Rhipicephalus microplus is the main ectoparasite of cattle and its parasitism can reduce weight gain of hosts, in addition to causing anemia, increasing the risk of myiasis, and resulting in the transmission of Babesia bovis and Anaplasma marginale, among other pathogens. The use of synthetic chemicals plays an important role in controlling these ticks. However, its frequent and indiscriminate use has contributed to the selection of resistant strains, resulting in greater interest in the search for natural-origin products. The shrub Callistemon viminalis (Myrtaceae), also known as weeping bottlebrush, is known for its antibacterial, acaricidal, repellent, and antifungal activities, but there are no reports in the literature about its effects on the internal morphology of ticks. This study aimed to extract and characterize the essential oil obtained from the leaves of C. viminalis. Additionally, its effects on the ovary morphology of engorged R. microplus were assessed through histological, histochemical, and morphometric techniques. Exposure to C. viminalis caused dose-dependent morphological changes, such as cellular alterations in the epithelial layer lining the ovary lumen and the pedicel, irregularity of the chorion and oocyte shape, changes in protein and carbohydrate content, decrease in oocyte size, reduction in the size of the nucleus as well as cytoplasmic and nucleolar vacuolation. Thus, C. viminalis essential oil exhibited a toxic effect on the reproductive system of R. microplus, which may result in the reproductive impairment of this tick species.

In addition to pesticidal activity, the capacity of natural compounds to inhibit the reproduction of parasites emerge as an important alternative tick control method. In this context, Satureja spp. stand out due to their recognized pesticidal properties. Among parasites of veterinary importance, the cattle tick, Rhipicephalus microplus, is responsible for great economic losses in livestock and transmission of relevant pathogens. Thus, the aim of this study was to evaluate the effects of different concentrations of the essential oil of Satureja montana L. on the ovary morphology of R. microplus engorged females through histological and histochemical techniques. The most remarkable morphological changes found were: cytoplasmic vacuolation of germ cells, irregular and thicker chorion, irregular oocyte shape, ring-shaped nucleolus, decrease in protein and carbohydrate content in oocytes, in addition to cellular changes in the oviduct and pedicel. All morphological changes were assessed using a semiquantitative method already established in the literature. Ticks exposed to 5.0 µl/ml of essential oil showed the most significant changes when compared to control groups. Thus, the essential oil of S. montana L. damaged the reproductive system of R. microplus, which may impair ticks' offspring production and promote a long-term control of this species. HIGHLIGHTS: The essential oil of Satureja montana L. affects the ovary morphology of the cattle tick. The main morphological alterations found were cytoplasmic vacuolation, irregular and thicker chorion and irregular oocyte shape. These alterations may impair the development of eggs.

Several studies searching for methods to control Rhipicephalus sanguineus s.l., (dog tick) infestations have been developed aiming to minimize the damages caused by these ectoparasites to the hosts and the environment, which is harmed by the indiscriminate use of toxic acaricide products. In this scenario, neem oil has been used as a natural alternative against ticks, once this chemical has repellent properties and interferes in the growth regulation of these ectoparasites, inhibiting ecdysis. The present study evaluated the effects of azadirachtin-enriched neem oil on the integument of semi-engorged R.sanguineus s.l., females through morphohistological techniques. The results showed the occurrence of significant morphological and histochemical alterations, mainly in the females exposed to higher concentrations, which demonstrates the dose-dependent action of the chemical. A decrease in the cuticle thickness was observed, as well as a modification in the distribution of the epithelial cells, which displayed pyknotic and fragmented nuclei, and intensely vacuolated cytoplasm, indicating that these cells would be undergoing death processes. These morphological alterations observed in the integument of the females exposed to the azadirachtin-enriched neem oil encourage the use of this chemical as a strategy to control these ectoparasites.

The present study performed an analysis of the ultrastructural changes induced by andiroba seed oil in the synganglion of Rhipicephalus sanguineus female ticks, aiming to provide scientific grounds to help in the creation of more specific and efficient methods of control. The synganglion consists of a mass of fused nerves externally covered by the neural lamella, a uniform and acellular layer. Just below, the perineurium is found, formed by glial cells. Internally, the synganglion is subdivided into an outer cortical region (cortex), which contains the cellular bodies of the neural cells and an inner region, the neuropile, formed by a set of nerve fibers (extensions of the neural cells). The results showed that the synganglion of females exposed to andiroba oil showed structural changes, such as: irregular and apparently thinner neural lamella, perineurium glial cells presenting large cytoplasmic vacuoles, decrease in the extensions of glial cells, separation of cortex cells, which were formerly attached through their membranes, neural cells presenting irregular plasma membranes and cytoplasm with autophagic vacuoles and mitochondria with disorganized cristae and in process of degeneration. This study confirmed the neurotoxic action of the andiroba oil, which would probably be able to impair the neural functions. Thus, it is suggested that this product has the potential to be used as an alternative method to control ticks.

O presente estudo teve como objetivo verificar a ação da aloxana em parâmetros metabólicos e do sistema imune 24 e 192 horas após sua aplicação em ratos Wistar. Para isso, oito ratos foram mantidos em jejum de 12 horas e receberam aloxana monoidratada, Sigma (32 mg/kg de peso corporal) via endovenosa. Foram registrados os valores de massa corporal, ingestão hídrica e alimentar e realizada a contagem de leucócitos totais previamente, 24 e 192 horas após a aplicação da aloxana. As análises de glicemia e trigliceridemia foram realizadas previamente e 192 horas após a aplicação da aloxana. Após 24 horas, a aplicação da aloxana causou aumento da ingestão hídrica e redução da massa corporal, ingestão alimentar e contagem de leucócitos. Na avaliação realizada 192 horas após a aplicação da aloxana houve recuperação da ingestão alimentar e contagem de leucócitos. Por outro lado, neste período houve aumento da glicemia e ingestão hídrica além de redução da massa corporal. Isso indica que parte dos sinais de diabetes causados pela aloxanamanifesta-se em curto prazo da administração da droga.


The present study aimed to verify action of alloxan in metabolic and immune parameters after 24 and 192 hours of the injection in Wistar rats. Thus, eight rats were fasted and received monohidrated alloxan Sigma (32 mg/kg body weight) via endovenous. Glycemia and trglyceridemia analyzes were performed before and 192 hours after alloxan application. After 24 hours, alloxan application increased water intake and decreased body mass, food intake and leucocytes counting. 192 hours after alloxan application, there was a recuperation in food intake and leucocytes counting. On the other hand, in this period there was an increase of glycemia and water intake and reduction of body mass. These results indicate that some of diabetic signs caused by alloxan occur in short-term after drug administration.
