential antigens to use in anti-tick vaccines. It is not only applicable against R. microplus but also against tick species that play important roles in spreading other diseases. ATAQ proteins should be considered as the antigen in new trials to develop a multi-antigenic vaccine. Although these peptides behave as hapten and are not able to be recognized by the immune system on its own, using carriers and adjuvants helps its presentation and induces strong immune responses. Furthermore, an efficiency of 35% reduction in overall life cycle parameters was reported for R. microplus (98% for ELISA responder animals) and 47% for R. sanguineus. Although not yet enough to prevent the environment to infestation of ticks, this still constitutes a promising strategy that could be applied to integrated measures on tick control and in new research that develops anti-tick vaccines.

Sand flies are recognized as the major vector of canine visceral leishmaniasis. However, in some areas of Brazil where sand flies do not occur, this disease is found in humans and dogs. There has been speculation that ticks might play a role in transmission of canine visceral leishmaniasis and the DNA of Leishmania spp. has been reported in whole ticks. We investigated the presence of Leishmania spp. promastigotes in the intestines, ovaries, and salivary glands of Rhipicephalus sanguineus ticks collected from tick-infested dogs in two cities of Brazil. We used 66 dogs that tested positive and 33 that tested negative for Leishmania spp. according to direct cytological examination assays. Ten ticks were collected from each dog and dissected to collect the intestines, ovaries, and salivary glands for immunohistochemistry (IHC) and diagnostic real-time polymerase chain reaction (RT-PCR). IHC results showed Leishmania spp. in 98, 14, and 8 % of the intestines, ovaries, and salivary glands, respectively. Real-time PCR showed that 89, 41, and 33 % of the tick intestine, ovary, and salivary glands, respectively, were positive for Leishmania spp. The verification of promastigotes of Leishmania spp. by two independent techniques in ticks collected from these urban region dogs showed that there is need for clarification of the role of ticks in the transmission of canine visceral leishmaniasis in Brazil.

Cattle ticks are responsible for great economic losses in cattle farming worldwide, and their main control method, chemicals, has been showing problems, whether resulting from the development of resistant strains of ticks or environmental contamination. Research studies directed toward developing vaccines against ticks are emerging. One way to evaluate those vaccines is to calculate the percentage of efficacy. The aim of this study was to analyze scientific publications archived in PubMed that used this method of assessment and discuss the main factors that may affect its calculation. Thus, 25 articles addressing this subject were selected. The percentage of efficacy was usually calculated in one of two ways, with one considering the reduced fertility of eggs and the other not. The latter method may underestimate the vaccine efficacy, and the most complete formula for calculating the efficacy reflects how much the vaccine actually affects the infestation. In our view, the use of the complete formula for calculating the percentage of efficacy is broader and more representative of the vaccine effect on the tick population.


Carrapatos de bovinos são responsáveis por grandes perdas econômicas para a pecuária bovina mundial e seu principal método de controle, o químico, vem apresentando problemas, seja pelo desenvolvimento de amostras de carrapatos resistentes ou pela contaminação ambiental. Na tentativa de diminuir a utilização dos acaricidas, surgem pesquisas direcionadas ao desenvolvimento de vacinas contra carrapatos. Uma maneira de avaliar essas vacinas é pelo cálculo de percentagem de eficácia. O objetivo deste trabalho foi analisar as publicações científicas indexadas no PubMed que utilizaram este método de avaliação e discutir os principais fatores que podem interferir no seu cálculo. Dessa maneira, selecionaram-se 25 artigos que tratavam desse assunto. A percentagem de eficácia apareceu sendo calculada de duas formas, uma considerando a redução da fertilidade dos ovos e a outra não. Essa última pode subestimar a eficiência da vacina, e a fórmula de cálculo da eficácia mais completa representa o quanto da infestação a vacina realmente reduziu. Em nosso entendimento, a utilização da fórmula completa para o cálculo da percentagem de eficácia é mais abrangente e representativa do efeito da vacina na população de carrapatos.

The Rhipicephalus microplus tick is globally regarded as the most economically important ectoparasite of livestock, and the evolution of resistance to commercial acaricides among cattle tick populations is of great concern. The essential oil derived from Tagetes minuta may be efficacious against cattle tick infestation, and the results of a cattle pen trial using this essential oil for the control of ticks are reported here. The chemical composition of the essential oil was determined by GC-MS and NMR spectroscopy analyses, which revealed the presence of four major components in the essential oil. These components represent more than 70% of the essential oil: limonene (6.96%), ß-ocimene (5.11%), dihydrotagetone (54.10%) and tagetone (6.73%). The results of the cattle pen trial indicated significant differences among the average values of the analyzed biological parameters, including the number of ticks, the average weight of the ticks, the average egg weight per engorged female and larval viability. Treatment with the T. minuta essential oil prepared in this study promoted significant effects on all biological indicators analyzed. Based on the biological indicators, the essential oil showed 99.98% efficacy compared to the control group when used at a 20% concentration. The results obtained in this study suggest that the T. minuta essential oil is a potential R. microplus tick control agent and may be used to mitigate the economic losses caused by tick infestation.

Herein, we report tick species found on wild and domestic animals and in the environment during a one-year sampling period at the Brazilian Farming Research Company beef cattle unit (Embrapa Beef Cattle), which is located within the urban area of Campo Grande, Mato Grosso do Sul, Brazil. From 55 wild hosts including six different species (Nasua nasua, Cebus spp., Cerdocyon thous, Myrmecophaga tridactyla, Tamandua tetradactyla and Dasyprocta aguti), 323 ticks were collected. Amblyomma ovale ticks were found solely on coatis, and Amblyomma nodosum was identified solely on anteaters. No ticks were found on capuchin monkeys. However, Amblyomma cajennense was found on all parasitized host species with the exception of capuchin monkeys. Giant anteaters displayed the highest infestation abundance, with a mean of 53 ticks/animal. Environmental sampling yielded 166 adult A. cajennense ticks. The tick species found on domestic animals (Rhipicephalus (Boophilus) microplus, R. sanguineus, Dermacentor nitens and A. cajennense) were those typically found on these hosts in Brazil. The most prevalent tick species, A. cajennense, was found on both wild and domestic animals and was also prevalent in the environment. Thus, this tick species is the primary vector that allows pathogens to bridge wild and domestic animals in the Cerrado.


Neste trabalho são descritas as espécies de carrapatos de animais selvagens e domésticos e do ambiente coletados por um ano na EMBRAPA Gado de Corte localizado na área urbana de Campo Grande, Mato Grosso do Sul, Brasil. Dos 55 hospedeiros selvagens de seis espécies diferentes (Nasua nasua, Cebus spp.,Cerdocyon thous, Myrmecophaga tridactyla,Tamandua tetradactyla e Dasyprocta aguti) foram coletados 323 carrapatos. Amblyomma ovale foi encontrado apenas em quatis e Amblyomma nodosum apenas sobre tamanduás. Nenhum carrapato foi encontrado sobre macacos-prego. Por outro lado, Amblyomma cajennense foi encontrado em todos os hospedeiros com exceção dos macacos-pregos. A maior abundancia de infestação foi aquela em tamanduás-bandeira com média de 53 carrapatos/animal. No ambiente foram capturados 166 carrapatos, todos da espécie A. cajennense. As espécies de carrapatos em animais domésticos (Rhipicephalus (Boophilus) microplus, Rhipicephalus sanguineus, Dermacentor nitens e A. cajennense) foram aquelas características nestes hospedeiros no Brasil. De forma geral a espécie de carrapato A. cajennense foi a mais prevalente sendo encontrada em animais domésticos e selvagens bem como no ambiente. Portanto, esta é a principal espécie de vetor a estabelecer uma ponte para bioagentes patogênicos entre animais domésticos e selvagens.

O controle de carrapatos que causa impacto na produção de bovinos possui importância econômica para a cadeia produtiva. Neste trabalho objetivou-se testar a atividade acaricida do óleo essencial das folhas e caules de Tagetes minuta contra várias espécies de carrapatos brasileiros, incluindo Rhipicephalus (Boophilus) microplus, Rhipicephalus sanguineus, Amblyomma cajennense e Argas miniatus. A composição química do óleo foi determinada por GC-MS e análises de espectroscopia de RMN, que revelaram a presença de monoterpenos. Na avaliação destas substâncias no controle do carrapato foram empregados os testes de imersão de adulto (TIA) e o de pacote de larvas (TPL) para o extrato de óleo de T. minuta nas concentrações de 2,5%; 5%; 10%; 20% e 40%. Os resultados do TPL e TIA demonstraram que o óleo essencial na concentração de 20% de T. minuta apresenta eficácia superior a 95% nas quatro espécies de carrapato. Estes resultados sugerem que o óleo essencial de T. minuta pode ser usado como um acaricida eficaz e com baixo impacto ambiental.(AU)

The present research investigated the presence of T. evansi antibodies in animals from the subregion of Nhecolandia, in the Pantanal Sul-mato-grossense, by means of an enzyme linked immunosorbent assay (ELISA) and indirect immunofluorescence antibody test (IFAT), and the pattern of polypeptide recognition by sera from experimentally and naturally infected hosts using Western blotting. Serum samples were obtained from bovines (n = 102), horses (n = 98), and dogs (n = 55), and from 32 free-ranging coatis (Nasua nasua). None of the bovines were found positive, while sera from 16 dogs (29%) and 23 horses (23.4%) were positive by ELISA. Sera from 8 coatis (25%) were found positive using IFAT. Western blotting revealed major polypeptides of T. evansi with molecular weight ranging from 74 to 38 kDa. The polypeptides of 66, 48-46, and 38 kDa were identified by sera from experimentally infected bovines, donkeys, dogs, and coatis. The 48-46 and 38 kDa bands were mainly recognized in chronic phase of infection. The antigen with apparent molecular weight of 66 kDa, revealed by antibodies from all experimental animals, was also recognized in sera of horses and dogs from the Pantanal. The 48-46 kDa polypeptide was identified by antibodies from all naturally infected animals and must be further evaluated for use in specific diagnosis of T. evansi infection.
