OBJECTIVES: Considering that γ-terpinene (γ-TPN) is a monoterpene found in Cannabis oil, with high lipophilicity and limited pharmacokinetics, our objective was to evaluate whether its complexation in ß-cyclodextrin (γ-TPN/ß-CD) could improve its physicochemical properties and action on cancer pain, as well as verify the mechanisms of action involved. METHODS: The γ-TPN/ß-CD was prepared and submitted to physicochemical characterization. Animals with sarcoma 180 were treated (vehicle, γ-TPN 50 mg/kg, γ-TPN/ß-CD 5 mg/kg or morphine) and assessed for hyperalgesia, TNF-α and IL-1ß levels, iNOS and c-Fos activity. The effects of γ-TPN on calcium channels were studied by patch-clamp and molecular docking. RESULTS: ß-CD improved the physicochemical properties and prolonged the anti-hyperalgesic effect of γ-TPN. This compound also reduced the levels of IL-1ß, TNF-α and iNOS in the tumour, and c-Fos protein in the spinal cord. In addition, it reduced Ca2+ current, presenting favourable chemical interactions with different voltage-dependent calcium channels. CONCLUSION: These results indicate that the complexation of γ-TPN into ß-CD increases its stability and time effect, reducing spinal neuroactivity and inflammation by blocking calcium channels.

Resumo Fundamento: A (-)-carvona é um monoterpeno encontrado em óleos essenciais com atividade antioxidante e anti-inflamátoria. Objetivos: O objetivo deste estudo foi analisar a propriedade antiarrítmica da (-)-carvona no coração de rato e seus efeitos sobre a sinalização de Ca+2 intracelular. Métodos: Os efeitos da (-)-carvona foram avaliados sobre a contratilidade atrial (0,01 - 4 mM) e ventricular (0,5 mM), e no eletrocardiograma (0,5mM). A fração de encurtamento, a corrente de cálcio do tipo L (ICa,L) e a sinalização de Ca+2 foram medidas no cardiomiócito isolado (0,5 mM). O efeito antiarrítmico foi avaliado no modelo de arritmia induzida por sobrecarga de cálcio (0,5 mM) (n = 5). Um p < 0,05 foi adotado como nível de significância estatística. Resultados: No átrio, a (-)-carvona causou inotropismo negativo de maneira concentração-dependente (EC50 0,44 ± 0,11 mM) e diminuiu o inotropismo positivo induzido pelo CaCl2 (0,1 - 8,0 mM) e BAY K8644 (5 - 500 nM), um agonista de canal de cálcio do tipo L. Em coração isolado, a (-)-carvona (0,5mM) reduziu a contratilidade ventricular em 73% e a frequência cardíaca (em 46%), aumentou o Pri (30,7%, tempo desde o início da onda P até a onda R) e o QTc (9,2%, uma medida de despolarização e repolarização dos ventrículos), sem mudar a duração do complexo QRS. A (-)-carvona diminuiu a fração de encurtamento (61%), a (ICa,L) (79%) e o transiente intracelular de Ca+2 (38%). Além disso, a (-)-carvona apresentou ação antiarrítmica, identificada pela redução do escore de arritmia (85%) e ocorrência de fibrilação ventricular. Conclusão: A (-)-carvona reduz a entrada de Ca+2 através de canais de Ca+2 do tipo L e, assim, diminui a contratilidade cardíaca e o Ca+2 intracelular e apresenta promissora atividade antiarrítmica no coração de ratos.


Abstract Background: (-)-Carvone is a monoterpene found in essential oils with antioxidant and anti-inflammatory activity. Objective: The aim of this paper was to analyze the antiarrhythmic property of (-)-carvone in the rat heart and its effects on the intracellular Ca2+ signaling. Methods: The effects of (-)-carvone were evaluated on the ventricular (0.5 mM) and atrial contractility (0.01 - 4 mM) and on electrocardiogram (0.5 mM). Fractional shortening, L-type calcium current (ICa,L) and Ca2+ signaling were measured in the isolated cardiomyocyte (0.5 mM). Antiarrhythmic effect was evaluated in arrhythmia model induced by calcium overload (0.5 mM) (n = 5). P < 0.05 was used as the significance level. Results: In the atrium, (-)-carvone evoked negative inotropism that was concentration-dependent (EC50 0.44 ± 0.11 mM) and decreased the positive inotropism evoked by CaCl2 (0.1 to 8.0 mM) or BAY K8644 (5 to 500 nM), an agonist of L-type Ca2+ channel. In isolated heart, (-)-carvone (0.5 mM) promoted reduction of ventricular contractility (73%) and heart rate (46%), increased PRi (30.7%, time from the onset of the P wave until the R wave) and QTc (9.2%, a measure of the depolarization and repolarization of the ventricles) without changing the QRS complex duration. (-)-Carvone decreased the fractional shortening (61%), ICa,L (79%) and Ca2+ intracellular transient (38%). Furthermore, (-)-carvone showed antiarrhythmic action, verified by decrease of the arrhythmia score (85%) and occurrence of ventricular fibrillation. Conclusion: (-)-Carvone decreases Ca2+ entry through L-type Ca2+ channels, reducing the cardiac contractility and intracellular Ca2+, and, therefore, presenting promising antiarrhythmic activity in the rat hearts.

Doxorubicin (DOX) is an anticancer chemotherapy drug that is widely used in clinical practice. It is well documented that DOX impairs baroreflex responsiveness and left ventricular function and enhances sympathetic activity, cardiac sympathetic afferent reflexes and oxidative stress, which contribute to hemodynamic deterioration. Because resistance training (RT)-induced cardioprotection has been observed in other animal models, the objective of this study was to assess the effects of RT during DOX treatment on hemodynamics, arterial baroreflex, cardiac autonomic tone, left ventricular function and oxidative stress in rats with DOX-induced cardiotoxicity. Male Wistar rats were submitted to a RT protocol (3 sets of 10 repetitions, 40% of one-repetition maximum (1RM) of intensity, 3 times per week, for 8 weeks). The rats were separated into 3 groups: sedentary control, DOX sedentary (2.5 mg/kg of DOX intraperitoneal injection, once a week, for 6 weeks) and DOX + RT. After training or time control, the animals were anesthetized and 2 catheters were implanted for hemodynamic, arterial baroreflex and cardiac autonomic tone. Another group of animals was used to evaluate left ventricular function. We found that RT in DOX-treated rats decreased diastolic arterial pressure, heart rate, sympathetic tone and oxidative stress. In addition, RT increased arterial baroreflex sensitivity, vagal tone and left ventricular developed pressure in rats with DOX-induced cardiotoxicity. In summary, RT is a useful non-pharmacological strategy to attenuate DOX-induced cardiotoxicity.

INTRODUCTION: Childhood obesity is a significant health problem worldwide, associated with significant metabolic and cardiovascular morbidity. Recent evidence points to metabolic and bariatric surgery as a safe and effective treatment for morbidly obese adolescents. We aim to report the initial results after laparoscopic sleeve gastrectomy (LSG) for adolescent patients in a pediatric center. MATERIAL AND METHODS: Retrospective data review of patients younger than 19 years who underwent LSG for treatment of morbid obesity between 2013 and 2019. RESULTS: A total of 16 adolescents (12 female, 4 male) with a median age of 17.5 years underwent LSG. Median preoperative weight and body mass index were 129kg and 48.6kg/m-2, respectively. All patients had at least one comorbidity. Median follow-up was 18.5 months. The overall percent total weight loss was 32.5% and percent excess weight loss was 68%. Resolution of comorbidities was noted in the majority of patients. No perioperative complications were reported. CONCLUSIONS: Laparoscopic sleeve gastrectomy is a safe and effective option for treatment of morbidly obese adolescents, resulting in significant weight loss and comorbidity resolution with a low risk of complications.

Abstract Background: D-limonene (DL) is a monoterpene and is the major component in the essential oil of citrus fruit. It presents antihyperglycemic and vasodilatation activities. Objectives: This study evaluated the cardiovascular effects and potential antiarrhythmic of DL in rats. Methods: Hemodynamic and electrocardiographic (ECG) parameters were measured in male Wistar rats, which under anesthesia had been cannulated in the abdominal aorta and lower vena cava and had electrodes subcutaneously implanted. In the in vitro approach, the heart was removed and perfused using the Langendorff technique. The significance level adopted was 5% (p < 0.05). Results: DL, in doses of 10, 20, and 40 mg/kg (i.v), produced intense and persistent bradycardia associated with hypotension. Bradycardia with prolonged QTc was observed in the ECG in vivo recording. In the in vivo model of arrhythmia induced by Bay K8644, DL (10 mg/kg) decreased the arrhythmia score from 15.33 ± 3.52 to 4.0 ± 2.64 u.a (p < 0.05, n = 4). In isolated perfused hearts, DL (10-3 M) promoted significant reductions in heart rate (from 228.6 ± 8.5 ms to 196.0 ± 9.3 bpm; p < 0.05) and left ventricular development pressure (from 25.2 ± 3.4 to 5.9 ± 1.8 mmHg; n = 5, p < 0.05). Conclusions: DL produces bradycardia and antiarrhythmic activity in rat heart.


Resumo Fundamento: O D-limoneno (DL) é um monoterpeno e o principal componente do óleo essencial de frutas cítricas. Ele apresenta atividades anti-hiperglicêmicas e vasodilatadoras. Objetivos: Este estudo avaliou os efeitos cardiovasculares e antiarrítmicos potenciais do DL em ratos. Métodos: Os parâmetros hemodinâmicos e eletrocardiográficos (ECG) foram mensurados em ratos Wistar machos que, sob anestesia, tiveram a aorta abdominal e a veia cava inferior canuladas e receberam eletrodos implantados subcutaneamente. Na abordagem in vitro, o coração foi removido e perfundido utilizando a técnica de Langendorff. O nível de significância adotado foi de 5% (p < 0,05). Resultados: DL, nas doses de 10, 20 e 40 mg/kg (i.v), produziu bradicardia intensa e persistente associada à hipotensão. A bradicardia com QTc prolongado foi observada no registro in vivo do ECG. No modelo in vivo de arritmia induzida por Bay K8644, DL (10 mg / kg) houve diminuição do escore da arritmia de 15,33 ± 3,52 para 4,0 ± 2,64 u.a (p < 0,05, n = 4). Em corações perfundidos isolados, o DL (10-3 M) promoveu reduções significativas na frequência cardíaca (de 228,6 ± 8,5 ms para 196,0 ± 9,3 bpm; p < 0,05) e na pressão desenvolvida do ventrículo esquerdo (de 25,2 ± 3,4 para 5,9 ± 1,8 mmHg; n = 5, p < 0,05). Conclusões: O DL produz bradicardia e atividade antiarrítmica no coração de ratos.

BACKGROUND: Oncological pain is one of the most prevalent and difficult-to-treat symptoms in patients with cancer. p-Cymene (PC) is a monoterpene found in more than 100 different plant species, endowed with various pharmacological properties-particularly antinociceptive. HYPOTHESIS/PURPOSE: PC has antinociceptive effect in a model of oncologic pain due to the activation of the descending inhibitory pathway of pain. STUDY DESIGN: A pre-clinical, longitudinal, blind and randomized study. METHODS: Male Swiss mice were induced with S180 cells in the right hind paw, then treated daily with PC (12.5, 25 and 50 mg/kg, s.c.) and screened for mechanical hyperalgesia, spontaneous nociception, nociception induced by non-noxious palpation, tumor growth, changes in the neuromuscular function and existence of bone degradation in the tumor area. The effect of PC on Ca2+ currents (electrophysiological records), histological and neurochemical changes (immunofluorescence for Fos) were also evaluated. RESULTS: PC reduced (p < 0.05) the mechanical hyperalgesia, the spontaneous (p < 0.001) and non-noxious palpation (p < 0.001) nociceptions, not changing the tumor development, neuromuscular function or histopathological aspects of the paw affected. PC reduced Fos expression in the spinal cord (p < 0.001) and increased this expression in the PAG (p < 0.05) and in the NRM (p < 0.01). PC decreased the density of calcium channel currents (p < 0.05). CONCLUSION: These results suggest the antinociceptive effect of PC on oncologic pain, probably acting in both ascending and descending pain pathways, and modulating the calcium channel currents in order to exert its effects.

Myrtenol is a monoterpene with multiple pharmacological activities. However, although monoterpenes have been proposed to play beneficial roles in a variety of cardiac disorders, pharmacological actions of myrtenol in the heart are not yet reported. Hence, the aim of this study was to evaluate whether myrtenol promotes cardioprotection against myocardial ischemia-reperfusion (IR) injury, and the mechanisms involved in these effects. Male Wistar rats were orally treated for seven consecutive days with myrtenol (50 mg/kg) or N-acetyl cysteine (1.200 mg/kg, NAC). Afterward, hearts were subjected to myocardial IR injury. Here, we show that the severe impairment of contractile performance induced by IR was significantly prevented by myrtenol or NAC. Moreover, myrtenol abolished aberrant electrocardiographic waveform (ST-segment elevation), as well as reduced life-threatening arrhythmias and infarct size induced by IR injury. Importantly, myrtenol fully prevented the massive increase of cardiac reactive oxygen species generation and oxidative stress damage. Accordingly, myrtenol restored the impairment of endogenous antioxidant enzymes (superoxide dismutase, catalase, glutathione peroxidase and reductase) activities and balance of pro- and anti-apoptotic pathways (Bax and Bcl-2), associated with decreased apoptotic cells. Taken together, our data show that myrtenol promotes cardioprotection against IR injury through attenuation of oxidative stress and inhibition of pro-apoptotic pathway.

Outbreaks of the Zika, dengue, and chikungunya viruses, especially in the Americas, pose a global threat due to their rapid spread and difficulty controlling the vector. Extreme phenotypes are often observed, from asymptomatic to severe clinical manifestations, which are well-studied in dengue. Host variations are also important contributors to disease outcomes, and many case-control studies have associated single nucleotide polymorphisms (SNPs) with severe dengue. Here, we found that the TC genotype and T-carriers for SNP rs1285933 in the C-type lectin superfamily member 5 (CLEC5A) gene was associated with severe dengue in a Northern Brazilian population (OR=2.75 and p-value=0.01, OR=2.11 and p-value=0.04, respectively). We also tested the functional effect of the CLEC5A protein and found that it is upregulated on the surface of human monocytes after in vitro dengue infection. CLEC5A was correlated with viral load inside the monocytes (Spearman r=0.55, p=0.008) and TNF production in culture supernatants (Spearman r=0.72, p=0.03). Analysis of mRNA in blood samples from DENV4-infected patients exhibiting mild symptoms showed that CLEC5A mRNA expression is correlated with TNF (r=0.67, p=0.0001) and other immune mediators. Monocytes from rs1285933 TT/TC individuals showed lower CLEC5A expression compared to CC genotypes. However, in these cells, CLEC5A was not correlated with TNF production. In summary, we confirmed that CLEC5A is genetically associated with dengue severity outcome, playing a central role during the immune response triggered by a dengue viral infection, and rs1285933 is a relevant SNP that is able to regulate signaling pathways after interactions between the dengue virus and CLEC5A receptors.

The zoonotic potential of multidrug resistant (MDR) bacteria is a worldwide concern and companion animals have been implicated in the spread of resistant bacteria. Therefore, surveillance is important, as there are reports of transmission of these bacteria from dog to men, as well as from men to dog. A 5-year-old mixed-breed male dog was admitted with obstructive struvite urolithiasis relapsing for over 18 months, in Botucatu, in the state of São Paulo, Brazil. The strain, biochemically identified as Staphylococcus spp., was MDR and was treated off-label with vancomycin, which resulted in clinical cure. The strain was molecularly identified as Staphylococcus pseudintermedius and the mecA gene was identified. This is the main gene responsible for methicillin-resistant S. pseudintermedius (MRSP), which is often resistant to multiple antimicrobials. The hypotheses for this clinical case are the transmission from man to animal, since the tutor was an intensivist doctor, or the bacterium itself could be part of the animal's microbiota and due to other factors, such as stress or obstructive urinary disease, opened a doorway to infection by S. pseudintermedius. Further studies should elucidate the transmission of MDR bacteria between human and pets.(AU)


O potencial zoonótico de bactérias multirresistentes é uma preocupação global e os animais de companhia têm sido implicados na disseminação de bactérias resistentes; assim, é importante a vigilância, pois já existem relatos de transmissão destas bactérias do cão para o homem e vice-versa. Um cão, sem raça definida e de cinco anos de idade, foi atendido na cidade de Botucatu, São Paulo, Brasil, apresentando urolitíase obstrutiva de estruvita recorrente há um ano e meio. Na urocultura do animal foi isolada uma estirpe de Staphylococcus spp. multirresistente; o tratamento com vancomicina possibilitou acura clínica. A estirpe de Staphylococcus spp. isolada foi identificada molecularmente como S. pseudintermedius e nela foi identificada a presença do gene mecA, o principal responsável por S. pseuidintermedius resistente à meticilina (MRSP), e que é frequentemente resistente à múltiplos antimicrobianos. As hipóteses para este caso clínico são a transmissão do homem para o animal, pois o tutor era um médico intensivista, ou que a própria bactéria fazia parte da microbiota do animal e, devido a outros fatores como estresse e doença urinária obstrutiva, abriu-se uma porta de entrada para a infecção pelo S. pseudintermedius. Mais estudos são necessários para a elucidação da transmissão de bactérias multirresistentes entre animais de companhia e o ser humano.(AU)

INTRODUCTION: Hypercholesterolemia results from an alteration, genetic or acquired, in lipoprotein metabolism. Evidence that hypercholesterolemia is associated with the atherosclerotic process from childhood justifies the screening of high-risk children and initiation of therapy at preschool ages. OBJECTIVE: To assess children referred for pediatric consultations due to hypercholesterolemia. METHODS: Children and adolescents referred for pediatric consultations with a diagnosis of hypercholesterolemia were enrolled. Information on family history and clinical, anthropometric and biochemical parameters was recorded and, when appropriate, molecular study was performed. RESULTS: A total of 168 children were assessed. Forty-six presented a familial hypercholesterolemia phenotype and in 22 of these, a mutation in the low-density lipoprotein (LDL) receptor gene was identified. The lipid profile of the group with mutations showed significantly higher values of total and non-high-density lipoprotein (HDL) cholesterol compared to the group without mutations (total cholesterol 316.5±75.9 mg/dl vs. 260.9±42,0 mg/dl; non-HDL cholesterol 268.3±72.6 mg/dl vs. 203.5±43.9 mg/dl; p<0.05). Of the total, 55 were prescribed pharmacological therapy and the others underwent diet and exercise interventions only. A greater reduction in LDL cholesterol was observed in individuals under pharmacological therapy compared to those prescribed diet and exercise only (30.3% vs. 18.1%). Drug side effects were insignificant. CONCLUSION: It is possible to maintain a normal lipid profile in most individuals with familial hypercholesterolemia in order to reduce the risk of early onset of atherosclerosis, which is associated with serious cardiovascular complications from childhood.

Avaliaram-se os hábitos e comportamentos pessoais, no que se refere à brucelose, toxoplasmose e leptospirose, relacionando-se com os resultados obtidos no inquérito sorológico realizado, em alunos e residentes da Faculdade de Medicina Veterinária da FMVZ, UNESP-Botucatu-SP. Foram entrevistados 174 alunos da graduação, 21 residentes e 13 pós-graduandos. Os dados dos entrevistados foram trabalhados em programa estatístico (EpiInfo®2002). Paralelamente, coletou-se sangue dos alunos, para a realização da pesquisa de anticorpos para leptospirose, toxoplasmose e brucelose. Verificou-se que o contato frequente com cães mostrou associação estatisticamente significante com a presença de anticorpos anti-Toxoplasma gondii. A correlação dos resultados sorológicos, com as variáveis consideradas como fatores de risco, mostrou que, para a brucelose, somente 20,0% dos reagentes consideraram a urina do cão uma via de eliminação do agente, 40,0% os alimentos crus ou mal cozidos uma via de transmissão, 33,4% e 20,0% a presença de animais no domicílio e o fato de dormir com eles, como risco de infecção. Em relação à toxoplasmose, verificou-se, para algumas situações, o mesmo conhecimento equivocado, pois os alimentos crus ou mal cozidos foram considerados via de transmissão por somente 26,3% dos soropositivos, e 56,2% a manutenção de animais de estimação no domicílio, e o fato de dormir com eles, como fatores de risco. Os resultados obtidos sugerem a conscientização, no que concerne aos aspectos de saúde pública.


The habits and personal behaviors to brucellosis, toxoplasmosis and leptospirosis were evaluated for the relation with the obtained results in the serological inquiry realized in classmates of the Veterinary Medicine course, FMVZ, UNESP-Botucatu-SP. 174 classmates, 21 residents and 13 postgraduating were interviewed. Data of the interviewed persons were analyzed in statistical program (EpiInfo®2002). Paralleling, the blood of all persons was harvested to realize the research of antibodies to leptospirosis, toxoplasmosis and brucellosis. The frequent contact with dogs showed a significant statistically association with the presence of antibodies anti-Toxoplasma gondii. The correlation of the serological results with the variables considered as risk factors, show that to brucellosis, only 20.0% of the reagents considered the urine of dog a potent route of elimination of the agent, 40.0% the raw or undercooked food a route of transmission, 33.4% and 20.0% the presence of animal at home and the fact of sleeping with it, as risk of infection. In relation to toxoplasmosis, the same mistaken knowledge was verified, because the raw or undercooked food were incriminated as route of transmission only for 26.3% positives serologically, and 56.2% the maintenance of pet animals at home, and the fact of to sleeping with it, as risk factor. The obtained results suggest the conscientization in relation to the aspects of public health.


Los hábitos y comportamientos personales fueron evaluados, en lo que se refiere a la brucelosis, toxoplasmosis y leptospirosis, relacionándose con los resultados obtenidos en las pruebas serológicas realizadas, en alumnos y residentes de la Facultad de Medicina Veterinaria FMVZ, UNESP-Botucatu-SP. Fueron entrevistados 174 alumnos de graduación, 21 residentes y 13 postgraduandos. Los datos de los entrevistados fueron estudiados en programa estadístico (EpiInfo®2002). Paralelamente, se colectó sangre de alumnos para la realización de la investigación de anticuerpos para leptospirosis, toxoplasmosis y brucelosis. Se verificó que el contacto frecuente con perros presentó asociación estadísticamente significante con la presencia de anticuerpos anti- Toxoplasma gondii. La correlación de los resultados serológicos, con las variables consideradas como factores de riesgo, evidenció que para la brucelosis, solamente 20,0% de los reactivos consideraron la orina del perro una vía de eliminación del agente, 40,0% los alimentos crudos o mal cocidos, 33,4% una vía de transmisión y 20,0% la presencia de animales en el domicilio y el hecho de dormir con los mismos, como riesgo de infección. En relación a la toxoplasmosis, se verificó, para algunas situaciones, el mismo conocimiento equivocado, pues los alimentos crudos o mal cocidos fueron considerados vía de transmisión por solamente 26,3% de los seropositivos, y 56,2% la manutención de mascotas en el domicilio, y el hecho de dormir con ellos, como factores de riesgo. Los resultados obtenidos sugieren concienciar cuanto a los aspectos de salud pública.

The negative inotropic effect of aqueous fraction (AqF) obtained from the acetic extract of Psidium guajava L leaf was investigated on the guinea pig left atrium. Myocardial force was measured isometrically (27 ± 0.1 ºC, 2 Hz). AqF (100 μg/ml) reduced contractility of about 85 ± 9.4 percent (n = 4, p < 0.001, Fcalc = 51.70, F(0.01; 4; 21) = 5.09, EC50 = 14.28 ± 3 μg/mL) in a concentration-dependent fashion. This effect was reduced by 20 mM of tetraethylammonium (TEA), increasing EC50 to 50 ± 7 μg/ml (n = 4, p < 0.001, Fcalc = 282.13; F(0.01; 21; 66) = 2.36). AqF (100 μg/ml) shifted to the right the CaCl2 concentration-effect curve, increasing the EC50 from 2170 ± 112 to 2690 ± 132 μM (n = 3, p < 0.001, Fcalc = 220.80 ; F(0.01; 29; 60) = 2.19). L-NAME (100 μM) did not modify the AqF inotropic effect (n = 3, p > 0.05) sugesting that the oxide nitric pathway did not participate of the action mechanism of AqF. We can conclude that AqF depresses the atrial contractile by reducing the calcium entry in myocardial cells and also by openenig potassium channels of cardiac tissue.


O efeito inotrópico da fração aquosa (AqF) do extrato acético das folhas de Psidium guajava L. foi investigado em átrio esquerdo de cobaia. A força miocárdica foi medida isometricamente (27 ± 0,1 ºC; 2 Hz). A AqF (100 μg/mL) reduziu a contratilidade em até 85 ± 9,4 por cento (n = 4; p < 0,001; Fcalc = 51,70; F(0,01; 4; 21) = 5,09; CE50 = 14,28 ± 3 μg/mL) de forma dependente da concentração. Este efeito foi reduzido pelo tetraetilamônio (TEA, 20 mM) que também aumentou a CE50 de 14,28 ± 3 μg/mL para 50 ± 7 μg/mL (n = 4; p < 0,001; Fcalc = 282,13; F(0,01; 21; 66) = 2,36). A AqF (100 μg/mL) deslocou para a direita a curva concentração-efeito do CaCl2, aumentando a CE50 de 2170 ± 112 para 2690 ± 132 μM (n = 3; p < 0,001; Fcalc = 220,80 ; F(0,01; 29; 60) = 2,19). Por outro lado, o L-NAME (100 μM) não alterou o efeito inotrópico da AqF (n = 3; p > 0,05), sugerindo que a via do óxido nítrico não participa do mecanismo de ação da AqF. Conclui-se que a AqF deprime a contratilidade atrial por reduzir a entrada de cálcio nas células miocárdicas e por abrir canais de potássio deste tecido.

It was previously showed that aqueous leaf extract (AqEx) of Averrhoa carambola depresses the guinea pig atrial inotropism. Therefore, experiments were carried out on guineapig left atrium and on pituitary GH3 cells in order to evaluate the effect of AqEx on the cellular calcium infl ux. The atrium was mounted in an organ chamber (5 mL, Tyrode, 27 ± 0.1 °C, 95% O2, 5 % CO2), stretched to 10 mN, and paced at 2 Hz (0.5 ms, 400 V) and GH3 cells were submitted to a whole cell voltage clamp confi guration. In the atrium, the AqEx (1500 μg/mL) shifted to the right the concentration-effect curve of the positive inotropic effect produced by (±) BAY K 8644, an L-type calcium channel agonist. The AqEx increased EC50 (concentration required to promote 50% of the maximum effect) of the inotropic effect of BAY K 8644 from 7.8 ± 0.38 to 115.1 ± 0.44 nM (N = 3; p < 0.05). In GH3 cells assayed with 500 μg/mL of AqEx, the L-type calcium inward current declined 30 % (from 282 to 190 pA). Nevertheless, the extract did not change the voltage correspondent to the peak current. These data suggest that, at least in part, the negative inotropic effect of AqEx on the guinea pig atrium is due to a reduction of the L-type calcium current.


Em estudo prévio mostrou-se que o extrato aquoso das folhas de Averrhoacarambola (ExAq) reduziu o inotropismo atrial da cobaia. Por isso, este trabalho avaliou se o ExAq interfere com o infl uxo de cálcio através da membrana celular. A investigação foi conduzidaem átrio esquerdo de cobaia, montado em cuba (5 mL, Tyrode, 27 ± 0,1 °C, 95 % O2, 5 % CO2), estirado para uma tensão de repouso de 10 mN e submetido a uma estimulação de 2 Hz (0,5 ms, 400 V). O efeito do ExAq sobre a entrada de cálcio nas células foi avaliado em átrio de cobaia e em células GH3, estas submetidas a ‘patch clamp’ na confi guração ‘whole cell’. No átrio, o ExAq (1500 μg /mL) deslocou para direita a curva concentração-efeito do (±) BAY K 8644 (agonista dos canais de cálcio tipo-L), aumentando a CE50 (concentração capaz de produzir 50 % do efeito máximo) de 7,8 ± 0,38 para 115,1 ± 0,44 nM (N = 3, p < 0,05). Em células GH3, este extrato (500 μg /mL) reduziu de 282 para 190 pA (30 %) a corrente de cálcio, sem contudo alterar a voltagem de pico da curva desta corrente. Estes resultados mostram que, pelo menos em parte, o efeito inotrópico negativo do ExAq em átrio de cobaia se deve a uma diminuição do infl uxo de cálcio pelos canais tipo-L.
