RESUMO

The foot-and-mouth disease virus is a highly contagious and economically devastating virus of cloven-hooved animals, including cattle, buffalo, sheep, and goats, causing reduced animal productivity and posing international trade restrictions. For decades, chemically inactivated vaccines have been serving as the most effective strategy for the management of foot-and-mouth disease. Inactivated vaccines are commercially produced in cell culture systems, which require successful propagation and adaptation of field isolates, demanding a high cost and laborious time. Cell culture adaptation is chiefly indebted to amino acid substitutions in surface-exposed capsid proteins, altering the necessity of RGD-dependent receptors to heparan sulfate macromolecules for virus binding. Several amino acid substations in VP1, VP2, and VP3 capsid proteins of FMDV, both at structural and functional levels, have been characterized previously. This literature review combines frequently reported amino acid substitutions in virus capsid proteins, their critical roles in virus adaptation, and functional characterization of the substitutions. Furthermore, this data can facilitate molecular virologists to develop new vaccine strains against the foot-and-mouth disease virus, revolutionizing vaccinology via reverse genetic engineering and synthetic biology.

BACKGROUND: Multidrug resistance (MDR) in the family Enterobacteriaceae is a perniciously increasing threat to global health security. The discovery of new antimicrobials having the reversing drug resistance potential may contribute to augment and revive the antibiotic arsenal in hand. This study aimed to explore the anti-Enterobacteriaceae capability of bioactive polyphenols from Punica granatum (P. granatum) and their co-action with antibiotics against clinical isolates of Enterobacteriaceae predominantly prevalent in South Asian countries. METHODS: The Kandhari P. granatum (Pakistani origin) extracts were tested for anti-Enterobacteriaceae activity by agar well diffusion assay against MDR Salmonella enterica serovar Typhi, serovar Typhimurium and Escherichia coli. Predominant compounds of active extract were determined by mass spectrometry and screened for bioactivity by agar well diffusion and minimum inhibitory concentration (MIC) assay. The active punicalagin was further evaluated at sub-inhibitory concentrations (SICs) for coactivity with nine conventional antimicrobials using a disc diffusion assay followed by time-kill experiments that proceeded with SICs of punicalagin and antimicrobials. RESULTS: Among all P. granatum crude extracts, pomegranate peel methanol extract showed the largest inhibition zones of 25, 22 and 19 mm, and the MICs as 3.9, 7.8 and 7.8 mg/mL for S. typhi, S. typhimurium and E. coli, respectively. Punicalagin and ellagic acid were determined as predominant compounds by mass spectrometry. In plate assay, punicalagin (10 mg/mL) was active with hazy inhibition zones of 17, 14, and 13 mm against S. typhi, S. typhimurium and E. coli, respectively. However, in broth dilution assay punicalagin showed no MIC up to 10 mg/mL. The SICs 30 µg, 100 µg, and 500 µg of punicalagin combined with antimicrobials i.e., aminoglycoside, ß-lactam, and fluoroquinolone act in synergy against MDR strains with % increase in inhibition zone values varying from 3.4 ± 2.7% to 73.8 ± 8.4%. In time-kill curves, a significant decrease in cell density was observed with the SICs of antimicrobials/punicalagin (0.03-60 µg/mL/30, 100, 500 µg/mL of punicalagin) combinations. CONCLUSIONS: The P. granatum peel methanol extract exhibited antimicrobial activity against MDR Enterobacteriaceae pathogens. Punicalagin, the bacteriostatic flavonoid act as a concentration-dependent sensitizing agent for antimicrobials against Enterobacteriaceae. Our findings for the therapeutic punicalagin-antimicrobial combination prompt further evaluation of punicalagin as a potent activator for drugs, which otherwise remain less or inactive against MDR strains.

Duodenal ulcer perforation, a frequent surgical emergency, needs simple closure with indirect Graham's Omentopexy which is effective with excellent results in majority of cases despite patients' late presentation. The objective of the study was to determine the frequency of postoperative complications of perforated duodenal ulcer, conducted in the Surgery Department, Jinnah Postgraduate Medical Centre, Karachi, from March 20, 2018 to September 20, 2018. The study was a descriptive case series of 108 patients of both genders with perforated duodenal ulcer > 1 week old with ASA score I & II. Patients with trauma and comorbidities were excluded. The patients underwent laparotomy and peritoneal toilet, and after noting the site of perforation indirect Graham's Omentopexy was performed. Complications like duodenal fistula, peritonitis, and paralytic ileus, and patient's death within 10 days of surgery were noted. Age ranged from 18 to 50 years with mean age of 35.027±5.13 years, mean weight 71.120±12.77 kg, mean height 1.541 ±0.09 metres, mean BMI 29.975±4.99 kg/m2, and the mean duration of complaint was 4.194±1.30 weeks. Male predominance in 75 (69.4%) patients. Duodenal fistula was seen in 10 (9.3%) patients, peritonitis 12 (11.1%), paralytic ileus 14 (13%) and mortality was in 11 (10.2%) patients.

SARS-CoV-2 enters the host cell through the ACE2 receptor and replicates its genome using an RNA-Dependent RNA Polymerase (RDRP). The functional RDRP is released from pro-protein pp1ab by the proteolytic activity of Main protease (Mpro) which is encoded within the viral genome. Due to its vital role in proteolysis of viral polyprotein chains, it has become an attractive potential drug target. We employed a hierarchical virtual screening approach to identify small synthetic protease inhibitors. Statistically optimized molecular shape and color-based features (various functional groups) from co-crystal ligands were used to screen different databases through various scoring schemes. Then, the electrostatic complementarity of screened compounds was matched with the most active molecule to further reduce the hit molecules' size. Finally, five hundred eighty-seven molecules were docked in Mpro catalytic binding site, out of which 29 common best hits were selected based on Glide and FRED scores. Five best-fitting compounds in complex with Mpro were subjected to MD simulations to analyze their structural stability and binding affinities with Mpro using MM/GB(PB)SA models. Modeling results suggest that identified hits can act as the lead compounds for designing better active Mpro inhibitors to enhance the chemical space to combat COVID-19.Communicated by Ramaswamy H. In the current study, we have evaluated the protective efficacy of the 'insertion domain' which is commonly found in the capsid penton base protein of many adenoviruses. Using the 'insertion domain' of the penton base protein of a representative fowl adenovirus, fowl adenovirus serotype 4 (FAdV-4), we find that the 'insertion domain' can readily be expressed in a soluble form in the bacterial system, and can be purified in sufficient quantities through simple chromatographic methods. We demonstrate that the 'insertion domain', when employed as a subunit vaccine candidate, provides complete protection against hydropericardium syndrome, caused by FAdV-4, in chickens. The data presented here indicate that the protein, adjuvanted with Montanide™ ISA71 VG, provides complete protection in chickens against a lethal FAdV-4 challenge after administration of two doses (100 µg of the protein per dose) two weeks apart (the first dose at the 7th day of life and a booster dose at the age of 21 days). Furthermore, the purified protein can be stored at low temperatures without any observable loss in the protein integrity up to one year, tested so far. Due to the conserved nature of the 'insertion domain' across the penton base protein of fowl adenoviruses, it is suggested that homologous insertion domains could be employed as highly stable and cost-effective subunit vaccine candidates against infections caused by respective fowl adenoviruses.

The generation of laser-induced plasma at the gas-liquid interface provides many fundamental and interesting scientific phenomena such as ionization, sharp explosion, shock wave radiation, bubble creation, and water splitting. However, despite the extensive research in this area, there is no reference on the effect of the surrounding environment on the chemical processes that occur during the laser-induced plasma-water interaction. In this work, we investigate the effect of the surrounding gas environment on femtosecond laser-induced plasma when generated at the pure water-gas interface. Ultrashort laser pulses were applied to water in the presence of air and N2 and Ar gas environments. Formation of a significant number of nitrate-based species in water was observed after exposure to femtosecond laser-induced plasma in air and N2 environments. The detected NO3 ions formed in the laser-treated water led to the appearance of an absorption peak in the UV range, a significant decrease in the water pH value, and a significant increase in water's electrical conductivity. All induced properties of water were stable for 3 months of monitoring after laser treatment. Our work shows that the generation of laser-induced plasma in water propagating into a gaseous medium facilitates the interaction between the two media, as a result of which the compositions of substances present in the gaseous medium can be dissolved in water without increasing the gas pressure. The presented approach may find applications in areas such as water purification, material synthesis, and environmental stewardship.

Background Chronic cholecystitis is inflammation of the gall bladder usually caused by stones. The aim was to find out the prevalence of mucocele and empyema in chronic cholecystitis using cholecystectomy findings and histopathological reports. Methodology This was a cross-sectional observational study conducted in Surgical Ward 1, Jinnah Postgraduate Medical Center, Karachi from December 2019 to December 2021 for two years. Patients above 12 years of age diagnosed with chronic cholecystitis with cholelithiasis on clinical examination and investigations were included. Patients who were diagnosed with acute cholecystitis, mucocele, or empyema on clinical examination and ultrasonography were excluded from the study. Laparoscopic cholecystectomy was done and operative findings were noted. Gall bladder specimens were examined for mucocele and empyema and were sent for histopathology. Results were recorded and analyzed. Results There were 241 patients diagnosed with chronic cholecystitis with cholelithiasis on clinical examination and investigations. On examination, tenderness in the right hypochondrium was absent in all patients. Chronic cholecystitis was proved on histopathology in 231 patients (95.85%). Other findings diagnosed on peroperative findings and histopathology reports were strawberry gall bladder (2.41%), empyema (0.83%), mucocele (0.41%), and polyp (0.41%). Two hundred eight patients were female (86.31%), 33 were male(13.69%). The male to female ratio was 1:6.43. The average age was 31 years. Conclusion Inflammation and fibrosis of the gallbladder around Calot's triangle increase the chances of vascular and common bile duct injury. In such cases, cholecystectomy can become difficult. It was concluded that empyema, mucocele, and strawberry gall bladder could be found in chronic cholecystitis, and cholecystectomy becomes difficult in such cases.

Introduction The aim of this study was to compare epidemiological characteristics of breast cancer in young adolescent women (YAW) versus older women (OW). Methods This was a cross-sectional prospective observational study, conducted in Ward 3, Jinnah Postgraduate Medical Center, Karachi, Pakistan, from September 2021 to February 2022. A total of 120 female patients were recruited in this study from the Outpatient Department of Jinnah Postgraduate Medical Center, out of which 22 patients were below the age of 40 years and 98 were above 40 years. For breast cancer diagnosis, we used the triple assessment method involving clinical examination, radiology, and histopathology. Diagnosed patients were further evaluated for hormonal status and metastatic workup. Results were noted on a performa, and differences between both age groups were analyzed. Results Out of 120 patients, 22 were younger than 40 years and 98 were older than 40 years. YAW used to present late after the appearance of symptoms. Patients of both age groups mostly presented with breast lumps (68.18% in YAW and 81.6% in OW). YAW presented with larger sizes of lumps and with more nodal involvement as compared to OW. BI-RADS IV (Breast Imaging Reporting and Data System Category IV) was the most commonly observed (27.27% in YAW and 48.97% in OW) mammographic finding in both age groups. Invasive ductal carcinoma was the most common histological type in both age groups (72.73% in YAW and 76.53% in OW). The triple-negative disease was more commonly found in YAW than OW (40.91% in YAW vs 21.43% in OW). We found that usually YAW presented at advanced stages (stages III and IV, 54.55%) and higher grades (grade III, 63.63%). Conclusion Breast cancer in young patients is rare but more aggressive with higher grades, advanced stages, and poor prognostic features. Heredity is mainly the risk factor in young breast cancer patients. There should be proper screening programs for high-risk group for early diagnosis and prompt treatment. Other age-specific concerns such as psychological impact of disease should be addressed as well.

The current study was dedicated to finding the effect of soil amendments (biochar and compost) on plants belonging to Poaceae and Fabaceae families. Plants selected for the phytoremediation experiment included wheat (Triticum aestivum), maize (Zea mays), white clover (Trifolium repens), alfalfa (Medicago sativa), and ryegrass (Lolium multiflorum). The physiological and microbial parameters of plants and soil were affected negatively by the 4 % TPHs soil contamination. The studied physiological parameters were fresh and dried biomass, root and shoot length, and chlorophyll content. Microbial parameters included root and shoot endophytic count. Soil parameters included rhizospheric CFUs and residual TPHs. Biochar with wheat, maize, and ryegrass (Fabaceae family) and compost with white clover and alfalfa (Poaceae family) improved plant growth parameters and showed better phytoremediation of TPHs. Among different plants, the highest TPH removal (68.5 %) was demonstrated by ryegrass with compost, followed by white clover with biochar (68 %). Without any soil amendment, ryegrass and alfalfa showed 59.55 and 35.21 % degradation of TPHs, respectively. Biochar and compost alone removed 27.24 % and 6.01 % TPHs, respectively. The interactive effect of soil amendment and plant type was also noted for studied parameters and TPHs degradation.

BACKGROUND: Classical homocystinuria (HCU) is an autosomal recessive inborn error of metabolism, which is caused by the cystathionine-ß-synthase (CBS: encoded by CBS) deficiency. Symptoms of untreated classical HCU patients include intellectual disability (ID), ectopia lentis and long limbs, along with elevated plasma methionine, and homocysteine. METHODS: A total of 429 ID patients (age range: 1.6-23 years) were sampled from Northern areas of Punjab, Pakistan. Biochemical and genetic analyses were performed to find classical HCU disease in ID patients. RESULTS: Biochemically, nine patients from seven unrelated families were identified with high levels of plasma methionine and homocysteine. Targeted exonic analysis of CBS confirmed seven causative homozygous mutations; of which three were novel missense mutations (c.451G>T; p.Gly151Trp, c.975G>C; p.Lys325Asn and c.1039 + 1G>T splicing), and four were recurrent variants (c.451 + 1G>A; IVS4 + 1 splicing, c.770C>T; p.Thr257Met, c.808_810del GAG; p.Glu270del and c.752T>C; p.Leu251Pro). Treatment of patients was initiated without further delay with pyridoxine, folic acid, cobalamin, and betaine as well as dietary protein restriction. The immediate impact was noticed in behavioral improvement, decreased irritability, improved black hair color, and socialization. Overall, health outcomes in this disorder depend on the age and symptomatology at the time of treatment initiation. CONCLUSIONS: With personalized treatment and care, such patients can reach their full potential of living as healthy a life as possible. This screening study is one of the pioneering initiatives in Pakistan which would help to minimize the burden of such treatable inborn errors of metabolism in the intellectually disabled patients.

Blunt abdominal trauma (BAT) refers to injuries without wounds entering the peritoneal cavity due to road traffic accidents (RTA) and falls, as a result of collision or counter collision. The objective of the present study was to determine the frequency of patients with visceral injuries in blunt abdominal trauma. This study was carried out in the Department of Surgery, including ward-3 of Jinnah Post Graduate Medical Centre, Karachi, from November 2017 till November 2020. The study design was descriptive case series. During the study period, the data of 112 patients was collected, which comprised of 102 males and 10 females. All the patients between 12 to 65 years of age (mean age:31.84±13.14 years) presenting to the emergency with < 24 hours of abdominal trauma, were included in the study. Organs involved during blunt abdominal trauma were observed and frequency was recorded. Liver injuries were found in 48(42.28%) patients, intestinal injuries in 40(35.7%), splenic injuries in 32(28.5%), kidney injuries in 24(21.4%), pancreatic injuries in 8(7.1%) and stomach injuries in 4(3.5%) patients. Grade 4 liver injury was found in 80(71%) patients, Grade 2 and 3 splenic injuries in 56(50%) patients whereas 47(42%) patients had grade 1 kidney injury. Liver was found to be the most frequent organ involved, followed by intestine and spleen.

The direct discharge of azo dyes and/or their metabolites into the environment may exert toxic, mutagenic, and carcinogenic effects on exposed fauna and flora. In this study, we analyzed the metabolites produced during the degradation of an azo dye namely Reactive Black 5 (RB5) in the bacterial-augmented floating treatment wetlands (FTWs), followed by the investigation of their underlying toxicity. To this end, a FTWs system was developed by using a common wetland plant Phragmites australis in the presence of three dye-degrading bacteria (Acinetobacter junii strain NT-15, Pseudomonas indoloxydans strain NT-38, and Rhodococcus sp. strain NT-39). We found that the FTW system effectively degraded RB5 into at least 20 different metabolites with the successful removal of color (95.5%) from the water. The fish toxicity assay revealed the nontoxic characteristics of the metabolites produced after dye degradation. Our study suggests that bacterially aided FTWs could be a suitable option for the successful degradation of azo dyes, and the results presented in this study may help improve the overall textile effluent cleanup processes.

This study investigates the potential of crude oil degrading capabilities of biosurfactant-producing strains of Pseudomonas aeruginosa MF069166 and Meyerozyma sp. MF138126. P. aeruginosa produced mono-/di-rhamnolipids congeners whereas, Meyerozyma sp. produced acidic and lactonic forms of sophorolipids with crude oil. The values of critical micelle concentrations of rhamnolipids and sophorolipids were 40 mg/L and 50 mg/L with reductions in surface tension of water to 29 mN/m and 33 mN/m. Dynamic light scattering revealed that the average diameter of micellar aggregates of rhamnolipids ranged between 300 and 350 nm and the average size of sophorolipids micelles was 309 nm and 380 nm. Biosurfactants from P. aeruginosa and Meyerozyma sp. exhibited emulsification activities of 87% and 84% in crude oil. Cell surface hydrophobicity of both strains was higher in the presence of hydrophobic contaminants. The biosurfactants showed stability under varying pH, NaCl concentrations and temperatures. Gravimetric and GC-MS analyses demonstrated that P. aeruginosa degraded 91% of the petroleum hydrocarbons while Meyerozyma sp. showed 87% biodegradation efficiency. P. aeruginosa and Meyerozyma sp. have also been found to degrade halogen-containing compounds and showed excellent crude oil degradation efficiency. It is concluded that both strains have high potential of applications in the bioremediation of hydrocarbons-contaminated sites.

Adenoviruses cause economically important diseases in vertebrates. Effective vaccines against adenoviral diseases are currently lacking. Here, we report a highly conserved epitopic region on hexon proteins of adenoviruses that generate a strong immune response when used as a virus-like-particle (VLP) vaccine, produced by inserting the epitopic region into the core protein of hepatitis B virus. For evaluation of its protective efficacy, the epitopic region from a representative adenovirus, fowl adenovirus serotype 4 (FAdV-4), was tested as a VLP vaccine which conferred 90% protection against challenge with a virulent FAdV-4 isolate in chickens. Importantly, such a high level of protection is not achieved when the epitopic region is employed as a part of a subunit vaccine. As the sequence and the structure of the epitopic region are highly conserved in hexon proteins of adenoviruses, the epitopic region could be employed as a promising VLP vaccine candidate against adenoviral diseases, in general.

There is lot of research work at enhancing the performance of energy conversion and energy storage devices such as solar cells, supercapacitors, and batteries. In this regard, the low bandgap and a high absorption coefficient of CdSe thin films in the visible region, as well as, the low electrical resistivity make them ideal for the next generation of chalcogenide-based photovoltaic and electrochemical energy storage devices. Here, we present the properties of CdSe thin films synthesized at temperatures (below 100°C using readily available precursors) that are reproducible, efficient and economical. The samples were characterized using XRD, FTIR, RBS, UV-vis spectroscopy. Annealed samples showed crystalline cubic structure along (111) preferential direction with the grain size of the nanostructures increasing from 2.23 to 4.13 nm with increasing annealing temperatures. The optical properties of the samples indicate a small shift in the bandgap energy, from 2.20 to 2.12 eV with a decreasing deposition temperature. The band gap is suitably located in the visible solar energy region, which make these CdSe thin films ideal for solar energy harvesting. It also has potential to be used in electrochemical energy storage applications.

BACKGROUND: Dysphagia after occipitocervical fixation (OCF) is a complex phenomenon and revision surgery in this context involves difficult decision-making. The pathogenesis is explored and surgical strategies discussed. A surgical strategy that has not been described before in the management of this condition, is discussed with two illustrative cases. METHODS: Two cases are presented where dysphagia occurred after OCF for C1/C2 instability. The preoperative imaging was not available to determine whether the optimal craniocervical angle had been achieved. Both had revision surgery with removal of the cranial fixation and fusion to the atlas instead. One of the cases had the revision surgery more than 10 years after the original OCF. RESULTS: The dysphagia recovered after the revision surgery in both cases. The patients gained weight and reported more satisfaction with their posture and head movements compared to before the revision surgery. There was no head ptosis and instead, patients reported better forward gaze and head position. CONCLUSION: In cases of C1/C2 instability, it is preferable to perform C1/C2 fusion rather than OCF. If performing an OCF, care must be taken to fix the head at the same O-C2 angle as preoperative. When an OCF done for C1 C2 instability ends up causing dysphagia, it is feasible and effective to shorten the rostral construct by removing the cranial fixation and fusing to C1 instead.

BACKGROUND: Hepatitis C virus genotype 3a (HCV G3a) is highly prevalent in Pakistan. Due to the elevated cost of available Food and Drug Administration-approved drugs against HCV, medicinal natural products of potent antiviral activity should be screened for the cost-effective treatment of the disease. Furthermore, from natural products, active compounds against vital HCV proteins like non-structural protein 3 (NS3) protease could be identified to prevent viral proliferation in the host. AIM: To develop cost-effective HCV genotype 3a NS3 protease inhibitors from citrus fruit extracts. METHODS: Full-length NS3 without co-factor non-structural protein 4A (NS4A) and codon optimized NS3 protease in fusion with NS4A were expressed in Escherichia coli. The expressed protein was purified by metal ion affinity chromatography and gel filtration. Citrus fruit extracts were screened using fluorescence resonance energy transfer (FRET) assay against the protease and polyphenols were identified as potential inhibitors using electrospray ionization-mass spectrometry (MS)/MS technique. Among different polyphenols, highly potent compounds were screened using molecular modeling approaches and consequently the most active compound was further evaluated against HCV NS4A-NS3 protease domain using FRET assay. RESULTS: NS4A fused with NS3 protease domain gene was overexpressed and the purified protein yield was high in comparison to the lower yield of the full-length NS3 protein. Furthermore, in enzyme kinetic studies, NS4A fused with NS3 protease proved to be functionally active compared to full-length NS3. So it was concluded that co-factor NS4A fusion is essential for the purification of functionally active protease. FRET assay was developed and validated by the half maximal inhibitory concentration (IC50) values of commercially available inhibitors. Screening of citrus fruit extracts against the native purified fused NS4A-NS3 protease domain showed that the grapefruit mesocarp extract exhibits the highest percentage inhibition 91% of protease activity. Among the compounds identified by LCMS analysis, hesperidin showed strong binding affinity with the protease catalytic triad having S-score value of -10.98. CONCLUSION: Fused NS4A-NS3 protease is functionally more active, which is effectively inhibited by hesperidin from the grapefruit mesocarp extract with an IC50 value of 23.32 µmol/L.

Oil-water separation using super-wetting and the selective permeability of membranes for oil or water has great ecological and economic significance. We report on the transition of wettability response, from superhydrophilic underwater-superoleophobic to superhydrophobic-superoleophilic state, by nanostructuring stainless steel and copper meshes using ultrashort femtosecond laser pulses. Our approach is environment-friendly, chemical free, and efficient as it exploits the benefit of aging the processed samples in a high vacuum environment. We optimized the laser scanning parameters, mesh pore size, and aging conditions to produce membranes exhibiting an extraordinary separation efficiency of 98% for the oil-water mixture. A variation in the water and oil contact angles for different meshes is presented as a function of the laser scanning speed. Stainless steel meshes with 150 µm pore size and copper meshes with 100 µm pore size have demonstrated an excellent wettability response for oil and water phases. Vacuum aging causes rapid chemisorption of hydrocarbons on laser-structured surfaces in the absence of water molecules, rapidly transforming the wetting state from superhydrophilic to superhydrophobic.

Birnaviridae is a family of viruses (birnaviruses) which consists of four genera, members of which cause diseases in fish, birds, mollusks, and insects. The genome of birnaviruses encodes the highly immunogenic VP2 capsid protein. In order to demonstrate that the VP2 protein can be exploited as a diagnostic antigen for birnaviruses, we developed a lateral flow assay based on the surface-exposed VP2 protrusion domain of a representative birnavirus, infectious bursal disease virus (IBDV) of serotype 1 which causes the highly devastating infectious bursal disease in chickens. The biophysical characterization of the purified domain reveals that the domain predominantly consists of ß-sheets, exists in a trimeric form, and remains folded at high temperatures, making it suitable for diagnostic purposes. Owing to its highly immunogenic nature and excellent biophysical properties, we employed the VP2 protrusion domain in a gold nanoparticle-based lateral flow assay for rapid detection of anti-IBDV antibodies in serum samples of infected chickens. Our results indicate that the domain binds anti-IBDV antibodies with high specificity during laboratory testing and on-site testing. The lateral flow assay reported here yields comparable results in a qualitative manner as obtained through a commercial enzyme-linked immunosorbent assay (ELISA). As VP2 is a common capsid protein of birnaviruses, the lateral flow assay can be generalized for other birnaviruses, and members of Tetraviridae and Nodaviridae families which contain homologous VP2 capsid proteins.

The use of ornamental plant will increase with the improvement in living standards in green and blue-green infrastructure of urban settings. Nicotiana alata is an ornamental plant, frequently grown as a model plant for horticulture, medicine, and scientific research studies throughout the world. Despite its popularity, little is known about the response of N. alata against heavy metals (HMs). This work is based on the hydroponic study to identify the impacts of selected HMs (Cd, Cr, Cu, Ni and Pb) on N. alata, at 0, 50 and 100 µM concentration, with the co-application of EDTA, at 0 and 2.5 mM in hydroponics system. The HMs uptake was found to be dose dependent, with significant higher uptake at 100 µM of respective HM. Highest cumulative uptake (mg kg-1 of HMs in root, shoot, and leaf dried weight) noted were 767.50 ±â€¯50.83, 862.30 ±â€¯23.83, 271.29 ±â€¯18.68, 1117.49 ±â€¯46.10 and 2166.81 ±â€¯102.09, for Cd, Cr, Cu, Ni, and Pb at 100 µM, respectively. It was identified that EDTA co-application with HMs resulted in boosted HMs uptake, with cumulative uptake percentage increment of 41.62, 54.67, 53.98, 34.48 and 19.92% for 100 µM of Cd, Cr, Cu, Ni, and Pb, respectively. Higher uptake led to negative impact on plant physiology, photosynthetic pigments, and higher lipid peroxidation, H2O2 contents, and electrolyte leakage that increased the stress. Higher HMs uptake induced higher antioxidant enzymatic response. It is recommended to incorporate appropriate soil modification to grow N. alata in sustainable infrastructures.
