RESUMO

Sea urchin injuries are common following accidental contact with sharp sea urchin spines. Immediate manifestations of injury include local erythema, pain, and myalgia. Failure to remove the spines from the skin may result in delayed systemic reactions, secondary infection, granulomas, and-if joint spaces are involved-inflammatory or infectious synovitis and arthritis. The majority of severe complications can be avoided if the spines are fully removed from the skin soon after injury, which can be difficult. This article aims to bring awareness to the myriad complications from sea urchin injuries as well as the mechanisms for successful spine removal.

RESUMO

Mycophenolate mofetil (MpM) is a medication used to prevent the rejection of transplanted organs, particularly in kidney, heart, and liver transplant surgeries. It is extremely important to be conscious that MpM can raise the risk of severe infections and some cancers if it exceeds the recommended dose while lower doses will result in organ rejections. So, it is essential to monitor the dosage of MpM in real time in the micromolar range. In this work, we have synthesized 3-aminopropyltriethoxysilane (APTES) functionalized nickel cobaltite (NiCo2O4) and this amino functionalization was chosen to enhance the stability and electrochemical activity of NiCo2O4. The enhanced activity of NiCo2O4 was used for developing an electrochemical sensor for the detection of MpM. APTES functionalized NiCo2O4 was coated on carbon cloth and used as the working electrode. Surface functionalization with APTES on NiCo2O4 was aimed at augmenting the adsorption/interaction of MpM due to its binding properties. The developed sensor showed a very low detection limit of 1.23 nM with linear ranges of 10-100 nM and 1-100 µM and its practical applicability was examined using artificial samples of blood serum and cerebrospinal fluid, validating its potential application in real-life scenarios.

Different bioactive molecules extracted from macroalgae, including oxylipins, showed interesting potentials in different applications, from healthcare to biomaterial manufacturing and environmental remediation. Thus far, no studies reported the effects of oxylipins-containing macroalgae extracts on embryo development of marine invertebrates and on neuroblastoma cancer cells. Here, the effects of an oxylipins-containing extract from Ericaria brachycarpa, a canopy-forming brown algae, were investigated on the development of Arbacia lixula sea urchin embryos and on SH-SY5Y neuroblastoma cells viability. Embryos and cells were exposed to concentrations covering a full 0-100% dose-response curve, with doses ranging from 0 to 40 µg mL-1 for embryos and from 0 to 200 µg mL-1 for cells. These natural marine toxins caused a dose-dependent decrease of normal embryos development and of neuroblastoma cells viability. Toxicity was higher for exposures starting from the gastrula embryonal stage if compared to the zygote and pluteus stages, with an EC50 significantly lower by 33 and 68%, respectively. Embryos exposed to low doses showed a general delay in development with a decrease in the ability to calcify, while higher doses caused 100% block of embryo growth. Exposure of SH-SY5Y neuroblastoma cells to 40 µg mL-1 for 72 h caused 78% mortality, while no effect was observed on their neuronal-like cells derivatives, suggesting a selective targeting of proliferating cells. Western Blot experiments on both model systems displayed the modulation of different molecular markers (HSP60, HSP90, LC3, p62, CHOP and cleaved caspase-7), showing altered stress response and enhanced autophagy and apoptosis, confirmed by increased fragmented DNA in apoptotic nuclei. Our study gives new insights into the molecular strategies that marine invertebrates use when responding to their environmental natural toxins and suggests the E. brachycarpa's extract as a potential source for the development of innovative, environmentally friendly products with larvicide and antineoplastic activity.

Microplastics, particles under 5 mm, pervade aquatic environments, notably in Tarragona's coastal region (NE Iberian Peninsula), hosting a major plastic production complex. To investigate weathering and yellowness impact on plastic pellets toxicity, sea-urchin embryo tests were conducted with pellets from three locations-near the source and at increasing distances. Strikingly, distant samples showed toxicity to invertebrate early stages, contrasting with innocuous results near the production site. Follow-up experiments highlighted the significance of weathering and yellowing in elevated pellet toxicity, with more weathered and colored pellets exhibiting toxicity. This research underscores the overlooked realm of plastic leachate impact on marine organisms while proposes that prolonged exposure of plastic pellets in the environment may lead to toxicity. Despite shedding light on potential chemical sorption as a toxicity source, further investigations are imperative to comprehend weathering, yellowing, and chemical accumulation in plastic particles.

Biomineralization had apparently evolved independently in different phyla, using distinct minerals, organic scaffolds, and gene regulatory networks (GRNs). However, diverse eukaryotes from unicellular organisms, through echinoderms to vertebrates, use the actomyosin network during biomineralization. Specifically, the actomyosin remodeling protein, Rho-associated coiled-coil kinase (ROCK) regulates cell differentiation and gene expression in vertebrates' biomineralizing cells, yet, little is known on ROCK's role in invertebrates' biomineralization. Here, we reveal that ROCK controls the formation, growth, and morphology of the calcite spicules in the sea urchin larva. ROCK expression is elevated in the sea urchin skeletogenic cells downstream of the Vascular Endothelial Growth Factor (VEGF) signaling. ROCK inhibition leads to skeletal loss and disrupts skeletogenic gene expression. ROCK inhibition after spicule formation reduces the spicule elongation rate and induces ectopic spicule branching. Similar skeletogenic phenotypes are observed when ROCK is inhibited in a skeletogenic cell culture, indicating that these phenotypes are due to ROCK activity specifically in the skeletogenic cells. Reduced skeletal growth and enhanced branching are also observed under direct perturbations of the actomyosin network. We propose that ROCK and the actomyosin machinery were employed independently, downstream of distinct GRNs, to regulate biomineral growth and morphology in Eukaryotes.

Gregory's diverticulum, a digestive tract structure unique to a derived group of sand dollars (Echinoidea: Scutelliformes), is filled with sand grains obtained from the substrate the animals inhabit. The simple methods of shining a bright light through a specimen or testing response to a magnet can reveal the presence of a mineral-filled diverticulum. Heavy minerals with a specific gravity of >2.9 g/cm3 are selectively concentrated inside the organ, usually at concentrations one order of magnitude, or more, greater than found in the substrate. Analyses of diverticulum content for thirteen species from nine genera, using optical mineralogy, powder X-ray diffraction, scanning electron microscopy and energy dispersive X-ray spectroscopy, as well as micro-computed tomography shows the preference for selection of five major heavy minerals: magnetite (Fe3O4), hematite (Fe2O3), ilmenite (FeTiO3), rutile (TiO2), and zircon (ZrSiO4). Minor amounts of heavy or marginally heavy amphibole, pyroxene and garnet mineral grains may also be incorporated. In general, the animals exhibit a preference for mineral grains with a specific gravity of >4.0 g/cm3, although the choice is opportunistic and the actual mix of mineral species depends on the mineral composition of the substrate. The animals also select for grain size, with mineral grains generally in the range of 50 to 150 µm, and do not appear to alter this preference during ontogeny. A comparison of analytical methods demonstrates that X-ray attenuation measured using micro-computed tomography is a reliable non-destructive method for heavy mineral quantification when supported by associated analyses of mineral grains extracted destructively from specimens or from substrate collected together with the specimens. Commonalities in the electro-chemical surface properties of the ingested minerals suggest that such characteristics play an important role in the selection process.

Resumen Introducción: El comportamiento de enderezamiento se ha utilizado como indicador de salud en respuesta a variables estresantes. La aplicación de este parámetro en acuicultura podría ser beneficiosa para reducir la mortalidad y mejorar el bienestar en el cultivo del erizo de mar Arbacia dufresnii. Objetivo: El objetivo de este estudio fue evaluar el efecto del sexo, el diámetro y tres factores estresantes sobre el comportamiento de enderezamiento del erizo de mar A. dufresnii. Métodos: Se midieron un total de 300 animales para evaluar el tiempo de comportamiento de enderezamiento completo (CRB) y 100 de ellos también registraron el tiempo de comportamiento de enderezamiento medio (HRB). Se aplicaron tres factores estresantes a los animales: repeticiones seriadas (tres giros sucesivos), temperatura (shock de 24 horas) e inducción del desove con inyección de KCl. El tiempo se midió con un cronómetro y el diámetro con un calibre de precisión. Resultados: El tiempo de enderezamiento resultó ser dependiente del diámetro, pero independiente del sexo. El límite superior de temperatura de 19 °C tuvo un efecto significativo en el comportamiento de enderezamiento en comparación con las temperaturas de 16 °C y 13 °C, con tiempos de CRB de hasta 150 segundos. Las repeticiones seriadas y el desove no tuvieron un efecto significativo. Sin embargo, con base en los tiempos registrados, se puede deducir que el desove tuvo un impacto en la salud de los animales con tiempos de CRB de hasta 150 segundos en comparación con el control, con tiempos inferiores. Conclusiones: El comportamiento de enderezamiento completo (CRB) parece ser un indicador óptimo para evaluar la salud y condición del erizo de mar A. dufresnii, sin embargo sería óptimo realizar más ensayos para corroborar el efecto del tratamiento control con respecto al desove.

Abstract Introduction: The genus Agassizia in Mexico is represented both in the fossil record by the species Agassizia regia† during the Miocene of Chiapas and by the extant species Agassizia excentrica on the Atlantic coast and Agassizia scrobiculata on the Pacific coast. Qualitative diagnosis and descriptions make it hard to distinguish morphological boundaries between species, especially in groups with fossils and recent representatives, increasing the level of complexity by having samples of disparate qualities and quantities. Objective: We propose the use of little explored statistical methods in the comparison of paleontological and biological populations. This methodology allowed us to resolve issues of missing values in a morphometric data set for the genus Agassizia. Methods: Using samples recently collected and specimens already housed in collections, we explore a routine of recovery of missing data MICE and the numerical and graphic analyses PERMANOVA, PCA, and SIMPER to compare morphometric parameters between these species for recognizing diagnostic characters. Results: Our results show a morphological difference in the length of the ambulacrum II and the length and width of the periproct and peristome structures, these being greater in A. scrobiculata, with a consistent pattern in both population samples not previously described. Conclusions: Quantitative morphometric comparisons can be an assertive and complementary tool to determine distinctive differentiation characteristics in species of the same genus. Comparative morphology reviews should be an ongoing exercise to keep taxonomic knowledge on both extinct and extant species up to date. Our research encourage the scientific community studying fossil populations to utilize quantitative and multivariate methods to strengthen their investigations.


Resumen Introducción: El género Agassizia en México está representado tanto en el registro fósil por la especie Agassizia regia† del Mioceno de Chiapas, como por las especies actuales Agassizia excentrica de la costa del Atlántico y Agassizia scrobiculata de la costa del Pacífico. Las descripciones y diagnosis cualitativas dificultan reconocer los limites morfológicos entre especies, especialmente en grupos con representantes fósiles y recientes, e incrementando el nivel de complejidad al tener muestras de cantidad y calidad desiguales. Objetivo: Proponemos el uso de métodos estadísticos poco explorados en la comparación de poblaciones paleontológicas y biológicas. Esta metodología nos permitió resolver problemas de valores faltantes en un conjunto de datos morfométricos para el género Agassizia. Métodos: Usando muestras recolectadas para este fin, así como provenientes de colecciones científicas, exploramos una rutina de recuperación de datos faltantes MICE, y los análisis numéricos y gráficos PERMANOVA, PCA y SIMPER para comparar parámetros morfométricos entre estas especies y reconocer caracteres de diagnóstico. Además, comparamos cuidadosamente los caracteres morfológicos descritos previamente en la literatura taxonómica y la descripción ambiental del hábitat actual de A. scrobiculata. Resultados: Nuestros resultados muestran una diferencia morfológica en la longitud del ambulacrum II y la longitud y anchura de las estructuras del periprocto y peristoma, siendo estas mayores en A. scrobiculata, con un patrón consistente en ambas muestras poblacionales no descrito previamente. El hábitat actual de las muestras de A. scrobiculata en la costa del Pacífico es un sistema costero poco profundo con sedimentos arenosos y temperaturas tropicales. Bahía Chamela comparte varias similitudes con la fauna y las condiciones ambientales previamente descritas en el Mioceno de Chiapas. Conclusiones: Las comparaciones morfométricas cuantitativas pueden ser una herramienta poderosa y complementaria para determinar caracteres distintivos de diferenciación en especies del mismo género. Las revisiones de morfología comparativa deben ser un ejercicio continuo para mantener actualizado el conocimiento taxonómico sobre las especies existentes y extintas. Nuestro trabajo busca incentivar a la comunidad científica que trabaja con poblaciones fósiles a explorar estos y otros métodos cuantitativos y multivariados para fortalecer sus investigaciones.

Abstract Introduction: Short-term gametes storage is an inexpensive and simple technique that allows the use of the same batch of eggs or sperm at different times, maximizing the application of research protocols and the use of gametes in production. Arbacia dufresnii is a sea urchin species with proven aquaculture potential and already used in the nutraceutical industry. Aging of its gametes is unknown and is a needed information to scale up the production. Objective: Determine the effect of male and female gamete aging on the fertilization success of Arbacia dufresnii. This will allow optimizing the use of gametes after collection decoupling spawning from fertilization. Methods: A. dufresnii individuals were induced to spawn and gametes were kept at 12 ± 1 °C throughout each bioassay. Sperm was separated into two treatments: activated sperm in seawater (AS), and dry sperm (DS). Two bioassays were made: Bioassay 1 evaluated the effect of time on fertility by performing fertilization tests at 0 h, 24 h, 48 h, 72 h, and 96 h after spawning. Bioassay 2 evaluated the contribution of each type of aged gamete on fertility, combining aged gametes (96 h) with fresh gametes (0 h). Results: Bioassay 1: the fertilization success obtained by combining eggs (E) with AS or DS presented important differences. While the fertilization success remained acceptable (greater than 50 %) for up to 72 h using ExDS, it only remained acceptable for up to 48 h using ExAS. Bioassay 2: acceptable fertilization success was found by combining aged E (96 h) with fresh sperm, or aged DS (96 h) with fresh E, but not using aged AS with fresh E. Conclusions: The findings of this work show that fertilization success in A. dufresnii gametes remains relatively unchanged for up to 48 h after spawning when combining ExAS, and for up to 72 h when combining ExDS. However, when combining aged E or aged DS with a fresh gamete, post-collection fertilization can be extended up to 96 h. In this work, the first steps have been taken to understand the conservation time of A. dufresnii gametes with minimum intervention.


Resumen Introducción: El almacenamiento de gametos a corto plazo es una técnica económica y sencilla que permite utilizar el mismo lote de óvulos o espermatozoides en diferentes momentos, maximizando la aplicación de protocolos de investigación y el uso de gametos en la producción. Arbacia dufresnii es una especie con probado potencial acuícola como fuente de gametos para la industria nutracéutica. Sin embargo, se desconoce el envejecimiento de sus gametos y es una información necesaria para escalar la producción. Objetivo: Determinar el efecto del envejecimiento de los gametos masculinos y femeninos en el éxito de la fecundación de Arbacia dufresnii con el fin de optimizar el aprovechamiento de los gametos después de la recolecta desincronizando el desove de la fecundación. Métodos: Se indujo el desove de individuos de A. dufresnii y los gametos se mantuvieron a 12 ± 1 °C durante cada bioensayo. El esperma se separó en dos tratamientos: esperma activado en agua de mar (AS) y esperma seco (DS). Se realizaron dos bioensayos: El Bioensayo 1 evaluó el efecto del tiempo sobre la fertilidad realizando pruebas de fecundación a las 0 h, 24 h, 48 h, 72 h y 96 h después del desove. El bioensayo 2 evaluó la contribución de cada tipo de gameta envejecida (96 h) sobre la fertilidad, combinando gametos envejecidas (96 h) con gametos frescas (0 h). Resultados: Bioensayo 1: el éxito de fecundación obtenido combinando huevos (E) con AS o DS presentó diferencias importantes. Si bien el éxito de la fecundación se mantuvo aceptable (más del 50 %) durante un máximo de 72 h con ExDS, solo permaneció aceptable hasta 48 h con ExAS. Bioensayo 2: se encontró un éxito de fecundación aceptable combinando E envejecidos (96 h) con esperma fresco, o DS envejecido (96 h) con E fresco (0 h), pero no usando AS envejecido con E fresco (0 h). Conclusiones: Los hallazgos de este trabajo muestran que el éxito de la fecundación en los gametos de A. dufresnii permanece relativamente sin cambios hasta 48 h después del desove cuando se combina ExAS, y hasta 72 h cuando se combina ExDS. Sin embargo, cuando se combina E envejecido o DS envejecido con un gameto fresco, el tiempo entre la recolección y la fecundación puede extenderse hasta 96 h. En este trabajo se han dado los primeros pasos para entender el tiempo de conservación de los gametos de A. dufresnii con mínima intervención.

Abstract Introduction: Sea urchin aquaculture is a rising industry, and in consequence, there is a need to establish optimal culture parameters to ensure the health of the cultured animals. Objective: To evaluate the bacterial counts in the seawater of sea urchin (Arbacia dufresnii) aquaculture recirculating systems (RAS). Methods: The bacteriological water quality of two RAS containing sea urchins was determined. For approximately two months, weekly water samples were taken. The bacteriological quality was determined by counting total aerobic heterotrophic populations, lactic acid bacteria, enterobacterias and genus Vibrio. Physicochemical parameters were also measured. Results: There was no presence of disease or mortality. Enterobacteria and lactic acid bacteria were not detected from both RAS systems. The number of animals had an important effect on the observed difference in the count of total bacteria and Vibrio spp. In RAS 1 the maximum counts of total bacteria and Vibrio spp. were 2.8 x 105 ± 1.7 x 105 and 1.45 x 105 ± 3.6 x 104 UFC ml-1, respectively. In RAS 2 total bacteria and Vibrio spp. exhibited repetitive behavior over time influenced in part by water exchange and mainly by feeding. The results indicate that periodic water changes ensure a limited growth of bacterial strains as Vibrio and other bacteria. Conclusions: Our results suggests that the bacterial count levels recorded in this study can be used as a threshold or safety limit for Arbacia dufresnii aquaculture.


Resumen Introducción: La acuicultura de erizos de mar es una industria en auge, y en consecuencia, existe la necesidad de establecer los parámetros de cultivo óptimos para garantizar la salud de los animales en cultivo. Objetivo: Evaluar los recuentos bacterianos en el agua de cultivo de los sistemas de recirculación acuícola (RAS) de erizo de mar Arbacia dufresnii. Métodos: Se determinó la calidad bacteriológica del agua de cultivo de dos RAS que contenían erizos de mar. Durante aproximadamente dos meses, se tomaron muestras de agua semanalmente. La calidad bacteriológica se determinó realizando recuento de las poblaciones heterótrofas aerobias totales, bacterias ácido lácticas, enterobacterias y bacterias del género Vibrio. También se midieron parámetros fisicoquímicos. Resultados: No se observaron patologías ni mortalidad. No se detectaron enterobacterias ni bacterias del ácido láctico en ninguno de los sistemas RAS. El número de animales cultivados tuvo un efecto importante en la diferencia observada en el recuento de bacterias totales y Vibrio spp. En el RAS 1 los recuentos máximos de bacterias totales y Vibrio spp. fueron 2.8 x 105 ± 1.7 x 105 y 1.45 x 105 ± 3.6 x 104 UFC ml-1, respectivamente. En RAS 2 los recuentos de bacterias totales y Vibrio spp. exhibieron un comportamiento repetitivo en el tiempo influenciado en parte por el recambio de agua y principalmente por la alimentación. Los resultados indican que los cambios periódicos de agua aseguran un crecimiento limitado de cepas bacterianas como Vibrio y otras bacterias. Conclusiones: Nuestros resultados sugieren que los niveles de recuento bacteriano registrados en este estudio pueden usarse como umbral o límite de seguridad para la acuicultura de Arbacia dufresnii.

Abstract Introduction: The sea urchin Diadema mexicanum, due to its bioerosion activity, is considered of ecological importance. This phenomenon could negatively or positively affect coral reef ecosystems. The bioerosion process varies according to the abundance and size of the sea urchin. Objective: Juvenile organisms possess different metabolic needs compared to adults, so knowing their stomach content according to size allows us to quantify the selection of substrate bioeroded. Methods: To determine this, D. mexicanum individuals were collected in 12 sites from January 2009 to September 2010 along the Eastern Tropical Pacific coast. The stomach content was categorized in Carbonated Fraction (CF), Non-Carbonated Fraction (NCF), and Organic Matter (OM). Stomach content was analyzed according to a) juvenile (< 2.5 cm) or adult (> 2.5 cm) stage and b) locality. Results: Juveniles presented the following stomach content average percentages: 20.7 % OM, 12 % NCF and 67.9 % CF; and adults: 11.4 % OM, 14.8 % NCF and 73.8 % CF. Based on a Wilcoxon test and a Kendall linear regression, the following results were obtained. The carbonated fraction in the stomach increased by 0.47 units on average for every cm of growth (p < 0.05). OM consumed by D. mexicanum increases only 0.05 units for every cm of growth (p < 0.05). We found a difference of stomach content depending on the site (p < 0.05) and life stage (p < 0.05). Localities like Huatulco and Coco presented significant differences that could be related to local oceanographic conditions. Conclusions: We relate these changes of the stomach fractions to the necessity of the juvenile sea urchins for nutrients to maintain their growth. The amount of OM is crucial for the development of early stages, meaning that there is a difference in substrate selection associated with growth.


Resumen Introducción: El erizo de mar Diadema mexicanum, por su actividad de bioerosión, es considerado de importancia ecológica. Este fenómeno podría afectar negativa o positivamente a los ecosistemas de arrecifes de coral. El proceso de bioerosión varía según la abundancia y el tamaño del erizo de mar. Objetivo: Los organismos juveniles poseen diferentes necesidades metabólicas en comparación con los adultos, por lo que conocer el contenido estomacal según el tamaño, nos permite cuantificar la selección de sustrato bioerosionado. Métodos: Para determinar esto, se recolectaron individuos de D. mexicanum en 12 sitios desde enero de 2009 hasta septiembre de 2010 a lo largo de la costa del Pacífico Tropical Oriental. El contenido estomacal se clasificó en Fracción Carbonatada (FC), Fracción No Carbonatada (NCF) y Materia Orgánica (MO). El contenido estomacal se analizó según a) estadio juvenil (< 3 cm) o adulto (> 3 cm) y b) localidad. Resultados: Los juveniles presentaron los siguientes porcentajes promedio del contenido estomacal: 20.7 % OM, 12 % NCF y 67.9 % CF; y adultos: 11.4 % OM, 14.8 % NCF y 73.8 % CF. Con base en una prueba de Wilcoxon y una regresión lineal de Kendall, se obtuvieron los siguientes resultados. La fracción carbonatada en el estómago aumentó en promedio 0.47 unidades por cada cm de crecimiento (p < 0.05). La MO consumida por D. mexicanum aumenta solo 0.05 unidades por cada cm de crecimiento (p < 0.05). Encontramos una diferencia en el contenido estomacal según el sitio (p < 0.05) y el estadio de vida (P < 0.05). Localidades como Huatulco y Coco presentaron diferencias significativas que podrían estar relacionadas con las condiciones oceanográficas locales. Conclusiones: Relacionamos estos cambios de la composición porcentual del contenido estomacal con la necesidad de los erizos de mar juveniles de nutrientes para mantener su crecimiento. La cantidad de MO es crucial para el desarrollo de las primeras etapas, lo que significa que existe una diferencia en la selección de sustrato asociada con el crecimiento.

Abstract Introduction: Care towards nutrition is essential for the quality of a sustainable aquaculture product. Since the balance in food affects the growth and production of gametes. The circular economy is made possible through the use of discarded materials. Objective: The aim of this research was to study the fatty acid composition and metabolic pathways in the gametes of Arbacia dufresnii, with a focus on the implications of incorporating shrimp byproducts into aquaculture feeds. Methods: Four different treatments were designed to maintain optimal nutritional quality, particularly in lipids and proteins, based on previous studies. The fatty acid profiles of the feeds and gametes were analyzed by using gas-chromatography, and statistical analyses were conducted to determine significant differences. Results: Significant differences were observed in the abundance (%) of omega-3 (ω-3) and omega-6 (ω-6) fatty acids. The (ω-3) metabolic pathway was more pronounced in the gametes of wild animals and those fed with the experimental feeds. In contrast, the (ω-6) metabolic pathway was less relevant in these groups. The (ω-3) /(ω-6) ratio was highest in the gametes of wild animals. Feeds enriched in fatty acids enhanced their bioaccumulation in the gametes reaching higher concentrations than wild animals. The availability of fatty acids in foods allowed their bioaccumulation in gametes, with concentrations equal to or higher than those observed in animals in their natural environment for certain fatty acids. Conclusions: Incorporating shrimp byproducts in aquaculture feeds demonstrated a promising strategy for resource utilization and organic input generation. The fatty acid composition in the gametes of A. dufresnii was influenced by the diet, highlighting the potential of balanced feeds to enhance the bioaccumulation of essential fatty acids. These findings provide valuable insights for the development of sustainable aquaculture practices and the production of nutritionally enriched seafood products.


Resumen Introducción: El cuidado hacia la nutrición es fundamental para la calidad de un producto acuícola sostenible. Ya que el balance en los alimentos afecta el crecimiento y producción de los gametos. A partir del aprovechamiento de materias de descarte se posibilita la economía circular. Objetivo: El objetivo de este estudio fue investigar la composición de ácidos grasos y las vías metabólicas en los gametos de Arbacia dufresnii, centrándose en las implicaciones de la incorporación de subproductos de camarones en los alimentos de acuicultura. Métodos: Se diseñaron cuatro tratamientos diferentes para mantener una calidad nutricional óptima, especialmente en lípidos y proteínas, basándose en estudios previos. Se analizaron los perfiles de ácidos grasos de los alimentos y los gametos mediante cromatografía de gases, y se realizaron análisis estadísticos para determinar diferencias significativas. Resultados: Se observaron diferencias significativas en la abundancia (%) de ácidos grasos omega-3 (ω-3) y omega-6 (ω-6). La vía metabólica de (ω-3) fue más pronunciada en los gametos de los animales en su entorno natural y aquellos alimentados con los piensos experimentales. Por el contrario, la vía metabólica de (ω-6) tuvo menos relevancia en estos grupos. La relación (ω-3) /(ω-6) fue más alta en los gametos de los animales en su entorno natural. La disponibilidad de ácidos grasos en los alimentos permitió su bioacumulación en los gametos, con concentraciones iguales o superiores a las observadas en los animales en su entorno natural para ciertos ácidos grasos. Conclusiones: La incorporación de subproductos de camarones en los alimentos de acuicultura demostró ser una estrategia prometedora para la utilización de recursos y la generación de insumos orgánicos. La composición de ácidos grasos en los gametos de A. dufresnii fue influenciada por la dieta, destacando el potencial de los alimentos balanceados para mejorar la bioacumulación de ácidos grasos esenciales. Estos hallazgos brindan información valiosa para el desarrollo de prácticas sostenibles en acuicultura y la producción de productos marinos enriquecidos nutricionalmente.

Resumen Introducción: Los equinoideos irregulares se caracterizan por tener una selección del sustrato en fondos blandos principalmente. En El Salvador se han registrado siete especies de equinoideos irregulares en las principales playas arenosas del país, M. stokesii es una de ellas. Objetivo: Determinar la abundancia, densidad y estructura de tallas, en playones de la bocana del estero El Tamarindo, El Salvador. Métodos: La investigación se realizó en época seca en el oriente del país en playones del estero El Tamarindo que se caracteriza por presentar grandes extensiones de arena mezclada con materia orgánica. En cada sitio, se trazaron transectos en banda paralelos a la costa mediante 9 cuadrantes de 10 m2 separados entre sí por 10 m (área total de 90 m2), en donde se contabilizaron y midieron los individuos de M. stokesii que se encontraran dentro del área delimitada. Resultados: Se contabilizaron 958 individuos de M. stokesii con una densidad total de 10.64 ± 13.22 ind/m2. El rango de tamaños fue de 1-6.5 cm. El hábitat se caracterizó por presentar arena con materia orgánica en los primeros mm del sustrato en compañía de otros invertebrados. Conclusiones: La abundancia y densidad de M. stokesii es similar a la registrada en otros estudios de la región.


Abstract Introduction: Irregular echinoids are characterized by having a selection of the substrate mainly on soft bottoms. In El Salvador, seven species of irregular echinoids have been recorded on the main sandy beaches of the country, M. stokesii is one of them. Objective: Determine the abundance, density and size structure of M. stokesii on beaches at the mouth of the El Tamarindo estuary, El Salvador. Methods: The research was carried out in the dry season in the east of the country on the beaches of the El Tamarindo estuary, which is characterized by large extensions of sand mixed with organic matter. At each site, band transects parallel to the coast were plotted by 9 quadrants of 10 m2 separated from each other by 10 m (total area of 90 m2), where individuals of M. stokesii found within the delimited area were counted and measured. Results: 958 individuals of M. stokesii were counted with a total density of 10.64 ± 13.22 ind/m2. The size range was 1-6.5 cm. The habitat was characterized by presenting sand with organic matter in the first mm of the substrate in the company of other invertebrates. Conclusions: The abundance and density of M. stokesii are similar to the ones recorded in other studies in the region.

Resumen Introducción: La industria acuícola está en constante crecimiento, registrando una producción mundial de casi 88 millones de toneladas para el año 2020. Esta industria trae consigo problemas ambientales si sus efluentes no son debidamente tratados. En el 2020, se constituyó la primera empresa de base tecnológica del CONICET en la Patagonia Argentina cuyo propósito es la producción acuícola del erizo verde de mar, Arbacia dufresnii con la finalidad de elaborar una gama de productos nutracéuticos. Su sistema de cultivo conlleva un compromiso de sustentabilidad desde su creación, y sin embargo genera efluentes con niveles altos de nitratos y fosfatos. Objetivo: Ante este escenario, y valorizando la biorremediación como herramienta de tratamiento de aguas, se propone en este trabajo la utilización de las microalgas marinas como agentes fitorremediadores del efluente acuícola. Métodos: Se utilizaron las microalgas Chaetoceros gracilis, Navicula sp., Tetraselmis suecica., Rhodomona salina., Nanochloropsis galvana y Cylindrotheca closterium, las cuales son usadas como alimento de las larvas del erizo en el proceso productivo. Se diseñó un experimento que compara el crecimiento microalgal y la capacidad de remoción de los nutrientes en el efluente en contraste con el medio de cultivo artificial actualmente usado en el ciclo productivo. Resultados: Es posible remediar el efluente de la industria acuícola mediante las microalgas seleccionadas, con porcentaje de eficacia de remoción del 100 % del nitrato y un porcentaje de eficacia de remoción promedio de 50 % para todas las microalgas testeadas. Asimismo, se obtuvieron valores de biomasa microalgal significativamente mayores cuando el cultivo fue realizado en el efluente respecto del cultivo en el medio artificial. Conclusiones: Los avances en investigación proporcionados en este trabajo ponen de manifiesto que es posible el aprovechamiento de un descarte para cultivar las microalgas, incluso mejorando la productividad microalgal para su uso como alimento, disminuyendo los costos involucrados en el sector de producción microalgal cambiando el uso del tipo de medio de cultivo actual (F/2) por el proveniente de un descarte. Estos avances si son escalados y validados, pueden mejorar los estándares de sustentabilidad de la industria en el marco de una economía circular.


Abstract Introduction: The aquaculture industry is constantly growing, registering a global production of almost 88 million tonnes by 2020. This industry brings environmental problems if its effluents are not properly treated. In 2020, the first technology-based company of CONICET was established in Argentine Patagonia whose purpose is the aquaculture production of the green sea urchin, Arbacia dufresnii to develop a range of nutraceutical products. Its cultivation system entails a commitment to sustainability since its creation, and yet it generates effluents with high levels of nitrates and phosphates. Objective: Given this scenario, and valuing bioremediation as a water treatment tool, the use of marine microalgae as phytoremediating agents of aquaculture effluent is proposed in this work. Methods: The microalgae Chaetoceros gracilis, Navicula sp., Tetraselmis suecica, Rhodomona salina, Nanochloropsis galvana and Cylindrotheca closterium were use; which are used as food for sea urchins larvae in the production process. An experiment was designed that compares the microalgal growth and the removal capacity of nutrients in the effluent in contrast to the artificial culture medium currently used in the production cycle. Results: It is possible to remedy the aquaculture industry's effluent by employing the selected microalgae, with a percentage of removal efficiency of 100 % of the nitrate and an average removal efficiency percentage of 50 % for all the microalgae tested. Likewise, significantly higher microalgal biomass values were obtained when the culture was carried out in the effluent the culture in the artificial environment. Conclusions: The advances in research provided in this work show that it is possible to take advantage of a discard to cultivate microalgae, even improving microalgal productivity for use as food, reducing the costs involved in the microalgal production sector by changing the use of the type of current culture medium (F/2) for that from a current discard. These advances, if scaled and validated, can improve industry sustainability standards within the framework of a circular economy.

Abstract Introduction: The coral reefs of Isla del Coco National Park are some of the most pristine ecosystems on Earth. The sea urchin Diadema mexicanum (Diadematoida: Diadematidae) is a common inhabitant with a pivotal role in the ecology of this unique ecosystem. Objective: In this study, we identified the predominant predators of D. mexicanum and estimated the predatory consumption rates. We also determined predation rates at different sea urchin sizes and at sites with contrasting refuge availability. Methods: We use field video recording observations and tethering experiments. Results: The balistid Sufflamen verres and the labrid Bodianus diplotaenia were identified as predators of small and medium size D. mexicanum; the labrids Thalassoma grammaticum and B. diplotaenia (juvenile) and the tetraodontid Arothron meleagris were attempted predators; and Canthigaster punctatissima and Holacanthus passer were scavengers. Larger sea urchins (> 30 mm) were also preyed upon during the tethering experiments. Furthermore, a clear effect of the site on survival of the different sea urchins' sizes was noted. No difference in the sea urchin predator biomass was found among sites, which highlights the importance of site complexity on survival. At high and medium complexity sites, large individuals had better survival, while at the low complexity site, there was almost no differences in survival rates among the three size classes. Our results also show that a high abundance of these predatory fishes, above 0.04 ind m-2, guarantees a low sea urchin density. Below this threshold, a higher variability in sea urchin density is observed Despite not being registered with the video recordings, lobsters were observed once preying upon a large sea urchin individual. Conclusions: This study identifies a keystone fish guild with high predation rate for Diadema in the National Park, which suggests that protective actions have positively benefited predatory fish and lobster populations.


Resumen Introducción: Los arrecifes de coral del Parque Nacional Isla del Coco son uno de los ecosistemas más prístinos de la Tierra. El erizo de mar Diadema mexicanum (Diadematoida: Diadematidae) es un habitante común con un papel ecológico esencial en este ecosistema único. Objetivo: En este estudio, identificamos los depredadores predominantes de D. mexicanum y estimamos las tasas de consumos predatorias. También determinamos las tasas predatorias de diferentes tamaños de erizo de mar en sitios con disponibilidad de refugio contrastante. Métodos: Utilizamos grabaciones de video de campo y experimentos de marcaje. Resultados: El pez ballesta Sufflamen verres y el lábrido Bodianus diplotaenia fueron identificados como depredadores de tamaños pequeños y medianos de D. Mexicanum; los lábridos Thalassoma grammaticum y B. diplotaenia (juvenil) y el tetraodóntido Arothron meleagris fueron intento de depredadores; y Canthigaster punctatissima y Holacanthus passer fueron carroñeros. Los erizos de mar de gran tamaño (> 30 mm) también fueron depredados durante el experimento de marcaje. Además, se encontró un efecto claro del sitio en la supervivencia de los diferentes tamaños de erizo de mar. No se encontraron diferencias en la biomasa de los depredadores del erizo de mar entre sitios, lo que señala la importancia de la complejidad del sitio en la supervivencia. En sitios con complejidad estructural alta y media, los individuos grandes tuvieron mejor supervivencia, mientras que en sitios de complejidad baja apenas hubo diferencias en las tasas de supervivencia entre los tres tamaños. Nuestros resultados también muestran que una alta abundancia de peces depredadores, por encima de 0.04 ind m-2, asegura bajas densidades de erizos de mar. A pesar de no ser registrado durante las grabaciones de video, se observó en una ocasión a una langosta depredando sobre un erizo de gran tamaño. Conclusiones: Este estudio identifica el grupo de peces clave con grandes tasas de depredación sobre Diadema en el Parque Nacional, lo que sugiere que las medidas de protección han beneficiado positivamente las poblaciones de peces depredadores y langostas.

Abstract Introduction: Even though only a few species are considered to be dangerous, pests or vectors, the majority of invertebrates produce a feeling of aversion in humans. This has contributed to the delay in the development of ethical considerations as regards this group in contrast with vertebrates, with the exception of cephalopods. Objective: In the present study, we provide an overview of the current situation on animal ethics and welfare in order to contribute to the development of a framework for ensuring invertebrate welfare. Methods: Today, animal welfare is multidisciplinary in nature to a very high degree as it includes ethology, physiology, pathology, biochemistry, genetics, immunology, nutrition, cognitive-neural, veterinary medicine, and ethics. Animal welfare is a complex concept, difficult to achieve successfully from one perspective. Results: As a consequence, we propose to include the five domains (nutrition, environment, health, behaviour and mental state) along with the three conceptions (basic health and functioning, affective state and natural living), as well as the 5R Principle (Replace, Reduction, Refinement, Respect and Responsibility) in seeking to achieve a comprehensive welfare state. Conclusions: We consider that in both research and animal production, the individual and collective ethical concerns coexist and, in fact, the main moral concern to account for is the collective one and that, within that collective view, the individual moral concern should be applied with responsibility and respect for the individual. Finally, we propose a practical example of invertebrate welfare production in sea urchin aquaculture with the aim of including animal production of invertebrates in this important discussion.


Resumen Introducción: Aunque sólo unas pocas especies son consideradas peligrosas, plagas o vectores, la mayoría de los invertebrados producen un sentimiento de aversión en el ser humano. Esto ha contribuido al retraso en el desarrollo de consideraciones éticas respecto a este grupo en comparación con los vertebrados, a excepción de los cefalópodos. Objetivo: En el presente trabajo, proporcionamos una visión general de la situación actual en materia de ética y bienestar animal con el fin de contribuir al desarrollo de un marco para garantizar el bienestar de los invertebrados. Métodos: Hoy en día, el bienestar animal es de naturaleza multidisciplinaria en un grado muy alto, ya que incluye etología, fisiología, patología, bioquímica, genética, inmunología, nutrición, cognitivo-neural, medicina veterinaria y ética. El bienestar animal es un concepto complejo, difícil de lograr con éxito desde una sola perspectiva. Resultados: Como consecuencia, proponemos incluir los cinco dominios (nutrición, ambiente, salud, comportamiento y estado mental) junto con las tres concepciones (Salud básica y funcionamiento, estado afectivo y vida natural), así como el Principio 5R (Reemplazar, Reducir, Refinar, Respetar y Responsabilidad) en la búsqueda de alcanzar un estado de bienestar integral. Conclusiones: Consideramos que tanto en la investigación como en la producción animal coexisten las preocupaciones éticas individuales y colectivas y, de hecho, la principal preocupación moral a dar cuenta es la colectiva y que, dentro de esa visión colectiva, se debe aplicar la preocupación moral individual. con responsabilidad y respeto por la persona. Finalmente, proponemos un ejemplo práctico de producción de bienestar de invertebrados en la acuicultura de erizos de mar con el objetivo de incluir la producción animal de invertebrados en esta importante discusión.

Without general adaptative immunity, invertebrates evolved a vast number of heterogeneous non-self recognition strategies. One of those well-known adaptations is the expansion of the immune receptor gene superfamily coding for scavenger receptor cysteine-rich domain containing proteins (SRCR) in a few invertebrates. Here, we investigated the evolutionary history of the SRCR gene superfamily (SRCR-SF) across 29 metazoan species with an emphasis on invertebrates. We analyzed their domain architectures, genome locations and phylogenetic distribution. Our analysis shows extensive genome-wide duplications of the SRCR-SFs in Amphimedon queenslandica and Strongylocentrotus purpuratus. Further molecular evolution study reveals various patterns of conserved cysteines in the sponge and sea urchin SRCR-SFs, indicating independent and convergent evolution of SRCR-SF expansion during invertebrate evolution. In the case of the sponge SRCR-SFs, a novel motif with seven conserved cysteines was identified. Exon-intron structure analysis suggests the rapid evolution of SRCR-SFs during gene duplications in both the sponge and the sea urchin. Our findings across nine representative metazoans also underscore a heightened expression of SRCR-SFs in immune-related tissues, notably the digestive glands. This observation indicates the potential role of SRCR-SFs in reinforcing distinct immune functions in these invertebrates. Collectively, our results reveal that gene duplication, motif structure variation, and exon-intron divergence might lead to the convergent evolution of SRCR-SF expansions in the genomes of the sponge and sea urchin. Our study also suggests that the utilization of SRCR-SF receptor duplication may be a general and basal strategy to increase immune diversity and tissue specificity for the invertebrates.

BACKGROUND: Some marine invertebrate organisms are considered not to develop tumors due to unknown mechanisms. To gain an initial insight into how tumor-related genes may be expressed and function during marine invertebrate development, we here leverage sea urchin embryos as a model system and characterize the expressions of Myc and p53/p63/p73 which are reported to function synergistically in mammalian models as an oncogene and tumor suppressor, respectively. RESULTS: During sea urchin embryogenesis, a combo gene of p53/p63/p73 is found to be maternally loaded and decrease after fertilization both in transcript and protein, while Myc transcript and protein are zygotically expressed. p53/p63/p73 and Myc proteins are observed in the cytoplasm and nucleus of every blastomere, respectively, throughout embryogenesis. Both p53/p63/p73 and Myc overexpression results in compromised development with increased DNA damage after the blastula stage. p53/p63/p73 increases the expression of parp1, a DNA repair/cell death marker gene, and suppresses endomesoderm gene expressions. In contrast, Myc does not alter the expression of specification genes or oncogenes yet induces disorganized morphology. CONCLUSIONS: p53/p63/p73 appears to be important for controlling cell differentiation, while Myc induces disorganized morphology yet not through conventional oncogene regulations or apoptotic pathways during embryogenesis of the sea urchin.

The intricate process of biomineralization, e.g. in sea urchins, involves the precise interplay of highly regulated mineralization proteins and the spatiotemporal coordination achieved through compartmentalization. However, the investigation of biomineralization effector molecules, e.g. proteins, is challenging, due to their very low abundance. Therefore, we investigate the functional mimicry in the bioinspired precipitation of calcium carbonate (CaCO3) with artificial peptides selected from a peptide library by phage display based on peptide-binding to calcite and aragonite, respectively. The structure-directing effects of the identified peptides were compared to those of natural protein mixes isolated from skeletal (test) structures of two sea urchin species (Arbacia lixula and Paracentrotus lividus). The calcium carbonate samples deposited in the absence or presence of peptides were analyzed with a set of complementary techniques with regard to morphology, polymorph, and nanostructural motifs. Remarkably, some of the CaCO3-binding peptides induced morphological features in calcite that appeared similar to those obtained in the presence of the natural protein mixes. Many of the peptides identified as most effective in exerting a structure-directing effect on calcium carbonate crystallization were rich in basic amino acid residues. Hence, our in vitro mineralization study further highlights the important, but often neglected, role of positively charged soluble organic matrices associated with biological and bioinspired CaCO3 deposition.
