RESUMO

The present work prepared a novel BiMgO-2MBD (X = 0.42) material for iodine vapor capture in temperature conditions related to spent nuclear fuel reprocessing and nuclear accidents. BiMgO-2MBD (X = 0.42) was synthesized by a solvothermal process and exhibited an exceptional ultrafast and high iodine uptake with a capacity of 4352.12 mg/g and 5147.08 mg/g after 5 h at 75 °C and 150 °C, respectively. The TGA analysis shows that Bi/Mg oxide substrate highly contributed to improving the thermal stability of the functionalized BiMgO-2MB (X = 0.42) as indicated by the weight losses of the material components of 3.77 wt%, 29.32 wt%, and 97.72 wt%, respectively for Bi/Mg oxide, BiMgO-2MBD, and 2-MBD. The material characterization and DFT calculations indicate that 2-MBD played a significant role towards improving iodine capture capacity. For long-term and safe waste disposal, a chemically durable waste form was made from etidronic acid and Bi2O3, and successfully immobilized the iodine-loaded wastes (I2 @BiMgO-2MBD) which exhibited a low normalized leaching rate of 1.098 × 10-6 g.m2/day for 7 days under the PCT-A method. In addition, BiMgO-2MBD (X = 0.42) showed an ability to be reused after several regeneration cycles. The comparison with previously reported materials shows that the current BiMgO-2MBD (X = 0.42) is the first functionalized metal oxide comparable to metal-organic and covalent organic frameworks for iodine uptake. BiMgO-2MBD (X = 0.42) shows promising results for practical applications in the gas phase capture of radioactive iodine.

BACKGROUND: Thrombocytopenia has long been considered an important complication of chemotherapy and radiotherapy, which severely limits the effectiveness of cancer treatment and the overall survival of patients. However, clinical treatment options are extremely limited so far. Ruxolitinib is a potential candidate. METHODS: The impact of ruxolitinib on the differentiation and maturation of K562 and Meg-01 cells megakaryocytes (MKs) was examined by flow cytometry, Giemsa and Phalloidin staining. A mouse model of radiation-injured thrombocytopenia (RIT) was employed to evaluate the action of ruxolitinib on thrombocytopoiesis. Network pharmacology, molecular docking, drug affinity responsive target stability assay (DARTS), RNA sequencing, protein blotting and immunofluorescence analysis were applied to explore the targets and mechanisms of action of ruxolitinib. RESULTS: Ruxolitinib can stimulate MK differentiation and maturation in a dose-dependent manner and accelerates recovery of MKs and thrombocytopoiesis in RIT mice. Biological targeting analysis showed that ruxolitinib binds directly to Toll Like Receptor 2 (TLR2) to activate Rac1/cdc42/JNK, and this action was shown to be blocked by C29, a specific inhibitor of TLR2. CONCLUSIONS: Ruxolitinib was first identified to facilitate MK differentiation and thrombocytopoiesis, which may alleviate RIT. The potential mechanism of ruxolitinib was to promote MK differentiation via activating the Rac1/cdc42/JNK pathway through binding to TLR2.

BACKGROUND: Cibotii rhizoma (CR) is a famous traditional Chinese medicine (TCM) used to treat bleeding, rheumatism, lumbago, etc. However, its therapeutic effects and mechanism against thrombocytopenia are still unknown so far. In the study, we investigated the effects of aqueous extracts of Cibotii rhizoma (AECRs) against thrombocytopenia and its molecular mechanism. METHODS: Giemsa staining, phalloidin staining, and flow cytometry were performed to measure the effect of AECRs on the megakaryocyte differentiation in K562 and Meg-01 cells. A radiation-induced thrombocytopenia mouse model was constructed to assess the therapeutic actions of AECRs on thrombocytopenia. Network pharmacology and experimental verification were carried out to clarify its mechanism against thrombocytopenia. RESULTS: AECRs promoted megakaryocyte differentiation in K562 and Meg-01 cells and accelerated platelet recovery and megakaryopoiesis with no systemic toxicity in radiation-induced thrombocytopenia mice. The PI3K/AKT, MEK/ERK, and JAK2/STAT3 signaling pathways contributed to AECR-induced megakaryocyte differentiation. The suppression of the above signaling pathways by their inhibitors blocked AERC-induced megakaryocyte differentiation. CONCLUSIONS: AECRs can promote megakaryopoiesis and thrombopoiesis through activating PI3K/AKT, MEK/ERK, and JAK2/STAT3 signaling pathways, which has the potential to treat radiation-induced thrombocytopenia in the clinic.

Efficient iodine gas capture is necessitated in many industries like spent nuclear fuel off-gas treatment in view of environmental protection and resource recycling. However, the adsorption efficiency and stability of the current adsorbents are limited. In the present work, efficient and stable barbituric and thiobarbituric acid-based UiO-66-NH2 adsorbents (i.e., UiO-66-NH-B.D and UiO-66-NH-T.D, respectively) have been synthesized by post-synthetic covalent modification. Characterization approaches, including SEM-EDS, TEM, XRD, FTIR, XPS, 1H NMR, TGA and BET, are used to obtain information on the properties and adsorption mechanisms of these metal-organic framework (MOF) adsorbents. The kinetics and mechanisms involved are studied in detail. The treatment efficiency and recyclability of the adsorbents are checked and compared with the adsorbents reported in previous works. The results show that the current adsorbents are potentially suitable for efficient iodine gas capture. High maximum iodine adsorption amount by UiO-66-NH-B.D and UiO-66-NH-T.D (1.17 and 1.33 g/g) was achieved under 75 °C. These new adsorbents are thermally stable for iodine adsorption and regenerated and reused with good performance. The adsorption mechanisms were revealed based on experimental results, indicating that iodine is adsorbed by both physisorption and chemisorption.

The current study analyzes the contribution of 10 water quality parameters (including pH, turbidity, conductivity, total dissolved solids (TDS), hardness, total organic carbon (TOC), alkalinity, calcium ions, chlorides and sulfates) to corrosion extent of stainless steel valves taken from different locations in a reverse osmosis system of a reclaimed water plant. The valves were in service for 5 years. Raman spectroscopy and X-ray photoelectron spectroscopy analyses are conducted to quantify corrosion products on different valves under various water quality conditions. On that basis, bivariate and multivariate regression analyses between the 10 water quality parameters and the corrosion extent of valve specimens (represented by metal loss percentage (MLP) values) are carried out to check the contribution of those water quality parameters to MLP. The results indicate that the proportions of metal oxides as corrosion products vary according to the corrosion extent of the valves. Although no linear correlation is found, all 10 water quality parameters except for pH show a significant positive correlation with the MLP values of the valve specimens. Moreover, results of multivariate regression suggest that the variation of MLP can be explained by turbidity, TDS, TOC and sulfates. A positive contribution of turbidity, TDS and TOC to MLP is observed, whereas the contribution of sulfates is negative. The results from the current work help to identify the reasons for water quality-induced failure of stainless steel equipment in RO systems.

Risk of cross-connection is becoming higher due to greater construction of potable-reclaimed water dual distribution systems. Cross-connection events can result in serious health concerns and reduce public confidence in reclaimed water. Thus, reliable, cost-effective and real-time online detection methods for early warning are required. The current study carried out pilot-scale experiments to simulate potable-reclaimed water pipe cross-connection events for different mixing ratios (from 30% to 1%) using machine learning methods based on multiple conventional water quality parameters. The parameters included residual chlorine, pH, turbidity, temperature, conductivity, oxidation-reduction potential and chemical oxygen demand. The results showed that correlated variation occurred among water quality parameters at the time of the cross-connection event. A single parameter-based method can be effective at high mixing ratios, but not at low mixing ratios. The direct supporting vector machine (SVM)-based method managed to overcome this drawback, but coped poorly with abnormal readings of water parameter sensors. In that respect, a Pearson correlation coefficient (PCC)-SVM-based method was developed. It provided not only high detection performance under normal conditions, but also remained reliable when abnormal readings occurred. The detection accuracy and true positive rate of this method was still over 88%, and the false positive rate was below 12%, given a sudden variation of an individual water quality parameter. The receiver operating characteristic curves further confirmed the promising practical applicability of this PCC-SVM-based method for early detection of cross-connection events.

OBJECTIVE: Percutaneous pedicle screw fixation is commonly used for upper lumber burst fractures. The direct decompression remains challenging with this minimally invasive surgery. The objective was to evaluate a novel paraspinal erector approach for effective and direct decompression in patients with canal compromise and neurologic deficit. METHOD: Patients (n = 21) with neurological deficiency and Denis B type upper lumbar burst fracture were enrolled in the study, including 14 cases in the L1 and 7 cases in the L2. The patients underwent removal of bone fragments from the spinal canal through intervertebral foramen followed by short-segment fixation. Evaluations included surgery-related, such as duration of surgery and blood loss, and 12-month follow-up, such as the kyphotic angle, the height ratio of the anterior edge of the vertebra, the ratio of sagittal canal compromise, visual analog scale (VAS), Oswestry Disability Index (ODI), and Frankel scores. RESULTS: All patients achieved direct spinal canal decompression using the paraspinal erector approach followed by percutaneous pedicle screw fixation. The mean operation time (SD) was 173 (23) min, and the mean (SD) blood loss was 301 (104) ml. Significant improvement was noted in the kyphotic angle, 26.2 ± 8.7 prior to operation versus 9.1 ± 4.7 at 12 months after operation (p < 0.05); the height ratio of the anterior edge of the injured vertebra, 60 ± 16% versus 84 ± 9% (p < 0.05); and the ratio of sagittal canal compromise, 46.5 ± 11.4% versus 4.3 ± 3.6% (p < 0.05). Significant improvements in VAS (7.3 ± 1.2 vs. 1.9 ± 0.7, p < 0.05), ODI (86.7 ± 5.8 vs. 16.7 ± 5.1, p < 0.05), and Frankel scores were also noted. CONCLUSIONS: The paraspinal erector approach was effective for direct spinal canal decompression with minimal injury in the paraspinal muscles or spine. Significant improvements in spinal function and prognostics were achieved after the percutaneous pedicle screw fixation.

OBJECTIVE: To observe and measure the approach next to the erector spinae in the thoracic and lumbar segments of the spine and adjacent anatomical structures by the topographic method, to clarify the positioning method and safe range so as to provide the anatomical basis of the approach for spinal canal decompression. METHODS: Twelve formaldehyde-treated adult cadaver specimens were selected, including 6 males and 6 females with an average age of 43 years (range, 27-52 years) and with an average height of 166 cm (range, 154-177 cm). The related data of the approach at T1-S1 levels were respectively measured: the distance between the lateral edge of the erector spinae and the spinous process, the length of the approach, the angle between the approach and the horizontal plane, the size of intervertebral foramen, and the vertical distance between the segmental artery and the upper edge of the vertebrae. RESULTS: The distance between the lateral edge of the erector spinae and the spinous process ranged from (41.75 +/- 3.29) mm to (74.54 +/- 7.08) mm. The length of the approach ranged from (66.75 +/- 10.81) mm to (97.13 +/- 13.35) mm. The angle between the approach and the horizontal plane ranged from (38.38 +/- 6.16) degrees to (53.67 +/- 4.40) degrees. The vertical distance between the segmental artery and the upper edge of the vertebrae ranged from (9.50 +/-0.60) mm to (18.30 +/- 1.56) mm. The size of foraminal was also measured. The spinal canal could reach when iliocostalis lateral edge was used as the starting point in the lumbar segments, and longissimus lateral edge as the starting point in the thoracic segments. It was confirmed that there was enough safe space for the spinal decompression without the resection of the articular process. CONCLUSION: The approach next to the erector spinae can reach spinal canal to achieve the purpose of decompression through the intervertebral foramen. The minimally invasive approach is feasible and safe. It has the value of the operative application.

Influenza infection after allogeneic hematopoietic cell transplantation (allo-HCT) can result in severe complications. The effectiveness of the annual vaccine depends on age, immune competence, and the antigenic potential of the 3 strains included. We hypothesized that a second vaccine dose, the standard of care for vaccine-naïve children, might improve post hematopoietic cell transplantation (HCT) immune responses. Patients >60 days post-HCT were randomized to receive either 1 (n = 33) or 2 (n = 32) influenza vaccine doses separated by 1 month. The primary endpoint was whether 2 vaccinations induced superior immunity; however, we found no difference. Secondary endpoints were to identify variables associated with responses. Both hemagglutination inhibition (HI; P < .005) and ELISpot responses (P = .03) were greater for patients vaccinated ≥ 1 year posttransplantation. Umbilical cord blood (UCB) recipients showed less IFN-γ responses (P < .001). Interestingly, there was a positive correlation between the total number of CD19(+) cells before vaccination and seroconversion (P = .01) and an inverse correlation for IFN-γ responses (P = .05). Variables not associated with vaccine responses included prevaccine CD4(+) cell counts (total, naïve, or memory), steroid usage at vaccination, age, or conditioning intensity. Time from transplantation to vaccination and absolute CD19(+) cell counts were the strongest predictors of vaccine responses. Methods to improve influenza vaccine responses after allo-HCT are needed.

RESUMO

We evaluated pandemic influenza A (H1N1) 2009 virus isolates and respiratory tissues collected at autopsy by electron microscopy. Many morphologic characteristics were similar to those previously described for influenza virus. One of the distinctive features was dense tubular structures in the nuclei of infected cells.
