BACKGROUND: Melioidosis is a serious bacterial infection caused by Burkholderia pseudomallei, a gram-negative bacterium commonly found in soil and water. It can affect both humans and animals, and is endemic in regions such as Southeast Asia and Northern Australia. In recent years, there have been reports of an emergence of human melioidosis in other areas, including New Caledonia. RESULTS: During standard laboratory analysis in New Caledonia in 2021, a strain of B. pseudomallei was isolated from a goat. The strain was characterized using both MLST and WGS techniques and was found to cluster with previously described local human strains from the area. In parallel, several serological tests (CFT, ELISA, Luminex (Hcp1, GroEL, BPSS1840), arrays assay and a latex agglutination test) were performed on animals from the farm where the goat originated, and/or from three other neighboring farms. Using two commercial ELISA kits, seropositive animals were found only on the farm where the infected goat originated and tests based on recombinant proteins confirmed the usefulness of the Hcp1 protein for the diagnosis of melioidosis in animals. CONCLUSIONS: Despite the regular reports of human cases, this is the first confirmed case of melioidosis in an animal in New Caledonia. These results confirm the presence of the bacterium in the region and highlight the importance of vigilance for both animal and human health. It is critical that all health partners, including breeders, veterinarians, and biologists, work together to monitor and prevent the spread of the disease.

The choice of the aggregation that defines the temporal unit of epidemiological surveillance is part of the more theoretical framework of the modifiable temporal unit problem (MTUP). It has been demonstrated that this choice influences temporal cluster detection and may lead to false-positive results and poor estimation of regression model parameters. In syndromic surveillance (SyS), despite the choice of which temporal aggregation to use being crucial, it has not yet been addressed in the literature. In most SyS systems, this choice is driven by the frequency of the data collection and/or human resources available, although neither the temporal unit's influence on the performance of anomaly detection algorithms nor on the efficiency of the SyS are known.The main objective of our study was to analyze the influence of the temporal aggregation unit on the performances of SyS detection algorithms used routinely, according to the characteristics of specific syndromes and outbreaks. Simulating daily time series of various syndromes, we tested three different time series aggregation methods. For each of four anomaly detection algorithms and their variants, we calculated seven performance indicators and multi-criteria scores to guide epidemiologists in their choice of which temporal aggregation of surveillance to use. From 19,200 analyzed time series, we observed an effect of temporal aggregation on the performance of the detection algorithms tested. Results also showed that the time aggregation unit was linked to the detection algorithm used, and that strong aggregation-algorithm interactions need to be taken into account when deciding on which aggregation-algorithm pair to use. Using theoretical data, our study also showed that no one ideal aggregation-algorithm pair exists for all contexts when deciding on which temporal unit of surveillance to use, and that the choice depends on several parameters.Our results can help public health practitioners choose the most appropriate time series aggregation and algorithm according to their specific needs. Finally, the present work enabled us to develop recommendations for a One Health project where the same time aggregation type and detection method could be used for both human and animal syndromic surveillance data.

Foot and mouth disease (FMD) is a highly contagious viral disease that affects domestic and wild artiodactyl animals and causes considerable economic losses related to outbreak management, production losses and trade impacts. In Tunisia, the last FMD outbreak took place in 2018-2019. The effectiveness of control measures implemented to control FMD depends, in particular, on the human resources used to implement them. Tunisia has the ultimate objective of obtaining OIE status as 'FMD-free with vaccination'. The aim of this study was to determine and compare the necessary and available human resources to control FMD outbreaks in Tunisia using emergency vaccination and to assess the gaps that would play a role in the implementation of the strategy. We developed a resources-requirement grid of necessary human resources for the management of the emergency vaccination campaign launched after the identification of a FMD-infected premises in Tunisia. Field surveys, conducted in the 24 governorates of Tunisia, allowed quantifying the available human resources for several categories of skills considered in the resources-requirement grid. For each governorate, we then compared available and necessary human resources to implement vaccination according to eight scenarios mixing generalised or cattle-targeted vaccination and different levels of human resources. The resources-requirement grid included 11 tasks in three groups: management of FMD-infected premises, organisational tasks and vaccination implementation. The available human resources for vaccination-related tasks included veterinarians and technicians from the public sector and appointed private veterinarians. The comparison of available and necessary human resources showed vaccination-related tasks to be the most time-consuming in terms of managing a FMD outbreak. Increasing the available human resources using appointed private veterinarians allowed performing the emergency vaccination of animals in the governorate in due time, especially if vaccination was targeted on cattle. The overall approach was validated by comparing the predicted and observed durations of a vaccination campaign conducted under the same conditions as during the 2014 Tunisian outbreak. This study could provide support to the Tunisian Veterinary Services or to other countries to optimise the management of a FMD outbreak.

This study aimed at estimating, in a prospective scenario, the potential economic impact of a possible epidemic of WNV infection in Belgium, based on 2012 values for the equine and human health sectors, in order to increase preparedness and help decision-makers. Modelling of risk areas, based on the habitat suitable for Culex pipiens, the main vector of the virus, allowed us to determine equine and human populations at risk. Characteristics of the different clinical forms of the disease based on past epidemics in Europe allowed morbidity among horses and humans to be estimated. The main costs for the equine sector were vaccination and replacement value of dead or euthanised horses. The choice of the vaccination strategy would have important consequences in terms of cost. Vaccination of the country's whole population of horses, based on a worst-case scenario, would cost more than EUR 30 million; for areas at risk, the cost would be around EUR 16-17 million. Regarding the impact on human health, short-term costs and socio-economic losses were estimated for patients who developed the neuroinvasive form of the disease, as no vaccine is available yet for humans. Hospital charges of around EUR 3,600 for a case of West Nile neuroinvasive disease and EUR 4,500 for a case of acute flaccid paralysis would be the major financial consequence of an epidemic of West Nile virus infection in humans in Belgium.

Aspergillus fumigatus remains a major respiratory pathogen in birds and treatment is still difficult. We challenged different groups of few-day-old turkeys via intratracheal aerosolisation with increasing concentrations (10(5) up to 10(8)) of conidia using a MicroSprayer(®) device. The fungal burden was assessed by real-time PCR, galactomannan dosage, CFU counting and histopathological evaluation in order to provide a comparison of these results within each inoculum groups. Significant mortality, occurring in the first 96h after inoculation, was only observed at the highest inoculum dose. Culture counts, GM index and qPCR results on the one hand and inoculum size on the other hand appeared to be clearly correlated. The mean fungal burden detected by qPCR was 1.3log10 units higher than the mean values obtained by CFU measurement. The new model and the markers will be used to evaluate the efficacy of antifungal treatments that could be used in poultry farms.

Infection due to the mold Aspergillus fumigatus remains a common and life-threatening infection in many animals, especially birds. Animal models are still required to better understand the physiopathology of infection and evaluate diagnostic tools and treatment procedures. The aim of the present study was to assess the pathogenicity of A. fumigatus in two lineages of chicken (Gallus gallus): SPF White Leghorn PA12 layers and conventional JA657 broilers. Four-day-old birds were experimentally infected in an inhalation chamber in order to reproduce a "natural" contamination and to obtain a large repartition of conidia into the respiratory tract. Half of the chicks were injected subcutaneously with dexamethasone for 4 days before the infective challenge. At days 0 and 7, the effects of chicken lineage and immunosuppressive treatment on pulmonary fungal burden were analyzed using two linear mixed models. The pathogenicity of A. fumigatus varied according to the lineage: no clinical signs and no mortality were observed in layer chickens whereas more than 50% of mortality occurred in broilers. The effect of immunosuppressive treatment was also demonstrated, notably on animals weight but also on mortality.

Current knowledge suggests that there is a low-level and recurrent circulation of West Nile virus (WNV) in Europe, with sporadic human and/or equines cases. However, recent events indicate that this picture is changing, raising the possibility that Europe could experience a modification in the virus' circulation patterns. We used an existing model of WNV circulation between Southern Europe and West Africa to estimate the sample size of equivalent West Nile surveillance systems, either passive (based upon horse populations and sentinel veterinarians) or active (sentinel horses, sentinel chickens, or WNV genome detection in trapped mosquito pools). The costs and calendar day of first detection of these different surveillance systems were compared under three different epidemiological scenarios: very low level circulation, low level recurrent circulation, and epidemic situation. The passive surveillance of 1000 horses by specialized veterinarian clinics appeared to be the most cost-effective system in the current European context, and estimated median dates of first detection appeared consistent with recent field observations. Our results can be used to optimize surveillance designs for different epidemiological requirements.

Fungal elements represent a significant part of the biological contaminants that could be detected in the air of animal facilities. The aim of this study was to assess the relative efficiencies of two air sampling methods and three culture conditions for the quantification of airborne culturable fungi in a poultry farmhouse in France. Air samples were collected every week throughout a 15-week period. Two devices were simultaneously used-a rotative cup air sampler (CIP 10-M, Arelco, France) and an air sampler based on filtration (AirPort MD8, Sartorius, Germany). Culture of airborne viable fungi was performed on malt extract agar (ME) and dichloran glycerol-18 (DG18) at 25 or 37°C. CIP 10-M and AirPort MD8 were shown to display comparable performances but significant differences were observed between culture conditions for Aspergillus spp. (p<0.01), Scopulariopsis spp. (p=0.02) and unidentified molds (p<0.01).

BACKGROUND: The public health threat represented by a potential circulation of bovine spongiform encephalopathy agent in sheep population has led European animal health authorities to launch large screening and genetic selection programmes. If demonstrated, such a circulation would have dramatic economic consequences for sheep breeding sector. In this context, it is important to evaluate the feasibility of qualification procedures that would allow sheep breeders demonstrating their flock is free from scrapie. Classical approaches, based on surveys designed to detect disease presence, do not account for scrapie specificities: the genetic variations of susceptibility and the absence of live diagnostic test routinely available. Adapting these approaches leads to a paradoxical situation in which a greater amount of testing is needed to substantiate disease freedom in genetically resistant flocks than in susceptible flocks, whereas probability of disease freedom is a priori higher in the former than in the latter. The goal of this study was to propose, evaluate and compare several qualification strategies for demonstrating a flock is free from scrapie. RESULTS: A probabilistic framework was defined that accounts for scrapie specificities and allows solving the preceding paradox. Six qualification strategies were defined that combine genotyping data, diagnostic tests results and flock pedigree. These were compared in two types of simulated flocks: resistant and susceptible flocks. Two strategies allowed demonstrating disease freedom in several years, for the majority of simulated flocks: a strategy in which all the flock animals are genotyped, and a strategy in which only founders animals are genotyped, the flock pedigree being known. In both cases, diagnostic tests are performed on culled animals. The less costly strategy varied according to the genetic context (resistant or susceptible) and to the relative costs of a genotyping exam and of a diagnostic test. CONCLUSION: This work demonstrates that combining data sources allows substantiating a flock is free from scrapie within a reasonable time frame. Qualification schemes could thus be a useful tool for voluntary or mandatory scrapie control programmes. However, there is no general strategy that would always minimize the costs and choice of the strategy should be adapted to local genetic conditions.

In France, the considerable economic losses due to paratuberculosis introduction in cattle herds may justify the development of a herd-level certification procedure. For a cost/benefit analysis purpose, a simple deterministic and stochastic simulation model for intra-herd paratuberculosis transmission has been developed to evaluate the economic consequences of the purchase of a single infected heifer in a French average herd. The values of the epidemiological parameters were provided by a panel of French paratuberculosis experts. The results were in adequacy with field observations. A sensitivity analysis was conducted. The model was however difficult to validate rigorously, since few data on the intra-herd paratuberculosis true prevalence level are currently available.

Paratuberculosis has received increasing attention in France because of the important losses this disease may provoke. The use of certification schemes has proven its effectiveness for the protection of healthy herds against diseases transmitted mainly by trade. The economic justification of such schemes in the particular case of paratuberculosis is studied, for French cattle herds, using a cost/benefit approach. The basic economical hypotheses and estimates have been proposed and carefully examined by a working group composed of paratuberculosis experts and field specialists. By adopting the point of view of a breeder that buys animals, we first estimated the benefits resulting from the non-introduction of the disease. They were then compared with the costs resulting from the fact that the vendor reports its own certification costs on the price of the animals he sells. Two average herds (the mean French beef herd and the mean French dairy herd), and two certification levels were studied. The results show that, currently, the use of the certification is not very economically profitable in French cattle herds. This conclusion, however should be reappraised if the certification costs decrease, for example with the commercialization of diagnostic tests on mixtures.
