RESUMO

A new species of nematode, Ascarophis morronei n. sp. (Cystidicolidae), is described from the stomach wall of the woolly sculpin Clinocottus analis (Cottidae) collected in the rocky intertidal from northwestern Baja California, Mexico. Collected nematodes were studied using both light and scanning electron microscopy. Sequence fragments for 18S rDNA molecular markers were obtained from the new nematode species, in order to test its position within the family Cystidicolidae under a phylogenetic context. Main characters distinguishing this new species include the reduced labia and the morphology of the eggs, distances of nerve ring and excretory pore from the anterior end, and left spicule of males. The new species described here is the second for the genus Ascarophis reported as adult in the Southern California Bight, and the first one recorded for the fish genus Clinocottus.

Cystidicoloides vaucheri collected in the stomach of the redtail catfish Phractocephalus hemioliopterus from River Acre, State of Acre, Brazil is redescribed, including the first description of males and the first genetic characterization based upon 18S and 28S genes of the rRNA. Newly collected females were biometrically smaller than those reported in the original description, but similar morphology shared by the two samples revealed that they belong to the same species. Scanning electron micrographs showed the accurate structure of the cephalic region, described here in detail. Furthermore, the morphology of males completed the specific diagnosis, strengthening the validity of the species. The three other congeners differ from C. vaucheri mainly as follows: in C. dlouhyi the area rugosa is absent, the cephalic structures in C. fischeri are completely distinct, and in both species the spicules have membranous outgrowths, absent in C. vaucheri. Despite the dubious generic assignment of C. izecksohni, it differs from C. vaucheri in several biometrical and morphological features. Because of data availability, only sequences of the 18S were used for phylogenetic reconstructions. Results showed that the genus Ascarophis and the families Cystidicolidae and Physalopteridae are not monophyletic. Cystidicoloides vaucheri formed an independent branch clustering with representatives of Cystidicolidae, confirming its validity. The inclusion of Salmonema and Spinitectus within Cystidicolidae should be reviewed, since they formed an assemblage with species from Rhabdochonidae. In fact, current classification of some taxa belonging to Habronematoidea, Physalopteroidea and Thelazioidea need to be re-evaluated, mainly based on molecular data from different genes.

Specimens of the genus Gongylonema were collected from the gastric mucosa of rodents of Rattus rattus Linnaeus, 1758, and Rattus norvegicus Berkenhout, 1769, collected in urban areas in Belém, Pará, in the eastern Brazilian Amazon. The helminths were processed for analysis using light microscopy and scanning electron microscopy (SEM) techniques and presented taxonomic characteristics of the species Gongylonema neoplasticum. The SEM analyses revealed the presence of 2 developed buccal plates (1 dorsal, 1 ventral), also called interlabia, with a prominent and bifurcated ventral plaque. The occurrence of the bifurcated ventral interlabium had not yet been identified by any other author from G. neoplasticum. As a result of our extensive research on published data on Gongylonema spp., we propose a taxonomic key for species of this genus that parasitize rodents. This is the first record of G. neoplasticum in urban areas of the Brazilian Amazon.

Nematodes of the cystidicolid Neoascarophis Machida, 1976 , are all parasites of macrourid fishes, making up at present 5 species. Several other unidentified species have also been reported in several fish species from the northern and southern Atlantic Ocean, including 1 from Macrourus carinatus (Günther) (Macrouridae) in the southwest Atlantic Ocean. During a parasitological survey carried out on samples of M. carinatus from Patagonian waters, nematodes referable to Neascarophis were found in ulcers in the gastric mucosa. These nematodes Neascarophis sphaerocaudata n. sp. closely resemble N. macrouri by the posterior position of the vulva and the dilated posterior extremity in females. However, the new species differs from N. macrouri mainly by its larger size, a larger muscular esophagus, and a widely globose posterior extremity in females. SEM study of cephalic structures also showed morphological differences between both species, especially in the morphology of the submedian labia and lateral pseudolabia. In view of these differences a new species is proposed.

Two new nematode species of the family Cystidicolidae, each representing a new genus, were recovered from marine perciform fishes off New Caledonia, South Pacific: Ascarophisnema tridentatum n. gen., n. sp. from the stomach of the Japanese large-eye bream, Gymnocranius euanus (Günther) (Lethrinidae) and Metabronemoides mirabilis n. gen., n. sp. from the stomach of the painted sweetlip, Diagramma pictum (Thunberg) (Haemulidae). Ascarophisnema is characterized mainly by its cephalic structures (presence of two tooth-like projections on either side of the base of each pseudolabium, dorsal and ventral inner extensions of each pseudolabium recurved laterally in apical view, and submedian sublabia fused together dorsally and ventrally) and the presence of trident-like deirids, and Metabronemoides by its unique cephalic structures (presence of one dorsal and one ventral labium and four large dorsolateral and ventrolateral labia, and absence of sublabia). Rhabdochona gymnocranius (considered a species inquirenda) is provisionally transferred to the former genus as Ascarophisnema gymnocranius (Yamaguti, 1935) n. comb. To date, a total of seven species of cystidicolids are reported from marine fishes off New Caledonia.

During a parasitological survey of Galaxias maculatus (Jenyns) from Patagonian Andean Lakes, specimens of a new species of nematode were collected from the stomach of fish and studied by light and scanning electron microscopy. This species is described as the only member of a newly proposed genus of Cystidicolidae by having oral opening dorsoventrally elongated, 2 lateral pseudolabia, 4 cephalic papillae, stoma (vestibule) elongated, esophagus divided into anterior muscular and posterior glandular sections, and caudal alae in males. Placonema n. gen. (Habronematoidea, Cystidicolidae) is characterized by the combination of the following features: oral opening dorsoventrally elongated demarcated by 4 sclerotized plates and 2 well-developed pseudolabia projected to the buccal cavity, each pseudolabium with conspicuous, conical, anterior protuberances. Four cephalic papillae and deirids simple. Male with caudal alae, area rugosa absent, 4 pairs of preanal papillae, unpaired papilla present on anterior cloacal lip, and 6 pairs of postanal papillae. Larvigerous eggs without filaments. Placonema pataguense n. gen. n. sp. infects the stomach of G. maculatus from Lake Patagua (Patagonia, Argentina) and is the first species of Cystidicolidae described from G. maculatus.

The cystidicolid nematode Cystidicoloides fischeri (Travassos, Artigas and Pereira, 1928) is redescribed from specimens collected from the stomach of the San Francisco piranha, Pygocentrus piraya (Cuvier), and the white piranha, Serrasalmus brandtii (Lütken) (both Characidae, Characiformes) (new host records), from the Três Marias Reservoir, Upper São Francisco River, Minas Gerais state, Brazil. The morphology of this type species of Cystidicoloides Skinker, 1931, studied with both light and scanning electron microscopy, is characterized by some taxonomically important, previously unreported features, such as the presence of a cephalic cuticular collarette, subdorsal and subventral cephalic spikes, cuticular tooth-like elevations inside the prostom, deirids, area rugosa, and details in the structure of the cephalic end. Heliconema izecksohni Fabio, 1982 is transferred to Cystidicoloides as C. izecksohni (Fabio, 1982) n. comb. Cystidicoloides uniseriata Valovaya and Valter, 1988 is considered a species inquirenda with uncertain generic appurtenance. The presence of subdorsal and subventral cephalic spikes and the collarette are characteristic of Cystidicoloides, comprising only species parasitizing Neotropical fishes. The species from salmonids in the Holarctic, hitherto reported mostly as Cystidicoloides ephemeridarum (Linstow, 1872), belongs to a different, newly erected genus, for which the name Salmonema n. gen. (type species S. ephemeridarum) is now proposed. Cystidicoloides prevosti (Choquette, 1951) is transferred to Salmonema as S. prevosti (Choquette, 1951) n. comb. Sterliadochona ssavini Skryabin, 1948 and Sterliadochona Skryabin, 1948 are considered as species inquirenda and a genus inquirendum, respectively. A key to species of Cystidicoloides is provided.

RESUMO

Scanning electron microscopy of the cystidicolid nematode Ascarophis mexicana Moravec, Salgado-Maldonado et Vivas-Rodríguez, 1995 enabled the first detailed study of its cephalic structure. In contrast to most Ascarophis species, its pseudolabia are highly reduced and sublabia are unlobed and weakly developed. Similascarophis is considered a synonym of Ascarophis, to which two its species are transferred as A. maulensis (Muñoz, González et George-Nascimento, 2004) comb. n. and A. chilensis (Muñoz, González et George-Nascimento, 2004) comb. n.

Specimens of a little-known nematode, Spinitectus oviflagellis Fourment, 1884, the type-species of Spinitectus Fourment, 1884, were collected mainly from the pyloric caeca of a marine deep-water fish, the onion-eye grenadier Macrourus berglax Lacépède (a new host record), in the eastern Greenland Sea, North Atlantic Ocean. Studies using light and scanning electron microscopy revealed some taxonomically important, previously unreported features of S. oviflagellis, such as the detailed structure of the cephalic end, the position of the excretory pore and the presence of ventral pre-anal cuticular ridges (area rugosa) in the male, which indicated a certain degree of intraspecific biometrical variability in this species. S. oviflagellis is compared with similar congeneric species parasitising marine fishes.

In this study, we describe 2 new species of Ascarophis van Beneden, 1871 (Nematoda: Cystidicolidae), found in fishes from southern Chile. Ascarophis carvajali n. sp. was found in Austrolycus depressiceps and Patagonotothen cornucola, whereas Ascarophis draconi n. sp. was taken from Champsocephalus gunnari. These new Ascarophis species differ from other species in a combination of several morphometric and morphological characteristics. Although A. carvajali n. sp. was morphologically close to Ascarophis minuta, the new species has a larger ratio between glandular and muscular esophagus, filaments on both egg poles, and a shorter right spicule than A. minuta. Ascarophis draconi n. sp. was morphologically similar to Ascarophis adioryx and Ascarophisfiliformis. However, A. adioryx has eggs without filaments, a smaller ratio between glandular and muscular esophagus length, and a smaller ratio between left and right spicule lengths in contrast to A. draconi n. sp., whereas A. filiformis has a shorter glandular esophagus and left spicule length than A. draconi n. sp. Only 1 Ascarophis species has been recorded in a single fish from Chile (i.e., Ascarophis sebastodis in Sebastes capensis). Consequently, this study constitutes not only new species and records of Ascarophis in fishes from Chile, but also new records for the Pacific coast of South America.

The cystidicolid nematode Metabronema magnum (Taylor, 1925) is redescribed from specimens collected from the swimbladder of the fish (golden trevally) Gnathanodon speciosus (Forsskål) (Carangidae, Perciformes) off New Caledonia, South Pacific (a new geographical record). The light and scanning electron microscopical examination made it possible to study in detail the morphology of this so far little-known species. Its pseudolabia were found to possess distinct anterior protrusions (protuberances), sublabia are absent, only four cephalic papillae are present, deirids are bifurcated, and the male possesses six pairs of postanal papillae. By its morphology, M. magnum seems to be most similar to species of Salvelinema Trofimenko, 1962, also from the swimbladder of fishes, differing from them mainly in the presence of median wedge-shaped outgrowths in the mouth, lateral alae, the longer spicule on the right side, and a fewer number of pairs of preanal papillae in the male. Since the morphology of M. magnum considerably differs from that of other representatives of the Cystidicolidae, Metabronema in Rasheed's (1966) conception is considered a valid genus.

A new species of parasitic nematode, Neoascarophis macrouri n. sp. (Cystidicolidae), is described from the stomach and stomach wall of the marine deep-water fish Macrourus berglax (onion-eye grenadier) in the eastern Greenland Sea (North Atlantic Ocean). The new species, studied using both light and scanning electron microscopy, is characterised mainly by the location of the vulva near the posterior end of the body (a short distance anterior to the anus), non-filamented eggs, the structure of the mouth, a short vestibule and the length of the spicules (567-615 and 144-156 mum). Metabronema insulanum Solov'eva, 1991 is transferred to Neoascarophis as N. insulana (Solov'eva, 1991) n. comb.

Ascarophis valentina n. sp. is described from Mullus surmuletus off the Valencian coast of Spain on the basis of both light and scanning electron microscopy. It can be distinguished from the other members of the genus by the length of the left (long) spicule of the males and by egg morphology. An updated grouping of the species of Ascarophis considered valid is provided with respect to these characters. The new species resembles Ascarophis capelanus, belonging to the group of species possessing eggs with a single polar knob with filaments, but is distinguished by the size of the body, the length of the esophagus (especially in relation to body length), the position of the vulva, and the size of the left spicule. The new species also shows substantial morphological differences compared with the 3 species, Ascarophis mullusi, Ascarophis upenei, and Ascarophis parupenei, previously described from mullid hosts.

An experimental infection model for the heteroecious spiruid nematode Tetrameres americana (Cram 1927) was developed. The cockroach Blattella germanica (L.) and the locust Locusta migratoria (L.) were found to serve as intermediate hosts for the parasite. T. americana larvae developed to full maturity in these intermediate hosts and were infective to young Lohman Brown chickens after 32 days in the cockroach and 28 days in the locust. The maximum length of the larvae was reached in the insects at 28-30 degrees C after 10-15 days, at which time the larvae measured up to 2.2 mm. The parasite did not develop in the cockroach Periplaneta americana (L.), the woodlouse Oniscus asellus (L.), or the pupal stage of the giant mealworm Zophobas morio (Fabricius). Trials in which chickens were infected directly without an intermediate host failed. Infection of 24 chickens with a dosage of 100 larvae was followed by weekly post-mortems until day 48 post-infection (p.i.) and used to describe the development of T. americana. The average establishment rate (%) and the average worm burden varied from 16.5 to 30.8. The total numbers of parasites recovered ranged from 9 to 40. During mating, in the first 2 weeks p.i. females and males were equally abundant, whereas from day 20 p.i. twice as many females were recovered. From day 13 p.i. the females average length fluctuated between 2.6 and 3.7 mm, whereas they reached their maximum width of 2.4 mm on day 48 p.i. Males reached their full length after 27 days p.i. and measured up to 6.7 mm.

Similascarophis (Cystidicolidae) n. gen. is proposed. In the mouth of specimens of this genus, submedial labia are absent and pseudolabia do not have any part projecting toward the central oral opening. These nematodes were obtained from the alimentary tract of 7 marine fish species along the coast of Chile: Bovichthys chilensis Regan, Eleginops maclovinus (Cuvier), Pinguipes chilensis (Valenciennes), Cilus gilberti (Abbott), Cheilodactylus variegatus Valenciennes, Girella laevifrons (Tschudi), and Graus nigra Philippi. Morphology and morphometry are compared between 2 new Similascarophis species: Similascarophis maulensis n. sp. and S. chilensis n. sp., which differ in the presence of sublabia and in the length of the glandular esophagus and left spicule. We also recorded Similascarophis sp. in 2 other host species, which showed some distinct proportional measurements, although these differences were not sufficiently clear to identify them as a new species.

Spinitectus macrospinosus n. sp., a parasite of the channel catfish Ictalurus punctatus, from the Red and Assiniboine rivers in southern Manitoba, Canada, is characterized by spines reaching a maximum length of 21.8-26.8 microm (means for males and females, respectively) in the anterior esophageal region, arranged in 4 sectors in the anterior region of the body. Spines in this region of the body increase in size and decrease in number (from a maximum of 6 to 3 per sector). The combination of spine length, number, and arrangement and the position of the excretory pore (between spine rows 6 and 7) readily distinguishes it from all its North American congeners, including the species it is most similar to, S. carolini. It is different from S. gracilis and S. acipenseri in having a posteriorly directed vagina, an excretory pore between spine rows 6 and 7, a longer stoma, a right spicule with a terminal ventral barb, and a heart-shaped caudal mucron. Spinitectus macrospinosus is similar to other North American species of Spinitectus, e.g., S. carolini, S. micracanthus, S. mexicanus, S. osorioi, and S. humbertoi, in having the anterior rows of spines arranged in 4 sectors. It is also similar to S. micracanthus and S. carolini in possessing a relatively long stoma, a posteriorly directed vagina, a right spicule with a terminal ventral barb, and a heart-shaped caudal mucron. Furthermore, it is similar to S. carolini in possessing an "area rugosa" with 2 rows of precloacal cuticular cleats. In southern Manitoba, S. macrospinosus appears to mature only in the channel catfish. Reexamination of museum specimens revealed that the nematode also is found in I. furcatus in Kentucky Lake (Kentucky-Tennessee) and in I. lacustris in Lake Texoma (Oklahoma-Texas).

Larval nematode parasites (Spiruroidea: Cystidicolidae) are recorded for the first time in Octopus vulgaris Cuvier, 1797 in the northeastern Atlantic Ocean. Prevalence was 16% and mean intensity was 1.46 worms/host. Body length of larval nematodes ranges from 8.3 to 9.3 mm, with a distance from the anterior end to nerve ring from 187.5 to 200 microm, and to excretory pore 194.6-350 microm. Anatomical characteristics, such as deirid, nerve ring, cephalic alae, excretory pore, pseudolabia amphids, sclerotized protuberance, and anus, examined using light microscopy (LM) or scanning electron microscopy (SEM), are illustrated. The nematode was designed as a cystidicolid "Type A" larva. The hemocytic infiltration present in the host tissue around the nematode capsule and the mechanical compression in the infected organs denote the pathogenicity of this nematode. In the study area, O. vulgaris may play the role of an intermediate or paratenic host in the nematode life cycle.

The histopathology induced by Camallanus oxycephalus and Spinitectus carolini in the intestine of green sunfish Lepomis cyanellus was examined. At what is assumed to be more recent attachment sites, penetration of C. oxycephalus was restricted to the mucosal layer, causing complete destruction of the columnar epithelium. At what appeared to be older attachment sites, the parasite penetrated deep into the intestinal wall, even to the circular muscle layer. At these sites of attachment, there were flask-shaped ulcers in the mucosal and submucosal layers. Surrounding the ulcers, there was granulomatous tissue, with extensive fibrosis at the surface; the fibrosis probably reduced the loss of body fluids and blood cells from the intestinal wall. The number of goblet cells in the posterior end of the intestine increased significantly, from 92.6 cells/mm in uninfected fish to 168.4 cells/mm in fish infected with more than 5 C. oxycephalus. Adult S. carolini also causes severe damage to the intestinal wall of green sunfish. Parasite penetration was not limited to the mucosal layer. Mature worms were observed invading both the muscular and serosal layers. Penetration by the parasite was usually associated with the local loss of columnar epithelium and infiltration by lymphocytes and granulocytic leukocytes. Fibrosis in the mucosal layer was commonly observed. There was goblet cell hyperplasia in the anterior portion of the intestine, with numbers of goblet cells increasing from 64.4 cells/mm in uninfected fish to 95.0 cells/mm in heavily infected fish.

Durante los períodos abril-agosto de 1989-1990 se examinaron los estómagos e intestinos de 129 ejemplares de Dusycion griseus, 22 de D. culpaeus y 22 de conepatus de la Provincia de Neuquén, Argentina. Se recolectaron cuatro especies de nematodes (Physaloptera clausa, Ph. maxillaris, Protospirura numidica criceticola y Toxascaris leonina). La posición sistemática de Ph. clausa Rudolphi, 1819, fue comentada habiendo concluído que representa a la especie tipo del género. Se consideró la posibilidad de que D. grisaseus y D. culpaeus fueran los hospedadores accidentales de la subespecie P. n. criceticola. Se dan los valores de prevalencia, intensidad media y frecuencia de cada especie en los hospedadores correspondientes. Se relacionaron esos valores con las dietas de cada hospedador y los ciclos de vida de los parásitos. Se determinó que la carga parasitaria de cada hospedador es independiente de su estado nutricional (prueba no paramétrica Tau de Kendall) y que existen diferencias significativas entre las dietas de los hospedadores y entre los valores de prevalencia, intensidad media y frecuencia de las especies parásitas (prueba de Chi cuadrado de Homogeneidad)
