RESUMO

Kelp forests (KFs) are one of the most significant marine ecosystems in the planet. They serve as a refuge for a wide variety of marine species of ecological and economic importance. Additionally, they aid with carbon sequestration, safeguard the coastline, and maintain water quality. Microplastic (MP) and polybrominated diphenyl ethers (PBDEs) concentrations were analyzed across trophic levels in KFs around Todos Santos Bay. Spatial variation patterns were compared at three sites in 2021 and temporal change at Todos Santos Island (TSI) in 2021 and 2022. We analyzed these MPs and PBDEs in water, primary producers (Macrocystis pyrifera), grazers (Strongylocentrotus purpuratus), predators (Semicossyphus pulcher), and kelp detritus. MPs were identified in all samples (11 synthetic and 1 semisynthetic polymer) and confirmed using Fourier-transform infrared microspectroscopy-attenuated total reflectance (µ-FTIR-ATR). The most abundant type of MP is polyester fibers. Statistically significant variations in MP concentration were found only in kelps, with the greatest average concentrations in medium-depth kelps from TSI in 2022 (0.73 ± 0.58 MP g-1 ww) and in the kelp detritus from TSI in 2021 (0.96 ± 0.64 MP g-1 ww). Similarly, PBDEs were found in all samples, with the largest concentration found in sea urchins from Punta San Miguel (0.93 ± 0.24 ng g-1 ww). The similarity of the polymers can indicate a trophic transfer of MPs. This study shows the extensive presence of MP and PBDE subtropical trophic web of a KF, but correlating these compounds in environmental samples is highly complex, influenced by numerous factors that could affect their presence and behavior. However, this suggests that there is a potential risk to the systems and the services that KFs offer.

RESUMO

Experiments conducted on triple-negative breast cancer have shown that fucoidan from Lessonia trabeculata (FLt) exhibits cytotoxic and antitumor properties. However, further research is necessary to gain a complete understanding of its bioactivity and level of cytotoxicity. The cytotoxic effect of FLt was determined by the 2,5-diphenyl-2H-tetrazolium bromide (MTT) assay. Apoptosis was analyzed using annexin V and caspase 3/7 staining kit and DNA fragmentation. In addition, transcriptional expression of antiapoptotic (Bcl-2 and XIAP) and proapoptotic (caspase 8, caspase 9, and AIF) genes were analyzed in TNBC 4T1 cells. After 72 h of culture, the IC50 for FLt was 561 µg/mL, while doxorubicin (Dox) had an IC50 of 0.04 µg/mL. In addition, assays for FLt + Dox were performed. Annexin V and caspase 3/7 revealed that FLt induces early and late-stage apoptosis. DNA fragmentation results support necrotic death of 4T1 cells. Similarly, transcripts that prevent cell death were decreased, while transcripts that promote cell death were increased. This study showed that FLt induces apoptosis by both caspase-dependent and caspase-independent mechanisms. These findings suggest that FLt may have potential applications in breast cancer treatment. Further research will provide more information to elucidate the mechanism of action of FLt.

RESUMO

We previously demonstrated the beneficial effects of U.S.-grown sugar kelp (Saccharina latissima), a brown seaweed, on reducing serum triglycerides (TG) and total cholesterol (TC) and protecting against inflammation and fibrosis in the adipose tissue of diet-induced obesity mice. In this current study, we aimed to explore whether the dietary consumption of sugar kelp can prevent atherosclerosis using low-density lipoprotein receptor knockout (Ldlr KO) mice fed an atherogenic diet. Eight-week-old male Ldlr KO mice were fed either an atherogenic high-fat/high-cholesterol control (HF/HC) diet or a HF/HC diet supplemented with 6% (w/w) sugar kelp (HF/HC-SK) for 16 weeks. Consumption of sugar kelp significantly increased the body weight gain without altering fat mass and lean mass. Also, there were no significant differences in energy expenditure and physical activities between the groups. The two groups did not show significant differences in serum and hepatic TG and TC levels or the hepatic expression of genes involved in cholesterol and lipid metabolism. Although serum alanine aminotransferase (ALT) activity did not differ significantly between the two groups, there were significant increases in the expression of macrophage markers, including adhesion G protein-coupled receptor E1 and cluster of differentiation 68, as well as tumor necrosis factor alpha in the HF/HC-SK group compared to the HF/HC mice. The consumption of sugar kelp did not elicit a significant effect on the development of aortic lesions. Moreover, lipopolysaccharide-stimulated splenocytes isolated from HF/HC-SK-fed mice showed no significant changes in the mRNA levels of pro-inflammatory genes compared with those from the HF/HC mice. In summary, the consumption of dietary sugar kelp did not elicit anti-atherogenic and hepatoprotective effects in Ldlr KO mice.

RESUMO

Obesity requires treatment to mitigate the potential development of further metabolic disorders, including diabetes, hyperlipidemia, tumor growth, and non-alcoholic fatty liver disease. We investigated the anti-obesity effect of a 30% ethanol extract of Eisenia bicyclis (Kjellman) Setchell (EEB) on 3T3-L1 preadipocytes and high-fat diet (HFD)-induced obese C57BL/6 mice. Adipogenesis transcription factors including peroxisome proliferator-activated receptor (PPAR)γ, CCAAT/enhancer-binding protein-alpha (C/EBPα), and sterol regulatory element-binding protein-1 (SREBP-1) were ameliorated through the AMP-activated protein kinase (AMPK) pathway by EEB treatment in differentiated 3T3-L1 cells. EEB attenuated mitotic clonal expansion by upregulating cyclin-dependent kinase inhibitors (CDKIs) while downregulating cyclins and CDKs. In HFD-fed mice, EEB significantly decreased the total body weight, fat tissue weight, and fat in the tissue. The protein expression of PPARγ, C/EBPα, and SREBP-1 was increased in the subcutaneous fat and liver tissues, while EEB decreased the expression levels of these transcription factors. EEB also inhibited lipogenesis by downregulating acetyl-CoA carboxylase (ACC) and fatty acid synthase (FAS) expression in the subcutaneous fat and liver tissues. Moreover, the phosphorylation of AMPK and ACC was downregulated in the HFD-induced mouse group, whereas the administration of EEB improved AMPK and ACC phosphorylation; thus, EEB treatment may be related to the AMPK pathway. Histological analysis showed that EEB reduced the adipocyte size and fat accumulation in subcutaneous fat and liver tissues, respectively. EEB promotes thermogenesis in brown adipose tissue and improves insulin and leptin levels and blood lipid profiles. Our results suggest that EEB could be used as a potential agent to prevent obesity.

RESUMO

Fucoidan, a sulphated polysaccharide, is found exclusively in brown seaweeds and has been reported to possess a wide range of biological functionalities. Fucoidans are found within the cell wall of brown seaweeds, which is composed of recalcitrant cellulose and hemicellulose. This hampers the recovery of fucoidans. In addition, fucoidans are found within a network of viscous hydrocolloids, alginates, further complicating their recovery. Traditionally, the hot water extraction method is used to recover fucoidans from brown seaweed, however, this is characterized by low yields and long extraction time. To combat these issues, several novel extraction technologies have been introduced, these include ultrasound-assisted extraction and enzyme-assisted extraction. Thus, the main aim of this study was to investigate and optimize fucoidan recovery from Ecklonia maxima based on ultrasound-assisted enzymatic extraction. The impact of temperature (40-65 °C), ultrasound intensity (0-118 W·cm-2), enzyme dosage (0-0.05 ml·g-1) and pH (4.5-6) on total dissolved, total carbohydrates and inorganic sulphates yields was studied. The application of ultrasound-assisted enzymatic extraction mainly improved the extraction of total carbohydrates. Ultrasound significantly improves the kinetics and extraction of fucoidan, but there was no merit when it was applied with enzymes. Results reveal that at optimized conditions, the fucoidan extracted 79.13 mg⋅g-1 (7.9 % w/w) of algal dry weight. The present study provides insight into the extraction potentials of enzyme-assisted extraction, ultrasound-assisted extraction, and ultrasound-assisted enzymatic extraction.

RESUMO

The fisheries history of the Giant Sea Bass, Stereolepis gigas (Telostei: Polyprionidae), is closely linked to its spatial ecology. Its overharvest is directly associated with formation of spatially distinct spawning aggregations during summer, while its subsequent recovery is hypothesized to be the result of spatially explicit gear restrictions. Understanding the spatial ecology of Giant Sea Bass is a key part of efforts to assess contemporary threats such as commercial harvest and incidental catch by recreational fisheries. In this study, we used acoustic telemetry to characterize Giant Sea Bass space use in the La Jolla kelp forest using an acoustic array that encompasses two marine protected areas (MPAs) and heavily trafficked recreational fishing grounds. Five of the seven fish we tagged remained in the La Jolla array for at least 6 months. Two fish were resident across multiple years, with one fish consistently detected for 4 years. Only one fish was detected in the broader network of regional acoustic receivers, moving north approximately 8 km to Del Mar. Most tagged fish had home ranges and core use areas indicating they spend considerable time outside MPAs, particularly in areas with high recreational fishing activity. During spawning season we detected fish less frequently in the La Jolla array and recorded higher movement rates. While the current MPA network in La Jolla by no means offers complete protection to this fish, it does appear to support long-term persistence of some individuals in a region of exceptionally high recreational fishing pressure.

RESUMO

Carbon dioxide removal (CDR) and emissions reduction are essential to alleviate climate change. Ocean macroalgal afforestation (OMA) is a CDR method already undergoing field trials where nearshore kelps, on rafts, are purposefully grown offshore at scale. Dissolved iron (dFe) supply often limits oceanic phytoplankton growth, however this potentially rate-limiting factor is being overlooked in OMA discussions. Here, we determine the limiting dFe concentrations for growth and key physiological functions of a representative kelp species, Macrocystis pyrifera, considered as a promising candidate for OMA. dFe additions to oceanic seawater ranging 0.01-20.2 nM Fe' ‒ Fe' being the sum of dissolved inorganic Fe(III) species ‒ result in impaired physiological functions and kelp mortality. Kelp growth cannot be sustained at oceanic dFe concentrations, which are 1000-fold lower than required by M. pyrifera. OMA may require additional perturbation of offshore waters via dFe fertilisation.

RESUMO

To assess the level of oxidative stress, expression of immune-related genes, histomorphology, and changes in the intestinal tract of hybrid snakeheads(Channa maculata ♀ × Channa argus ♂) under stress from kelp powder in place of flour against Aeromonas hydrophila. We set up experimental diets: a control (C) diet of 20% flour, an experimental (MR) diet of 10% kelp powder and 10% flour, and an experimental (FR) diet of 0% starch and 15% kelp powder. The experimental fish in each group were infected with Aeromonas hydrophila after 60 days of feeding. For this experiment, some of the experimental fish in group C were injected with PBS as a negative control group (PBS). The results showed that the C group had significantly higher SOD, CAT, and T-AOC activity and expression of TAK1, IKKß, IL-1ß, and TNF-α genes in the MyD88 pathway than the PBS group. CAT activity and the expression of TAK1, IL-1ß and TNF-α genes in the MyD88 pathway were significantly lower in the MR group than in the C group. Furthermore, the number of goblet cells in the MR group was significantly higher than in the C group. Furthermore, microorganisms such as Bacteroidota and Actinobacteriota were significantly lower in the C group than in the PBS and FR groups, as were beneficial bacteria such as Clostridium_sensu_stricto_1 and Sphingomonas. Replacing flour with kelp powder increases hybrid snakehead gut resistance to Aeromonas hydrophila.

RESUMO

This study evaluated the efficacy of Monascus purpureus fermentation on Saccharina japonica (SJ). Healthy substances and antioxidant activity of fermented SJ (FSJ) were determined. Results showed that fermentation caused the release of phenolic compounds and flavonoids, which resulted in the enhancement of antioxidant activity. Essential amino acids and γ-aminobutyric acid also greatly accumulated in FSJ. Sensory evaluation and gas chromatography-ion mobility spectrometry (GC-IMS) were used to evaluate flavor properties of FSJ. A lexicon consisted of 24 descriptors was established for SJ and FSJ, of which 14 descriptors were regarded as odor attributes. A total of 46 volatile compounds were identified by GC-IMS and showed positive correlation with odor attributes. Fifteen volatile compounds were screened as key compounds, tricarboxylic acid cycle, embden-meyerhof-parnas and amino acid catabolism were main formation metabolisms of them. Advanced properties of FSJ indicated that fermentation is a promising approach for the production of SJ food.

RESUMO

Cystophora is the second largest genus of fucoids worldwide and, like many other forest-forming macroalgae, is increasingly threatened by a range of anthropogenic impacts including ocean warming. Yet, limited ecological information is available from the warm portion of their range (SW Western Australia), where severe range contractions are predicted to occur. Here, we provide the first insights on the abundance, diversity, productivity, and stand structure of Cystophora forests in this region. Forests were ubiquitous over more than 800 km of coastline and dominated sheltered and moderately-exposed reefs. Stand biomass and productivity were similar or greater than that of kelp forests in the temperate reef communities examined, suggesting that Cystophora spp. play a similarly important ecological role. The stand structure of Cystophora forests was, however, different than those of kelp forests, with most stands featuring an abundant bank of sub-canopy juveniles and only a few plants forming the canopy layer. Stand productivity followed an opposite seasonal pattern than that of kelps, with maximal growth in late autumn through early winter and net biomass loss in summer. Annually, stands contributed between 2.2 and 5.7 kg · m-2 (fresh biomass) to reef productivity depending on the dominant stand species. We propose that Cystophora forests play an important and unique role in supporting subtidal temperate diversity and productivity throughout temperate Australia, and urge a better understanding of their ecology and responses to anthropogenic threats.

RESUMO

The potential contribution of kelp forests to blue carbon sinks is currently of great interest but interspecific variance has received no attention. In the temperate Northeast Atlantic, kelp forest composition is changing due to climate-driven poleward range shifts of cold temperate Laminaria digitata and Laminaria hyperborea and warm temperate Laminaria ochroleuca. To understand how this might affect the carbon sequestration potential (CSP) of this ecosystem, we quantified interspecific differences in carbon export and decomposition alongside changes in detrital photosynthesis and biochemistry. We found that while warm temperate kelp exports up to 71% more carbon per plant, it decomposes up to 155% faster than its boreal congeners. Elemental stoichiometry and polyphenolic content cannot fully explain faster carbon turnover, which may be attributable to contrasting tissue toughness or unknown biochemical and structural defenses. Faster decomposition causes the detrital photosynthetic apparatus of L. ochroleuca to be overwhelmed 20 days after export and lose integrity after 36 days, while detritus of cold temperate species maintains carbon assimilation. Depending on the photoenvironment, detrital photosynthesis could further exacerbate interspecific differences in decomposition via a potential positive feedback loop. Through compositional change such as the predicted prevalence of L. ochroleuca, ocean warming may therefore reduce the CSP of such temperate marine forests.

RESUMO

A Gram-stain-positive, aerobic, non-sporulating, yellow-pigmented and rod or cocci-shaped bacterium, designated Arc0846-15T, was isolated from the kelp Laminaria japonica. Strain Arc0846-15T was found to grow at 16-35 °C (optimum, 28 °C), at pH 6.0-9.5 (optimum, 7.0) and in the presence of 0-6 % (w/v) NaCl (optimum, 2 %). Cells were positive for catalase and negative for oxidase activity. Phylogenetic analyses, based on 16S rRNA gene sequence comparisons, revealed that the nearest phylogenetic neighbour strains of strain Arc0846-15T were Ornithinimicrobium murale 01 Gi-040T (96.2 %), Ornithinimicrobium kibberense K22-20T (96.1 %) and Ornithinimicrobium humiphilum HKI 0124T (95.2 %). Based on phylogenomic analysis, the average nucleotide identity values between strain Arc0846-15T and the neighbour strains were 69.8, 69.7 and 69.8 %, respectively; the digital DNA-DNA hybridization values between strain Arc0846-15T and its three closest neighbour strains were 18.8, 19.1 and 19.3 %, respectively. The predominant menaquinone was MK-8 (H4). The dominant cellular fatty acids were C17 : 1 ω8c, iso-C15 : 0, iso-C16 : 0 and C17 : 0. The polar lipids contained diphosphatidylglycerol, phosphatidylglycerol, phosphatidylinositol, glycolipid, one unidentified aminolipid and four unidentified lipids. The DNA G+C content of strain Arc0846-15T was 61.6 mol% based on the whole genome sequence. Based on the phylogenetic and phenotypic characteristics, strain Arc0846-15T is considered to represent a novel species of the genus Ornithinimicrobium, for which the name Ornithinimicrobium laminariae sp. nov. is proposed, with Arc0846-15T (=KCTC 49655T=MCCC 1K06093T) as the type strain.

RESUMO

The anti-amnesic effect of a mixture (4:6 = phlorotannin:fucoidan from Ecklonia cava, P4F6) was evaluated on amyloid-beta peptide (Aß)-induced cognitive deficit mice. The cognitive function was examined by Y-maze, passive avoidance, and Morris water maze tests, and the intake of the mixture (P4F6) showed an ameliorating effect on Aß-induced learning and memory impairment. After the behavioral tests, superoxide dismutase (SOD) activity and thiobarbituric acid-reactive substances (TBARS) contents were confirmed in brain tissue, and in the results, the mixture (P4F6) attenuated Aß-induced oxidative stress. In addition, mitochondrial activity was evaluated by mitochondrial reactive oxygen species (ROS) content, mitochondrial membrane potential (MMP), adenosine triphosphate (ATP) content, and mitochondria-mediated apoptotic signaling pathway, and the mixture (P4F6) enhanced mitochondrial function. Furthermore, the mixture (P4F6) effectively regulated tau hyperphosphorylation by regulating the protein kinase B (Akt) pathway, and promoted brain-derived neurotrophic factor (BDNF) in brain tissue. Moreover, in the cholinergic system, the mixture (P4F6) ameliorated acetylcholine (ACh) content by regulating acetylcholinesterase (AChE) activity and choline acetyltransferase (ChAT) expression in brain tissue. Based on these results, we suggest that this mixture of phlorotannin and fucoidan (P4F6) might be a substance for improving cognitive function by effectively regulating cognition-related molecules.

RESUMO

One of the well-known causes of hearing loss is noise. Approximately 31.1% of Americans between the ages of 20 and 69 years (61.1 million people) have high-frequency hearing loss associated with noise exposure. In addition, recurrent noise exposure can accelerate age-related hearing loss. Phlorofucofuroeckol A (PFF-A) and dieckol, polyphenols extracted from the brown alga Ecklonia cava, are potent antioxidant agents. In this study, we investigated the effect of PFF-A and dieckol on the consequences of noise exposure in mice. In 1,1-diphenyl-2-picrylhydrazyl assay, dieckol and PFF-A both showed significant radical-scavenging activity. The mice were exposed to 115 dB SPL of noise one single time for 2 h. Auditory brainstem response(ABR) threshold shifts 4 h after 4 kHz noise exposure in mice that received dieckol were significantly lower than those in the saline with noise group. The high-PFF-A group showed a lower threshold shift at click and 16 kHz 1 day after noise exposure than the control group. The high-PFF-A group also showed higher hair cell survival than in the control at 3 days after exposure in the apical turn. These results suggest that noise-induced hair cell damage in cochlear and the ABR threshold shift can be alleviated by dieckol and PFF-A in the mouse. Derivatives of these compounds may be applied to individuals who are inevitably exposed to noise, contributing to the prevention of noise-induced hearing loss with a low probability of adverse effects.

RESUMO

Nonalcoholic steatohepatitis (NASH), closely associated with obesity, is a health concern worldwide. We investigated whether the consumption of U.S.-grown sugar kelp (Saccharina latissima), an edible brown alga, can prevent obesity-associated metabolic disturbances and NASH in a mouse model of diet-induced NASH. Male C57BL/6J mice were fed a low-fat diet, a high-fat/high-sucrose/high-cholesterol diet (HF), or a HF diet containing sugar kelp (HF-Kelp) for 14 weeks. HF-Kelp group showed lower body weight with increased O2 consumption, CO2 production, physical activity, and energy expenditure compared with the HF. In the liver, there were significant decreases in weight, triglycerides, total cholesterol, and steatosis with HF-Kelp. The HF-Kelp group decreased hepatic expression of a macrophage marker adhesion G protein-coupled receptor E1 (Adgre1) and an M1 macrophage marker integrin alpha x (Itgax). HF-Kelp group also exhibited decreased liver fibrosis, as evidenced by less expression of fibrogenic genes and collagen accumulation than those of HF group. In epididymal white adipose tissue (eWAT), HF-Kelp group exhibited decreases in eWAT weight and adipocyte size compared with those of the HF. HF-Kelp group showed decreased expression of collagen type VI alpha 1 chain, Adgre1, Itgax, and tumor necrosis factor α in eWAT. We demonstrated, for the first time, that the consumption of U.S-grown sugar kelp prevented the development of obesity and its associated metabolic disturbances, steatosis, inflammation, and fibrosis in the liver and eWAT of a diet-induced NASH mouse model.

RESUMO

Heavy metals (HMs) and polycyclic aromatic hydrocarbons (PAHs) are persistent toxicants in coastal environments. Notably, in comparison to individual metal toxicity, knowledge about the effects of HMs and PAHs mixtures on kelps remains scarce. Accordingly, we performed in vitro experiments to determine the individual and combined effects of Cu, Cd, and PAHs on spore release, settlement, and germination on Macrocystis pyrifera and Lessonia spicata, two key-habitat forming kelp species of the coast of the Valparaíso Region in Chile. This region concentrates highly polluting industries, mainly due to unrestrained mining and fossil-fuel energy production. Single Cu, Cd, and PAHs treatments included concentrations in the ranges 5-200, 0.125-2000, and 0.05-100 µg/L, respectively, and a toxic-free treatment. Cu, Cd, and PAHs concentrations causing 20-50% (IC20, IC50) arrested spore release, settlement, and germination were determined, and the results shown in both species that single Cu, Cd, and PAHs IC20 values were generally lower on spore release than on spore settlement and germination, probably due to the absence of a cell wall in spores compared to later stages. Binary equitoxic IC20s mixture treatments changed from an antagonistic response to another with a greater inhibitory effect on spore release, from hour 1 to 7, whereas in IC50 treatments, the response was always antagonistic. The tertiary IC20 mixture of Cu+Cd+PAHs produced generally an antagonistic effect. Remarkably, all IC20 equitoxic mixture treatments showed a synergistic response on spore settlement in both kelps, suggesting that these toxicants are extremely harmful to kelp population persistence near highly polluted sites.

RESUMO

Phlorotannins are a group of major polyphenol secondary metabolites found only in brown algae and are known for their bioactivities and multiple health benefits. However, they can be oxidized due to external factors and their bioavailability is low due to their low water solubility. In this study, the potential of utilizing nanoencapsulation with polyvinylpyrrolidone (PVP) to improve various activities of phlorotannins was explored. Phlorotannins encapsulated by PVP nanoparticles (PPNPS) with different loading ratios were prepared for characterization. Then, the PPNPS were evaluated for in vitro controlled release of phlorotannin, toxicity and antioxidant activities at the ratio of phlorotannin to PVP 1:8. The results indicated that the PPNPS showed a slow and sustained kinetic release of phlorotannin in simulated gastrointestinal fluids, they were non-toxic to HaCaT keratinocytes and they could reduce the generation of endogenous reactive oxygen species (ROS). Therefore, PPNPS have the potential to be a useful platform for the utilization of phlorotannin in both pharmaceutical and cosmetics industries.

RESUMO

In the present study, effects of riboflavin (RF) and kelp polyphenol extracts (KPE) on mackerel (Scomberomorus Niphonius) myofibrillar protein (MP) gel were studied with or without ultraviolet A (UVA) irradiation treatment. The gel strength was increased with the addition of RF and KPE under UVA irradiation. Analysis of the proteins in the gel indicated that the carbonyl content increased, while the contents of total sulfhydryl and amino groups decreased. The proteins appeared to have no α-helix structures, and the endogenous tryptophan content appeared to decrease. The results of SDS-PAGE indicated that the RF and KPE treated samples under UVA irradiation showed massive MP cross-linking by covalent bonds. Electron spin resonance (ESR) results indicated that UVA irradiation generated free radicals in RF and KPE, which ultimately led to an improvement in MP gel properties. It also indicated that KPE could prevent the occurrence of peroxidation to improve the gel properties.

RESUMO

The subtidal kelp Nereocystis luetkeana (hereafter Nereocystis) maintains an upright stature by producing a single gas-filled float (pneumatocyst) that provides buoyancy. The ability of Nereocystis pneumatocysts to inflate with gas underwater is peculiar, and the gas composition of pneumatocysts has been the topic of several studies over the last 100 years. Past studies of pneumatocyst gases only examined large sporophytes, leaving open questions about the origins of these gases and how gas composition may change during development. In this study, we use developmental time as a means to understand the origin and physiological mechanisms that give rise to different gases within Nereocystis pneumatocysts. Total gas composition was measured across a range of pneumatocyst sizes (5-725 mL). Contrary to previous studies that documented pneumatocyst gas concentrations to be similar to air, this study found internal gas levels of CO, CO2 , and O2 to be 1.2 ± 0.8%, 0.6 ± 0.2%, and 59.9 ± 13.6%, respectively. Our data suggest that the composition of gases does not change as pneumatocysts grow and that the rate of each gas added is approximately proportional to changes in pneumatocyst volume. Therefore, cells constituting the pneumatocyst wall are likely producing more gas (per surface area) to fill pneumatocysts as they expand, maintaining proportional gas composition and corresponding internal pressure.

RESUMO

Nontargeted analysis of food safety requires selective removal of interference matrices and highly efficient recovery of chemical hazards. Porous materials such as covalent organic frameworks (COFs) show great promise in selective adsorption of matrix molecules via size selectivity. Considering the complexity of interference matrices, we prepared crystalline heteropore COFs whose two kinds of pores have comparable sizes to those of several common phytochromes, main interference matrices in vegetable sample analysis. By controlling the growth of COFs on the surface of Fe3O4 nanoparticles or by utilizing a facile co-electrospinning method, heteropore COF-based magnetic nanospheres or electrospun nanofiber films were prepared, respectively. Both the nanospheres and the films maintain the dual-pore structures of COFs and show good stability and excellent reusability. Via simple magnetic separation or immersion operation, respectively, they were successfully used for the complete removal of phytochromes and highly efficient recovery of 15 pesticides from the extracts of four vegetable samples, and the recoveries are in the range of 83.10-114.00 and 60.52-107.35%, respectively. Film-based immersion operation gives better sample pretreatment performance than the film-based filtration one. This work highlights the great application potentials of heteropore COFs in sample pretreatment for nontargeted analysis, thus opening up a new way to achieve high-performance sample preparation in many fields such as food safety analysis, environment monitoring, and so on.
