RESUMO

A profound concern in developing microbially-assisted phytoextraction is that introduced microbes not only remove heavy metals from contaminated soils but also enhance metal uptake into plant tissues from the treated soils. Cadmium (Cd) removal efficiencies were compared after leaching with deionized water (CK), acidified basal salts medium (acid control), cell-free spent medium (spent bioleaching) and mixotrophic acidophiles (two-step bioleaching). Two-step bioleaching using the mixotrophic acidophiles removed 34% of total Cd and 87% of available Cd, significantly more than CK (3% and 4%), acid control (12% and 51%) and spent bioleaching (26% and 75%). Pot experiments of water spinach growing in four treated soils were conducted to evaluate the Cd uptake performance in plants. Notably, the mixotrophic acidophiles increased Cd concentration in plant tissues by 78% compared to the CK group. More interestingly, the mixotrophic acidophiles were not colonized in soils but caused the compositional increase of indigenous microbes such as the genera of Alicyclobacillus, Clostridium sensu strict and Streptacidiphilus. Meanwhile, two-step bioleaching had little effects on soil structure and physicochemical properties determined by the spectroscopy characteristics analysis. These results implied that the mixotrophic acidophiles facilitated the development of microbially-assisted phytoextraction technology.

RESUMO

Spontaneous plants of Ipomoea acuminata ("morning glory") exhibiting white rust pustules were found in a field crop area of Planaltina, DF, in the fall season of 2010 and the disease causal agent was identified as Albugo ipomoea-panduratae (Oomycota). No reports of the association between A. ipomoea-panduratae and I. acuminata were known in Brazil previously to 2010. A reference specimen was deposited at the University of Brasilia Mycological Reference Collection.

Studies are presented on new types of anaerobic digesters in which chopped or dry crushed Ipomoea carnea was fed without any other pretreatment, in an attempt to develop commercially viable means of utilizing the otherwise very harmful plant. Two types of solid-feed anaerobic digesters (SFADs) were studied. The first type had a single vessel in which the bottom 35% portion was separated from the top portion by a perforated PVC disk. The weed was charged from the top and inoculated with anaerobically digested cowdung-water slurry. The fermentation of the weed in the reactor led to the formation of volatile fatty acids (VFAs) plus some biogas. The leachate, rich in the VFAs, was passed through the perforated PVC sheet and collected in the lower portion of the vessel. The other type of reactors had two vessels, the first one was fully charged with the weed and the second received the VFA leachate. With both types were attached upflow anaerobic filters (UAFs) which converted the leachate into combustible biogas consisting of approximately 70% methane. All SFADs developed very consistent performance in terms of biogas yield within 17 weeks of start. The two-compartment reactors yielded significantly more biogas than the single-compartment reactors of corresponding total volume, and the reactors with which anaerobic filters (AF) were attached yielded more biogas than the ones without AF. The best performing units generated 2.41m(3) of biogas per m(3) of digester volume, as compared to 0.1-0.2m(3) of biogas, m(-3)d(-1), obtainable with conventional digesters. This indicates the viability of this technology. The spent weed can be vermicomposted directly to obtain good soil-conditioner cum fertilizer; earthworm Eudrilus eugeniae produced 540mg vermicast per animal every day, achieving near total conversion of feed to vermicast in 20 days. The proposed systems, thus, makes it possible to accomplish total utilization of ipomoea.

OBJECTIVE: To identify the level of contamination with thermotolerant coliforms (ThC), intestinal helminth eggs and protozoan parasites in water spinach (Ipomoea aquatica) cultivated in a wastewater-fed lake in Phnom Penh, Cambodia. METHODS: The investigation was carried out from July 2004 to May 2005 in Boeng Cheung Ek lake, a main recipient of wastewater from the city of Phnom Penh. Water spinach samples were collected near two wastewater entry points in the northern part of the lake and near a wastewater exit point in the southern part of the lake, where water is discharged from the lake into a stream. Water spinach samples were harvested at each of the three locations with and without wastewater contact according to the normal practices of farmers, and analysed for ThC, protozoan parasites (Giardia, Cryptosporidium and Cyclospora), and helminth eggs (Ascaris lumbricoides, Trichuris trichura and hookworm) using standard methods. Wastewater samples were also collected at each of the three locations and analysed for ThC and helminth eggs. RESULTS: High concentrations of ThC (approximately 10(5)-10(7)/g) were found in water spinach samples. ThC mean counts did not differ significantly, neither between sample types at each location, nor between locations. ThC mean concentration of water spinach samples collected in the rainy season of 2005 was higher than that in the dry season of 2004. Protozoan parasites were detected in the samples from all three locations and in water spinach harvested with and without wastewater contact. Giardia spp. were present in 56% (20) of samples with an average cyst concentration of 6.6/g. Mean counts of Giardia were not significantly associated with wastewater contact status of water spinach. Cryptosporidium oocysts were found in 17% (six) of samples with an average concentration of 0.5 oocyst per gram of water spinach. Cyclospora oocysts were detected in 8% (three) of samples. Four (11%) water spinach samples contained helminth eggs (average concentration: 0.1 egg per gram). Water samples contained approximately 10(4)-10(7) ThC per 100 ml and a mean of 27.4 helminth eggs per litre. There was a 2-3 log reduction of ThC between the water samples collected at the wastewater entry and exit point(s) of the lake. CONCLUSIONS: Water spinach cultivated in the lake was highly contaminated with feces, as indicated by high ThC concentrations and the presence of protozoan parasites. A reduction in ThC numbers almost to WHO guideline levels for irrigation water occurred through natural biological and physical processes in the lake, as shown by differences in ThC counts at wastewater entry and exit points.
