RESUMO

Silver oxide doped iron oxide (Ag2O-Fe2O3) nanocatalyst was prepared and coated on cotton cloth (CC) as well as wrapped in sodium alginate (Alg) hydrogel. Ag2O-Fe2O3 coated CC (Ag2O-Fe2O3/CC) and Ag2O-Fe2O3 wrapped Alg (Ag2O-Fe2O3/Alg) were utilized as catalysts in reduction reaction of 4-nitrophenol (4-NP), congo red (CR), methylene blue (MB) and potassium ferricyanide (K3[Fe(CN)6]). Ag2O-Fe2O3/CC and Ag2O-Fe2O3/Alg were found to be effective and selective catalyst for the reaction of K3[Fe(CN)6]. Further amount of catalyst, K3[Fe(CN)6] quantity, amount of NaBH4, stability of catalyst and recyclability were optimized for the reaction of K3[Fe(CN)6] reduction. Ag2O-Fe2O3/Alg and Ag2O-Fe2O3/CC were appeared to be the stable catalysts by maintaining high activity during recyclability tests showing highest reaction rate constants (kapp) of 0.3472 and 0.5629 min-1, correspondingly. However, Ag2O-Fe2O3/CC can be easily recovered as compared to Ag2O-Fe2O3/Alg by simply removing from the reaction which is the main advantage of Ag2O-Fe2O3/CC. Moreover, Ag2O-Fe2O3/Alg and Ag2O-Fe2O3/CC were also examined in real samples and found useful for K3[Fe(CN)6] reduction involving real samples. The Ag2O-Fe2O3/CC nanocatalyst is a cost and time saving material for economical reduction of K3[Fe(CN)6] and environmental safety.

RESUMO

Peanut (Arachis hypogaea L.) is an important food and feed crop worldwide and is affected by various biotic and abiotic stresses. The cellular ATP levels decrease significantly during stress as ATP molecules move to extracellular spaces, resulting in increased ROS production and cell apoptosis. Apyrases (APYs) are the nucleoside phosphatase (NPTs) superfamily members and play an important role in regulating cellular ATP levels under stress. We identified 17 APY homologs in A. hypogaea (AhAPYs), and their phylogenetic relationships, conserved motifs, putative miRNAs targeting different AhAPYs, cis-regulatory elements, etc., were studied in detail. The transcriptome expression data were used to observe the expression patterns in different tissues and under stress conditions. We found that the AhAPY2-1 gene showed abundant expression in the pericarp. As the pericarp is a key defense organ against environmental stress and promoters are the key elements regulating gene expression, we functionally characterized the AhAPY2-1 promoter for its possible use in future breeding programs. The functional characterization of AhAPY2-1P in transgenic Arabidopsis plants showed that it effectively regulated GUS gene expression in the pericarp. GUS expression was also detected in flowers of transgenic Arabidopsis plants. Overall, these results strongly suggest that APYs are an important future research subject for peanut and other crops, and AhPAY2-1P can be used to drive the resistance-related genes in a pericarp-specific manner to enhance the defensive abilities of the pericarp.

RESUMO

Over time, molecular biology and genomics techniques have been developed to speed up the early diagnosis and clinical management of cancer. These therapies are often most effective when administered to the subset of malignancies harboring the target identified by molecular testing. Important advances in applying molecular testing involve circulating-free DNA (cfDNA)- and cell-free RNA (cfRNA)-based liquid biopsies for the diagnosis, prognosis, prediction, and treatment of cancer. Both cfDNA and cfRNA are sensitive and specific biomarkers for cancer detection, which have been clinically proven through multiple randomized and prospective trials. These help in cancer management based on the noninvasive evaluation of size, quantity, and point mutations, as well as copy number alterations at the tumor site. Moreover, personalized detection of ctDNA helps in adjuvant therapeutics and predicts the chances of recurrence of cancer and resistance to cancer therapy. Despite the controversial diagnostic values of cfDNA and cfRNA, many clinical trials have been completed, and the Food and Drug Administration has approved many multigene assays to detect genetic alterations in the cfDNA of cancer patients. In this review, we underpin the recent advances in the physiological roles of cfDNA and cfRNA, as well as their roles in cancer detection by highlighting recent clinical trials and their roles as prognostic and predictive markers in cancer management.

RESUMO

Though the incidence of several cancers in Western societies is regulated wisely, some cancers such as breast, lung, and colorectal cancer are currently rising in many low- and middle-income countries due to increased risk factors triggered by societal and development problems. Surgery, chemotherapy, hormone, radiation, and targeted therapies are examples of traditional cancer treatment approaches. However, multiple short- and long-term adverse effects may also significantly affect patient prognosis depending on treatment-associated clinical factors. More and more research has been carried out to find new therapeutic agents in natural products, among which the bioactive compounds derived from plants have been increasingly studied. Naringin and naringenin are abundantly found in citrus fruits, such as oranges and grapefruits. A variety of cell signaling pathways mediates their anti-carcinogenic properties. Naringin and naringenin were also documented to overcome multidrug resistance, one of the major challenges to clinical practice due to multiple defense mechanisms in cancer. The effective parameters underlying the anticancer effects of naringenin and naringin include GSK3ß inactivation, suppression of the gene and protein activation of NF-kB and COX-2, JAK2/STAT3 downregulation, downregulation of intracellular adhesion molecules-1, upregulation of Notch1 and tyrocite-specific genes, and activation of p38/MAPK and caspase-3. Thus, this review outlines the potential of naringin and naringenin in managing different types of cancers.

RESUMO

In the current study, five different plants, Syzygium Cumini, Fagonia cretica, Acacia modesta, Withania coagulans, and Olea europaea aqueous extracts were prepared and applied against the anticancer and antibacterial activities. It was observed that O. Europaea extract shows the highest anticancer activity with cell viability of 21.5%. All the five plants extract was also used against the inhibition of Bacillus subtilis where O. Europaea extract shows a promising inhibitory activity of 3.2 cm followed by W. coagulans. Furthermore, W. coagulans was subjected to the process of column chromatography as a result a withanolide was isolated. The fast atom bombardment mass spectrometry (FAB-MS) and high resolution fast atom bombardment (HRFAB-MS) [M + 1] indicated molecular weight at m/z 453 and molecular formula C28H37O5. The UV-Vis. spectrum shows absorbance at 210 nm suggesting the presence of conjugated system, and Fourier-transform infrared spectroscopy (FTIR) was recorded to explore the functional groups. Similarly, 1D and 2D NMR spectroscopy techniques such as 1H, 13C NMR, correlation spectroscopy (COSY-45°), heteronuclear single quantum correlation (HSQC), heteronuclear multiple bond correlation (HMBC) and Nuclear Overhauser effect Spectroscopy (NOESY) techniques was carried out to determine the unknown natural product. The collective data of all these techniques established the structure of the unknown compound and recognized as a withanolide.

RESUMO

This study concentrates on biosynthesis of Silver Nanoparticles (AgNPs) from stem extract of Acacia nilotica (A. nilotica). The reaction was completed at a temperature ~40-45 °C and time duration of 5 h. AgNPs were thoroughly investigated via advanced characterization techniques such as UV-Vis spectrophotometry (UV-Vis), Fourier Transform Infrared spectroscopy (FTIR), X-ray Diffractometry (XRD), Field Emission Scanning Electron Microscopy (FESEM), High Resolution Transmission Electron Microscopy (HRTEM), X-ray Photoelectron Spectroscopy (XPS), Thermo Gravimetric Analysis (TGA), Diffuse Reflectance Spectroscopy (DRS), Brunner-Emmett-Teller (BET), Dynamic Light Scattering (DLS), and Zeta potential analysis. AgNPs with average size below 50 nm were revealed by all the measuring techniques. Maximum surface area ~5.69 m2/g was reported for the as synthesized NPs with total pore volume ~0.0191 mL/g and average pore size ~1.13 nm. Physical properties such as size and shape have changed the surface plasmon resonance peak in UV-visible spectrum. Antimicrobial activity was reported due to denaturation of microbial ribosome's sulphur and phosphorus bond by silver ions against bacterium Methicillin Resistant Staphylococcus aureus (MRSA) and fungus Candida Albican (CA). Furthermore, AgNPs degraded toxic pollutants such as 4-nitrophenol (4-NP), 2-nitrophenol (2-NP) and various hazardous dyes such as Congo Red (CR), Methylene Blue (MB) and Methyl Orange (MO) up to 95%. The present work provided low cost, green and an effective way for synthesis of AgNPs which were utilized as potential antimicrobial agents as well as effective catalyst for detoxification of various pollutants and dyes.

RESUMO

High oil and protein content make tetraploid peanut a leading oil and food legume. Here we report a high-quality peanut genome sequence, comprising 2.54 Gb with 20 pseudomolecules and 83,709 protein-coding gene models. We characterize gene functional groups implicated in seed size evolution, seed oil content, disease resistance and symbiotic nitrogen fixation. The peanut B subgenome has more genes and general expression dominance, temporally associated with long-terminal-repeat expansion in the A subgenome that also raises questions about the A-genome progenitor. The polyploid genome provided insights into the evolution of Arachis hypogaea and other legume chromosomes. Resequencing of 52 accessions suggests that independent domestications formed peanut ecotypes. Whereas 0.42-0.47 million years ago (Ma) polyploidy constrained genetic variation, the peanut genome sequence aids mapping and candidate-gene discovery for traits such as seed size and color, foliar disease resistance and others, also providing a cornerstone for functional genomics and peanut improvement.

RESUMO

A facile and highly porous heterostructure nanocomposite was designed for the support of zero-valent metal nanoparticles. This nanocomposite comprising of ZnO/carbon black (ZnO/CB) embedded in cellulose acetate polymer (CA), named as ZCA sheet, supported the metallic Cu, Ag and bimetallic CuAg nanoparticles (NPs). The ZnO/CB was incorporated to CA polymer host molecule in 3, 4 and 5 wt% and are designated as ZCA-3, ZCA-4 and ZCA-5. The catalytic tunability was evaluated for CuAg/ZCA-5 by adjusting the concentration of Cu and Ag ions in different molar ratio. Therefore, the CuAg/ZCA-5 was further selected for the removal of eight model pollutants comprising of o-nitrophenol (ONP), m-nitrophenol (MNP), p-nitrophenol (PNP), 2,6-dinitrophenol (DNP), methyl orange (MO), congo red (CR), methylene blue (MB) and rhodamine B (RB). The Kapp value for PNP was 1.9 × 10-1 min-1 and 9.0 × 10-2 min-1 for MO. Among the various nitrophenols, the rate of reaction with CuAg/ZCA-5 followed the ordered as PNP ˃ ONP ˃ MNP ˃ DNP, while methyl orange (MO) is degraded faster as compared to other dyes. The morphology, shape, elemental analysis, functional groups and peak crystallinity were scrutinized through FESEM, EDS, ATR-FTIR and XRD respectively.

RESUMO

The eco-friendly biosynthesis of silver nanoparticles (AgNps) from bark extract of Albizia chevalier are reported here for their anti-proliferative, antibacterial and pollutant degradation potentials. The synthesized AgNps were characterized by FTIR spectroscopy, transmission electron microscopy (TEM), field emission scanning electron microscopy (FESEM), energy dispersive X-rays spectrometry (EDS) and X-ray diffraction studies. The TEM and FESEM images show a monodispersed spherical shaped particles of approximately 30 nm. Crystalline peaks were obtained for the synthesized AgNps in XRD spectrum. The AgNps were investigated for in vitro anticancer and antibacterial activities and its potential to degrade 4-nitrophenol (4-NP) and congo red dye (CR). The MTT results shows a significant dose-dependent antiproliferation effect of the AgNps on the cell lines HepG2, MDA-MB-231 and MFC7. The effect was found more pronounced in MDA-MB-231 as compared to MFC-7 cell lines. The antibacterial results indicated 99 and 95% killing of Escherichia coli (E. coli) and Staphylococcus aureus (S. aureus) respectively, after 24 h of incubation with the AgNps. The AgNps were found to speed up the reductive degradation of 4-NP and CR dye, which give an alternative route for the removal of toxic organic pollutants from the wastewater. The synthesized AgNps were not only used as a bactericidal and anticancer agent, but also effectively used for the reductive degradation of carcinogenic compounds which are listed as the priority pollutants. Therefore, AgNps have the potential for the treatment of various cancers, bacterial infections and for industrial detoxification of wastewater.

RESUMO

The study concentrate on the biosynthesis of silver nanoparticles (AgNps) from the leaves extract of Guiera senegalensis with focus on its; antiproliferation effect on prostate (PC3), breast (MCF7) and liver (HepG2) cancer cell lines, antibacterial effect on Escherichia coli (E. coli) and Staphylococcus aureus (S. aureus) and the degradation on 4-nitrophenol (4-NP) and congo red dye (CR). The synthesized AgNps were characterized by FTIR, TEM, FESEM, XRD and EDX analysis. The EDS spectrum revealed that the synthesized nanoparticles (Nps) were composed of 55.45% Ag atoms of spherical shape with approximately 50nm size, identified from TEM and FESEM data. The antiproliferation effect of the AgNps varies with cell lines in a concentration dependent manner. The result showed that the AgNps were more effective on PC3 (IC50 23.48µg/mL) than MCF7 (29.25µg/mL) and HepG2 (33.25µg/mL) by the virtue of their IC50 values. The AgNps were highly effective against E. coli and S. aureus by killing 99% colonies. The AgNps also shows a good catalytic reduction of the toxic organic pollutants in which only 3mg of the AgNps degraded 95% of both CR dye and 4-NP in 22 and 36min respectively. Therefore, the green synthesis of AgNps may have potential applications in pharmacology and industries for the treatment of cancers, bacterial infections and in degrading toxic organic pollutants in water.

RESUMO

We present here the biosynthesis of AgNps from the aqueous extract of H. thebaica fruit, and monitored through UV-Vis spectrophotometer. The functional group were characterized through ATR-FTIR spectroscopy, the particle size, morphologies and elemental composition of the nanoparticles were investigated by using TEM, FESEM and EDS respectively. The anti-proliferation activity of the synthesized AgNps was carried out using MTT assay on human prostate (PC3), breast (MCF7) and liver (HepG2) cancer cell lines. The anti-proliferation assay showed that the AgNps were able to inhibit the proliferation of the cancer cell lines in a dose depending manner. The effect was found more pronounced on prostate (IC50 2.6 mg/mL) followed by breast (IC50 4.8 mg/mL) and then liver cancer cell lines (IC50 6.8 mg/mL). The prepared AgNps were found to inhibit 99% growth of both E. coli and S. aureus after 24 h of incubation. The nanoparticles were used for the degradation of 4-nitrophenol (4-NP) and Congo red dyes (CR), which efficiently degrade CR, but make complex formation with 4-NP. Therefore, the AgNps synthesized from the aqueous fruit extract of H. thebaica have potential application in pharmacology and waste water treatment.

RESUMO

Present study was aimed to screening the population of 25 wheat genotypes from Baluchistan region of Pakistan along with five commercial cultivars for leaf rust adult plant resistance (APR) through gene postulation using natural inoculation of Puccinia triticina Erikss local pathotype. Infection severity was recorded on scale in comparison with susceptible control "Morroco" cultivar. On the basis of phenotypic score, seven accessions and four varieties (Zardana-89, Sariab-92, Zarlashta-99 and Raskoh-05) with AUDPC values up to 20% were characterized as resistant genotypes. Coefficient of infection (CI) score ranged from 0-10 for some accessions and cultivars showing high level of adult plant resistance. Furthermore, bi-allelic STS marker csLV34 having close linkage with Lr34 (0.4cM). This marker amplified one gene specific allele of 150bp in 21 genotypes, including 19 accessions and two commercial varieties (Sariab-92 and Zarghoon-79) which confirmed presence of Lr34 gene conferring adult plant resistance against leaf rust. The rust pathogenicity scale varied for accessions from resistant to moderately susceptible. However, beside Lr34, phenotypic gene postulation, in combination with marker assisted selection for leaf rust resistance, has revealed presence of some other unknown resistance genes in local wheat germplasm which signified its use in wheat improvement programs both locally and abroad.

BACKGROUND: Potable, clean and safe water is the basic need for all human beings. Major portion of the earth is occupied by water, however, this is contaminated by rapid industrialization, improper sewage and natural calamities and man-made activates, which produce several water-borne and fetal diseases. In this review we presented some recent patent for environmental remediation. METHODS: Various technologies have been developed for the treatment of waste water consist of chemical, membrane, filtration, sedimentation, chlorination, disinfection, electrodialysis, electrolysis, reverse osmosis and adsorption. Among these entire phenomenon's, adsorption was the most efficient method for wastewater treatment, because it is a quick and cheap technology which signifies extensive practical applications. Adsorption phenomenon has been tactfully used for the removal of biological waste as well as soluble and insoluble material with a removal efficacy of 90-99%. RESULTS: Clean water supply is limited to human beings. The people in the developing countries have less or no access to the clean and potable water. The shortage of potable water resources and long term safe water deficiencies are some of the leading problems worldwide. In this review, we have explained in the detail adsorption phenomena of chitosan, pharmaceutical importance and other applications. It is worth to say that adsorption technologies using chitosan and its derivative is one of the quickest and cost effective methods for the wastewater treatment. The review comprises of ninety eight references. This review also covers various patents vis-a-vis the role of chitosan-nanocomposite in environmental application for wastewater treatment. CONCLUSION: Chitosan is a pseudo-neutral cationic polymer which is formed by the de-acetylation of chitin polymer. Various patent on chitosan and chitosan-nanocomposite were taken into account related to wastewater purification. We have found that chitosan and chitosan-nanocomposite are used for the removal of viruses, bacteria, cryptosporidium oocysts and giardia cysts, soluble and insoluble organic pollutants, poly aromatic hydrocarbons and heavy metals from wastewater. In this study, we also found that chitosan and chitosan-nanocomposite are selected for the removal of transition metals.
