RESUMO

The rapid and reliable detection of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) seroconversion in humans is crucial for suitable infection control. In this sense, many studies have focused on increasing the sensibility, lowering the detection limits and minimizing false negative/positive results. Thus, biosensors based on nanoarchitectures of conducting polymers are promising alternatives to more traditional materials since they can hold improved surface area, higher electrical conductivity and electrochemical activity. In this work, we reported the analytical comparison of two different conducting polymers morphologies for the development of an impedimetric biosensor to monitor SARS-CoV-2 seroconversion in humans. Biosensors based on polypyrrole (PPy), synthesized in both globular and nanotubular (NT) morphology, and gold nanoparticles are reported, using a self-assembly monolayer of 3-mercaptopropionic acid and covalently linked SARS-CoV-2 Nucleocapsid protein. First, the novel hybrid materials were characterized by electron microscopy and electrochemical measurements, and the biosensor step-by-step construction was characterized by electrochemical and spectroscopic techniques. As a proof of concept, the biosensor was used for the impedimetric detection of anti-SARS-CoV-2 Nucleocapsid protein monoclonal antibodies. The results showed a linear response for different antibody concentrations, good sensibility and possibility to quantify 7.442 and 0.4 ng/mL of monoclonal antibody for PPy in the globular and NT morphology, respectively. The PPy-NTs biosensor was able to discriminate serum obtained from COVID-19 positive versus negative clinical samples and is a promising tool for COVID-19 immunodiagnostic, which can contribute to further studies concerning rapid, efficient, and reliable detections.

RESUMEN Introducción: Cuidar completamente de niños con microcefalia es un gran desafío para sus cuidadores. La situación de vulnerabilidad social junto con la complejidad de la atención puede llevar a sobrecarga con pérdida de la calidad de vida. Objetivo: Analizar la relación entre aspectos socioeconómicos, dominio social de la calidad de vida y la sobrecarga de madres cuidadoras de niños con microcefalia. Metodología: Estudio transversal con 105 participantes, mediante aplicación de cuestionarios sociodemográficos, WHOQOL-BREF, sobrecarga del cuidador. Se utilizaron pruebas ANOVA, t de Student, coeficiente de correlación de Pearson (r). Resultados: Población de madres mestizas (65.7%), desempleadas (88.6%), dependientes de la prestación social (76.2%), bajo nivel educativo (50.5%) e insuficiencia financiera (90.5%). El promedio del dominio social se consideró bajo (51.11). Hubo asociación entre beneficio social y puntaje de calidad de vida total (p=0.046). Las madres sufren sobrecarga severa (39%) e intensa (30.4%). Se encontró asociación entre niveles de sobrecarga y variables etnia (p= 0.027), ocupación (p= 0.043) y medios de transporte (p <0.0001). La sobrecarga tiene impacto negativo en la calidad de vida (r= -.463, p <0.0001), percepción de la calidad de vida (r= -.476, p= <0.0001), calidad de vida total (r= -.576, p= <0.0010). Conclusiones: Las madres cuidadoras manifiestan sobrecarga con pérdida de calidad de vida. Los aspectos sociales influyen directamente y pueden potenciar la sobrecarga del cuidado. El personal de enfermería tiene un papel fundamental para crear e implementar líneas de atención con énfasis en promover la salud y alivio de la sobrecarga en el cuidador.


ABSTRACT Introduction: Taking care of children with microcephaly is a great challenge for their care providers because of the associated social vulnerability, work overload, and loss of quality of life. Objective: To analyze the associations related to the social aspects, the quality of life, and the work overload among care providing mothers of children with microcephaly. Methodology: This is a transversal study with 105 participants. Social and demographic questionnaires and WHOQOL-BREF were used. ANOVA tests, Student's t-test, and Pearson's correlation coefficient were calculated. Results: 65.7% were of mixed ethnicity; 88.6% were unemployed; 76.2% were dependent on Social Welfare; 50.5% had a low level of education; and 90.5% were financially insufficient. The social domain average was considered low (51.11). An association was found between social benefits and quality of life (p=0.046). 39% of these mothers suffer from severe work overload while 30.4% suffer from intense work overload. An association was found between the levels of work overload and ethnicity (p= 0.027), occupation (p= 0.043), and transportation means (p <0.0001). Work overload had a negative impact on the quality of life (r= -.463, p <0.0001), the perception of quality of life (r= -.476, p= <0.0001), and total quality of life (r= -.576, p= <0.0010). Conclusions: These care providing mothers suffer work overload with an associated loss of quality of life. Social aspects have a direct influence and can increase work overload. Nursing professionals have a fundamental role in the creation and implementation of attention initiatives aimed at promoting health and relief to this kind of care providing mothers.

Foi encontrada associação entre os níveis de sobrecarga e as variáveis de etnia (p= 0.027), ocupação (p= 0.043) e meio de transporte (p <0.0001). A sobrecarga tem um impacto negativo na qualidade de vida (r= -.463, p <0.0001), percepção de qualidade de vida (r= -.476, p= <0.0001), qualidade de vida total (r= -.576, p= <0.0010). Conclusões: As mães cuidadoras sofrem sobrecarga com perda de qualidade de vida. Os aspectos sociais influenciam diretamente e podem potenciar a sobrecarga do cuidado. Kahweol is a compound derived from coffee with reported antinociceptive effects. Based on the few reports that exist in the literature regarding the mechanisms involved in kahweol-induced peripheral antinociceptive action, this study proposed to investigate the contribution of the endocannabinoid system to the peripheral antinociception induced in rats by kahweol. Hyperalgesia was induced by intraplantar injection of prostaglandin E2(PGE2) and was measured with the paw pressure test. Kahweol and the drugs to test the cannabinoid system were administered locally into the right hind paw. The endocannabinoids were purified by open-bed chromatography on silica and measured by LC-MS. Kahweol (80 µg/paw) induced peripheral antinociception against PGE2-induced hyperalgesia. This effect was reversed by the intraplantar injection of the CB1 cannabinoid receptor antagonist AM251 (20, 40, and 80 μg/paw), but not by the CB2 cannabinoid receptor antagonist AM630 (100 μg/paw). Treatment with the endocannabinoid reuptake inhibitor VDM11 (2.5 μg/paw) intensified the peripheral antinociceptive effect induced by low-dose kahweol (40 μg/paw). The monoacylglycerol lipase (MAGL) inhibitor, JZL184 (4 μg/paw), and the dual MAGL/fatty acid amide hydrolase (FAAH) inhibitor, MAFP (0.5 μg/paw), potentiated the peripheral antinociceptive effect of low-dose kahweol. Furthermore, kahweol increased the levels of the endocannabinoid anandamide, but not of the other endocannabinoid 2-arachidonoylglycerol nor of anandamide-related N-acylethanolamines, in the plantar surface of the rat paw. Our results suggested that kahweol induced peripheral antinociception via anandamide release and activation of CB1 cannabinoid receptors and this compound could be used to develop new drugs for pain relief.

Melanoma is the most lethal form of skin cancer, with a worldwide increase in incidence. Despite the increased overall survival of metastatic melanoma patients given recent advances in targeted and immunotherapy, it still has a poor prognosis and available treatment options carry diverse severe side effects. Polysaccharides from seaweed have been shown to exert antitumor activities. Here we show in vitro and in vivo antitumor activities of a sulfated homogalactan (named 3G4S) from Codium isthmocladum seaweed in the B16-F10 murine melanoma cell line. 3G4S did not induce cytotoxicity or proliferation changes; however, it was able to reduce solid tumor growth and metastasis, while not inducing side effects in mice. B16-F10 cells traits related to the metastatic cascade were also impaired by 3G4S, reducing cell invasion, colony-forming capacity and membrane glycoconjugates. Therefore, 3G4S shows promising antitumor activities without the commonly associated drawbacks of cancer treatments and can be further explored.

Melanoma is a form of skin cancer with high mortality owing to its fast progression and metastatic capacity. The treatments available nowadays are only palliative in advanced stages of the disease. Thus, alternative therapies for cancer treatment are in demand, and molecules from natural sources, such as polysaccharides, could represent new possible therapeutic approaches. Polysaccharides of freshwater and marine algae with biological activities, such as antitumor properties, are greatly reported in the scientific literature. In the present study, a sulfated heterorhamnan obtained from the green seaweed Gayralia brasiliensis (Gb1 fraction) was chemically characterized and its biological activities in the B16-F10 murine melanoma cell line were evaluated. The Gb1 polysaccharidic fraction tested concentrations presented low or absence of cytotoxicity to B16-F10 cells and neither cell proliferation nor cell cycle were altered. Interestingly, Gb1 treatment decreased B16-F10 cells migration and invasion capabilities and CD44 labeling, showing to be a promising compound for further in vitro and in vivo antitumor studies.

The Maytenus genus is a member of the Celastraceae family. Numerous medicinal uses were assigned to species of this genus, with the use of roots, bark, and leaves for the treatment of gastric ulcers, as anti-inflammatory, analgesic, antiallergic, antitumor, among others. Several studies have demonstrated that natural products derived from plants have an important role in the prevention and treatment of obesity. Accordingly, we evaluated the effect of Maytenus imbricata extracts in the treatment of obesity induced by diet rich in refined carbohydrate (HC). BALB/c mice were fed chow or HC diet for 8 weeks. At the beginning of the 9th week, the HC group was subdivided into three groups: (i) group of animals that continued to consume only HC diet; (ii) the group of animals fed HC diet supplemented with ethyl acetate extract of M. imbricata roots (HC + EAE); (iii) the group of animals fed HC diet supplemented with extract in hexane/ethyl ether (HC + HEE). The period of extracts supplementation was 4 weeks. It was observed that EAE and EHE when added to the HC diet modulated the metabolic and inflammatory changes, such as: reduced the adipocytes area, improved glucose intolerance, reduced the levels of triglycerides and resistin in serum, and the number of total leukocytes in blood. In the epididymal adipose tissue, the extracts reduced proinflammatory mediators' concentration. According to the results, it was concluded that the species Maytenus imbricata has the potential to be used for the treatment of obesity.

RESUMO

The scientific publication landscape is changing quickly, with an enormous increase in options and models. Articles can be published in a complex variety of journals that differ in their presentation format (online-only or in-print), editorial organizations that maintain them (commercial and/or society-based), editorial handling (academic or professional editors), editorial board composition (academic or professional), payment options to cover editorial costs (open access or pay-to-read), indexation, visibility, branding, and other aspects. Additionally, online submissions of non-revised versions of manuscripts prior to seeking publication in a peer-reviewed journal (a practice known as pre-printing) are a growing trend in biological sciences. In this changing landscape, researchers in biochemistry and molecular biology must re-think their priorities in terms of scientific output dissemination. The evaluation processes and institutional funding for scientific publications should also be revised accordingly. This article presents the results of discussions within the Department of Biochemistry, University of São Paulo, on this subject.

BACKGROUND: Maringa oleifera leaves are rich in antioxidant substances; however, when lyophilized leaves were used in flour form in meat products, they presented no antioxidant effect and even accelerated the oxidation process of the product. Thus, the objective of this study was to evaluate the effect of chlorophyll extraction on the physicochemical composition and antioxidant activity of Moringa leaves. METHODS: Moringa leaves were dried and ground in order to obtain uniform flour. A treatment using chlorophyll extraction (decolorized) was tested versus a control treatment(non-decolorized) for proximate composition, instrumental color, and antioxidant activity using ANOVA followed by Tukey'stest. RESULTS: Higher crude fiber, ash, and protein contents were observed for decolorized flour (19.41 and 38.13%, 11.87 and14.02%, and 28.81 and 31.33%, respectively) when compared to those for the control. Chlorophyll extraction significantly affected (p< 0.05) the instrumental color of the leaves flour. The half maximal effective concentration (EC50) of both decolorized and control flour was 3.74 and 4.30 mg/L, respectively. The equivalent of antioxidant per gramof non-decolorized leaves was higher than that observed for the decolorized leaves (0.36 and 0.32 g/g DPPH, respectively). The antioxidant activity (AA%) of the extract from non-decolorized leaves was higher in the concentrations of 5 and 2.5 mg/0.1 mL, while the decolorized leaves was higher in the extract concentration 5 and 2 mg/0.1 ml. CONCLUSION: The decolorization process affected the chemical composition and color of Moringa oleifera leaves flours however did not improve its antioxidant activity

A heteropolysaccharide was isolated by cold aqueous extraction from edible mushroom Pleurotus eryngii ("King Oyster") basidiocarps and its biological properties were evaluated. Structural assignments were carried out using mono- and bidimensional NMR spectroscopy, monosaccharide composition, and methylation analyses. A mannogalactan having a main chain of (1→6)-linked α-d-galactopyranosyl and 3-O-methyl-α-d-galactopyranosyl residues, both partially substituted at OH-2 by ß-d-Manp (MG-Pe) single-unit was found. Biological effects of mannogalactan from P. eryngii (MG-Pe) were tested against murine melanoma cells. MG-Pe was non-cytotoxic, but reduced in vitro melanoma cells invasion. Also, 50mg/kg MG-Pe administration to melanoma-bearing C57BL/6 mice up to 10days decreased in 60% the tumor volume compared to control. Additionally, no changes were observed when biochemical profile, complete blood cells count (CBC), organs, and body weight were analyzed. Mg-Pe was shown to be a promising anti-melanoma molecule capable of switching melanoma cells to a non-invasive phenotype with no toxicity to melanoma-bearing mice.

OBJECTIVES: Characterize the cell profile and immunostaining of proinflammatory markers in an experimental model of bisphosphonate-related osteonecrosis of the jaw (BRONJ). MATERIALS AND METHODS: Male Wistar rats (n = 6-7) were treated chronically with saline solution or zoledronic acid (ZA) at 0.04, 0.20, and 1.00 mg kg(-1) (1.4 × 10(-7) , 6.9 × 10(-6) , and 3.4 × 10(-5)  mol kg(-1) ), and subsequently, the first left inferior molar was extracted. Were performed counting of viable and empty osteocyte lacunae, viable and apoptotic osteoclasts, polymorphonuclear neutrophil, mast cells (toluidine blue), and the positive presence cells for CD68, tumor necrosis factor-alpha (TNF-α), IL (interleukin)-1ß, inducible nitric oxide synthase (iNOS), nuclear factor-kappa B (NF-kB) and IL-18 binding protein (IL-18 bp). RESULTS: BRONJ was showed in ZA treated with 0.20 and 1.00 mg kg(-1) . There is a dose dependent increase in percentage of empty osteocyte lacunae (P < 0.001) and apoptotic osteoclasts (P < 0.001), counting of total osteoclasts (P = 0.003), polymorphonuclear neutrophil cells (P = 0.009), cytoplasmic-positive cells of CD68 (P < 0.001), TNF-α (P = 0.001), IL-1ß (P = 0.001), iNOS (P < 0.001), NF-kB (P = 0.006), and nuclear-positive cells of NF-kB (P = 0.011). Consequently, there is no difference in mast cells (P = 0.957), and IL-18 bp immunostaining decreases dose dependently (P = 0.005). CONCLUSIONS: BRONJ is characterized by increases in immunostaining for proinflammatory markers and NF-kB and inversely associated with cells exhibiting IL-18 bp.

BACKGROUND: Denosumab, an anti-resorptive agent, IgG2 monoclonal antibody for human Receptor activator of nuclear factor-kappa B ligand (RANKL), has been related to the occurrence of osteonecrosis of the jaws. Thus, the aim of this study was to review the literature from clinical case reports, regarding the type of patient and the therapeutic approach used for osteonecrosis of the jaws induced by chronic use of Denosumab. MATERIAL AND METHODS: For this, a literature review was performed on PubMed, Medline and Cochrane databases, using the keywords "Denosumab" "anti-RANK ligand" and "Osteonecrosis of jaw". To be included, articles should be a report or a serie of clinical cases, describing patients aged 18 years or over who used denosumab therapy and have received any therapy for ONJ. RESULTS: Thirteen complete articles were selected for this review, totaling 17 clinical cases. The majority of ONJ cases, patients receiving Denosumab as treatment for osteoporosis and prostate cancer therapy. In most cases, patients affected by ONJ were women aged 60 or over and posterior mandible area was the main site of involvement. Diabetes pre-treatment with bisphosphonates and exodontia were the most often risk factors related to the occurrence of this condition. Systemic and local antibiotic therapy with or without surgical debridement was the most used treatment for ONJ resolution. CONCLUSIONS: It is concluded that the highest number of ONJ cases caused by the use of anti-RANKL agents occurred in female patients, aged 60 years or older, under treatment for osteoporosis and cancer metastasis, and the most affected region was the mandible posterior.

In this study, genotyping techniques including staphylococcal chromosomal cassette mec (SCCmec) typing, pulsed-field gel electrophoresis (PFGE), multilocus sequence typing (MLST) and restriction-modification tests were used to compare the molecular characteristics of methicillin-resistant Staphylococcus aureus (MRSA) isolates recovered at two times within a 10-year interval (1998 and 2008) from a tertiary Brazilian hospital. In addition, the antimicrobial susceptibility profiles were analyzed. All 48 MRSA isolates from 1998 and 85.7% from 2008 (48/56 isolates) displayed multidrug-resistance phenotypes and SCCmec III. All but one of the 13 representative SCCmec III isolates belonged to CC8 and had PFGE patterns similar to that of the BMB9393 strain (Brazilian epidemic clone of MRSA; BEC). In 2008, we found an increased susceptibility to rifampicin and chloramphenicol among the SCCmec III isolates. In addition, we detected the entrance of diverse international MRSA lineages susceptible to trimethoprim-sulfamethoxazole (SXT), almost all belonging to CC5. These non-SCCmec III isolates were related to the USA 300 (ST8-SCCmec IV; PFGE-type B), USA 800 (ST5-SCCmec IV; subtype D1), USA 100 (ST5-SCCmec II; subtype D2), and EMRSA-3/Cordobes (ST5-SCCmec I, type C) clones. To the best of our knowledge, this is the first report of the emergence of isolates genetically related to the EMRSA-3/Cordobes clone in southeast Brazil. In this regard, these isolates were the most common non-SCCmec III MRSA in our institution, accounting for 8.9% of all isolates recovered in 2008. Thus, despite the supremacy of BEC isolates in our country, significant changes may occur in local MRSA epidemiology, with possible consequences for the rationality of MRSA empiric therapy.

O uso indiscriminado de medicamentos acarreta riscos diretos e indiretos à população, tornando-se um importante problema de saúde pública. O conhecimento do padrão de uso de medicamentos contribui para ações educativas com a finalidade de reduzir riscos e danos à saúde da população. O objetivo deste estudo foi investigar o perfil de consumo de medicamentos em estudantes adolescentes e correlacioná-lo com o perfil sócio-educacional entre os familiares. Estudo de natureza transversal com amostra representativa de 102 alunos da população de escolares de ensino médio no Bairro Maruípe em Vitória-ES. O questionário aplicado contemplou as variáveis: idade, gênero, série, consumo de medicamentos nos sete dias anteriores à entrevista, quem orientou o consumo e a automedicação, aos quais foram avaliados por meio de perguntas dicotômicas, em caso afirmativo, especificando o(s) fármaco(s) utilizado(s) e sua finalidade. O uso de medicamentos ocorreu em 42,3% da amostra, sendo que em 53,3% dos casos houve automedicação. Os grupos farmacológicos mais consumidos foram analgésicos/antiinflamatórios, sendo o paracetamol a substância com maior freqüência de consumo. A incidência de automedicação no grupo representado pelo gênero feminino (66,7%) foi superior ao grupo do gênero masculino (33,3%). A relação entre a idade e automedicação mostrou uma maior prevalência entre os jovens de 15 a 17 anos de idade, representando 70,8% dos adolescentes que fazem automedicação. O consumo de medicamentos sem orientação médica pelos familiares mostrou ser um fator de influência na automedicação dos adolescentes.


The indiscriminate use of medicines poses direct and indirect risks to the population, making it a major public health problem. Knowledge of the pattern of drug use in a given population would contribute to educational action aimed at reducing such risks and the resulting damage to the health of that population. The objective of this study was to investigate the profile of consumption of medicines by adolescents in a Brazilian city and to correlate it with the socio-educational profile of the students and the consumption of drugs among their relatives. A horizontal study was carried out on a representative sample population of 102 students of the Maruípe district in the city of Vitória (ES, Brazil). The questionnaire included the variables: age, sex, consumption of drugs in the seven days preceding the interview, who (if anyone) guided the consumption and self-medication; data were collected by means of dichotomous questions - if drug(s) were consumed, the interviewee was asked to name them and their purpose. Drugs were consumed in the week before the interview in 42.3% of the sample, 53.3% of these cases by self-medication. The drug groups most consumed by the adolescents were analgesics and anti-inflammatory drugs, paracetamol being the most frequent. The incidence of self-medication among the females (66.7%) was higher than among the males (33.3%). Analysis of the relationship between age and self-medication showed a high prevalence among young people aged 15 to 17 years, representing 70.8% of the adolescents who practiced self-medication. The consumption of drugs without medical advice by families was a factor influencing adolescent self-medication.

A crescente demanda por produtos cosméticos estáveis, seguros e eficazes tem exigido da comunidade científica estudos cada vez mais complexos e utilização de técnicas mais eficientes para determinação da estabilidade dos produtos. O uso de ativos da biodiversidade brasileira levou ao desenvolvimento de inúmeros produtos, nas mais diferentes formas cosméticas, o que dificulta, ainda mais, a padronização de protocolos experimentais para atestara estabilidade das preparações cosméticas. Embora não exista no Brasil um protocolo que padronize os ensaiosa serem realizados para se determinar a estabilidade dos produtos, vários estudos foram realizados nos meios acadêmicos para determinação da estabilidadede matérias-primas específicas. As características reológicas são propriedades importantes a serem consideradas na fabricação, estocagem e aplicação de produtos de uso tópico. Além de ensaios comumente empregados, a determinação do comportamento reológico da formulação auxilia na avaliação da natureza físico-química do veículo, de tal forma que torna possível detectar sinais precoces de instabilidade física, possibilitando o controle de qualidade dos constituintes, das formulações teste e dos produtos finais. A análise térmica também é usada para auxiliar o estudo da estabilidade de produtos cosméticos e, de modo geral, a calorimetria exploratória diferencial para orientar o desenvolvimento de novos produtos. Outras ferramentas, como a fluorimetria e a granulometria a laser podem auxiliar o estudo e o desenvolvimento de sistemas, emulsionados ou não. O objetivo deste estudo é o desenvolvimento de protocolo para o estudo da estabilidade físico-química de fitocosméticos - sistemas contendo ativos da biodiversidade brasileira.

This work investigated the treatability of real textile effluents using several systems involving advanced oxidation processes (AOPs) such as UV/H2O2, UV/TiO2, UV/TiO2/H2O2, and UV/Fe2+/H2O2. The efficiency of each technique was evaluated according to the reduction levels observed in the UV absorbance of the effluents, COD, and organic nitrogen reduction, as well as mineralization as indicated by the formation of ammonium, nitrate, and sulfate ions. The results indicate the association of TiO2 and H2O2 as the most efficient treatment for removing organic pollutants from textile effluents. In spite of their efficiency, Fenton reactions based treatment proved to be slower and exhibited more complicated kinetics than the ones using TiO2, which are pseudo-first-order reactions. Decolorization was fast and effective in all the experiments despite the fact that only H2O2 was used.

Canova is a homeopathic complex medicine, used as an immune modulator. We studied its effects in normal and sarcoma 180-bearing mice. Three control groups were also evaluated. The mice were examined at daily intervals and the tumours observed histologically. Peripheral blood was analysed by flow cytometry. A delay in the development, and a reduction in size of the tumours, and increased infiltration by lymphoid cells, granulation tissue, and fibrosis surrounding the tumour were observed with active treatment compared to control. All animals from the treated group survived, 30% of control groups died. In 30% of treated animals, a total regression of the tumour was confirmed using light microscopy, no regression was found in the control groups. Treatment with Canova increased total numbers of leukocytes and lymphocytes. Among lymphocytes, TCD4, increased in normal-treated group and B and NK cells in S180-treated groups. The results reflect enhanced immune response of the host after treatment with Canova.

The regulation of the synthesis of ferritin and erythroid 5-aminolevulinate synthase in mammalian cells is mediated by the interaction of the iron regulatory factor (IRF) with a specific recognition site, the iron responsive element (IRE), in the 5' untranslated regions (UTRs) of the respective mRNAs. A new modular expression system was designed to allow reconstruction of this regulatory system in Saccharomyces cerevisiae. This comprised two components: a constitutively expressed reporter gene (luc; encoding luciferase) preceded by a 5' UTR including an IRE sequence, and an inducibly expressed cDNA encoding human IRF. Induction of the latter led to the in vivo synthesis of IRF, which in turn showed IRE-binding activity and also repressed translation of the luc mRNA bearing an IRE-containing 5' UTR. The upper stem-loop region of an IRE, with no further IRE-specific flanking sequences, sufficed for recognition and repression by IRF. Translational regulation of IRE-bearing mRNAs could also be demonstrated in cell-free yeast extracts. This work defines a minimal system for IRF/IRE translational regulation in yeast that requires no additional mammalian-specific components, thus providing direct proof that IRF functions as a translational repressor in vivo. It should be a useful tool as the basis for more detailed studies of eukaryotic translational regulation.
