The onion thrips, Thrips tabaci (Lindeman, 1889), is a cosmopolitan pest of economic importance on a wide range of crops. Despite being one of the most studied thrips species, there is very limited knowledge available about its ability to perceive light. The T. tabaci cryptic species complex consists of a tobacco-associated (T) and two leek-associated (L1, L2) biotypes. We made electroretinogram recordings on the most widespread thelytokous (where unfertilized eggs produce females) T. tabaci L2 biotype and measured attraction to light sources in this biotype as a function of wavelength in behavioural experiments. The spectral sensitivity of the T. tabaci L2 biotype shows a unimodal curve peaking at λmax = 521 nm. Contrary to this spectral sensitivity curve, L2 biotype attraction in an arena is bimodal with local maxima at 368 nm (UV) and 506-520 nm (green) being practically of the same magnitude. Although being similar to the arrhenotokous (where unfertilized eggs produce males) L1 biotype in phototaxis, significant differences regarding photoreceptor cell responses emerged. This study contributes to our understanding of light perception in Thysanoptera as well as to the development of more effective monitoring tools for this economically important pest species.

BACKGROUND: Differences in floral traits, such as petal color, scent, morphology, or nectar quality and quantity, can lead to specific interactions with pollinators and may thereby cause reproductive isolation. Petunia provides an attractive model system to study the role of floral characters in reproductive isolation and speciation. The night-active hawkmoth pollinator Manduca sexta relies on olfactory cues provided by Petunia axillaris. In contrast, Petunia exserta, which displays a typical hummingbird pollination syndrome, is devoid of scent. The two species can easily be crossed in the laboratory, which makes it possible to study the genetic basis of the evolution of scent production and the importance of scent for pollinator behavior. RESULTS: In an F2 population derived from an interspecific cross between P. axillaris and P. exserta, we identified two quantitative trait loci (QTL) that define the difference between the two species' ability to produce benzenoid volatiles. One of these loci was identified as the MYB transcription factor ODORANT1. Reciprocal introgressions of scent QTL were used for choice experiments under controlled conditions. These experiments demonstrated that the hawkmoth M. sexta prefers scented plants and that scent determines choice at a short distance. When exposed to conflicting cues of color versus scent, the insects display no preference, indicating that color and scent are equivalent cues. CONCLUSION: Our results show that scent is an important flower trait that defines plant-pollinator interactions at the level of individual plants. The genetic basis underlying such a major phenotypic difference appears to be relatively simple and may enable rapid loss or gain of scent through hybridization.

Regulation of female mosquito feeding and reproduction plays a central role in their disease-vector competence. In this study we show that Anopheles gambiae mosquitoes engorged on albumin, amino acid and saline meals the same way as on blood, whereas sucrose evoked a typical plant nectar feeding response. Among the artificial diets, only the albumin-containing ones allowed follicular development. The target of rapamycin (TOR)/p70 S6 kinase (S6K) pathway has been identified as an essential nutrient-sensing tool controlling egg development in mosquitoes under the control of regulating inputs from the insulin pathway. We assayed the early response of TOR, S6K, tuberous sclerosis (TSC2), insulin receptor (INR) and two insulin-like peptides (ILPs) by quantitative real-time PCR assessment of mRNA levels and immunoblotting of phosphorylated active TOR and S6K in An. gambiae ovary and brain 3 h after engorgement. We show that transcript levels of s6k and members of the insulin pathway are readily affected by nutrients (especially one ILP in the head) and that the TOR/S6K phosphorylation is able to react quickly to a meal to an extent which depends on the true nutritive value.

In the genus Petunia, distinct pollination syndromes may have evolved in association with bee-visitation (P. integrifolia spp.) or hawk moth-visitation (P. axillaris spp). We investigated the extent of congruence between floral fragrance and olfactory perception of the hawk moth Manduca sexta. Hawk moth pollinated P. axillaris releases high levels of several compounds compared to the bee-pollinated P. integrifolia that releases benzaldehyde almost exclusively. The three dominating compounds in P. axillaris were benzaldehyde, benzyl alcohol and methyl benzoate. In P. axillaris, benzenoids showed a circadian rhythm with an emission peak at night, which was absent from P. integrifolia. These characters were highly conserved among different P. axillaris subspecies and P. axillaris accessions, with some differences in fragrance composition. Electroantennogram (EAG) recordings using flower-blends of different wild Petunia species on female M. sexta antennae showed that P. axillaris odours elicited stronger responses than P. integrifolia odours. EAG responses were highest to the three dominating compounds in the P. axillaris flower odours. Further, EAG responses to odour-samples collected from P. axillaris flowers confirmed that odours collected at night evoked stronger responses from M. sexta than odours collected during the day. These results show that timing of odour emissions by P. axillaris is in tune with nocturnal hawk moth activity and that flower-volatile composition is adapted to the antennal perception of these pollinators.

The behaviour of Ixodes ricinus nymphs was recorded in 10-day experiments using computer-assisted video-tracking, in the absence of any host stimuli. These ticks switch spontaneously from questing in a desiccating atmosphere to quiescence in a water-saturated atmosphere after dark. Quantification of both questing and quiescence duration demonstrates that questing duration is inversely related to saturation deficit whereas quiescence duration is not. Distance walked after quiescence increased with desiccating conditions, while the distance walked after questing remained unchanged. Almost all locomotor activities of I. ricinus occurred during darkness under either a 14 h:10 h L:D or a 8 h:4 h L:D cycle. We established that all life stages of I. ricinus are equipped to sense shifts in light intensity with bilaterally placed strings of photoreceptors. This permits I. ricinus to use onset of darkness to trigger mobility when desiccation risk is reduced in nature.
