BACKGROUND & AIMS: The present study evaluated the lipid profile of enteral nutrition formulas with added fish oil used in a public hospital, with an emphasis on the fatty acid (FA) composition. METHODS: FA composition was determined using gas chromatography coupled to a flame ionization detector (GC-FID). Nine enteral nutrition formulas were evaluated and the results obtained were compared with those reported on the formulas labels. RESULTS: The sample with the highest percentage of added fish oil according to the label information had the lowest total amounts of eicosapentaenoic acid and docosahexaenoic acid (EPA + DHA) (p < 0.05). In the evaluation of the total amount of polyunsaturated fatty acids/saturated fatty acids (ΣPUFA/ΣSFA) ratio, five samples were not within the values recommended by Brazilan and international health regulatory agencies. Regarding the n-6/n-3 fatty acids ratio, five samples showed values higher than the recommended ratio. It was observed that EPA + DHA content was positively correlated with the cost of the diet. Importantly, we also found that there was a significant difference between the results of our analysis and the descriptions found on the labels for fatty acids n-6, n-3, EPA, DHA, SFA, PUFA and MUFA. CONCLUSIONS: We conclude that the discrepancies for fatty acids between the values obtained in the analyses and the values reported on the labels highlight the need for more rigorous inspection when public hospitals purchase enteral nutrition formulas with added fish oil, since the administration of formulas with inadequate levels of FA in hospitalized patients can compromise clinical results during the hospitalization period.

This study evaluated the antibacterial activity of cinnamaldehyde (CIN) and biogenic silver nanoparticles (BioAgNP), alone and in combination, against Escherichia coli, Salmonella Typhimurium, and Staphylococcus aureus in vitro. Their sanitation activities on fresh sweet grape tomatoes were also evaluated. CIN and BioAgNP inhibited the growth of the tested bacteria, and at low concentrations, their combinations presented a synergistic effect. In the sanitization of fresh sweet grape tomatoes, CIN (156 µg/mL) combined with BioAgNP (31.25 µM) at subinhibitory concentrations inhibited the growth of E. coli after only 5 min of contact. Exposed samples showed no growth of E. coli during their shelf life. The combination of these compounds did not change significantly (p > 0.05) the physicochemical properties of sweet grape tomatoes and showed that CIN combined with BioAgNP could represent an effective method for decontaminating fruits and vegetables. This combination has great potential for application in the prevention of foodborne diseases.

Essential oils (EOs) have strong antibacterial properties and can be potential sanitizers to reduce pathogen load and prevent cross-contamination during postharvest washing. The objective of this study was to investigate the efficacy of emulsions containing oregano (OR; Origanum vulgare) and winter savory (WS; Satureja montana) EOs at different concentrations (0.94 and 1.88 µL/mL) and storage times (0 h, 24 h, and 7 days), in reducing Escherichia coli O157:H7 on the surface of three types of lettuce (romaine, crisphead, and butterhead). The EO emulsions were compared with one no-rinse treatment and three rinse treatments using water, 200 ppm chlorine, and 80 ppm peroxyacetic acid (PAA), respectively, in a simulated washing system. The results showed that while the EO emulsions significantly reduced E. coli O157:H7 on crisphead lettuce over time, not all treatments were effective for romaine and butterhead lettuce. The mixture of OR and WS at concentrations of 0.94 and 1.88 µL/mL was found to be the most effective in reducing E. coli O157:H7 on inoculated lettuce, resulting in reductions of 3.52 and 3.41 log CFU/g, respectively. Furthermore, the PAA and the mixture of OR and WS at 1.88 µL/mL effectively limited bacterial cross-contamination close to the detection limit for all lettuce types during all storage times. These results suggest that OR and WS EOs could serve as potential alternatives to chemical sanitizers for postharvest lettuce washing.

We evaluated whether linseed oil (LO) modulates the effects of a high-carbohydrate diet (HCD) on liver inflammation, fatty acid (FA) accumulation, and lipid distribution in periportal and perivenous hepatocytes. The control group (control high-carbohydrate diet [HCD-C]) received an HCD with lard and soybean oil as the lipid source. The L10 and L100 groups received the HCD with 10% and 100% of LO as the lipid source, respectively. The animals were killed by decapitation before (day 0) and after receiving the diets. Liver FA composition, inflammation, and fibrogenesis gene expression were evaluated. Also, the percentage of lipid-occupied area in periportal end perivenous hepatocytes were measured. The L100 group exhibited a higher (P < .05) liver amount of omega-3 polyunsaturated FA (n-3 PUFA) and lower (P < .05) amounts of saturated FA (SFA), monounsaturated FA (MUFA), and omega-6 polyunsaturated FA (n-6 PUFA) compared with L10 or HCD-C mice. On day 56, interleukin 10 and type IV collagen gene expression were significantly upregulated and downregulated, respectively in L100. Also, the L100 group showed lower (P < .05) FA accumulation (i.e., total FA, SFA, MUFA, and n-6 PUFA). Also, L10 and L100 presented lower (P < .05) percentage of high lipid-containing portion in periportal and perivenous hepatocytes. We concluded that LO attenuation of liver inflammation promoted by an HCD is associated with increased liver n-3 PUFA levels, so modulating FA composition, deposition, and distribution in periportal and perivenous hepatocytes.

The direct methylation method developed in this work for coffee samples allowed the determination of fatty acids using smaller sample amounts (100 mg), lower solvent volumes (5 mL) and shorter experimental run time (~12 min). Hence, the experiments are more economical and collaborate with green chemistry, besides favoring the health of the analyst who handles a smaller amount of solvents in a short period. Design Expert software was employed to determine the optimal concentration of the acid and base (0.7 e 1.75 molL-1) and evaluate the sonication time (5 min for both procedures). The experiments were carried out on different coffee samples in which the fat content ranging from 10.18% to 14.86%. The relative standard deviation values for intraday and interday were 2.78% and 7.8%, respectively, confirming the good precision of the proposed method for fatty acid derivatization.

A high-carbohydrate diet (HCD) is a well-established experimental model of accelerated liver fatty acid (FA) deposition and inflammation. In this study, we evaluated whether canola oil can prevent these physiopathological changes. We evaluated hepatic FA accumulation and inflammation in mice fed with a HCD (72.1% carbohydrates) and either canola oil (C group) or soybean oil (S group) as a lipid source for 0, 7, 14, 28, or 56 days. Liver FA compositions were analyzed by gas chromatography. The mRNA expression of acetyl-CoA carboxylase 1 (ACC1) was measured as an indicator of lipogenesis. The mRNA expression of F4/80, tumor necrosis factor-α (TNF-α), interleukin (IL)-1ß, IL-6, and IL-10, as mediators of liver inflammation, were also measured. The C group stored less n-6 polyunsaturated FAs (n-6 PUFAs) and had more intense lipid deposition of monounsaturated FAs (MUFAs), n-3 PUFAs, and total FAs. The C group also showed higher ACC1 expression. Moreover, on day 56, the C group showed higher expressions of the inflammatory genes F4/80, TNF-α, IL-1ß, and IL-6, as well as the anti-inflammatory IL-10. In conclusion, a diet containing canola oil as a lipid source does not prevent the fatty acid accumulation and inflammation induced by a HCD.

Despite the numerous studies that have shown a wide range of biological activities to berry fruits, scientific data focusing on modern, rapid and simple extraction methods followed by a clean-up step is still lacking. Therefore, the present work was aimed at investigating the use of a fast one-step solid-liquid extraction procedure followed by a dispersive solid-phase extraction (d-SPE) clean-up step to evaluate the phenolic composition, antioxidant and antiproliferative activities from three of the principal berries found in Brazil, pomegranate (Punica granatum L.), blackberry (Rubus ulmifolius Schott.), and strawberry (Fragaria × ananassa Duch.). Under the optimized extraction conditions, sixteen phenolic compounds were determined by UHPLC-MS/MS analysis and all berry extracts showed antioxidant activity and antiproliferative effects on cervical (HeLa) and colon (HT-29) cancer cells. Overall, these results highlight the importance of the clean-up step for more reliable data in studies of health-promoting proprieties from berry fruits.

Coconut oil has properties that are beneficial to human health. It assists in reducing total cholesterol, triacylglycerol (TAG), phospholipids, low-density lipoprotein (LDL) cholesterol, and very low-density lipoprotein (VLDL) cholesterol in serum and tissues. So its production, and consequently consumption, have increased in recent years. However, it has been a target for intentional adulteration with lower priced oils and fats, such as soybean oil and palm kernel oil (PKO). Coconut oil (CO) and PKO have similar chemical and physical characteristics that make it difficult to verify adulteration of CO with PKO. This study demonstrates a simple, sensitive, and fast technique that uses direct infusion electrospray ionisation mass spectrometry (ESI-MS) in conjunction with principal component analysis (PCA), in order to detect CO adulterated with PKO. Among the seven commercial coconut oil samples analysed, three were adulterated with PKO. Therefore, the suggested direct infusion ESI-MS method can be used in routine analysis to guarantee the quality of coconut oil.

The effects on the viability of promastigote and amastigote forms of Leishmania (Leishmania) amazonensis and citotoxicity in J774.A1 macrophages of ethanolic extract, fractions and isolated compounds that were obtained from the leaves of Nectandra cuspidata were determined. The total phenolics content (TP) and antioxidant activity were assessed using different assays. The ethyl acetate fraction (EAF) had the best antiproliferative activity (amastigote IC50: 4.42 ± 0.60 µg/mL, promastigote IC50: 33.33 ± 0.78) and presented no cytotoxicity at the highest concentration tested (1000 µg/mL). The EAF also had the greatest antioxidant capacity, which was comparable to butylated hydroxytoluene and quercetin (DPPH IC50 6.54 ± 0.10 µg/mL, ABTS 4.75 ± 0.11 mmol ET/g; FRAP 2.37 ± 0.03 mmol ET/g; ORAC = 33.52 ± 1.99 mmol ET/g; TP 387.11 ± 0.25 mg GAE/g). Vitexin, epicatechin and isovitexin isolated from EAF contribute to both activities.

The aims of this study were the optimization and validation of a new ultrasound assisted phytosterol method through analysis of response surface generated from Box-Behnken design. Variables quantity of sample (g), volume of KOH/MeOH (mL) and ultrasound time (s) were investigated in three levels. The evaluation process of the methodology was executed through determination of analytical parameters, such as selectivity, linearity, accuracy, dynamic linear range, "intra-day" accuracy and limits of detection and quantification. Optimized conditions for stigmasterol determination were 0.25g; 2.5mL; 300s; and ß-sitosterol were 0.25g; 5.4mL; 300s. The evaluated conditions and all investigated analytical parameters showed that the method is effective for determination of analytes ß-sitosterol and stigmasterol. This new analytical method has the same efficiency of the traditional method, while significantly reducing the total time of extraction for 300s regarding saponification step when compared to 3600s from the traditional method.

Fast and innovative methodology to monitors the addition of soybean oil in extra virgin olive oil was developed employing ESI-MS with ionization operating in positive mode. A certified extra virgin olive oil and refined soybean oil samples were analyzed by direct infusion, the identification of a natural lipid marker present only in soybean oil (m/z 886.68 [TAG+NH4]+) was possible. The certified extra virgin olive oil was purposely adulterated with soybean oil in different levels (1, 5, 10, 20, 50, 70, 90%) being possible to observe that the new methodology is able to detect even small fraud concentration, such as 1% (v/v). Additionally, commercial samples were analyzed and were observed the addition of soybean oil as a common fraud in this segment. This powerful analytical method proposed could be applied as routine analysis by control organization, as well as food industries, considering its pronounced advantages; simplicity, rapidity, elevated detectability and minor amounts of sample and solvent consumed.

OBJECTIVE: To investigate the antiinflammatory effects of a single administration of fish oil (FO) on the acute inflammatory response. METHODS: The paw edema and pleurisy models were used to evaluate the effects of FO dissolved in olive oil (FOP) orally administered in a single dose in rats. Nitric oxide (NO) concentrations in the pleural exudate were performed according to the Griess method and the cytokine concentrations were determined by Luminex bead-based multiplex assay. RESULTS: FOP treatment (30 and 300 mg/kg) significantly reduced paw edema. FOP treatment at 18.75, 37.5, 75.0, 150.0, and 300 mg/kg decreased both the volume of pleural exudate and cellular migration into the pleural cavity and each of these doses presented the same effectiveness. Treatment with FOP (300 mg/kg) reduced NO, TNF-α, IL-1ß, and IL-6 concentrations in the pleural exudate. CONCLUSIONS: The present data provide evidence that FO has inhibitory effects on the acute inflammatory response when administered in a single dose in rats. This effect might be attributable to a direct inhibitory effect of FO on the production or release of inflammatory mediators that are involved in the pathological processes evaluated herein.

ABSTRACT: This study evaluated the yield, color, and chemical composition of commercial cuts (tail, sirloin cut, back fillet, and thigh) of Pantanal caiman meat in both sexes. The yield of tail was higher than other cuts, and the yield of females (17.0%) was higher than males (15.9%). The thigh of males had lower protein content (20.8%) compared with other cuts. Females showed a higher lipid content in the tail (2.4%) and thigh (0.8%) compared with males (1.7% and 0.4%, respectively). The tail presented the greatest content of monounsaturated fatty acids (45.2%) and higher n6/n3 ratio (4.6). Although lightness was not different among cuts or between sexes, there were differences in color. Males have more yellowish meat compared with females. Thigh and back fillet were more reddish when compared to sirloin cut and tail, regardless of sex. In conclusion, female tail meat provided greater yield and lipid content than males, and this result was statistically significant. These findings can help producers and consumers alike, better understand yield, quality, and nutritional quality of Pantanal caiman meat.


RESUMO: Objetivando avaliar rendimento, cor e composição química dos cortes comerciais (cauda, filé de lombo, filé de dorso e coxa) da carne do jacaré-do-Pantanal nos diferentes sexos, foram processados 20 animais, sendo estes, dez fêmeas (3677g) e dez machos (3510g). Não houve interação significativa (P>0,05) entre cortes comerciais e sexo para peso dos cortes, teor de umidade e cor. O rendimento da cauda foi superior aos demais cortes, sendo o rendimento das fêmeas (17.0%) superior ao dos machos (15,9%). A coxa dos machos apresentou menor teor de proteína (20,8%), em relação aos demais cortes. As fêmeas apresentaram maior teor de lipídeos na cauda (2,4%) e na coxa (0,8%), em relação aos machos (1,7% e 0,4%, respectivamente). A cauda apresentou maior quantidade de ácidos graxos monoinsaturados (45,2%) e maior razão n6/n3 (4,6). A luminosidade não foi diferente entre cortes e sexos. Os machos apresentaram carne com coloração mais amarelada em relação às fêmeas, e a coxa e o filé de dorso tiveram cor mais avermelhada, comparadas ao filé de lombo e cauda, independente do sexo. Concluiu-se que houve influência do sexo no corte de cauda, para o teor de lipídios e rendimento, sendo que a fêmea apresentou rendimento e lipídeos maiores que o macho.

Both high-carbohydrate diet (HCD) and high-fat diet (HFD) modulate liver fat accumulation and inflammation, however, there is a lack of data on the potential contribution of carbohydrates and lipids separately. For this reason, the changes in liver fatty acid (FA) composition in male Swiss mice fed with HCD or HFD were compared, at the time points 0 (before starting the diets), and after 7, 14, 28 or 56 days. Activities of stearoyl-CoA desaturase-1 (SCD-1), ∆-6 desaturase (D6D), elongases and de novo lipogenesis (DNL) were estimated. Liver mRNA expression of acetyl-CoA carboxylase 1 (ACC1) was evaluated as an additional indicator of the de novo lipogenesis. Myeloperoxidase activity, nitric oxide (NO) production, and mRNA expressions of F4/80, type I collagen, interleukin (IL)-6, IL-1ß, IL-10, and tumor necrosis factor-α (TNF-α) were measured as indication of the liver inflammatory state. The HCD group had more intense lipid deposition, particularly of saturated fatty acids (SFAs) and monounsaturated fatty acids (MUFAs). This group also showed higher DNL, SCD-1, and D6D activities associated with increased NO concentration, as well as myeloperoxidase activity. Livers from the HFD group showed higher elongase activity, stored more polyunsaturated fatty acids (PUFAs) and had a lower omega-6/omega-3 fatty acid (n-6/n-3) ratio. In conclusion, liver lipid accumulation, fatty acids (FA) composition and inflammation were modulated by the dietary composition of lipids and carbohydrates. The HCD group had more potent lipogenic and inflammatory effects in comparison with HFD.

In order to analyze the composition of fatty acids employing gas chromatography as the separation method, a derivatization of lipids using esterification and transesterification reactions is needed. The methodologies currently available are time consuming and use large amounts of sample and reagents. Thus, this work proposes a new procedure to carry out the derivatization of fatty acids without the need for prior extraction of lipids. The use of small amounts of sample (100mg) allows the analysis to be performed in specific parts of animals, in most cases without having them slaughtered. Another benefit is the use of small amounts of reagents (only 2mL of NaOH/Methanol and H2SO4/Methanol). The use of an experimental design procedure (Design Expert software) allows the optimization of the alkaline and acid reaction times. The procedure was validated for five minutes in both steps. The method was validated for bovine fat, beef, chicken, pork, fish and shrimp meats. The results for the merit figures of accuracy (from 101.07% to 109.18%), precision (RSDintra-day (from 0.65 to 3.93%), RSDinter-day (from 1.57 to 5.22%)), linearity (R(2)=0.9864) and robustness confirmed that the new method is satisfactory within the linear range of 2-30% of lipids in the sample. Besides the benefits of minimizing the amount of samples and reagents, the procedure enables gas chromatography sample preparation in a very short time compared with traditional procedures.

Solanum americanum Mill. is a plant that belongs to the Solanaceae family, its respective ripe fruit is dark purple. Ripe S. americanum Mill. fruits were submitted to physicochemical characterisation, and their trans-resveratrol contents were quantified by high-performance liquid chromatography. Such determination was executed with fruits at different stages of ripeness and freeze-stored fruits as well. In natura ripe fruit pulp and peel presented average trans-resveratrol amounts of 1.07 and 0.7960 µg per gram of sample, respectively. These amounts are significantly higher when compared to freeze-stored fruit (0.1353 µg of trans-resveratrol per gram of sample) and to other berries. All ripe fruits showed significant amounts of total anthocyanins and total antioxidants. Thus, for the first time, trans-resveratrol has been identified and quantified in S. americanum Mill. fruit samples.

Flaxseed (Linum usitatissimum L.) oil was obtained via subcritical n-propane fluid extraction (SubFE) under different temperatures and pressures with an average yield of 28% and its composition, purity and oxidative stability were compared to oils obtained via conventional solvent extraction methods (SEMs). When the oxidative stability was measured by differential scanning calorimetry, the oil was found to be up to 5 times more resistant to lipid oxidation as compared to the SEM oils. Direct infusion electrospray ionization mass spectrometry (ESI-MS) analysis showed characteristic and similar TAG profiles for SubFE and SEMs oils but higher purity for the SubFE oil. The flaxseed oil content of ß-tocopherol, campesterol, stigmasterol and sitosterol were quantified via GC-MS. SubFE showed to be a promising alternative to conventional SEM since SubFE provides an oil with higher purity and higher oxidation stability and with comparable levels of biologically active components.

This study evaluated the effect of the dietary replacement of sunflower oil with perilla oil in Nile tilapia (GIFT strain) at 0, 10, 20 and 30 days. There was an increase in total omega 3 fatty acids (n-3) in muscle tissue (from 63.6 to 181.5 mg g(-1)) and a decrease in total concentration of omega 6 fatty acids (n-6) ranging between 255.6 and 196.1 mgg(-1). Amongst the n-3 fatty acids, the concentrations of alpha-linolenic acid (18:3n-3) and docosahexaenoic acid (20:6n-3) increased by 9.4 and 1.9 times, respectively, after 30 days for the perilla oil-enriched diet. The n-6/n-3, polyunsaturated and saturated fatty acids (PUFA/SFA), and polyunsaturated and monounsaturated fatty acids (PUFA/MUFA) ratios tended to stabilize after 20 days under the perilla oil-enriched diet. Incorporation of perilla oil as a lipid source in the diet of tilapia for 20 or 30 days resulted in significant changes in the fatty acid composition of the fish muscle tissue, contributing to increase its nutritional value.

This study was conducted to evaluate the performance, carcass characteristics and chemical composition of Longissimusmuscle (LM) of the bulls. Twenty-four Nellore bulls were used in a complete randomised design. The bulls were randomly assigned to one of the three diets containing 0, 5 or 12% glycerin. Final BW and ADG were similar (P>0.05) between the bulls fed with 5 or 12% of glycerin but were higher (P<0.10) compared to the bulls fed without glycerin. DMI and feed efficiency was not affected (P>0.05) by glycerin level. Hot carcass weight increased (P<0.10) when glycerin was fed at 5%. Glycerin level did not affect (P<0.05) the dressing percentage and fat thickness. LM increased (P<0.10) when glycerin was fed at 12%. Dietary glycerin did not affect (P>0.05) the conformation, colour, texture, marbling and pH. There was difference (P>0.05) for moisture, ashes and crude protein among glycerin levels. Bulls fed 12% glycerin present the highest (P<0.03) total lipids on LM. The percentage of saturated fatty acids (SFA), monounsaturated acids (MUFA), polyunsaturated acids (PUFA), n-6 and n-3 fatty acids and PUFA/SFA and n-6:n-3 ratios of the LM were similar among the diets. In conclusion, glycerin level did not affect the animal performance and carcass characteristics of Nellore bulls finished in feedlot.

Omega-3 polyunsaturated fatty acids (n-3 PUFA) can modulate the immune system and their primary effect is on macrophage function. Paracoccidioidomycosis (PCM) is an endemic systemic mycosis in Latin America that is caused by the dimorphic fungus Paracoccidioides brasiliensis (Pb). Macrophages are the main defence against this pathogen and have microbicidal activity that is dependent on interferon-Γ and tumour necrosis factor (TNF)-α. These cytokines stimulate the synthesis of nitric oxide (NO) and hydrogen peroxide (H2O2), leading to the death of the fungus. To study the effect of n-3 PUFA on the host immune response during experimental PCM, macrophages that were obtained from animals infected with Pb18 and fed a diet enriched by linseed (LIN) oil were cultured and challenged with the fungus in vitro. The macrophage function was analysed based on the concentrations of TNF-α, NO and H2O2. LIN oil seems to influence the production of TNF-α during the development of disease. A diet enriched with LIN oil influences the microbicidal activity of the macrophages by inducing the production of cytokines and metabolites such as NO and H2O2, predominantly in the chronic phase of infection.
