RESUMO

The effect of within-person variance on prevalence estimates from population distributions based on a single measurement was examined for hematologic and iron-status indicators collected in the Hispanic Health and Nutrition Examination Survey (HHANES). Within-person to between-person variance ratios (W:Bs) were estimated for 11 indicators by using data from 80 persons who provided blood twice in HHANES. Distributions of selected iron-status indicators from 20-44-y-old Mexican American females from HHANES were adjusted for within-person variance by using the W:B estimates, and prevalences of low values based on the original and adjusted distributions were compared. W:B were less than 1 for the majority of the indicators. Nonetheless, prevalences of low values from the original distributions were inflated when commonly used cutoff values were applied. Within-person variance in serum analytes needs to be controlled when the prevalence of a condition in populations is assessed.

RESUMO

The primary objective of this study was to determine whether it is the fish oil itself or the alpha-tocopherol that is added to the fish oil preparations (to prevent peroxidation) that is responsible for the beneficial effects of dietary supplementation with fish oil in patients with rheumatoid arthritis (RA). One group of RA patients took fish oil supplements and another group took alpha-tocopherol-enriched coconut oil supplements (placebo controls), both for 3 months. Clinical and laboratory indices of RA activity in relation to cellular and plasma vitamin E levels were assessed at the beginning and the end of the trial. The results of the study provide evidence that the beneficial effects of fish oil supplementation cannot be ascribed to the antioxidizing properties of the alpha-tocopherol per se.

The concentrations of ATP, ADP, AMP; NADP and NADPH; NAD and NADH were determined in erythrocytes from healthy newborns and compared with those obtained in healthy adults. No significant differences were found for the adenine nucleotide concentrations, but NADH levels were reduced in newborn erythrocytes, with a consequent increase in the NAD/NADH ratio. Moreover, in newborn erythrocytes increased levels of NADP were observed, with a consequent increase in the NADP/NADPH ratio and a decrease in the NAD/NADP ratio. These results indicate the need to use reference values of the ratios NAD/NADH, NADP/NADPH and NAD/NADP from healthy newborns in the study of syndromes affecting the metabolism of erythrocytes in the newborn.

Sixty-four male, healthy volunteers aged 35-45 y were randomly assigned to receive (as 1-g capsules) either 14 g fish-oil concentrate/d (55% n-3 fatty acids) or 14 g olive oil/d for 6 wk. Plasma fibrinogen was reduced by 13% and serum triglycerides by 22% after fish-oil supplementation ended. Three weeks after supplementation ended both variables were back to baseline values. An appreciable increase in the ratio of eicosapentaenoic acid to arachidonic acid (EPA:AA) in plasma eicosapentaenoic acid to arachidonic acid (EPA:AA) in plasma and red blood cell phospholipids occurred during the fish-oil intake. High-density-lipoprotein (HDL) cholesterol and HDL2 activity tended to be lowered by fish-oil supplementation. Systolic and diastolic blood pressures, serum cholesterol, gamma-glutamyltransferase, blood glucose, and monocyte low-density-lipoprotein receptor activity did not differ significantly between the two groups. The reduction in plasma fibrinogen concentration seems of special interest because this variable in several recent studies emerges as a separate cardiovascular risk factor with a high predictive value.

Se determinaron las concentraciones de sodio y potasio en plasma sanguíneo, saliva y eritrocitos en 3 grupos de pacientes. Grupo A (n=27) normotensos sin antecedentes familiares de hipertensión arterial y grupo C (n=35), individuos que padecían de hipertensión arterial. Se encontró un aumento estadísticamente significativo (p < 0,05) del sodio y una disminución del potasio intraeritrocitario, así como un aumento del sodio y potasio en la saliva en el grupo C con respecto a los demás grupos estudiados. No hubo diferencias significativas entre los grupos A y B en ninguno de los parámetros.

To investigate the claim that equine erythrocytes released from the spleen are older cells than those found at rest in the circulation, the 2,3, diphosphoglycerate (2,3 DPG), adenosine triphosphate (ATP) and creatine concentration of erythrocytes before and following splenic emptying were examined. Normal values for thoroughbreds (43) and ponies (10) at rest were established. Following either exercise or intravenous injection of adrenaline in six thoroughbreds, there was an increase in erythrocyte creatine content and a decrease in ATP concentration. Exercise produced a slight increase in 2,3 DPG while no change occurred with adrenaline. In two splenectomised ponies adrenaline only produced a decrease in erythrocyte ATP, with no change in creatine or 2,3 DPG content. It was concluded that erythrocytes released from the spleen are not aged cells. However, the stimuli used produced a decrease in ATP content which may affect the properties of the erythrocytes.

In the present study we aimed at evaluating the intracellular concentrations of magnesium, potassium and sodium in 50-year-old, otherwise healthy white men with never treated, essential hypertension (n = 12) and in normotensive control subjects (n = 12) matched for age, sex, race, height, weight and smoking habits. Intraerythrocyte magnesium was significantly increased in the hypertensive group (P less than .001) and correlated positively and significantly to blood pressure in the total group (P less than .01). The intracellular potassium to sodium ratio tended to be lower in the hypertensive group (P less than .05). Thus, the present study supports increased intracellular magnesium probably unrelated to intracellular potassium-sodium imbalance in never treated, essential hypertension.

Female rhesus monkeys were fed a commercial monkey diet and given selenium (Se) as either selenite or selenomethionine (SeMet) in the drinking water for 11 mo. Muscle and liver biopsies were taken initially and at the end of the experiment for determination of Se levels and glutathione peroxidase (GPX) activity. Blood was collected at monthly to bimonthly intervals, and the plasma and erythrocytes were subjected to gel filtration to determine the distribution of Se among proteins of various molecular weights. At the end of the experiment, there was significantly more Se in liver, muscle and hair from the monkeys given SeMet than in tissues from those given selenite, but there were no differences in liver or muscle GPX activity between the two treatment groups. The erythrocyte and plasma Se levels were significantly higher in the monkeys given SeMet than in those receiving selenite, but there were no differences in the GPX levels between these groups. About 68% of erythrocyte Se was associated with GPX in monkeys given selenite whereas only 34% was associated with GPX in those administered SeMet. The correlation coefficient for blood Se level and erythrocyte GPX activity was 0.92 in monkeys given selenite but only 0.37 in those given SeMet. Gel filtration of plasma revealed only one Se peak for plasma from the monkeys given selenite but at least two major Se peaks for plasma from monkeys receiving SeMet. The possible implications of these results for humans are discussed, including the reasons for poor correlations of GPX activity and blood Se levels.

A particle obtained by nuclease digestion of nucleohistone complexes prepared by direct mixing of histones with DNA in 0.15 M NaCl was indistinguishable by composition and physical properties from nucleosome cores prepared under the same conditions from nucleohistone preannealed in 0.6 M NaCl. We show here that different photo-cross-links form when these particles are prepared from H3 labeled with photoaffinity reagents on the unique histone H3 cysteine. H3-H3 histone dimers were dominant when the particles were prepared by dilution of the nucleohistone from 0.6 M NaCl while H3-H2B and H3-H4 histone dimers were prominent if the nucleohistone complex was prepared directly in 0.15 M NaCl. Peptide mapping of the novel H3-H4 and H3-H2B dimers showed that Cys-110 of histone H3 is cross-linked to the 18 amino acid C-terminal end of H4 or to the 66 amino acid C-terminal half of H2B.

Iron-depleted donors are at increased risk of developing anemia; if these donors could be identified by a screening test, iron supplementation or decreased donation frequency could be considered. Tests to determine serum ferritin, blood hemoglobin, and erythrocyte (Erc)-zinc protoporphyrin concentrations were examined in 679 consecutive female blood donors to identify donors with non-anemic iron deficiency. The test to determine serum ferritin is expensive and slow, whereas the two latter tests are rapid and less costly and could therefore be used for screening. Women in the fertile age groups had the lowest average serum ferritin values. In all, 93 women (13.7%) had depleted iron stores, as indicated by serum ferritin concentrations less than 14 micrograms/L. In these women, a much better correlation was found between Erc-zinc protoporphyrin and serum ferritin (rs = -0.49, P less than 0.001) than between blood hemoglobin and serum ferritin (rs = 0.31, P less than 0.01). These findings suggest that measurement of Erc-zinc protoporphyrin is superior to that of blood hemoglobin in identifying donors with non-anemic iron deficiency.

The association between breast cancer and selenium, measured in the diet, plasma, erythrocytes, and toenails, was investigated in a case-control study in the Netherlands during 1985-1987. Dietary selenium intake was assessed by a structured dietary history technique among 133 breast cancer cases and 238 population controls. Absolute and energy-adjusted selenium intake and selenium concentrations in plasma, erythrocytes, and toenails were similar in cases and controls. The multivariate adjusted odds ratios of breast cancer for subjects in the lowest quartile compared with the highest quartile were 1.6 (95% confidence interval (CI) 0.8-3.4) for dietary selenium, 2.0 (95% CI 0.9-4.4) for plasma selenium, 0.9 (95% CI 0.4-1.9) for erythrocyte selenium, and 1.1 (95% CI 0.6-2.1) for toenail selenium. No statistically significant trend was observed in the odds ratios for any of the four indicators of selenium. These results do not suggest a substantial association between selenium and breast cancer for both short- and long-term markers of selenium status.

We investigated the vitamin B-6 status in smokers, nonsmokers, and exsmokers by measuring both B-6 aldehyde vitamers, pyridoxal-5'-phosphate (PLP) and pyridoxal (PL), in the plasma as well as in the erythrocyte compartment. Two hundred eighty-six healthy, sedentary male workers from a middle-income group were investigated. There were 159 smokers, 59 exsmokers, and 68 nonsmokers. Plasma PLP and PL concentrations were significantly lower in smokers than in the nonsmokers and exsmokers whereas erythrocyte PLP and PL did not differ significantly between groups. Because PLP mainly functions as an intracellular coenzyme, the clinical significance of a depressed plasma PLP concentration alone is uncertain. It is concluded that circulating plasma PLP is labile and not necessarily indicative of intracellular PLP concentrations. The measurement of erythrocyte PLP and/or PL may be more informative about vitamin B-6 status than is plasma PLP alone.

We assessed quantitative aspects of blood salvage retrospectively in 270 consecutive coronary bypass patients over one year. They were assigned either to group A (n = 10, Cellsaver [Haemonetics]), B (n = 189, centrifugated oxygenator blood), C (n = 107, retransfusion of shed mediastinal blood) or D (n = 74, combination of method B and C). In addition, blood quality was studied prospectively in group A (n = 5), B (n = 10) and C (n = 10) and each group compared to a control group. Results (mean values/patient) were: Blood salvage with A yielded 570 +/- 230 ml (hematocrit = 50%), B 509 +/- 156 ml (69%) and C 593 +/- 430 ml (26%). The required homologous blood products, i.e. packed cells (PC) and fresh frozen plasma (FFP) diminished significantly when combined autologous blood salvage was used (PC 6.0 +/- 3.4 vs 3.4 +/- 1.9, p less than 0.05; FFP 3.4 +/- 3.8 vs 2.6 +/- 3.0). Autologous erythrocyte function assessed by 2,3DPG was normal in all three methods (range 14.77-16.03 mumol/gHb). ATP was nearly normal in A (3.34 +/- 0.45 mumol/gHb), reduced by 20% in B (3.21 +/- 1.14 mumol/gHb) and by 30% in C (2.56 +/- 0.78 mumol/gHb) compared to the corresponding preoperative patient value. Hemolysis (free plasma hemoglobin) (was elevated in A (63 +/- 7 mg/dl), B (202 +/- 57 mg/dl) and C (211 +/- 44 mg/dl). However, no increase of free plasma hemoglobin was encountered in our patients after retransfusion of either A, B or C. No side effects were detected and bacteriology remained negative in all examined blood samples in C.(ABSTRACT TRUNCATED AT 250 WORDS)

The RhD polypeptide and LW glycoprotein were separately immunopurified with monoclonal antibodies and compared by two-dimensional (2-D) iodopeptide mapping after digestion with alpha-chymotrypsin. These proteins have distinct 2-D maps, as seen after 125I-labeling tyrosine residues (chloramine-T procedure), and even more strikingly after labeling primary amine residues (Bolton-Hunter procedure). Of the more than 20 iodopeptides visualized, only five migrated identically when preparations of RhD and LW were directly compared, suggesting that RhD and LW are different proteins that may share some common protein domains. N-glycanase treatment of the iodopeptides did not modify the 2-D map of the RhD protein but greatly affected the LW map, further indicating that LW, but not RhD, carries N-linked carbohydrate chains. After deglycosylation the LW map was different from the RhD map, confirming that the RhD and LW polypeptides are different proteins. These findings demonstrate that LW is neither a glycosylated form of Rh protein nor is Rh a precursor of LW.

La técnica de Waaler-Rose donde se emplean eritrocitos de carnero formalinizados y sensibilizados con la fracción IgG de un antisuero de conejo anti-eritrocitos de carnero (WRECFS), se desarrolló sobre láminas portaobjetos, método propuesto por Milgrom y Tönder (1964) para la determinación del factor reumatoideo. Se compararon obtenidos mediante esta técnica y la prueba del Látex, en un grupo de pacientes con el diagnóstico de artritis reumatoide (AR) y en donantes de sangre sanos. El porcentaje de positividad de las técnicas ensayadas y de la prueba del Látex en pacientes con AR, fue de 47,7 y 50,8, respectivamente, y en donantes, de 0 y 1,5. Se encontró una correlación positiva y con coeficiente de correlación * = 0,88, p < 0,005 entre ambas pruebas, valores todos similares a los comunicados por otros autores

Hybrid hemoglobins, containing mesoheme in one type of subunit and protoheme in the partner subunits, have been studied by resonance Raman spectroscopy. These hybrids have been studied in both the met hybrid and fully reduced, deoxy forms. Judicious choice of laser excitation frequency permits selective enhancement of modes associated with each type of subunit; i.e., either meso- or protoheme-containing subunit. The assignments of low-frequency modes of meso- and protoheme are briefly discussed with special reference to the iron-histidine linkage. Despite functional differences between the hybrids, no significant changes in the strength of the iron-histidine linkages are detected by resonance Raman spectroscopy. These results are discussed with reference to recent high-resolution NMR studies of these same hybrids.

RESUMO

The influence of a new antitumor enzyme L-lysine alpha-oxidase on Lewis lung carcinoma spreading was studied in mice in which primary tumor had been removed. The enzyme was found to significantly decrease the extent and number of lung metastases as compared to mice which hadn't received L-lysine alpha-oxidase. This was matched by recovery of alveolar macrophages functional activity, as assessed by adenosine deaminase and 5' nucleotidase levels in these cells. Moreover, antimetastatic and cytostatic effect was confirmed by the measuring of polyamine concentration in mice erythrocytes.
