Resumen El aumento de la resistencia a los antibióticos por parte de bacterias patógenas ha motivado la búsqueda de alternativas para disminuir su utilización. Dentro de las opciones propuestas se encuentra la terapia de antiadherencia, en la cual se utilizan moléculas análogas a los glicoepítopes que son reconocidos por las bacterias para impedir la unión de éstas al tejido celular. En este estudio se llevó a cabo la obtención de glicoconjugados por medio de la reacción de Maillard partiendo de albúmina sérica bovina (BSA) y oligosacáridos de quitosano (oligosacáridos sin ultrafiltrar, ultrafiltrados y ultrafiltrados acetilados), en proporción 1:1 (p/p) a tres temperaturas de incubación (50, 60 y 70 °C) por 30 min. La caracterización de los conjugados sintetizados se realizó utilizando electroforesis (SDS-PAGE), espectroscopía de infrarrojo y espectroscopía de fluorescencia. Se realizaron ensayos de reconocimiento usando aglutinina de germen de trigo (WGA) y bacterias [Escherichia coli (K88ac y K88+)]. La caracterización por medio de electroforesis y espectroscopía de infrarrojo evidenció la unión de los oligosacáridos de quitosano a la estructura de la BSA. Además, los ensayos evidenciaron el reconocimiento de las moléculas sintetizadas tanto por la lectina WGA como por las bacterias. Los glicoconjugados sintetizados sin ultrafiltrar ni acetilar mostraron resultados muy favorables en el reconocimiento por ambas bacterias, lo que constituye una ventaja práctica, ya que su implementación a mayor escala reduciría costos de producción


Abstract The increase in antibiotic resistance by pathogenic bacteria has motivated the search for alternatives to reduce the use of antibiotics. Among these alternatives is anti-adhesion therapy, in which molecules that mimic the glycoepitopes that recognise bacteria are used to prevent their binding to cellular tissue. In this study, glycoconjugates were obtained by means of the Maillard reaction starting from bovine serum albumin (BSA) and chitosan oligosaccharides (unfiltered oligosaccharides, ultrafiltered and acetylated ultrafiltered), in a ratio of 1:1 (w/w) at three incubation temperatures (50, 60 and 70 °C) per 30 min. The characterisation was performed using the techniques of electrophoresis (SDS-PAGE), infrared spectroscopy and fluorescence spectroscopy. Recognition assays were performed using wheat germ agglutinin (WGA) and Escherichia coli bacteria (K88ac and K88+). Characterisation by electrophoresis and infrared spectroscopy demonstrated the binding of chitosan oligosaccharides to the structure of BSA. In addition, the tests showed the recognition of the molecules synthesised by both the WGA lectin and the E. coli bacteria. The glycoconjugates synthesised without ultrafiltration or acetylation showed very favourable results in recognition with both bacteria, which is an advantage, since its implementation on a larger scale would reduce production costs.


Resumo O aumento da resistência aos antibióticos por bactérias patogênicas tem motivado a busca de alterna­tivas para reduzir seu uso. Entre essas alternativas está a terapia anti-adesão, na qual são utilizadas moléculas análogas aos glicoepítopos que são reconhecidas pelas bactérias para impedir sua união ao tecido celular. Neste estudo, os glicoconjugados foram obtidos por meio da reação de Maillard a partir de albumina sérica bovina (BSA) e oligossacarídeos de quitosana (oligossacarídeos não ultra­filtrados, ultrafiltrados e acetilados ultrafiltrados), na proporção de 1:1 (p/p) em três temperaturas de incubação (50, 60 e 70 °C) durante 30 min. A caracterização dos conjugados sintetizados foi realizada utilizando a eletroforese (SDS-PAGE), espectroscopia de infravermelho e espectroscopia de fluorescência. Os ensaios de reconhecimento foram realizados utilizando aglutinina de germe de trigo (WGA) e bactérias [Escherichia coli (K88ac e K88+)]. A caracterização por meio de eletroforese e espectroscopia de infravermelho demonstrou a união dos oligossacarídeos de quitosana à estrutura da BSA. Além disso, os testes evidenciaram o reconhecimento das moléculas sintetizadas tanto pela lectina WGA quanto pelas bactérias. Os glicoconjugados sintetizados sem ultrafiltração ou acetilação apresentaram resultados muito favoráveis no reconhecimento por ambas as bactérias, o que é uma vantagem, visto que sua implementação em maior escala reduziria custos de produção.

Acute monocytic leukemia is a type of myeloid leukemia that develops in monocytes. The current clinical therapies for leukemia are unsatisfactory due to their side effects and nonspecificity toward target cells. Some lectins display antitumor activity and may specifically recognize cancer cells by binding to carbohydrate structures on their surface. Therefore, this study evaluated the response of the human monocytic leukemia cell lines THP-1 to the Olneya tesota PF2 lectin. The induction of apoptosis and reactive oxygen species production in PF2-treated cells was evaluated by flow cytometry, and the lectin-THP-1 cell interaction and mitochondrial membrane potential were evaluated by confocal fluorescence microscopy. PF2 genotoxicity was evaluated by DNA fragmentation analysis via gel electrophoresis. The results showed that PF2 binds to THP-1 cells, triggers apoptosis and DNA degradation, changes the mitochondrial membrane potential, and increases reactive oxygen species levels in PF2-treated THP-1 cells. These results suggest the potential use of PF2 for developing alternative anticancer treatments with enhanced specificity.

Nanomedicine has led to the development of new biocompatible and biodegradable materials able to improve the pharmaceutical effect of bioactive components, broadening the options of treatment for several diseases, including cancer. Additionally, some snake venom toxins have been reported to present cytotoxic activity in different tumor cell lines, making them an auspicious option to be used as cancer drugs. The present study aims to evaluate the cytotoxic activity of the northern black-tailed rattlesnake (Crotalus molossus molossus) venom-loaded chitosan nanoparticles (Cs-Venom NPs) against the T-47D breast carcinoma cell line. To do so, we first identified the significant proteins composing the venom; afterward, hemocompatibility and cytotoxic activity against tumoral cells were evaluated. The venom was then loaded into chitosan nanoparticles through the ionotropic gelation process, obtaining particles of 415.9±21.67 nm and ζ-potential of +28.3±1.17 mV. The Cs-Venom complex delivered the venom into the breast carcinoma cells, inhibiting their viability and inducing morphological changes in the T-47D cells. These features indicate that these nanoparticles are suitable for the potential use of C. m. molossus venom toxins entrapped within polymer nanoparticles for the future development and research of cancer drugs.

Doxorubicin (Dox) is the most widely used chemotherapeutic agent and is considered a highly powerful and broad-spectrum for cancer treatment. However, its application is compromised by the cumulative side effect of dose-dependent cardiotoxicity. Because of this, targeted drug delivery systems (DDS) are currently being explored in an attempt to reduce Dox systemic side-effects. In this study, DDS targeting hepatocellular carcinoma (HCC) has been designed, specifically to the asialoglycoprotein receptor (ASGPR). Dox-loaded albumin-albumin/lactosylated (core-shell) nanoparticles (tBSA/BSALac NPs) with low (LC) and high (HC) crosslink using glutaraldehyde were synthesized. Nanoparticles presented spherical shapes with a size distribution of 257 ± 14 nm and 254 ± 14 nm, as well as an estimated surface charge of -28.0 ± 0.1 mV and -26.0 ± 0.2 mV, respectively. The encapsulation efficiency of Dox for the two types of nanoparticles was higher than 80%. The in vitro drug release results showed a sustained and controlled release profile. Additionally, the nanoparticles were revealed to be biocompatible with red blood cells (RBCs) and human liver cancer cells (HepG2 cells). In cytotoxicity assays, Dox-loaded nanoparticles decrease cell viability more efficiently than free Dox. Specific biorecognition assays confirmed the interaction between nanoparticles and HepG2 cells, especially with ASGPRs. Both types of nanoparticles may be possible DDS specifically targeting HCC, thus reducing side effects, mainly cardiotoxicity. Therefore, improving the quality of life from patients during chemotherapy.

The aim of this study was to determine the Cd2+ removal capacity of a biosorbent system formed by Saccharomyces cerevisiae in calcium alginate beads. The adsorption of Cd2+ by a S. cerevisiae-alginate system was tested either by batch or fixed-bed column experiments. The S. cerevisiae-alginate system was characterized using dynamic light scattering (DLS, zeta potential), size, hardness, scanning electron microscopy (SEM), and Fourier-transform infrared spectroscopy. Beads of the S. cerevisiae-alginate system showed a spherical-elliptical morphology, diameter of 1.62 ± 0.02 mm, 96% moisture, negative surface charge (-29.3 ± 2.57 mV), and texture stability during storage at 4 °C for 20 days. In batch conditions, the system adsorbed 4.3 µg of Cd2+/g of yeast-alginate beads, using a Cd2+ initial concentration of 5 mg/L. Adsorption capacity increased to 15.4 µg/g in a fixed-bed column system, removing 83% of total Cd2+. In conclusion, the yeast-alginate system is an efficient option for the removal of cadmium at low concentrations in drinking water.

Hepatocellular carcinoma (HCC) ranks fifth in occurrence and second in mortality of all cancers. The development of effective therapies for HCC is urgently needed. Anticancer drugs targeted to the liver-specific asialoglycoprotein receptors (ASGPRs) are viewed as a promising potential treatment for HCC. ASGPRs facilitate the recognition and endocytosis of molecules, and possibly vehicles with galactose end groups, by the liver. In this study, bovine serum albumin (BSA) was conjugated with lactose using a thermal treatment. The formation of lactosylated BSA (BSA-Lac) was confirmed by a change of the chemical structure, increased molecular mass, and Ricinus communis lectin recognition. Subsequently, the low-crosslinking BSA-Lac nanoparticles (LC BSA-Lac NPs) and high-crosslinking BSA-Lac nanoparticles (HC BSA-Lac NPs) were synthesized. These nanoparticles presented spherical shapes with a size distribution of 560 ± 18.0 nm and 539 ± 9.0 nm, as well as an estimated surface charge of -26 ± 0.15 mV and -24 ± 0.45 mV, respectively. Both BSA-Lac NPs were selectively recognized by ASGPRs as shown by biorecognition, competition, and inhibition assays using an in vitro model of HCC. This justifies pursuing the strategy of using BSA-Lac NPs as potential drug nanovehicles with selective direction toward hepatocellular carcinoma.

Herein, silica nanoparticles were synthesized and chemically modified with iminodiacetic acid (IDA) and Ni2+ ions surrounded by a bis-acrylamide polymeric shell to obtain a new core-shell immobilized metal affinity chromatography (IMAC) based material. These Ni2+-IDA-core-shell silica nanoparticles (Ni2+-IDA-CSS-NP) represent a new alternative for purification of His-tagged proteins and exclusion of high molecular weight (HMW) proteins at the same time. Nanoparticles presented a final size of 479.6 ± 6.9 nm determined by dynamic light scattering (DLS) and a surface charge of -37.2 ± 0.5 mV. Successful incorporation of the different compounds at every phase of synthesis was evidenced by ATR-FTIR analysis. Ni2+-IDA-CSS-NP were used for isolation of His-tagged spo0F (6His-spo0F) from E. coli lysate. Ni2+-IDA-CSS-NP presented a capacity of 4.16 ± 0.45 µg mg-1. Purification of 6His-spo0F with high selectivity and the effective exclusion of HMW proteins were evidenced by SDS-PAGE and validated through mass spectrometry analysis.

Adhesion of enterotoxigenic (ETEC) E. coli to host intestinal cells is mediated by lectin-like fimbriae that bind to specific glycan moieties on the surfaces of enterocytes. To prevent in vitro binding of E. coli F4 fimbriae (F4 ETEC+) to piglet enterocytes, neoglycans were synthesized by the Maillard reaction conjugating lactose (Lac), galacto-oligosaccharides (GOS) or chitin oligosaccharides (Ochit) to porcine serum albumin (PSA). Neoglycans were characterized by SDS-PAGE, intrinsic tryptophan fluorescence and recognition by plant lectins, as well as by F4 ETEC variants. Electrophoretic patterns suggested the binding to PSA of 63, 13 and 2 molecules of Lac, GOS and Ochit, respectively. All neoglycans displayed quenching of tryptophan fluorescence consistent with the degree of glycation estimated by SDS-PAGE. Plant lectins recognized the neoglycans according to their specificity, whereas antigenic variants of F4 ETEC (ab, ac and ad) recognized PSA-Ochit and PSA-Lac with higher affinity than that for GOS. Neoglycans partially hindered the in vitro binding of F4+ ETEC to piglet enterocytes in a dose-dependent manner. The most effective blocking was observed with PSA-Lac that partially inhibited the adhesion of bacteria to enterocytes in a dose dependent manner, as quantified by flow cytometry. Increased production of the cytokines IL-6 and TNF-α was observed in response to F4+ ETEC infection of enterocytes and production was reduced in the presence of PSA-Ochit and PSA-GOS. These results suggest that neoglycans synthesized by the Maillard reaction could be useful in the prophylaxis of diarrhea in piglets.

Silica nanoparticles were functionalized with immobilized molecular bait, Cibacron Blue, and a porous polymeric bis-acrylamide shell. These nanoparticles represent a new alternative to capture low molecular weight (LMW) proteins/peptides, that might be potential biomarkers. Functionalized core-shell silica nanoparticles (FCSNP) presented a size distribution of 243.9 ± 11.6 nm and an estimated surface charge of -38.1 ± 0.9 mV. The successful attachment of compounds at every stage of synthesis was evidenced by ATR-FTIR. The capture of model peptides was determined by mass spectrometry, indicating that only the peptide with a long sequence of hydrophobic amino acids (alpha zein 34-mer) interacted with the molecular bait. FCSNP excluded the high molecular weight protein (HMW), BSA, and captured LMW proteins (myoglobin and aprotinin), as evidenced by SDS-PAGE. Functionalization of nanoparticles with Cibacron Blue was crucial to capture these molecules. FCSNP were stable after twelve months of storage and maintained a capacity of 3.1-3.4 µg/mg.

RESUMO

Lectins are a diverse class of proteins distributed extensively in nature. Among these proteins; legume lectins display a variety of interesting features including antimicrobial; insecticidal and antitumor activities. Because lectins recognize and bind to specific glycoconjugates present on the surface of cells and intracellular structures; they can serve as potential target molecules for developing practical applications in the fields of food; agriculture; health and pharmaceutical research. This review presents the current knowledge of the main structural characteristics of legume lectins and the relationship of structure to the exhibited specificities; provides an overview of their particular antimicrobial; insecticidal and antitumor biological activities and describes possible applications based on the pattern of recognized glyco-targets.

Long-beaked common dolphins (Delphinus capensis) in the Gulf of California have been exposed to persistent contaminants that originated in large agricultural areas near the coast. Live common dolphins were sampled by remote dart biopsies to determine concentrations of tDDT in blubber. Life stage and initial gender identification was determined by field observations. Gender was confirmed by genetic analysis of the skin. Concentration of tDDT in blubber was analyzed by gas chromatography. The 16 samples collected consisted of: 2 adult males, 6 adult females, and 8 juveniles. 4,4'-DDE was detected in most of the samples with 4,4'-DDD and 4,4'-DDT under detection levels. Concentrations of DDE varied from non-detectable to 87.3 µg/g lipid weight with a median of 16 µg/g lipid weight. The highest concentration was detected in an immature female. No differences were detected between gender or life stage but this could be attributed to small sample size. We recommend continued sampling of D. capensis blubber biopsies from the Gulf of California in order to relate these levels with affected in vitro biomarkers such as mixed function oxidase activity.

Enterotoxigenic (ETEC) Escherichia coli (E. coli) causes traveller's diarrhoea and high mortality among baby animals. ETEC adhesion is mediated by lectins (adhesins) that bind to glycoconjugates on the surface of host cells. Glycans that compete for adhesion could be used for disease prevention. Neoglycans of porcine albumin (PSA) that were conjugated with prebiotic galactooligosaccharides (GOS) were synthesised using the Maillard reaction. PSA glycation was confirmed by a reduction in the number of available free amino groups, decreased tryptophan intrinsic fluorescence, increased molecular mass and Ricinus communis lectin recognition. The adhesion of four ETEC strains (E. coli H10407, CFA(+), K99 and K88) to PSA-GOS was examined by an enzyme-linked lectin assay. E. coli K88 bound to PSA-GOS with greater affinity (P<0.05) than did E. coli H10407, CFA(+) and K99. In addition, PSA-GOS partially inhibited the adherence of the K88 strain to intestinal mucins. Pig ETEC strain was unable to ferment galactooligosaccharide-neoglycans. These results suggest that neoglycans obtained by the Maillard reaction may serve in the prophylaxis of ETEC K88 diarrhoea.

Recently, glyco-therapy is proposed to prevent the interaction of bacterial lectins with host ligands (glycoconjugates). This interaction represents the first step in infection. Neoglycans referred to as PSA-Lac (PSA-Glu (ß1-4) Gal) were obtained by conjugation of porcine serum albumin (PSA) with lactose at 80 °C, 100 °C and 120 ºC. Characterization studies of the products showed that PSA could contain 1, 38 or 41 added lactoses, depending on the reaction temperature. These neoglycans were approximately 10 times more glycated than PSA-Lac obtained in previous work. Lactose conjugation occurred only at lysines and PSA-Lac contained terminal galactoses as confirmed by Ricinus communis lectin recognition. Furthermore, Escherichia coli K88+, K88ab, K88ac and K88ad adhesins showed affinity toward all PSA-Lac neoglycans, and the most effective was the PSA-Lac obtained after 100 ºC treatment. In vitro, this neoglycan partially inhibited the adhesion of E. coli K88+ to piglet mucin (its natural ligand). These results provide support for the hypothesis that glycated proteins can be used as an alternative for bioactive compounds for disease prevention.

Se comparó el efecto antibacteriano de las lactoferrinas (Lfs) bovina y porcina sobre Escherichia coli K88+ (E. coli K88+), uno de los principales agentes etiológicos de las diarreas en lechones en el hemisferio norte. Las Lfs se purificaron por cromatografía de intercambio iónico, confirmando su pureza por electroforesis en condiciones desnaturalizantes y reductoras (SDS-PAGE) en geles al 8% y por inmuno-detección con anticuerpos anti- lactoferrina. La actividad bacteriostática se probó utilizando concentraciones de 0,5 y 1,0 mg/mL de Holo (saturada de hierro) y Apo-Lf (libre de hierro). En todos los casos la actividad bacteriostática de las Holo-Lfs fue insignificante, mientras que en ambas concentraciones, la Apo-Lf bovina mostró un mayor efecto en la inhibición del crecimiento de la E. coli K88+ que la Apo-Lf porcina. La actividad bactericida se ensayó utilizando concentraciones de 2,0; 4,0; 6,0 y 8,0 mg/mL de Lfs bovina o porcina. La Lf bovina mostró efecto bactericida a una concentración de 8 mg/mL, mientras que la Lf porcina no presentó este efecto. Los resultados indican que la fuente bovina puede ser útil en la prevención de diarreas en lechones.


The antibacterial effect of bovine lactoferrin (Lf) towards Escherichia coli K88+ (E. coli K88+) was compared with that of porcine Lf. E. coli K88+ is one of the main etiological agents of piglet diarrhea in the northern hemisphere. Lactoferrins (Lfs) were purified by ion exchange chromatography and further analyzed by electrophoresis using 8% sodium dodecyl sulfate polyacrylamide gel (SDS-PAGE) and immuno detection with anti-lactoferrin antibodies. Bacteriostatic effect was assayed using 0.5 and 1.0 mg/mL of Holo-Lf (iron saturated) or Apo-Lf (iron free). In all test the holo-LFs showed negligible bacteriostatic activity while for Apo-Lfs the bovine protein had the highest activity. Bactericide activity was assayed using bovine or porcine Lf concentrations of 2.0, 4.0, 6.0 y 8.0 mg/mL. Bovine Lf had bactericide activity at 8.0 mg/mL while no growth inhibition was observed with porcine Lf. These results suggest that bovine Lf may serve in the prophylaxis of piglets’ diarrhea.

Se analizó el efecto de la sustitución de clara de huevo por albúmina sérica porcina (ASP) en panqués de chocolate. La ASP se obtuvo mediante un método escalado de aislamiento por cromatografía de interacción hidrofóbica. En la formulación del panqué se reemplazó el 50 y 100% de la clara de huevo con ASP. Todos los panqués presentaron valores similares (P >0.05) de los parámetros de color en la miga: L (25,7-26,2), a* (9,8-10,1) y b* (14,5-15,0) y en la costra: L (25,7-26,2), a* (9,8-10,1) y b* (14,5-15,0). La textura (2,9 N) y el volumen (148,9 ± 1,8 cm ³) de los panqués con 50% de ASP fueron similares (P> 0,05) a los de los controles. El análisis sensorial indicó que los panqués en los que se reemplazó 50% de la clara por ASP, gustaron tanto como los controles. Los panqués con un reemplazo del 100%, gustaron menos. La excelente calidad microbiológica de los panqués muestra las óptimas condiciones sanitarias durante la obtención de la ASP y su elaboración.


The effect of porcine serum albumin (PSA) as a substitute for egg white (EW) in chocolate cakes was examined. PSA was obtained by a lab-scaled method of Hydrophobic Interaction Chromatography. 50 and 100% of the normal level of EW was replaced with PSA in cake formulation. All cakes had similar (P > 0.05) crumb L (25.7-26.2), a* (9.8-10.1) y b* (14.5-15.0) and crust: L (25.7-26.2), a* (9.8-10.1) y b* (14.5-15.0) color values. Texture (2.9 N) and volume (148.9 1.8 cm ³) of cakes with 50% PSA replacing EW were similar (P > 0.05) to those of the controls. Sensory analysis indicated that cakes replaced with 50% EW for ASP were as well liked as control cakes. The excellent microbiological quality of formulated cakes points out the optimal sanitary conditions in the PSA isolation and in the cake elaboration process.

Las enfermedades gastrointestinales siguen siendo un problema de salud pública mundial. El avance de la ciencia muestra que cambios en el balance adecuado de la microflora intestinal (MI) juegan un papel crucial en la patogénesis. La evidencia apunta a que una manera de modular esta MI es a través del uso de oligosacáridos prebióticos, que estimulan el crecimiento de bacterias benéficas y que a la vez aumentan la resistencia a la invasión por patógenos. Estudios con animales indican que el consumo de carbohidratos prebióticos podría estar implicado en la prevención y tratamiento de diarreas. En infantes humanos sanos, los estudios revelan que el consumo de mezclas de prebióticas (galactooligosacáridos/fructooligosacáridos, inulina/galactooligosacáridos) disminuyen la incidencia de fiebre, de infecciones y de patógenos gastrointestinales. Lo anterior representa un gran potencial para los alimentos funcionales que los contienen, principalmente las fórmulas infantiles. Sin embargo, los estudios de prevención de diarreas mediante el suministro de prebióticos en personas con una microflora intestinal alterada no son concluyentes, sobre todo aquellos practicados en ancianos, personas con problemas crónicos de inflamación intestinal y personas con diarreas asociadas a la toma de antibióticos. Lo anterior nos indica la necesidad de estudios bioquímicos y microbiológicos más profundos en humanos de diferentes edades y condiciones de salud intestinal, a fin de determinar en que condiciones, los prebióticos tienen algún efecto sobre las infecciones.


Gastrointestinal disorders are still a main world public health problem. Scientific progress shows that and inadequate balance in intestinal microbiota (IM) plays a crucial role in its pathogenesis. Evidence indicates that one way to modulate the IM is through the use of prebiotics. These oligosaccharides stimulate the growth of benefic bacteria and increase the resistance to invading pathogens. Research using animals show that the consumption of prebiotics could be implicated in prevention and treatment of diarrhea. Studies in healthy infants also indicate that the consumption of prebiotic mixtures (galactooligosaccharides/ fructooligosaccharides, inulin/ galactooligosaccharides) decreases the incidence of fever, infections and pathogens. These results represent a great potential for functional foods that contain prebiotics, mainly the infant formulas. However, results of other clinical studies for prebiotics effects on diarrhea are not conclusive. Specially those studies that include patients with an altered IM (like the elderly), patients with chronic intestinal inflammation and with diarrhea associated to antibiotic treatments. There is a need for more biochemical and microbiological studies in humans at different ages and intestinal health conditions, in order to determine when prebiotics may effectively function on infections.

Escherichia coli (E. coli) that expresses galactose-reactive lectins, like K88 adhesin, causes high mortality among piglets. Carbohydrates that compete for adhesion could serve as an alternative for disease prevention. Porcine serum albumin (PSA) was modified by non-enzymatic glycation with lactose to produce PSA-Lac or PSA-Glc beta (1-4) Gal, as confirmed by reduction of available free amino groups, increased molecular mass and by Ricinus communis lectin recognition. E. coli K88 binds to PSA-Lac treatments containing three and four lactoses, respectively. In addition, PSA-Lac partially inhibited K88 strain adherence to mucins. These results suggest that neoglycoconjugates obtained by non-enzymatic glycation of proteins may serve in the prophylaxis of piglets' diarrhea.

In this study, we have analyzed the chemical composition and antiproliferative activity of propolis from three different arid and semiarid regions of Sonora, Mexico. We identified and quantitated the main chemical constituents of propolis by HPLC-MS. The most abundant constituents of propolis were pinocembrin, pinobanksin 3-acetate, and chrysin. Sonoran propolis had a strong antiproliferative activity on both murine and human cancer cell lines in a concentration-dependent manner. The propolis constituents CAPE, galangin, xanthomicrol and chrysin showed significant antiproliferative activity on most of the cancer cells tested. DNA harvested from cancer cell cultures treated with Sonoran propolis exhibited a ladder of internucleosomal DNA cleavage characteristic of apoptosis. In summary, we have identified and quantitated the main constituents of Sonoran propolis. These propolis samples possess a strong antiproliferative activity on cancer cell lines.

Iron is an essential element of living cells and organisms as a component of numerous metabolic pathways. Hemoglobin and ferric-transferrin in vertebrate host blood are the two major iron sources for female mosquitoes. We used inductively coupled plasma mass spectrometry (ICP-MS) and radioisotope labeling to quantify the fate of iron supplied from hemoglobin or as transferrin in Aedes aegypti. At the end of the first gonotrophic cycle, approximately 87% of the ingested total meal heme iron was excreted, while 7% was distributed into the eggs and 6% was stored in different tissues. In contrast, approximately 8% of the iron provided as transferrin was excreted and of that absorbed, 77% was allocated to the eggs and 15% distributed in the tissues. Further analyses indicate that of the iron supplied in a blood meal, approximately 7% appears in the eggs and of this iron 98% is from hemoglobin and 2% from ferric-transferrin. Whereas, of iron from a blood meal retained in body of the female, approximately 97% is from heme and <1% is from transferrin. Evaluation of iron-binding proteins in hemolymph and egg following intake of (59)Fe-transferrin revealed that ferritin is iron loaded in these animals, and indicate that this protein plays a critical role in meal iron transport and iron storage in eggs in A. aegypti.

Se analizó suero obtenido de 14 muestreos de sangre de cerdo (10 L) cada uno, obtenido en la línea de sacrificio de una sala de matanza Tipo Inspección Federal (TIF) de Hermosillo, México, con sistema HACCP (Análisis de Riesgos, Identificación y Control de puntos Críticos) implementado. El análisis proximal del suero liofilizado fue: humedad 2,3 g por ciento, proteína 84,3 g por ciento, grasa 4,0 g por ciento y cenizas 4,6 g por ciento. El contenido graso fue mayor (P<0,05) en las muestras de invierno que en las de los meses cálidos, mientras que el contenido proteico no varió. Las concentraciones de IgA, IgG e IgM en suero fresco y liofilizado corresponden a los extremos más altos de los niveles reportados, sin presentarse diferencia (P<0,05) entre los dos tratamientos. La cuenta de mesofílicos aerobios fue < 10UFC/g, los organismos coliformes < 3 NMP/g y no se observó la presencia de Salmonella spp ni de Staphylococcus aureus en ninguna de las muestras. La calidad del suero indica el éxito del Sistema HACCP implementado en la planta y la potencialidad alimentaria del suero porcino
