Antimicrobial peptides (AMPs) constitute a complex network of 10-100 amino acid sequence molecules widely distributed in nature. While over 300 AMPs have been described in mammals, cathelicidins and defensins remain the most extensively studied. Some publications have explored the role of AMPs in COVID-19, but these findings are preliminary, and in vivo studies are still lacking. In this study, we report the plasma levels of five AMPs (LL-37, α-defensin 1, α-defensin 3, ß-defensin 1, and ß-defensin 3), using the ELISA technique (MyBioSource, San Diego, CA, United States, kits MBS2601339 (beta-defensin 1), MBS2602513 (beta-defensin 3), MBS703879 (alpha-defensin 1), MBS706289 (alpha-defensin 3), MBS7234921 (LL37)), and the measurement of six cytokines (tumor necrosis factor-α, interleukin-1ß, interleukin-6, interleukin-10, interferon-γ, and monocyte chemoattractant protein-1), through the magnetic bead immunoassay Milliplex® and the MAGPIX® System (MilliporeSigma, Darmstadt, Germany, kit HCYTOMAG-60 K (cytokines)), in 15 healthy volunteers, 36 COVID-19 patients without Acute Kidney Injury (AKI) and 17 COVID-19 patients with AKI. We found increased levels of α-defensin 1, α-defensin 3 and ß-defensin 3, in our COVID-19 population, when compared to healthy controls, along with higher levels of interleukin-6, interleukin-10, interferon-γ, and monocyte chemoattractant protein-1. These findings suggest that these AMPs and cytokines may play a crucial role in the systemic inflammatory response and tissue damage characterizing severe COVID-19. The levels of α-defensin 1 and α-defensin 3 were significantly higher in COVID-19 AKI group in comparison to the non-AKI group. Furthermore, IL-10 and the product IL-10 × IL-1B showed excellent performance in discriminating AKI, with AUCs of 0.86 and 0.88, respectively. Among patients with COVID-19, AMPs may play a key role in the inflammation process and disease progression. Additionally, α-defensin 1 and α-defensin 3 may mediate the AKI process in these patients, representing an opportunity for further research and potential therapeutic alternatives in the future.

This study aimed to evaluate the effects of essential oils from the fruits of Citrus sinensis and Xylopia aromatica, included in broiler feed, on blood parameters, the biometrics of digestive organs, bone analyses, and the biochemical profiles of the viscera, as well as the histomorphometry of the small intestine. In this study, 180 one-day-old male chicks of the Cobb 500 strain were fed a corn and soybean meal over three treatments and six replications, and the experimental design was completely randomized. The data were subjected to an analysis of variance and a Tukey test at a 5% significance level. The effect of the experimental diets on performance, blood parameters, biometrics of the digestive organs, bone analysis, and biochemical profiles of the viscera, as well as the histomorphometry of the small intestine, were evaluated. The compounds identified in the essential oil of X. aromatica were sylvestrene, α-pinene, and ß-pinene, while in C. sinensis they were limonene and myrcene. The essential oils of C. sinensis and X. aromatica had no significant effect on performance at 14 days. The effects of the presence of the essential oils of C. sinensis and X. aromatica on the response were beneficial: there were reductions in liver lipids, cholesterol, and triglycerides, and in the depths of the crypts in the jejunum of chickens. So, the essential oils from the fruits of C. sinensis and X. aromatica can be used in broiler chickens to improve the lipid profiles of birds without affecting their performance.

INTRODUCTION: Although fewer children have been affected by the severe form of the coronavirus disease 2019 (COVID-19), community-acquired pneumonia (CAP) continues to be the leading global cause of child hospitalizations and deaths. AIM: This study investigated the incidence of respiratory syncytial virus (RSV) as well its subtypes (RSV A and B), adenovirus (ADV), rhinovirus (HRV), metapneumovirus (HMPV), coronavirus (NL63, OC43, 229E and HKU1), parainfluenza virus subtypes (PI1, PI2 and PI3), bocavirus and influenza A and B viruses (FluA and FluB) in children diagnosed with CAP during the COVID-19 pandemic. METHODS: A total of 200 children with clinically confirmed CAP were initially recruited, of whom 107 had negative qPCR results for SARS-CoV-2 and were included in this study. Viral subtypes were identified using a real-time polymerase chain reaction in the nasopharyngeal swab samples. RESULTS: Viruses were identified in 69.2% of the patients. RSV infections were the most frequently identified (65.4%), with type RSV B being the most prevalent (63.5%). In addition, HCoV 229E and HRV were detected in 6.5% and 3.7% of the patients, respectively. RSV type B was associated with severe acute respiratory infection (ARI) and a younger age (less than 24 months). CONCLUSIONS: New strategies for preventing and treating viral respiratory infections, particularly RSV infections, are necessary.

Encapsulation via nanotechnology offers a potential method to overcome limited thermal and photo-stability of botanical pesticides. In this study, nanospheres of essential oils (NSEO) derived from Zanthoxylum rhoifolium Lam. fruit were characterized and evaluated for their photostability and insecticidal activity against Bemisia tabaci. Three major compounds of Z. rhoifolium fruits were detected by CG-MS: ß-phellandrene (76.8%), ß-myrcene (9.6%), and germacrene D (8.3%). The nanoprecipitation method was used to obtain homogeneous spherical NSEO, with ≥98% encapsulation efficiency. Tests with UV/Vis spectrophotometry showed significantly reduced photodegradation from exposed NSEO samples when compared with essential oil (EO) controls. Whitefly screenhouses bioassays with bean plants treated with 0.25, 0.5, 1 and 1.5% suspensions showed EO treatments in both free and nanoencapsulated forms reduced adult whitefly oviposition by up to 71%. In further tests, applications at 1.5% caused ≥64% mortality of second instar nymphs. When the test was conducted under high temperature and light radiation conditions, the insecticidal effect of NSEO treatments was improved (i.e., 84.3% mortality) when compared to the free form (64.8%). Our results indicate the insecticidal potential of EO-derived from Z. rhoifolium fruits with further formulation as nanospheres providing greater photostability and enhanced insecticidal activity against B. tabaci under adverse environmental conditions.

Belonging to the Brazilian flora, the species Hancornia speciosa (Gomes), known as mangabeira, has bioactive compounds of interest, such as flavonoids, xanthones, and proanthocyanidins. The objective of this study was to determine how the supplementation of sugars in culture medium affects the osmotic potential of the medium, as well as its influence on cell growth and on the concentration of phenolic compounds. For this purpose, after 90 days of subculture, 20 mL aliquots of the cultures were added to flasks containing 20 mL of medium with different sugars (glucose, fructose, sucrose, mannitol, and sorbitol) under a 16-h photoperiod with a spectral range between 400 and 700 nm of photosynthetically active radiation (45-55 µmol m-2 s-1) in a shaker at 110 rpm. After 30 days, the pH, electrical conductivity, osmotic potential, biomass accumulation, and concentrations of phenolic compounds were evaluated. Regardless of their concentration in the medium, the sugars sorbitol and mannitol provided more unfavorable conditions for water absorption at the cellular level, reducing the water potential of the medium. Sucrose favored greater water absorption and biomass accumulation. Among the various sugar concentrations, 3% (30 g/L) sucrose or glucose improved the accumulation of fresh and dry cell weight and the production of polyphenols such as chlorogenic acid, epicatechin, rosmarinic acid, hesperidin, rutin, and quercetin. In addition, they resulted in a higher osmotic potential of the medium and larger cells than other carbon sources. Despite the differences in cell size, no culture conditions compromised cell survival.

BACKGROUND: The progression and severity of COVID-19 vary significantly in the population. While the hallmarks of SARS-CoV-2 and severe COVID-19 within routine laboratory parameters are emerging, the impact of sex and age on these profiles is still unknown. METHODS: A multidimensional analysis was performed involving millions of records of laboratory parameters and diagnostic tests for 178 887 individuals from Brazil, of whom 33 266 tested positive for SARS-CoV-2. Analyzed data included those relating to complete blood cell count, electrolytes, metabolites, arterial blood gases, enzymes, hormones, cancer biomarkers, and others. FINDINGS: COVID-19 induced similar alterations in laboratory parameters in males and females. CRP and ferritin were increased, especially in older men with COVID-19, whereas abnormal liver function tests were common across several age groups, except for young women. Low peripheral blood basophils and eosinophils were more common in the elderly with COVID-19. Both male and female COVID-19 patients admitted to intensive care units displayed alterations in the coagulation system, and higher values for neutrophils, CRP, and lactate dehydrogenase. CONCLUSIONS: Our study uncovered the laboratory profiles of a large cohort of COVID-19 patients, which formed the basis of discrepancies influenced by aging and biological sex. These profiles directly linked COVID-19 disease presentation to an intricate interplay between sex, age, and immune activation.

Hyptis suaveolens (HS), Hyptis pectinata (HP) and Hyptis marrubioides (HM) are plants used in folk medicine for treatment of several diseases. Here, we tested the in vivo antioxidant and neuroprotective potential of methanolic extracts from these plants, containing several rosmarinic acid derivatives and isoquercetin. In C. elegans, HS, HP and HM leaf extracts enhanced the antioxidant responses through the induction of specific antioxidant enzymes and demonstrated neurotherapeutic potential in transgenic models of genetically determined human neurodegenerative diseases - Frontotemporal dementia with parkinsonism linked to chromosome 17 and Machado-Joseph disease. Chronic treatment of disease models with HS, HP and HM leaf extracts improved the animals' motor function and increased their tolerance to an oxidative insult. The restorative effect of HM extract in motor performance of both disease models required the presence of glutathione reductase (gsr-1), an enzyme that assures the glutathione redox cycle, highlighting the role of this pathway and unveiling a common candidate therapeutic target for these diseases. Our findings strengthen the relevance of plant-derived bioactive compound discovery for neurodegenerative disorders that remain without effective treatment.

The giant otter, Pteronura brasiliensis, (Zimmermann, 1780), is a semiaquatic carnivore that feeds mainly on fish. Mercury has high toxicity and high potential for bioaccumulation in tissues and biomagnification in organisms through food chains. Thus, as a top predator in the trophic chain, the giant otter has the potential to accumulate mercury by biomagnification. The objective of the present study was to measure the total mercury concentration in giant otter fur samples from an area in the southern Brazilian Pantanal. Fur samples from 19 otters from different social groups were collected from captured animals or were sampled with biopsy darts. Total mercury determination was performed by cold steam spectrophotometry. Mercury concentrations found in the giant otter fur were 7.15 ± 3.41 µg g-1 (2.01-12.06 µg g-1) dry weight. The values are above to the upper limit found in fur samples of otter species not exposed to contamination sources, which typically range from 1 to 5 µg g-1. The concentrations found in the study area indicate that even in the southern Pantanal, which is approximately 475 km from gold mining activity, mercury can be considered a threat for giant otters, as well as for riverside populations. It is also important to consider that other anthropogenic sources of mercury, such as pesticides used in agriculture, could affect this population.

ABSTRACT Introduction The rural worker involved in the coffee harvest is an important person for the economy of the many countries. However, scientific studies on the health of this worker are rare. Objective To analyze the lumbar paravertebral musculature and abdominal rectus of rural workers during coffee harvesting with the manual machines with different footrest bases (inclined, declined and horizontal), by means of electromyography. Methods Eight male rural workers from the region of Muzambinho participated in the study. All procedures and electromyographic routines were applied according to the SENIAN protocol. The workers made the usual coffee harvesting movements, using the manual machine for one minute. The data are expressed as means and standard deviations. The RMS (root mean square) signal and the median frequency (FM) were also analyzed. Results It was observed that the type of footrest base did not influence the electromyographic activity RMS or the FM, but we found that the paravertebral muscles have greater electromyographic activation when compared to the abdominal muscles, regardless of the type of footrest base. Conclusion For the task performed, the paravertebral muscles present greater electromyographic activation compared to the abdominal rectus. RESUMO Introdução O trabalhador rural envolvido na colheita de café é um personagem importante para a economia de muitos países. Contudo, são raros os estudos que envolvam a saúde desse trabalhador. Objetivo Analisar através da eletromiografia a musculatura paravertebral lombar e reto abdominal dos trabalhadores rurais durante a colheita de café com a derriçadeira manual, em diferentes bases de apoio para os pés (inclinado, declinado e horizontal). Métodos Participaram do estudo oito trabalhadores rurais da região de Muzambinho, do sexo masculino. Todos os procedimentos e rotinas eletromiográficas foram aplicados de acordo com o protocolo de SENIAN. Os trabalhadores realizaram os movimentos usuais da colheita do café utilizando a derriçadeira manual durante um minuto. Os dados foram expressos em média e desvio padrão com o sinal RMS (root mean square), além disso, foi analisada a frequência mediana (FM). Resultados Observou-se que os planos não influenciaram na atividade eletromiográfica RMS e nem na FM, no entanto, foi constatado que os músculos paravertebrais possuem maiores níveis de ativação eletromiográfica quando comparados aos músculos abdominais, independentemente dos planos. Conclusão Conclui-se que para a tarefa realizada os músculos paravertebrais apresentam maior ativação eletromiográfica em comparação aos retos abdominais. RESUMEN Introducción El trabajador rural involucrado en la cosecha de café es un personaje importante para la economía de muchos países. Sin embargo, son raros los estudios que abarquen la salud de este trabajador. Objetivo Analizar a través de electromiografía la musculatura paravertebral lumbar y recto abdominal de los trabajadores rurales durante la cosecha de café con la cosechadora manual, en diferentes bases de apoyo para los pies (inclinado, declinado y horizontal). Métodos Participaron en el estudio ocho trabajadores rurales de la región de Muzambinho, del sexo masculino. Todos los procedimientos y rutinas electromiográficas fueron aplicadas de acuerdo con el protocolo de SENIAN. Los trabajadores realizaron los movimientos usuales de la cosecha de café utilizando una cosechadora manual durante un minuto. Los datos fueron expresados en promedio y desviación estándar con la señal RMS (root mean square) y también fue analizada la frecuencia mediana (FM). Resultados Se observó que los planos no influenciaron en la actividad electromiográfica RMS y tampoco en la FM, sin embargo, fue constatado que los músculos paravertebrales poseen mayores niveles de activación electromiográfica cuando comparados a los músculos abdominales, independientemente de los planos. Conclusión Se concluye que para la tarea realizada los músculos paravertebrales presentan mayor activación electromiográfica en comparación a los rectos abdominales.
