BACKGROUND AND OBJECTIVE: Healthy diets and diets rich in phytochemicals can have health-promoting benefits in prostate cancer. Therefore, this study aimed to explore the possible association between Healthy Diet Indicator (HDI) and Phytochemical Index (PI) with prostate cancer odds ratio. METHODS: This is a case-control study conducted in Shiraz, Iran, involving 62 newly diagnosed prostate cancer cases and 63 hospital-based controls. The study collected demographic and anthropometric data, as well as dietary intake information via a semi-quantitative food frequency questionnaire. Logistic regression models were employed to evaluate the association between HDI and PI with prostate cancer. RESULTS: The study included 120 participants and found that individuals with higher HDI and PI scores had a lower odds ratio of prostate cancer (HDI: odds ratio (OR): 0.322 - confidence interval (CI) 95%: 0.14-0.700 - PI: OR: 0.426 - CI 95%: 0.204-0.888). After adjusting for potential confounders, a lower odds ratio of prostate cancer was observed specifically among those with higher HDI scores (OR: 0.376 - CI 95%: 0.163-0.868). CONCLUSION: The findings of the present study suggest that adopting healthier dietary habits rich in dietary phytochemicals could be effective in preventing and halting the progression of prostate cancer.

One of the leading causes that complicate the treatment of some malignancies, including breast cancer, is tumor heterogeneity. In addition to inter-heterogeneity and intra-heterogeneity of tumors that reflect the differences between cancer cell characteristics, heterogeneity in the tumor microenvironment plays a critical role in tumor progression and could be considered an overlooked and a proper target for the effective selection of therapeutic approaches. Due to the difficulty of completely capturing tumor heterogeneity in conventional detection methods, Tumor-on-Chip (TOC) devices with culturing patient-derived spheroids could be an appropriate alternative. In this research, human-derived spheroids from breast cancer individuals were cultured for 6 days in microfluidic devices. To compare TOC data with conventional detection methods, immunohistochemistry (IHC) and ITRAQ data were employed, and various protein expressions were validated using the transcriptomic databases. The behavior of the spheroids in the collagen matrix and the cell viability were monitored over 6 days of culture. IHC and immunocytochemistry (ICC) results revealed that inter and intra-heterogeneity of tumor spheroids are associated with HER2/ER expression. HER2 expression levels revealed a more important biomarker associated with invasion in the 3D culturing of spheroids. The expression levels of CD163 (as a marker for Ma2 macrophages) and CD44 (a marker for cancer stem cells (CSCs)) were also evaluated. Interestingly, the levels of M2a macrophages and CSCs were higher in triple-negative specimens and samples that showed higher migration and invasion. Cell density and extracellular matrix (ECM) stiffness were also important factors affecting the migration and invasion of the spheroids through the matrix. Among these, rigid ECM revealed a more crucial role than cell density. To sum up, these research findings demonstrated that human-derived spheroids from breast cancer specimens in microfluidic devices provide a dynamic condition for predicting tumor heterogeneity in patients, which can help move the field forward for better and more accurate therapeutic strategies.

INTRODUCTION: According to our knowledge, the relationship between dietary patterns such as pro-healthy, pro-vegetarian, and non-healthy dietary patterns and prostate cancer risk has not been clearly investigated in Iranian men. Therefore, we aimed to investigate the relationship between adherence to a pro-healthy (PHDI), pro-vegetarian (PDP), and non-healthy dietary indices (NHDI) and the risk of prostate cancer. METHOD: In this matched case-control study, 125 participants (62 cases and 63 hospital-based controls) were enrolled from April to September 2015. Participants' dietary intakes were evaluated using a valid and reliable 160-item semi-quantitative food frequency questionnaire. Dietary indices calculated based on previous studies. The relationship between dietary indices (PHDI, NHDI and PDP) and prostate cancer risk was assessed using binary regression models. RESULTS: According to adjusted model, significant negative correlations were found between PHDI and PDP with prostate cancer (PHDI: OR = 0.31; 95% CI; 0.11-0.85; P = 0.023 - PDP: OR = 0.34; 95% CI; 0.15-0.75; P = 0.008). Also, a positive association was seen between NHDI and prostate cancer (OR = 3.01; 95% CI; 1.20-7.57; P = 0.019). CONCLUSION: We found that adherence to healthy dietary indices which includes high amounts of fruits, vegetables, and whole grains reduces the risk of prostate cancer. While adherence to a dietary pattern high in red and processed meat, refined grains, and sweetened beverages increases the risk of prostate cancer.

OBJECTIVE: Nail salons offer a developing and diverse occupation for many women, especially the new generation. Due to the increasing apprehension surrounding heavy metals in dust caused by filing nails containing dried nail polish, the present study was designed aimed to health risk assessment of heavy metals in breathing zone of nail salon technicians (NSTs). METHODS: This is a cross-sectional study that was conducted in NSTs. The concentration of Cadmium (Cd), Lead (Pb), Nickel (Ni), Chromium (Cr) and Manganese (Mn)in breathing zone of 20 NSTs was determined using ICP-OES. RESULTS: The metal concentrations were in the following order: Mn > Pb > Ni > Cr > Cd with corresponding arithmetic mean values of0.008, 0.0023, 0.0021, 0.001 and 0.0006 mg m-3, respectively, which are exceeded the recommended levels stated in the indoor air guidelines. The average lifetime carcinogenic risk (LCR) for Cr, Cd, Ni and Pb was calculated 0.0084, 0.00054, 0.00026 and 1.44 E - 05, respectively. The LCR values of all metals (except Pb) exceeded the acceptable level set by the USEPA. The mean of Hazard quotients (HQ) for Mn, Cd, Cr, Ni and Pb were calculated to be23.7, 4.74, 2.19, 0.51 and 0.0.24, respectively. The sensitivity analysis showed that, the exposure frequency (EF) for Cr and Ni had the strong effects on generation of both LCR and HQ. Furthermore, the concentrations of Mn, Cd and Pb had strong impacts on the HQ generation and the concentration of Cd and Pb had main effects on LCR generation. CONCLUSION: To effectively reduce pollutant concentration, it is recommended to install a ventilation system near nail salon work tables and conduct continuous monitoring and quality control of nail products.

BACKGROUND: The incidence of colorectal cancer (CRC) has increased in Iran, and determining the dietary patterns that can contribute to reducing or increasing the risk of CRC will help better control this disease. Therefore, in the current study, we assessed the association between the empirical lifestyle index for hyperinsulinemia (ELIH) and the empirical dietary index for hyperinsulinemia (EDIH) with the CRC odds. METHODS: The present case (n = 71)-control (n = 142) study was carried out in several CRC surgical units of hospitals in Tehran, Iran. A semi-quantitative food frequency questionnaire containing 168 items was used to assess participants' dietary intakes. The EDIH and ELIH scores were calculated by food groups and some variables such as body mass index and physical activity. Logistic regression models were applied to evaluate the association between the EDIH and ELIH scores with CRC odds. RESULTS: According to baseline features of the study participants, there were significant differences between the controls and cases in ELIH score, fiber intake, taking aspirin, and family history of CRC in first- and second-degree relatives. Also, we found that the odds of CRC increased significantly in the last tertile compared to the first tertile in EDIH and ELIH in the adjusted model (odds ratio (OR) = 3.12; 95% confidence interval (CI): 1.30-7.48 and OR = 4.72; 95% CI: 1.15-19.39, respectively). CONCLUSIONS: In conclusion, the result of this study indicated that CRC odds was significantly greater in subjects with higher EDIH and ELIH scores. Also, according to the results of this study, lifestyle and diet with insulinemic potential can influence the CRC risk.

Triclosan (TCS) has been increased in the water during the COVID-19 pandemic because it cannot remove by conventional water treatment. In addition, it can accumulate in the human body over time through long-term exposure. Therefore, the occurrence of TCS in the water treatment plant (WTP) and tap water, and its human health risk assessment through tap water ingestion, dermal absorption, and inhalation routes in Isfahan, Iran, were investigated. Moreover, spatial regression methods were used for the prediction of water quality parameters, TCS concentration, and total hazard quotient (HQ). The average TCS concentration in the influent and effluent of WTP and tap water was 1.6, 1.4, and 0.4 µg/L, respectively. Conventional WTP has low efficiency in the removal of TCS (12.6%) from water. The average values of total HQ for males were 7.79×10-5, 4.97×10-4, and 4.97×10-5 and for females were 3.31×10-5, 2.11×10-4, and 2.11×10-5 based on RfDEPA, RfDMDH, and RfDRodricks, respectively that were in the low-risk levels (HQ<1). Furthermore, TCS concentration in tap water and the ingestion rate of drinking water had the highest effect on the risk of TCS exposure from tap water. The non-carcinogenic health risk of TCS in water was low. The results of this study may be useful for promoting WTP processes to remove emerging pollutants.

This study aimed to assess the triclosan (TCS) health risk in an Iranian pregnant women sample by Monte Carlo simulation (MCS). The urinary TCS of 99 women after the 28th week of pregnancy was detected by gas chromatography/mass spectrometry detector (GC/MS), and the MCS model implemented a health risk assessment. The corresponding hazard quotient (HQ) and the sensitivity analysis were calculated. TCS was measured in 100% of the urine samples with a median concentration of 2.89 µg/L. The median of HQ was obtained at 1.93 × 10-4. The TCS exposure risk in the studied population was lower than the allowable limit. A comparison between HQ values in the two weight subgroups of pregnant women showed that the risk level is almost equal, and there was minimal health risk in pregnant women from exposure to TCS.

The purpose of this study was to evaluate the microbial characteristics of coastal waters which are impacted by anthropogenic pollution as well as estimate the health risks associated with exposure to enteric and non-enteric microorganisms during swimming. Fecal indicator bacteria were highly detected in samples. Moreover, pathogenic and opportunistic microorganisms were found, with the highest frequency for Pseudomonas aeruginosa followed by Adenovirus 40/41, Acanthamoeba spp., Salmonella enterica, and Cryptosporidium parvum. The median risk of gastrointestinal illness through ingestion of water was estimated to be above the benchmark value of 0.05 per event recommended by WHO. Cryptosporidium followed by Adenovirus, showed higher illness risks than Salmonella. The potential risks of Acanthamoeba and P. aeruginosa were estimated to be low for both dermal and ocular exposure routes. However, there are uncertainties about the infectious fraction of pathogens existing in coastal waters and the delivered dose of microorganisms from dermal/ocular exposure during recreational activities.

This study aimed to model the removal of formaldehyde as an indoor air pollutant by Nephrolepis obliterata (R.Br.) J.Sm. plant using response surface methodology (RSM) and artificial neural network (ANN) models, and optimization of the models by particle swarm optimization algorithm (PSO). The data obtained in pilot-scale experiments under a controlled environment were used in this study. The effects of parameters on the removal efficiency such as formaldehyde concentration, relative humidity, light intensity, and leaf surface area were empirically investigated and considered as model parameters. The results of the RSM model, with power transformation, were in meaningful compromise with the experiments. A multilayer perceptron (MLP) neural network was also designed, and the mean of squared error (MSE), mean absolute error (MAE), and R2 were used to evaluate the network. Several training algorithms were assessed and the best one, the Levenberg Marquardt (LM), was selected. The PSO algorithm proved that the highest removal efficiency of formaldehyde was obtained in the presence of light, maximum leaf surface area and relative humidity, and at the lowest inlet concentration. The empirical system breakthrough occurred at 15 mg/m3 of formaldehyde, and the maximum elimination capacity was about 0.96 mg per m2 of leaves. The findings indicated that the ANN model predicted the removal efficiency more accurately compared to the RSM model.

Surgery is the standard treatment for breast malignancies, although local and distant relapses might occur. Previous studies have shown that surgery-induced wound fluid (WF) contains tumor-initiating and progressing factors; however, these experiments have only been performed on breast cancer cell lines. Since a cancerous tumor includes various components like malignant cells, recruited non-malignant cells and extracellular matrix, those investigations that only focused on cancer cell lines themselves are not adequate to establish WF's effects. We conducted a 3D model study where we mimicked the tumor microenvironment to re-assess previous in-vitro findings. We generated human-derived breast tumor spheroids from 23 patient specimens, dissociated and cultured them in microfluidic devices. The spheroids from each sample were treated with the patients' WF or RPMI medium. The proportion of live and dead cells was assessed using live/dead assays and fluorescent imaging on day 6. In 22 samples, the percentage of live cells was significantly higher in the WF-treated group than in the RPMI-treated group. In one sample, we observed an opposite trend. The results were contrary in one of the samples, and we reported that case with more details. We compared the two groups using the 3D culture environment of human-derived tumor spheroids prepared from different microfluidic devices to mimic the tumor environment heterogeneity. Our findings showed that most patients with breast cancer benefit from surgical wound healing. However, removal of the surgical-induced serum may not be a method of inhibiting the tumor in all patients.

Intraoperative radiotherapy (IORT) could abrogate cancer recurrences, but the underlying mechanisms are unclear. To clarify the effects of IORT-induced wound fluid on tumor progression, we treated breast cancer cell lines and human-derived tumor spheroids in 2D and microfluidic cell culture systems, respectively. The viability, migration, and invasion of the cells under treatment of IORT-induced wound fluid (WF-RT) and the cells under surgery-induced wound fluid (WF) were compared. Our findings showed that cell viability was increased in spheroids under both WF treatments, whereas viability of the cell lines depended on the type of cells and incubation times. Both WFs significantly increased sub-G1 and arrested the cells in G0/G1 phases associated with increased P16 and P21 expression levels. The expression level of Caspase 3 in both cell culture systems and for both WF-treated groups was significantly increased. Furthermore, our results revealed that although the migration was increased in both systems of WF-treated cells compared to cell culture media-treated cells, E-cadherin expression was significantly increased only in the WF-RT group. In conclusion, WF-RT could not effectively inhibit tumor progression in an ex vivo tumor-on-chip model. Moreover, our data suggest that a microfluidic system could be a suitable 3D system to mimic in vivo tumor conditions than 2D cell culture.

Land application of MSW compost promotes the soil fertility and productivity, but there is concern about its chemical and microbial quality. Although, current microbial indicators of MSW compost are generally considered to be protective of human health, the use of these indicators doesn't adequately represent the presence of pathogens that might be more resistant to inactivation during composting process. This work aims evaluation of the microbial characteristics of MSW compost and estimation of the health risks associated with exposure to pathogens of potential concern in MSW compost. Although the recommended standards for fecal coliforms and Salmonella were met in a relatively high percentage of samples, there were detectable levels of microbial pathogens. The highest daily infection risk was estimated for Cryptosporidium (1.25 × 10-3 per person) followed by Salmonella and Campylobacter, while the lowest related to adenovirus. Infection risks were low for both farmers and children in one-time exposure and all pathogen risks were decreased with increasing waiting time to near insignificant levels. However, the median annual risk of cryptosporidiosis or gastroenteritis was above the recommended value of 10-3 per person per year. Because of potential presence of pathogens in MSW compost, some level of pathogen monitoring beyond conventional indicators may be required to estimate potential risks. The findings of this study could provide information to governments and local authorities in making decisions on measures to reduce risk from application of MSW compost. Further research is needed to obtain the reliable information about the inactivation of microorganisms in environment.

PURPOSE: Heavy metals, as significant toxic environmental contaminants, can cause serious adverse health outcomes on the human body even in trace concentrations. There is limited evidence on heavy metal concentrations existing in the body fluids of pregnant women. This study aims to evaluate the urinary levels of arsenic (As) and lead (Pb), as two main toxic heavy metals, among pregnant women and their lifestyle determinants. METHODS: The study was performed in 2019-2020 in Isfahan, Iran. A number of 140 urine samples of pregnant women who were in their first pregnancy trimester were examined. Inductively coupled plasma optical emission spectrometry (ICP-OES) was applied to analyze the urinary concentrations of As and Pb. Socio-demographic data including age, pre-pregnancy body mass index (BMI), education status, and family income, as well as the use of cleaning products (cosmetic and household cleaning products), and lifestyle habits (food intake, smoking, and physical activity) were collected using a validated questionnaire. RESULTS: The mean concentration of As and Pb were 8.14 ± 10.8 and 9.6 ± 7.1 µg/g creatinine, respectively. The mean urinary concentration of Pb indicated significant differences in the levels of cosmetic usage, second-hand smoking exposure, and the use of Copper, Aluminum, Teflon, Steel, and Enameled utensils for cooking (p-value < 0.05). Furthermore, the mean of urinary Pb concentrations at high levels of physical activity and scratched utensils using was significantly different from the other categories (p-value = 0.02). No significant differences were found between As and Pb concentration with other socio-demographic factors. CONCLUSION: The lifestyle determinants and cosmetic products use are important predictors of urinary heavy metals in pregnant women, rather than sociodemographic characteristics. Additional research is necessary to determine long-term adverse birth outcomes of exposure to these heavy metals.

Heavy metals have been well documented to pose detrimental health effects. The current study aimed to measure the concentration of chromium (Cr) and nickel (Ni) in urinary samples of Iranian pregnant females and determine their potential correlations with different lifestyle variables. The study was conducted in 2019-2020 in Isfahan, Iran, and the urine samples were collected from 140 pregnant women. The concentrations of Cr and Ni in the urinary samples were measured by inductively coupled plasma optical emission spectrometry (ICP-OES). Data on socio-demographic characteristics, use of cleaning products, and lifestyle profiles was collected by validated questionnaires. Cr and Ni were detected in 100% of urinary samples with the mean concentration of 4.1±3.4 and 7.5±4.8 µg/g creatinine, respectively. Significant associations were found between the mean concentration of Cr and Ni with using cooking utensils made of copper, aluminum, Teflon, steel, and enameled, as well as with cosmetic use, and second-hand smoking exposure during pregnancy. The results also showed that the mean urinary Ni and Cr concentrations were significantly different among individuals who consumed seafood and canned food (p-value <0.05). Furthermore, the mean of urinary Cr and Ni concentrations at high levels of physical activity and scratched utensils used was significantly different from the other categories (p-value <0.05). According to our findings, the lifestyle determinants and cosmetic products had superiority to socio-demographic characteristics in predicting urinary heavy metals in Iranian pregnant women.

One of the main sites of the magnetic fields influence on living cells is the cell cycle. The intensity of this influence however, varies depending on the cell type and the duration of the treatment. Suspension of cultured tobacco cells (Nicotiana tabacum cv. Barley 21) were synchronized via sucrose starvation at their stationary growth phase. The cells were then exposed to 0.2 m T SMF up to 24 h. The progression of different cell cycle phases was monitored through flow cytometry in a time course manner. Expression of cell cycle controlling genes and amounts of certain signaling molecules were measured as well. Exposure to SMF delayed G1.S transition which was accompanied by decrease of cyclin-dependent kinases A (CDK A) and D-type cyclin, but an increase in the adenylyl cyclase (AC), transcription factor E2F, retinoblastoma protein (Rbp), and CDK-inhibitor protein 21 (p21) transcript accumulation. Exposure to SMF also increased the contents of nitric oxide (NO), hydrogen peroxide (H2O2), and salicylic acid (SA), compared to the control group. The results suggest a signaling pathway triggered by SMF starting from accumulation of NO and H2O2 followed by downstream events including the increase of cyclic nucleotides and subsequent decrease of both CDKA and CycD.

INTRODUCTION: Ectopic Cushing`s syndrome (ECS) secondary to neuroendocrine tumours (NETs) of the lung and mediastinum are rarely encountered. In this study, we present our experience in Iran on 15 patients with ECS secondary to lung and mediastinal tumours over a period of 27 years. MATERIAL AND METHODS: Since 1985, 15 patients with ECS secondary to lung and mediastinal tumours have been diagnosed and prospectively followed by the endocrinology team of Taleghani Hospital, Tehran, Iran. The clinical signs and symptoms, laboratory findings, radiological features, immunohistochemical characteristics, management strategies and outcome data are here presented. RESULTS: There were six women and nine men, aged 26-70 years, all presenting with typical features of Cushing's syndrome. Based on histopathologic evaluations, four patients had small cell lung cancer (SCLC) and seven patients had pulmonary NETs, one patient had a thymic NET, and one case was diagnosed as a lung tumourlet. The mean ± SD serum cortisol, 24h urine free cortisol and plasma ACTH were 47.2 ± 20.5 µg/dL, 2,702 ± 5,439 µg/day, and 220 ± 147 pg/mL, respectively. Pulmonary lesions ranged in diameter from 1.1 to 4 cm (mean 1.9 ± 1.1 cm). One patient had a 10 cm mediastinal mass. The duration of follow up in these cases was between one month and seven years (mean 29.9 ± 27.5 months). The four patients with SCLC died within three months of diagnosis. CONCLUSION: Our data demonstrates the protean clinical and laboratory manifestations of ECS secondary to lung and mediastinal tumours, the problems encountered in diagnosis, and the need for a multidisciplinary approach. This study confirms other series from Western Europe and North America that, unlike the SCLC patients who show a poor outlook, ECS secondary to lung carcinoids has a more favourable prognosis.

BACKGROUND: Hepatitis C Virus (HCV), a public health problem, is an enveloped, single-stranded RNA virus and a member of the Hepacivirus genus of the Flaviviridae family. Liver cancer, cirrhosis, and end-stage liver are the outcomes of chronic infection with HCV. HCV isolates show significant heterogeneity in genetics around the world. Therefore, determining HCV genotypes is a vital step in determining prognosis and planning therapeutic strategies. OBJECTIVES: As distribution of HCV genotypes is different in various geographical regions and HCV genotyping of patients has not been investigated in Zanjan City, this study was designed for the first time, to determine HCV genotypes in the region and to promote the impact of the treatment. MATERIALS AND METHODS: Serum samples of 136 patients were collected and analyzed for anti-HCV antibodies using ELISA (The enzyme-linked immunosorbent assay) method. Then, positive samples were exposed to RT-PCR, which was performed under standard condition. Afterwards, they investigated for genotyping using allele-specific PCR (AS-PCR), and HCV genotype 2.0 line probe assay (LiPA). RESULTS: Samples indicated 216 bp bands on 2% agarose gel. Analyses of the results demonstrated that the most dominant subtype was 3a with frequency of 38.26% in Zanjan Province followed by subtypes of 1b, 1a, 2, and 4 with frequencies of 25.73%, 22.05%, 5.14%, and 4.41%, respectively. The frequency of unknown HCV genotypes was 4.41%. CONCLUSIONS: According to the results, it was found that HCV high prevalent genotype in Zanjan is subtype 3a. Analysis of the results provides identification of certain HCV genotypes, and these valuable findings could affect the type and duration of the treatment.

BACKGROUND: Estrogen plays a fundamental role in the pathogenesis of type 2 diabetes mellitus (T2DM). Very few studies have shown the association between estrogen receptor α (ERα), PvuII and XbaI gene polymorphisms with T2DM in both men and women. We evaluated the hypothesis that PvuII and XbaI polymorphisms of ERα gene may be associated with T2DM in adult. METHODS: From spring of 2010 to the fall of 2011, a case-control study was performed at clinical centers of Jahrom University of Medical Sciences. We included 174 patients with T2DM including men and women and 174 age, sex, and body mass index frequency-matched health controls. We analyzed the PvuII and XbaI polymorphisms of ERα by using the polymerase chain reaction-based restriction fragment length polymorphism method. RESULTS: No significant differences between demographic characteristics of control and patients groups were observed. Allele frequencies of both PvuII and XbaI polymorphisms were significantly different between patients and control subjects (P=0.014 vs. P=0.002, respectively). When the group was separated into women and men, logistic regression analysis of genotype distribution of PvuII (pp vs. Pp+PP) in both sexes revealed that there was no significant association of PvuII genotype with men (odds ratio [OR], 1.67; confidence interval [CI], 0.86 to 3.28; P=0.89) and women (OR, 0.96; CI, 0.53 to 1.74; P=0.12). CONCLUSION: PvuII and XbaI polymorphisms in ERα are related with T2DM in the inpatient population.
