In their natural habitat, insects may bioaccumulate toxins from plants for defence against predators. When insects are accidently raised on feed that is contaminated with toxins from co-harvested herbs, this may pose a health risk when used for human or animal consumption. Plant toxins of particular relevance are the pyrrolizidine alkaloids (PAs), which are genotoxic carcinogens produced by a wide variety of plant species and the tropane alkaloids (TAs) which are produced by a number of Solanaceae species. This study aimed to investigate the transfer of these plant toxins from substrates to black soldier fly larvae (BSFL) and lesser mealworm (LMW). PAs and the TAs atropine and scopolamine were added to insect substrate simulating the presence of different PA- or TA-containing herbs, and BSFL and LMW were grown on these substrates. Bioaccumulation from substrate to insects varied widely among the different plant toxins. Highest bioaccumulation was observed for the PAs europine, rinderine and echinatine. For most PAs and for atropine and scopolamine, bioaccumulation was very low. In the substrate, PA N-oxides were quickly converted to the corresponding tertiary amines. More research is needed to verify the findings of this study at larger scale, and to determine the potential role of the insect and/or substrate microbiome in metabolizing these toxins.

The presence of carcinogenic aflatoxins in food and feed is a major issue. In prior studies, aflatoxin B1 (AfB1) and known primary metabolites were absent from Lesser Mealworm (LMW, Alphitobius diaperinus) reared on contaminated diets. LMW is a promising alternative protein source. The objectives of this stu\dy were to determine whether LMW can be reared on AfB1-contaminated feed in each life-stage, and to gather more insight into potential metabolites formed. Results suggested no adverse effects in terms of survival/growth when three stages of LMW (larvae, pre-pupae, beetles) were exposed to feed containing AfB1 concentrations of 200 and 600 µg/kg for 48 h. Insect and frass samples were analyzed by LC-MS/MS and high-resolution MS to, respectively, quantify concentrations of AfB1 and its major metabolites, and determine secondary metabolites. No AfB1 or major metabolites were quantified in the insect samples. Mass balance calculations showed that up to 40% of spiked AfB1 could be recovered in the frass, in the form of AfB1, aflatoxicol and AfM1. HRMS results suggested the presence of additional metabolites in the frass, but, due to lack of commercially available reference standards for these compounds, exact identification and quantification was not possible. More research is needed to verify the absence of toxicity.

This study aimed to develop a mitigation strategy for the formation of 2-monochloropropane-1,3-diol esters (2-MCPDE), 3-monochloropropane-1,2-diol esters (3-MCPDE), and glycidol fatty acid esters (GE) during palm oil refining. Single physical refining was the starting point (the control) for this study. Experimental treatments including a double refining repeating the entire single refining process (T1), double refining with a high-low deodorization temperature (T2), and double deodorization (T3) with similar temperature settings as T2 were performed. Compared with the control experiment, T2 successfully reduced the formation of GE by 87%; in particular, the second degumming and bleaching were crucial for eliminating GE. Both 2- and 3-MCPDE were formed prior to the deodorization process in all treatments. MCPDE concentrations remained stable throughout the refining process and, hence, they require a different mitigation approach as compared to GE. These results provide useful insights which can directly be implemented by the oil industry.

The refining of vegetable oils leads to the formation of 2- and 3-monochloropropane-1,2-diol esters (2- and 3-MCPD-E), and glycidyl esters (Gly-E). A literature review was performed aiming to provide up-to-date knowledge on mitigation strategies during oil refining that can reduce the formation of these three processing contaminants. The review used the database Scopus and covered the period from 2009 to 2017. Most of the 18 papers dealt with palm oil and two papers with vegetable oil. Most studies focused on 3-MCPD-E, some on Gly-E, and none on 2-MCPD-E. Water degumming was able to reduce the concentrations of 3-MCPD-E by 84% and Gly-E by 26%. Neutralization of the oil reduced concentrations of 3-MCPD-E by 81% and Gly-E by 84%. Bleaching with synthetic magnesium silicate reduced the 3-MCPD-E concentration by 67%. For the deodorization step, several mitigation strategies, such as double-deodorization, the addition of various antioxidants, or a longer deodorization time, can reduce the formations of 3-MCPD-E by 82% and Gly-E by 78%. Postrefining mitigation, including the use of absorbents, enzymes, or rebleaching of the oil, has also been reported to produce desirable contaminant reduction. Postrefining treatment with calcinated zeolite was able to reduce the 3-MCPD-E concentration by 19% and the Gly-E concentration by 77%. Applying combined mitigation strategies to multiple steps of oil refining is likely crucial in order to adequately reduce levels of 3-MCPD-E and Gly-E.

This study aimed to obtain insights into the presence of cadmium, lead, mercury and arsenic in feed materials and feed over time for the purpose of guiding national monitoring. Data from the Dutch feed monitoring programme and from representatives of the feed industry during the period 2007-13 were used. Data covered a variety of feed materials and compound feeds in the Netherlands. Trends in the percentage of samples that exceeded the maximum limit (ML) set by the European Commission, and trends in average, median and 90th percentile concentrations of each of these elements were investigated. Based on the results, monitoring should focus on feed material of mineral origin, feed material of marine origin, especially fish meal, seaweed and algae, as well as feed additives belonging to the functional groups of (1) trace elements (notably cupric sulphate, zinc oxide and manganese oxide for arsenic) and (2) binders and anti-caking agents. Mycotoxin binders are a new group of feed additives that also need attention. For complementary feed it is important to make a proper distinction between mineral and non-mineral feed (lower ML). Forage crops in general do not need high priority in monitoring programmes, although for arsenic grass meal still needs attention.

Insects have potential as a novel source of protein in feed and food production in Europe, provided they can be used safely. To date, limited information is available on the safety of insects, and toxic elements are one of the potential hazards of concern. Therefore, we aimed to investigate the potential accumulation of cadmium, lead and arsenic in larvae of two insect species, Tenebrio molitor (yellow mealworm) and Hermetia illucens (black soldier fly), which seem to hold potential as a source of food or feed. An experiment was designed with 14 treatments, each in triplicate, per insect species. Twelve treatments used feed that was spiked with cadmium, lead or arsenic at 0.5, 1 and 2 times the respective maximum allowable levels (ML) in complete feed, as established by the European Commission (EC). Two of the 14 treatments consisted of controls, using non-spiked feed. All insects per container (replicate) were harvested when the first larva in that container had completed its larval stage. Development time, survival rates and fresh weights were similar over all treatments, except for development time and total live weight of the half of the maximum limit treatment for cadmium of the black soldier fly. Bioaccumulation (bioaccumulation factor > 1) was seen in all treatments (including two controls) for lead and cadmium in black soldier fly larvae, and for the three arsenic treatments in the yellow mealworm larvae. In the three cadmium treatments, concentrations of cadmium in black soldier fly larvae are higher than the current EC maximum limit for feed materials. The same was seen for the 1.0 and 2.0 ML treatments of arsenic in the yellow mealworm larvae. From this study, it can be concluded that if insects are used as feed materials, the maximum limits of these elements in complete feed should be revised per insect species.

Accurate and reliable quantification methods for gluten in food are necessary to ensure proper product labeling and thus safeguard the gluten sensitive consumer against exposure. Immunochemical detection is the method of choice, as it is sensitive, rapid and relatively easy to use. Although a wide range of detection kits are commercially available, there are still many difficulties in gluten detection that have not yet been overcome. This review gives an overview of the currently commercially available immunochemical detection methods, and discusses the problems that still exist in gluten detection in food. The largest problems are encountered in the extraction of gluten from food matrices, the choice of epitopes targeted by the detection method, and the use of a standardized reference material. By comparing the available techniques with the unmet needs in gluten detection, the possible benefit of a new multiplex immunoassay is investigated. This detection method would allow for the detection and quantification of multiple harmful gluten peptides at once and would, therefore, be a logical advancement in gluten detection in food.
