RESUMO

Fatty acid photodecarboxylase in Chlorella variabilis NC64A (CvFAP) performed excellent ability to exclusively decarboxylate renewable fatty acids for C1-shortened hydrocarbons fuel production under visible light. However, the large-scale application by such an approach is limited by the free state of CvFAP catalyst, which is unstable for efficient biofuel production. In this study, CvFAP was immobilized in magnetic nickel ferrite (NiFe2O4) nanoparticles for facile recovery by a simple procedure. The shift of Ni 2p in electron binding energy was detected to clarify the interaction between Ni2+ and histidine of CvFAP. The coordination of NiFe2O4 and CvFAP contributed to an efficient affinity binding with an immobilization capacity of 98 mg/g carrier. Hydrocarbon fuel concentration of 3.7 mM was obtained by NiFe2O4@CvFAP-induced photoenzymatic decarboxylation. The high stability of CvFAP in terms of residual enzyme activity of 79.7% at pH 9.0 and that of 68% at organic solvent ratio of 60%, respectively, were observed.

OBJECTIVE: This study aimed to report the prognostic predictors and compare the long-term outcomes of complete cytoreductive surgery (CCRS) vs. debulking surgery (DS) in patients with pseudomyxoma peritonei (PMP) of appendiceal origin. METHODS: A retrospective analysis of 1008 consecutive patients with PMP undergoing primary surgery from January 2008 to December 2019 was performed. A propensity score-matched (PSM) analysis (1:1) was performed, and oncologic outcomes were compared between the CCRS and DS groups. RESULTS: Out of 1008 patients, 258 patients were excluded. Baseline characteristics differed significantly between the CCRS and DS groups (total n = 750). After PSM, 106 patients were selected from each group and the baseline characteristics were matched between groups. There were significant differences between groups in operative time, the incidence of major complications (P = 0.017), and the numbers of organs removed. The median follow-up was 28 (1-131) months. Median overall survival (OS) for the 212 patients was 52.0 months (95% CI 40.2-63.8), and 10-year OS was 39.0%. Median OS could not be calculated for the CCRS group; in the DS group, this value was 41 months (P = 0.010). The 10-year OS rate was 54.2% in the CCRS group and 31.2% in the DS group. Multivariate analyses identified CCRS (P = 0.012) and histopathologic subtype (P < 0.001) as independent prognostic factors for OS. CONCLUSIONS: In this matched-pair analysis of patients with appendiceal PMP, CCRS was safe and associated with better prognosis than DS. The completeness of cytoreduction and histopathologic subtype were two independent prognostic factors for OS.

Pseudomyxoma peritonei (PMP) is a rare disorder with unique pathological and genetic changes. Although several studies have reported the clinical features and mutational changes of PMP that originates from the appendix, few studies on PMP originating from the ovary have been reported due to its extreme rarity. In order to characterize the somatic mutational landscape and to investigate the prognosis predicting factors of ovary-originating PMP, we examined 830 cases of PMP and identified 16 patients with PMP that originated from the ovary. Whole-exome sequencing (WES) was performed on 12 cases using formalin-fixed, paraffin-embedded (FFPE) tissue samples. We found that 25% (3/12) of the patients carried mutations in cancer driver genes, including TP53, ATM and SETD2, and 16.7% (2/12) of the patients carried mutations in cancer driver genes, including ATRX, EP300, FGFR2, KRAS, NOCR1 and RB1. The MUC16 (58.33%), BSN (41.67%), PCNT (41.67%), PPP2R5A (41.67%), PRSS36 (41.67%), PTPRK (41.67%) and SBF1 (41.67%) genes presented the highest mutational frequencies. The PI3K-Akt signaling pathway, human papillomavirus infection pathway, cell skeleton, cell adhesion, and extracellular matrix and membrane proteins were the major pathways or functions that were affected. Patients were followed up to 174 months (median: 48.26 months). The 5-year OS rate for all patients was 71.2% and the median OS was not reached. PTPRK mutations, presurgical CA199 level, completeness of cytoreduction (CCR) and peritoneal cancer index (PCI) were identified as potential predictive factors for patient survival. In conclusion, the mutational landscape for ovary-originating PMP was revealed and exhibited unique features distinct from appendix-originating PMP. PTPRK, CA199, CCR and PCI may predict patient survival.

Stimulating direct interspecies electron transfer with conductive materials is a promising strategy to overcome the limitation of electron transfer efficiency in syntrophic methanogenesis of industrial wastewater. This paper assessed the impact of conductive foam nickel (FN) supplementation on syntrophic methanogenesis and found that addition of 2.45 g/L FN in anaerobic digestion increased the maximum methane production rate by 27.4 % (on day 3) while decreasing the peak production time by 33 % as compared to the control with no FN. Cumulative methane production from day 2 to 6 was 14.5 % higher with addition of 2.45 g/L FN than in the control. Levels of FN in excess of 2.45 g/L did not show benefits. Cyclic voltammetry results indicated that the biofilm formed on the FN could generate electrons. The dominant bacterial genera in suspended sludge were Dechlorobacter and Rikenellaceae DMER64, whereas that in the FN biofilm was Clostridium sensu stricto 11. The dominant archaea Methanosaeta in the FN biofilm was enriched by 14.1 % as compared to the control.

Heavy metals inevitably cause invisible or visible damage to plants, leading to significant economic losses. Therefore, it is necessary to develop a method for timely monitoring the damage of plants under the stress of heavy metals. Here, vitronectin-like proteins (VN) on the surface of plant cells is as an important biomarker for monitoring damage of plants under the stress of heavy metals. A living plant cell-based biosensor is constructed to monitor invisible damage of plant cells induced by cadmium [Cd(II)] or lead [Pb(II)]. To fabricate this sensor, l-cysteine was first modified on the glassy carbon electrode followed by the modification of anti-IgG-Au antibody. Then, the living plant cells, incubated with the anti-VN, were modified onto the electrode. The sensor worked by determining the change in electrochemical impedance. Cd(II) and Pb(II) was detected in the linear dynamic range of 45-210 and 120-360 µmol·L-1, respectively. And the detection limit of Cd(II) and Pb(II) of this biosensor was 18.5 nmol·L-1 [with confidence interval (95%) 18.4-18.6 nmol·L-1] and 25.6 nmol·L-1 [with confidence interval (95%) 25.4-25.8 nmol·L-1], respectively. In both Arabidopsis and soybean, when the content of VN increased by about 20 times under the stress of Cd(II) or Pb(II), which means when the electron-transfer resistance increased by 35%, chlorophyll content showed significant decrease about 17%. Therefore, by establishing a quantitative relationship among the content of biomarker, the electron-transfer resistance and chlorophyll content in plant cells, the invisible damage of plants under the stress of heavy metals was detected. These results can provide a reference method for early-onset warning systems for heavy metal pollution in the environment.

OBJECTIVE: Mild hypothermia has a protective effect on ischemic stroke, but the mechanisms remain elusive. Here, we investigated microRNA (miRNA) profiles and the specific role of miRNAs in ischemic stroke treated with mild hypothermia. MATERIALS AND METHODS: Male adult Sprague Dawley rats were subjected to focal transient cerebral ischemia. Mild hypothermia was induced by applying ice packs around the neck and head of the animals. miRNAs expression profiles were detected in ischemic stroke treated with mild therapeutic hypothermia through miRNA chips. Reverse transcription-polymerase chain reaction (RT-PCR) was used to verify the change of miRNA array. Western blot and adenosine triphosphate (ATP) assay kits were used to detect the changes of protein expression and ATP levels, respectively. miR-15b mimic and its control were injected into the right lateral ventricle 60 min before the induction of ischemia. RESULTS: The results showed that mild hypothermia affected miRNAs profiles expression. We verified the expression of miR-15b and miR-598-3p by miRNA RT-PCR. miR-15b mimic inhibited the expression of its target, ADP ribosylation factor-like 2 (Arl2) protein, and decreased ATP levels in PC12 cells. Compared with the control, miR-15b mimic increased the infarct volume and aggravated the neurological function under normothermia or hypothermia treatment. Furthermore, the expression of Arl2 was decreased in the miR-15b mimic group under normothermia or hypothermia treatment. CONCLUSIONS: Mild therapeutic hypothermia affected miRNA profiles and protected against cerebral ischemia/reperfusion by inhibiting miR-15b expression in rats. miR-15b may be a potential target for therapeutic intervention in stroke.

Ascophyllum nodosum, an abundant Irish brown seaweed, shows significant seasonal variation in chemical composition and biogas production. The polyphenol content is shown to be a more important factor in biogas production than ash content. High polyphenol content in summer months adversely affected biogas production; suggesting two potential harvest dates, March and October. A. nodosum harvested in October showed a relatively low level of polyphenols (2% of TS) and ash (23% of volatile solids), and exhibited a specific methane yield of 215LCH4kgVS(-1), which was 44% of theoretical yield. The highest yield per wet weight of 47m(3)CH4t(-1) was achieved in October, which is 2.9 times higher than the lowest value (16m(3)CH4t(-1)), obtained in December. The gross energy yield of A. nodosum based on the optimal biogas production can achieve 116GJha(-1)yr(-1) in October.

We prepared poly-L-aspartic acid (PASP) functionalized NaYF4:Yb(3+),Er(3+) upconversion nanoparticles (UCNP-PASP). These nanoparticles can give red upconversion emission under excitation at 915 nm, whose wavelength of emission and excitation is located in the optical window of biological tissue. Dynamic laser scatting and zeta potentials of UCNP-PASP were used to study their stabilities in different aqueous solution. To understand the mechanism of the red emission of UCNP-PASP, photoluminescence spectra of samples were recorded before and after modification with PASP, poly acrylic acid (PAA), and poly(ether imide) (PEI) ligands under excitation at 915 and 980 nm, respectively. The cytotoxicity of the UCNP-PASP was also examined on a A549 cell and KB cell by 3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyl-tetrazolium bromide (MTT) assay. Moreover, the PASP-functionalized UCNP was employed as a potential biomarker for in vitro and in vivo experiments of upconversion luminescence imaging.

Dual-modality imaging with magnetic resonance (MR) and upconversion luminescence (UCL) is a promising technique for molecular imaging in biomedical research. Multifunctional lanthanide-based nanoparticles have been widely investigated as agents for contrast enhanced MR and fluorescence imaging. However, the use of rare earth fluoride nanoparticles for dual-modality imaging of T2-weighted MR and UCL is rarely reported. We find that NaYF4:Yb(3+),Tm(3+),Co(2+) (MUC) nanorods can be applied as a high-performance dual contrast agent for both T2-weighted MR and UCL dual-modality imaging. After modification with 6-O-carboxymethyl chitosan (OCC), MUC nanorods can be endocytosed by cells without showing signs of cytotoxicity. High-quality UCL images of living cells incubated with MUC-OCC nanorods were acquired on a near-infrared (NIR) confocal microscopy under the excitation at 980 nm. Moreover, MUC-OCC nanorods display high transverse (r2) relaxivities in vitro. The application of low-dose MUC-OCC nanorods for NIR-to-NIR UCL and MR dual-modality in vivo imaging was also carried out successfully. In addition, the toxicity of MUC-OCC nanorods was evaluated by MTT assay, serological tests and histological analysis of visceral organs.
