RESUMO

Deep learning is currently being used to automate surface defect detection in aluminum. The common target detection models based on neural networks often have a large number of parameters and a slow detection speed, which is not conducive to real-time detection. Therefore, this paper proposes a lightweight aluminum surface defect detection model, M2-BL-YOLOv4, based on the YOLOv4 algorithm. First, in the YOLOv4 model, the complex CSPDarkNet53 backbone network was modified into an inverted residual structure, which greatly reduced the number of parameters in the model and increased the detection speed. Second, a new feature fusion network, BiFPN-Lite, is designed to improve the fusion ability of the network and further improve its detection accuracy. The final results show that the mean average precision of the improved lightweight YOLOv4 algorithm in the aluminum surface defect test set reaches 93.5%, the number of model parameters is reduced to 60% of the original, and the number of frames per second (FPS) detected is 52.99, which increases the detection speed by 30%. The efficient detection of aluminum surface defects is realized.

RESUMO

Este artigo pretende conhecer como a rede de cuidados em saúde tem se operacionalizado a partir da percepção de familiares de crianças com demanda de cuidado em saúde mental (SM). Foram realizados dois grupos focais, um com familiares da Atenção Básica (AB) e outro com familiares do Centro de Atenção Psicossocial Infantojuvenil (CAPSij), totalizando 15 participantes. Seguiu-se com a análise lexical do tipo classificação hierárquica descendente, com o auxílio do software R Interface, a fim de análises multidimensionais de textos e questionários (IRaMuTeQ), resultando em cinco classes: A Pílula Mágica; Forças e Fraquezas dos serviços; Procurando por ajuda; Aceitando o diagnóstico da criança e Onde procurei ajuda. Os resultados apontam para dificuldades presentes na AB em identificar e manejar situações de Saúde Mental Infantojuvenil (SMIJ), por meio de uma lógica ainda medicalizante. Ressalta-se que a escola é apresentada como lugar de destaque na produção da demanda por cuidado e a família ainda é pouco convocada à construção das ações. Conclui-se, então, que avanços ainda são necessários para operacionalização de um cuidado pautado nas diretrizes da política de SMIJ.(AU)


This article aims to know how the healthcare network has been operationalized from the perception of family members of children with demand for mental health care (MH). Two focus groups were held, one with family members from Primary Care (PC) and the other with family members from the Child Psychosocial Care Center (CAPSij), totaling 15 participants. A lexical analysis of the descending hierarchical classification type was performed with the help of the software R Interface for multidimensional analyzes of texts and questionnaires (IRAMUTEQ), resulting in five classes: The Magic Pill; Strengths and Weaknesses of services; Looking for help; Accepting the child's diagnosis; and Where did I look for help. The results point to difficulties present in PC in identifying and managing situations of mental health in children and adolescents (MHCA), with a medicalization logic. Note that the school is presented as a prominent place in producing the demand for care, and the family is still not very much involved in the actions. It is, thus, concluded that advances are still needed for operationalization of care guided by MHCA policy guidelines.(AU)


Este artículo tuvo por objetivo conocer cómo opera una red asistencial a partir de la percepción de familiares de niños con demanda de atención en salud mental (SM). Se realizaron dos grupos focales, uno con familiares de Atención Primaria (AP) y otro con familiares del Centro de Atención Psicosocial Infantojuvenil (CAPSij), totalizando 15 participantes. Se realizó análisis léxico del tipo clasificación jerárquica descendente con la ayuda del software Interface de R pour les Analyses Multidimensionnelles de Textes et de Questionnaires (IRAMUTEQ), lo que resultó en cinco clases: "La píldora mágica"; "Fortalezas y debilidades de los servicios"; "En busca de ayuda"; "Aceptar el diagnóstico del niño" y "¿Dónde busqué ayuda?". Los resultados apuntan las dificultades presentes en AP para identificar y manejar situaciones de salud mental infantojuvenil (SMIJ) mediante una lógica aún medicalizante. La escuela tiene un lugar destacado en la producción de la demanda de cuidados y la familia aún no está muy involucrada en la construcción de acciones. Se concluye que se necesitan avances para ofertar una atención guiada por lineamientos de la política del SMIJ.(AU)

A inclusão educacional de estudantes com autismo e/ou deficiência intelectual no ensino regular, em classe comum, evidencia a urgência de programas de formação docente em práticas educacionais baseadas em evidências. O objetivo foi avaliar a estrutura de um processo formativo remoto para elaboração, aplicação e avaliação de uma intervenção comportamental na perspectiva educacional inclusiva e, mais especificamente, caracterizar as atitudes sociais de agentes educacionais participantes da formação. A formação ocorreu em dois formatos. No primeiro, todo o material foi disponibilizado em uma pasta virtual compartilhada e, no segundo, em um Ambiente Virtual de Aprendizagem (AVA). Nos dois formatos, a estrutura contou com parte teórica e prática, por meio de leitura de textos, realização de exercícios e discussões. Na parte prática, participantes tiveram a oportunidade de trabalhar com famílias e estudantes (crianças e jovens) com autismo e/ou deficiência intelectual visando identificar as necessidades e prioridades da família para elaboração colaborativa de um plano de intervenção (sistematização de práticas), implementação e análise das práticas. Para a caracterização da amostra foram levantadas informações pessoais, experiências prévias, familiaridade com ferramentas tecnológicas, conhecimento sobre os conceitos teóricos da Análise do Comportamento. Foi feita uma caracterização das atitudes sociais em relação à inclusão e analisada a validade social de participantes sobre a formação. Os resultados da formação identificaram correlações entre o nível de atitudes sociais e o engajamento nas atividades práticas entregues, além da relação entre o número de participantes que concluíram a formação, o cumprimento de tarefas e dificuldades de contato com a família. O estudo discutiu sobre estratégias que tenham em vista possibilidades de escalabilidade da formação docente de maneira sistemática e científica nesta área de atuação profissional.(AU)


The educational inclusion of students with autism and/or intellectual disabilities in regular education has highlighted the urgency of initial and continuing education programs with evidence-based educational practices. The objective was to evaluate the structure of a remote training process for the elaboration, application and evaluation of a behavioral intervention in the inclusive educational perspective and, complementarily, a specific objective was to characterize the social attitudes of educational agents participating in the training. The training took place in two formats. In the first, all material was made available in a shared folder and in the second, in a Virtual Learning Environment (VLE). In both formats, the structure had a theoretical and practical part, by reading and carrying out exercises and discussions. In the practical part, participants had the opportunity to work with families and students with autism and/or intellectual disability to identify the needs and priorities of the family for the collaborative elaboration of an intervention plan (systematization of practices), implementation and analysis of practices. For the characterization of the sample, personal information, previous experiences, familiarity with technological tools, and knowledge about the theoretical concepts of Behavior Analysis were collected. A characterization of social attitudes towards inclusion was carried out and the social validity of participants regarding training was analyzed. The results of the training identified correlations between the level of social attitudes and engagement in the practical activities delivered, in addition to the relationship between the drop in the number of participants, the fulfillment of tasks and difficulties in contacting the family. The study discussed strategies that aim at possibilities of scaling continuing education in a systematic and scientific way in this area of professional activity.(AU)


La inclusión educativa de estudiantes con autismo y/o discapacidad intelectual en la educación regular, en aulas regulares, ha puesto de relieve la urgencia de programas de educación inicial y continua en prácticas educativas basadas en evidencia. El objetivo de este estudio fue evaluar la estructura de un proceso de formación a distancia para la elaboración, aplicación y evaluación de una intervención conductual en la perspectiva educativa inclusiva y, de forma complementaria, el objetivo específico fue caracterizar las actitudes sociales de los agentes educativos participantes en la formación. La capacitación se llevó a cabo en dos formatos. En el primer, todo el material estaba disponible en una carpeta compartida y, en el segundo, en un entorno virtual de aprendizaje (EVA). En los dos formatos, la estructura tuvo una parte teórica y práctica, mediante la lectura de textos, la realización de ejercicios y discusiones. En la parte práctica, los participantes tuvieron la oportunidad de trabajar con las familias y los alumnos con autismo y/o discapacidad intelectual con el fin de identificar las necesidades y prioridades de la familia para la elaboración colaborativa de un plan de intervención (sistematización de prácticas), su implementación y el análisis de prácticas. Para la caracterización de la muestra, se planteó información personal, experiencias previas, familiaridad con herramientas tecnológicas, conocimiento sobre los conceptos teóricos del análisis de comportamiento. Se realizó una caracterización de las actitudes sociales hacia la inclusión y se analizó la validez social de los participantes respecto a la formación. Los resultados de la capacitación identificaron correlaciones entre el nivel de actitudes sociales y el compromiso en las actividades prácticas impartidas, además de la relación entre la disminución del número de participantes, el cumplimiento de las tareas y las dificultades de contacto con la familia. Este estudio discutió estrategias que apuntan a posibilidades de escalar la formación docente de forma sistemática y científica en esta área de actuación profesional.(AU)

Antitumor immune polarization is a key predictor of clinical outcomes to cancer therapy. An emerging concept influencing clinical outcome involves the spatial location of CD8+ T cells, within the tumor. Our earlier work demonstrated immunosuppressive effects of NOS2 and COX2 tumor expression. Here, we show that NOS2/COX2 levels influence both the polarization and spatial location of lymphoid cells including CD8+ T cells. Importantly, elevated tumor NOS2/COX2 correlated with exclusion of CD8+ T cells from the tumor epithelium. In contrast, tumors expressing low NOS2/COX2 had increased CD8+ T cell penetration into the tumor epithelium. Consistent with a causative relationship between these observations, pharmacological inhibition of COX2 with indomethacin dramatically reduced tumor growth of the 4T1 model of TNBC in both WT and Nos2- mice. This regimen led to complete tumor regression in ∼20-25% of tumor-bearing Nos2- mice, and these animals were resistant to tumor rechallenge. Th1 cytokines were elevated in the blood of treated mice and intratumoral CD4+ and CD8+ T cells were higher in mice that received indomethacin when compared to control untreated mice. Multiplex immunofluorescence imaging confirmed our phenotyping results and demonstrated that targeted Nos2/Cox2 blockade improved CD8+ T cell penetration into the 4T1 tumor core. These findings are consistent with our observations in low NOS2/COX2 expressing breast tumors proving that COX2 activity is responsible for limiting the spatial distribution of effector T cells in TNBC. Together these results suggest that clinically available NSAID's may provide a cost-effective, novel immunotherapeutic approach for treatment of aggressive tumors including triple negative breast cancer.

PURPOSE: During our transthoracic echocardiography (TTE) courses, medical students showed difficulty in spatial orientation. We implemented the use of 3D printed cardiac models of standard TTE views PLAX, PSAX, and A4C and assessed their efficacy in TTE-teaching. METHODS: One hundred fifty-three participants were split into two groups. A pre-test-retest of anatomy, 2D -, and 3D orientation was conducted. The intervention group (n = 77) was taught using 3D models; the control group (n = 76) without. Both were comparable with respect to baseline parameters. Besides test-scores, a Likert scale recorded experiences, difficulties, and evaluation of teaching instruments. RESULTS: From the 153 students evaluated, 123 improved, 20 did worse, and ten achieved the same result after the course. The median overall pre-test score was 29 of 41 points, and the retest score was 35 (p < 0.001). However, the intervention group taught with the 3D models, scored significantly better overall (p = 0.016), and in 2D-thinking (p = 0.002) and visual thinking (p = 0.006) subtests. A backward multivariate linear regression model revealed that the 3D models are a strong individual predictor of an excellent visual thinking score. In addition, our study showed that students with difficulty in visual thinking benefited considerably from the 3D models. CONCLUSION: Students taught using the 3D models significantly improved when compared with conventional teaching. Students regarded the provided models as most helpful in their learning process. We advocate the implementation of 3D-printed heart models featuring the standard views for teaching echocardiography. These findings may be transferable to other evidence based medical and surgical teaching interventions.

BACKGROUND: Three-dimensional endoscopes provide a stereoscopic view of the operating field, facilitating depth perception compared to two-dimensional systems, but are not yet widely accepted. Existing research addresses performance and preference, but there are no studies that quantify anatomical orientation in endoscopic ear surgery. METHODS: Participants (n = 70) were randomised in starting with either the two-dimensional or three-dimensional endoscope system to perform one of two tasks: anatomical orientation using a labelled three-dimensional printed silicone model of the middle ear, or simulated endoscopic skills. Scores and time to task completion were recorded, as well as self-reported difficulty, confidence and preference. RESULTS: Novice surgeons scored significantly higher in a test of anatomical orientation using three-dimensional compared to two-dimensional endoscopy (p < 0.001), with no significant difference in the speed of simulated endoscopic skills task completion. For both tasks, there was lower self-reported difficulty and increased confidence when using the three-dimensional endoscope. Participants preferred three-dimensional over two-dimensional endoscopy for both tasks. CONCLUSION: The findings demonstrate the superiority of three-dimensional endoscopy in anatomical orientation, specific to endoscopic ear surgery, with statistically indistinguishable performance in a skills task using a simulated trainer.

Animals have a remarkable ability to use local cues to orient in space in the absence of a panoramic fixed reference frame. Here we use the mechanosensory lateral line in larval zebrafish to understand rheotaxis, an innate oriented swimming evoked by water currents. We generated a comprehensive light-microscopy cell-resolution projectome of lateralis afferent neurons (LANs) and used clustering techniques for morphological classification. We find surprising structural constancy among LANs. Laser-mediated microlesions indicate that precise topographic mapping of lateral-line receptors is not essential for rheotaxis. Recording neuronal-activity during controlled mechanical stimulation of neuromasts reveals unequal representation of water-flow direction in the hindbrain. We explored potential circuit architectures constrained by anatomical and functional data to suggest a parsimonious model under which the integration of lateralized signals transmitted by direction-selective LANs underlies the encoding of water-flow direction in the brain. These data provide a new framework to understand how animals use local mechanical cues to orient in space.

BACKGROUND: The effect of three-dimensional (3D) vs. two-dimensional (2D) video on performance of a spatially complex procedure and perceived cognitive load were examined among residents in relation to their visual-spatial abilities (VSA). METHODS: In a randomized controlled trial, 108 surgical residents performed a 5-Flap Z-plasty on a simulation model after watching the instructional video either in a 3D or 2D mode. Outcomes included perceived cognitive load measured by NASA-TLX questionnaire, task performance assessed using Observational Clinical Human Reliability Analysis and the percentage of achieved safe lengthening of the scar. RESULTS: No significant differences were found between groups. However, when accounted for VSA, safe lengthening was achieved significantly more often in the 3D group and only among individuals with high VSA (OR = 6.67, 95%CI: 1.23-35.9, p = .027). CONCLUSIONS: Overall, 3D instructional videos are as effective as 2D videos. However, they can be effectively used to enhance learning in high VSA residents.

PURPOSE: To describe a simple finding that can be used to determine donor tissue orientation in Descemet membrane endothelial keratoplasty (DMEK). This involves the appearance of a highly reflective round curved line from an overlapping graft edge within the anterior chamber using light from an endoilluminator. We here name this Kobayashi sign (K-sign). METHODS: Twelve consecutive eyes from 12 patients who underwent DMEK were evaluated for the presence of a K-sign. The presence of Berrospi sign (B-sign), a double-line reflection from the peripheral curls of the Descemet membrane (DM) roll that has been reported to be useful for correct graft orientation, was also evaluated. RESULTS: Of 12 cases, 3 donors showed a loosely scrolled roll soon after DM donor insertion; all 3 of these showed a positive B-sign only when the endoilluminator was used. Nine donors showed a tightly scrolled DM roll without a B-sign; among these 9 donors, a K-sign was visible in 7 cases only when the endoilluminator was used. The remaining 2 cases with a tight scroll configuration showed no K-sign even with the use of endoillumination, indicating that the roll edge was located completely downward; rotation of the roll using a small jet of fluid from paracentesis revealed K-sign in these cases. CONCLUSIONS: This technique is useful for determining the correct orientation of a tightly scrolled DM roll, subsequently enabling rapid DMEK surgery. For loosely scrolled DM roll, endoillumination enhanced B-sing is proven useful.

In different vertebrate species, hippocampus plays a crucial role for spatial orientation. However, even though cognitive lateralization is widespread in the animal kingdom, the lateralization of this hippocampal function has been poorly studied. The aim of the present study was to investigate the lateralization of hippocampal activation in domestic chicks, during spatial navigation in relation to free-standing objects. Two groups of chicks were trained to find food in one of the feeders located in a large circular arena. Chicks of one group solved the task using the relational spatial information provided by free-standing objects present in the arena, while the other group used the local appearance of the baited feeder as a beacon. The immediate early gene product c-Fos was employed to map neural activation of hippocampus and medial striatum of both hemispheres. Chicks that used spatial cues for navigation showed higher activation of the right hippocampus compared to chicks that oriented by local features and compared to the left hippocampus. Such differences between the two groups were not present in the left hippocampus or in the medial striatum. Relational spatial information seems thus to be selectively processed by the right hippocampus in domestic chicks. The results are discussed in light of existing evidence of hippocampal lateralization of spatial processing in chicks, with particular attention to the contrasting evidence found in pigeons.

PURPOSE: In patients with open growth plates, the direction of tunneling that avoids distal femoral physis (DFP) damage in anatomic reconstructions of the medial patellofemoral ligament (MPFL) has been a topic of discussion. The objective of this study was to determine the ideal orientation for anatomic reconstructions of MPFL tunneling that minimized DFP damage while avoiding breaching the intercondylar notch. METHODS: Eighty magnetic resonance images of patients aged 10 through 17 were obtained, randomly sampled from the institutional database. A de novo software was developed to obtain 3D models of the distal femur and DFP. In each model, the anatomical insertion point of the MPFL was determined as defined by Stephen. A 20-mm-depth drilling was simulated, starting from the insertion point at every possible angle within a 90° cone using 5-, 6- and 7-mm drills. Physeal damage for each pair of angles and each drill size was determined. Damage was expressed as a percentage of total physis volume. Statistical analysis was conducted using Student's t test and one-way ANOVA. RESULTS: Maximum physeal damage (5.35% [4.47-6.24]) was obtained with the 7-mm drill when drilling 3° cephalic and 15° posterior from insertion without differences between sexes (n.s.). Minimal physeal damage (0.22% [0.07-0.37]) was obtained using the 5-mm drill aimed 45° distal and 0° anteroposterior, not affected by sex (n.s.). Considering intra-articular drilling avoidance, the safest zone was obtained when aiming 30°-40° distal and 5°-35° anterior, regardless of sex. CONCLUSION: Ideal femoral tunnel orientation, avoiding physeal damage and breaching of the intercondylar notch, was obtained when aiming 30°-40° distal and 5°-35° anterior, regardless of sex. This area is a safe zone that allows anatomic MPFL reconstruction of patients with an open physis.

Abstract Spatial disorientation has been observed in mild cognitive impairment (MCI) and is associated with a higher risk of progression to Alzheimer's disease (AD). However, there is no gold standard assessment for spatial orientation and paper-and-pencil tests lack ecological validity. Recently, there has been an increasing number of studies demonstrating the role of spatial disorientation as a cognitive marker of pathological decline, shedding new light on its importance for MCI. This systematic review aimed to investigate the accuracy of spatial orientation tasks for the diagnosis of MCI by comparison with cognitively healthy elderly. The search was conducted in the databases Medical Literature Analysis and Retrieval System Online (MEDLINE/PubMed), Web of Science, Scopus, Excerpta Medica Database (Embase), Literatura Latino-Americana e do Caribe em Ciências da Saúde (Lilacs) and Scientific Electronic Library Online (SciELO). Only original studies reporting spatial orientation assessment in MCI patients compared to a healthy control group were included. Studies were excluded if the MCI classification did not follow well described criteria and/or if accuracy results of spatial orientation assessment were not provided. Seven studies met the eligibility criteria, describing a variety of spatial orientation assessments including questionnaires, paper-and-pencil, office-based route learning, and computer-based and virtual reality tasks. Spatial orientation tasks demonstrated moderate to high accuracy in detecting elderly with MCI compared to cognitively healthy elderly, with areas under the curve (AUC) ranging from 0.77 to 0.99. However, important methodological issues were found in the selected studies which should be considered when interpreting results. Although the inclusion of spatial orientation assessments in MCI evaluations seems to have significant value, further studies are needed to clarify their true capacity to distinguish pathological from non-pathological aging.


RESUMO A ocorrência de desorientação espacial foi observada no comprometimento cognitivo leve (CCL) e está associada a um maior risco de progressão para a doença de Alzheimer (DA). No entanto, não há um padrão ouro para avaliação da orientação espacial e os testes em papel e caneta não apresentam validade ecológica. Recentemente, um número cada vez maior de estudos têm apontado o papel da desorientação espacial como um marcador cognitivo do declínio patológico, lançando uma nova luz sobre sua importância para o CCL. Esta revisão sistemática teve como objetivo investigar a acurácia de tarefas de orientação espacial para se estabelecer o diagnóstico de CCL entre idosos cognitivamente saudáveis. A pesquisa foi realizada através das bases de dados Medline/PubMed, Web of Science, Scopus, Embase, Lilacs e Scielo. Apenas artigos originais que reportassem avaliação da orientação espacial em idosos CCL comparados a um grupo controle saudável foram incluídos. Foram excluídos os estudos que não utilizassem a classificação de CCL segundo critérios bem descritos e/ou que não reportassem resultados de acurácia da avaliação da orientação espacial. Sete estudos atenderam aos critérios de elegibilidade, descrevendo uma variedade de formas de avaliação da orientação espacial, incluindo questionários, tarefas em papel e lápis, tarefas de aprendizado de rotas no escritório, tarefas baseadas em computador e com realidade virtual. As tarefas de orientação espacial demonstraram acurácia moderada a alta na detecção de CCL em comparação com idosos cognitivamente saudáveis, com áreas sob a curva (area under the curve — AUC) variando de 0,77 a 0,99. No entanto, um viés metodológico importante foi identificado nos estudos selecionados, o que deve ser levado em consideração na interpretação dos resultados. Apesar da inclusão da orientação espacial na avaliação cognitiva em CCL parecer ter um valor significativo, mais estudos são necessários para esclarecer sua verdadeira capacidade de distinguir o envelhecimento patológico do não patológico.

The anterior pelvic plane (APP) defined by both iliac spines and the pubic symphysis, is essential in total hip arthroplasty (THA) for the orientation of the prosthetic cup. However, the APP is nowadays still difficult to determine in computer assisted orthopedic surgery (CAOS). We propose to use a statistical shape model (SSM) of the pelvis to estimate the APP from ipsilateral anatomical landmarks, more easily accessible during surgery in computer assisted THA with the patient in lateral decubitus position. A SSM of the pelvis has been built from 40 male pelvises. Various ipsilateral anatomical landmarks have been extracted from these data and used to deform the SSM. Fitting the SSM to several combinations of these landmarks, we were able to reconstruct the pelvis with an accuracy between 2.8mm and 4.4mm, and estimate the APP inclination with an angular error between 1.3° and 2.8°, depending on the landmarks fitted. Results are promising and show that the APP could be acquired during the intervention from ipsilateral landmarks only.

BACKGROUND: Changes in knee joint line orientation (KJLO) resulting in excessive joint line obliquity are a well-known consequence of high tibial osteotomy (HTO) procedures and could lead to degenerative changes. The precise effect of the correction on final KJLO changes is poorly understood. The goal of this study was to identify radiographical parameters that could help to explain the size of KJLO changes after HTO surgery. METHODS: A total 117 HTO patients were radiographically examined preoperatively and three months postoperatively. Radiographic parameters were KJLO, medial proximal tibial angle (MPTA), hip-knee angle (HKA), mechanical lateral distal femoral angle (mLDFA), lateral distal tibial angle (LDTA), knee joint line congruence angle (KJLCA), ankle joint line congruence angle (AJLCA) and ankle joint line orientation (AJLO). Four new radiographic parameters were introduced to describe knee and foot position on long-leg X-ray: malleolar distance to midline (MDTM), intermalleolar distance (IMD), condylar distance to midline (CDTM) and intercondylar distance (ICD). Correlations of these parameters and changes in KJLO were assessed. RESULTS: Strong correlations are found between KJLO changes and MDTM (r = 0.709), IMD (r = 0.691), CDTM (r = 0.711) and ICDM (r = 0.702), in contrast to weak correlations between changes in KJLO and MPTA (r = -0.342). These results suggest an important impact of foot and knee position changes on the final alteration of KJLO after HTO. CONCLUSIONS: The final change of KJLO after HTO is the effect of adaptation of the lower limb, which is driven by the (maximum) alterations of foot and knee position rather than the size of correction of the procedure.

The orientation of microtubule (MT) networks is exploited by motors to deliver cargoes to specific intracellular destinations and is thus essential for cell polarity and function. Reconstituted in vitro systems have largely contributed to understanding the molecular framework regulating the behavior of MT filaments. In cells, however, MTs are exposed to various biomechanical forces that might impact on their orientation, but little is known about it. Oocytes, which display forceful cytoplasmic streaming, are excellent model systems to study the impact of motion forces on cytoskeletons in vivo. Here we implement variational optical flow analysis as a new approach to analyze the polarity of MTs in the Drosophila oocyte, a cell that displays distinct Kinesin-dependent streaming. After validating the method as robust for describing MT orientation from confocal movies, we find that increasing the speed of flows results in aberrant plus end growth direction. Furthermore, we find that in oocytes where Kinesin is unable to induce cytoplasmic streaming, the growth direction of MT plus ends is also altered. These findings lead us to propose that cytoplasmic streaming - and thus motion by advection - contributes to the correct orientation of MTs in vivo. Finally, we propose a possible mechanism for a specialized cytoplasmic actin network (the actin mesh) to act as a regulator of flow speeds to counteract the recruitment of Kinesin to MTs.

Lipid bilayers form the basis of cell membranes and the phase behaviour of the membrane has been linked to proper cell function. Model membranes composed of relatively simple mixtures of phospholipids and cholesterol can already exhibit complex phase behaviour. Specifically, liquid ordered-liquid disordered fluid phase coexistence occurs in mixtures which contain one saturated long chain phospholipid and one unsaturated long chain phospholipid and cholesterol. This fluid-fluid two phase region persists over a broad range of temperatures and sample compositions and can be observed experimentally in various sample preparations including multilamellar dispersions, bicelles, and multi-lamellae stacked on glass slides. In order to explore the practicality of using oriented samples with different concentrations of the peptide, we investigated the effect of the addition of a synthetic 22 residue amphiphilic peptide on the orientability and phase behaviour of the lipid mixtures, as well as the orientation and dynamics of the peptide itself via 2H NMR. Increasing the peptide concentration promoted the formation of the liquid ordered phase, suggesting a preferential interaction of the peptide with the thicker ordered phase. However, higher peptide content (> 4 mol%) had a significant negative effect on the alignment of bicelles with their bilayer normal perpendicular to the external magnetic field. In the stacked bilayer samples, 6 mol% peptide eliminated the two phase coexistence region altogether and a single liquid ordered phase was observed from 285 to 311 K. Even so, 2H spectra of the peptide itself did not reveal any preference for the peptide to partition into either the liquid disordered or liquid ordered phase and we found two populations of the peptide, one which undergoes rapid axial reorientation about the bilayer normal and a second (powder component) which does not.

Sound radiation patterns have ecological implications in the effective communication between conspecifics, like optimization of the sound propagation, increase the likelihood to reach mates and to mitigate effects of sound scattering by environmental factors like vegetation. The territorial frog Allobates femoralis advertises its territory against conspecific males and attract females with advertisement calls. Here we report the nearly omnidirectional sound-radiation pattern of the advertisement call of A. femoralis. This sound spreading pattern allows the males to attract mates and repel rivals in all directions. Furthermore, A. femoralis males direct the advertisement call to conspecific neighbours after phonotactic orientation.

Resumo Este estudo propôs a avaliar o nível de atividade física e o estado cognitivo de idosos usuários das Unidades Básicas de saúde (UBS) do Município de Maringá, Paraná. Trata-se de um estudo epidemiológico descritivo, de corte observacional e transversal, realizado com 654 idosos, de ambos os sexos, usuários das UBS. Foi utilizado um questionário sociodemográfico, o Mini exame do estado mental e o International Activity Questionnaire (IPAQ), versão curta. A análise dos dados foi realizada mediante o teste Kolmogorov-Smirnov, Qui quadrado, Kruskal-Wallis e "U" de Mann-Whitney, adotando significância quando p < 0,05. Os idosos não realizam atividades físicas vigorosas e poucas atividades moderadas durante a semana. Porém, apresentaram alto escore na orientação temporal (Md = 5,0), Orientação espacial (Md = 5,0), memória imediata (Md = 3,0), evocação (Md = 3,0) e linguagem (Md = 8,0). Ao comparar o estado mental em função do nível de atividade física dos idosos verificou-se que os Muito ativo/ativo possuem melhor atenção e cálculo (p = 0,036), evocação (p = 0,001) e estado cognitivo geral (p = 0,002), se comparado aos irregularmente ativos e sedentários. Níveis adequados de atividade física podem estar relacionados a melhores escores de funções cognitivas de sujeitos idosos.


Abstract This study proposed to evaluate the level of physical activity and the cognitive state of elderly users of Primary Care Facilities (UBS) of the Municipality of Maringá, State of Paraná, Brazil. This is a descriptive, cross-sectional, epidemiological study with 654 elderly men and women UBS users. A sociodemographic questionnaire, the Mini Mental State Examination (MMSE) and the short version of the International Physical Activity Questionnaire (IPAQ) were employed. Data was analyzed using the Kolmogorov-Smirnov, Chi-square, Kruskal-Wallis and Mann-Whitney "U" tests, with a significance level of p<0.05. However, they evidenced a high score in temporal orientation (Md = 5.0), spatial orientation (Md = 5.0), immediate memory (Md = 3.0), recall (Md = 3.0) and language (Md = 8.0). When comparing the mental state according to the level of physical activity of the elderly, we observed that very active/active individuals had better attention and calculation (p = 0.036), recall (p = 0.001) and general cognitive status (p = 0.002) against irregularly active and sedentary elderly. Adequate levels of physical activity may be related to better scores of cognitive functions of elderly subjects.

INTRODUCTION: Anxiogenic and anxiolytic effects of cannabinoids are mediated by different mechanisms, including neural signaling via cannabinoid receptors (CBRs) and nicotinic cholinergic receptors (nAChRs). This study examined the effects of prior nicotine (the psychoactive component in tobacco) exposure on behavioral sensitivity to delta-9-tetrahydrocannabinol (THC; the psychoactive component of cannabis) challenge in animals. METHODS: Male and female adult Sprague-Dawley rats (N = 96) were injected daily with nicotine (1 mg/kg, i.p.) or vehicle for 14 days, followed by a 14-day drug-free period. On test day, rats were injected with THC (0.5, 2.0, or 5.0 mg/kg, i.p.) or vehicle and anxiety-related behavior was assessed in the emergence (EM), elevated plus maze (EPM), and social interaction (SI) tests. RESULTS: Chronic nicotine pretreatment attenuated some of the anxiogenic effects induced by THC challenge which can be summarized as follows: (a) THC dose-dependently affected locomotor activity, exploratory behavior, and social interaction in the EM, EPM, and SI tests of unconditioned anxiety; (b) these effects of acute THC challenge were greater in females compared with males except for grooming a conspecific; (c) prior nicotine exposure attenuated the effects of acute THC challenge for locomotor activity in the EPM test; and (d) prior nicotine exposure attenuated the effects of THC challenge for direct but not indirect physical interaction in the SI tests. CONCLUSIONS: The ability of nicotine prior exposure to produce long-lasting changes that alter the effects of acute THC administration suggests that chronic nicotine may induce neuroplastic changes that influence the subsequent response to novel THC exposure.
