RESUMO

Angiogenesis is considered one of the hallmarks of cancer, assisting tumor progression and metastasis. The mesoionic compound, MI-D, can induce cell death and provoke cytoskeletal and metabolic changes in cancer cells. Using in vitro and in vivo models, this study aimed to evaluate the effects of MI-D on the viability of human endothelial cells (EC) and its ability to inhibit tumor-induced angiogenesis induced by tumoral cells. For in vitro analysis, colon carcinoma (HT29) and endothelial (EA.hy926) cells were used as the tumoral and angiogenesis models, respectively. To evaluate cytotoxicity, methylene blue viability stain and annexin-V/7AAD tests were performed with both cell types. For the angiogenesis experiments, scratch wound healing and capillary tube-like formation assays were performed with the EC. The in vivo tests were performed with the chorioallantoic membrane (HET-CAM) methodology, wherein gelatin sponge implants containing MI-D (5, 25, and 50 µM), HT29 cells, or both were grafted in the CAM. Our data showed that MI-D induced apoptosis in both endothelial and colon carcinoma cells, with a strong cytotoxic effect on the tumoral lineage. The drug inhibited the EC's migration and capillary-like structure formation in vitro. In the HET-CAM assays, MI-D reduced the number of blood vessels in the membrane when grafted alone and accompanied by tumor cells. In this study, MI-D interfered in important steps of angiogenesis, such as maintenance of endothelial cell viability, migration, formation of capillary-like structures, as well tumor-induced neovascularization, reinforcing the hypothesis that MI-D might act as an inhibitor of angiogenesis, and a potential antitumor agent.

BACKGROUND: Chagas disease kills around 10,000 people yearly, primarily in Latin America, where it is prevalent. Current treatment has limited chronic effectiveness, is unsafe, and has substantial side effects. As a result, the use of oxadiazole derivatives and similar heterocyclic compounds as bioisosteres are well known, and they are prospective candidates in the hunt for novel anti-Trypanosoma cruzi chemicals. Recent research has revealed that the cysteine protease cruzain from T. cruzi is a validated target for disease treatment. OBJECTIVE: Thus, using a molecular dynamics simulation, the current study attempted to determine if a significant interaction occurred between the enzyme cruzain and its ligand. RESULTS: Interactions with the catalytic site and other critical locations were observed. Also, the RMSD values suggested that the molecule under research had stable interactions with its target. CONCLUSION: Finally, the findings indicate that the investigated molecule 2b can interfere enzymatic activity of cruzain, indicating that it might be a promising antichagasic drug.

PURPOSE: To develop an ABP-MRI to evaluate response to NAC for invasive breast carcinoma. STUDY TYPE: A single-center, cross-sectional study. SUBJECTS: A consecutive series of 210 women with invasive breast carcinoma who underwent breast MRI after NAC between 2016 and 2020. FIELD STRENGTH/SEQUENCE: 1.5 T / Dynamic contrast-enhanced. ASSESSMENT: MRI scans were independently reevaluated, with access to dynamic contrast-enhanced without contrast and to the first, second, and third post-contrast time (ABP-MRI 1-3). STATISTICAL TESTS: The diagnostic performance of the ABP-MRIs and the Full protocol (FP-MRI) were analyzed. The Wilcoxon non-parametric test (p-value <0.050) was used to compare the capability in measuring the most extensive residual lesion. RESULTS: The median age was 47 (24-80) years. ABP-MRI 1 showed higher specificity (84.6%; 77/91) but a higher probability of false-negatives (16.8%) and lower sensitivity (83.2%; 99/119) than ABP-MRI 2,3 and the FP-MRI, which were identical in specificity (81.3%; 74/91), probability of false-negatives (8.4%), and sensitivity (91.6%; 109/119). ABP-MRI 2 showed a mean underestimation of only 0.03 cm in the measurement of the longest axis of the residual lesion (p = 0.008) with an average reduction in the acquisition time of 75%, compared with the FP-MRI. CONCLUSION: ABP-MRI 2 showed diagnostic performance equivalent to the FP-MRI with a 75% reduction in the acquisition time.

This study evaluated the effects of palm kernel oil (PKO) in the diet of lambs on carcass characteristics, quality, and fatty acid profile of the meat. Forty uncastrated male Santa Inês lambs were used and divided among the treatments: PKOzero without inclusion; PKO1.3­added 1.3%; PKO2.6­added 2.6%; PKO3.9­added 3.9%; PKO5.2­added 5.2%. The carcass characteristics, the variables related to meat color, and the chemical composition of the Longissimus lumborum of lambs were not affected by the PKO inclusion. The weight of the carcasses at slaughter, hot and cold, half carcass, loin-eye area, and commercial cuts decreased linearly when PKO was added to the lamb diet (p < 0.01). CCY decreased linearly to the inclusion level of 2.66% PKO (RMSE 2.204). Total conjugated linoleic acid CLA and C18:3 n-3 GA concentrations remained stable until the inclusion levels of 3.44% PKO (RMSE 0.0956) and 2.17% (RMSE 0.0637), decreasing its concentrations as the increased level of PKO. The presence of PKO in the lambs' diet up to the level of 5.2% did not change the meat quality characteristics; thus, from the point of view of lamb meat production and fatty acid profile, the inclusion of PKO is not beneficial.

This work describes the synthesis, characterization and in vitro anticancer activity of two platinum(II) complexes of the type [Pt(L1)2(1,10-phen)] 1 and [Pt(L2)2(1,10-phen)] 2, where L1 = 5-heptyl-1,3,4-oxadiazole-2-(3H)-thione, L2 = 5-nonyl-1,3,4-oxadiazole-2-(3H)-thione and 1,10-phen = 1,10-phenanthroline. As to the structure of these complexes, the X-ray structural analysis of 1 indicates that the geometry around the platinum(II) ion is distorted square-planar, where two 5-alkyl-1,3,4-oxadiazol-2-thione derivatives coordinate a platinum(II) ion through the sulfur atom. A chelating bidentate phenanthroline molecule completes the coordination sphere. We tested these complexes in two breast cancer cell lines, namely, MCF-7 (a hormone responsive cancer cell) and MDA-MB-231 (triple negative breast cancer cell). In both cells, the most lipophilic platinum compound, complex 2, was more active than cisplatin, one of the most widely used anticancer drugs nowadays. DNA binding studies indicated that such complexes are able to bind to ct-DNA with Kb values of 104 M-1. According to data from dichroism circular and fluorescence spectroscopy, these complexes appear to bind to the DNA in a non-intercalative, probably via minor groove. Molecular docking followed by semiempirical simulations indicated that these complexes showed favorable interactions with the minor groove of the double helix of ct-DNA in an A-T rich region. Thereafter, flow cytometry analysis showed that complex 2 induced apoptosis and necrosis in MCF-7 cells.

Introduction: The new coronavirus pandemic (COVID-19) is unprecedented in recorded human history. It spread from Wuhan, China, in early December, 2019, crossing the entire planet and reaching Brazilian shores in the following February. It was declared a pandemic on March 11, 2020, with the first case  recorded in the city of Uberaba, state of Minas Gerais, Brazil, on March 18, 2020. Since then, we have been collecting data and assessing the evolution of this fatal disease. Objective: In this work, we report the epidemiological characteristics of one year of the COVID-19 in Uberaba, and discuss its implications to the general public. Method: This is an observational, descriptive, documentary and retrospective study to describe the epidemiological profile of COVID-19 cases in the city of Uberaba from March 18, 2020 to March 17, 2021. Results: The study shows that the young-working age population are those who most spread the virus; however, the elderly are those who suffer and die the most, with slight differences regarding sex. This is in line with the reported national and international epidemiological profiles that show a shifting tendency of younger generations to be increasingly active on the evolution of the pandemic. We observed two major peaks on the two epidemiological time-series, confirmed cases and deaths, with an average age of 41 years old for the confirmed cases and 68 for the confirmed deaths. It was also reported that the lethality rate was 2.45%, and 80.00% of the confirmed deaths suffered from some previous health condition. Conclusions: In this sense, a permanent epidemiological surveillance has to take place in order to guide public health counter-measurements. Introdução: A pandemia do novo coronavírus (COVID-19) é inédita na história humana registrada. Espalhou-se de Wuhan, na China, no início de dezembro de 2019, cruzando todo o planeta e chegando à costa brasileira no mês de fevereiro seguinte. Foi declarada pandemia em 11 de março de 2020, com o primeiro caso registrado na cidade de Uberaba, estado de Minas Gerais, Brasil, em 18 de março de 2020. Desde então, estamos coletando dados e avaliando a evolução dessa fatalidade doença. Objetivo: Neste trabalho, relatamos as características epidemiológicas de um ano da COVID-19 em Uberaba e discutimos suas implicações para o público em geral. Método: Trata-se de um estudo observacional, descritivo, documental e retrospectivo para descrever o perfil epidemiológico dos casos de COVID-19 na cidade de Uberaba no período de 18 de março de 2020 até 17 de março de 2021. Resultados: O estudo mostra que a população jovem em idade ativa é a que mais espalha o vírus, no entanto, os idosos são os que mais sofrem e morrem, com pequenas diferenças em relação ao sexo. Isso está de acordo com os perfis epidemiológicos nacionais e internacionais relatados que mostram uma tendência de mudança das gerações mais jovens a serem cada vez mais ativas na evolução da pandemia. Observamos dois grandes picos nas duas séries temporais epidemiológicas, casos confirmados e óbitos, com média de idade de 41 anos para os casos confirmados e 68 para os óbitos confirmados. Também foi relatado que a taxa de letalidade foi de 2,45%, e 80,00% das mortes confirmadas sofriam de alguma condição de saúde anterior. Conclusões: Nesse sentido, uma vigilância epidemiológica permanente deve ocorrer para orientar as contramedidas de saúde pública. The aim of this study was to evaluate the effects of the inclusion of palm kernel oil (PKO) in a lamb diet on nutrient intake, digestibility, ingestive behavior, nitrogen balance, blood metabolites, rumen fermentation parameters, and animal performance. Three experimental trials were conducted. The treatments consisted of varying levels of PKO included in the diet, with PKOzero = no PKO inclusion, PKO1.3 = 1.3% addition, PKO2.6 = 2.6% addition, PKO3.9 = 3.9% addition, and PKO5.2 = 5.2% addition, based on the total dry matter (DM) of the diet. With the inclusion of PKO in the diet, linear decreases in DM (p < 0.001), crude ash (p < 0.001), crude protein (CP) (p < 0.001), neutral detergent fiber (NDF) (p < 0.001), nonfibrous carbohydrate (NFC) (p < 0.001), and total digestible nutrient (TDN) (p = 0.021) intake were observed, as was an increase in ether extract (EE) intake (p < 0.001). The digestibility coefficients of NDF and NFC were not affected by PKO addition to the diet. However, the digestibility of DM (p = 0.035), EE (p < 0.001), CP (p < 0.001), and TDNs (p < 0.001) increased when PKO was added to the lambs' diet. Reductions in N intake (p < 0.001), fecal nitrogen excretion (p < 0.001), and microbial protein production (p < 0.001) were noted with increasing PKO levels. Serum cholesterol increased (p < 0.001) while serum GGT enzyme concentrations in the blood decreased (p = 0.048) with increasing PKO levels. PKO addition had no effect on total weight gain and average daily gain; however, feed conversion improved (p = 0.001) with increasing PKO levels. The intake, digestibility, ingestive behavior, and growth performance of lambs with PKO1.3 added to their diet were similar to animals that did not receive PKO, meaning that PKO could be an alternative energy source for growing lambs because it does not harm animal performance and can lower the cost of feed.

Vegetable oils can be used to increase energy density in diets; manipulate rumen fermentation; and alter the capacity for degradation, digestion and absorption of nutrients. Two experiments were conducted to evaluate palm kernel oil (PKO) in the diet of confined bulls with the inclusion of 0.0, 11.5, 23.0 and 34.6 g PKO/kg dry matter (DM). The first experiment evaluated nutrient intake, performance, ingestive behavior and carcass characteristics. In the second experiment, steers crossbred with a ruminal cannula were used to evaluate digestibility, nitrogen balance, microbial protein synthesis, short-chain fatty acid levels and protozoal counts. The results showed that the inclusion of PKO linearly reduced intake in kg/day (DM, crude protein-CP, neutral detergent fiber-NDFap, nonfibrous carbohydrates-NFC and total digestible nutrient-TDN) and digestibility (DM, NDFap and TDN). Ether extract intake increased quadratically with the predicted maximum intake of 15.4 g/kg DM. Regarding ingestive behavior, there was a quadratic increase in rumination time and a quadratic reduction in idle time. Nitrogen balance, nitrogen intake, nitrogen retention, microbial protein production, acetate, butyrate, acetate/propionate ratio and protozoa count showed linear decreases due to dietary PKO inclusion. Regarding the carcass characteristics, linear decreases were observed for the final weight, average daily gain, hot carcass weight, cold carcass weight, hot carcass yield, cold carcass yield, loin eye area and subcutaneous fat thickness. The inclusion of PKO at up to 34.6 g/kg DM in diets for confined bulls reduces intake, negatively affecting digestibility, performance and carcass characteristics.

Lipid supplementation through vegetable oils in diets for ruminants can be a nutritional strategy to increase energy density, manipulate ruminal fermentation and change the physicochemical composition and sensorial properties of meat. This study evaluated the optimal dietary inclusion of palm kernel oil (PKO) for Nellore bulls on meat quality. The diets consisted of 0.0, 11.5, 23.0, and 34.6 g/kg dry matter (DM) PKO levels. PKO inclusion did not influence the centesimal composition, pH, color indices, water holding capacity, cooking loss, or shear force of the beef. There were linear increases in the concentrations of lauric acid (C12:0) and myristic acid (C14:0) in the bull's meat. However, palmitic acid (C16:0), oleic acid (C18:0), vaccenic acid (t-11-C18:1) and conjugated linoleic acid (CLA), ∑n - 6, ∑n - 3, ∑n - 6/∑n 3, the hypocholesterolemic: hypercholesterolemic ratio of the fatty acid content, and the thrombogenicity index were not affected. There were linear reductions in the oleic acid meat concentration (c-9-C18:1) and elongated enzymatic activity when PKO was added to the bull diet. The atherogenicity index increased linearly due to PKO inclusion in the bull diet. No effect of the inclusion of PKO on meat flavor, perception of tenderness, juiciness, or global acceptance from the sensorial evaluation was recorded. The inclusion of PKO up to 34.6 g/kg DM can be recommended to supplement young bulls with no effects on meat composition and quality characteristics.

Exposure to mercury (Hg) and pesticides (o.p'DDT, p.p'DDT, o.p'DDE, and p.p'DDE) in the Amazon through eating fish is of concern due to the large participation of this food in the diet of traditional fishing communities. The aim of this study was to evaluate the estimated daily intake (EDI) and the incremental lifetime cancer risk associated with Hg and o.p'DDT, p.p'DDT, o.p'DDE, and p.p'DDE in an Amazonian community. The results showed that for Hg, the EDI from carnivorous and detritivorous fish had the highest values, while for pesticides, the EDI from detritivorous fish intake had the highest value. The incremental lifetime cancer risk was below the permitted limit. A recommendation for controlling the high risk of exposure includes the reduction of detritivorous fish ingestion and/or replacement with herbivorous fish, which had lower EDI. We highlight the importance of investigating the human dietary patterns when estimating risk of exposure to Hg and pesticides.

Abstract Background: Inclusion of urea and calcium oxide (CaO) during sugarcane ensilage has been hypothesized to improve dairy cow performance. Objective: To evaluate the use of urea and calcium oxide as additives to preserve sugarcane silage quality and determine whether they improve dairy cow performance and metabolic efficiency. Methods: Four diets were used: sugarcane silage without additive (SS), sugarcane silage with 10 g/kg urea (SU), sugarcane silage with 5 g/kg urea + 5 g/kg CaO (SUC), and sugarcane silage with 10 g/kg CaO (SC). Eight crossbred cows at approximately 100 days of lactation were distributed in two 4x4 Latin squares, and their diets were formulated to reach an average milk production of 15 kg/day. Results: Dry matter, non-fibrous carbohydrates, organic matter, total carbohydrates, and total digestible nutrient intake were the highest (p<0.05) for the cows fed SC. Urinary excretion of nitrogen was higher (p<0.05) for cows fed diets based on SUC than those fed diets with SC. Milk urea nitrogen content was the highest (p<0.05) in animals fed SU diets. Nitrogen balance was positive in all treatments; however, the microbial protein synthesis was low for all diets. Conclusions: The diet with SU showed the highest nitrogen loss through the milk. Among the evaluated silages, those with SC and SUC are recommended to promote a high intake of nutritional components.


Resumen Antecedentes: La hipótesis fue que una dieta con combinación de urea y óxido de calcio (CaO) para el ensilaje de caña de azúcar, puede mejorar la producción de vacas lecheras cruzadas. Objetivo: Evaluar la urea y el óxido de calcio como aditivos para preservar la calidad del ensilaje de caña de azúcar y determinar si mejoran el rendimento y la eficiencia metabólica de vacas lecheras. Métodos: Cuatro dietas fueron utilizadas: ensilaje de caña de azúcar sin aditivo (SS), ensilaje de caña de azúcar con 10 g/kg de urea (SU), ensilaje de caña de azúcar con 5 g/kg de urea + 5 g/kg de CaO (SUC) y ensilaje de caña de azúcar con 10 g/kg de CaO (SC). Ocho vacas cruzadas fueron evaluadas con una producción media de leche de 15 kg/día y aproximadamente 100 días de lactación, distribuidas en dos cuadrados latinos 4x4. Resultados: El consumo de materia seca, carbohidratos no fibrosos, materia orgánica, carbohidratos totales y nutrientes digestibles totales fue mayor (p<0,05) cuando las vacas fueron alimentadas con dietas conteniendo SC. La excreción de nitrógeno en la orina fue mayor (p<0,05) cuando las vacas fueron alimentadas con dietas basadas en SUC que las alimentadas con dietas SC. El nitrógeno ureico de la leche fue mayor (p<0,05) en animales alimentados con dietas con SU. El balance de nitrógeno fue positivo en todos los tratamientos, sin embargo, la síntesis de proteína microbiana fue baja para todas las dietas. Conclusión: La dieta con SU presentó la mayor pérdida de nitrógeno por medio de la leche. Entre los ensilajes evaluados, aquellos con SC y SUC son recomendados por promover un mayor consumo de componentes nutricionales.

Objetivo: Avaliar ureia e óxido de cálcio como aditivos para preservar a qualidade da silagem de cana de açúcar: melhoria da produção e eficiência metabólica de vacas leiteiras. Métodos: Quatro dietas foram utilizadas: silagem de cana de açúcar sem aditivo (SS), silagem de cana de açúcar com 10 g/kg de ureia (SU), silagem de cana de açúcar com 5 g/kg de ureia + 5 g/kg de CaO (SUC) e silagem de cana de açúcar com 10 g/kg de CaO (SC). Oito vacas mestiças foram avaliadas com uma produção media de 15 kg/dia e aproximadamente 100 dias de lactação, distribuídas em dois quadrados latinos 4x4. Resultados: O consumo de matéria seca, carboidratos não fibrosos, matéria orgânica, carboidratos totais e nutrientes digestíveis totais foi maior (p<0,05) quando as vacas foram alimentadas com dietas contendo SC. A excreção de nitrogênio na urina foi maior (p<0,05) quando as vacas foram alimentadas com dietas baseadas em SUC do que aqueles animais alimentados com dietas SC. O nitrogênio ureico do leite foi maior (p<0,05) em animais alimentados com dietas com SU. O balanço de nitrogênio foi positivo em todos os tratamentos, no entanto, a síntese de proteína microbiana foi baixa para todas as dietas. Conclusão: A dieta com SU apresentou maior perda de nitrogênio por meio do leite. Entre as silagens avaliadas, aquelas com SC e SUC são recomendadas por promover maior consumo de componentes nutricionais.

Objetivo: Descrever possíveis contribuições da religiosidade no enfrentamento da Síndrome de Burnout (SB) em profissionais da Enfermagem. Métodos: Trata-se de uma revisão de literatura, realizou-se a busca de estudos em periódicos nacionais nas bases de dados: SCIELO e Google Acadêmico, publicados no período de 2010 a 2020. Para critérios de inclusão foram utilizados: Síndrome de Burnout; Enfermagem; Transtornos Mentais, de exclusão: Trauma; Convivência; Contentamento. Tomamos a Análise de Conteúdo Temática proposta por Minayo para analisar o material. Resultados: Foi possível identificar que a dinâmica laboral dos profissionais de enfermagem provoca fatores estressores, logo a premência de intervenções. Conclusão: As estratégias de enfrentamento ao diagnóstico da Síndrome de Burnout em profissionais da enfermagem necessitam de ações que favoreçam o equilíbrio emocional e espiritual, assim estes trabalhadores poderão atender as responsabilidades que a função exige. Ressalta-se que, independente da crença, busca-se um tratamento preventivo e ou curativo a partir da vivência religiosa.(AU)


Objective: To describe the possible contributions of religiosity in coping with Burnout Syndrome in nursing professionals. Methods: This is a literature review and the search for studies in national journals was carried out in a database: SCIELO and Google Scholar, published in the period from 2010 to 2020. For inclusion criteria we used: Burnout Syndrome; Nursing; Mental Disorders, exclusion criteria: Trauma, Coexistence, Contentment. We took the Thematic Content Analysis proposed by Minayo to analyze the material. Results: It was possible to identify that the work dynamics of nursing professionals causes stressors, hence the urgency of interventions. Conclusion: The strategies for coping with the diagnosis of Burnout Syndrome in nursing professionals need actions that favor the emotional and spiritual balance, so that these workers will be able to meet the responsibilities that the function requires. It is noteworthy that, regardless of belief, a preventive and / or curative treatment is sought from the religious experience.(AU)


Objetivo: Describir las posibles contribuciones de la religiosidad en el afrontamiento del Síndrome de Burnout en profesionales de enfermería. Conclusión: Las estrategias para afrontar el diagnóstico de Síndrome de Burnout en los profesionales de enfermería necesitan acciones que favorezcan el equilibrio emocional y espiritual, para que estos trabajadores puedan cumplir con las responsabilidades que la función requiere. Es de destacar que, independientemente de la creencia, se busca un tratamiento preventivo y o curativo a partir de la experiencia religiosa.(AU)

A total of forty-three compounds were synthesized, including thirty-two new ones. Among those compounds, seventeen were selected and tested on human tumor cell lines: PC-3 (prostate adenocarcinoma), HCT-116 (colorectal tumor), NCIH-460 (lung carcinoma), SKMEL-103 (melanoma) and AGP-01 (gastric tumor). Alkynylated 1,2,4-oxadiazoles 2m, 3g and 3k exhibited antiproliferative activities against NCIH-460 in culture. Alkynylated N-cyclohexyl-1,2,4-oxadiazoles 3a-m and bis-heterocycle glucoglycero-1,2,3-triazole-N-cyclohexyl-1,2,4-oxadiazole derivatives 5a-k and 6-11 were evaluated for their in vitro efficacy towards Mycobacterium tuberculosis (Mtb) H37Ra and H37Rv strains. In general, glycerosugars conjugated to 1,2,4-oxadiazole via a 1,2,3-triazole linkage (5a, 5e, 5j, 5k, and 7) showed in vitro inhibitory activity against Mtb (H37Rv). The largest molecules bis-triazoles 10 and 11, proved inactive against TB. Probably, the absence of the N-cyclohexyl group in compound 8 and 1,2,4-oxadiazole nucleus in compound 9 were responsible for its low activity. Glucoglycero-triazole-oxadiazole derivatives 5e (10 µM) and 7 (23.9 µM) were the most promising antitubercular compounds, showing a better selective index than when tested against RAW 264.7 and HepG2 cells. Vero cell were used to investigate cytotoxicity of compounds 5a, 5h, 5j, 5k, and these compounds showed good cell viability. Further, in silico studies were performed for most active compounds (5e and 7) with potential drug targets, DprE1 and InhA of Mtb to understand possible interactions aided with molecular dynamic simulation (100ns).

Objetivou-se no presente estudo comparar as técnicas de coleta de urina via sondagem uretral e cistocentese guiada por ultrassom, afim de verificar se o método de coleta pode influir nos resultados laboratoriais. Foram utilizados 12 cães machos, sem histórico de enfermidades, dos quais coletou-se cinco mililitros (mL) de urina via sondagem uretral e cinco mL via cistocentese guiada por ultrassom, ambas no mesmo momento. Posteriormente foi realizada a análise física (cor, odor, densidade, turbidez), química (urobilinogênio, glicose, corpos cetônicos, bilirrubina, proteína, nitrito, pH, sangue e leucócitos) e sedimentoscopia (avaliação de 10 campos de luz, objetiva de 40x). Cilindros urinários, cristais, corpúsculos gordurosos, espermatozoides, bactérias e células vesicais foram classificados qualitativamente como: ausentes (0), discretos (1), moderados (2) e intensos (3). Hemácias, leucócitos, e células de descamação foram quantificadas a partir da média dos campos analisados. As análises bioquímicas de microalbuminúria, creatinina e proteína total urinárias foram realizadas a partir do sobrenadante urinário, removido das amostras após centrifugação, e utilizados kits reagentes, conforme recomendação do fabricante, sendo a leitura em espectrofotômetro. Em todos os testes realizados os valores de p encontrados foram superiores 0,05 (p>0,05), excluindo-se a possibilidade de haver diferenças significativas dos resultados laboratoriais obtidos pelas duas formas de coleta.

Red blood cells, leukocytes and desquamation cells were quantified from the mean of the analyzed fields. The urine supernatants were obtained after centrifugation and were used for biochemical analyzes of microalbuminuria, urinary protein and creatinine. The reagent kits were used as recommended by the manufacturer and the samples were read by spectrophotometry. All tests presented p values higher than 0,05 (p>0,05), excluding the possibility of significant differences between the laboratory results of both forms of urine collection.

Angiotensin-converting enzyme 2 (ACE2) is the host cellular receptor that locks onto the surface spike protein of the 2002 SARS coronavirus (SARS-CoV-1) and of the novel, highly transmissible and deadly 2019 SARS-CoV-2, responsible for the COVID-19 pandemic. One strategy to avoid the virus infection is to design peptides by extracting the human ACE2 peptidase domain α1-helix, which would bind to the coronavirus surface protein, preventing the virus entry into the host cells. The natural α1-helix peptide has a stronger affinity to SARS-CoV-2 than to SARS-CoV-1. Another peptide was designed by joining α1 with the second portion of ACE2 that is far in the peptidase sequence yet grafted in the spike protein interface with ACE2. Previous studies have shown that, among several α1-based peptides, the hybrid peptidic scaffold is the one with the highest/strongest affinity for SARS-CoV-1, which is comparable to the full-length ACE2 affinity. In this work, binding and folding dynamics of the natural and designed ACE2-based peptides were simulated by the well-known coarse-grained structure-based model, with the computed thermodynamic quantities correlating with the experimental binding affinity data. Furthermore, theoretical kinetic analysis of native contact formation revealed the distinction between these processes in the presence of the different binding partners SARS-CoV-1 and SARS-CoV-2 spike domains. Additionally, our results indicate the existence of a two-state folding mechanism for the designed peptide en route to bind to the spike proteins, in contrast to a downhill mechanism for the natural α1-helix peptides. The presented low-cost simulation protocol demonstrated its efficiency in evaluating binding affinities and identifying the mechanisms involved in the neutralization of spike-ACE2 interaction by designed peptides. Finally, the protocol can be used as a computer-based screening of more potent designed peptides by experimentalists searching for new therapeutics against COVID-19.

OBJECTIVES: The aim of this study was to contribute to the design of specialized parenteral nutrition protocols in surgery by evaluating the dynamics of polyunsaturated fatty acid (PUFA) concentrations in different body pools after the infusion of fish oil-containing lipid emulsion (FOLE) in rats that had undergone surgical central venous catheterization (CVC). METHODS: After 5-d adaptation in metabolic cages, 78 male Lewis rats (300-450 g) fed a standard diet were sacrificed (baseline control) or had only CVC (surgical control) or also received a 72-h infusion of a parenteral lipid emulsion with or without fish oil. The catheterized animals were sacrificed 0 (T0), 2 (T2), 6 (T6), and 12 h (T12) after the infusion ended. Gas chromatography was used to determine the concentrations of eicosapentaenoic (EPA), docosahexaenoic (DHA), and arachidonic (ARA) acids and the ω-3 to ω-6 ratio in plasma, liver, and blood leukocytes. Kruskal-Wallis and Wilcoxon tests were applied to plasma and liver data and descriptive analysis to leukocyte data. RESULTS: Plasma, liver, and leukocytes exhibited almost undetectable EPA and DHA and detectable ARA concentrations at baseline. Immediately after FOLE infusion (T0), these PUFAs changed in all pools, resulting in a higher ratio of ω-3 to ω-6 compared with rats with no FOLE infusion (P < 0.05). All these changes decreased over time, with residual effects remaining until T6 in plasma, T12 in liver, and only until T2 in leukocytes. CONCLUSION: Data from this study suggest that ω-3 PUFAs are cleared early after the end of FOLE infusion, mainly in leukocytes. This should be considered when FOLEs are applied for immunomodulatory purposes in surgery.

Objetivos analisar as características epidemiológicas e distribuição dos casos novos de hanseníase na população de Paulo Afonso, Bahia, entre 2000 e 2015. Método estudo descritivo com todos os casos novos de hanseníase notificados no Sistema de Informações de Agravos de Notificação. Foram utilizadas variáveis sociodemográficas e clínicas e calculados os coeficientes de detecção anuais. Estimativas populacionais foram obtidas no Instituto Brasileiro de Geografia e Estatística. Resultados a maioria dos 1.069 casos novos notificados foi do sexo feminino (57,2%), residiam na zona urbana (92%), raça/cor parda (66,92%), ensino fundamental (67,6%), maiores de quinze anos (92,5%), forma clínica tuberculoide (48,26%), paucibacilares (62,3%), grau de incapacidade física no diagnóstico (45,93%) e na cura (29,75%). Conclusão o perfil epidemiológico da hanseníase em Paulo Afonso é similar ao nacional, houve variações nos coeficientes de detecção da hanseníase entre os anos e a análise espacial evidenciou distribuição heterogênea, com maior concentração de casos nos bairros periféricos.

Se utilizaron variables sociodemográficas y clínicas y se calcularon los coeficientes anuales de detección. Las estimaciones de población se obtuvieron del Instituto Brasileño de Geografía y Estadística. Resultados la mayoría de los 1.069 nuevos casos notificados fueron mujeres (57,2%), que viven en el área urbana (92%), raza/color parda (66,92%), escuela primaria (67,6%), con más de quince años (92,5%), forma clínica de tuberculoide (48,26%), paucibacilario (62,3%), grado de discapacidad física al diagnóstico (45,93%) y en la cura (29,75%). Conclusión el perfil epidemiológico de la lepra en Paulo Afonso es similar al nacional, hubo variaciones en los coeficientes de detección de lepra entre años y el análisis espacial mostró una distribución heterogénea, con una mayor concentración de casos en barrios periféricos.

Conclusion the epidemiological profile of leprosy in Paulo Afonso is similar to the national one, with variations in leprosy detection coefficients between years and spatial analysis showing heterogeneous distribution, with a higher concentration of cases in peripheral neighborhoods.
