RESUMO

Aim: This study aimed to perform an in vitro comparative analysis of the antifungal activity of different calcium silicate-based endodontic sealers against three fungal species. Methods: The antifungal properties of three calcium silicate-based sealers were tested: Bio-C Sealer, Cambiar a Sealer Plus BC, and MTA-Fillapex. Two commonly used sealers were used as controls: AH Plus and Endomethasone. An agar diffusion test was performed to analyze the antifungal activity of the sealers against Candida albicans, Candida glabrata, Candida tropicalis, and a mixed microbial culture medium. The results were analyzed using ANOVA (p <0.05). Results: Endomethasone exhibited the highest inhibition against all strains examined, maintaining a consistent level of inhibition throughout 7 days. MTA-Fillapex demonstrated the best performance among the calcium silicate-based sealers for the three fungal species (p < 0.05), maintaining stable values over the 7 days, surpassing that of Endomethasone. Nevertheless, MTA-Fillapex only exhibited antimicrobial effect against the mixed culture for the first 24 hours, and no antimicrobial activity was observed at 48 hours, being surpassed by all tested sealers (p < 0.05). Conclusion: Of all silicate-based sealers tested, only MTA-Fillapex exhibited promising antifungal activity. Nevertheless, care must be taken when extrapolating these results, as MTA-Fillapex exhibited poor antimicrobial activity when tested in mixed microbial cultures

RESUMO

Calcium silicate-based cement (CSC) has attracted much interest because of its favourable osteogenic effect that supports its clinical use. Although CSC has antibacterial activity, this activity still needs to be improved when used in an infected bone defect. Natural polyphenols have been considered antimicrobial reagents. To this end, three different types of polyphenols (gallic acid (GA), pyrogallol (PG) and tannic acid (TA)) with different concentrations as a liquid phase were mixed with bioactive calcium silicate to enhance the antibacterial activity of CSC. The setting time, antibacterial activity, and osteogenic activity of CSC were studied. Evaluation of antibacterial ability and reactive oxygen species (ROS) was performed using Gram-negative Escherichia coli (E. coli) and Gram-positive Staphylococcus aureus (S. aureus) bacteria, while a human osteoblast-like cell line (MG63) was used to examine osteogenic activity. The experimental results showed that the addition of polyphenols did not remarkably affect the phase composition and morphology of CSC, but changed the setting time and diametral tensile strength. At the same concentration of 1 wt%, the setting time of TA (21 min) was significantly shorter than that of PG (26 min) and GA (68 min), and was indistinguishable from the control cement (20 min). GA had a significantly higher antioxidant activity than PG and TA. As expected, higher concentrations of polyphenols had a more positive impact on ROS generation. More importantly, the incorporation of polyphenols greatly enhanced the antibacterial activity of CSC against E. coli and S. aureus, but had little effect on the in vitro osteogenic activity of MG63 cells and the cytotoxicity of L929 cells. It was concluded that among the three phenolic compounds, the optimal concentration of the liquid phase in the hybrid cement was 5 wt% TA in terms of setting time, strength, antibacterial activity and in vitro osteogenic activity.

RESUMO

Abstract Cetrimide (CTR) is a cationic surfactant detergent with antimicrobial and antibiofilm activity. The aim of this study was to evaluate setting time, pH, solubility and antibiofilm activity of tricalcium silicate cement (TSC) with zirconium oxide (ZrO2) and its association with 0.2 and 0.4% cetrimide. Initial and final setting times (IST and FST) were assessed based on ISO-6876. pH was evaluated at periods of 1,3,7,14 and 21 days. Solubility was analyzed by weight loss. A modified direct contact test (MDCT) on the biofilm of Enterococcus faecalis formed on bovine root dentin blocks was performed, after 6 hours of manipulation and 15 hours of contact time. The analysis was performed by UFC mL ¯¹ counting. The data were analyzed by ANOVA and Tukey's tests (α=0.05). Higher IST was observed for TSC/ZrO2+CTR in both concentrations than for TSC/ZrO2 and lower FST for TSC/ZrO2+0.4% CTR (p0.05). All materials exhibited increased mass. TSC/ZrO2+CTR 0.4% had lower mass gain than the other materials (p<0.05). The highest antibiofilm activity was observed for TSC/ZrO2+CTR in both concentrations, when compared with the positive control (p<0.05). In conclusion, CTR exhibited potential to promote greater antibiofilm activity to tricalcium silicate cement, without harming its physicochemical properties of setting time, pH and solubility.

Introduction: Root perforation is a technical-surgical accident that results in communication of the pulp cavity with periodontal tissues. This type of accident may have an unfavorable prognosis because it significantly affects the floor of the coronary chamber or the cervical, middle and apical thirds of the root canal. Factors that interfere with the prognosis of root perforations include location, extension, presence or absence of periodontal pocket, time elapsed between perforation and treatment, and type of obturator material. In this context, bioceramic canal sealers have emerged as an innovative proposal for treatment of perforations previously considered to have an unfavorable prognosis. Objective: The aim of this study was to report the treatment of a lower molar root perforation with the EndoSequence BC Sealer bioceramic sealer after a 3-year follow up. Results: Radiographic images and patient clinical data showed success in the treatment of perforation with repair of the periradicular lesion. Conclusion: Endo-Sequence BC Sealer bioceramic endodontic sealer associated with the resumption of the original conduit path and its instrumentation favored the repair of periradicular tissues, suggesting promising biological properties of compatibility and bioactivity (AU).

Aim: To compare the microhardness of two silicate-based endodontic materials: MTA Densell Endo Sealer and Total Fill BC Sealer. Materials and methods: Samples of MTA Densell Endo Sealer (Group 1, n=5) and Total Fill BC Sealer (Group 2, n=5) contained into cow dentine cylinders were subjected to the Vickers Microhardness Indentation test at room temper- ature and 14 days after setting. Five indentations per sample were performed under a load of 100 g for 10 s. The indenta- tion measurements were converted into hardness values (HV). Mean HV values of each group were compared using the Stu- dent t test at a significance level of P <0,05. Results: 14 days after setting, the microhardness of MTA Densell Endo Sealer was significantly greater (P=0,001) than that of Total Fill BC Sealer. Conclusions: The results of this study suggest that due to its hardness the removal of MTA Densell Endo Sealer could be difficult to perform when a root canal retreatment is indicated (AU)

RESUMO

Introduction: This study aims to analyze the influence of particles size of sol-gel derived calcium silicate particles in the setting reaction of bioactive endodontic cements. Materials and Methods: Sol-gel derived calcium silicate particles were synthesized and sieved to separate the particles in different sizes: CS400, CS200, and CS100. A commercial MTA (Control) was used as control. The particle size and the specific surface area were assessed by laser diffraction and nitrogen adsorption. The cements were prepared with water as the liquid for the reaction. The setting time was conducted according to ISO 6876, and the setting kinetics was analyzed by Fourier trans-formed infrared spectroscopy (FTIR) at different time points between 120s to 72h. Results: The particle size varied from 9.45µm (CS400 ) to 31.01 (Control). The higher specific surface area valuer reached 15.14g/cm2 in the CS400. The smallest particle sizes, the higher specific surface area, and the lowest setting time were found for CS400 (p < 0.05). Control presented the highest setting time (p < 0.05). The FTIR analyses showed the differences in materials structure over time, with faster hydration and crystallization for CS400. The setting kinetics was slower for Control even when compared to a sol-gel derived group with similar particle size. Conclusion:The route of synthesis and the particle size influences the setting reaction of calcium silicate-based cements. The reduction of particle size for sol-gel derived calcium silicates lead to the acceleration of the setting reaction of the produced bioactive endodontic cement.

Biosilicate is a bioactive glass-ceramic used in medical and dental applications. This study evaluated novel reparative materials composed of pure tricalcium silicate (TCS), 30% zirconium oxide (ZrO2 ) and 10 or 20% biosilicate, in comparison with Biodentine. Setting time was evaluated based on ISO 6876 standard, radiopacity by radiographic analysis, solubility by mass loss, and pH by using a pH meter. Cytotoxicity was evaluated by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide and NR assays. Alkaline phosphatase (ALP) activity and alizarin red were used to evaluate cell bioactivity. Antimicrobial activity was assessed on Enterococcus faecalis by the direct contact test. The data were submitted to analysis of variance (ANOVA)/Tukey; Bonferroni and Kruskal-Wallis, and Dunn tests (α = 0.05). The association of Biosilicate with TCS + ZrO2 had appropriate setting time, radiopacity, and solubility, alkaline pH, and antimicrobial activity. TCS and Biodentine showed higher ALP activity in 14 days than the control (serum-free medium). All cements produced mineralized nodules. In conclusion, Biosilicate + TCS ZrO2 decreased the setting time and increased the radiopacity in comparison to TCS. Biosilicate + TCS ZrO2 presented lower solubility and higher radiopacity than Biodentine. In addition, these experimental cements promoted antimicrobial activity and mineralization nodules formation, suggesting their potential for clinical use.

(AU)

Aim: To compare the sealing capacity of mineral trioxide (MTA®) and tricalcium silicate (Biodentine®) for retrograde filling in single-root permanent teeth. Materials and methods: Thirty-eight permanent single-root teeth with complete apical closure were selected and randomly divided into two experimental groups (n = 15): Group 1: MTA®, and Group 2: Biodentine®, in addition to positive and negative controls. Root treatment was performed with the MTWO rotary system, and obturation was carried out by means of the continuous wave thermoplastic system (Beefill) using Grossman-type cement as a sealant. Apical end apicoectomy was performed followed by the preparation of standardized retro-apical cavities and the respective fillings with MTA ® and Biodentine® according to the study group. Subsequently, the specimens were subjected to an apical filtration process of Chinese ink and transparency, while the apical filtration was evaluated with a stereoscopic microscope. The data were analyzed with the Mann-Whitney U test. Results: Inferential analysis showed that Biodentine® had less microleakage than MTA® with a statistically significant difference of 95% (p <0.034). Conclusion: Biodentine® cement showed greater apical sealing capacity than MTA® cement in retrograde fillings of single-root teeth ex vivo. (AU)

RESUMO

Calcium silicate-based cements are known for their wide applications in dentistry and orthopedics. The alkaline pH (up to 12) of these cements limits their application in other orthopedic areas. In this study, the effect of dicalcium phosphate dihydrate (DCPD) coating on set cement on pH reduction and biocompatibility improvement was examined. Samples with 0 and 10 weight ratio DCPD were prepared and characterized by XRD, FTIR, and SEM. The DCPD coating on the set cement was performed by a 7 d immersion in 1% monocalcium phosphate (MCP) solution and characterized by XRD, FTIR, SEM, and EDX. Also, the compressive strength and cytotoxicity of the samples were tested. The results showed that DCPD coating did not significantly change the compressive strength of the cement, but by decreasing the pH of the culture medium to the physiological range, it led to enhance adhesion, spreading and proliferation of human osteosarcoma cell line (Saos-2). The novel DCPD coated calcium silicate-based cement could be served as a bulk or porous bone substitute and scaffold.

Objective: this study aimed to compare the dentin bond strength of a resin-modified light-curable calcium-silicate-based material (TheraCal LC®; Bisco, Schaumburg, IL, USA) with White MTA (WMTA®; Angelus, Londrina, PR, Brazil). Materials and Methods: sixteen human maxillary incisors and canines were selected and three 1-mm-discs were obtained from the middle third of each root. On the coronal surface of each disc, two 1.2-mm-wide-holes were drilled through the dentin. Then, artificial holes were filled with one of the tested materials: WMTA® and TheraCal LC®. The filled dental slices were stored in a phosphate-buffered saline (PBS) solution (pH 7.2) for 7 days at 37°C. After that, push-out assessment was performed with a 1.0-mm-plunger-tip. Load was applied at a crosshead speed of 0.5 mm/min until sealer displacement. The results were expressed in MPa. Mann-Whitney U test was applied to rank materials regarding dentin push-out bond strength. Significance level was set at a = 5%. Results: All specimens showed measurable results and no premature failure occurred. TheraCal LC® demonstrated superior push-out bond strength values to dentin when compared to WMTA® (P<0.0001). Conclusions: there is advantage of TheraCal LC® over WMTA® as regards to the push-out bond strength and, therefore it may be taken as a promising and innovative reparative material

RESUMO

The aim of this study was to evaluate the effect of different commercial silanes on microshear bond strength of resin cement to lithium disilicate ceramic. Twenty ceramic samples with 10 mm in length, 10 mm wide and 3 mm in thickness were fabricated, etched with 10% hydrofluoric acid for 20 seconds, and divided into 5 groups, according to the commercial silane applied: G1- RelyX Ceramic Primer (3M ESPE), G2- Angelus Silane, G3- Prosil (FGM), G4- Dentsply Silane (Dentsply) and G5- Bis-Silane (Bisco). Silanes were applied in accordance to manufacturers' recommendations. Addition silicone molds with 1 mm in thickness, 10 mm in diameter and 3 perforations with 1 mm in diameter each one, were placed on ceramic and filled with the resin cement RelyX ARC (3M ESPE) in all groups. Light activation was performed with LED Bluephase G2 (Ivoclar Vivadent) at 1200 mW/cm2, for 30 seconds. Samples were maintained in 100% humidity at 37°C for 24 hours and submitted to microshear test. The data (MPa) were submitted to one-way ANOVA and Tukey test (α=0.05). The results obtained for each group were: G1: 22.39±2.99; G2: 23.35±4.08; G3: 26.05±5.46; G4: 18.56±4.09; G5: 25.26±4.10. Statistical analysis showed significantly lower microshear bond strength for G4. Fracture pattern analysis showed predominance of adhesive failures in G1 and G2. G3 and G5 presented higher percentage of cohesive failures in ceramic, and G4 showed mixed, adhesive and cohesive fractures with similar percentages. It was concluded that different silanes showed influence on the lithium disilicate ceramic resin cement bond strength

The aim of the present study was to investigate the new silicate cement mineral trioxide aggregate (MTA Repair HP) with respect to its effect on the inflammation process involving the tooth and periodontal tissues. The composition of MTA Repair HP was supplemented with plasticizer agents which can have a negative effect on the modulation of tooth inflammation. The silicate-based material in question is widely used in regeneration of the pulp-dentin complex, treatment of perforations of various locations in the tooth, as well as in surgical treatment of the complications of periapical tissue. The improved bioceramic restorative cement can affect the expression of metalloproteinases MMP-2 and MMP-9 in monocytes/macrophages involved in modulation of inflammation and regenerative processes of the tooth and periodontal tissues. The novel aspect of the present study lies in the application of the model of THP-1 monocyte/macrophage and applying the biomaterial in direct contact with the cells. Hence, it provides a representation of clinical conditions with respect to regenerative pulp and periodontal treatment with the use of MTA Repair HP. A lack of macrophage activation (as measured with flow cytometry) was found. Moreover, the study identified a lack of expression stimulation of the studied metalloproteinases (with the use of Western blotting and fluorescent microscopy). Similarly, no increase in MMP-2 and MMP-9 concentration was found (measured by ELISA method) in vitro when incubated with MTA Repair HP. Based on the results it can be concluded that new MTA Repair HP does not increase the inflammatory response in monocytes/macrophages associated with the activity of the described enzymes. It can also be speculated that they do not affect the process of dentin regeneration in which MMP-2 and MMP-9 play significant roles.

The mechanical properties of the materials have made the technique of veneer cementation, so changing and so studied. This technique has been modifi ed due to several clinical studies; especially in investigations that have been carried out in this respect. The changes made in the cementation technique, have not been only for the substrate of the dental organ, but also for the chosen ceramic material. This confuses the clinician by modifying the cementing procedures. In recent years, as a result of the rapid progress in CAD/CAM technology, a large number of new ceramics and hybrid materials, have been introduced into the dental market. This is refl ected in benefi t to the patients, because we have more opportunities to off er diff erent kinds of restorative materials; and at the same time, their indications have expanded, but this, increases the diffi culty for clinicians and ceramists to stay up-to-date on material selection, and the most important in the correct adhesion protocols for the treatment of diff erent surfaces. This paper presents, a nine months clinical case report to evaluate the effi cacy of the new monocomponent conditioner, for etching and silanizing in a single step the Lithium Disilicate (LD), the new Monobond Etch & Prime (MBEP), contains in the same bottle suffi cient amount of conditioning acid and silane. This simplifi es the adhesion procedures by reducing, not only the number of steps but also the working time (AU)

RESUMO

ABSTRACT: The aim of the study was to evaluate the chemical composition and radiopacity of new calcium-silicatebased cements. Discs of 10 mm x 1 ± 0.1mm were prepared of BiodentineTM, TheraCal, Dycal and GC Fuji IX (n=5). The samples were radiographed directly on an PSP occlusal plate adjacent to an aluminium step wedge. The radiopacity of each specimen was determined according to ISO 9917/2007. Statistical analyses were carried out using ANOVA and Tukey's test at a significance level of 5 %. The chemical constitution of materials was determined by scanning electron microscopy (SEM) and energy dispersive x-ray element mapping. The radiopacities of the materials in decreasing order were: GC Fuji IX (3.45 ± 0.16 mm), Dycal (3.18 ± 0.17), BiodentineTM (2.79 ± 0.22), and TheraCal (2.17 ± 0.17). TheraCal showed the lowest radiopacity compared to the other materials, followed by BiodentineTM. Dycal and GC Fuji IX radiopacity values did not present significant statistical differences. Scanning electron microscopy and energy dispersive X-ray analysis revealed the presence of zirconium in BiodentineTM; and strontium, barium and zirconium in TheraCal as radiopacifying elements. The new calcium silicate cements present distinctive chemical composition. BiodentineTM contains zirconium as a radiopacifying element and has higher radiopacity values than TheraCal, which contains barium and strontium as radiopacifiers.

(AU)

Objective: The aim of this in vitro study was to evaluate the fracture resistance of two root reinforcement materials in mono-radicular teeth with endodontic treatment. Methods: Thirty freshly extracted human premolars were divided into three groups (n=10). The teeth in group 1 were endodontically treated without any root reinforcement and a resin restora-tion was placed in the coronal portion. In group 2 only the apical 5-mm portion of the root canal was filled with gutta-percha, and the remaining portion was filled with calcium silicate-based cement (BiodentineTM), which was used as root reinforcement material. In group 3 a fiber post was cemented as a root reinforce-ment material. f racture resistance (kn) was evaluated using a universal testing machine exerting a compression load at a velocity of 0.5 mm / min until frac-ture occurred. a one-way anova test and Tukey's post-hoc test were used to compare the mean root fracture resis-tance (kn). Results: The mean values and standard deviation of each group were as follows: group 1 gutta-percha (gp) 1.01 ± 0.19 Kn; group 2 biodentine (bd) 1.41 ± 0.25 Kn, and; group 3 fiber post (pfv) 1.27 ± 0.16 Kn. statistically significant differences were not confirmed between the Biodentine (BD) group and fiber post (PFV) group (p 0.179). Conclu-sion: BiodentineTM exhibited better root fracture resistance values compared to fiber post and gutta-percha. (AU)
