Safety assessment requires information on both the chronic and acute effects of chemicals. Chemical materials such as food additives have become one of the most critical and compelling issues in the continuing debate on food safety. Calcium lactate (CL) and Sodium lactate (SL) are approved chemicals used in various products. The present study, in vitro study, was designed to evaluate the cyto-genotoxicity effects of CL and SL on human umbilical vein endothelial cells (HUVECs). The cytotoxicity effect of these additives was determined by MTT and Annexin V-FITC apoptosis assays. Besides, genotoxicity parameters such as morphological change of DNA and DNA fragmentation were studied with DAPI staining and DNA ladder assays, respectively. The results showed that the growth of HUVECs decreased upon treatment with CL at higher concentrations, but SL did not significantly alter HUVECs cell number. Annexin V-FITC apoptosis assays revealed that CL could induce cell death based on necrosis rather than apoptosis. In SL, the early and late apoptosis was not considerable in treated cells. Exposure to CL and SL did not lead to morphological change and DNA fragmentation in the HUVECs cell line. Overall, it is concluded from these results that CL and SL have not been cytotoxic and genotoxic effects in low concentrations in human umbilical vein endothelial cells in vitro.

Considering the importance of vegetables as a source of micronutrients and fibers in a balanced diet, there is still a concern that vegetables could also be a source of toxic heavy metal contaminants. The study aimed to determine the concentrations of lead (Pb), cadmium (Cd), arsenic (As), chromium (Cr), copper (Cu), nickel (Ni), and zinc (Zn) in the salad vegetables sold in Tabriz city, Iran, and to evaluate the probabilistic health risk assessment. The amount of toxic metals in 240 samples was evaluated by atomic absorption spectrophotometer (AAS) method. The average levels of toxic metals in the samples were found to be 1.59, 1.26, 1.42, 4.89, 13.38, 1.01, and 32.65 mg/kg for Pb, Cd, As, Cr, Cu, Ni, and Zn, respectively. According to the results, Zn and Cu had the highest concentration, whereas the lowest concentration belonged to Ni. The rank order of the toxic elements in the samples based on target hazard quotient (THQ) values was Cr > Cd > As > Pb > Cu> Zn > Ni, for both females and males. Leafy vegetables had a higher amount of total target hazard quotient (TTHQ) than tuber crops for both males and females. The carcinogenic risks of As and Pb were 0.032 and 0.03 in the females and 0.22 and 0.19 in males. According to the finding, there may be a potential risk of toxic metals, especially Cr, Cd, and As, for both females and males in Tabriz through the consumption of vegetables.

Purpose: Ascorbyl palmitate (AP) is a widely used food additive in food industry. In this study, AP was evaluated for potential cyto-genotoxicity on Human Umbilical Vein Endothelial Cells (HUVECs). Methods: MTT assay and flow cytometry analysis was used for cytotoxicity evaluation, while genotoxicity was carried out using DAPI staining assays and real time PCR. Results: The growth of HUVECs was decreased upon treatment with AP in dose-and time-dependent manner. Early/late apoptosis percentage in HUVECs treated with this additive was detected using flow cytometry analysis. Also morphology of DAPI stained HUVECs clearly showed chromatin fragmentation. Furthermore, real time PCR results showed that AP induces apoptosis by up-regulation of caspase-3, 9 and down-regulation of Bcl-2 ratio. Conclusion: The present results indicated that AP has capability to induce apoptosis in HUVECs and its better to make a thorough analysis about its extensive application in food industry.

Cytotoxicity and genotoxicity of sodium acetate (SA), sodium diacetate (SDA), and potassium sorbate (PS) was tested on Human Umbilical Vein Endothelial Cells (HUVEC). Cytotoxicity was investigated by MTT assay and flow cytometry analysis, while genotoxicity was evaluated using DNA fragmentation and DAPI staining assays. The growth of treated HUVECs with various concentrations of SA, SDA and PS decreased in a dose-and time-dependent manner. The IC50 of 487.71, 485.82 and 659.96 µM after 24 h and IC50 of 232.05, 190.19 and 123.95 µM after 48 h of treatment were attained for SA, SDA and PS, respectively. Flow cytometry analysis showed that early and late apoptosis percentage in treated cells was not considerable. Also neither considerable DNA fragmentation nor DNA smear was observed using DAPI staining and DNA ladder assays. Overall, it can be concluded that the aforementioned food additives can be used as safe additives at low concentration in food industry.

There is limited evidence on the effects of prebiotics on inflammation. Therefore, the aim of this study was to evaluate the effects of inulin supplementation on inflammatory indices and metabolic endotoxemia in patients with type 2 diabetes mellitus. The participants included diabetic females (n = 49). They were divided into an intervention group (n = 24) as well as a control group (n = 25) and received 10 g/d inulin or maltodextrin for 8 weeks, respectively. Fasting blood sugar (FBS), HbA1c, insulin, high-sensitive C-reactive protein (hs-CRP), tumor necrosis factor-alpha (TNF-α), interleukin-10 (IL-10), and plasma lipopolysaccharide (LPS) were measured pre and post intervention. Inulin-supplemented patients exhibited a significant decrease in FBS (8.5%), HbA1c (10.4%), fasting insulin (34.3%), homeostasis model assessment of insulin resistance (HOMA-IR) (39.5%), hs-CRP (35.6%), TNF-α (23.1%), and LPS (27.9%) compared with the maltodextrin group (p < 0.05). Increase in IL-10 was not significant in inulin compared with the maltodextrin group. It can be concluded that inulin supplementation seems to be able to modulate inflammation and metabolic endotoxemia in women with type 2 diabetes.
