OBJECTIVE: Somatotroph tumors are the second most common type of pituitary neuroendocrine tumors, which can be further classified into 2 subtypes-densely granulated somatotroph tumors (DGSTs) and sparsely granulated somatotroph tumors (SGSTs). The aim of this study was to investigate the clinical significance of the 2 subtypes in a retrospective analysis. METHODS: From the database of the Ningbo Clinical Pathology Diagnosis Center, we collected patients diagnosed with pituitary somatotroph tumors. We then compiled pertinent clinical and radiological data and proceeded with histopathological examination involving hematoxylin-eosin staining and immunohistochemical staining. Subsequent analysis compared the 2 subtypes using either χ2 test or Fisher exact test. RESULTS: We analyzed 40 cases of somatotroph tumors, 18 cases DGSTs and 22 SGSTs. Male-to-female ratio was 5:4 for DGSTs and 4:7 for SGSTs. Mean age was 52.83 years for DGSTs and 47.18 years for SGSTs. Statistically significant differences were observed between the DGST and SGST groups in invasiveness (P = 0.0267) and postoperative remission (P = 0.007). Cells of both DGSTs and SGSTs exhibited coexpression of PIT1, growth hormone, and CAM5.2, although the patterns of CAM5.2 expression differed between the 2 subtypes. CONCLUSIONS: The efficacy of CAM5.2 staining in distinguishing between DGSTs and SGSTs was demonstrated. SGSTs, with their increased invasiveness and lower remission rate, are a high-risk subtype. The histological subtype of somatotroph tumors plays a crucial role in guiding treatment decisions and prognostic evaluation in affected patients.

Pancreatic ductal adenocarcinoma (PDAC) is one of the most lethal cancers and its dismal prognosis indicates the urgent need to elucidate the potential oncogenic mechanisms. SIRT7 is a classic NAD+-dependent deacetylase that stabilizes the transformed state of cancer cells. However, its functional roles in PDAC are still unclear. Here, we found that SIRT7 expression is upregulated and predicts poor prognosis in PDAC. Then we screened the new interacting proteins of SIRT7 by mass spectrometry and the results showed that SIRT7 can interact with O-GlcNAc transferase (OGT). O-GlcNAcylation stabilizes the SIRT7 protein by inhibiting its interaction with REGγ to prevent degradation, and hyper-O-GlcNAcylation in pancreatic cancer cells leads to hypoacetylation of H3K18 via SIRT7, which promotes transcriptional repression of several tumour suppressor genes. In addition, SIRT7 O-GlcNAcylation at the serine 136 residue (S136) is required to maintain its protein stability and deacetylation ability. In vivo and in vitro experiments showed that blocking SIRT7 O-GlcNAcylation at S136 attenuates tumour progression. Collectively, we demonstrate that O-GlcNAcylation is an important post-translational modification of SIRT7 in pancreatic cancer cells, and elucidating this mechanism of SIRT7 is expected to pave the way for the development of novel therapeutic methods in the future.

The co-digestion of corn straw and sewage sludge with different additives (biochar, magnetic biochar, Fe3O4) were investigated. The highest cumulative methane yield of 245.15 mL/g VSadded was obtained with the Fe3O4 addition ratio of 5 g/kg, which was 60.47% higher than that of the control run (without additives). The lag phase time was shortened from 5.46 to 3.82 days with a biochar dosage of 5 g/kg. The performance of Fe3O4 on methane production from the co-digestion process was better than that of the biochar and magnetic biochar. The direct interspecies electron transfer (DIET) was enhanced with regard to the increased concentration of acetic acid and decreased concentration of propionic acid. Microbial community analysis showed that the Geobacter and Methanosarcina were selectively enriched on the surface of Fe3O4, promoting the DIET and acetoclastic methanogenesis pathway. The cost-benefit analysis proved that the strategy of recycling Fe3O4 additive has the best economic benefit.

Plant growth-promoting rhizobacteria (PGPR) are a specific category of microbes that improve plant growth and promote greater tolerance to metal stress through their interactions with plant roots. We evaluated the effects of phytoremediation combining the cadmium accumulator Solanum nigrum L. and two Cd- and Pb-resistant bacteria isolates. To understand the interaction between PGPR and their host plant, we conducted greenhouse experiments with inoculation treatments at Nanjing Agricultural University (Jiangsu Province, China), in June 2018. Two Cd- and Pb-resistant PGPR with various growth-promoting properties were isolated from heavy metal-contaminated soil. 16S rRNA analyses indicated that the two isolates were Bacillus genus, and they were named QX8 and QX13. Pot experiments demonstrated that inoculation may improve the rhizosphere soil environment and promote absorption of Fe and P by plants. Inoculation with QX8 and QX13 also enhanced the dry weight of shoots (1.36- and 1.7-fold, respectively) and roots (1.42- and 1.96-fold) of plants growing in Cd- and Pb-contaminated soil, and significantly increased total Cd (1.28-1.81 fold) and Pb (1.08-1.55 fold) content in aerial organs, compared to non-inoculated controls. We also detected increases of 23% and 22% in the acid phosphatase activity of rhizosphere soils inoculated with QX8 and QX13, respectively. However, we did not detect significant differences between inoculated and non-inoculated treatments in Cd and Pb concentrations in plants and available Cd and Pb content in rhizosphere soils. We demonstrated that PGPR-assisted phytoremediation is a promising technique for remediating heavy metal-contaminated soils, with the potential to enhance phytoremediation efficiency and improve soil quality.

BACKGROUND: Pancreatic cancer is a highly malignant tumor of the digestive system. Early pancreatic cancer is often difficult to diagnosis due to its atypical clinical symptoms. Patients with pancreatic cancer have a very poor prognosis because they have lost the opportunity for radical surgical tumor resection and they are less sensitive to the clinically used radiotherapy and chemotherapy. METHODS: In this study, a peptide targeting pancreatic cancer cells was screened by phage display technology, and its targeting property was evaluated in vitro using PANC1 cells by fluorescence imaging and flow cytometry. Furthermore, the targeting peptide was conjugated to the pro-apoptotic KLAKLAKKLAKLAK (KLA), the fusion peptide and its targeting ability that allowing KLA to specifically enter pancreatic tumor cells in vitro and in vivo was confirmed by fluorescence imaging and in vivo imaging system (IVIS). Its mechanism was determined using flow cytometry, mitochondrial membrane potential evaluation and Western blot. The inhibitory effect on pancreatic tumor growth and toxic effects were evaluated by animal experiment. RESULTS: Due to the internalization facilitated by the targeting mechanism of the targeting peptide, KLA specifically entered pancreatic cancer cells, destroyed mitochondria and induced apoptosis. The fusion peptide and its targeting ability that allowing KLA to specifically enter pancreatic tumor cells and exert a significant inhibitory effect on pancreatic tumor growth with reduced toxic effects. CONCLUSION: This approach possesses potential advantages in the clinical diagnosis and treatment of pancreatic cancer.

Conventional chemical soil amendments and novel material biochars have been widely reported for the immobilisation of cadmium (Cd) and lead (Pb) in polluted soil. However, information regarding their comparative effectiveness is poor. In the present study, rice husk biochar (RHB) was compared with two chemical soil amendments including hydroxyapatite (HAP) and hydrated lime (HDL) for their effectiveness to enhance plant growth and the reduction of Cd uptake and translocation by Triticum aestivum L. grown in heavy-metal-polluted soil. Compared with control and two chemical soil amendments, RHB rapidly improved wheat growth. The HAP, HDL, and RHB treated plants retained Cd and Pb in roots and restricted their translocation. The RHB treatment had the best effect on growth, yield promotion and the reduction of Cd and Pb in wheat grain. Furthermore, the soils treated with RHB and HAP showed lower DTPA-extracted Cd concentrations, and the maximum reduction was observed in HAP-amended soil. However, the DTPA-extracted Pb concentration was not significantly decreased after the application of two chemical soil amendments for 40 days; the maximum reduction was found in soil treated with RHB for 80 days. In all treatments, Cd in post-harvest soil was mainly present in exchangeable, carbonate bound, and Fe-Mn oxide Cd, while the dominant chemical form of Pb was Fe-Mn oxide Pb. Three soil amendments application decreased exchangeable and organic bound- Cd and Pb levels. HAP and RHB displayed significantly immobilisation for soil Cd and reduced translocation of heavy metal as well as its availability in soil, but the HAP had significant inhibition on growth of wheat in contaminated soil. Therefore, RHB shows a promising potential for the reduction of Cd and Pb bioaccumulation in grains from wheat grown on heavy-metal-polluted soils.

Cancer-associated fibroblasts (CAFs), which are an important component of the tumor microenvironment, have been identified in the blood circulation of patients with cancer metastasis, and metastatic cancer cells can recruit circulating CAFs. However, primary carcinoma sites usually regulate the behavior of metastatic cancer cells through exosomes. Here, we hypothesized that cancer-derived exosomes could enhance CAF recruitment. Exosomes secreted by pancreatic cancer cells (PANC-1 and MIA PaCa-2) were isolated and characterized. The ability of pancreatic cancer to recruit pancreatic stellate cells (PSCs) was assessed with Transwell assays in vitro and bioluminescent imaging in a mouse model in vivo, and the underlying molecular mechanism was also investigated. The results showed that pancreatic cancer cell-derived exosomes (Exo-Pan and Exo-Mia) promoted the pancreatic cancer recruitment of PSCs. This effect was mediated partially by the transfer of the exosomal protein Lin28B to the recipient cells to activate the Lin28B/let-7/HMGA2/PDGFB signaling pathway. These results suggested that exosomes derived from local cancer could promote the formation of distant metastases through transferring the exosomal protein Lin28B to the metastatic cancer cells.

Traditional metal chelators, such as ethylenediaminetetraacetic acid (EDTA), have been gradually replaced due to their poor biodegradability in soil and high risk of heavy metal leaching into groundwater, which pose high environmental risks to the health of humans and animals. In this study, a liquid amino acid fertilizer (LAAF, waste proteins from hydrolysates of animal carcasses) and polyaspartate (PASP) were used as additives to enhance the phytoextraction of cadmium (Cd) and lead (Pb) from contaminated soil. We conducted pot experiments to investigate the phytoextraction capacity of Solanum nigrum, a Cd accumulator, grown on soil highly contaminated with Cd and Pb in the absence (as controls) or presence of PASP and LAAF. Both PASP and LAAF significantly improved plant growth, Cd accumulation, and total Cd and Pb content in S. nigrum shoots and roots. PASP and LAAF application promoted Cd translocation from roots to shoots in S. nigrum and Cd bio-accessibility in rhizosphere soils, but this was not the case for Pb. Both PASP and LAAF increased Cd and Pb phytoextraction by S. nigrum plants, and Cd phytoextraction was more effective in LAAF-assisted S. nigrum than in PASP-assisted S. nigrum. These findings demonstrate that the low cost and ecofriendly features of recycled waste proteins make them good candidates for chelant-enhanced phytoextraction from heavy metal-contaminated soils.
