The vegetable leafminer (Liriomyza sativae [Burgess]) is a highly polyphagous pest that threatens vegetables and horticultural plants. Although sexual communication is a key component of the animal behavioral repertoire, the mechanism underlying sexual communication in L. sativae remains to be elucidated. Here, we used laser vibrometry to characterize the vibrational signals emitted by L. sativae during pair formation. By emitting trains of vibrational pulses (male calling) the male initiated communication on the host plant. The female then became immobile and responded to the male calling by emitting replies (female replies), which in turn triggered male replies consisting of a rapid series of chirps and trills. If the female replied, a continuous exchange of male and female replies ensued, representing a duet. In playback trials, a playback signal caused responses from the opposite sex. Moreover, scanning electron microscopy revealed vibration-producing stridulatory organs in both male and female individuals. The files in males were more developed than those in females, and older male specimens had more signs of abrasion. The results provide new insight into the mating biology of L. sativae.

The Liriomyza trifolii is a highly invasive polyphagia pest. Understanding the physiological functions of odorant binding proteins (OBPs) in the chemical communication of L. trifolii can lead to effective pest management strategies. Seven full-length OBPs were identified by transcriptome screening of L. trifolii adults. Bioinformatics analyses classified the seven OBPs into two subfamilies (six classic OBPs, one minus-C OBP). The analysis of their expression in different development stages revealed that LtriOBP5 was highly expressed in the larval stage, LtriOBP4 in the pupa stage, and LtriOBP1, 2, 3, 6, 7 in the adult stage; the expression levels were higher in male adults than in females. The analysis of different tissues showed high expression of LtriOBP1, 3, 6, 7 in the antennae, which were selected for in vitro purification. To explore the ligand compounds of OBPs, fluorescence competitive binding experiments were performed. Immunofluorescence localization revealed that LtriOBP1, 3, 6, 7 showed strong binding abilities to plant volatiles and were located in the antennae, implying that LtriOBP1, 3, 6, 7 may play key roles in olfaction, such as host location. LtriOBP6 and LtriOBP7 had strong binding abilities to specific herbivore-induced plant volatiles, suggesting LtriOBP6 and LtriOBP7 may also play critical roles in chemoreception. This study provides preliminary exploration of the olfactory perception mechanism of L. trifolii, which can be used as a basis to design insect behavior regulators and develop highly effective insecticides using mixture of ligands and known pesticides.

ABSTRACT The sensilla of insects play important roles in odor, taste, the feeling environment, and some sensory functions, which are closely associated with insect host locating, feeding, habitat searching, courtship, mating, and oviposition. In this study, we used a scanning electron microscope to observe the external morphology of nymph and adult Frankliniella occidentalis sensilla on antennae, compound eyes, legs, mouthparts, wings and tail. The results show that three main types of sensilla are located on the antennae: sensilla trichodea, sensilla chaetica, and sensilla basiconica. Among them, sensilla trichodea are the most abundant, followed by sensilla chaetica. Sensilla basiconica on antennae are divided into seven sub-styles, including longer sensilla (L-sensilla basiconica, long sensilla basiconica), shorter sensilla (angle sensilla basiconica, bud sensilla basiconica), and thicker and bigger sensilla (stick sensilla basiconica, fork sensilla basiconica, and finger sensilla basiconica). Only two fork sensilla basiconica were found, located on the dorsal part of the first flagellum subsegment and the ventral part of the second flagellum subsegment, respectively. Seven sensillum types were found on the legs: sensilla trichodea, sensilla chaetica, sensilla basiconica, sensilla ear washing buob-shaped, mamillary sensilla, sensilla campaniform, and Böhm bristle. About 60% of them are sensilla chaetica. Only one sensillum type was found on compound eyes: sensilla chaetica. In addition, some sensillum types were also found on the mouthparts, wings and tail, respectively. In the study, we observed the type, morphology, and distribution of sensilla on antennae, legs, compound eyes, and other regions of nymph and adult F. occidentalis, forming a base for future electrophysiological and behavior experiments on F. occidentalis.

The Western flower thrips, Frankliniella occidentalis Pergande (Thysanoptera: Thripidae) is an invasive polyphagous pest with an expanding global range that damages a wide variety of crops. F. occidentalis was first reported in China from Yunnan province during 2000, and has rapidly expanded its range since then. It is currently distributed across at least 10 provinces in China and has become a particularly devastating pest, causing substantial damage and economic losses. At present, the still heavy reliance on frequent use of insecticides for control of F. occidentalis, has lead to a series of ecological problems stemming from insecticide resistance, interspecific displacement and non-target effects. Thus, integrated pest management (IPM) programmes, multiple complementary tactics, including preventive tactics, biological controls, and judicious use of insecticides will likely provide a viable IPM strategy for control of F. occidentalis in the near future. This review provides an overview for information gained during the 10+ years since the invasion of F. occidentalis into China, reviews lessons that have been learned enhancing our overall understanding of the biology and ecology of F. occidentalis and discusses IPM practices relative to this widespread invasive insect pest.

Using RT-PCR and RACE-PCR strategies, we cloned and identified a new chemosensory protein (FoccCSP) from the Western flower thrips, Frankliniella occidentalis, a species for which no chemosensory protein (CSP) has yet been identified. The FoccCSP gene contains a 387 bp open-reading frame encoding a putative protein of 128 amino acids with a molecular weight of 14.51 kDa and an isoelectric point of 5.41. The deduced amino acid sequence contains a putative signal peptide of 19 amino acid residues at the N-terminus, as well as the typical four-cysteine signature found in other insect CSPs. As FoccCSP is from a different order of insect than other known CSPs, the GenBank FoccCSP homolog showed only 31-50% sequence identity with them. A neighbor-joining tree was constructed and revealed that FoccCSP is in a group with CSPs from Homopteran insects (e.g., AgosCSP4, AgosCSP10, ApisCSP, and NlugCSP9), suggesting that these genes likely developed from a common ancestral gene. The FoccCSP gene expression profile of different tissues and development stages was measured by quantitative real-time PCR. The results of this analysis revealed this gene is predominantly expressed in the antennae and also highly expressed in the first instar nymph, suggesting a function for FoccCSP in olfactory reception and in particular life activities during the first instar nymph stage. We expressed recombinant FoccCSP protein in a prokaryotic expression system and purified FoccCSP protein by affinity chromatography using a Ni-NTA-Sepharose column. Using N-phenyl-1-naphthylamine (1-NPN) as a fluorescent probe in fluorescence-based competitive binding assay, we determined the binding affinities of 19 volatile substances for FoccCSP protein. This analysis revealed that anisic aldehyde, geraniol and methyl salicylate have high binding affinities for FoccCSP, with KD values of 10.50, 15.35 and 35.24 µM, respectively. Thus, our study indicates that FoccCSP may play an important role in regulating the development of the first instar nymph and mediate F. occidentalis host recognition.

BACKGROUND: Biopesticides containing Cry insecticidal proteins from the bacterium Bacillus thuringiensis (Bt) are effective against many lepidopteran pests, but there is a lack of Bt-based pesticides for efficient control of important coleopteran pests. Based on the reported increase in Bt toxin oligomerization by a polypeptide from the Cry3Aa receptor cadherin in Tenebrio molitor (Coleoptera: Tenebrionidae), it was hypothesized that this cadherin peptide, rTmCad1p, would enhance Cry3Aa toxicity towards coleopteran larvae. To test this hypothesis, the relative toxicity of Cry3Aa, with or without rTmCad1p, against damaging chrysomelid vegetable pests of China was evaluated. RESULTS: Cry3Aa toxicity was evaluated in the spotted asparagus beetle (Crioceris quatuordecimpunctata), cabbage leaf beetle (Colaphellus bowringi) and daikon leaf beetle (Phaedon brassicae). To assess the effect of rTmCad1p on Cry3Aa toxicity, neonate larvae were fed Cry3Aa toxin alone or in combination with increasing amounts of rTmCad1p. The data demonstrated that Cry3Aa toxicity was significantly increased in all three vegetable pests, resulting in as much as a 15.3-fold increase in larval mortality. CONCLUSION: The application of rTmCad1p to enhance Cry3Aa insecticidal activity has potential for use in increasing range and activity levels against coleopteran pests displaying low susceptibility to Bt-based biopesticides.
