RESUMO

Organic materials such as bark and biochar can be effective filter materials to treat stormwater. However, the efficiency of such filters in retaining microplastics (MPs) - an emerging stormwater pollutant - has not been sufficiently studied. This study investigated the removal and transport of a mixture of MPs commonly associated with stormwater. Different MP types (polyamide, polyethylene, polypropylene, and polystyrene) were mixed into the initial 2 cm material of horizontal bark and biochar filters of 25, 50, and 100 cm lengths. The MP types consisted of spherical and fragmented shapes in size ranges of 25-900 µm. The filters were subjected to a water flow of 5 mL/min for one week, and the total effluents were analyzed for MPs by µFTIR imaging. To gain a deeper insight, one 100 cm bark filter replica was split into 10 cm segments, and MPs in each segment were extracted and counted. The results showed that MPs were retained effectively, >97%, in all biochar and bark filters. However, MPs were detected in all effluents regardless of filter length. Effluent concentrations of 5-750 MP/L and 35-355 MP/L were measured in bark and biochar effluents, respectively, with >91% of the MP counts consisting of small-sized (25 µm) polyamide spherical particles. Combining all data, a decrease in average MP concentration was noticed with longer filters, likely attributed to channeling in a 25 and 50-cm filter. The analyses of MPs in the bark media revealed that most MPs were retained in the 0-10 cm segment but that some MPs were transported further, with 19% of polyamide retained in the 80-90 cm segment. Overall, this study shows promising results for bark and biochar filters to retain MPs, while highlighting the importance of systematic packing of filters to reduce MP emissions to the environment from polluted stormwater.

Microplastics accumulate in stormwater and can ultimately enter freshwater recipients, and pose a serious risk to aquatic life. This study investigated the effectiveness of lab-scale horizontal flow sand filters of differing lengths (25, 50 and 100 cm) in retaining four types of thermoplastic microplastics commonly occurring in stormwater runoff (polyamide, polyethylene, polypropylene, and polyethylene terephthalate). Despite the differences in particle shape, size and density, the study revealed that more than 98% of the spiked microplastics were retained in all filters, with a slightly increased removal with increased filter length. At a flow rate of 1 mL/min and after one week of operation, 62-84% of the added microplastics agglomerated in the first 2 cm of the filters. The agglomerated microplastics included 96% of high-density fibers. Larger-sized particles were retained in the sand media, while microplastics smaller than 50 µm were more often detected in the effluent. Microplastics were quantified and identified using imaging based micro Fourier Transform Infrared Spectroscopy. The efficient retention of microplastics in low-flow horizontal sand filters, demonstrated by the results, highlights their potential importance for stormwater management. This retention is facilitated by various factors, including microplastic agglomeration, particle sedimentation of heavy fibers and favorable particle-to-media size ratios.

Microplastics (MPs) are ubiquitous pollutants in the ocean, and there is a general concern about their persistence and potential effects on marine ecosystems. We still know little about the smaller size-fraction of marine MPs (MPs <300 µm), which are not collected with standard nets for MPs monitoring (e.g., Manta net). This study aims to determine the concentration, composition, and size distribution of MPs down to 10 µm in the Kattegat/Skagerrak area. Surface water samples were collected at fourteen stations using a plastic-free pump-filter device (UFO sampler) in October 2020. The samples were treated with an enzymatic-oxidative method and analyzed using FPA-µFTIR imaging. MPs concentrations ranged between 11 and 87 MP m-3, with 88% of the MPs being smaller than 300 µm. The most abundant shape of MPs were fragments (56%), and polyester, polypropylene, and polyethylene were the dominant synthetic polymer types. The concentration of MPs shows a significant positive correlation to the seawater density. Furthermore, there was a tendency towards higher MPs concentrations in the Northern and the Southern parts of the study area. The concentration of MPs collected with the UFO sampler was several orders of magnitude higher than those commonly found in samples collected with the Manta net due to the dominance of MP smaller size fractions. Despite the multiple potential sources of MPs in the study area, the level of MPs pollution in the surface waters was low compared (<100 MP m-3) to other regions. The concentrations of MPs found in the studied surface waters were six orders of magnitude lower than those causing negative effects on pelagic organisms based on laboratory exposure studies, thus is not expected to cause any impact on the pelagic food web.

The seafloor is the major sink for microplastic (MP) pollutants. However, there is a lack of robust data on the historical evolution of MP pollution in the sediment compartment, particularly the sequestration and burial rate of small MPs. By combining a palaeoceanographic approach and state-of-the-art analytical methods for MP identification down to 11 µm in size, we present the first high-resolution reconstruction of MP pollution from an undisturbed sediment core collected in the NW Mediterranean Sea. Furthermore, we investigate the fate of MPs once buried in the sediments by evaluating the changes in the size distribution of the MPs and the weathering status of the polyolefins, polyethylene, and polypropylene. Our results indicate that the MP mass sequestered in the sediment compartment mimics the global plastic production from 1965 to 2016. We observed an increase in the weathering status of the polyolefins as the size decreased. However, the variability in the size and weathering status of the MPs throughout the sedimentary record indicated that these pollutants, once incorporated into sediments, remain preserved with no further degradation under conditions lacking remobilization.

Analysis of microplastics (MP) in environmental samples is an emerging field, which is performed with various methods and instruments based either on spectroscopy or thermoanalytical methods. In general, both approaches result in two different types of data sets that are either mass or particle number related. Depending on detection limits of the respective method and instrumentation the derived polymer composition trends may vary. In this study, we compare the results of hyperspectral Fourier-transform infrared (FTIR) imaging analysis and pyrolysis gas chromatography-mass spectrometry (Py-GC/MS) analysis performed on a set of environmental samples that differ in complexity and degree of microplastic contamination. The measurements were conducted consecutively, and on exactly the same sample. First, the samples were investigated with FTIR using aluminum oxide filters; subsequently, these were crushed, transferred to glass fiber filters, in pyrolysis cups, and measured via Py-GC/MS. After a general data harmonization step, the trends in MP contamination were thoroughly investigated with regard to the respective sample set and the derived polymer compositions. While the overall trends in MP contamination were very similar, differences were observed in the polymer compositions. Furthermore, polymer masses were empirically calculated from FTIR data and compared with the Py-GC/MS results. Here, a most plausible shape-related overestimation of the calculated polymer masses was observed in samples with larger particles and increased particle numbers. Taking into account the different measurement principles of both methods, all results were examined and discussed, and future needs for harmonization of intermethodological results were identified and highlighted. Since plastics degrade very slowly, they remain in the environment on much longer timescales than most natural organic substrates and provide a novel habitat for colonization by bacterial communities. The spectrum of relationships between plastics and bacteria, however, is little understood. The first objective of this study was to examine plastics as substrates for communities of Bacteria in estuarine surface waters. We used next-generation sequencing of the 16S rRNA gene to characterize communities from plastics collected in the field, and over the course of two colonization experiments, from biofilms that developed on plastic (low-density polyethylene, high-density polyethylene, polypropylene, polycarbonate, polystyrene) and glass substrates placed in the environment. Both field sampling and colonization experiments were conducted in estuarine tributaries of the lower Chesapeake Bay. As a second objective, we concomitantly analyzed biofilms on plastic substrates to ascertain the presence and abundance of Vibrio spp. bacteria, then isolated three human pathogens, V. cholerae, V. parahaemolyticus, and V. vulnificus, and determined their antibiotic-resistant profiles. In both components of this study, we compared our results with analyses conducted on paired samples of estuarine water. This research adds to a nascent literature that suggests environmental factors govern the development of bacterial communities on plastics, more so than the characteristics of the plastic substrates themselves. In addition, this study is the first to culture three pathogenic vibrios from plastics in estuaries, reinforcing and expanding upon earlier reports of plastic pollution as a habitat for Vibrio species. The antibiotic resistance detected among the isolates, coupled with the longevity of plastics in the aqueous environment, suggests biofilms on plastics have potential to persist and serve as focal points of potential pathogens and horizontal gene transfer.

Microplastic pollution within the marine environment is of pressing concern globally. Accordingly, spatial monitoring of microplastic concentrations, composition and size distribution may help to identify sources and entry pathways, and hence allow initiating focused mitigation. Spatial distribution patterns of microplastics were investigated in two compartments of the southern North Sea by collecting sublittoral sediment and surface water samples from 24 stations. Large microplastics (500-5000 µm) were detected visually and identified using attenuated total reflection (ATR) Fourier transform infrared (FTIR) spectroscopy. The remaining sample was digested enzymatically, concentrated onto filters and analyzed for small microplastics (11-500 µm) using Focal Plane Array (FPA) FTIR imaging. Microplastics were detected in all samples with concentrations ranging between 2.8 and 1188.8 particles kg-1 for sediments and 0.1-245.4 particles m-3 for surface waters. On average 98% of microplastics were <100 µm in sediments and 86% in surface waters. The most prevalent polymer types in both compartments were polypropylene, acrylates/polyurethane/varnish, and polyamide. However, polymer composition differed significantly between sediment and surface water samples as well as between the Frisian Islands and the English Channel sites. These results show that microplastics are not evenly distributed, in neither location nor size, which is illuminating regarding the development of monitoring protocols.

Although mounting evidence suggests the ubiquity of microplastic in aquatic ecosystems worldwide, our knowledge of its distribution in remote environments such as Polar Regions and the deep sea is scarce. Here, we analyzed nine sediment samples taken at the HAUSGARTEN observatory in the Arctic at 2340-5570 m depth. Density separation by MicroPlastic Sediment Separator and treatment with Fenton's reagent enabled analysis via Attenuated Total Reflection FTIR and µFTIR spectroscopy. Our analyses indicate the wide spread of high numbers of microplastics (42-6595 microplastics kg-1). The northernmost stations harbored the highest quantities, indicating sea ice as a possible transport vehicle. A positive correlation between microplastic abundance and chlorophyll a content suggests vertical export via incorporation in sinking (ice-) algal aggregates. Overall, 18 different polymers were detected. Chlorinated polyethylene accounted for the largest proportion (38%), followed by polyamide (22%) and polypropylene (16%). Almost 80% of the microplastics were ≤25 µm. The microplastic quantities are among the highest recorded from benthic sediments. This corroborates the deep sea as a major sink for microplastics and the presence of accumulation areas in this remote part of the world, fed by plastics transported to the North via the Thermohaline Circulation.

In the present study, Xenopus laevis tadpoles were chronically exposed to four concentrations of the synthetic gestagen Levonorgestrel (LNG; 10(-11), 10(-10), 10(-9), and 10(-8)M) starting at Nieuwkoop and Faber (NF) stage 48 until completion of metamorphosis. At NF 58 and 66, brain-pituitary and gonad samples were taken for gene expression analyses of gonadotropins and gonadal steroidogenic enzymes. Exposure to 10(-9) and 10(-8)M LNG until NF 58 repressed messenger RNA (mRNA) expression of luteinizing hormone (LH) ß in both genders. This decrease was persistent after further treatment until NF 66 in the 10(-8)M LNG treatment. Expression of follicle-stimulating hormone (FSH) ß was affected sex-specifically. No effect was present in NF 58 females, whereas LNG at 10(-9) and 10(-8)M significantly increased FSHß mRNA levels in males. In NF 66 females, 10(-9)M LNG treatment increased FSHß gene expression, whereas a decrease was observed in NF 66 males exposed to 10(-8)M LNG. In gonads, expression of steroid-5-alpha-reductase was affected sex-specifically with increased mRNA levels in females but repressed levels in males. Gene expression of further gonadal steroidogenic factors was decreased by 10(-8)M LNG in both genders at NF 66. Assessment of gonad gross morphology and histology revealed poorly developed testes in the 10(-8)M LNG treatment. Our results reveal considerable effects of chronic LNG exposure on sexual development of amphibians. The persistent inhibition of LHß expression concomitant with decreased mRNA levels of gonadal steroidogenic enzymes is suggested to result in the disruption of reproduction in adult amphibians.

Synthetic gestagens, including levonorgestrel (LNG), are active compounds in contraceptives, and several studies report their occurrence in surface waters. However, information about endocrine-disrupting effects in nontarget organisms is scarce. The present study investigated effects of LNG exposure on thyroid hormone-dependent metamorphosis of Xenopus laevis. Premetamorphic X. laevis tadpoles at Nieuwkoop and Faber (NF) stage 48 were exposed in a flow-through culture system to four LNG concentrations (10(-11), 10(-10), 10(-9), and 10(-8)M) over the period of metamorphosis. At NF 58 and 66, tadpoles were examined sex specifically. Developmental time and organismal responses were recorded and correlated with molecular and histopathological endpoints. Exposure to 10(-8)M LNG caused an inhibition of metamorphosis resulting in developmental arrest at early climax stages as giant tadpoles or tailed frogs. In brain-pituitary tissue of NF 58 tadpoles, gene expression of thyroid-stimulating hormone (ß-subunit; TSHß), TH receptor ß (TRß), and deiodinase type 3 (D3) was not changed. Instead, prolactin (PRL) messenger RNA (mRNA) was significantly increased by 10(-9)M LNG in females and by 10(-8)M LNG in both sexes. In NF 66 tadpoles, mRNA levels of TSHß mRNA were significantly increased in the 10(-9) and 10(-8)M LNG treatment groups indicating a hypothyroid state. No changes of TRß, D3, and PRL gene expression were detected. Histopathological evaluation of thyroid gland sections revealed no typical sign of hypothyroidism but rather an inactivated appearance of the thyroid. In conclusion, our data demonstrate for the first time a completely new aspect of thyroid system disruption caused by synthetic gestagens in developing amphibians.

The uptake of iodide represents the first step in thyroid hormone synthesis by thyroid follicular cells and is mediated by the sodium-iodide symporter (NIS). In mammals, expression of NIS is stimulated by TSH and transcription of the NIS gene involves regulation by the thyroid-specific transcription factors Pax8 and Nkx2.1. In this study, we examined the mRNA expression of NIS, Pax8 and Nkx2.1 in the thyroid gland of Xenopus laevis tadpoles by semi-quantitative reverse transcriptase (RT)-PCR. During spontaneous metamorphosis, NIS mRNA expression was low in premetamorphic tadpoles, increased throughout prometamorphosis, and peaked at climax stage 60. Analysis of TSH beta-subunit (TSHbeta) mRNA in the pituitary of the same tadpoles revealed a close temporal relationship in the expression of the two genes during metamorphosis, suggesting a regulatory role of TSH in the developmental expression of NIS. Treatment of tadpoles with goitrogenic compounds (sodium perchlorate and ethylenethiourea) increased TSHbeta mRNA expression (approximately twofold) and caused thyroid gland hyperplasia, confirming that feedback along the pituitary-thyroid axis was operative. Analysis of gene expression in the thyroid gland revealed that goitrogen treatment was correlated with increased expression of NIS mRNA (approximately 20-fold). In the thyroid gland organ culture experiments, bovine TSH (bTSH; 1 mU/ml) strongly induced NIS mRNA expression. This effect was mimicked by co-culture of thyroid glands with pituitaries from stage 58 tadpoles and by agents that increase intracellular cAMP (forskolin, dibutyryl-cAMP). In addition, it could be shown that thyroid glands of X. laevis tadpoles express Pax8 and Nkx2.1 mRNA in a developmentally regulated manner and that ex vivo treatment of thyroid glands with bTSH, forskolin, and cAMP analogs increased the expression of Pax8 and Nkx2.1 mRNA. This is the first report on developmental profiles and hormonal regulation of thyroid gland gene expression in amphibian tadpoles and, together, results reveal a critical role of TSH in the regulation of NIS mRNA expression in the thyroid gland of X. laevis tadpoles.
