OBJECTIVE: To describe left recurrent laryngeal neurectomy (LRLn) performed under standing sedation and evaluate the effect of LRLn on upper respiratory tract function using a high-speed treadmill test (HST). We hypothesized that (1) unilateral LRLn could be performed in standing horses, resulting in ipsilateral arytenoid cartilage collapse (ACC); and (2) HST after LRLn would be associated with alterations in upper respiratory function consistent with dynamic ACC. ANIMALS: 6 Thoroughbred horses. METHODS: The horses were trained and underwent a baseline HST up to 14 m/s at 5% incline until fatigue. Evaluation included; airflow, pharyngeal and tracheal pressures, and dynamic upper respiratory tract endoscopy. Trans-laryngeal impedance (TLI) and left-to-right quotient angle ratio (LRQ) were calculated after testing. The following day, standing LRLn was performed in the mid-cervical region. A HST was repeated within 4 days after surgery. RESULTS: Standing LRLn was performed without complication resulting in Havemayer grade 4 ACC at rest (complete paralysis) and Rakestraw grade C or D ACC (collapse up to or beyond rima glottis midline) during exercise. Increasing treadmill speed from 11 to 14 m/s increased TLI (P < .001) and reduced LRQ (P < .001). Neurectomy resulted in an increase in TLI (P = .021) and a reduction in LRQ (P < .001). CLINICAL RELEVANCE: Standing LRLn induces laryngeal hemiplegia that can be evaluated using a HST closely after neurectomy. Standing LRLn may be useful for future prospective evaluations of surgical interventions for laryngeal hemiplegia.

Sex effects on ventilatory and oxygen consumption (V̇O2) measurements during exercise have been identified in humans. This study's aim was to evaluate the hypothesis that there are sex effects on ventilatory and V̇O2 measurements in exercising, untrained yearling Thoroughbreds (Tb). Forty-one Tbs (16 colts, 25 fillies; 19.8 ± 1.4 months old) were recruited. Physiological, ventilatory and exercise data were gathered from horses exercising unridden at high intensity on an all-weather track from a global positioning-heart rate unit and a portable ergospirometry system. Data were analysed with an unpaired Student's t-test and the Benjamini-Hochberg correction for multiple testing (P ≤ 0.05 significant). Mean bodyweight (BW, P = 0.002) and wither height (P = 0.04) were greater for colts than fillies. There were no differences in physiological and exercise data and absolute peak V̇O2 between groups. However, fillies had a higher mass specific peak V̇O2 (P = 0.03) than colts (121.5 ± 21.6 mL/kg.min vs. 111.9 ± 27.4 mL/kg.min). The peak breathing frequency was greater for fillies (P < 0.001) while the peak inspiratory (P < 0.001) and expiratory air flow (P < 0.001), peak expiratory tidal volume (VTE; P < 0.001) and peak minute ventilation (V̇E; P = 0.01) were greater for colts; there were no differences for peak VTE and V̇E when adjusted for BW. Differences in BW explain the differences in mass specific peak V̇O2 between groups. Given their morphological differences, it is likely that lung volumes and airway diameters are smaller for fillies, resulting in greater resistance and lower air flows and volumes. Further research is required to investigate the ventilatory differences and how they may change with maturation and impact performance.

BACKGROUND: Incline treadmill and underwater treadmill (UWTM) exercises are common canine rehabilitation modalities , which are often used in isolation in dogs recovering from spinal surgery. Early use of an incline during UWTM exercise may have the potential to improve rehabilitation outcomes in dogs, but, it is hypothesised that dorsoventral movement of the spine may be excessive meaning it is unsuitable in some circumstances. OBJECTIVES: The purpose of this study was to identify changes in canine spinal kinematics in dogs when using a dry treadmill at different angles of incline compared to an underwater treadmill using the same inclines. METHODS: Eight dogs were encouraged to walk on a dry, horizontal, underwater treadmill as well as under the same conditions with both a 10% and 20% incline. This was then repeated at a 10% and 20% incline with the addition of water to hock level. Data were collected using reflective anatomical markers placed at the occipital protuberance, T1, T13, L3, L7 and sacral apex, captured by a high-speed camera facing the lateral aspect of the treadmill. Dorsoventral motion of the spine as well as flexion, extension and range of motion (ROM) of T1, T13, L3 and L7 were recorded. RESULTS: We found significant differences in dorsoventral spinal ROM at T1, L3 and L7, but no significant differences in T13 ROM. No significant differences were found in flexion and extension of any of the joints assessed when comparing dry conditions to the use of water (P>0.05). CONCLUSIONS: The lack of significant differences in joint flexion and extension at T1, T13, L3 and L7 indicates the potential safe use of combining underwater treadmill and incline exercise in canine rehabilitation. However, a lack of uniformity in results makes distinguishing any patterns of significance difficult. More research is needed to establish the effects of these exercises in additional planes of motion before a treatment protocol can be established.

BACKGROUND: Despite its importance, there is no agreed definition of recovery of ambulation in dogs with spinal cord injury. OBJECTIVES: To validate a new walking test in dogs recovering from thoracolumbar spinal cord injury. ANIMALS: Two hundred twenty-four dogs weighing <20 kg: 120 normally ambulatory dogs, plus 104 dogs undergoing decompressive surgery for acute thoracolumbar intervertebral disc herniation. METHODS: Prospective cohort studies. The distance each freely-ambulatory dog walked during 50 step cycles was regressed on ulna length. For each postsurgical dog, we recorded when the calculated 50-step distance was completed without falling, or their inability to complete this distance by 4 months or more after surgery. Bayesian analysis compared outcomes for presurgical neurologic categories; association of recovery with several preoperative variables was explored using logistic and time-to-event regression. RESULTS: For control dogs, 50-step distance (m) = 1.384 × ulnar length (cm) + 2.773. In postsurgical dogs, the 50-step test provided decisive evidence that deep pain-negative dogs were less likely to recover ambulation than dogs with intact pain perception (12/29 recovered vs 71/75; Bayes factor [BF] = 5.9 × 106 ) and, if they did recover, it took much longer (median 91 days vs median 14 days; BF = 1.5 × 103 ). Exploratory analysis suggested that presurgical neurologic status (subhazard ratio [SHR] = 0.022; P < .001) and duration of presurgical anesthesia (SHR = 0.740; P = .04) were associated with rapidity of recovery. CONCLUSIONS AND CLINICAL IMPORTANCE: This straightforward 50-step walking test provides robust data on ambulatory recovery well-suited to large scale pragmatic trials on treatment of thoracolumbar spinal cord injury in dogs.

BACKGROUND: Glycogen in skeletal muscle is a major source of energy during exercise and an important determinant of endurance capacity, so that its measurement may provide a meaningful marker of athletes' preparation and a possible predictor of performance, both in humans and in equines. Gold standard of glycogen concentration measurement is the histochemical and biochemical analysis of biopsy-derived muscle tissue, an invasive and potentially injuring procedure. Recently, high-frequency ultrasound (US) technology is being exploited in human sports medicine to estimate muscle glycogen content. Therefore, aim of the present study is to evaluate the feasibility of US assessment of muscle glycogen in equines. RESULTS: US images of gluteus medius (GL) and semitendinosus (ST) muscles were obtained on eight healthy horses (3-10 years) before and after a steady-state exercise on treadmill (velocity: 4.0-12.5 m/s; duration: 2-20 min; heart rate: 137-218 b/min). Average image greyscale intensity was significantly different between GL and ST, both before and after exercise (p < 0.001). Comparing baseline and post-exercise US images, significant increase in greyscale intensity has been observed in ST (p < 0.001), but not in GL (p = 0.129). The volume of the exercise was significantly correlated with exercise-dependent change in image intensity (R2 = 0.891), consistent with a reduction of glycogen muscle stores resulting from aerobic activity. CONCLUSIONS: US technique evidences also in horses muscle changes possibly associated to glycogen utilisation during exercise. Present results on a small sample need to be further confirmed and provide preliminary data warranting future validation by direct glycogen measurement through biopsy technique.

Studies on biochemical constituents and hydration of horses in training or competitions are essential for the knowledge of their adaptive physiological and metabolic responses to the type of exercise inherent to the sport. The present research evaluated the effects of exercise on hydric status and physiologic and biochemical profiles in Quarter Horses during a barrel race training session. The design consisted of three evaluation times (T): at rest, before the start of physical activity (T0); immediately after a 10-minute warm-up trot and then running the barrel race courses twice with a 10-minute interval (T1); and after one hour's rest after the second course (T2). The parameters analyzed were: heart rate (HR), respiratory rate (RR), body temperature (BT) and mucose color; the capillary refill time (CRT), packed cell volume (PCV), total plasma proteins (TPP), serum creatinine (CRE) and blood urea nitrogen (BUN) were evaluated and used to infer water status; plasma osmolarity (Osm) was measured; the electrolytes sodium (Na), potassium (K), chloride (Cl), total calcium (tCa), total magnesium (tMg) and phosphorus (P), plasma glucose (Glu) and lactate (Lac) and the muscle enzymes creattine kinase (CK) and aspartate aminotransferase (AST) completed the evaluated parameters. The data for the variables studied were submitted to ANOVA and the Tukey test at 5% probability. After exercise (in T1), HR, RR, BT, PCV, TPP, Glu, Lac, Cre increased (P<0.05), there was also increase in phosphorus and decrease in K (P<0.05), but there was no hypophosphatemia and hipokalemia, respectively. After rest (in T2), only glucose returned to baseline levels (T0) and, with lower mean values, HR, RR, BT, PCV, K and Lac did not return to baseline levels (T0), while P decreased (P<0.05). Osm, BUN, Na, Cl, tCa, tMg, CK and AST did not change between times (P>0.05). It was concluded that the maximum intensity exercise performed on two barrel race courses caused mild hemoconcentration, did not changed plasma osmolality or concentrations of important electrolytes and muscle enzymes of horses, but caused hyperlactatemia without the plasma lactate concentrations being fully recomposed after resting.(AU)


O perfil bioquímico e o status de hidratação de equinos em treinamentos ou competições fornecem dados importantes das suas respostas fisiológicas e metabólicas em relação ao tipo específico de exercício em uma modalidade esportiva. No presente estudo, o status hídrico e o perfil fisiológico-bioquímico de equinos foram avaliados durante treinamento de três tambores, em três tempos (T): em repouso, antes do início da atividade física (T0); imediatamente após 10 minutos de aquecimento ao trote seguido de duas corridas na pista de tambor, intervaladas em 10 minutos (T1); e após uma hora de descanso do segundo percurso (T2). Os parâmetros avaliados foram: frequência cardíaca (FC), frequência respiratória (FR), temperatura corporal (BT) e cor da mucosa; o tempo de enchimento capilar (TEC), volume globular ou hematócrito (VG), proteínas plasmáticas totais (TPP), creatinina sérica (Cre) e a ureia (Ure) foram avaliados e utilizados para inferir o estado hídrico dos animais; a osmolaridade plasmática (Osm) também foi mensurada; as concentrações dos eletrólitos sódio (Na), potássio (K), cloreto (Cl), cálcio total (tCa), magnésio total (tMg) e fósforo (P), e das enzimas musculares foram mensurados no soro, e as de glicose (Gli) e lactato (Lac) no plasma. Os dados das variáveis estudadas foram submetidos à ANOVA e ao teste de Tukey a 5% de probabilidade. Após o exercício (T1), a FC, FR, TC, VG, TPP, Gli, Lac, Cre aumentaram. O P também aumentou (P<0,05), mas não alcançou nível de hiperfosfatemia, e o K diminuiu (P<0,05), mas não ao nível de hipocalemia. Embora com médias menores, a FC, FR, TC, VG, K e o Lac não retornaram aos níveis basais (T0) após o repouso (T2), apenas a Gli, enquanto o fósforo diminuiu (P<0,05). A Osm, ureia, Na, Cl, tCa, tMg, CK e AST não variaram entre os tempos (P>0,05). Concluiu-se que, o exercício de intensidade máxima realizado em dois percursos de três tambores causou leve hemoconcentração, não alterou a osmolaridade plasmática nem importantes concentrações de eletrólitos e de enzimas musculares dos equinos, porém, causou hiperlactatemia, sem completa recomposição da concentração do lactato após uma hora de descanso.(AU)

In young Cavalier King Charles spaniels (CKCS), intermittent mitral regurgitation (iMR; defined as moderate to severe mitral regurgitation [MR] in a small proportion of heartbeats), has been associated with an increased risk of cardiac death due to myxomatous mitral valve disease (MMVD). It is associated with increased R-R interval variability. Little is known about response to physiological factors and whether iMR is a precursor for developing significant MR. The aim of this study was to determine the effect of stress testing on the presence of iMR and heart rate, and short-term (1-2 year) progression of MR in CKCS with and without iMR. In total, 52 CKCS were included. Substudy 1 enrolled six dogs with iMR and 11 dogs without iMR. Substudy 2 enrolled 14 dogs with iMR and 28 dogs without iMR. Substudy 1 prospectively assessed the influence of stress testing on the presence of iMR and heart rate. Substudy 2 retrospectively evaluated short-term progression of iMR. During stress testing, iMR disappeared in 50% of CKCS and no iMR was recorded at mean heart rates >150 beats/min. Heart rate response did not differ between CKCS with or without iMR. CKCS with iMR did not have a higher odds (odds ratio = 5.2; 95% confidence interval, 0.7-38.2) of MR progression compared to controls (P = 0.1). In conclusion, physical stress influenced the occurrence of iMR in CKCS, but heart rate response was not different from CKCS without iMR. Intermittent mitral regurgitation did not significantly predict short-term MR progression. In stressed CKCS with early disease, iMR may be overlooked.

BACKGROUND: Despite the use of water treadmills (WT) in conditioning horses, the intensity of WT exercise has not been well documented. The workload on a WT is a function of water height and treadmill speed. Therefore, the purpose of this study was to determine the effects of these factors on workload during WT exercise. Fifteen client-owned Quarter Horses were used in a randomized, controlled study. Three belt speeds and three water heights (mid cannon, carpus and stifle), along with the control condition (dry treadmill, all three speeds), were tested. Measured outcomes were oxygen consumption (V̇O2), ventilation (respiratory frequency, tidal volume (VT)), heart rate (HR), and blood lactate. An ergospirometry system was used to measure V̇O2 and ventilation. Linear mixed effects models were used to examine the effects of presence or absence of water, water height and speed (as fixed effects) on measured outcomes. RESULTS: Water height and its interaction with speed had a significant effect on V̇O2, VT and HR, all peaking at the highest water level and speed (stifle at 1.39 m/s, median V̇O2 = 16.70 ml/(kg.min), VT = 6 L, HR = 69 bpm). Respiratory frequency peaked with water at the carpus at 1.39 m/s (median 49 breaths/min). For a given water height, the small increments in speed did not affect the measured outcomes. Post-exercise blood lactate concentration did not change. CONCLUSIONS: Varying water height and speed affects the workload associated with WT exercise. The conditions utilized in this study were associated with low intensity exercise. Water height had a greater impact on exercise intensity than speed.

BACKGROUND: Severity of pulmonary hypertension (PH) in dogs is related to clinical signs and prognosis. HYPOTHESIS/OBJECTIVES: We hypothesized that Doppler echocardiographic (DE) indices of pulmonary artery pressure (PAP) and pulmonary vascular resistance (PVR) are influenced by independent factors that create clinically important variability of DE-based estimates of PH in dogs. ANIMALS: Thirty-eight client owned dogs with naturally acquired degenerative atrioventricular valve disease and tricuspid regurgitation (TR). METHODS: Dogs were prospectively enrolled, and target variables were acquired during 4 echocardiographic study periods (lateral recumbency, standing, lateral recumbency after a 6-minute walk test [6MWT], and lateral recumbency after sedation with butorphanol 0.25 mg/kg IM). Statistical methods included repeated measures ANOVA, mixed model analysis, and Chi-squared test of association. RESULTS: There was a significant increase in peak TR flow velocity (TRFV; P < 0.01) after sedation in 78% of dogs, with TRFV increasing by >0.4 m/s in 42% of dogs, independent of stroke volume. A significant effect of study period on DE-estimated PVR was not found (P = 0.15). There were negligible effects of sonographer, body position, and 6MWT on echocardiographic variables of PH. Clinically relevant cyclic variation of TRFV was found. There was an association between estimation of right atrial pressure based on subjective assessment and estimation based on cranial vena cava collapsibility (P = 0.03). CONCLUSIONS AND CLINICAL IMPORTANCE: The increase in TRFV observed with sedation could change assessment of PH severity and impact prognostication and interpretation of treatment response. Further studies with invasive validation are needed.

OBJECTIVE: To determine the association of a radiographic score derived from tibial plateau angle (TPA) and femoral anteversion (FAA) with an outcome of cranial cruciate ligament deficiency (CCLD) in large dogs. STUDY DESIGN: Cross-sectional study. ANIMALS: 167 Labrador Retrievers. METHODS: Hind limbs of sound Labrador Retrievers over 6 years of age were considered at low risk for CCLD. Limbs were considered high risk for CCLD if they were affected or predisposed (sound contralateral limb in dogs with unilateral CCLD). The radiographic CCLD score was calculated for each limb. The TPA, FAA, and CCLD scores were compared between limbs of the same dog and between risk categories. A contingency table was used to evaluate the association of the CCLD score with the CCLD status of limbs. RESULTS: TPA, FAA, and CCLD scores were greater in limbs categorized as high risk for CCLD than in normal limbs. The sensitivity and specificity of the CCLD score was 87% and 79%, respectively. The positive predictive value was 69% and the negative predictive value was 92%. Scores were similar between paired right and left limbs, but did not agree for predicted status in 14/106 dogs. DISCUSSION: Our study supports an association between TPA, FAA, and CCLD in Labrador Retrievers. The negative predictive value of the CCLD score supports its application for screening dogs considered at low risk for CCLD. Positive CCLD scores should be interpreted with caution and the status of a dog may be undetermined if scores obtained on each limb disagree.

A previous report suggests a substantial incidence of exertional rhabdomyolysis (ER) in Arabian horses performing endurance racing. This study compared formalin histopathology and clinical and metabolic responses to a standardised field exercise test (SET) between Arabians with and without ER. Arabian horses with (n = 10; age 15.4 ± 5.6 years) and without (n = 9; 12.9 ± 6.1 years) prior ER were stall-rested for 24-48 h, after which paired ER and control horses were fitted with a telemetric ECG and performed a 47 min submaximal SET. Plasma glucose, lactate, electrolyte and total protein concentrations and packed cell volume were measured before and immediately after exercise. Blood and percutaneous gluteal muscle samples were also obtained before and 3 h after exercise for measurement of plasma creatine kinase (CK) activity and muscle glycogen concentration, respectively. Histopathologic analysis of formalin-fixed pre-exercise muscle sections was performed. Data were analyzed by ANOVA and non-parametric tests (P <0.05). No horses displayed clinical signs of ER during exercise, and plasma CK increased similarly in ER and control Arabians. Muscle glycogen, heart rate, and remaining plasma variables did not differ between horses with ER and control horses. Horses with ER had more internalised nuclei in mature myofibers, more aggregates of cytoplasmic glycogen and desmin, and higher myopathic scores than control horses. Although many horses with ER had histopathologic evidence of chronic myopathy, muscle glycogen concentrations and metabolic exercise responses were normal. Results did not support a consistent metabolic myopathy or a glycogen storage disorder in Arabians with ER.

OBJECTIVE: To compare effects of training on conventional and underwater treadmills on fiber properties and metabolic responses of the superficial digital flexor (SDF) and gluteal muscles to high-speed exercise in horses. SAMPLE: 6 unconditioned Quarter Horse-type horses. PROCEDURES: 6 horses were walked on underwater and conventional treadmills for 5 d/wk (maximum, 40 min/d) for 8 weeks in a randomized crossover design (60-day detraining period). Horses underwent a standardized exercise test (SET) at high speed before and after training. Analyte concentrations and fiber characteristics were measured in muscle biopsy specimens obtained from horses before and after each SET. RESULTS: Lactate concentration increased 2- to 3-fold in SDF and gluteal muscle after SETs. No training effect was identified on muscle fiber type composition, type II fiber diameter, muscle analyte concentrations, blood lactate concentration, or heart rate responses. Maximum diameters of type I fibers decreased significantly in gluteal muscle with conventional treadmill training and decreased in SDF muscle with both types of training, with maximum diameters greater for horses after underwater versus conventional treadmill training. No change was identified in minimum fiber diameters. CONCLUSIONS AND CLINICAL RELEVANCE: SETs involving near-maximal exertion resulted in an anaerobic response in SDF and gluteal muscles of horses. Eight weeks of conventional or underwater treadmill training resulted in minor changes in type I muscle fiber sizes, with no effect on muscle metabolic or heart rate responses to SETs. After rehabilitation involving underwater treadmills, training at progressing speeds is recommended for horses to develop the required fitness for speed work.

OBJECTIVE: To assess the effects of sodium pentosan polysulfate (PPS), N-acetyl glucosamine (NAG), and sodium hyaluronan (HA) in horses with induced osteoarthritis (OA). STUDY DESIGN: Experimental. ANIMALS: Adult Standard bred horses (n = 16). METHODS: OA was induced arthroscopically in 1 intercarpal joint; 8 horses were administered 3 mg/kg PPS, 4.8 mg/kg NAG, and 0.12 mg/kg HA (PGH), intravenously (IV), weekly and 8 horses were administered an equivalent volume of saline IV until study completion (day 70). Horses underwent a standardized treadmill exercise program. Clinical and radiographic findings and synovial fluid analysis were evaluated throughout the study. Macroscopic, histologic, histochemical, and biochemical findings were evaluated after necropsy. Comparisons of interest included OA and non-OA joints of saline treated horses and OA joints of PGH treated horses and OA joints of saline treated horses. Results were statistically analyzed with significance set at P < .05. RESULTS: OA caused increases in clinical assessment scores, synovial fluid variables, radiographic, macroscopic, and histologic cartilage scores, synovial fluid and cartilage chondroitin sulfate 846-epitope and glycosaminoglycan concentration. Total radiographic scores, total macroscopic joint pathology and macroscopic cartilage pathology scores were significantly reduced in horses treated with PGH compared with saline treated horses. Synovial fluid total protein concentration and white blood cell count were higher in OA joints of PGH treated horses compared with saline treated horses. There were no other significant differences between treatment groups. CONCLUSIONS: Improvements in macroscopic variables were not supported by other outcomes. Further evidence is needed before PGH can be recommended as a therapeutic option for osteoarthritis in horses.

BACKGROUND: Idiopathic pulmonary fibrosis (IPF) is an incurable interstitial lung disease occurring mainly in West Highland White Terriers (WHWTs). The effects of IPF on survival and on exercise tolerance in WHWTs are unknown. OBJECTIVES: To evaluate survival, prognostic factors, and exercise tolerance in WHWTs with IPF. ANIMALS: Privately owned WHWTs; 15 with IPF and 11 healthy controls. METHODS: Prospective case-control study conducted in 2007-2012. For survival, descriptive statistics and Kaplan-Meier (KM) survival curves with Cox proportional hazard ratios were performed. For the prognostic factor study, KM curves, Cox regression analysis, and logistic regression models were used. The 6-minute walk test (6MWT) was used for measurement of exercise tolerance. RESULTS: The median IPF-specific survival of deceased WHWTs (7/15) with IPF was 32 (range 2-51) months from onset of clinical signs. The risk of death from birth in WHWTs with IPF in age-adjusted Cox model was significantly higher (hazard ratio 4.6; 95% confidence interval 1.05-19.74, P = .04) than in control WHWTs. No significant prognostic factors were identified. In 6MWT, WHWTs with IPF walked a shorter distance, median 398 m (range 273-519 m), than healthy controls, median 492 m (420-568 m), P = .05, and the partial pressure of oxygen in arterial blood in diseased dogs had a moderate positive correlation with walking distance (Kendall's tau-b = 0.69, P = .06). CONCLUSION AND CLINICAL IMPORTANCE: IPF had a negative impact on life expectancy, but individual survival varied considerably. 6MWT proved to be a well-tolerated, noninvasive test to evaluate exercise tolerance.

BACKGROUND: Systolic dysfunction is associated with poor outcomes in dogs with myxomatous mitral valve disease. However, assessment of systolic variables by conventional echocardiographic methods is difficult in these dogs because of mitral regurgitation (MR). HYPOTHESIS: We hypothesized that assessment of systolic function by dobutamine stress may identify systolic dysfunction in dogs with MR, and that 2-dimensional speckle-tracking echocardiography (2D-STE) could quantitatively evaluate myocardial function. ANIMALS: Anesthetized dogs with experimentally induced MR. METHODS: Dogs were examined for systolic myocardial deformations using 2D-STE during dobutamine infusion before and 3 and 6 months after MR induction. We evaluated peak systolic rotation and rotation rate in each basal and apical view; peak systolic torsion and torsion rate were also calculated. RESULTS: Invasive peak positive first derivatives of left ventricular pressure (dp/dt) were significantly decreased in dogs 6 months after induction of MR compared with pre-MR results. After 3 and 6 months of MR, dogs had diminished peak systolic torsion values and torsion rates in response to dobutamine infusion compared with pre-MR results (3 months, P < .001 and P = .006; 6 months, P = .003 and P = .021). These results were significantly correlated with overall invasive dp/dt (r = 0.644, P < .001; r = 0.696, P < .001). CONCLUSIONS AND CLINICAL IMPORTANCE: Decreased torsion during dobutamine infusion in dogs with MR may reflect latent systolic dysfunction. Dobutamine infusion, therefore, may be useful for the assessment of systolic function in dogs with MR.

BACKGROUND: Few studies show the detrimental effect of canine obesity on cardiopulmonary function (CPF). The 6-Minute Walk Test (6MWT) is a noninvasive exercise test easy to perform in clinical settings. OBJECTIVE: The aim of this study was to investigate the effect of obesity and body weight loss (BWL) on CPF assessed by the 6MWT and arterial blood gas analysis. ANIMALS: Six experimental Beagles and 9 privately owned obese dogs were enrolled in a diet-induced BWL program. METHODS: Arterial blood gas analysis and 6MWT were repeated in obese subjects (BCS 8-9/9), in the middle of BWL (overweight, BCS 6-7/9), and in lean dogs (BCS 5/9). Heart rate (HRp) and oxygen saturation (SpO2 ) were measured by pulse oximetry before the 6MWT, at midtest, and during a 5-minute recovery period. RESULTS: Twelve dogs completed the BWL program (initial BW, 27.3 ± 2.9 kg; final BW, 20.85 ± 2.9, lsmeans ± SE, P ≤ .001). BWL caused a significant increase in 6MWT walked distance (WD; obese: 509 ± 35 m; overweight: 575 ± 36 m; lean: 589 ± 36 m; P ≤ .05). Resting arterial blood gas results were not influenced by BWL. Including all time points, obese dogs showed higher HRp and lower SpO2 compared to overweight and lean dogs. SpO2 at the end of the walk was significantly lower in obese dogs. CONCLUSION AND CLINICAL IMPORTANCE: Obesity negatively affects 6MWT performances in dogs. The 6MWT may be used to demonstrate the efficacy of BWL to improve CPF and quality of life in obese dogs. Although BWL induced significant improvement of cardiopulmonary parameters before ideal BW, WD improved until the end of the BWL program.

Fish repeatedly experience stressful situations under experimental and aquaculture conditions, even in their natural habitat. Fish submitted to sequential stressors can exhibit accumulation or habituation on its cortisol response. We posed a central question about the cortisol response profiles after exposure to successive acute stressors of a similar and different nature in Rhamdia quelen. We have shown that successive acute stressors delivered with 12-h, 48-h, and 1-week intervals provoked similar cortisol responses in juvenile R. quelen, without any habituation or accumulation. The cumulative stress response is more associated to short acute stressors with very short intervals of minutes to hours. In our work, we used an interval as short as 12h, and no cumulative response was found. However, if the length of time between stressors is of a day or week as used in our work the most common and an expected phenomenon is the attenuation of the response. Thus, also, the absence of both accumulation of the stress response and the expected habituation is an intriguing result. Our results show that R. quelen does not show habituation or accumulation in its stress responses to repeated stressors, as reported for other fish species.


Os peixes, tanto em condições de cultivo quanto em condições experimentais ou na natureza, podem experimentar situações de estresse. Peixes submetidos a estressores sequenciais podem apresentar acumulação de sua resposta de cortisol com aumento dos níveis desse hormônio em cada resposta sucessiva ou habituação da resposta do cortisol, com redução dos níveis de cortisol em cada resposta sequencial. Abordamos a questão central de como é o perfil de resposta do cortisol de R. quelen após a exposição a estressores agudos sucessivos de natureza semelhante e diferente. Os dados mostraram que sucessivos estressores agudos com intervalos de 12 e 48h e de uma semana, provocaram respostas de cortisol semelhantes em juvenis de R. quelen, sem qualquer habituação ou acumulação. No presente trabalho usamos intervalos tão curtos quanto 12h e não verificamos qualquer tipo de acumulação da resposta. No entanto, se o intervalo entre estressores forem de um dia ou semana, o fenômeno mais comum e esperado é o da atenuação da resposta. Assim, tanto a ausência de acumulação da resposta ao estresse quanto à ausência da habituação esperada são resultados interessantes. Nossos resultados mostraram que juvenis de R. quelen não mostram habituação ou acumulação em suas respostas de estresse a estressores repetidos, como relatado para outras espécies de peixes.

The aim of this study was to illustrate the surgical implantation of Swan Ganz introducers into the jugular vein, as well as the pulmonary artery cannulation procedure using ten mixed breed (MB) adult horses. We also proposed to demonstrate pulmonary arterial pressure (PAP) values measured through the implanted Swan Ganz during one incremental submaximal exercise test performed on a treadmill. Surgical implantation of the Swan Ganz introducers has demonstrated to be easy and quick to perform. None of the animals showed signs of discomfort following the surgery and ergometric test was carried out as proposed. PAP increased proportionally with exercise intensity, and the major average values were recorded at the higher effort time points. The PAP average values increased (P<0.05) when treadmill speed reached 7.5m/s (39±3mmHg) and 8.5m/s (41±3mmHg), comparing to average values at rest (26±3mmHg). However, the PAP values observed at the maximal effort performed by the MB horses of this trial (41±3mmHg), are under the values described for other breeds at similar effort tests. The technique described in the present study contributes for the standardization of research protocols using arterial pulmonary access via Swan Ganz catheter implantation in horses. The ergometric test was effective to promote PAP changes according to the effort, showing PAP values for MB horses that can guide further investigations.


Objetivou-se ilustrar a técnica cirúrgica de implantação de introdutores para cateter de Swan Ganz na veia jugular, bem como o procedimento de canulação da artéria pulmonar de dez equinos adultos sem raça definida (SRD). Realizando medidas através do cateter de Swan Ganz implantado, objetivou-se ainda demonstrar os valores de pressão arterial pulmonar (PAP) durante um teste de esforço progressivo de intensidade submáxima realizado em esteira rolante. A técnica cirúrgica de implantação de introdutores para o cateter de Swan Ganz empregada, demonstrou-se de fácil e rápida realização. Os animais não apresentaram complicações frente ao introdutor implantado e o teste ergométrico foi realizado conforme proposto. A PAP se elevou ao longo do exercício seguindo o incremento de velocidade, sendo os maiores valores médios observados nos momentos de maior intensidade do esforço. Os valores médios de PAP aumentaram (p<0,05) às velocidades de 7,5m/s (39±3 mmHg) e 8,5m/s (41±3 mmHg), quando comparados aos valores mensurados no repouso (26±3mmHg). Entretanto, os valores de PAP encontrados no máximo de esforço realizado pelos equinos SRD deste ensaio (41±3mmHg), encontram-se abaixo dos valores descritos para outras raças para a mesma intensidade de esforço. A descrição detalhada da técnica ilustrada no presente trabalho, pode contribuir substancialmente na padronização de protocolos experimentais que propõem o acesso arterial pulmonar por meio da implantação de cateter de Swan Ganz em equinos. O teste ergométrico proposto foi eficaz em promover variações de PAP de acordo com o esforço realizado, demonstrando valores para equinos SRD que poderão ser usados como base para futuras avaliações.

The effects of three different blood-guided conditioning programmes on ultrastructural and histochemical features of the gluteus medius muscle of 2-year-old sport horses were examined. Six non-trained Haflinger horses performed three consecutive conditioning programmes of varying lactate-guided intensities [velocities eliciting blood lactate concentrations of 1.5 (v1.5 ), 2.5 (v2.5 ) and 4 (v4 ) mm respectively] and durations (25 and 45 min). Each conditioning programme lasted 6 weeks and was followed by a 5-week resting period. Pre-, post- and deconditioning muscle biopsies were analysed. Although training and detraining adaptations were similar in nature, they varied significantly in magnitude among the three different conditioning programmes. Overall, the adaptations consisted in significant increases in size of mitochondria and myofibrils, as well as a hypertrophy of myofibrillar ATPase type IIA muscle fibres and a reduction in number of type IIx low-oxidative fibres. Together, these changes are compatible with a significant improvement in both muscle aerobic capacity and muscle strength. The use of v1.5 and v2.5 as the exercise intensities for 45 min elicited more significant adaptations in muscle, whereas conditioning horses at v4 for 25 min evoked minimal changes. Most of these muscular adaptations returned towards the pre-conditioning status after 5 weeks of inactivity. It is concluded that exercises of low or moderate intensities (in the range between v1.5 and v2.5 ) and long duration (45 min) are more effective for improving muscle features associated with stamina and power in sport horses than exercises of higher intensity (equivalent to v4 ) and shorter duration (25 min).

OBJECTIVE: To compare kinematics, net moments, powers, ground and joint reaction forces (GRF, JRF) across the hock, stifle and hip joints in Labrador Retrievers at low risk or predisposed to cranial cruciate ligament disease (CCLD). STUDY DESIGN: Cross-sectional clinical study. ANIMALS: Clinically normal Labradors predisposed (n = 11) or at low risk for CCLD (n = 9). METHODS: Right pelvic limbs were classified as predisposed or not to CCLD using a predictive score equation based on combining tibial plateau angle (TPA) and femoral anteversion angle (FAA) measured on radiographs. Kinematic, GRF, and morphometric data were combined in an inverse dynamics approach to compute hock, stifle and hip kinematics, net moments, powers, and JRF while trotting. RESULTS: The extensor moment of the hock joint was greater in predisposed limbs compared with nonpredisposed limbs (0.37 versus 0.28 Nm/kg). The power generated around the hock and stifle joints in predisposed limbs was increased (0.69 versus 0.44 W/kg for the hock, 1.59 versus 1.05 W/kg for the stifle). Trotting velocity, stance time, vertical and craniocaudal GRF and JRF did not differ between groups. CONCLUSIONS: Extensor moment at the hock was increased in predisposed limbs compared with nonpredisposed limbs. Predisposed limbs generated more energy than nonpredisposed limbs around the hock and stifle joints.
