RESUMO

In this article, we introduce and analyze an emerging polycentric governance model for addressing wastewater challenges in textile industrial parks in China. Unlike the conventional two-tier model with government and polluters only, the emerging model emphasizes multi-actor participation and presents new possibilities in enhancing the cost-effectiveness and alleviating the compliance deficiency. We introduce the model through a case study of the Binhai textile industrial park. The features of the model include levering the subject of monitoring, allowing collaborative wastewater treatment, employing the administrative power of local governing bodies, and engaging multiple local actors. To explain its formation, we adopt event sequence analysis, and point out the important role of local government as a facilitator and a regulator in the externally generated collaborative governance. We identified its success factors with pattern matching based on polycentric governance theories. We find the involvement of varied actors in the decision-making process of internal wastewater emission standard could lead to applicable policy making with high acceptance, which contributes to the environmental performance. To initiate the transition towards the new model, three conditions are required, including policy support from the government, the establishment of monitoring systems, and capacity building for actors.

In the present study, we performed a cross-sectional survey to determine the occurrence and genotype distribution of T. gondii DNA in soil samples collected from different sources from six geographic regions in China. Between March 2015 and June 2017, 2100 soil samples were collected from schools, parks, farms and coastal beaches, and examined for T. gondii DNA using three PCR assays targeting 529-bp repeat element (RE) sequence, B1 gene and ITS-1 gene sequences. Also, we investigated whether geographic region, soil source and type, and sampling season can influence the prevalence of T. gondii DNA in the soil. Soil samples collected from farms and parks had the highest prevalence, whereas samples collected from school playgrounds and coastal beaches had the lowest prevalence. PCR assays targeting 529-bp RE and ITS-1 gene sequences were more sensitive than the B1 gene-based assay. Positive PCR products were genotyped using multi-locus PCR-RFLP, and ToxoDB #9 was the predominant genotype found in the contaminated soil samples. Multiple logistic regression identified factors correlated significantly with the presence of T. gondii DNA in the soil to be the source of the soil, including farms (odds ratio 3.10; 95% confidence interval [CI], 1.52 to 6.29; p = 0.002) and parks (2.59; 95% CI 1.28 to 5.27; p = 0.009). These results show that Chinese soil hosts T. gondii of the most prevalent genotype in China (ToxoDB#9) and that the soil type influences infection patterns.

RESUMO

We describe a visible dye-based staining method for DNA in polyacrylamide gels using ethyl violet (EV). The novel method is a background-free, sensitive, economical, and simple procedure involving only staining and washing steps that can be completed within 30 min. As little as 0.8-1.6 ng of phiX174 DNA/HaeIII can be detected by EV, which is about eightfold more sensitive than Nile blue (NB) stain and twofold less sensitive than ethidium bromide (EB) stain.

A convenient silver staining method for protein in sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) gels is described. The method is previsible, sensitive, and mass spectrometry (MS) compatible. Two visible counter ion dyes, ethyl violet (EV) and zincon (ZC), were used in the first staining solution with a detection limit of 2 to 8 ng/band in approximately 1h. The dye-stained gel can be further stained by silver staining, which is based on acidic silver staining employing ZC with sodium thiosulfate as silver ion sensitizers. Especially, ZC has silver ion reducing power by cleavage of the diazo bond of the dye during silver reduction. The second silver staining can be completed in approximately 1h with a detection limit of 0.2 ng/band.

RESUMO

One simple but efficient carbon-supplying device was designed and developed, and the correlative carbon-supplying technology was described. The absorbing characterization of this device was studied. The carbon-supplying system proved to be economical for large-scale cultivation of Spirulina sp. in an outdoor raceway pond, and the gaseous carbon dioxide absorptivity was enhanced above 78%, which could reduce the production cost greatly.
